Phosphorus (P) fractions were quantified in water samples collected on four occasions from sites at the lower tidal limit of seven Scottish East Coast rivers. Individual catchment characteristics ranged from those dominated by semi-natural land use to those where agriculture predominated. Together the rivers displayed attributes ranging from nearly pristine to those impacted by point and diffuse sources of pollutants. Sampling times were chosen to coincide with periods of low river flows where conditions should result in low concentrations of suspended particulate matter (SPM) but favourable for phytoplankton growth. Total phosphorus (TP) concentrations were < 0.004 mg l–1, 0.005–0.048 mg l–1 and 0.28–2.2 mg l–1 for pristine, agricultural and point source impacted rivers respectively. Soluble reactive phosphorus (SRP) represented from < 5% to > 90% of TP and dissolved P dominated all samples. The total phosphorus content (TPC) of SMP ranged from 0.1 to 1.1% and was significantly related to SRP. Organic matter was a significant component of SPM and organically bound phosphorus was the dominant form of particulate P. The C/P ratio of organic matter was wide, between 500–1200 for the more pristine systems which narrowed to < 400 for heavily impacted rivers. Exchangeable P increased during the summer but was generally a minor component of TP and therefore likely to be a significant source of SRP only in pristine rivers. Phytoplankton constituted between 5 and 46% of organic matter and concentrations of chlorophyll-a were significantly correlated with both TP and SRP.  相似文献

ABSTRACT

EDTA is useful to assess mobile metal pools in polluted soils and sediments. There is a need to enhance our understanding of the significance of metal fractions released. The impact of single reagent extraction with 0.05 mol L?1 EDTA on the solid phase distribution of trace metals in surface soils sampled from confined dredged sediment disposal sites was investigated. Not extracted and EDTA extracted soils were subjected to sequential extraction to fractionate the total contents into: (1) easily exchangeable and carbonate bound fraction; (2) reducible fraction; (3) oxidisable fraction; and (4) residual fraction. With EDTA, significant portions of metals associated with the acid extractable and reducible fractions were released. The oxidisable and residual fractions remained unaffected for most of the investigated metals except for the organic matter associated metals (Cu and Pb). A decrease in the residual fraction after EDTA-extraction for Cu and Pb was attributed to artifacts of the sequential extraction procedure.  相似文献

A method was developed to evaluate the cumulative effect of wetland mosaics in the landscape on stream water quality and quantity in the nine-county region surrounding Minneapolis—St. Paul, Minnesota. A Geographic Information System (GIS) was used to record and measure 33 watershed variables derived from historical aerial photos. These watershed variables were then reduced to eight principal components which explained 86% of the variance. Relationships between stream water quality variables and the three wetland-related principal components were explored through stepwise multiple regression analysis. The proximity of wetlands to the sampling station was related to principal component two, which was associated with decreased annual concentrations of inorganic suspended solids, fecal coliform, nitrates, specific conductivity, flow-weighted NH4 flow-weighted total P, and a decreased proportion of phosphorus in dissolved form(p < 0.05). Wetland extent was related to decreased specific conductivity, chloride, and lead concentrations. The wetland-related principal components were also associated with the seasonal export of organic matter, organic nitrogen, and orthophosphate. Relationships between water quality and wetlands components were different for time-weighted averages as compared to flow-weighted averages. This suggests that wetlands were more effective in removing suspended solids, total phosphorus, and ammonia during high flow periods but were more effective in removing nitrates during low flow periods.  相似文献

