Oriented fibres of extracted nucleohistone were employed as test material in a study of satisfactory fixation, embedding, and staining methods for structures containing a high proportion of nucleic acid. Fixation in buffered osmium tetroxide solution at pH 6, containing 10-2 M Ca++, and embedding in Araldite enabled sections of the fibres to be cut in which the orientation was well preserved. These could be strongly stained in 2 per cent aqueous uranyl acetate, and showed considerable fine structure. Certain regions in the nuclei of whole thymus tissue could also be strongly stained by the same procedure, and were identical with the regions stained by the Feulgen procedure in adjacent sections. Moreover, purified DNA was found to take up almost its own dry weight of uranyl acetate from 2 per cent aqueous solution. Strongest staining of whole tissue was obtained with very short fixation times-5 minutes or so at 0°C. Particularly intense staining was obtained when such tissue stained in uranyl acetate was further stained with lead hydroxide. Although the patterns of staining by lead hydroxide alone and by uranyl acetate were similar in tissues fixed for longer times (½ hour to 2 hours, at 0°C or 20°C), in briefly fixed material the DNA-containing regions appeared relatively unstained by lead hydroxide alone, whilst often there was appreciable staining of RNA-containing structures. Observations on the staining of some viruses by similar techniques are also described.  相似文献

Summary Tissues of rats given 3H-oestradiol were prepared for autoradiography according to methods commonly used in light and electron microscopy.By formalin fixation large amounts of radioactive material were lost, both in the fixative and during dehydration. Altogether 78.6±7.5 per cent was extracted from uterine tissue, while 49.0±4.6 per cent was lost from liver tissue removed 15 minutes after the injection. Significantly more radioactivity was lost in the fixative from liver tissue than from uterine. In the former fixation accounted for about 60 per cent of the loss, whereas in the latter it was responsible for about 25 per cent.Osmium tetroxide fixation was found to retain the radioactivity of liver and uterine tissue almost completely. However, large amounts were invariably extracted during dehydration. Although only 3.9±1.2 per cent of the radioactivity of uterine tissue diffused into the fixative, 72.8±12.4 per cent was extracted during ethanol dehydration. A heavy loss was also registered when dehydration and infiltration were carried out in glycol methacrylate.Glutaraldehyde perfusion and postfixation with osmium tetroxide retained almost completely the radioactivity of uterine and pituitary tissue. Nevertheless, nearly all of it was extracted during ethanol/propylene oxide dehydration and Epon embedding.The methods studied are not adequate for accurate autoradiographic localization of oestradiol.This work was supported by grants from The Norwegian Cancer Society and by Nordisk Insulinfond. Rat liver mitochondria allowed to accumulate maximal amounts of Ca++ and HPO4= ions from the suspending medium in vitro during respiration have a considerably higher specific gravity than normal mitochondria and may be easily separated from the latter by isopycnic centrifugation in density gradients of sucrose or cesium chloride. When the mitochondria are allowed to accumulate less than maximal amounts of Ca++ and HPO4= from the medium, they have intermediate specific gravities which are roughly proportional to their content of calcium phosphate. Maximally "loaded" mitochondria are relatively homogeneous with respect to specific gravity. Correlated biochemical and electron microscopic studies show that Ca++-loaded mitochondria contain numerous dense granules, of which some 85 per cent are over 500 A in diameter. These granules are electron-opaque not only following fixation and staining with heavy metal reagents, but also following fixation with formaldehyde, demonstrating that the characteristic granules in Ca++-loaded mitochondria have intrinsic electron-opacity. The dense granules are almost always located within the inner compartment of the mitochondria and not in the space between the inner and outer membranes. They are frequently located at or near the cristae and they often show electron-transparent "cores." Such granules appear to be made up of clusters of smaller dense particles, but preliminary x-ray diffraction analysis and electron diffraction studies have revealed no evidence of crystallinity in the deposits. The electron-opaque granules decrease in number when the Ca++-loaded mitochondria are incubated with 2,4-dinitrophenol; simultaneously there is discharge of Ca++ and phosphate from the mitochondria into the medium.  相似文献

