Selected behavior of Empoasca fabae (Harris) (Homoptera: Cicadellidae) was examined to elucidate resistance of commercially‐available glandular‐haired alfalfa to this key forage pest. The overall objective was to assess the effects of the glandular trichomes on the behavior of nymphs and adults. Studies of host‐plant acceptance by E. fabae nymphs found first and third instars to show a higher degree of change in settling location on the glandular‐haired FGplh13 alfalfa than on the nonglandular P5373 alfalfa. Nymphs also cleaned their tarsi more frequently when in contact with the glandular trichomes on FGplh13 alfalfa, and in addition a larger number of nymphs jumped/fell off the surface of FGplh13 alfalfa. The glandular trichomes on FGplh13 also impeded nymphal mobility more effectively than the nonglandular trichomes on P5373. No choice, time‐course analysis of adult host‐plant acceptance behavior found that adults settled less frequently on FGplh13 alfalfa with the glandular trichomes intact, than on either FGplh13 with the glandular trichomes removed, or P5373 alfalfa with the nonglandular trichomes intact or removed. Free choice, time‐course analysis of adult host‐preference behavior determined that at each observation, stems of the nonglandular P5373 alfalfa were preferred over FGplh13 alfalfa. Similarly, at each observation, stems of FGplh13 alfalfa with the glandular trichomes removed were preferred over FGplh13 with the glandular trichomes intact. These data provide additional evidence for the localization of a resistance factor in the glandular trichomes of FGplh13 alfalfa. An antixenotic resistance mechanism also appears to be present, which may function, in part, through a tactile avenue.  相似文献

Experiments were performed to elucidate resistance of glandular-haired alfalfa, Medicago sativa L., to the potato leafhopper, Empoasca fabae (Harris). The primary objective was to examine the potential role of the glandular trichomes and stem lignification for imparting resistance to this pest. During free-choice foliar discoloration experiments, the resistant alfalfa clone FGplh13 expressed lower levels of injury than the susceptible P5373 after 13 d. No-choice tests performed using nymphal potato leafhoppers showed higher levels of mortality associated with the resistant glandular-haired clone FGplh13, and no nymphs survived after 48 h. Additional no-choice experiments using FGplh13 and P5373 alfalfa with the glandular and nonglandular trichomes intact or removed show that mortality of nymphs and adults decreased after removal of the glandular trichomes from FGplh13. Nymphal and adult mortality and nymphal development time did not differ on FGplh13 with the trichomes removed and P5373 with the trichomes intact or removed. Firstinstar, and possibly second-instar, potato leafhoppers were entrapped by an exudate produced by the glandular trichomes on the FGplh13 alfalfa. Stem lignification and the number of vascular bundles did not differ between FGplh13 and P5373 alfalfas. The glandular trichomes on FGplh13 alfalfa appeared to provide the major host resistance factor, with resistance to adults being chemically based and resistance to nymphs being chemically and mechanically based. To maintain levels of potato leafhopper resistance, breeders would appear to benefit by continuing to select for the expression of the glandular trichome phenotype.  相似文献

New cultivars of alfalfa, Medicago sativa L., have been released with glandular trichomes for resistance to potato leafhopper, Empoasca fabae (Harris). Yet, the impact of the glandular trichomes on the primary natural enemy of the leafhopper, Anagrus nigriventris Girault, is unknown. We compared the host searching behavior of the egg parasitoid on four alfalfa clones varying in trichome characters. Female wasps were videotaped on Ranger, a susceptible clone with relatively sparse trichomes, B14, a resistant clone with dense but nonglandular trichomes, and FG12 and FG18, two resistant clones with glandular trichomes. Although the number of leafhopper eggs per stem exposed to wasps did not significantly differ among the four clones, the frequency of foraging and total foraging time were less on the two clones with glandular trichomes than on the two clones with nonglandular trichomes. In addition, an analysis of covariance demonstrated that, although the number of ovipositional probes increased with egg density on a stem, the number of probes on stems with glandular trichomes was significantly less than that on stems without glandular trichomes. The allocation of time by wasps among drumming, probing, and grooming behaviors was similar among the clones. Wasps tended to fly off of clones with glandular trichomes more often than off of clones with nonglandular trichomes. This study suggests that cultivars with glandular trichomes may interfere with host searching by A. nigriventris.  相似文献

