TwoVibrio species identified asV. damsela and a new sucrose-positiveVibrio sp.,V. carchariae sp. nov., were simultaneously isolated from a brown shark which died while being held in captivity at a large aquarium. Pathogenicity studies were subsequently conducted using a variety of elasmobranchs, including smooth dogfish and lemon sharks. Both bacterial strains proved pathogenic, causing death in nearly all of the elasmobranch hosts challenged. Virulence studies revealed that both bacterial strains were cytotoxic for Y-1 mouse adrenal cells. TheV. damsela strain was highly cytotoxic causing Y-1 cellular damage at culture supernatant dilutions up to 1 : 128. Both strains were hemolytic, but neither exhibited the Kanagawa phenomenon. They were both capable of urea hydrolysis, an interesting trait, considering that elasmobranchs retain large (ca 300 milliosmolal) urea concentration in their tissue.  相似文献

Three species of Antarctic mites, Alaskozetes antarcticus, Hydrogamasellus antarcticus and Rhagidia gerlachei, are abundant in the vicinity of Palmer Station, Antarctica. No single mechanism for reducing water stress was shared by all three species. A. antarcticus and R. gerlachei (both ca. 200 μg) are over twice as large as H. antarcticus (ca. 90 μg), but all had similar body water content (67%) and tolerated a loss of up to 35% of their body water before succumbing to dehydration. All imbibed free water and had the capacity to reduce water loss behaviorally by forming clusters. Alaskozetes antarcticus was distinct in that it relied heavily on water conservation (xerophilic classification) that was largely achieved by its thick cuticular armor, a feature shared by all members of this suborder (Oribatida), and abundant cuticular hydrocarbons. In comparison to the other two species, A. antarcticus was coated with 2–3× the amount of cuticular hydrocarbons, had a 20-fold reduction in net transpiration rate, and had a critical transition temperature (CTT) that indicates a pronounced suppression in activation energy (E a) at temperatures below 25°C. In contrast, H. antarcticus and R. gerlachei lack a CTT, have lower amounts of cuticular hydrocarbons and have low E as and high net transpiration rates, classifying them as hydrophilic. Only H. antarcticus was capable of utilizing water vapor to replenish its water stores, but it could do so only at relative humidities close to saturation (95–98 %RH). Thus, H. antarcticus and R. gerlachei require wet habitats and low temperature to counter water loss, and replace lost water behaviorally through predation. Compared to mites from the temperate zone, all three Antarctic species had a lower water content, a feature that commonly enhances cold tolerance.  相似文献

