SA浸种对盐胁迫下小麦种子萌发和幼苗生长的影响   总被引:5,自引:0,他引:5

以小麦盐敏感品种鲁麦15为材料,研究了外源水杨酸(salicylic acid,SA)浸种对100 mmol/L NaCl胁迫下小麦种子萌发和幼苗生长的影响。研究结果表明:盐胁迫下,无论经SA浸种还是未经SA浸种,小麦幼苗的生长均受到明显抑制,干、鲜重显著下降;0.1 mmol/L、0.2 mmol/L和0.3 mmol/L SA溶液浸种均能显著缓解NaCl胁迫对小麦幼苗生长的抑制,其中以0.2 mmol/L SA溶液浸种预处理效果最好。实验中,0.2 mmol/L SA浸种可显著提高盐胁迫下小麦种子β-淀粉酶的活性和吸胀速率。此外,与未经SA浸种的盐胁迫小麦幼苗相比,0.2 mmol/L SA浸种的盐胁迫小麦幼苗整株的干、鲜重显著增加,幼苗体内Na~+含量降低,K~+含量和K~+/Na~+比值显著提高;同时,小麦幼苗叶片中超氧化物歧化酶(superoxide dismutase,SOD)、过氧化氢酶(catalase,CAT)、过氧化物酶(peroxidase,POD)活性升高,而丙二醛(MDA)含量则显著降低。由此可以得出,SA浸种能有效提高盐胁迫下小麦幼苗体内K~+/Na~+比值,提高SOD、CAT和POD的活性,减轻膜脂过氧化程度,以缓解盐胁迫对小麦幼苗生长的抑制作用,从而提高耐盐性。  相似文献

以小白菜(Brassica campestris ssp.chinensis L.)为材料,通过采用不同浓度的外源赤霉素(GA3)浸种处理,研究了GA3对混合盐碱胁迫下小白菜种子的发芽率(Gr)、发芽势(Gv)、发芽指数(Gi)以及根毛区宽度和胚长的影响。结果表明:混合盐碱胁迫下,小白菜各发芽指标均显著低于对照,GA,处理后,各指标均显著提高,且随外源GA3浓度的增加,各指标均表现出先增后减的趋势,当GA3浓度达80mg/L时,各发芽指标达到峰值,说明80mg/LGA3可使混合盐胁迫下小白菜种子萌发的缓解作用达到最佳。同时外源GA3处理还可不同程度地缓解盐胁迫对小白菜种子a-淀粉酶活性的抑制。  相似文献

盐胁迫下,DK961(耐盐)和LM15(盐敏感)小麦种子的发芽率(Gr)、发芽指数(Gi)和活力指数(Vi)均显著下降,LM15下降的幅度大于DK961.外源100 mg/L GA3、1×10-7 mol/L ABA和0.1% Ca(NO3)2处理均能缓解盐对小麦种子萌发的抑制作用,对盐胁迫下LM15种子萌发的缓解作用显著好于DK961,并且不同程度地缓解盐处理引起的种子内源GA 1+3含量和α淀粉酶的活性下降,从而降低盐胁迫对种子萌发的抑制作用.表明盐抑制小麦种子萌发的主要原因是盐胁迫导致种子内源GA 1+3含量和α淀粉酶的活性下降.  相似文献

采用多因素交叉试验设计,探讨了不同盐分类型、盐分浓度以及温度对盐节木(Halocnemum strobilaceum)种子萌发的影响。结果表明:15~25℃范围是盐节木种子萌发的最适温度范围,5~15℃和25~35℃的变温不利于种子萌发;最适温度下,单盐和土盐浓度分别小于0.96%和0.6%(电导率小于9.375dS/m)时可促进种子萌发,当浓度分别高于0.96%和0.6%(电导率大于9.375dS/m)时,单盐处理下的种子萌发率随浓度升高而降低,土盐处理下的种子萌发率随浓度升高表现为先升高后下降,但2种盐处理下的平均种子萌发率差异不显著(P>0.05),非最适温度下,2种盐处理下的平均种子萌发率差异显著(P<0.05)。由此得出:盐分浓度和温度是影响盐节木种子萌发的关键因素,土盐对种子萌发的抑制作用较单盐的小。  相似文献