Methyl green-pyronin staining has been used for localization of RNA and DNA in chick and mouse embryonic tissues and in insect larval salivary glands. Glutaraldehyde or tricholoracetic acid-lanthanum acetate (TCA-LA) was used as fixative and paraffin wax or Araldite was used as embedding medium. For good results the following are specially desirable: fixation with 2.5% glutaraldehyde, dehydration in alcohols for short time, and the use of fresh staining solutions. After TCA-LA fixation the final results are much less specific. The digestion with RNAse appears essential for the detection of RNA because pyronin does not seem to be entirely specific to RNA. The results show that glutaraldehyde a common fixative for electron microscopic work, is particularly suitable fixative for light microscopic cytochemical investigations if followed by methyl green-pyronin staining; furthermore, methyl green-pyronin staining after glutaraldehyde fixation can be carried out on Araldite sections.  相似文献

Thin strips of frog ventricle were isolated and bathed for 15 min in a solution containing 140 mM KCl, 5 mM Na2ATP, 3 mM EDTA, and 10 mM Tris buffer at pH 7.0. The muscle was then exposed to contracture solutions containing 140 mM KCl, 5 mM Na2ATP, 1 mM MgCl2, 10 mM Tris, 3 mM EGTA, and CaCl2 in amounts to produce concentrations of free calcium from 10-4.8 M to 10-9 M. The muscles developed some tension at approximately 10-8 M, and maximum tension was achieved in 10-5 M Ca++. They relaxed in Ca++ concentrations less than 10-8 M. The development of tension by the EDTA-treated muscles was normalized by comparison with twitch tension at a stimulation rate of 9 per min before exposure to EDTA. In 10-5 M Ca++ tension was always several times the twitch tension and was greater than the contracture tension of a frog ventricular strip in KCl low Na-Ringer. Tension equal to half-maximum was produced at approximately 10-6.2 M Ca++. Intracellular recording of membrane potential indicated that after EDTA treatment the resting potential of cells in Ringer solution with 10-5 M Ca or less was between 5 and 20 mv. Contracture solutions did not produce tension without prior treatment with EDTA. The high permeability of the membrane produced by EDTA was reversed and the normal resting and action potentials restored in 1 mM Ca-Ringer. Similar studies of EDTA-treated rabbit right ventricular papillary muscle produced a similar tension vs. Ca++ concentration relation, and the high permeability state reversed with exposure to normal Krebs solution.  相似文献

1. Reasons are given for believing that the uptake of Na+, Cl-, and NaCl by the crayfish occurs through the gills. 2. A crayfish in fresh water, with a Cl concentration of about 0.2 mEq./l., can) by active Cl absorption, compensate entirely for Cl lost in the urine. 3. The carbonic anhydrase activity of the gills is markedly higher than that of other tissues of the crayfish, but the equivalent CO2 output of the crayfish is far in excess of the equivalent Cl absorption per unit time and weight and thus fails to warrant the supposition that Cl absorption is of respiratory importance. 4. The carbonic anhydrase activity of the soft integument of the lobster, before and after molting, and of the hypodermis of the hard-cuticled animal is almost identical and of the same order as that of other tissues of the lobster. 5. The concentration of the electrolytes was about 7.5 mEq./l.; i.e., considerably lower than in the blood of the crayfish. Cl- can be taken up independently of the complementary cation. Na+ can be taken up independently of the complementary anion. K+ and SO4 = are not taken up at all. In pure NaCl, the Na+ and Cl- are absorbed evidently largely together. Ca++ is absorbed only in newly molted animals and in animals preparing to molt but is not absorbed by hard-cuticled animals not preparing to molt. Ca++ is taken up independently of Cl- in pure CaCl2. 6. Newly molted animals absorb Ca++ at a rate exceeding that of the absorption of other absorbable ions (Na+ and Cl-) in the same equivalent concentration. 7. A crayfish utilizes the Ca++ in fresh water in the calcification of its cuticle. Since the animal does not swallow water, the Ca++ must enter through the exterior. Reasons are given for believing that, unlike Na+ and Cl-, Ca++ is absorbed directly from the exterior by the integument and does not enter the body through the gills. 8. During molting, only about 4 per cent of the raw ash and 2.3 per cent of the organic material of the old cuticle is resorbed.  相似文献