The structures on leaf surfaces, e.g. trichomes, can act as effective antiherbivory mechanisms as chemical repellents. Structural defences usually represent constitutive resistance, but there are also a few cases of inducible morphological defences. We tested whether defoliation may induce changes in trichome production in white birch (Betula pubescens). The studied birches were either 0, 50 or 100% defoliated during the previous or current summer, and we measured the alterations in the production of glandular vs. nonglandular leaf trichomes, developmental instability (fluctuating asymmetry, FA) and leaf and shoot growth. We detected a clear shift from glandular to nonglandular leaf trichomes following previous‐year defoliation but not after current‐year defoliation. Furthermore, the density of nonglandular trichomes around the mid‐vein of leaves increased following previous‐year defoliation but decreased after current‐year defoliation. While leaf and shoot growth showed a distinct decrease in response to defoliation, FA turned out to be less sensitive. Consequently, previous‐year defoliation can induce the production of nonglandular trichomes in birch leaves. Because this response was accompanied by a reduction in glandular trichomes, the present results may suggest a trade‐off between the different trichome types of birch leaves.  相似文献

Eight proprietary genotypes of glandular-haired alfalfa, Medicago sativa L., supplied by two different companies, were compared for the degree and types of resistance to the potato leafhopper, Ernpoasca fabae (Harris), and hopperburn. A tube cage no-choice bioassay was developed to test leafhopper mortality, feeding, settling preferences, severity of hopperburn symptoms (in this case, defined as both yellowing and stem growth reduction), and trichome density and type on feeding sites. Leafhopper mortality was both strongly and significantly associated with feeding and leaf trichome density; decreased hopperburn symptom severity was weakly, although significantly, associated with increased mortality. To quantify hopperburn in terms of both yellowing and stem growth reduction, we developed a ranking system that reduces overall hopperburn expression to a single number that considers the varying responses to both types of symptoms. Great variability in leafhopper settling, leafhopper mortality, and stem glandular trichome density was detected among alfalfa genotypes, suggesting that genotypic differences may be based on the concentration and/or chemical constituency of the trichome exudates. We postulate that, among variably resistant genotypes of glandular-haired alfalfa, differences among leafhopper responses and hopperburn severity are linked to forced movement from the stems to the leaves as refuge feeding sites. Principal component analysis was performed to reduce the 10 variables down to five biologically significant factors. Scores for these factors were then used to develop resistance indices for potato leafhopper resistance, hopperburn resistance, and an overall glandular-haired alfalfa resistance index.  相似文献

The structure, ontogeny, classification and organographic distribution of trichomes in 9 genera and 12 species of the Cucurbitaceae are presented. The vegetative and floral trichomes are classified into glandular and non-glandular. In all 13 main types and 26 subtypes of glandular and nonglandular trichomes are observed. All the types of trichomes originate from a single papillate hair initial. The trichomes are classified on the basis of form, structure and contents.  相似文献

Trichoderma species are usually considered soil organisms that colonize plant roots, sometimes forming a symbiotic relationship. Recent studies demonstrate that Trichoderma species are also capable of colonizing the above ground tissues of Theobroma cacao (cacao) in what has been characterized as an endophytic relationship. Trichoderma species can be re-isolated from surface sterilized cacao stem tissue, including the bark and xylem, the apical meristem, and to a lesser degree from leaves. SEM analysis of cacao stems colonized by strains of four Trichoderma species (Trichoderma ovalisporum-DIS 70a, Trichoderma hamatum-DIS 219b, Trichoderma koningiopsis-DIS 172ai, or Trichoderma harzianum-DIS 219f) showed a preference for surface colonization of glandular trichomes versus non-glandular trichomes. The Trichoderma strains colonized the glandular trichome tips and formed swellings resembling appresoria. Hyphae were observed emerging from the glandular trichomes on surface sterilized stems from cacao seedlings that had been inoculated with each of the four Trichoderma strains. Fungal hyphae were observed under the microscope emerging from the trichomes as soon as 6 h after their isolation from surface sterilized cacao seedling stems. Hyphae were also observed, in some cases, emerging from stalk cells opposite the trichome head. Repeated single trichome/hyphae isolations verified that the emerging hyphae were the Trichoderma strains with which the cacao seedlings had been inoculated. Strains of four Trichoderma species were able to enter glandular trichomes during the colonization of cacao stems where they survived surface sterilization and could be re-isolated. The penetration of cacao trichomes may provide the entry point for Trichoderma species into the cacao stem allowing systemic colonization of this tissue.  相似文献