用不同浓度的NaCl、Na2CO3混合盐胁迫梯度,对荆条、白蜡和沙枣种子进行了种子萌发实验,观察了3个物种在盐胁迫下的发芽率、发芽速度和胚根生长,比较了光照和黑暗条件下3种植物种子的萌发差异。结果表明:光照对3种植物种子的萌发有明显的促进作用,但对胚根生长的影响却各不相同。低浓度混合盐溶液对荆条、白蜡种子萌发有促进作用,对沙枣有轻度抑制。当盐胁迫浓度超过0.8%,3种植物种子的萌发均受到不良影响,尤其对荆条的影响较大。3种植物均能在盐渍化生境中生长,3者均可作为园林植物在天津试验栽培。沙枣在天津塘沽地区盐碱地上进行广泛种植完全具有可行性。  相似文献

以野生乌拉尔甘草种子为材料,研究不同温度、覆土厚度、干旱胁迫、pH值、盐胁迫、干旱和盐双重胁迫下种子发芽与萘乙酸、3-吲哚乙酸、褪黑素浸种对种子发芽特性的影响。结果表明,(1)乌拉尔甘草种子最适发芽温度为20~25℃,发芽率≥90%,发芽时滞显著低于其他温度;(2)乌拉尔甘草种子对pH适应范围较宽,pH值在4~8发芽率较高;(3)覆土厚度在0~0.5 cm时发芽率显著高于其他覆土厚度;(4)盐碱、干旱或盐碱和干旱双重胁迫下乌拉尔甘草种子发芽势和发芽率显著降低;(5)100~300μmol/L褪黑素预处理可显著提高盐胁迫处理下乌拉尔甘草种子的发芽率和发芽势;(6)萘乙酸对乌拉尔甘草种子发芽无促进作用,但当浓度>50 mg/L时,乌拉尔甘草种子发芽势和发芽率显著降低;(7)吲哚乙酸浓度为50 mg/L时,乌拉尔甘草种子发芽势和发芽率最高。综上所述,乌拉尔甘草种子发芽具有一定的抗非生物胁迫能力,且适宜的外源植物激素浓度预处理可促进种子发芽。  相似文献

研究了不同浓度盐(NaCl)处理对蓝蓟种子发芽势、发芽率及幼苗生长的影响。结果表明:(1)不同盐胁迫处理对蓝蓟种子的萌发具有显著影响。随着盐处理浓度的增加,发芽率呈下降趋势,蓝蓟在无盐环境(蒸馏水)和低盐环境中(0.05 mol/L)长势最好、发芽率最高、发芽速度最快。(2)将不同盐溶液处理5 d的未萌发种子转移到蒸馏水后,蓝蓟种子的萌发能力均可恢复,原来较高盐浓度(0.3 mol/L~0.5 mol/L)下的种子在恢复后,其萌发恢复率均在75%以上,发芽率基本上随着原浓度的升高呈上升的趋势。研究表明,NaCl处理对蓟种子没有造成伤害,其抑制萌发是通过渗透效应而不是离子毒害。  相似文献

为了确定沙拐枣植物种子的萌发特性及最优播前预处理方法,在实验室条件下,对10种沙拐枣植物的种子进行了磨砺、硫酸和热水浸泡、冷藏、种子浸出液处理,然后进行发芽实验研究。萌发实验的结果表明,10种沙拐枣植物对于不同的种子预处理,均表现出相似的萌发反应。磨砺、硫酸浸泡和冷藏处理对种子萌发有明显地促进作用。与对照相比,种子浸出液处理对种子的发芽率、发芽速度均具有明显地抑制作用,并能增强种子的休眠。冷藏处理具有打破有活力的种子休眠、促进种子萌发的作用,但它与热水浸泡处理一样,对有活力种子表现出一定的致死作用。沙拐枣植物的萌发模式在不同种子预处理问均表现出明显的差异性。机械磨擦和硫酸处理能够促进种子的萌发率及发芽势。泡果沙拐枣(Callingonum junceum)在本项实验中表现出很强的萌发能力。  相似文献