Correlation of the localization of La+++ with its effects on Ca++ exchange in cultured rat heart cells is examined with the use of a recently developed technique. 75% of cellular Ca++ is exchangeable and is completely accounted for by two kinetically defined phases. The rapidly exchangeable phase has a t ½ = 1.15 min and accounts for 1 1 mmoles Ca++/kg wet cells or 43% of the exchangeable Ca++ (cells perfused with [Ca++]o = 1 mM) Phase 2 has a t ½ = 19.2 min and accounts for 1.5 mmoles Ca++/kg wet cells or 57% of the exchangeable Ca++. 0.5 mM [La+++]o displaces 0 52 mmoles Ca++/kg wet cells—all from phase 1—and almost completely abolishes subsequent Ca++ influx and efflux The presence of La+++ in the washout converts the washout pattern to a single phase system with a t ½ = 124 min. The effects upon Ca++ exchange are coincident with abolition of contractile tension but regenerative depolarization of the tissue is maintained Electron microscope localization of the La+++ places it exclusively in the external lamina or basement membrane of the cells. Summary Primary cultures of embryonic chick pectoral skeletal muscle were used to study calcium regulation of myoblast fusion to form multinucleated myotubes. Using atomic absorption spectrometry to measure total cellular calcium and the45Ca-exchange method to determine free cellular Ca++, our data suggest that only the free cellular calcium changes significantly during development under conditions permissive for myotube formation (0.9 mM external Ca++). Increases in calcium uptake occurred before and toward the end of the period of fusion with the amount approximating 2 to 4 pmol per cell in mass cultures. If the medium [Ca++] is decreased to 0.04 mM, as determined with a calcium electrode, a fusion-block is produced and free cell Ca++ decreased 5- to 10-fold. Removal of the fusion-block by increasing medium [Ca++] results in a release of the fusion-block and an increase in cellular Ca++ to approximately 1 pmol per cell during fusion, and higher thereafter. Cation ionophore A23187 produced transient increases in cellular calcium and stimulated myoblast fusion and the final extent of myotube formation only when added at the onset of culture. Results suggest that transient increased calcium uptake alone is insufficient for fusion because critical cellular content in conjunction with permissive amounts of medium [Ca++] must exist. The latter suggests further that cell surface Ca++ was also critical.  相似文献

The membrane excitability and contraction were examined in single barnacle muscle fibers with different internal Ca++ concentrations by using buffer solutions made up with EGTA and Ca-gluconate in various proportions. During the passage of dc currents the membrane shows all-or-none spike potentials for internal Ca++ concentrations below about 8 x 10-8 M, oscillatory potential changes in the range between 8 x 10-8 to 5 x 10-7 M, but neither oscillatory nor spike potentials were seen for concentrations above 5 x 10-7 M. All-or-none spike potentials were suppressed when the internal Mg++ concentration exceeded 5 mM. The suppression threshold of the internal Ca++ concentration for the Sr spike is much higher than that for the Ca spike. The threshold concentration of internal Ca++ for contraction was about 8 x 10-7 M.  相似文献

Summary The Cu++ retaining power of the three soils used in our experiments was found to be of the order: alkali soil > black soil > red soil. The alkali soil retained the applied Cu++ in basic copper carbonate and hydroxide forms due to its high carbonate (soluble + insoluble) and high pH values, and the red soil retained the least amount of Cu++ because of its low pH value and negligible carbonate content, whilst the black soil, being fairly rich in CaCO3, organic matter and suitable pH, occupied an intermediate position.When the original samples were treated with H2O2, H2O2 + HCl or were ignited at 600°C for 1 hour the retention of applied Cu++ decreased more or less as a result of destruction of organic matter, carbonate and dehydration of sesquioxides leaving an inert material.Saturation of original soils with H+ (by HCl) resulted in lower Cu++-retention, whilst the conversion of H-soils to Ca++-soils showed a higher Cu++-retention but never approached the amount of Cu retained by original soils. This is due to lowering of pH of the samples, removal of carbonates as well as due to antagonistic effect of H+-ions. A greater percentage of the Cu++ retained by these samples exists in the exchangeable forms in comparison to original soils.It has also been observed that addition of CaCO3, at the rate of 1 to 2 per cent (to the hydrogen samples) resulted in a precipitation of practically all the applied Cu++ and non-existence of exchangeable forms of Cu++.  相似文献