Summary The water balance of three different sized coexisting species of heteromyid rodents (Dipodomys merriami ca. 39 g;Perognathus fallax ca. 23 g;Perognathus longimembris ca. 9 g) was assessed while consuming two different diets (either wheat or hulled sunflower) at ambient temperatures of 15–30°C. The metabolism of wheat as the sole food source was calculated to provide a greater metabolic water production (MWP) than the consumption of sunflower seed because of their different composition. The state of water balance was assessed by measuring urine concentrations and body weight maintenance on each diet at each temperature. Both measures showed that (i) all species were able to maintain a more positive water on the higher MWP seed, (ii) for all species there was an ambient temperature above which water balance could no longer be maintained, (iii) that this temperature was higher with the higher MWP food source and (iv) water regulatory efficiency was negatively correlated with body mass.Dipodomys showed a reduced digestive efficiency compared toPerognathus. When presented with both seedsDipodomys showed no preference for either seed irrespective of the state of water balance whilst thePerognathus species showed a tendency for an increased preference for the high MWP food source at the higher ambient temperatures. A number of cationic derivatives of cholesterol containing polar residues of N-methylimidazole, pyridine, N-methylmorpholine, and 4-N,N-dimethyaminopyridine were synthesized by the interaction of the corresponding heterocyclic bases with cholesterol 5-bromopentanoate followed by the treatment with methyl iodide in the case of tertiary amines. In order to ascertain whether the major species of continental antarctic macrolichens are photosynthetically active during summer conditions, the chlorophyll fluorescence of three lichen species [Umbilicarin decussata (Vill. Zahlbr., Pseudephebe minuscula (Ny-l. ex Arnold) Brodo and Hawksw. and Usnea sphacetala R. Br.] was monitored in the vicinity of Casey Station. Wilkes Land, continental Antarctica using a PAM-2000 modulated fluorescence system. Lichens were studied when in equilibrium with the atmosphere as well as when moistened by snow showers. Photochemical quantum yield was estimated as ΔF/F′m and related to thallus water content as well as microclimatic conditions. Lichens were photosynthetically active only when moistened by snow fall or by run-off from snow melt. The levels of photosynthetic activity in the field for all species were influenced by microenvironmental conditions and patterns in response were site and species specific. Highest levels of photosynthetic efficiency occurred when thalli were at intermediate water contents. Photosynthetic activity was reduced by cold as well as warm, bright conditions. Highest thallus water contents occurred during the middle of the day after substantial “falls of snow. P. minuscula maintained highest thallus water contents at all sites and appears to have a high water compensation point which is related to its observed distribution patterns. Umbilicaria decussata studied in the laboratory did not become photosynthetically active even when exposed to 95% relative humidity (RH) for 51 h and. when dehydrating after artificial wetting, showed an optimum thallus water content for photosynthesis of ca 90% dry weight and a thallus water compensation point of about 35% dry weighl. In the field U. decussata did not become pholosynthetically active except when moistened by snow. Usnea sphacelata exposed to the atmosphere had low thallus water contents (ca 30%) which was not related to RH. The results indicate that the lichens are photosynthetically inactive for most of the summer period and are totally reliant on snow as a water supply. This i.s important when modelling carbon gain and growth rates of continental antarctic lichens.  相似文献

Soil samples from an agricultural field contaminated with 10 ppm14C-benz(a)anthracene in glass tubes were brought into contact with cultures of wood-rotting fungi, precultivated on wheat straw substrate. Forty-five strains of white-rot fungi and four brown-rot fungi were tested for their ability to colonize the soil and to mineralize14C-benz(a)anthracene to14CO2 within a 20-week incubation time. Twenty-two white-rot fungi and all brown-rot fungi were unable to colonize the soil. Twenty-three strains of white-rot fungi, all belonging to the genusPleurotus, colonized the soil. During the experiment the noncolonizing fungi and their substrate disintegrated more and more to a nonstructured pulp from which water diffused into the soil. The same phenomenon was observed in the control which contained only straw without fungus and contaminated soil. In samples with colonizing fungi the substrate as well as the mycelia in the soil remained visibly unchanged during the entire experiment. Surprisingly, most samples with fungi not colonizing the soil and the control without fungus liberated between 40 and 58 % of the applied radioactivity as14CO2 whereas the samples with the colonizing fungi respired only 15–25 % as14CO2. This was 3–5 times more14CO2 than that liberated from the control (4.9 %) which contained only contaminated soil without straw and fungus. A similar result was obtained with selected colonizing and noncolonizing fungi and soil contaminated with 10 ppm14C-pyrene. However, in pure culture studies in which14C-pyrene was added to the straw substrate,Pleurotus sp. (P2), as a representative of the colonizing fungi, mineralized 40.3 % of the added radioactivity to14CO2. The noncolonizing fungiDichomitus squalens andFlammulina velutipes liberated only 17.2 or 1.7 %, respectively, as14CO2. These results lead to the hypothesis that the native soil microflora stimulated by the formed products of straw lysis is responsible for high degradation rates found with noncolonizing fungi.  相似文献

