Aims: To develop probiotics for the control of vibriosis caused by Vibrio anguillarum and Vibrio ordalii in finfish. Methods and Results: Kocuria SM1, isolated from the digestive tract of rainbow trout, was administered orally to rainbow trout (Oncorhynchus mykiss) for 2 weeks at a dose equivalent to c. 108 cells per g of feed and then challenged intraperitoneally with V. anguillarum and V. ordalii. Use of SM1 led to a reduction in mortalities to 15–20% compared to 74–80% mortalities in the controls. SM1 stimulated both cellular and humoral immune responses in rainbow trout, by elevation of leucocytes (5·5 ± 0·8 × 106 ml?1 from 3·7 ± 0·8 × 106 ml?1), erythrocytes (1·2 ± 0·1 × 108 ml?1 from 0·8 ± 0·1 × 108 ml?1), protein (23 ± 4·4 mg ml?1 from 16 ± 1·3 mg ml?1), globulin (15·7 ± 0·2 mg ml?1 from 9·9 ± 0·1 mg ml?1) and albumin (7·3 ± 0·2 mg ml?1 from 6·1 ± 0·1 mg ml?1) levels, upregulation of respiratory burst (0·05 ± 0·01 from 0·02 ± 0·01), complement (56 ± 7·2 units ml?1 from 40 ± 8·0 units ml?1), lysozyme (920 ± 128·8 units ml?1 from 760 ± 115·3 units ml?1) and bacterial killing activities. Conclusions: Kocuria SM1 successfully controlled vibriosis in rainbow trout, and the mode of action reflected stimulation of the host innate immune system. Significance and Impact of the Study: Probiotics can contribute a significant role in fish disease control strategies, and their use may replace some of the inhibitory chemicals currently used in fish farms.  相似文献

Abstract

The purpose of the study was to analyze the relationship between the serum concentration of soluble tumour necrosis factor-α type 1 (sTNF-R1), the severity of plaque-type psoriasis and therapeutic response. We compared sTNF-R1 in 25 patients treated with narrowband ultraviolet B (NB-UVB) radiation and 25 patients treated with systemic photochemotherapy (psoralen plus UVA – PUVA). The pretreatment Psoriasis Area and Severity Index (PASI) score and sTNF-R1 concentration were 16.32±5.26 and 1.99±0.40 ng ml?1, respectively, in the group treated with NB-UVB, and 17.22±3.48 and 2.07±0.31 ng ml?1, respectively, in the group treated with PUVA. The concentration of sTNF-R1 in healthy controls was 1.49±0.34 ng ml?1 (p<0.05 compared with patients with psoriasis). The pretreatment PASI score correlated with sTNF-R1 in both treatment groups (r=0.46 and r=0.44, p<0.05). NB-UVB and PUVA gave similar therapeutic effects (the PASI score after 20 treatments was 4.42±1.67 in the NB-UVB-treated group and 5.55±2.10 in PUVA-treated patients); however, the sTNF-R1 concentration at the same time differed significantly: 1.52±0.37 ng ml?1 and 1.98±0.39 ng ml?1 (p<0.001), respectively. Moreover, the decline in sTNF-R1 in both treatment groups was significant only in patients in whom the duration of skin lesions was less than 3 months. The results suggest that the value of serum sTNF-R1 concentration as a marker of response to phototherapy may depend on duration of skin lesions and the treatment method.  相似文献

Background: Pulmonary hypertension (PH) may be associated with subendocardial ischaemia. We investigated whether ischaemia-modified albumin (IMA), an established marker of ischaemia, is elevated in stable patients with PH.

Methods: We studied 32 patients with PH and an equal number of age-matched normal volunteers. We assessed serum IMA levels with the albumin cobalt-binding test.

Results: Patients’ mean ± SD (range) pulmonary arterial pressure was 56?±?12 (33–73) mmHg and their exercise capacity was 394?±?145 (121–688) m in the 6-min walk test. IMA was 92?±?14 (69–115) U ml?1 in the patient group and 93?±?9.4 (76–122) U ml?1 in the control group with no significant difference between the two (p?=?0.85), although almost one-third of the patients had detectable troponin-I.

