Seasonal changes of fatty acid composition and thermotropic behavior of polar lipids from marine macrophytes 总被引:1,自引:0,他引:1
Major glyco- and phospholipids as well as betaine lipid 1,2-diacylglycero-O-4'-(N,N,N-tri-methyl)-homoserine (DGTS) were isolated from five species of marine macrophytes harvested in the Sea of Japan in summer and winter at seawater temperatures of 20-23 and 3 degrees C, respectively. GC and DSC analysis of lipids revealed a common increase of ratio between n-3 and n-6 polyunsaturated fatty acids (PUFAs) of polar lipids from summer to winter despite their chemotaxonomically different fatty acid (FA) composition. Especially, high level of different n-3 PUFAs was observed in galactolipids in winter. However, the rise in FA unsaturation did not result in the lowering of peak maximum temperature of phase transition of photosynthetic lipids (glycolipids and phosphatidylglycerol (PG)) in contrast to non-photosynthetic ones [phosphatidylcholine (PC) and phosphatidylethanolamine (PE)]. Different thermotropic behavior of these lipid groups was accompanied by higher content of n-6 PUFAs from the sum of n-6 and n-3 PUFAs in PC and PE compared with glycolipids and PG in both seasons. Seasonal changes of DSC transitions and FA composition of DGTS studied for the first time were similar to PC and PE. Thermograms of all polar lipids were characterized by complex profiles and located in a wide temperature range between -130 and 80 degrees C, while the most evident phase separation occurred in PGs in both seasons. Polarizing microscopy combined with DSC has shown that the liquid crystal - isotropic melt transitions of polar lipids from marine macrophytes began from 10 to 30 degrees C mostly, which can cause the thermal sensitivity of plants to superoptimal temperatures in their environment. 相似文献

The lipid compositions of Rothia dentocariosa was investigated. All of the strains tested possessed closely related lipid profiles consisting of predominantly straight-chain saturated and methyl branched long-chain fatty acids, unsaturated menaquinones with seven isoprene units and a polar lipid composition comprising diphosphatidylglycerol, phosphatidylglycerol and a diglycosyldiacylglycerol. The results of the present study indicate Rothia dentocariosa is a good and distinct taxon. The lipid data however does not support the classification of Rothia dentocariosa in the family Actinomycetaceae. 相似文献

Representative strains of Gardnerella vaginalis were degraded using both an alkaline and an acid methanolysis and the fatty acid methyl esters released examined by thin-layer and gas chromatography. The profiles obtained were both qualitatively and quantitatively similar and were comprised of straight chain saturated and unsaturated non-hydroxylated fatty acids with hexadecanoic acid (16:0) and octadecenoic acid (18:1) the major components. All of the strains contained very characteristic polar lipid patterns consisting of diphosphatidylglycerol, phosphatidylglycerol, phosphatidylinositol, five partially identified glycolipids and an uncharacterised phospholipid. Analyses of wall amino acid preparations using gas chromatography showed that Gardnerella vaginalis strains contain major amounts of alanine, glycine, glutamic acid and lysine. The chemical data support the integrity of the genus Gardnerella. 相似文献

The fatty acid composition of two motile (strains WH 8113 and WH 8103) and one nonmotile (strain WH 7803) marine cyanobacteria has been determined and compared with two freshwater unicellular Synechocystis species (strain PCC 6308 and PCC 6803). The fatty acid composition of lipid extracts of isolated membranes from Synechocystis PCC 6803 was found to be identical to that of whole cells. All the marine strains contained myristic acid (14:0) as the major fatty acid, with only traces of polyunsaturated fatty acids. This composition is similar to Synechocystis PCC 6308. The major lipid classes of the nonmotile marine strain were identified as digalactosyl diacylglycerol, monogalactosyl diacylglycerol, phosphatidylglycerol, and sulfoquinovosyl diacylglycerol, identical to those found in other cyanobacteria.Abbreviations DGDG

Detailed fatty acid analyses are reported for ten benthic algae from Australian waters of which the data for Cladophora fascicularis (Chlorophyta); Polysiphonia pungens, Porphyra sp., Centroceras clavatum (Rhodophyta); Hormosira banksii, Ralfsia sp., and Dictyota dichomota (Phaeophyta) are presented for the first time. The analyses report the exact molecular structure of component acids which is essential for taxonomic and food chain studies. The acid 16:4ω3 could be taxonomically distinguishing for species of the Chlorophyta. The occurrence of cis-vaccenic acid (18:1ω7) in the algae reported here suggests a distribution in marine benthic algae which is wider than hitherto realised, with particular taxonomic importance for Chlorophyta species in which it occurs in high levels. Corallina officinalis was found to contain the non-methylene interrupted acids 20:2 and 22:2. 相似文献