Fragmented sarcoplasmic reticulum (FSR) membranes isolated from rabbit skeletal muscle are impermeable to inulin-14C (mol wt 5,000), and dextran-14C (mol wt 15,000–90,000) at pH 7.0–9.0, yielding an excluded space of 4–5 µl/mg microsomal protein. In the same pH range urea and sucrose readily penetrate the FSR membrane. EDTA or EGTA (1 mM) increased the permeability of microsomes to inulin-14C or dextran-14C at pH 8–9, parallel with the lowering of the FSR-bound Ca++ content from initial levels of 20 nmoles/mg protein to 1–3 nmoles/mg protein. EGTA was as effective as EDTA, although causing little change in the Mg++ content of FSR. The permeability increase caused by chelating agents results from the combined effects of high pH and cation depletion. As inulin began to penetrate the membrane there was an abrupt fall in the rate of Ca++ uptake and a simultaneous rise in ATPase activity. At 40°C inulin penetration occurred at pH 7.0 with 1 mM EDTA and at pH 9.0 without EDTA, suggesting increased permeability of FSR membranes. This accords with the higher rate of Ca++ release from FSR at temperatures over 30°C. The penetration of microsomal membranes by anions is markedly influenced by charge effects. At low ionic strength and alkaline pH acetate and Cl are partially excluded from microsomes when applied in concentrations not exceeding 1 mM, presumably due to the Donnan effect. Penetration of microsomal water space by acetate and Cl occurs at ionic strengths sufficiently high to minimize charge repulsions.  相似文献

A hyperpolarization can be recorded intracellularly following either a single bright light stimulus or the intracellular injection of Na+. This after-hyperpolarization is abolished by bathing in 5 x 10-6 M strophanthidin or removal of extracellular K+. Both treatments also lead to a small, rapid depolarization of the dark-adapted cell. When either treatment is prolonged, light responses can still be elicited, although with repetitive stimuli the responses are slowly and progressively diminished in size. The rate of diminution is greater for higher values of [Ca++]out; with [Ca++]out = 0.1 mM, there is almost no progressive diminution of repetitive responses produced by either K+-free seawater or strophanthidin. We propose that an electrogenic Na+ pump contributes directly to dark-adapted membrane voltage and also generates the after-hyperpolarizations, but does not directly generate the receptor potential. Inhibition of this pump leads to intracellular accumulation of sodium ions, which in turn leads to an increase in intracellular Ca++ (provided there is sufficient extracellular Ca++). This increase in intracellular calcium probably accounts for the progressive decrease in the size of the receptor potential seen when the pump is inhibited.  相似文献

The process of fixation of DNA-containing plasms is investigated by macroscopical and electron microscopical observations on solutions of DNA, nucleohistones, as well as on bacterial nuclei. The following treatments were found to produce a gelation of a solution of DNA or nucleohistones: (a) OsO4 fixation at pH 6 in the presence of amino acids (tryptone) and Ca++. (b) Exposure to aqueous solutions of uranyl acetate. (c) Exposure to aqueous solutions of indium chloride. Observed in the electron microscope, these gels show a fine fibrillar material. From experiments in which solutions of DNA or nucleohistones are mixed with bacteria and treated together, it is concluded that the behavior of the bacterial nucleoplasm is similar to that of the DNA solutions. The appearance of birefringence indicates that uranyl acetate and indium chloride produce an orientation of the molecules of a DNA solution during gelation. Bacterial chromosomes fixed by these agents also show a certain order, while those fixed by the OsO4-amino acid-Ca++ formula do not. Whether or not the order can be considered to be artificial is discussed, and a tentative conclusion is presented: (a) Uranyl acetate may induce artificial order. (b) Fixatives which do not gel DNA probably result in the grossest artifacts. (c) OsO4 fixation at pH 6 in the presence of amino acids (tryptone) and Ca++ may give the most accurate preservation of the in vivo disposition of DNA (RK+ fixation).  相似文献

The role of calcium in excitation-contraction coupling of lobster muscle   总被引:2,自引:1,他引:1  
Potassium contractures were induced in lobster muscle bundles under conditions which produced varying KCl fluxes into the fibers. The presence or absence of chloride fluxes during depolarization by high concentrations of potassium, had no effect on the tensions developed. The curve relating tension to the membrane potential had a typical sigmoid shape with an apparent "threshold" for tension at -60 mv. Soaking the muscles in low (0.1 mM) calcium salines for 30 min completely eliminated the potassium contractures but the caffeine contractures were only slightly reduced under these conditions. The potassium contracture could be completely restored in less than 2 min by return of the calcium ions to the saline. Evidence is presented for independent, superficial, and deep calcium sites; the superficial sites appear to be involved in the coupling mechanisms associated with potassium contractures. These sites are highly selective for Ca++, and attempts to substitute either Cd++, Co++, Mg++, Ba++, or Sr++ for Ca++ were unsuccessful. However, K+ appeared to compete with Ca++ for these sites, and the evoked tension could be reduced by prestimulation of the muscle fibers with high K+ salines. The results of studies on the influx of 45Ca during potassium contractures were compatible with the view of muscle activation by the entry of extracellular calcium.  相似文献