Summary Trichomes of Tremandra R.Br. ex DC., Platytheca Steetz and Tetratheca Sm. (Elaeocarpaceae, former Tremandraceae), together with two outgroup species of Elaeocarpus L., are illustrated using scanning electron microscopy, and their distribution on various plant organs is documented. Various trichomes types were identified that relate taxa: simple hairs, stellate hairs, short glandular trichomes, long glandular trichomes, and three forms of tubercules. Both outgroup and ingroup taxa have simple hairs. Stellate hairs are confirmed as unique to Tremandra. Prominent and sculptured multicelled tubercules, some bearing a stout hair, are characteristic of Platytheca. Smaller multicelled tubercules occur in both Platytheca and Tetratheca, except for the Western Australian taxon Te. filiformis Benth. (possibly plesiomorphic). Unicellular tubercules (papilla) characterise two species of Tetratheca. Short glandular trichomes, usually found on the ovary, also occur in both of these genera but not in all species (possibly secondary losses), while long glandular trichomes, usually on stems and leaves, occur only in some groups of Tetratheca. Within Tetratheca, Western Australian taxa that have five-merous flowers fall into three ‘groups’: seven species (together with one from South Australia) that have short glandular trichomes but no long glandular trichomes; six species that have long glandular trichomes but no short glandular trichomes; and four species that have both trichome types. All other species of Tetratheca have four-merous flowers and form two ‘groups’: 12 eastern species (including one from South Australia) that have both short glandular trichomes and long glandular trichomes; 4 western species and six eastern species that lack short glandular trichomes. On the basis of these characters, a phylogenetic hypothesis for the three genera is presented.  相似文献

Laboratory experiments were conducted to characterize the trichome-based defense of glandular-haired alfalfa, Medicago sativa L., against the potato leafhopper, Empoasca fabae (Harris). Within-plant variability in leafhopper resistance was examined by caging adult leafhoppers to either basal or apical stem internodes of the leafhopper-resistant, glandular-haired M. sativa genotype G98A or the susceptible, nonglandular-haired M. sativa 'Ranger'. Young, actively secreting glandular trichomes are located on apical internodes of G98A, whereas senesced gland heads are found on older, basal internodes of G98A. After 96 h, the highest cumulative leafhopper mortality and lowest number of excretory droplets were associated with apical internodes of G98A. No difference was detected in mortality and feeding levels among insects caged to basal internodes of G98A and basal and apical internodes of Ranger. The influence of abiotic factors on leafhopper resistance was evaluated by caging adult leafhoppers to either G98A or Ranger under four combinations of low and high light (250 and 1,000 micromol s(-1) m(-2)) and temperature regimes (17 and 30 degrees C). After 96 h, the highest cumulative mortality was associated with leafhoppers confined to G98A under high light and high temperature conditions. Temperature level and plant type also had an effect on the production of excretory droplets, resulting in the highest number of excretory droplets being associated with Ranger under the high temperature regime. These results indicate that certain regions of M. sativa G98A are better protected against the potato leafhopper than others and that temperature influences resistance levels of glandular-haired alfalfa.  相似文献