Density, biomass and primary productivity of eelgrass (Zostera marina L.) and density and biomass of Zostera noltii Hornem. were studied at three locations in Sevastopol Bay, Crimean Peninsula, Ukraine, in April 2002, late September 2002, and late January 2003 at 3 m and 5 m depths. All three growth parameters varied by station, depth, and month. The aboveground biomass (AGB) was greater in September than in April or January at 3 m. It was greater at 5 m than at 3 m in April at one station. For Z. noltii, results were more variable as to month. For belowground biomass (BGB), month and station relationships were not clear cut. It was generally true that total biomass was greatest in September. Significant differences in density, AGB and total biomass were observed for station and month in eelgrass, while in BGB, these differences were significant for depth and for month. Primary production in eelgrass was greatest at both depths in April as compared to September or January. Reproductive shoots were found only during April. Overall, seedlings were found in all collection periods and depths, but were most abundant at the three stations in April. 相似文献

We examined variations in the lipid composition of the marine red alga Tichocarpus crinitus exposed to different levels of photon irradiance: 70-80% and 8-10% of the incident photosynthetically active radiation (PAR). The content of storage and structural lipids was significantly affected by the light intensity. Exposure of T. crinitus to low light conditions induced an increase in the abundance of structural components of the cell membranes, especially sulfoquinovosyldiacylglycerol, phosphatidylglycerol (PG) and phosphatidylcholine, while growth of algae at high light intensity resulted in a 1.5-folder increase in the level of storage lipids, i.e. triacylglycerols. There were no significant differences in the fatty acid composition of the total lipid pool in algae grown under different light conditions. However, the content of the most unsaturated acid, 20:5n-3, was slightly higher in T. crinitus under 8-10% PAR compared to those at 70-80% PAR. Each lipid class was found to have a characteristic fatty acid composition. The relative proportions of fatty acids esterifying monogalactosyldiacylglycerol (MGDG) and PG were significantly affected by irradiance conditions. Exposure of algae to low light resulted in increase in the content of 20:5n-3 in MGDG and in decrease in the level of this acid in PG. The concentration of trans-16:1 acid in PG increased in algae grown under high light intensity. Light conditions influenced on total lipid content, which made up 4.2+/-0.5 and 3.4+/-0.3 mg g-1 fresh weight in algae exposed to 8-10% PAR and 70-80% PAR, respectively. We suggest that variations in the lipid composition of T. crinitus exposed to different levels of light intensity may be a response of alga to light conditions and it can be considered as one of the mechanisms of adaptation of T. crinitus to varying incident light intensity. 相似文献

The free fatty acid and phospholipid composition of 5 psychrotrophic marine Pseudomonas spp. have been determined in chemostat culture with glucose as the limiting substrate over the range 0–20°C. The predominant fatty acid present in all the isolates was hexadecenoic acid (C16:1) together with lesser quantities of octadecenoic acid (C 18:1) whilst none contained acids with chain lengths exceeding 18 carbon atoms. Decreasing the growth temperature from 20°C to 0°C resulted in little significant change in fatty acid composition. The principal phospholipid components of the five psychrotrophic pseudomonads have been identified as phosphatidylserine, phosphatidylglycerol, phosphatidylethanolamine and diphosphatidylglycerol. Decreasing the growth temperature did not elicit significant changes either in the total quantities of phospholipid synthesized or in the concentration of individual phospholipid components in any of the isolates. All the psychrotrophs showed maximum glucose uptake between 15°C and 20°C and the rate decreased rapidly as the temperature was decreased towards 0°C.Abbreviations PS

This paper investigates the correlation between mycelial age and fatty acid biosynthesis. The correlation was investigated by analyzing the lipid composition lengthwise the mycelium of the oleaginous fungus Mortierella isabellina, a potential producer of γ-linolenic acid (GLA). Young mycelia were rich in polar lipids (glycolipids plus sphingolipids and phospholipids), while neutral lipid content increased in aged mycelia. In young mycelia, each polar lipid fraction contained almost 40% (w/w) polyunsaturated fatty acids (PUFAs), but this content decreased to less than 30% (w/w) in aged mycelia. On the other hand, PUFA content in neutral lipids fluctuated slightly with age. These results indicate that PUFA biosynthesis is favored in young, fast growing mycelia, while it decreases significantly in aged mycelia. This trend was also observed when we grew M. isabellina on pear pomace, an agro-industrial waste. Pear pomace cultures yielded significant amounts of lipid, which reached 12% (w/w) in dry fermented mass. The produced lipid was rich in GLA and the maximum GLA content in dry fermented mass was 2.9 mg/g. 相似文献