A review is presented on the occurrence of macroalgae in dune pools, ditches and moorland pools in The Netherlands, which were studied in the period 1980–1987. There are clear effects of eutrophication on biomass as well as on species composition. A general trend is that under mesotrophic or moderately eutrophic conditions, the filamentous macroalgae form a diversified component of the primary producers in shallow water bodies. Especially spore forming genera asSpirogyra andOedogonium can be represented with up to about 20 species per site. In hardwater ditches alsoVaucheria is an important component, especially the sediment dwellingVaucheria dichotoma. Under distinctly eutrophic conditions dense floating algal masses contain only one or a few species asCladophora glomerata, Enteromorpha intestinalis, Hydrodictyon reticulatum. Under hypertrophic conditions the water surface is covered by dense blanketing mats of duckweeds in summer and autumn. Besides trophic factors, other factors as alkalinity and chloride concentration play a role in species composition. The results are discussed in the framework of water quality assessment of polder waters and possible applications for water purification or use as manuring substance. The need for experimental ecophysiological studies is underlined.  相似文献

Seven Gram-negative bacterial strains, capable of using lupanine, the predominant quinolizidine alkaloid in white lupin, as sole carbon and energy source, were isolated from soil in whichLupinus albus andL. luteus had been grown. A metabolic profile system (BIOLOG) identified only three of the seven isolates, two asXanthomonas oryzae pvoryzae E and one asGluconobacter cerinus. The maximum specific growth rates of the seven isolates when incubated at 27°C in a medium containing as sole carbon source 2 g L–1 of lupanine, ranged from 0.05 to 0.13 h–1 and the concentration of dry biomass at the stationary phase ranged from 0.7 to 1.1 g L–1. Unidentified strains IST20B and IST40D exhibited the highest maximum specific growth rates (0.13h–1), removed 99% of the initial lupanine after 30 h of incubation, and the dry biomass yields did not exceed 0.4 g per g lupanine consumed. Strain IST20B is of potential use forL. albus debittering because, after 32 h growth in aqueous extracts ofL. albus, 85% of initial alkaloids were removed while the concentration of soluble protein was only reduced by 8%.  相似文献

A novel biocatalytic reaction of transamidation of non-activated amides with amines is reported. Among 45 different lipolytic and proteolytic enzymes tested, only the lipase from Candida antarcticawas able to catalyze this reaction. The reaction proceeded with up to ca. 80% conversion in anhydrous methyl tert-butyl ether and worked with both N-substituted and unsubstituted amides. The biocatalytic transamidation is an equilibrium process and, therefore, higher conversions to the desired amide were achieved by using increased concentrations of the amine nucleophile.  相似文献

Abstract

Various comb-type copolymer containing a polycation as a main chain was design to construct delivery systems of DNAs. The comb-type copolymers having cell-specific polysaccharides were proved to be useful to deliver DNA to the target cells in vivo. Of interest, the copolymers with abundant side chains of hydrophilic polymers are capable of stabilizing DNA triplex. Further, injectable nanoparticles for controlled releases of DNAs were fabricated from the copolymer and a biodegradable polymer.  相似文献

In chernozem soil, vanillin was decomposed via vanillic and protocatechuic acid before the aromatic ring opened. The rate curves of oxygen consumption for the oxidation of vanillin were seen to have more than one maximum. During incubation of the soil with vanillin, the number of bacteria increased, especially those capable of utilizing vanillin as the sole carbon source. Of the 21 such strains isolated, 15 were identified asPseudomonas sp., five asCellulomonas sp. and one asAchromobacter sp. It was found that the course of the oxidation of vanillin varied at different p.H values and in different strains was found that the course of the oxidation of vanillin varied at different p.H values and in different strains of bacteria. In some cases, the phase of the oxidation of vanillin to vanillic acid was clearly differentiated from the subsequent decomposition of vanillic acid.  相似文献

In the Netherlands peat was excavated for fuel until 1950. This gave rise to waterbodies (called turf ponds) which were then colonized by aquatic plants. Succession resulted in different aquatic plant communities and more terrestrialized stages such as floating fens. Nature conservation authorities started to excavate new turf ponds in 1990(ca. 2 ha y–1) with the aim to restore calcareous, mesotraphent ecosystems by totally setting back succession. A sequence of new species was revealed by mapping the aquatic vegetation from 1990 onwards.Chara spp. proved early colonizers, which was not expected because they have not been present in ditches and ponds in the area for the last 20 years. The denseChara vegetation prevents the resuspension of organic soil and contributes to keep the water column nutrient-poor and clear. ability of species such asStratiotes aloides to colonise the ponds from adjacent waterbodies is not possible because no open contact exists between a turf pond and a ditch. Management measures, such as re-introduction, have to be considered if the full-range of aquatic plant communities remains the goal.  相似文献