A survey of 11 sites covering three large (>10 km long, 6 sites) playa lakes and four pans (<1 km2, 5 sites) of the Yarra Yarra salt lake system in the Northern Agricultural Region of Western Australia commenced in 2001. These salt lakes are shallow and ephemeral, with inundation being more regular following winter rainfall, but summer inundation also occurred in 2001. Salinity was generally higher in playas (156–368 g l−1) than pans (30–284 g l−1), but salinity responded noticeably to heavy rainfall events, especially in pans. pH values in the playa lakes (6.68–7.82) were less variable than in the pans (6.81–8.08). The range of dissolved oxygen concentrations was greater in pans (3.7–14.4 mg l−1) than in playas (3.9–8.2 mg l−1). Cationic concentrations generally followed the pattern of sea water cation dominance. Benthic microbial communities comprised either cohesive to loosely mucilaginous mats, or thin films of diatoms. The speciation of cadmium, lead, copper, zinc, manganese and iron into exchangeable, carbonate, reducible and organic bound fractions was studied in sediments from coastal and freshwater environments in Ghana. This was relevant as the species in which metals are stored within specific sediment components is important in determining their impact on the environment. For both coastal and inland sediments, a higher percentage of cadmium was associated with the more available exchangeable and carbonate fractions, while iron, zinc and manganese were mainly associated with the reducible and organic fractions. Lead and copper were found to have the greatest ability to form different species in the samples examined and were more evenly associated with all the fractions. The metals generally showed more ability to form different species in inland freshwaters than in coastal relatively saline waters. However, differences between inland and coastal waters were based more on whether the environments were oxidising or reducing than on whether they were fresh or saline. The metals may be divided into three groups of high mobility consisting of lead and copper; moderate mobility made up of cadmium, manganese and zinc; and low mobility, represented by iron.  相似文献

Chronic exposure to morphine in mice produced an increase in Ca2+ content of synaptosomes, synaptic plasma membranes (SPM), and synaptic vesicles. Ca2+ binding capacity was significantly reduced in tolerant SPM fractions. Naloxone significantly reversed the increased calcium content and reduced binding capacity of SPM when administered to 72-h-treated mice. Scatchard analysis of binding curves reveals three distinct classes of Ca2+ binding sites. During tolerance, the high- and low-affinity sites exhibit a reduced capacity to bind calcium, which may be reversed by in vivo and in vitro administration of naloxone. The increase in SPM and synaptic vesicle calcium content may reflect adaptive changes in the cell membrane during tolerance development, which may contribute to changes in neurotransmitter and second messenger function.  相似文献

Abstract: Specific binding of tritiated dopamine, spiperone, and N-propylnorapomorphine was examined in subcellular fractions from bovine caudate nucleus. All fractions contained at least two sets of specific binding sites for [3H]spiperone (KD 1aPP= 0.2 nM, KD 2aPP= 2.2 nM), the higher affinity sites accounting for one-third to one-eighth of the total. [3H]Spiperone binding was slightly enriched over the total particulate fraction in P2, P3, SPM, and a crude fraction of synaptic mitochondria. A microsomal subfraction (P3B2) exhibited the highest specific binding capacity obtained, representing a fourfold enrichment over the total particulate fraction. [3H]Dopamine exhibited apparent binding to a single class of high-affinity sites in all fractions examined (KDaPP= 4.0 nM). A greater than twofold enrichment was observed in all fractions except myelin and P3, with a fivefold enrichment in SPM and P3B2. At least two classes of receptors were labeled by [3H]-N-propylnorapomorphine (KD 1aPP= 0.55 nM, KD 2aPP= 20 nM), using 50 nM-spiperone together with 100 nM-dopamine to define nonspecific binding. Although binding to the higher affinity site was displaced by spiperone, and lower affinity binding by dopamine, comparison of receptor densities with values obtained by using [3H]spiperone and [3H]dopamine directly suggested that [3H]-N-propylnorapomorphine labeled additional sites. We have also examined a postsynaptic membrane (PSM) fraction obtained from SPM by successive extraction with salt and EGTA followed by sonication and separation on a density gradient. [3H]Spiperone binding in PSM was enriched two- to threefold over unfractionated SPM with a concomitant decrease in [3H]dopamine binding. The enrichment in spiperone receptors was almost entirely due to an increase in the number of lower affinity binding sites, suggesting that these sites may be associated with the postsynaptic membrane.  相似文献