A study to assess which environmental or developmental factors predominate in the biosynthesis of lipids of Laminaria japonica Aresch. blades was undertaken by means of 14C-labelling technique. In experiment 1, kelp blades at different growth stages were collected in different cultural seasons. In experiment 2, kelp blades of different sizes and maturity cultured simultaneously for two months in the same sea area were collected at the same time.The following results were obtained. In experiment 1, the 14C-incorporation into whole lipids was lowest in juvenile blades collected at the end of autumn and highest in blades of middle size collected in winter. However, the highest counts were incorporated in PC among complex lipid classes from all size classes of blades in both experiments 1 and 2. In experiment 2, 14C-incorporation patterns of individual lipid classes were characteristically different depending on the sizes of blades even under the same cultural condition. Thus, the biosynthesis of lipids in this kelp seems to be affected essentially by developmental factors.Abbreviation Comp. lip.

Dry wt, total oil and fatty acid composition of the oil was determined during kernel development of three maize inbreds. There was significant variability among these inbreds for duration and rate of dry wt, oil and fatty acid accumulation. Relative quantities of the component fatty acids changed as the kernels developed. Palmitic and linolenic decreased while oleic and stearic increased with maturity. Inbred C103, the higher oil inbred, appeared to accumulate oil over a longer period of time while inbred Hy2, the lower oil inbred, accumulated oil over a shorter period of time. However, the latter had higher daily rates of synthesis for some of the unsaturated fatty acids. 相似文献

Adductor muscle fatty acid (FA) composition was compared in Pecten maximus juveniles originating from five locations along the Norwegian coast from 59°N to 65°N to detect possible population differences. Broodstock sized scallops were translocated to sea conditions by a scallop hatchery near Bergen (60°N) before spawning in February 2006. The scallop larvae and juveniles were reared in the same environment for two years and 10 individuals from each population were sampled in May 2007 and in May 2008 for analysis of the FAs in the adductor muscle. The total lipid content determined as total amount of FAs were 5.7 ± 0.3 mg per g tissue, and no significant difference was found among the five populations. The polyunsaturated FAs made up close to 60% of the total, with 20:5n3 and 22:6n3 dominating. The saturated FA content was approximately 30%, while the monounsaturated FA were less abundant (7–10%). The FA composition of the muscles of the five populations was similar within each year, with larger differences between the years. Multivariate, supervised learning method PLS, applied pairwise, showed distinct FA composition, between the scallops from the five locations, indicating population differences. The relatedness between the populations was different in the two years, but the distinct FA profiles of the adductor muscle could be used to distinguish between scallop populations on a local scale. The results indicate habitat-specific lipid metabolism which may have important implications for the scallop aquaculture industry in the context of producing well adapted individuals for the specific locations. 相似文献

A study is reported on the incorporation of14C-acetate into lipid classes from three different growth stages ofLaminaria japonica, a species long used for food in Japan. This was done because of the possible utilization of its lipids.Radioactivity incorporated into whole lipids in the three growth stages under the same experimental conditions (10 °C, 500 lux) increased with maturity of the thalli. The radioactivity was found mainly in PC, TG and 1,2-DG and subsequently distributed into other lipid classes (PG,PI,PE,MGDG,SQDG and DGDG) to a lesser extent. The incorporation patterns of the former group were similar at all stages, but those of the latter group differed slightly according to growth stage.In juvenile thalli,14C was incorporated to a much higher extent into PG, MGDG, PI and fucosterol than PE, SQDG, DGDG and MG, while the14C-incorporation into MG, SQDG, DGDG and PS in the mature growth stage was higher than into the other lipid classes. The absolute level of incorporation was higher for all these compounds in mature thalli than the thalli of other growth stages. 相似文献