Scenedesmus acutus contains about 10 major amines and at least 20 other amines which are present in very small quantities. The following amines were identified by mass spectrometry after separation of the trifluoroacetyl derivatives by gas-liquid chromatography and of the dansyl 2 derivatives by thin-layer chromatography: methylamine, dimethylamine, ethylamine, ethanolamine, putrescine, cadaverine, spermidine, N-(3-aminopropyl)-1,3-diaminopropane, N-(4-aminobutyl)-1,4-diaminobutane, 2-phenylethylamine, tyramine, piperazine, adenine, and γ-butyrolactam. The methods applied for the analyses of these amines are described and discussed.  相似文献

This paper aims at comparing inedited maps of macrophytes and phytoplankton distribution in the lagoon of Venice in 1980 and 2003. The macrophyte distribution is displayed with reference to different biomass intervals which allow the calculation of the occupied surface, standing crop (SC), net (NPP) and gross (GPP) production. In 1980 the total macroalgal SC was ca. 841 ktonnes whereas the annual NPP and GPP were estimated to be ca. 2912 and 18498 ktonnes, respectively. In 2003 macroalgae displayed a marked regression and the SC, NPP and GPP decreased to ca. 89, 471 and 2336 ktonnes. Maps of the seagrass distribution date back to 1990 but their biomass and production have been quantified only in 2003. On the whole, in 2003, on a surface of ca. 56 km2 the three species living in the lagoon accounted for a SC and a NPP of ca. 209 and 821 ktonnes, whereas the GPP estimated according to literature was about twice as high as the NPP. Cymodocea nodosa was the most abundant phanerogam in the lagoon. Its SC was higher than the total of the macroalgae, although the latter were more productive. Zostera marina covered the highest surface in the lagoon but its biomass and production were a little lower than that of C. nodosa. Nanozostera noltii, which was common and widespread in the past, displayed an evident regression trend which was mainly due to the increase of the water turbidity and the disruptions of its habitat. Similarly, phytoplankton, underwent a descending trend, although data quoted in the present paper display its distribution only in the summer period.  相似文献

Summary The effects of water restriction on nitrogen metabolism were compared in the semi-arid adapted tammar wallaby (Macropus eugenii) and a wallaby from a moist forest environment, the red-necked pademelon (Thylogale thetis).On a medium (9 to 13%) protein diet water restriction depressed dry matter and nitrogen intakes in both species. Nitrogen balance remained positive, but fell further in the pademelons. Urea excretion decreased in the tammars, but increased in the pademelons. Urea recycling as a proportion of urea entry rate tended to increase in the tammars but not in the pademelons. These findings suggest thatT. thetis is not as well adapted asM. eugenii to coping with water shortages.In a second experiment water restriction depressed nitrogen balance in tammars on both high (15%) and low (5%) protein diets, but only on the latter diet did nitrogen balance become negative. Urea recycling was greater on the low than on the high protein diet, but was unaffected by water restriction. Although better able to withstand water stress,M. eugenii was unable to cope with both water stress and a low protein diet together. The reported ability ofM. eugenii to maintain water intake by drinking sea water during the dry season when fresh water is unavailable and vegetation is of low protein content is thus of great ecological significance.  相似文献