以新疆北部4种十字花科短命植物为材料,研究不同浓度NaCl胁迫下其种子萌发及植株生长特性,以明确它们的耐盐性强弱。结果显示:(1)随着NaCl浓度的增加,4种植物种子萌发率、萌发指数和相对萌发率均不同程度下降,而相对盐害率则逐渐升高,并以绵果荠的耐盐性最好,其在300 mmol.L-1NaCl胁迫下仍有22.22%的萌发率。(2)4种植物的生长在50~150 mmol.L-1NaCl胁迫下没有受到明显抑制,在200 mmol.L-1浓度下受不同程度的抑制,而在300 mmol.L-1NaCl胁迫下抑制作用更严重,其中绵果荠在各浓度处理下长势均最好,能够在300 mmol.L-1NaCl胁迫下存活10 d。(3)在盐胁迫条件下,4种植物细胞质膜透性不同程度增加,而叶片相对含水量和光合色素含量均不同程度降低,并以绵果荠各项指标的变化幅度最小。可见,4种新疆短命植物种子萌发和植株生长均受到盐胁迫的伤害,但它们的耐盐阈值明显不同,并以绵果荠的耐盐性最强。  相似文献

研究外源GA对NaCl胁迫下黄瓜种子萌发和幼苗生长的影响.结果显示:(1)0和75mmol·L-1NaCl处理可促进种子萌发,浓度为100mmol·L-1及以上时,随着浓度的增加,种子萌发受抑程度越严重;(2)150mmol·L-1NaCl胁迫下,添加外源GA可显著提高黄瓜种子发芽率、发芽势、发芽指数、活力指数和a-淀粉酶活性;促进种子萌发,以100和150mg·L-1GA处理效果较好;(3)外源GA显著提高幼苗生物量,提高SOD和POD活性,降低MDA含量,以100mg·L-1GA处理效果较好.研究表明一定范围的外源GA可缓解盐害对黄瓜种子萌发和幼苗生长的抑制作用,诱导其耐盐性的提高.  相似文献

The aerial surfaces of the common or crystalline ice plant Mesembryanthemum crystallinum L., a halophytic, facultative crassulacean acid metabolism species, are covered with specialized trichome cells called epidermal bladder cells (EBCs). EBCs are thought to serve as a peripheral salinity and/or water storage organ to improve survival under high salinity or water deficit stress conditions. However, the exact contribution of EBCs to salt tolerance in the ice plant remains poorly understood. An M. crystallinum mutant lacking EBCs was isolated from plant collections mutagenized by fast neutron irradiation. Light and electron microscopy revealed that mutant plants lacked EBCs on all surfaces of leaves and stems. Dry weight gain of aerial parts of the mutant was almost half that of wild-type plants after 3 weeks of growth at 400 mM NaCl. The EBC mutant also showed reduced leaf succulence and leaf and stem water contents compared with wild-type plants. Aerial tissues of wild-type plants had approximately 1.5-fold higher Na(+) and Cl(-) content than the mutant grown under 400 mM NaCl for 2 weeks. Na(+) and Cl(-) partitioning into EBCs of wild-type plants resulted in lower concentrations of these ions in photosynthetically active leaf tissues than in leaves of the EBC-less mutant, particularly under conditions of high salt stress. Potassium, nitrate, and phosphate ion content decreased with incorporation of NaCl into tissues in both the wild type and the mutant, but the ratios of Na(+)/K(+) and Cl(-)/NO(3)(-)content were maintained only in the leaf and stem tissues of wild-type plants. The EBC mutant showed significant impairment in plant productivity under salt stress as evaluated by seed pod and seed number and average seed weight. These results clearly show that EBCs contribute to succulence by serving as a water storage reservoir and to salt tolerance by maintaining ion sequestration and homeostasis within photosynthetically active tissues of M. crystallinum.  相似文献

