The pine wood nematode, Bursaphelenchus xylophilus, is an invasive plant parasitic nematode and a worldwide quarantine pest. An indigenous species in North America and the causal agent of pine wilt disease, B. xylophilus has devastated pine production in Southeastern Asia including Japan, China, and Korea since its initial introduction in the early 1900s. The reactive oxygen species (ROS) is the first line of defense utilized by host plants against parasites, while nematodes, counteractively, employ antioxidants to facilitate their infection. Peroxiredoxins (Prxs) are a large class of antioxidants recently found in a wide variety of organisms. In this report, a gene encoding a novel 2-cysteine peroxiredoxin protein in B. xylophilus was cloned and characterized. The 2-cysteine peroxiredoxin in B. xylophilus (herein refers to as "BxPrx") is highly conserved in comparison to 2-cysteine peroxiredoxins (Prx2s) in other nematodes, which have two conserved cysteine amino acids (Cp and Cr), a threonine-cysteine-arginine catalytic triad, and two signature motifs (GGLG and YF) sensitive to hydrogen peroxide. In silico assembly of BxPrx tertiary structure reveals the spatial configuration of these conserved domains and the simulated BxPrx 3-dimensional structure is congruent with its presumed redox functions. Although no signal peptide was identified, BxPrx was abundantly expressed and secreted under the B. xylophilus cuticle. Upon further analysis of this leader-less peptide, a single transmembrane α-helix composed of 23 consecutive hydrophobic amino acids was found in the primary structure of BxPrx. This transmembrane region and/or readily available ATP binding cassette transporters may facilitate the transport of non-classical BxPrx across the cell membrane. Recombinant BxPrx showed peroxidase activity in vitro reducing hydrogen peroxide using glutathione as the electron donor. The combined results from gene discovery, protein expression and distribution profiling (especially the "surprising" presence under the nematode cuticle), and recombinant antioxidant activity suggest that BxPrx plays a key role in combating the oxidative burst engineered by the ROS defense system in host plants during the infection process. In summary, BxPrx is a genetic factor potentially facilitating B. xylophilus infestation.  相似文献

We have characterized a family of GHF45 cellulases from the pine wood nematode Bursaphelenchus xylophilus. The absence of such genes from other nematodes and their similarity to fungal genes suggests that they may have been acquired by horizontal gene transfer (HGT) from fungi. The cell wall degrading enzymes of other plant parasitic nematodes may have been acquired by HGT from bacteria. B. xylophilus is not directly related to other plant parasites and our data therefore suggest that horizontal transfer of cell wall degrading enzymes has played a key role in evolution of plant parasitism by nematodes on more than one occasion.  相似文献

In our previous study, water-soluble extracts from Bursaphelenchus xylophilus (B. xylophilus), a pine wood nematode, were shown to enhance interleukin (IL)-4 plus lipopolysaccharide-induced polyclonal immunoglobulin (Ig) E production in vitro in mice and to increase serum levels of an antigen-nonspecific IgE in vivo. Here we examined whether the nematode extracts stimulate immunofunctions of murine peritoneal macrophages. In both resident and inflammatory macrophages, Fcgamma receptor-mediated phagocytosis was markedly activated by B. xylophilus extracts, while non-specific phagocytosis was not. The enhancement of specific phagocytosis was accompanied by an increase in the formation of IgG-Fcgamma receptor rosettes. B. xylophilus extracts also stimulated IL-1beta production in both types of macrophages, and enhanced NO production and mRNA expression of inflammatory cytokines in inflammatory macrophages. These results indicate that the extracts of B. xylophilus contain an activating substance(s) for immunofunctions in macrophages, besides an enhancing factor for polyclonal IgE production.  相似文献

Most Bursaphelenchus species are fungal feeding nematodes that colonize dead or dying trees. However, Bursaphelenchus xylophilus, the pine wood nematode, is also a pathogen of trees and is the causal agent of pine wilt disease. B. xylophilus is native to North America and here it causes little damage to trees. Where it is introduced to new regions it causes huge damage. The most severely affected areas are found in the Far East but more recently B. xylophilus has been introduced into Portugal and the potential for damage here is also high. As incidence and severity of pine wilt disease are linked to temperature we suggest that climate change is likely to exacerbate the problems caused by B. xylophilus and, in addition, will extend (northwards in Europe) the range in which pine wilt disease can occur. Here we review what is currently known about the interactions of B. xylophilus with its hosts, including recent developments in our understanding of the molecular biology of pathogenicity in the nematode. We also examine the potential developments that could be made by more widespread use of genomics tools to understand interactions between B. xylophilus, bacterial pathogens that have been implicated in disease and host trees.  相似文献