The photobiont of the lichen, Dictyonema glabratum (Scytonema sp.), was isolated and cultivated in a soil-extract medium and submitted to chemical analysis. Successive extractions with CHCl3-MeOH, aqueous MeOH, and H2O gave rise to solutions of lipids (25%), low-molecular-weight carbohydrates (22%), and polysaccharides (4%), respectively. TLC of the lipid extract showed the presence of glycolipids, which were further purified and examined by NMR spectroscopy and GC-MS. Monogalactosyldiacylglycerol (1%), digalactosyldiacylglycerol (0.8%), trigalactosyldiacylglycerol (0.4%), and sulfoquinovosyldiacylglycerol (0.5%) were identified. The most abundant fatty acid ester in each fraction was palmitic (C16:0), but a great variation of the ester composition from one to another was found. Others present were those of C12:0, C14:0, C15:0, C16:1, C17:0, C18:0, C18:1, C18:2, C18:3, C22:0, C22:2, and C24:0. The lipid extract was also subjected to acid methanolysis, which gave rise to dodecane, 2-Me-heptadecane, 2,6-Me2-octadecane, and 8-Me-octadecane, methyl esters of C14:0, C15:0, C16:0, C16:1, C17:0, C18:0, C18:1, C18:2, C20:0, and C24:0 fatty acids, and the dimethyl ester of decanedioic acid. The polysaccharide had mainly Glc, Gal, and Man, with small amounts of 3-O-methylrhamnose and 2-O-methylxylose, both found in plants, and unexpectedly, some of the units were beta-galactofuranose, typical of fungal, but not cyanobacterial polysaccharides. The low-molecular-weight carbohydrates showed mannose as the main free reducing sugar, which differs from Nostoc sp. and Trebouxia sp. photobionts. 相似文献

Abstract Streptomyces viridochromogenes was grown both as submerged and surface culture. Mycelia from these cultures were analysed for the composition of lipids and fatty acids. An increase in ornithinolipid content according to incubation time was observed. The addition of phosphate inhibited the ornithinolipid synthesis. A mutant strain with bald phenotype did not exhibit the phosphate inhibition. At the same time, the mutant strain had a higher content of 12-methyltetradecanoic acid. 相似文献

Fatty acid composition and degree of fatty acid saturation during temperature stress in thermo-intolerant (Phaeodactylum tricornutum) and thermo-tolerant (Chaetoceros muelleri) marine diatoms were investigated. A greater number of fatty acids were observed in C. muelleri than in P. tricornutum regardless of treatment. The major fatty acids detected were 14:0, 16:0, 16:1, 16:2, 16:3, 18:0, 18:1(n-9)c, 18:2(n-6) and 20:5(n-3) with additional fatty acids 18:1(n-9)t and 20:4(n-6) detected in C. muelleri. Short duration (2 h) temperature increase above optimal growth temperature had a greater effect on fatty acid composition in C. muelleri than in P. tricornutum and the degree of fatty acid saturation was affected more by temperature in C. muelleri than in P. tricornutum during both short and long duration (24 h) treatments. Total protein assay results suggest that P. tricornutum, but not C. muelleri, was undergoing stress under our growing conditions although lipids in both diatoms were affected by increased temperature. Immunodetection of proteins with anti-rubisco indicates that the rubisco large subunit was undergoing greater turnover in C. muelleri than in P. tricornutum. However, the integrity of rubisco as a suitable indicator of lipid status needs further study. This work supports the hypothesis that a particular temperature, and not treatment duration, has the greater effect on changes in fatty acid composition. Furthermore, changes in fatty acid composition and degree of fatty acid saturation occurred more quickly in the diatoms in response to increased temperature than previously observed in nutrient starvation studies. Since diatom lipids represent an important resource for growth and reproduction of marine animals, the rapid alteration of their lipid composition under temperatures normally encountered in marine environments warrants further study. 相似文献

