Over the past decade, molecular, biochemical and cellular markers have been extensively used in pollution monitoring of aquatic environments. Biochemical markers have been selected among early molecular events occurring in the toxicological mechanisms of main contaminants. This paper assesses the marine environment quality along the Tunisian coasts using a statistical approach. Clams (Ruditapes decussatus) were collected during the four seasons of 2003 on seven different sites from the Tunisian coasts. Oxidative stress was evaluated in gills using catalase activity (Cat), neutral lipids and malonedialdehyde accumulation. Glutathione S-transferase activity is related to the conjugation of organic compounds and was evaluated in both, gills and digestive glands. Acetylcholinesterase activity was evaluated as the biomarker of exposure to organophosphorous, carbamate pesticides and heavy metals. For each biomarker, a discriminatory factor was calculated and a response index allocated. For each site, a global response index was calculated as the sum of the response index of each biomarker. Discriminant analysis shows significant differences between sites and seasons compared with control sample. Faroua (site 1) and Menzel Jemile (site 2) seem to be the less polluted with respect to the other sites for all seasons. Gargour (site 6) shows the highest Multimarker Pollution Index during the four seasons, indicating higher contamination level.  相似文献

As a result of our calculation method, the quality of the coastal environment at each site can be classified with a five level scale. Mussels were collected during five cruises in 1995-1996 on the Baltic and Mediterranean coasts. The results show that water quality ranged from class 1 (clean areas in some sites of the French Riviera, Spanish Costa Brava and the Baltic coast) to class 5 (high pollution in main harbours, e.g. Kiel and Toulon). Some areas fall into class 4, e.g. Carteau, Cortiou, Barcelona, Warnemunde, Swinemunde, Ebro delta. The global Biomarker Index was positively correlated with the level of PAHs in mussels in Baltic transects. A number of other contaminants or stressors may be present in the marine environment and the Biomarker Index appeared to be relevant to classify coastal environmental pollution.  相似文献

As the result of our calculation method, the quality of the coastal environment at each site can be classified according to a five levels scale. Samples collected for five cruises in May 2001, 2002, 2003, and September 2001 and 2002 showed MPI evolutions. The results show that water quality can be classified from class 1 (clean areas in some sites of France, Italy and Spain) to class 4 (high pollution in main harbours). Results of the use of the biomarker scale in WP3 (Work Package Concernant Biomonitoring Programmes in Mediterranean Sea) during the BEEP programme make a strong contribution to the establishment of standardized strategies and methods for internationally agreed protocols for biomarker-based monitoring programmes. In comparison with scale pollution methodology used in the BIOMAR programme, the main contribution of BEEP was (1) to select from discriminatory analysis the biomarkers to be included in calculation of scale pollution; (2) to improve the use of the biomarker index in order to identify the main contaminants by analysis of individual contributions to the MPI; and (3) to apply methodology for temporal trends at sampled sites.  相似文献

Phoebis sennae (Pieridae), Agraulis vanillae (Heliconidae) and Urbanus proteus (Hesperiidae) migrate into peninsular Florida each fall from summer breeding areas throughout the south-eastern United States. Fall flight directions of the three species were studied at 78 sites for clues to their means of orientation and migratory routes. Mean flight directions for a visit to a site were calculated using conventional circular statistics. The distribution of flight directions permitted using linear procedures to combine visit means. The overall mean direction (OMD) of a species at a site was defined as the mean of visit means, weighted by frequency/angular variance, with a lower limit of 0.2 imposed on angular variance. In intensive studies at two sites at Gainesville, Florida, mean flight directions during fall migration were largely independent of time of day and crosswinds. The OMDs of the three species at the two Gainesville sites were similar (142 to 156°) and not significantly different among species at a site or, for the same group of dates, between sites for a species. However, day-to-day variation among species and between sites was not concordant. Visits to 65 sites on a grid throughout the southeastern United States and to 11 sites along transects inland from the Gulf and Atlantic coasts yielded 10 cases of a P. sennae OMD at a grid or transect site differing significantly from its OMD at the main Gainesville site. In each of these, the OMD was more easterly at sites inland from the Gulf coast or more southerly or south-westerly at sites inland from the Atlantic coast. By unknown means and with large, unexplained day-to-day variations in directions, P. sennae converge on peninsular Florida from widespread summer breeding grounds. They do this without closely following the Atlantic or Gulf coasts.  相似文献