Abstract The green peach aphid ( Myzus persicae ) is becoming resistant to many of the pesticides used in the tomato ( Lycopersicon esculentum ) industry. A potential alternative method of control is host-plant resistance, which may be mediated by trichomes. Resistance to the green peach aphid was assessed for six accessions of L. hirsutum, three of L. pennellii and a L. esculentum control. Mortality, entrapment and number of aphids unaffected by trichomes were recorded 24, 48 and 72 h after placement. Using multiple regression against counts of glandular and non-glandular trichomes, the role of foliar pubescence in resistance to the aphid was established. Lycopersicon pennellii possessed the highest level of resistance after 72 h and type IV trichome density accounted for 26.5% of variance in mortality ( P  < 0.001). Low densities of type IV and high densities of type VII trichomes increased the number of aphids 'unaffected' by trichomes, a relationship accounting for 29% of variance ( P  < 0.001). Information on which types of trichomes most strongly affect resistance to insect pests will aid future breeding and L. pennellii shows potential for use in breeding programs to increase levels of host-plant resistance in L. esculentum.  相似文献

The leaf epidermis of 14 species ofCodonanthe and 10 species of Nematanthus has been examined. Species of Codonanthe section Codonanthe are geographically restricted to south-eastern Brazil, and are diploid. They possess multicellular-uniseriate nonglandular trichomes, glandular trichomes with a four-celled head and a short body, anisocytic stomata and lack extrafloral nectaries. Species of Codonanthe section Spathuliformae and Codonanthe subgenus Codonanthella are distributed from southern Mexico through Central America to north-western South America and are tetraploid. They possess unicellular non-glandular trichomes (except C. caribaea), glandular trichomes with a two-celled head (except C. caribaea) and a short body, anisocytic stomata and extrafloral nectaries (except C. caribaea). All Nematanthus species are distributed in south-eastern Brazil and are diploid (N=8). Six species of Nematanthus consistently have multicellular-uniseriate nonglandular trichomes, glandular trichomes with a four-celled head and a short (unicellular) or long (multicellular) body, anisocytic stomata and lack extrafloral nectaries. Four species of Nematanthus have multicellular-uniseriate non-glandular trichomes, glandular trichomes with a head of more than four cells and a short body, anisocytic and helicocytic stomata and lack extrafloral nectaries.  相似文献

This study demonstrated how simple, lanate trichomes on the stem of Anaphalis margaritacea (D.C.) inhibit feeding by young nymphs of Philaenus spumarius (L.). Nymphs were caged on stems which were subsequently sectioned and examined for feeding marks. The lack of stylet punctures at sites where nymphs were unable to feed indicated that trichomes prevented the start of stylet penetration rather than interfering with the progress of the stylets after penetration had begun. Trichome resistance declined progressively with spittlebug instar from complete resistance to first instar nymphs through complete absence of resistance to fifth instar nymphs. Discriminant analysis of the effect of number of trichomes per mm2, vertical density, and height on trichome resistance to third instar nymphs revealed that trichome height was the critical factor in resistance. Variation in the percent of nymphs in each instar able to feed when caged on the stem was (1) positively correlated with degree to which the average rostrum length of the instar exceeded the trichome height, (2) independent of trichome density, which was uniformly high on all stems. Trichomes prevented stylet penetration where trichome height exceeded the length of the nymph's rostrum, probably because the necessary stimuli were not received when the sensilla on the tip of the rostrum could not touch the surface of the stem. A similar mechanism may explain resistance of simple trichomes to crop pests in the Cicadellidae.
Résumé Ce travail montre comment des trichomes simples et laineux sur les tiges d'Anaphalis margaritacea (D.C.) inhibent la prise d'aliments par les jeunes larves de Philaenus spumarius L. Les larves ont été encagées sur des tiges qui ont été ultérieurement coupées et les piqûres alimentaires dénombrées. L'absence de piqûres par les stylets aux lieux où les larves ont été incapables de s'alimenter a montré que les trichomes ont empêché le début de la pénétration des stylets plus qu'ils n'ont entravé leur progression après le début de la pénétration. La résistance des trichomes diminue progressivement: totale pour le premier stade larvaire, elle est devenue nulle au cinquième stade. Une analyse discriminante des effets de la densité des trichome par mm2, de la densité verticale, de leur hauteur sur la résistance des trichomes aux larves de 3ème stade, a révélé que la hauteur du trichome est le facteur de résistance. La variation dans le pourcentage de larves de chaque stade encagées sur la tige, capables de consommer, présentait une relation positive avec le degré par lequel la longueur du rostre dépasse la hauteur du trichome, quelle que soit la densité des trichomes qui est toujours élevée sur toutes les tiges. Les trichomes ont empêché la pénétration des stylets quand leur hauteur a dépassé la longueur du rostre, probablement parce que les stimuli nécessaires n'ont pas été reçus quant l'extrémité du rostre ne pouvait pas atteindre la surface de la tige. Un mécanisme semblable peut expliquer la résistance de trichomes simples aux Cicadellidae.