Conclusions: We conclude that IMA, a marker of ischaemia, does not differ in patients with advanced clinically stable PH compared with normal subjects.  相似文献

Glutathione belongs to a vital intra‐ and extra‐cellular protective antioxidant and is found almost exclusively in its reduced form. The ratio between its reduced and oxidized within cells is often used as a marker of cellular toxicity. The objectives of the study were to (i) determine both the reduced (GSH) and oxidized glutathione (GSSG) and cysteine (Cys) in the sperm of the Acipenser baerii and Acipenser ruthenus, as well as in Perca fluviatilis and Sander lucioperca, and (ii) to demonstrate the differences in concentration levels between representatives of acipenseriform and teleost species. High performance liquid chromatography with electrochemical detection was employed. The average content of the thiols determined in the sperm samples were as follows: Acipenser baerii (cysteine 55 ± 8 μg ml?1; GSH 126 ± 19 μg ml?1; GSSG 49 ± 7 μg ml?1), Acipenser ruthenus (cysteine 62 ± 9 μg ml?1; GSH 768 ± 115 μg ml?1; GSSG 180 ± 16 μg ml?1), Sander lucioperca (cysteine 251 ± 38 μg ml?1; GSH 185 ± 28 μg ml?1; GSSG 93 ± 14 μg ml?1), Perca fluviatilis (cysteine 281 ± 42 μg ml?1; GSH 496 ± 74 μg ml?1; GSSG 138 ± 21 μg ml?1). Based on the results obtained it can be concluded that this method is sensitive and selective for the determination of these compounds in real samples. Results revealed differences in cysteine content between species of the two systematic categories but also showed that ratios between GSH and GSSG can vary between species while potentially predict oxidative stress in fish sperm.  相似文献

Injection of somatostatin‐14 (SS‐14) at 5 ng g?1 body mass (BM) into rainbow trout Oncorhynchus mykiss decreased (P < 0·05, cubic, r2 = 0·54) levels of growth hormone (GH) (1·5 ± 0·9 ng ml?1v. 6·6 ± 0·6 ng ml?1) over time when compared to controls. Somatostatin‐14 at 50 ng g?1 BM also decreased (P = 0·064, quadratic; r2 = 0·30) levels of GH (3·6 ± 2·1 ng ml?1v. 6·6 ± 0·6 ng ml?1) over time compared to controls. In a second study, passive immunization against SS‐14 (1 : 25 dose) increased (P = 0·10, cubic, r2 = 0·12) levels of GH (11·0 ± 4·8 ng ml?1v. 5·2 ± 1·4 ng ml?1) over time. Passively immunizing against SS‐14 (1 : 50 dose) increased (P < 0·05, cubic, r2 = 0·10) levels of GH (8·2 ± 2·3 ng ml?1v. 5·2 ± 1·4 ng ml?1) over time compared to controls. Overall, in the active immunization study there was no difference (P > 0·10) in specific growth rate (G) or feed conversion ratio (FCR) between the three treatment groups during the 9 weeks of the study. Only four of the fish immunized against SS‐14, however, developed antibody titres against SS. Compared to controls, these fish exhibited a G of 0·89 ± 0·09 v. 0·56 ± 0·09% per 3 weeks and FCR of 0·80 ± 0·04 v. 1·20 ± 0·05 g g?1. In SS‐14 immunized fish, levels of GH decreased (P < 0·05) by day 63 while levels of insulin like growth factor‐I (IGF‐I) increased (P < 0·05) by day 42 and 63. These results indicate the hypothalamic hormone SS‐14 regulates GH secretion similarly in rainbow trout as it does in mammals. Active immunization against SS‐14 could improve growth performance in rainbow trout but enhanced G and FCR is dependent upon generation of antibody titres.  相似文献