The aim was to apply a multimarker scoring approach as complementary to freshwater monitoring programmes carried out by the Water Agency Adour–Garonne. Fish (chub, barbel and trout) were collected in 11 sites in rivers in south-west France. Five biomarkers of response were measured either in muscle or brain for acetylcholinesterase (AChE) and in liver for glutathione S-transferase, catalase and 7-ethoxyresorufine O-deethylase. As a result of multivariate analysis, sites were clearly discriminated mainly by 7-ethoxyresorufine O-deethylase and acetylcholinesterase activities. According to the scoring approach, a multimarker pollution index was calculated for each sampling site as the sum of the response index of the five measured biomarkers (pollution index). Sorting was established by ranging the sites from lightly to highly contaminated locations.  相似文献

Responses of biochemical biomarkers were evaluated in gills of immature adult mangrove oyster Crassostrea rhizophorae collected in three estuarine regions along the Brazilian coast. In each region, ten oysters were collected in one reference site (R) located far from pollution sources, and in two polluted sites (P-I and P-II sites) located in another water body with similar characteristics. P-I site is located close to recognized pollution sources while P-II site is in the same water body, but far from pollution sources. At the Paranaguá Bay (Southern Brazil), polluted sites receive domestic, harbor and phosphate fertilizer plant discharges. High lipid peroxides (LPO) content was observed in winter oysters from the P-I site. In summer, higher catalase activity was observed in these oysters. In the Piraquê region (Southeastern Brazil), polluted sites receive domestic and agricultural effluents. Lower total oxyradical scavenging capacity (TOSC) towards peroxyradicals was observed in summer oysters from both P-I and P-II sites. In the Itamaracá region (Northeastern Brazil), polluted sites receive paper mill and caustic soda and chlorine factories effluents. Increased glutathione S-transferase (GST) activity was observed in oysters from the P-I site in both summer and winter. At Paranaguá Bay (higher latitude), no seasonal differences were observed in oysters from the R site, suggesting that temperature was not an important factor influencing biomarkers levels. Lower GST activity was observed in oysters from the R site of the Itamaracá Bay (lower latitude) in winter and summer. Taken together, data obtained point to responses of biomarkers in oysters from polluted sites of the three estuarine regions analyzed, indicating the need for future monitoring of the biological effects of contaminants in these environments. They also point to the relevance to consider both season and latitude as factors influencing biomarker responses in environmental contamination monitoring programs.  相似文献

Seasonal variations of pollution biomarkers have been studied in two populations of Corbicula fluminea (Müller) for 2 years. Clams were collected from the Sanguinet lake and the Dronne river (Southwest France), sites characterised by water of excellent and passable quality, respectively. Biological parameters studied were enzymatic activity (catalase, propionylcholinesterase, glutathione S-transferase, NADPH-cytochrome c reductase and NADH-cytochrome c reductase activities) and indicators of physiological (protein, carbohydrate, lipid and ash contents, fresh and dry tissue weights, condition factor) and reproductive (amount of oocytes present in gonads and clams incubating larvae in gills) status. Environmental parameters (e.g. water temperature, chlorophyll a concentration) were also monitored. Effects of site, sampling date and season (from spring to winter) were studied, as well as relationships between abiotic and biotic factors. Finally, an index of variability was calculated for each biomarker. Catalase, propionylcholinesterase, NADH-cytochrome c reductase and lipids experienced the most pronounced seasonal fluctuations, whereas glutathione S-transferase, NADPH-cytochrome c reductase and proteins suffered less pronounced ones. For some parameters, results differed depending on the site. Most seasonal changes were related to the reproductive cycle and/or water temperature. Implications of such variations in the use of pollution biomarkers in environmental surveys are discussed.  相似文献