The differences in radiocaesium uptake between species were analysed in a series of solution culture and pot trials. Since radiocaesium uptake is very sensitive to the solution potassium (K) concentration, it was hypothesised that species depleting K in the rhizosphere to a larger extent, will have a higher radiocaesium uptake. Five species (bean, lettuce, winter barley, ryegrass and bentgrass) were grown for 18–21 days in nutrient solution spiked with 137Cs and at 4 K concentrations between 0.025 and 1.0 mM. Shoot 137Cs activities all decreased between 17- and 81-fold with increasing K supply. Shoot 137Cs activities were 4-fold different between species at the lowest K supply and 3.4-fold different at high K supply. The same five species were grown in two 134Cs spiked soils with contrasting exchangeable K but similar clay content. Shoot 134Cs activities were up to 19-fold higher in the soil with lowest exchangeable K. Differences in shoot activity concentrations between the species were only 4.5-fold in the high K soil, but were 15-fold in the low K soil. Bulk soil solution 134Cs and K concentration data were combined with radiocaesium uptake characteristics measured in solution culture to predict radiocaesium uptake from soil. Predictions were within 1.6-fold of observations in the high K soil but largely underestimated 134Cs uptake in lettuce, ryegrass and barley in the low K soil. A solute transport model was used to estimate K and radiocaesium concentrations in the rhizosphere. These calculations confirmed the assumption that higher radiocaesium uptake is found for species that deplete K in the rhizosphere to a larger extent.  相似文献

In this study, total phosphorus (P) concentrations and P fractions in the bed sediments of the Anllóns River (NW Spain) have been examined. The Lowest Effect Level of the Ontario Sediment Quality Guidelines was trespassed for total phosphorus (600 mg kg?1) at three out of the five sampling sites which, in addition, showed an average of 43% of bioavailable phosphate. The inorganic P fractions (loosely bound P, redox-sensitive P, metal oxide-bound P, carbonate and apatite P, and residual P) showed that inorganic P was mostly found as metal oxide bound P. The difference between total phosphorus and the sum of the inorganic P forms determined in these extracts is attributed to P associated to organic compounds being solubilized in some of the extractants, and represented in average 58% of total phosphorus. With the exception of loosely bound P, all the P fractions showed significant positive correlations with each other and with total phosphorus.  相似文献

Measurements of sedimentation were combined with water samples to calculate settling velocity of suspended particulate matter (SPM) in lakes. The study sites were open water stations and enclosures in Lake Erken (Sweden) and Lake Balaton (Hungary). Settling velocities were found to vary considerably both inside and outside the enclosures. Within sites, the differences between 25th and 75th percentiles of measured settling velocities of SPM were two‐ or three‐fold. Median settling velocities of SPM ranged from around 0.5 m/d in the enclosures of Lake Erken to more than 8 m/d in the open water of Lake Balaton. Special relevance was attributed to flocculation, which is known to be affected by, e.g., SPM concentration and turbulence. Even though not directly measured, the less turbulent environment inside the enclosures was suggested to explain the low settling velocity compared to the open water environment. Settling velocity apparently correlated with water current speed (r2 = 0.66; n = 12). Stepwise multiple linear regressions were used to relate the variability in settling velocity of SPM to the variability of possible controlling factors in a number of data subsets. In most cases, one variable describing the total amount of settling material (e.g., SPM) and one variable reflecting the composition of settling material (e.g., total phosphorus) were chosen. The use of suspended solids concentration to predict settling velocity in mass balance models was discussed. It was found that the mean slope between SPM and vSPM was close to 0.1 (m4d–1 g–1). Sediments from Haihe River mainstream, located in Xingjiaquan, Zhangjiazui, Tianjin, were collected and examined on the basis of P fractionation. SMT (standards, measurements and testing) procedure was adopted to investigate the changes in P concentration with depth in the core sediments collected from the different sampling sites of Haihe River mainstream. The relationships among different P fractions, such as exchangeable P (Ex-P), metal oxide bounded P (NaOH-P), organic matter and grain size, were also discussed. The results indicate that in both sites the rank order of P fractions was HCl-P > Organic P (OP) > NaOH-P > Ex-P in terms of their concentration. The Ex-P represented < 4% of the sediment total P, while the NaOH-P ranged 5–21%. The calcium bound phosphorus (HCl-P) showed considerable contribution (53–80%) to the sediment total P loads. Silt/clay sized sediments exhibited significantly higher concentrations of HCl-P and Ex-P in both sites. However, coarse-sand-sized sediments exhibited significantly higher concentrations of OP in both sites and NaOH-P in Xingjiaquan. The Huanghe River (Yellow River) had been the second largest river in the world in terms of sediment load to the sea; however, the river water discharge and sediment flux to the sea and their seasonal variability have been significantly altered by the dam activities and recent water–sediment regulation. These changes are believed to have important impacts on the flux of phosphorus that is generally transported in particulate form. In this article, the samples of suspended particulate matter (SPM) were collected at the Lijin Station during two high-discharge events in 2005 and were separated by particle size. Sequential extractions were applied to determine the forms of P in different particle size fractions and to assess the potential bio-availability of particulate phosphorus (PP). Based on the in-laboratory measurement, the impacts of different hydrological regimes on the source of PP and its bio-availability were also analyzed. The results indicate that exchangeable, organic, authigenic, and refractory P were preferentially associated with clay, very fine, and fine silt fractions. Detrital P was mainly associated with the medium and coarse silt fractions. Detrital P and authigenic P (two forms of calcium bound phosphorus) were the dominant fractions in all samples. Thus, the potential bio-available PP (exchangeable P and organic P) was mainly associated with the finer particles, such as clay. Higher content of exchangeable, organic, authigenic, and residual P and lower content of detrital P were found during the period of rainstorm compared to that of WSR. P forms and partitioning of P forms among different particle size fractions were assumed to depend on the sources of SPM. It is likely that the pathways and fates of PP forms were controlled by damming and by the related changes of hydrological regime. Therefore, anthropogenic changes of hydrological regime and particle size dominanted the amount and distribution pattern of bio-available P transportation to the estuary and the adjacent sea, which will have profound impacts on the marine ecosystems at the Huanghe River Estuary and even the Bohai Sea.  相似文献

