Abstract Bacteriodes gingivalis W50 was grown in a chemostat at pH 7.5 under haemin-limited and haemin excess conditions in order to provide cells with a known high or low inefectivity and virulence for mice, respectively. The activities of enzymes and formation of cytotoxic fermentation products by these cells were compared. No significant differences in chymotrypsin activity or in the ability to degrade hyaluronic acid were found; neither were there any significant differences in the production of butyrate, propionate or succinate. At pH 7.5, trypsin activity was 3.5-fold higher in cells grown under haemin excess conditions whereas collagenolytic activity was nearly 3-fold higher in haemin-limited cells. Although collagenolytic activity may be important in tissue damage, a high ratio of trypsin to collagen breakdown activities was associated with virulent cells grown under an excess of haemin.  相似文献

Porphyromonas gingivalis strain W50 was grown in a chemostat either under haemin limitation or haemin excess at pH 7.3. Cells and the extracellular vesicle (ECV) and extracellular protein (EP) fractions were separated, quantified, and assayed for haemagglutination, protease activity and haemin binding. Under haemin-limitation, despite a reduction in cell yield, there was a 2.5-fold increase in the gravimetric yield of extracellular vesicles. Cells and vesicles from haemin-limited cultures, haemagglutinated sheep red blood cells to higher titres than their haemin-excess counterparts. Growth in haemin-excess conditions resulted in increased haemin-binding capacities of ECV, cells and EDTA-extracted outer membrane. Cells grown under haemin-excess showed a 2-fold elevation in specific activity towards the substrate N-alpha-benzoyl-L-arginine-p-nitroanilide (L-BAPNA) compared to haemin-limited cells. The specific activities against L-BAPNA for haemin-limited ECV were 3-fold greater than their haemin-excess counterparts. These vesicle activities represented 25% and 3% of the total culture protease activity under haemin limited and haemin excess conditions respectively.  相似文献

The focus of this review is on the recent data from the omics approaches, measuring the presence of methylotrophs in natural environments. Both Bacteria and Archaea are considered. The data are discussed in the context of the current knowledge on the biochemistry of methylotrophy and the physiology of cultivated methylotrophs. One major issue discussed is the recent metagenomic data pointing toward the activity of “aerobic” methanotrophs, such as Methylobacter, in microoxic or hypoxic conditions. A related issue of the metabolic distinction between aerobic and “anaerobic” methylotrophy is addressed in the light of the genomic and metagenomic data for respective organisms. The role of communities, as opposed to single-organism activities in environmental cycling of single-carbon compounds, such as methane, is also discussed. In addition, the emerging issue of the role of non-traditional methylotrophs in global metabolism of single-carbon compounds and the role of methylotrophy pathways in non-methylotrophs is briefly mentioned.  相似文献

The balance between facilitation and competition in plants changes with species characteristics and environmental conditions. Facilitative effects are common in natural ecosystems, particularly in stressful environments or years. Contrarily, in artificial associations of plants, such as agroforestry systems, some authors have suggested that even when facilitative effects may occur, net balance of tree effects on grasses is usually negative, particularly in dry environments. The aim of this study was to determine the net effect of the exotic ponderosa pine on the native grass Festuca pallescens (St. Ives) Parodi in agroforestry systems in Patagonia. Soil water content, plant water status, and relative growth were measured in the grass growing in different treatments (determined by tree cover level) during two growing seasons with contrasting climatic conditions. Facilitative effects of trees over grass water status were recorded only when water availability was high. A net negative effect was detected on dates when soil water content was very low and evaporative demand was high. The strength of these negative effects depended on tree density and climatic conditions, being higher in treatments with lower tree canopy cover. These results indicate that the positive effect of trees could only be expected under relatively low stress conditions. However, relative growth of grasses was always similar in plants growing in forested plots than in open grassland. Differences in biomass allocation for grasses growing in shade and open habitats may reconcile these contrary results. Our results highlight the importance of the physiology of a species (relative drought and shade tolerance) in determining the response of a plant to a particular interacting species.  相似文献

Thermophysiological traits, particularly thermal tolerances and sensitivity, are key to understanding how organisms are affected by environmental conditions. In the face of ongoing climate change, determining how physiological traits structure species’ ranges is especially important in tropical montane systems. In this study, we ask whether thermal sensitivity in physiological performance restricts montane lizards to high elevations and excludes them from the warmer environments reported at low elevations. For three montane lizard species in the Brazilian Atlantic Forest, we collect thermophysiological data from lizards in the highest elevation site of each species’ distribution, and ask how well the individuals exhibiting those traits would perform across the Atlantic Forest. We use microclimatic and organism‐specific models to directly relate environmental conditions to an organism's body temperature and physiological traits, and estimate measures of thermophysiological performance. Our findings demonstrate that thermophysiological constraints do not restrict montane lizards to high elevations in this system, and thus likely do not determine the warm boundaries of these montane species’ distributions. Results also suggest that competition may be important in limiting the warm boundaries of the species’ ranges for two of the focal species. These experimental results suggest that caution should be used when claiming that physiology drives patterns of diversity and endemism within montane environments. They also highlight the importance of interdisciplinary experimental studies that bridge the fields of evolution and ecology to improve predictions of biological responses to future environmental shifts.  相似文献