We gathered sequence information from the nuclear 5.8S rDNA gene and associated internal transcribed spacers, ITS-1 and ITS-2 (5.8S rDNA/ITS), and the chloroplast maturase K (matK) gene, from Zostera samples collected from subtidal habitats in Monterey and Santa Barbara (Isla Vista) bays, California, to test the hypothesis that these plants are conspecific with Z. asiatica Miki of Asia. Sequences from approximately 520 base pairs of the nuclear 5.8S rDNA/ITS obtained from the subtidal Monterey and Isla Vista Zostera samples were identical to homologous sequences obtained from Z. marina collected from intertidal habitats in Japan, Alaska, Oregon and California. Similarly, sequences from the matK gene from the subtidal Zostera samples were identical to matK sequences obtained from Z. marina collected from intertidal habitats in Japan, Alaska, Oregon and California, but differed from Z. asiatica sequences accessioned into GenBank. This suggests the subtidal plants are conspecific with Z. marina, not Z. asiatica. However, we found that herbarium samples accessioned into the Kyoto University Herbarium, determined to be Z. asiatica, yielded 5.8S rDNA/ITS sequences consistent with either Z. japonica, in two cases, or Z. marina, in one case. Similar results were observed for the chloroplast matK gene; we found haplotypes that were inconsistent with published matK sequences from Z. asiatica collected from Japan. These results underscore the need for closer examination of the relationship between Z. marina along the Pacific Coast of North America, and Z. asiatica of Asia, for the retention and verification of specimens examined in scientific studies, and for assessment of the usefulness of morphological characters in the determination of taxonomic relationships within Zosteraceae. 相似文献

Seagrasses are considered important indicators of decline in water quality resulting in increased light attenuation that negatively influences their growth and survival. Chronic light-limitation interspersed with unpredictable acute attenuation events have had poorly understood effects on seagrass recovery dynamics. Zostera marina (eelgrass) and Halodule wrightii (shoalgrass) were subject to a matrix of light-deprivation events followed by recovery periods to mimic repeated acute shading events. Plant survival, morphology, biomass, chlorophyll content, and Fv/Fm were assessed over time to determine recovery. At the end of the experiment, all plants were harvested and species-specific treatment effects were determined. Significant differences due to treatments were noted in all parameters measured. In general, responses were similar for both life-stages and between species, suggesting similar physiological tolerance to repeated acute light-attenuation events. Only plants in treatments where light-deprivation was followed by a recovery interval of at least the same duration showed signs of long-term survival. Chlorophyll fluorescence (Fv/Fm) was an important metric for assessing recovery, but it failed to detect the onset of mortality in many plants. Other metrics of plant condition need to be assessed and coupled with chlorophyll fluorescence data to assess seagrass “health”. This is of particular importance in field studies, where the history of the plants is largely unknown. 相似文献

The cellular fatty acid composition of 14 strains of Caulobacter species and types, two species of Prosthecomicrobium, and two species of Asticcacaulis was determined by gas-liquid chromatography. In most of these bacteria, the major fatty acids were octadecenoic acid (C18:1), hexadecenoic acid (C16:1) and hexadecanoic acid (C16:0). Some cyclopropane and branched chain fatty acids were detected in addition to the straight chained acids. The fate of ingested eukaryotic photoautotrophic fatty acids during gut transit in the lugworm Arenicola marina (L.) and the influence of A. marina's faeces on the evolution of fatty acid distribution and bacterial community structure in superficial sediments were studied under laboratory conditions. Dead phytoplanktonic cells (food portions) were fed to individual A. marina and subsequently incubated, or allowed to directly incubate in the presence of fresh egesta or non-ingested sediment. Changes in fatty acid composition and genetic structure of bacterial communities during gut transit and/or incubation were monitored using gas-chromatography/mass-spectrometry and a DNA fingerprint approach (RISA), respectively. Results, supported by principal component analyses, suggest that A. marina's feeding activity can directly and indirectly affect the lipid biomarker composition and the bacterial community structure of inhabited sediments. Faecal casts produced from food portions appeared qualitatively enriched in saturated fatty acids relative to (poly)unsaturated ones due, partly, to an increase of some bacterial fatty acids and to the preferential removal of some polyunsaturated fatty acids (PUFAs). The incubation of food portions in the presence of fresh A. marina's egesta (designed to study the indirect impact of feeding by A. marina) induced a significant increase in the concentrations of C20 and C22 polyunsaturated fatty acids (PUFAs), whereas these compounds almost disappeared following direct feeding and subsequent incubation, indicating that some dietary fatty acids may be more accessible to biodegradation following passage through the gut of A. marina. The aforementioned increase in PUFAs was attributed to a bacterial production during incubation, suggesting the presence of PUFA-producing bacteria in the fresh egesta of A. marina. Those bacteria were either enteric bacteria that were released with the egesta or ingested bacteria that have survived gut passage, as suggested by the variations of the bacterial community structure (i.e. RISA profiles) during incubation. The results suggest that aged faeces from A. marina might be, in some circumstances, of relatively high nutritional value to trophic levels which are unable to synthesize essential PUFAs de novo. The presence of PUFA-producing bacteria in guts of marine lugworms deserves further attention. 相似文献