The risk (including bioavailability and eco-toxicity) of heavy metals (Pb, Zn, Cu, Cd, Cr and Ni) in liquefaction residues (LR) of sewage sludge (SS) was estimated, according to both the speciation of heavy metals and the local environmental characteristics. The amount of organic matters in LR was lower than that in SS, resulting in a smaller calorific value, while the total content of heavy metals in LR nearly doubled. High residual rates of heavy metals (about 80%) indicated that the heavy metals in SS were concentrated into LR after liquefaction. The comparisons of sequential extraction results between SS and LR showed that after liquefaction, the mobile and easily available heavy metal fractions (acid soluble/exchangeable and reducible fractions) were mainly transformed into the relatively stable heavy metal fractions (oxidizable and residual fractions). The bioavailability and eco-toxicity of heavy metals in LR were relieved, though the total concentrations of heavy metals increased.  相似文献

A detailed record of suspended particulate matter (SPM) concentrations in the benthic boundary layer (BBL) 1.5 m above an accumulation bottom and 13.5 m below the surface was obtained from frequent (30 min interval) beam attenuation measurements made with a Sea Tech transmissometer in the main basin of Lake Erken, a moderately deep (mean depth 9 m, maximum depth 21 m) dimictic lake in central Sweden. Concentrations of SPM (g m–3) were not as strongly correlated to the beam attenuation coefficient (c, [m–1]), as were concentrations of the inorganic SPM fraction. Apparently, this was caused by large optically inactive organic particles which significantly affected the measurements of SPM, but had little effect on the attenuation of light.When the water column was thermally stratified, SPM concentrations in the BBL showed a seasonal increase which was related to an increase in the thermocline depth. As the epilimnion deepened, there was also a marked increase in the occurrence of rapid and large changes in SPM concentration. After the loss of stratification, the amount of SPM and the temporal variability in its concentration was reduced. Since surface waves could not influence sediment resuspension at the depth of measurement, these data show the importance of internal waves in promoting sediment resuspension in areas of sediment accumulation. The relatively short period in each summer, when the thermocline reaches a sufficient depth to allow for resuspension over accumulation bottoms, can have important consequences for both the redistribution of lake sediments and the internal loading of phosphorus.  相似文献