Myosmine is a minor tobacco alkaloid with widespread occurrence in the human diet. Myosmine is genotoxic in human cells and is readily nitrosated and peroxidated yielding reactive intermediates with carcinogenic potential. For biomonitoring of short-term and long-term exposure, analytical methods were established for determination of myosmine together with nicotine and cotinine in plasma, saliva and toenail by gas chromatography–mass spectrometry (GC/MS). Validation of the method with samples of 14 smokers and 10 non-smokers showed smoking-dependent differences of myosmine in toenails (66?±?56 vs 21?±?15?ng?g?1, p?<0.01) as well as saliva (2.54?±?2.68 vs 0.73?±?0.65?ng ml?1, p <0.01). However, these differences were much smaller than those with nicotine (1971?±?818 vs 132?±?82?ng g?1, p <0.0001) and cotinine (1237?±?818 vs <35?ng?g?1) in toenail and those of cotinine (97.43?±?84.54 vs 1.85?±?4.50?ng ml?1, p <0.0001) in saliva. These results were confirmed in plasma samples from 84 patients undergoing gastro-oesophageal endoscopy. Differences between 25 smokers and 59 non-smokers are again much lower for myosmine (0.30?±?0.35 vs 0.16?±?0.18?ng?ml?1, p <0.05) than for cotinine (54.67?±?29.63 vs 0.61?±?1.82?ng ml?1, p <0.0001). In conclusion, sources other than tobacco contribute considerably to the human body burden of myosmine.  相似文献

ABSTRACT

The dissipation and detoxification of nicosulfuron (NS) by Pseudomonas aeruginosa B9 isolated from a cornfield soil was investigated. The fastest decline of NS occurred at 40 µg ml?1 in liquid media with 0.25% glucose plus 0.05% yeast extract (DT50 = 4 days) with a notable pH reduction (pH ? 5). Bioassay tests showed considerable phytotoxicity of NS for Cress (Lepidium sativum L.) with 50% shoot growth inhibition (SGI) at 40 µg ml?1. The dissipation of NS (40 µg ml?1) by the B9 isolate reduced the SGI significantly (SGI: up to 45 ± 3%) compared to the non-inoculated media (SGI: up to 58 ± 4%). In soils with the B9 isolate, NS dissipation, especially at 0.3 µg g?1, was faster with a more significant SGI reduction (k = 0.08 ± 0.00 day?1; SGI = 2 ± 1%) compared to non-inoculated samples (k = 0.03 ± 0.00 day?1; SGI = 8 ± 1%). NS initially inhibited soil respiration, microbial biomass carbon, and dehydrogenase activity. The effect was however transient, and these parameters recovered within 10 days, especially in the presence of the isolate. Overall, this study proves Pseudomonas aeruginosa B9 as a suitable candidate for bioremediation of NS in contaminated sites.  相似文献

For a period of 1 year, some blood parameters were evaluated on a monthly basis in a population of adult European sea bass (Dicentrarchus labrax L.) intensively reared in floating marine cages (Ionian Sea, Mediterranean). From April (13 months old) to July (16 months old) males (35–50%) and sexually indeterminate individuals were collected. From August to March (24 months old) only males were sampled. During this period the percentage of spermiated males was highest (100%) from November (20 months old) to January (22 months old). Plasma testosterone in males was inversely related to sunlight (h month?1) and was elevated between October and January, when males first achieved sexual maturity. Testosterone showed the highest value (0.49 ± 0.02 ng ml?1) in January and the lowest (0.09 ± 0.02 ng ml?1) in March. Haematocrit, red blood cell counts and haemoglobin concentration were elevated from November to March, being inversely related to sunlight. The two latter parameters were also inversely related to daily food intake. Haematocrit, red blood cell counts and haemoglobin concentration were highest in December [53 ± 1%, (5.36 ± 0.06) × 106 mm?3, 10.08 ± 0.14 g 100 ml?1, respectively] and lowest in June [35 ± 1%, (3.33 ± 0.05) × 106 mm?3, 6.47 ± 0.13 g 100 ml?1, respectively]. White blood cell counts were not correlated with sea water temperature, sunlight or daily food intake. They were highest in February [(8.45 ± 0.20) × 104 mm?3] and lowest in April [(6.07 ± 0.14) × 104 mm?3]. Total plasma protein concentration (4.88 ± 0.11–5.93 ± 0.10 g 100 ml?1) and mean cell volume (93.3 ± 0.9–105.5 ± 1.8 μm3) showed small fluctuations throughout the year. Sexual maturity appears to be a major factor that significantly affects haemopoiesis of D. labrax. This study contributes to the evaluation of normal levels of some blood parameters in European sea bass, which are helpful for the assessment of physiological status and health of this species.  相似文献