Resource availability and the trichome defenses of tomato plants   总被引:10,自引:0,他引:10  
We conducted two experiments to determine how resource availability influenced allocation by tomato (Lycopersicon esculentum) to trichomes, and how different patterns of trichome allocation by plants grown in different resource environments might then influence the behavior of tobacco hornworm (Manduca sexta) caterpillars. In the first experiment we used high and low levels of light and water, and then, using scanning electron microscopy, determined trichome densities on the leaves and stems. We sampled leaves and stems at several places throughout the plant to determine whether there were within-plant differences in allocation to trichomes. The results of the first experiment showed that resource availability influenced allocation to trichome growth. Patterns in high and low-light supported both the growth-differentiation balance hypothesis (GDBH) and the carbon-nutrient balance hypothesis (CNBH). However, the GDBH was not supported by differences among water treatments. Contrary, to predictions of the GDBH, plants with intermediate growth did not have the highest trichome densities, and plants with similar growth differed in trichome density. Possible biological and artifactual explanations are discussed. The first experiment also showed that there was within-plant variation in allocation to trichomes, and that plant resource availability may influence within-plant variation in allocation to trichomes. In the second experiment, we grew plants in high and low-light, and then monitored the behavior of tobacco hornworms on the stems of these plants in the laboratory. This experiment demonstrated that the light environment that tomato plants were grown in influenced the resting behavior of caterpillars. Furthermore, it demonstrated that both glandular and non-glandular trichomes impeded caterpillars from searching for food. Overall, this study indicated that plant resource availability can influence allocation to trichome defenses, and that these differences may affect insect herbivores.  相似文献

Glandular trichomes on foliage of the wild potato species, Solanum berthaultii Hawkes, deter oviposition by the potato tuber moth (PTM), Phthorimaea operculella Zeller and negatively affect other important performance parameters. Oviposition deterring factors are localized in the glandular trichomes of S. berthaultii. When mechanically transferred to foliage of a susceptible potato cultivar, trichome contents reduced egg laying by 97%. Removal of glandular trichomes from S. berthaultii foliage using a combination of chemical and mechanical procedures increased oviposition rates ca. 210-fold. Removal of trichomes also led to increased mobility of larvae on the leaf surface, more leaf feeding, shorter larval development and larger pupae. The resistance conferred by glandular trichomes of S. berthaultii provides an important genetic trait potentially useful for management of PTM.  相似文献

Trichomes and their distribution patterns have been observed in the aerial parts of plants of the genera Striga, Buchnera, Rhamphicarpa, Euphrasia, Melampyrum and Rhinanthus, using both light (LM) and scanning electron microscopy (SEM). All trichomes belong to the multicellular, uniseriate category and can be classified as glandular and nonglandular types. On nearly all organs, the two types can be observed side by side. Apart from a large group of ubiquitous trichomes, some characteristic trichome types are reported; they are restricted either to chlorophyllous organs (foliage leaves, bracts, calyx and stem) or to the corolla. In some nonglandular trichomes mineral deposits have been found. Other morphological and anatomical trichome features are discussed with regard to their possible function.  相似文献