This study characterises the genetic variability of fig, Ficus carica L., using simple sequence repeat (SSR) and amplified fragment length polymorphism (AFLP) markers. It compares the efficiency and utility of the two techniques in detecting variation and establishing genetic relationships among Tunisian fig cultivars. Our results show that using both marker systems, the Tunisian fig germ plasm is characterised by having a large genetic diversity at the deoxyribonucleic acid level, as most of AFLP bands were detected and all SSR markers were polymorphic. In fact, 351 (342 polymorphic) and 57 (57 polymorphic) bands were detected using AFLP and SSR primers, respectively. SSR markers were the most polymorphic with an average polymorphic information content value of 0.94, while AFLP markers showed the highest effective multiplex ratio (56.9) and marker index (45.2). The effective marker index was recorded highest (4.19) for AFLP markers and lowest (0.70) for the SSR ones. Our results demonstrate that (1) independent as well as combined analyses of cluster analyses of SSR and AFLP fragments showed that cultivars are clustered independently from their geographical origin, horticultural classifications and tree sex; (2) the analysis of molecular variance allowed the partitioning of genetic variation within and among fig groups and showed greater variation within groups and (3) AFLP and SSR markers datasets showed positive correlation. This study suggests the SSR and AFLP markers are suitable for diversity analysis and cultivars fingerprinting. An understanding of the genetic diversity and population structure of F. carica in Tunisia can also provide insight into the conservation and management of this species.  相似文献

The genetic characterization of natural populations of common pandora (Pagellus erythrinus, Sparidae) was studied using allozyme markers. Fish samples (n = 360) were collected from five locations along the Tunisian coasts and one from the Libyan coast. From eight enzymatic systems, 12 putative enzyme-coding loci were resolved, 8 of which were polymorphic at a level of 99%. F statistics indices revealed a significant departure from panmixia with heterozygote deficiencies and a slight genetic differentiation between samples. Neighbor-joining dendrogram constructed by Reynolds genetic distance showed that the populations sampled from the southeastern Mediterranean Sea were more differentiated from the others.  相似文献

Lead and cadmium concentrations in seawater and algae of the Tunisian coast   总被引:1,自引:0,他引:1  
Both lead and cadmium are toxic trace metals, even in very weak concentrations. The aim of this study was to estimate lead and cadmium pollution in various sites of the Tunisian coast and to verify the possibility of modification of the algae bioconcentration power according to water physico-chemical conditions. Our study concerned 99 samples of algae and 99 samples of seawater, taken in different sites of the Tunisian littoral. The analysis was realized by atomic absorption spectrophotometry (oven graphite). In algae, Sfax site presented the highest concentrations of lead when Sousse site showed the lowest ones. In seawater, the most amounts of lead were observed in Bizerte, Mahdia and Sfax sites, and those of cadmium in Bizerte and Medenine coasts. Bizerte's coast seems to be the most exposed zone to pollution. Indeed, the intensification of sea traffic may take place on this pollution because hydrocarbons derived from petroleum contain some tetraethylic lead characterised by its great toxicity. Sousse's region is the least polluted zone; it might be due to the development of tourism and a strict regulation of pollution in this district.  相似文献

Accumulation and effects of polychlorinated biphenyls (PCBs) in avian species were evaluated at a Superfund site located at Crab Orchard National Wildlife Refuge, Illinois, and seven criteria were used to assess whether there was a causal relationship between PCB exposure and observed reproductive effects. European starlings (Sturnus vulgaris) were monitored at nest boxes constructed at each of two exposed and two reference sites. During the breeding season, starling productivity (number of chicks produced per nest) and adult nest attentiveness behavior (provisioning behavior) were monitored. At 15 days post-hatch, chicks were collected for contaminant and biomarker analyses. Chicks were necropsied, ethoxyresorufin-O-deethylase (EROD) activity measured in liver tissue, and PCB (Aroclor 1254) and 34 chlorinated biphenyl (CB) congener concentrations measured in carcasses using gas chromatography. PCB and CB concentrations also were measured in eggs that failed to hatch. Mean Aroclor 1254 and quantified CB concentrations were greater (P<0.001) in eggs that failed to hatch and 15-day-old chicks collected from PCB sites compared to those collected from reference sites. EROD activity was greater (P=0.005) in 15-day-old chicks collected from PCB sites and was correlated with carcass PCB concentrations. Reduced adult nest attentiveness behavior and decreased chick survival were observed at PCB sites. Six of the seven causal criteria evaluated provided evidence that observed reproductive effects resulted from exposure to PCBs. Using this weight-of-evidence approach provided a means for establishing the likely cause of effects and thus provided managers with information needed in decision-making processes.  相似文献