In the design of polyethyleneimine derivatives for use as catalysts and binding media, the placement of reactive and hydrophobic groups previously has been limited by the specificity of the addition reaction. In this paper is described a protocol to limit the sites of addition of nucleophiles and long-chain alkanes to tertiary amines and the less reactive of the secondary amines. Three blocking groups for the primary and secondary amines were tested, but only trifluoroacteylating reagents left the polymer reactive to substitution on the tertiary amines with halogenated alkanes. The secondary amines that resisted trifluoroacetylation were blocked with either tert-butyloxy carbonate or trimethylsilyl carbonate. The tertiary amines were quaternized with iodododecane or dodecyl benzyl chloride. After removal of the trifluoroacetyl groups, the polymer amines were inactivated by methylation, which proceeded to 93% completion. The 7% of the amines that were not quaternized were largely tertiary, since propylene sulfide, which reacts only with secondary and primary amines, was substituted onto the polymer only to the extent of 0.2% total amine, as quantified by the indirect method of P. H. Butterworth, F. Baum, and J. W. Porter (1967, Arch. Biochem. Biophys., 118, 716–723). The sulfhydryl group did not oxidize over at least 14 days. This is the first stable sulfhydryl-containing synthetic polymer that has been reported.  相似文献

Five species ofAspergillus were isolated from post-harvest decays of pears, apples, peaches, grapes, strawberries, melons and tomatoes.A. niger was the most common species and was isolated from all types of fruits. Next in frequency wasA. flavus, which was infrequently isolated from stored grapes, tomatoes and peaches.A. wentii, A. ochraceus andA. tamarii were each cultured from only one type of stored fruit.All the fruit-decay Aspergilli were typical inhabitants of the atmospheres of store-rooms. Of these,A. niger was by far the most ubiquitous species, comprising about 80% of the total airborne Aspergilli. Each of the four other pathogenic Aspergilli comprised 0.2–0.4% of the total. However, some of the most common airborne Aspergilli such asA. versicolor (6.6% of the total), have never been isolated from any fruit decay and were also very limited in their potential pathogenicity.The highest potential pathogenicity was attributed toA. niger, which was capable of infecting all inoculated fruits except eggplants, followed by a 2–3-day incubation period. The other Aspergilli cultured, pathogenic as well as airborne, showed selective potential pathogenicity when artificially inoculated into the fruit. Fruit tissues of apples, pears and tomatoes, followed by those of peaches and grapes, were found the most suitable for Aspergilli development, whereas strawberries and, more so, eggplants were mostly resistant to infection.  相似文献

Aim Hybridization is a common and potent mechanism of plant evolution that has the potential to be evolutionary significant in its own right, and hybrids are common between invasive and native congeneric species. Our aims were to document the existence and nature of new Spartina hybrids arising between introduced Spartina densiflora and native S. maritima in tidal marshes of the Iberian Peninsula, to examine the actual and potential range of hybrids in the intertidal zone, and to analyse the seed set of hybrids. Location South‐West Iberian Peninsula. Methods Hybrids were characterized using chloroplast and nuclear DNA, and ploidy assessments. The ecological tolerance of the hybrids was studied using vegetation surveys and transplant experiments. Results We found that both parental species have been seed parents to hybrids and that all hybrids had an additive pattern of species‐specific nuclear markers consistent with F1 hybrids. Hybrid chromosome numbers varied between 2n = ca. 65 and 2n = ca. 94–97, while S. maritima had 2n = ca. 60 and S. densiflora had 2n = ca. 70. Hybrids grew in three discrete locations along the intertidal zone but were capable of growing throughout the ranges of both parental species in transplanted gardens, and in most cases, grew better than the parental species. While the potential exists for the origination of another invasive Spartina hybrid species, thus far hybrid plants are not fertile, limiting their ability to invade and spread. Main conclusions We recommend the eradication of all of the currently quite limited hybrid cordgrass and to fight the S. densiflora invasion in the Iberian Peninsula to prevent the origination of a new invasive allopolyploid Spartina species.  相似文献

Phototrophic sulfur bacteria were isolated from Black-Sea mud at depths of 660 and 2,240 m. The species obtained in pure cultures were identified asChromatium warmingii andThiocapsa roseopersicina. In addition,Chlorobium phaeovibrioides occurred in enrichment cultures. The findings prove that Chromatiaceae and Chlorobiaceae are capable of survival in 2000 m depth, in the dark, in the presence of H2S and organic materials. There is, however, no evidence for growth or reproduction of these organisms in this environment.  相似文献