Abstract

A stable recombinant Chinese hamster ovary (CHO) cell model system expressing the human type-1 receptor for parathyroid hormone and parathyroid hormone-related peptide (hPTH-R) was established for the analysis of human PTH (hPTH) variants. The cell lines showed receptor expression in the range from 105 to 1.9xl06 receptors per cell. The affinity of the receptors for hPTH-(l-34) was independent of the receptor number per cell (K<j = 8 nmol/1). The induction of cAMP by hPTH-(l-34) is maximal in clones expressing >2xl05 receptors per cell and Ca++ signals were maximal in cell lines expressing >1.4xl06 receptors per cell. Second messenger specific inhibitors demonstrated that PTH-induced increases in intracellular cAMP and Ca++ are independent and Ca++ ions are derived from intracellular stores. The cAMP-specific receptor activator hPTH-(l-31) showed also an increase in intracellular Ca++. Even in cell lines expressing more than 10 receptors per cell the Ca++/PKC specific activator hPTH-(28-48) did not activate hPTH-Rs. Based on these results, synthesis of further derivatives of PTH is required to identify pathway-specific ligands for the type-1 hPTH-R.  相似文献

In Chaos chaos streaming, motility, and cytokinesis were inhibited nearly 100% for several hours by 2.5–5 mM sodium adenosine triphosphate (ATP)1 added to culture fluid. All three effects were completely prevented by the addition of equimolar Mg++ or Ca++ ions but not Na+ to the ATP/culture fluid solution. The effects of ATP were not reproduced by EDTA, EGTA, colchicine, or AMP. Sodium pyrophosphate produced about 50% inhibition at 5 mM. Studies with 14C-ATP showed that 5 x 10-5 to 5 x 10-4 mmole of ATP was firmly associated with each milliliter of packed cells after an hour''s incubation at 24°C. Labeling studies also showed that prevention of the ATP effects by Mg++ ions was not due to a decrease in the amount of ATP associated with the cells.  相似文献

Pancreatic ribosomes (guinea pig) aggregate and lose upon treatment with polyamines, particularly spermine, their bound secretory enzymes. Spermine, at 0.5 mM, for example, causes the release of about 85 per cent of the chymotrypsinogen and RNase, and from 85 to 100 per cent of the ribosomal amylase. At the same time, the particles lose about 10 per cent of their RNA, 7 to 24 per cent of their total protein, and from 75 to 100 per cent of their Mg++. Observations with the electron microscope confirm the heavy agglutinating of the ribosomes but otherwise show little change in the structure of the particles. Using radioactive spermine it was found that, concomitant with the loss of bound enzymes and Mg++ from the ribosomes, spermine became bound to the particle. The extent of binding ranged from 0.29 to 1.49 µmoles per 10µmoles RNA-P. The bound radioactive spermine can be removed by subsequent treatment of the ribosomes with GTP, ATP, or P-P, which treatment also removes most of the RNA of the particles, leaving behind ribosomes with a much lower RNA/protein ratio. From this evidence it was inferred that spermine, in releasing the Mg++ of the particle, becomes salt-linked to the free phosphate hydroxyl groups of the RNA. Freshly isolated pancreatic and hepatic ribosomes contain very little spermine, about 0.1 to 0.2 µmoles polyamine/10 µmoles RNA-P. The results are discussed in terms of the linkages between the structural protein, the bound secretory enzymes, and the RNA of the ribosomes.  相似文献

Experiments have been performed on the apyrase activity of E. coli, strain B. Although the dependence on pH and substrate is similar to that of rat tissue, the bacterial extracts are inhibited by Ca++ and stimulated by Mg++. In bacterial extracts the rate of phosphate release decreases in the course of the reaction, possibly owing to product inhibition. With multiple bacteriophage infection, the apyrase activity of the intact cells increased several fold, and the activity of extracts increased about 30 per cent. It is suggested that the changes could be attributed to an increase in the amount of enzyme although other alternatives cannot be precluded at present.  相似文献

The possible role of calcium ions in the induction of ornithine decarboxylase (ODC) in rat hepatoma cells in culture (HTC) has been investigated by manipulating cellular calcium levels as follows: a) use of the calcium chelating agent EGTA to inhibit induction of ODC by dibutyryl cyclic AMP (cAMP), b) addition of Ca++ to reverse the inhibition of cAMP induction of ODC by EGTA, c) use of a calcium ionophore in the presence of Ca++ to induce ODC. In each case there was positive evidence for the participation of Ca++ in the induction of ODC.  相似文献