In order to characterise the sensitivity of antioxidative systems to temperature-induced oxidative stress, two species (Coleus blumei and Fagus sylvatica, L.) representative of environments with contrasting temperature characteristics have been exposed to low or high temperatures of 10 or 35 °C, respectively. Beech leaves were harvested in light and darkness. Coleus leaves were separated into green and white leaf tissue. The thermal dependencies of the activities of protective enzymes and chlorophyll fluorescence over a temperature range from 10 to 35 °C were determined. Ascorbate peroxidase activities were activated at low temperatures in vitro and, thereby, may provide an instantaneous protection against H2O2 accumulation which is faster than de novo synthesis. Monodehydroascorbate radical reductase was apparently not involved in short-term acclimation to low or high temperature. After short-term acclimation to low temperature, glutathione reductase and glutathione were more diminished in Coleus than in beech. Both species contained higher concentrations of ascorbate and glutathione at high temperatures than at low temperatures whereas glutathione reductase activity increased. Ascorbate peroxidase activity from Coleus leaves, though detectable under standard assay conditions (25 °C), failed at 35 °C in vitro. The results suggest that the higher temperature susceptibility of Coleus than that of beech was associated with a differential loss in glutathione reductase/glutathione at low temperature and an inhibition of ascorbate peroxidase at high temperature. Since the thermal dependencies of antioxidative enzymes were significantly affected by the preceding environmental conditions, the relative enzymatic activities determined under standard assay conditions may not be representative of enzymatic activities in foliage exposed to varying environmental temperatures.  相似文献

Seasonal changes in stress steroid hormone secretions are thought to reflect investment in self-maintenance versus reproduction. The capricious conditions hypothesis (CCH) posits that reduced corticosterone (CORT) secretion during stress coincident with parental phases of breeding is necessary in harsh environments because a full response would otherwise trigger repeated nest abandonments. To test this hypothesis, we measured seasonal changes in stress physiology in free-living red crossbills (Loxia curvirostra), an opportunistically breeding songbird that regularly breeds in summer and winter. This species allows unique comparisons of breeding physiology under very different seasonal environmental conditions within locations. We found strong support for the CCH: red crossbills showed reduced CORT secretion only when in high reproductive condition in the winter, when compared with summer breeders and winter non-breeders. These data demonstrate that behavioural status and local environmental conditions interact to affect mechanisms underlying investment trade-offs, presumably in a way that maximizes lifetime reproductive success.  相似文献

The binding of Congo red to P. gingivalis W50 grown in a chemostat under haemin-limitation and haemin-excess was quantified. Congo red bound to both haemin-excess and haemin-limited cells with similar capacity and affinity. Binding of Congo red was greater than for ferri- (haemin) or ferroprotoporphyrin IX (haem), and was not influenced by redox potential at low added ligand concentrations. Both haemin-limited and haemin-excess cells showed positive co-operativity towards Congo red binding. Pre-exposure of haemin-limited and haemin-excess cells to sub-saturating concentrations of ferriprotoporphyrin IX did not affect Congo red binding, whereas pre-exposure of haemin-excess cells to ferroprotoporphyrin IX increased binding. Iron protoporphyrin IX binding was enhanced after exposure of both haemin-excess and haemin-limited cells to Congo red, especially under reducing conditions. These results confirm that Congo red binding cannot be used as an indirect measure of haemin binding, nor can Congo red be used to inhibit haemin binding to P. gingivalis.  相似文献

We examined patterns of seedling root architecture, morphology and anatomy in Australian perennial plants chosen as phylogenetically independent contrasts (PICs) for rainfall in the areas they inhabit. Our objective was to assess whether there are consistent evolutionary patterns in structure of seedling root systems in species from different rainfall environments when examined across multiple evolutionary lineages. Seedlings were grown to a standardised developmental stage under controlled conditions. We found that seedling root systems of species restricted to low rainfall environments are characterised by greater proportional allocation to main root axis and have proportionally smaller main root axis diameter and areas of stele and xylem. Species of low rainfall environments also had higher specific root length (SRL) of the main axis, but lower SRL when the entire root system was considered. Seedling root system elongation rates were higher in species of high rainfall relative to those of low rainfall environments, paralleling expected differences in relative growth rate. The higher root system elongation rates in species of high rainfall environments were associated with greater numbers of growing tips in the root system, but not with differences in elongation rates of individual tips, relative to species of low rainfall environments.  相似文献