Abstract: Subcellular fractions from rat cerebellum and other tissues were examined for the presence of a 240K glycoprotein, designated GP-A. Previous results have shown that GP-A is enriched in cerebellum synaptic junction (SJ) fractions when compared to parent synaptic plasma membrane (SPM) fractions and is not detected in forebrain SPM or SJ fractions. In the present studies, GP-A was not detected in myelin, mitochondria, purified nuclei, or cytosolic fractions from cerebellum, but was present in microsomal fractions. GP-A is partially soluble in the non-ionic detergent Triton X-100 and is completely soluble when cerebellum SPMs are treated with the ionic detergent N-lauryl sarcosinate. The solubilization of GP-A from cerebellum membranes was shown to be a function of bound calcium ions, e.g., pretreating SPMs with 100 μM-1mM Ca2+ decreased the solubility of GP-A in Triton by approximately threefold. GP-A is a major concanavalin A (Con A)-binding glycoprotein in cerebellum SJ fractions and migrates on sodium dodecyl sulfate (SDS) gels with a slower relative mobility than the 235K/ 230K fodrin doublet. Comparisons between purified fodrin and the 235K/230K doublet in cerebellum and fore-brain synaptic fractions by two-dimensional peptide mapping indicated that they were identical. The Con A-binding property of GP-A was exploited to purify it by affinity chromatography with agarose-Con A. Peptide mapping comparisons between affinity-purified GP-A and GP-A in SPM and SJ fractions indicated that GP-A in synaptic fractions is apparently homogeneous. Peptide map comparisons between GP-A and 235K fodrin polypeptide indicated that these two synaptic components are highly related (50% of their respective peptides are shared). The 235K fodrin polypeptide in SJs reacted with anti-fodrin antisera on Western blots; however, GP-A failed to cross-react. These observations, together with results from previous studies, indicate that GP-A is highly enriched in cerebellum compared to other neuronal and nonneural tissues. Moreover, GP-A is enriched in SJs relative to SPM fractions, is related to fodrin, and is most likely a cell-surface glycoprotein at asymmetric synapses in cerebellum. GP-A may be involved in neuronal recognition or synaptic transmission in the cerebellum. The important role of calcium in synaptic transmission, together with the decreased solubility of GP-A in Triton that results from micromolar concentrations of calcium, suggest that GP-A may play a role in stabilizing cerebellar synaptic junctions.  相似文献

This article reports the results of a study focused on the presence and bioavailability of arsenic in agricultural soil in the mining and industrial regions of northern Kosovo and southern Serbia, as well as uptake and bioaccumulation of arsenic in two commonly cultivated plant species (Zea mays L. and Solanum tuberosum L.). This area was one of the most important mining districts in Europe. The collected soil samples were subjected to a modified BCR three-step sequential extraction procedure in order to investigate the chemical partitioning of arsenic in the soils. The general distribution of arsenic in various fractions was: exchangeable < reducible < oxidizable fractions. Highest concentrations of total arsenic in soil were found close to industrial facilities and tailing ponds. In addition, fluvisols were significantly more enriched with arsenic than soils at a distance from the river flows. The edible parts of the plant specimen showed different As contents, suggesting that these plant species have different propensities for the uptake and bioaccumulation of arsenic from soil.  相似文献

Aim To present a general, process‐based river model for suspended particulate matter (SPM). Location General approach based on processes; data from Europe and Israel. Methods The model has been tested and calibrated using an empirical river model for SPM and validated (blind‐tested) using data from seven European sites. This modelling gives mean monthly SPM concentrations in water for defined river sites. The model is based on processes in the entire upstream river stretch (and not for given river segments) and calculates the transport of SPM from land to water, primary production of SPM (within the upstream river stretch), resuspension, mineralization and retention of SPM in the upstream river stretch (but not bed load of friction materials, such as sand). The catchment area is differentiated into inflow (~ dry land) areas and outflow area (~ wetland areas dominated by relatively fast horizontal SPM‐fluxes). The model is simple to apply in practice as all driving variables may be accessed readily from maps. The driving variables are: latitude, altitude, continentality, catchment area and mean annual precipitation. Results Modelled values have been compared to independent empirical data from sites covering a relatively wide domain (catchment areas from 93 to 5250 km2, precipitation from 400 to 660 mm year?1, altitudes from ?210 to 150 m a.s.l., latitudes from 47 to 59° N and continentalities from 200 to 1000 km from the ocean). When blind‐tested, the model predicts annual SPM‐fluxes well. Conclusion When modelled values are compared to empirical data, the slope is almost perfect (1.03) and the r2‐value is 0.9996. This is good, given the fact that there are several simplifications in the model structure. It must, however, be stressed that there are only seven validation cases and that this model has not been tested for small catchments.  相似文献