The use of oyster gills for the analytical determination of metallothionein (MT) concentration as a biomarker of metal exposure was investigated. Temporal variations in MT and metal concentrations (which can interfere with inter-site differences) were examined over a 7 month period (from spring to autumn) in Japanese oysters from a clean site (Bay of Bourgneuf, France) and a metal-rich site (Gironde estuary, France) as well as in individuals translocated from the clean to the contaminated area. The ratio between the annual average of MT concentrations in specimens from the clean and the metal-rich sites was 1.3. During the last 3 months of the experiment, significant differences were no longer registered between transplants and residents from the Gironde estuary. Metals concentrations in oyster gills differed consistently between the clean and the metal-rich sites (annual average ratios of 1.5, 2.7 and 9.8, respectively, for zinc, copper and cadmium) and a fast increase in metal concentrations (over a few months) was observed in transplants, mainly for cadmium. MT and soluble metal concentrations were found to be positively and significantly correlated over the period of the study. This relationship is a positive argument for a possible use of gill MT concentration as a biomarker of metal pollution in contrast to previous findings on the digestive gland, there being a smaller amount of seasonal variability in the weight of oyster gills.  相似文献

Fish contamination has been extensively investigated along the Saudi coasts, but studies pertaining to bacterial pathogens are scarce. We conducted qualitative assessment and molecular identification of culture-dependent bacteria in 13 fish species from three coastal sites and a local fish market in Jeddah, Saudi Arabia. Bacterial counts of gills, skin, gut and muscle were examined on agar plates of Macconkey’s (Mac), Eosin Methylene Blue (EMB) and Thiosulfate Citrate Bile Salts (TCBS) culture media. Bacterial counts significantly differed between species, sources and feeding habits of examined fishes. Mugil cephalus exhibited higher counts on TCBS (all body parts), Mac (gills, muscle and gut) and EMB (gills and muscle). Fishes from Area I had higher bacterial loads, coinciding with those in seawater and sediment from the same site, indicating direct association between habitat conditions and the levels of bacterial contamination. By feeding habit, detritivorous fish harbored higher counts than herbivorous and carnivorous species. Bacterial counts of skin were higher in fish from market than field sites, and positively correlated with other body parts indicating the relation of surface bacterial load on the overall quality of fish. Rahnella aquatilis (Enterobacteriaceae) and Photobacterium damselae (Vibrionaceae) were among the dominant species from fish muscle based on 16S rRNA sequencing. These species are known human pathogens capable of causing foodborne illness with severe antibiotic resistance. Opportunistic pathogens, e.g. Hafnia sp. (Enterobacteriaceae) and Pseudomonas stutzeri (Pseudomonadaceae) also occurred in fish muscle. The inclusion of bacterial contamination in future monitoring efforts is thus crucial.  相似文献

Aspergillus flavus is second only to A. fumigatus in causing invasive aspergillosis and it is the major agent responsible for fungal sinusitis, keratitis and endophthalmitis in many countries in the Middle East, Africa and Southeast Asia. Despite the growing challenge due to A. flavus, data on the molecular epidemiology of this fungus remain scarce. The objective of the present study was to develop a new typing method based on the detection of VNTR (Variable number tandem repeat) markers. Eight VNTR markers located on 6 different chromosomes (1, 2, 3, 5, 7 and 8) of A. flavus were selected, combined by pairs for multiplex amplifications and tested on 30 unrelated isolates and six reference strains. The Simpson index for individual markers ranged from 0.398 to 0.818. A combined loci index calculated with all the markers yielded an index of 0.998. The MLVA (Multiple Locus VNTR Analysis) technique proved to be specific and reproducible. In a second time, a total of 55 isolates from Chinese avian farms and from a Tunisian hospital have been evaluated. One major cluster of genotypes could be defined by using the graphing algorithm termed Minimum Spanning Tree. This cluster comprised most of the isolates collected in an avian farm in southern China. The MLVA technique should be considered as an excellent and cost-effective typing method that could be used in many laboratories without the need for sophisticated equipment.  相似文献