The record of life during the Proterozoic is preserved by several different lithologies, but two in particular are linked both spatially and temporally: chert and carbonate. These lithologies capture a snapshot of dominantly peritidal environments during the Proterozoic. Early diagenetic chert preserves some of the most exceptional Proterozoic biosignatures in the form of microbial body fossils and mat textures. This fossiliferous and kerogenous chert formed in shallow marine environments, where chert nodules, layers, and lenses are often surrounded by and encased within carbonate deposits that themselves often contain kerogen and evidence of former microbial mats. Here, we review the record of biosignatures preserved in peritidal Proterozoic chert and chert-hosting carbonate and discuss this record in the context of experimental and environmental studies that have begun to shed light on the roles that microbes and organic compounds may have played in the formation of these deposits. Insights gained from these studies suggest temporal trends in microbial-environmental interactions and place new constraints on past environmental conditions, such as the concentration of silica in Proterozoic seawater, interactions among organic compounds and cations in seawater, and the influence of microbial physiology and biochemistry on selective preservation by silicification.  相似文献

Species may be able to respond to changing environments by a combination of adaptation and migration. We study how adaptation affects range shifts when it involves multiple quantitative traits evolving in response to local selection pressures and gene flow. All traits develop clines shifting in space, some of which may be in a direction opposite to univariate predictions, and the species tracks its environmental optimum with a constant lag. We provide analytical expressions for the local density and average trait values. A species can sustain faster environmental shifts, develop a wider range and greater local adaptation when spatial environmental variation is low (generating low migration load) and multitrait adaptive potential is high. These conditions are favoured when nonlinear (stabilising) selection is weak in the phenotypic direction of the change in optimum, and genetic variation is high in the phenotypic direction of the selection gradient.  相似文献

The predictive adaptive response (PAR) hypothesis proposes that animals adjust their physiology and developmental trajectory during early life in anticipation of their future environments. Accordingly, when environmental conditions in early life match environmental conditions during adulthood, individual fitness should be greater. Here, we test this hypothesis in a long-lived mammal, the roe deer, using data from two contrasting populations, intensively monitored for more than 35 years. In the highly productive site, the fitness of female roe deer increased with the quality of environment during adulthood and, contrary to predictions of PAR, individuals born in good conditions always outperformed those born under poor conditions. In the resource-limited site, the fitness of female roe deer born in poor years was better than those born in good conditions in poor years when the animals were adult, but not in good years. Although consistent with predictions of PAR, we showed that this pattern is likely to be a consequence of increased viability selection during the juvenile stage for animals born in poor years. While PARs are often advanced in evolutionary medicine, our findings suggest that detailed biological processes should be investigated before drawing conclusions about the existence of this phenomenon.  相似文献

Arid environments provide ideal ground for investigating the mechanisms of adaptive evolution. High temperatures and low water availability are relentless stressors for many endotherms, including birds; yet birds persist in deserts. While physiological adaptation probably involves metabolic phenotypes, the underlying mechanisms (plasticity, genetics) are largely uncharacterized. To explore this, we took an intraspecific approach that focused on a species that is resident over a mesic to arid gradient, the Karoo scrub‐robin (Cercotrichas coryphaeus). Specifically, we integrated environmental (climatic and primary productivity), physiological (metabolic rates: a measure of energy expenditure), genotypic (genetic variation underlying the machinery of energy production) and microbiome (involved in processing food from where energy is retrieved) data, to infer the mechanism of physiological adaptation. We that found the variation in energetic physiology phenotypes and gut microbiome composition are associated with environmental features as well as with variation in genes underlying energy metabolic pathways. Specifically, we identified a small list of candidate adaptive genes, some of them with known ties to relevant physiology phenotypes. Together our results suggest that selective pressures on energetic physiology mediated by genes related to energy homeostasis and possibly microbiota composition may facilitate adaptation to local conditions and provide an explanation to the high avian intraspecific divergence observed in harsh environments.  相似文献

High-temperature-oriented production of bacterial penicillin acylase (PAC), which is usually expressed at low temperatures (less than 30 degrees C), was demonstrated in this study via heterologous expression of the Providencia rettgeri (P. rettgeri) pac gene in Escherichia coli (E. coli). While it is possible to produce PAC at a temperature as high as 37 degrees C, the environmental condition (specifically, culture pH) critically affected culture performance. Production of PAC at 37 degrees C was feasible only when culture pH was close to neutral (i.e., 6.5-7.5). Outside this pH range, cell physiology for the host/vector system was seriously affected, resulting in poor culture performance. In acidic culture environments, temperature significantly affected the pac expression level and specific PAC activity decreased with an increase in culture temperature. In basic culture environments, cell growth was seriously inhibited though the pac expression level was minimally affected by temperature. Such unusual types of pH and temperature effects on pac expression were never reported for bacterial PACs. The results suggest that culture pH should be precisely controlled for the current host/vector systems being applied on the overproduction of P. rettgeri PAC in E. coli at high temperatures.  相似文献