The study purpose was the analysis of barbel Barbus barbus (L.) milt quality and quantity with regard to the time following stimulation with [(D‐Ala6, Pro9NEt)‐mGnRH+metoclopramide] i.e. Ovopel. Selected parameters such as total volume of milt (TVM, ml), volume of milt per kg of body weight (VOM, ml kg?1 b.w.), sperm concentration (×109 ml?1), total sperm production (TSP, ×109), osmolality (mOsm kg?1) and pH of seminal plasma were determined. Sperm motility was analyzed by the CASA system, i.e. the percentage of sperm motility (MOT, %) and their progressive motility (PRG, %), curvilinear velocity (VCL, μm s?1) and straight line velocity (VSL, μm s?1), amplitude of lateral head displacement (ALH, μm), and beat cross frequency (BCF, Hz). Milt was collected from 12 specimens (N = 12), with the first portion obtained 12 h following treatment with Ovopel (1 granule kg?1 b. w.). Subsequent portions of milt were taken at 24 h intervals, i.e. after 36, 60, 84, 108, and 132 h. The control group (Control, N = 12) was injected with 0.9% NaCl at 0.5 ml kg?1b.w. from which milt was taken 12 h post injection. The highest TVM, VOM and TSP values were recorded 12 h after Ovopel treatment (3.2 ± 0.7 ml, 36.7 ± 10.5 ml kg?1 b.w. and 39.1 ± 9.4 × 109, respectively); lowest values were recorded after 132 h (0.8 ± 0.4 ml, 11.1 ± 6.5 ml kg?1b.w. and 13.7 ± 7.5 × 109, respectively). The highest seminal plasma osmolality values (300.0 ± 42.6 mOsm kg?1) as well as the lowest sperm concentration (12.5 ± 1.5 × 109 ml?1) were observed 12 h after Ovopel treatment. No significant differences in the percentage of sperm motility (MOT) were noted during any of the periods after hormonal stimulation, however, a change in the character of their movement (PRG) was observed. The lack of significant differences (P > 0.05) in VCL and VSL values between 12 h and 60–132 h indicates that the lengthening of time does not lead to a decrease in sperm velocity and, therefore, is not likely to have a negative impact on their quality. The highest ALH (1.9 ± 0.2 μm) and BCF (11.5 ± 1.1 Hz) values were observed when the effect of stimulation was most noticeable, i.e. 12 h after Ovopel treatment. Based on the total milt volume and sperm production, the best time for milt collection from barbel is 12 h post‐hormonal treatment; 84 h post‐hormonal treatment, TVM, VOM, TSP and some CASA parameters decreased, which suggest the same aging process in sperm.  相似文献

Transforming growth factor-β1 (TGFβ1) is thought to be an inhibitor of the keratinocyte hyperproliferation associated with psoriasis. The aim of this study was to evaluate plasma TGFβ1 and TGFβ2 concentrations in psoriatic patients as possible indicators of treatment efficacy. TGFβ concentrations were measured in the plasma of 26 patients with psoriasis using an enzyme immunoassay and analysed with respect to the psoriasis area and severity index (PASI) before and after treatment with salicylic acid and/or sulphur followed by dithranol ointment. Baseline plasma concentrations of both TGFβ1 and TGFβ2 (20.3±2.2 ng ml?1 and 0.14±0.02 ng ml?1, respectively) did not differ significantly from control values (18.3±1.6 ng ml?1 and 0.14±0.03 ng ml?1, respectively). However, a significant positive correlation (r=0.69) between the baseline PASI and TGFβ1, but not TGFβ2, values was demonstrated. The pretreatment TGFβ1 concentration in patients with a PASI ≥15 (26.6±3.2 ng ml?1) was significantly higher than control values. There were no significant elevation of pretreatment TGFβ1 concentrations in patients with a PASI<15, or with respect to TGFβ2 in both groups. Treatment caused a significant decrease in TGFβ1, but only in patients with a PASI≥15. Patients with baseline TGFβ1 concentrations exceeding the mean of the control group had a PASI value that was significantly higher than that of patients with a TGFβ1 concentration below the mean of the controls. These results confirmed an association between plasma TGFβ1 concentration and psoriasis severity, and demonstrated its normalization during treatment. Measurement of TGFβ1 in plasma should be considered as a possible biomarker of psoriasis activity during its management.  相似文献