The costs and benefits of defensive traits in plants can have an ecological component that arises from the effect of defenses on the natural enemies of herbivores. We tested if glandular trichomes in Datura wrightii, a trait that confers resistance to several species of herbivorous insects, impose an ecological cost by decreasing rates of predation by the natural enemies of herbivores. For two common herbivores of D. wrightii, Lema daturaphila and Tupiocoris notatus, several generalized species of natural enemies exhibited lower rates of predation on glandular compared to non-glandular plants. Lower rates of predation were associated with reductions in the residence time and foraging efficiency of natural enemies on plants with glandular trichomes, but not with direct toxic effects of glandular exudate. Our results suggest that the benefit of resistance to herbivores conferred by glandular trichomes might be offset by the detrimental effect of this trait on the natural enemies of herbivores, and that the fitness consequences of this trichome defense might depend on the composition and abundance of the natural-enemy community.  相似文献

Pubescence characteristics for six accessions of Lycopersicon hirsutum Dunal and five accessions of L. hirsutum f. glabratum CH Mull. were determined and compared with those of an accession of cultivated tomato (L. esculentum Mill.). Removal of trichome exudates from excised leaflets using ethanol solution resulted in a reduced mortality and increased survival of potato moth (Phthorimaea operculella (Zeller)) neonates for the accessions that were most lethal when not treated with ethanol solution. No such treatment effect was evident for L. esculentum or for the L. hirsutum accession with least effect on neonates when its trichomes were intact. In a glasshouse experiment with caged intact plants, mortality of neonate P. operculella placed on the abaxial surface was greater on seven accessions than for L. esculentum.Neonates were less severely affected on the adaxial surface. Eleven days after inoculation, no live larvae were found on LA 1927, PI 127827, PI 134418, and PI 134428, and numbers on other accessions were lower than for L. esculentum. Eventual emergence of adults followed a similar trend. Multiple regression of insect data against pubescence indicated a significant correlation between density of type IV and VI trichomes and neonate mortality, decreased larval development and decreased adult emergence. Non-glandular type V trichomes were positively correlated with high survival of insects to 11 days and to adult. Though factors other than glandular trichomes are likely to be important, increased density of type IV and VI, along with reduced type V, are shown to be important to select in breeding for P. operculella resistance.  相似文献

The leaves of Artemisia nova Nelson are covered with a dense layer of nonglandular trichomes that obstruct the observation of glandular trichomes on the leaf surfaces. An acid-staining technique was developed that eliminates interference from the nonglandular trichomes and stains the glandular ones red, allowing them to be seen under a light microscope. Using this method it was found that these glands develop on both leaf surfaces, except over veins, and cover 21–35% of the leaf surface area at densities of 50–70 per mm2. Mature glands are sac-like structure with a thin sheath or cuticle surrounding a clear transparent fluid. This fluid contains all of the sesquiterpene lactones present in the leaves, and some of the monoterpenes. These terpenoids were responsible for the red color of the glands after acid treatment.  相似文献

A method based on the laser microdissection pressure catapulting technique has been developed for isolation of whole intact cells. Using a modified tissue preparation method, one outer pair of apical cells and two pairs of sub-apical, chloroplast-containing cells, were isolated from glandular secretory trichomes of Artemisia annua. A. annua is the source of the widely used antimalarial drug artemisinin. The biosynthesis of artemisinin has been proposed to be located to the glandular trichomes. The first committed steps in the conversion of FPP to artemisinin are conducted by amorpha-4,11-diene synthase, amorpha-4,11-diene hydroxylase, a cytochrome P450 monooxygenase (CYP71AV1) and artemisinic aldehyde Δ11(13) reductase. The expression of the three biosynthetic enzymes in the different cell types has been studied. In addition, the expression of farnesyldiphosphate synthase producing the precursor of artemisinin has been investigated. Our experiments showed expression of farnesyldiphosphate synthase in apical and sub-apical cells as well as in mesophyl cells while the three enzymes involved in artemisinin biosynthesis were expressed only in the apical cells. Elongation factor 1α was used as control and it was expressed in all cell types. We conclude that artemisinin biosynthesis is taking place in the two outer apical cells while the two pairs of chloroplast-containing cells have other functions in the overall metabolism of glandular trichomes.  相似文献