Maternal nutrition can strongly influence embryo development and offspring fitness. The environmental matching hypothesis posits that developmental conditions affect offspring in ways that enable them to appropriately deal with similar post‐developmental conditions, although mismatches between developmental and post‐developmental environments will reduce fitness. To test this hypothesis, reproductive lizards (Anolis sagrei) were reared in environments with high versus low prey availability. The resultant offspring were then reared reciprocally under the same two prey conditions that their mothers experienced. High levels of prey available to mothers increased egg production, although the survival of eggs was low compared to those produced by mothers in the low‐prey treatment. Low prey availability to offspring reduced growth, regardless of the amount of prey available to their mothers. Low prey availability also compromised offspring survival, although this negative effect was only present when mothers experienced high‐prey conditions, whereas matching of low‐prey conditions in maternal and offspring stages resulted in high survival. However, because the mismatch of low maternal and high offspring prey availability resulted in similar offspring survival to the matched treatments, our results do not fully support the environmental matching hypothesis. The effect of haemin and related porphyrins on the activity of the trypsin-like enzyme activity in cell sonicates of Porphyromonas gingivalis was examined using a spectrophotometric assay and by following the degradation of human IgG. Haemin was inhibitory in both assay systems and the effect was shown to be reversible. The high concentration of haemin accumulated by P. gingivalis may protect the organism against autodegradative effects of the trypsin-like protease.  相似文献

The two TonB systems in Vibrio cholerae were found to have unique as well as common functions. Both systems can mediate transport of haemin and the siderophores vibriobactin and ferrichrome. However, TonB1 specifically mediates utilization of the siderophore schizokinen, whereas TonB2 is required for utilization of enterobactin by V. cholerae. Although either TonB system was sufficient for the use of haemin as an iron source, in vitro competition between TonB1 and TonB2 system mutants indicates a preferential role for TonB1 in haemin utilization. This was most pronounced in conditions of high osmolarity, in which TonB1 system mutants were unable to grow with haemin as the sole iron source. Sequence analysis predicted that the two TonB proteins differ in both amino acid sequence and protein size. An internal deletion in TonB1 was constructed in order to generate a protein of approximately the same size as TonB2. A strain expressing the TonB1 deletion protein, and no other TonB, used haemin as the iron source in low-osmolarity medium, but could not use haemin in high osmolarity. This is the same phenotype as a strain expressing only TonB2 and suggests that TonB1, but not TonB2, can span the increased periplasmic space in high osmolarity and thus mediate haemin transport. Mouse colonization assays indicated a role for both TonB systems, and mutations in either system resulted in reduced ability to compete with the wild type in vivo.  相似文献

Many environments are undergoing rapid environmental change and there is a need to understand the mechanisms by which species can persist in altered environments. Model systems, such as amphibian metamorphosis, which can be generalized across many types of environmental change and across many species, are a powerful tool for understanding mechanisms that facilitate persistence in altered and disturbed environments. Amphibian larvae respond to environmental change by varying age at metamorphosis, or size at metamorphosis. Differential selection pressures on age or size at metamorphosis may result in a differential response among taxa to environmental change. Using a meta‐analysis, we investigated whether age at metamorphosis, size at metamorphosis, and larval growth rate vary within and among taxonomic families of amphibians in experiments that modified the environmental temperature, density of individuals, food, hydroperiod and the presence of predators. For all environmental factors except predators, the direction of the response was consistent across most of the studied taxa. However, there was considerable variation in effect size both within and among families. Results demonstrate that amphibian metamorphosis is a valuable model system for studying the effects of environmental change. Yet, we stress the need for caution in making generalizations about how individuals respond to environmental factors that have an indirect effect on physiology and require the perception of an environmental cue, such as the presence of predators. Synthesis As the current conditions of the environment are rapidly changing there is a need to understand how organisms respond to environmental change, and whether response of one species can be generalized to other species. Using a meta‐analyses, we tested whether the phenotypic response of amphibian larvae to five types of environmental change is consistent among and within taxonomic families. The phenotypic response to changes in environmental factors was consistent when the environmental factor has a direct effect on physiology, but varies among and within family if the environmental factor has an indirect effect on physiology or requires the perception of an environmental cue.  相似文献