The aim of the study was to compare the physico‐chemical parameters of milt from sea trout (Salmo trutta m. trutta), brook trout (Salvelinus fontinalis) and rainbow trout (Oncorhynchus mykiss). Milt was collected by stripping and spermatozoa concentrations, were determined and compared with sperm motility and spermatocrit values along with seminal plasma indices (pH, osmolality, sodium, potassium, chlorine, calcium, magnesium, glucose and protein concentrations). The highest spermatozoa concentration of 22.3 ± 6.7 × 109 ml?1 was found in the sea trout milt, and was significantly different of those observed in brook trout (11.9 ± 3.3 × 109 ml?1) and rainbow trout (10.7 ± 4.4 × 109 ml?1). The values for pH and K+ did not differ significantly among species. The mean pH was 8.0 in the milt of each species and the K+ concentrations ranged from 24.8 ± 7.2 to 30.5 ± 7.6 mm L?1. Considerable differences were determined for the Ca2+ ions concentrations. The highest value was found in sea trout (1.7 ± 0.3 mm L?1), while in the rainbow trout it was 0.7 ± 0.5 and in the brook trout 0.4 ± 0.1 mm L?1. The most pronounced differences were found in the glucose concentration cause of its unnaturally low concentration in rainbow trout of the mean value of 6.0 ± 15.2 mg L?1. The mean value in sea trout and brook trout was 185.0 ± 172.4 and 231.2 ± 148.4 mg L?1 respectively. For all species, protein mean values were below 1.3 g L?1. The mean osmolality was between 230.6 ± 98.6 and 272.0 ± 26.4 mOsm kg?1 in the species studied. No correlation was found between any components determined in milt and the spermatozoa motility (P > 0.05). The sperm concentration was positively correlated with the protein content in the milt of the three species studied, other less exhibited correlation was found.  相似文献

This study investigated the antimicrobial effects of the ethanolic extract of Brazilian red propolis (BRP) on multispecies biofilms. A seven-day-old subgingival biofilm with 32 species was grown in a Calgary device. Biofilms were treated with BRP (1,600, 800, 400 and 200?μg ml?1) twice a day for 1?min, starting from day 3. Chlorhexidine (0.12%) and dilution-vehicle were used as positive and negative controls, respectively. On day 7, metabolic activity and the microbial composition of the biofilms by DNA-DNA hybridization were determined. The viability data were analyzed by one-way ANOVA followed by Tukey’s post hoc, whereas the microbial composition data were transformed via BOX-COX and analyzed using Dunnett’s post hoc. BRP (1,600?μg ml?1) decreased biofilm metabolic activity by 45%, with no significant difference from chlorhexidine-treated samples. BRP (1,600?μg ml?1) and chlorhexidine significantly reduced levels of 14 bacterial species compared to the vehicle control. Taken together, BRP showed promising antimicrobial properties which may be useful in periodontal disease control.  相似文献

The reliability of self-reported smoking behaviour can vary and may result in bias if errors in misclassification vary with outcome. We examined whether self-report was an accurate measure of current smoking status in patients with malignant or non-malignant respiratory disease. Smoking behaviour was assessed by self-report and by analysis of whole blood for cotinine, a biomarker of exposure to cigarette smoke, in 166 patients attending a bronchoscopy clinic. Cotinine levels ranged from 2.5 to &gt;400 ng ml?1 blood and were higher in self-reported current smokers (173±123 ng ml?1) than in never smokers (3.7±8.7 ng ml?1) or ex-smokers (20.5±49.0 ng ml?1). Cotinine levels in self-reported current smokers increased with the numbers of cigarettes smoked (p=0.06), and levels in smokers and ex-smokers decreased with the reported length of time since the last cigarette (p=0.001). Using a cotinine level of 20 ng ml?1 and self-report as the gold standard, the sensitivity and specificity for defining current smoking status were 90.2% and 82.4%, respectively. Out of a total of 125 self-reported current non-smokers, 23 (18.4%) had cotinine levels greater than 20 ng ml?1. Smoking prevalence was significantly underestimated by self-report (24.7%) when compared with that defined using blood cotinine levels (36.1%: p&lt;0.001). Misclassification of current smoking status was particularly high in ex-smokers, in patients without malignant respiratory disease, in men, and in those below the median age. Such differential misclassification may result in bias in studies examining associations between current smoking habits and disease risk.  相似文献