The shift from C3 to CAM was investigated as a function of both leaf and plant age in well-watered and salt-stressed (300 mM NaCl solution) plants of Mesembryanthemum crystallinum. Initiation of a night-time accumulation of malic acid, the decisive criterion of CAM, was followed in plants that were continuously stressed at different points in their life cycle. The deinducibility of CAM was examined after the release from stress by extensively rinsing the potting soil with de-mineralized water. Our results show that in M. crystallinum CAM is under strict developmental control, since CAM appeared only when a certain stage of development of the whole plant was reached. CAM was not present in any plant before this threshold, which was the same in salt-stressed as well as in well-watered plants. The metabolic shift coincided with the change from the seedling to the juvenile growth phase, and not with that from vegetative to reproductive growth, represented by the start of branching. The latter is timed to the end of extension growth. In well-watered plants, after this decisive point in development, a weak nighttime accumulation of malic acid could be measured (? 0.05 mol kgDW?1) in the oldest, mature leaves but not in young, developing ones. This “CAM capacity” gradually increased up to 0.2 mol kgDE?1 with further plant ageing. Leaf senescence, characterized by wilting and yellowing, diminished the CAM activity. In mature leaves salt stress drastically enhanced the magnitude of diurnal fluctuation in malic acid content. Removal of salt stress did not deinduce CAM activity, but diminished the amplitude of malic acid oscillations to some extent in those plants which had been stressed from early in their life cycle. In these plants, salt stress delayed plant development and growth thus retarding the life cycle. Well-watered plants, for example, branched about three weeks earlier than those that had been stressed continuously from one week after germination. After removal of stress a quasi-preserved earlier developmental stage in relation to the control plants determined the weaker CAM expression.  相似文献

比较3种打破滇青冈种子休眠方法的结果表明,经40℃处理后的效果最好,发芽率达74.5%.100 mg·L-1的赤霉素浸种处理的次之,发芽率为70.2%.种子种皮切伤和去除种皮后,发芽率增大,分别为96.0%和95.0%,并提早5 d萌发.种子抑制物质实验的结果显示,滇青冈种子的内种皮和种仁含有抑制种子萌发的物质.  相似文献

The effects of NaCl, putrescine (Put), and the combination of two agents on the contents of free polyamines (PA), peroxidase activity, and the ultrastructure of the mesophyll apoplast were studied in the third leaf pair of Mesembryanthemum crystallinum L. plants. The NaCl solution was added to soil daily for three days (100 ml of the 100 mM solution), and plants were sprayed with the 1 mM Put solution twice per day for a six-day period. The accumulation of Put and especially spermidine (Spd) by day 3 of salinization was followed by a dramatic drop in Put and Spd contents by day 6. In contrast, the activities of soluble and ion-bound peroxidases increased following a long lag-period. Treatments with Put and NaCl plus Put considerably enhanced this rise in two peroxidase activities. An electron microscopic examination of cell walls in the control and stressed plants demonstrated that a gap developed at the middle lamella with pockets filled with amorphous polysaccharides (AP), presumably pectins. At the maximum gap width, the pockets fused with the intercellular spaces, and, in this case, the intercellular spaces also contained AP. Following salinization, AP in the apoplast swelled and expanded. Apparently the genetic determination of high AP content in M. crystallinum plants is the basis for the ability of juvenile plants to bind Na+ and Cl ions and excess PA and also to accumulate water. In the plants treated with NaCl plus Put, the number of pockets and their volume increased, and the surface of some cell walls became plated with suberin, thus providing an additional barrier for ion transport from the pockets and intercellular spaces into cells. The formation of suberin plates was correlated with the high activity of ion-bound peroxidase essential for suberin deposition. The authors presume that H2O2 results from PA oxidation and, in its turn, induces the activity of peroxidase involved in the suberin plate formation.  相似文献