The aims of this study were to document the mainly chemical behaviour of two linked artificial lakes used for both stormwater management and recreation in the new town of Craigavon. Further, the understanding of their behaviour should help in their management and the design of other similar lakes.The lake mean total phosphorus (73 µg P l–1), nitrate (0.50 mg N l–1) and chlorophyll a (25 µg l–1) concentrations, Secchi depth (1.2 m) and the estimated total phosphorus loading (1.98 g m–2 a–1) all classify the main lake as eutrophic. An important source of the phosphorus load on the lakes is the urban area of Craigavon (52% of the total load). The interrelationships between total phosphorus, chlorophyll a and Secchi depth in the main lake are similar to those in natural ones. In addition, the lake follows the total phosphorus load — trophic state relationships (lake total phosphorus and chlorophyll a concentrations and Secchi depth) found to apply elsewhere. These two points indicate that the artificial lakes in Craigavon behave similarly to natural ones.  相似文献

The revised Community Bureau of Reference (BCR) sequential extraction procedure has been applied to investigate the effectiveness of two soil remediation strategies to reduce the amounts of potentially toxic elements in three contrasting contaminated soils (soils A, B and C) from derelict chemical manufacturing sites in the UK. Soil A was from the 35–45 cm deep layer of a site used for the manufacture of sulfuric acid. Soils B and C were topsoils from a site used for the manufacture of explosives, nitric acid and nylon The remediation strategies were flushing with EDTA in a column experiment (applied to soils A, B and C) and EDTA enhanced phytoremediation with Taraxacum officinale in pots (applied to soil B). Soil B, which contained the least amounts of aqua regia extractable metals, except for lead, but higher proportions of analytes in non-residual (i.e. acid exchangeable, reducible and oxidisable) forms was found to release greater amounts of analytes when flushed with EDTA. Comparison of the BCR sequential extraction fractionation patterns obtained before and after flushing of soil B, suggested that EDTA removed calcium mainly from the acid exchangeable pool, manganese mainly from the reducible pool, zinc from both acid exchangeable and reducible pools, and copper and lead from acid exchangeable, reducible and oxidisable pools. The chelate enhanced phytoremediation pot experiment conducted using soil B showed that EDTA treatment was significantly positively, correlated (p?<?0.05) with an increase in the proportion of analytes recovered from the soil in step 1 of the BCR extraction scheme, for all analytes, and also enhanced metal uptake by plants. The sum of the amounts of analyte released in the first three steps of the sequential extraction, commonly regarded as the maximum amount of elements potentially available for plant uptake, was not positively correlated with plant-uptake.  相似文献

为分析岩溶石漠化区不同植被恢复模式土壤无机磷的形态特征,评价植被恢复的土壤供磷潜力,阐明有机碳及钙素在无机磷形态转化中的作用,选取研究区内8种有代表性的样地,采用蒋柏藩无机磷分级方法对土壤无机磷形态特征及影响因素进行研究。结果表明:研究区土壤全磷与速效磷含量分别在0.25—1.35 g/kg、1.05—53.01 mg/kg范围,无机磷总量在123.94—934.61 mg/kg,耕地与退耕地以及各退耕地之间全磷、速效磷、各形态无机磷含量水平差异明显,各退耕地磷素含量水平介于耕地与次生马尾松林地之间,退耕地中桃林地、花椒林地磷素含量水平较高、樟树林地、柳杉林地、撂荒草地次之、撂荒灌丛地较低。各样地土壤无机磷占全磷比例在51.2%—72.4%,不同形态的无机磷含量表现为O-PFe-PCa-PAl-P,其中Ca2-P、Al-P对速效磷的贡献率大,Fe-P、Ca8-P贡献较小,O-P、Ca10-P献率最小。不同活性土壤有机碳与不同形态钙素对各形态无机磷在总无机磷中比例的影响较大,p H、容重、粘粒含量、含水量等其它理化性质影响较小。  相似文献