The hemolymph of last instar Manduca sexta larvae contains a protein factor that enhances ecdysone synthesis by prothoracic glands in vitro. The titer of the factor fluctuates during development in a pattern that suggests that it is regulated by juvenile hormone (JH). In untreated control larvae, the titer drops from 2.17 U ml?1 on day 1 to 0.27 U ml?1 on day 3. When larvae were treated with (7S)-hydroprene (a JH analog), the titer remained elevated (2.09 U ml?1 on day 3). JH I, however, was ineffective in preventing the precommitment drop in the titer of the factor. After pupal commitment, the titer of the factor increases in untreated larvae from 0.84 U ml?1 on day 5 to 1.62 U ml?1 on day 7. This increase was blocked when the sources of JH (the corpora allata) were removed on day 5 by head ligation. When head-ligated day 5 larvae were treated with either (7S)-hydroprene or JH I, the titer of the factor was driven to a level (1.88 U ml?1 and 2.05 U ml?1, respectively) that was not significantly different from that found in untreated day 7 larvae (1.62 U ml?1). The combined results indicate the titer of the hemolymph factor is regulated by JH.  相似文献

Abstract

Anaerobic ruminal fungi may play an active role in fibre degradation as evidenced by the production of different fibrolytic enzymes in culture filtrate. In the present study, 16 anaerobic fungal strains were isolated from ruminal and faecal samples of sheep and goats. Based on their morphological characteristics they were identified as species of Anaeromyces, Orpinomyces, Piromyces and Neocallimastix. Isolated Neocallimastix sp. from goat rumen showed a maximum activity of CMCase (47.9 mIU ml?1) and filter paper cellulase (48.3 mIU ml?1), while Anaeromyces sp. from sheep rumen showed a maximum xylanolytic activity (48.3 mIU ml?1). The cellobiase activity for all the isolates ranged from 178.0 – 182.7 mIU ml?1. Based on the enzymatic activities, isolated Anaeromyces sp. from sheep rumen and Neocallimastix sp. from goat rumen were selected for their potential of in vitro fibre degradation. The highest in vitro digestibility of NDF (23.2%) and DM (34.4%) was shown for Neocallimastix sp. from goat rumen, as compared to the digestibility of NDF and DM in the control group of 17.5 and 25.0%, respectively.  相似文献

Objectives

To investigate the potential role and underlying mechanism of Sirtuin2 (SIRT2) in regulating high glucose (HG)-induced vascular endothelial cell injury by using human umbilical vein endothelial cells (HUVECs).

Results

SIRT2 mRNA and protein expression levels were decreased in HG-treated HUVECs. SIRT2 overexpression increased viability, decreased apoptosis and reduced levels of reactive oxygen species in HG-treated HUVECs. SIRT2 overexpression decreased TNF-α expression (146.5 ± 22.8 pg TNF-α ml?1) relative to that in the empty vector group (263.5 ± 18.5 pg TNF-α ml?1) and decreased MCP-1 expression (63.8 ± 9.85 pg MCP-1 ml?1) relative to that in the empty vector group (105.8 ± 8.5 pg MCP-1 ml?1). SIRT2 overexpression decreased the acetylation of p53 by 33% and decreased the acetylation of NF-κB p65 by 58% in HG-treated HUVECs.

Conclusion

SIRT2 prevents HG-induced vascular endothelial cell injury through suppressing the p53 and NF-κB signaling pathways.

Abstract

The aim of the study was to assess plasma and scales levels of interleukin (IL) 18 collected from psoriatic patients with different disease activity. IL-18 concentrations were measured using an enzyme immunoassay in the plasma and scales of 34 patients with chronic plaque type psoriasis. IL-18 levels were analysed with respect to plasma-transforming growth factor β1 (TGF-β1), the disease duration and the duration of the present relapse, and psoriasis area and severity index (PASI). Plasma IL-18 concentration varied from 90 to 1300 pg ml?1 and means (368.2±42.4 pg ml?1) were significantly elevated in comparison with healthy controls (205.9±31.8 pg ml?1). The presence of IL-18 was also demonstrated in scales from skin lesions. Treatment caused a significant decrease of plasma IL-18 concentration to 250.2±13.8 pg ml?1. There was a significant correlation between plasma IL-18 levels and PASI values (r=0.554). There was no correlation between IL-18 concentration in scales and PASI, between IL-18 concentrations in plasma and scales, and between plasma IL-18 and the disease duration or duration of present relapse. Plasma TGF-β1 concentration demonstrated a significant correlation with PASI (r=0.353), but not with IL-18 levels in plasma (r=0.063) and scales (0.141). The sum of plasma levels of IL-18 and TGF-β1 divided by the optimal coefficient demonstrated a statistically significant correlation with the highest r-value. The findings confirm an association between plasma IL-18 concentration and psoriasis severity. Moreover, it was shown that combined measurement of IL-18 and TGF-β1 in plasma can be considered as a possible biomarker of psoriasis activity.  相似文献

1. We offered Scenedesmus obliquus in five densities, from 0.5 to 8 ± 106 cells ml?1, to the rotifer Anuraeopsis fissa. Growth rates (r) during the exponential phase (first 7 days) were significantly and positively related to food density. The r values (mean ± SD) varied between 0.454 ± 0.067 and 0,856 ± 0.090, from the lowest to the highest food concentration, respectively. Population growth went through a phase of exponential increase which lasted from 7 to 10 days before a plateau and, in some cases, a decrease occurred. 2. There was a linear relation between food density and rotifer plateau density. At the highest food density, a peak abundance (mean ± SD) of 2312 ± 226 individuals ml?1 was reached, while at the lowest food density there was no identifiable single peak but a plateau, at a density of 361 ± 62 ind. ml?1. 3. The egg ratio decreased with increasing population density. The ratio of loose eggs to eggs attached to females indicated that more eggs became detached at higher food densities. 4. As in many other rotifer species studied so far, population density of A. fissa was less stable at higher algal food concentrations. Cost-effective production of proteases, which are robust enough to function under harsh process conditions, is always sought after due to their wide industrial application spectra. Solid-state production of enzymes using agro-industrial wastes as substrates is an environment-friendly approach, and it has several advantages such as high productivity, cost-effectiveness, being less labor-intensive, and less effluent production, among others. In the current study, different agro-wastes were employed for thermoalkali-stable protease production from Bacillus subtilis K-1 under solid-state fermentation. Agricultural residues such as cotton seed cake supported maximum protease production (728?U?ml?1), which was followed by gram husk (714?U?ml?1), mustard cake (680?U?ml?1), and soybean meal (653?U?ml?1). Plackett–Burman design of experiment showed that peptone, moisture content, temperature, phosphates, and inoculum size were the significant variables that influenced the protease production. Furthermore, statistical optimization of three variables, namely peptone, moisture content, and incubation temperature, by response surface methodology resulted in 40% enhanced protease production as compared to that under unoptimized conditions (from initial 728 to 1020?U?ml?1). Thus, solid-state fermentation coupled with design of experiment tools represents a cost-effective strategy for production of industrial enzymes.  相似文献

A blood sample was taken from each of 15 stallions at monthly intervals for 14 consecutive months. Plasma concentrations of estrogens and testosterone were measured by radioimmunoassay methods. Estrogens in peripheral blood were present in much higher amounts than testosterone and were principally in a water-soluble, solvolyzable form (> 98%). The major component in the solvolyzed extracts behaved chromatographically as estrone. The mean plasma level (± S.E.) of estrogens averaged across months was 52.9 ± 4.5 ng ml?1. Individual stallions showed considerable month-to-month variation; for example, single monthly samples ranged from 29.5 to 160.6 ng ml?1 for the stallion with the highest single value.The highest mean monthly concentration was 69 ± ng ml?1 in May, and plasma levels were < 40 ng ml?1 during November and December. For the 11 Thoroughbred stallions in the study, the mean concentrations of estrogens were 73 ± 5.8 ng ml?1 for May to July and 45 ± 4.1 ng ml?1 for November to January (P > 0.001).The mean monthly concentrations (± S.E.) of testosterone ranged from 0.22 ± 0.05 to 0.90 ± 0.14 ng ml?1, and individual samples ranged from < 0.02 to 2.8 ng ml?1 of plasma. While the highest mean level of testosterone was seen in September, there was a significant difference (P < 0.01) between the values in the breeding season (May–July, 0.73 ± 0.07 ng ml?1) and the non-breeding season (November–January, 0.38 ± 0.08 ng ml?1). No marked seasonal changes were observed, however, in testosterone levels in several stallions. It was concluded that plasma estrogen levels may provide a more sensitive index of endocrine function of the testes in the stallion.  相似文献