【目的】本研究旨在建立携带与未携带松材线虫Bursaphelenchus xylophilus的松墨天牛Monochamus alternatus成虫各组织的转录组数据库,揭示松墨天牛对松材线虫响应的转录组整体表达特征。【方法】以携带和未携带松材线虫的松墨天牛成虫的表皮、气管和脂肪体为材料,采用Illumina HiSeq TM 2000测序平台开展转录组测序,利用Trinity软件对RNA-Seq数据进行从头组装,利用NCBI数据库进行基因注释。通过DEG-Seq分析携带和未携带松材线虫的松墨天牛成虫中的差异表达基因,对上调表达基因进行GO和KEGG代谢途径富集分析,并通过实时荧光定量PCR技术对部分差异表达的基因包括ENV,CDK1,HSP70-C,HSP70,HSP75,DUO,PABPN1和IGFP基因的表达水平进行验证。【结果】测序过滤后共获得55059条单基因簇(unigene),平均长度为1536 bp。将unigene与数据库中的序列进行BLASTX比对,成功注释24354条unigenes,其中4022条unigene在GO数据库中获得注释,6098条unigene在KEGG数据库中获得注释。经过GO与KEGG富集分析显示,松墨天牛成虫携带松材线虫后,差异表达基因主要为与其气管中压力调节、组织与DNA修复、激素响应方面相关的基因。实时荧光定量PCR分析结果显示CDK1,HSP70,HSP75,DUO,PABPN1和IGFP基因的表达量在携带松材线虫的松墨天牛气管中相比未携带松材线虫的松墨天牛气管中的显著上调。【结论】本研究初步阐明松墨天牛携带松材线虫后转录组的整体表达模式,为进一步研究松墨天牛的基因功能及对松材线虫的胁迫响应过程奠定了基础。  相似文献

Here we report the draft genome sequence of Serratia sp. strain M24T3, which is associated with pinewood nematode Bursaphelenchus xylophilus, the causative agent of pine wilt disease. 【背景】在溴甲烷面临禁用的情形下,探讨新熏蒸剂甲酸乙酯对松材线虫的处理效果,可以科学地评估甲酸乙酯的使用前景。【方法】采用松材线虫分离液和带疫松木段,设置甲酸乙酯5个剂量梯度、5个处理温度和5个处理时间,测定其对松材线虫的毒力及CT值。【结果】在25℃下处理3、6、12、24、48h,甲酸乙酯对松材线虫的Lc50分别为2.63、1.60、0.99、0.41、0.20mg·L-1。温度对毒力有显著影响,在10~29℃,随温度升高,甲酸乙酯对松材线虫的毒力降低,19和29℃下,1.85mg·L-1甲酸乙酯处理松材线虫的死亡率分别为63%和100%。甲酸乙酯熏蒸12h内,能完全杀灭木段中的松材线虫。在23℃下处理6和12h,松材线虫死亡率达到99%时,甲酸乙酯的cr值分别为453.94和424.14mg·h-1·L-1。【结论与意义】甲酸乙酯可用于松材线虫的检疫处理。  相似文献

Cellulases are pathogenic substances suspected to be responsible for the development of the early symptoms of nematode disease. The pine wood nematode, Bursaphelenchus xylophilus (Parasitaphelenchidae), is the causal agent of pine wilt disease, which kills millions of pine trees. We used RNA interference (RNAi), a reverse genetic tool, to analyze the function of the endo-β-1,4-glucanase gene of B. xylophilus, which causes the most serious forest tree disease in China and the rest of eastern Asia. Silencing of this gene was detected through real-time PCR and cellulase activity assays after soaking for 24 h in dsRNA. The cellulase gene silencing effects differed among various siRNAs. The propagation and dispersal ability of these nematodes decreased when the endo-β-1,4-glucanase gene was silenced. It is important to select an effective siRNA before performing an RNAi test.  相似文献

The content of NO and H(2)O(2) as well as the activities of nitric oxide synthase (NOS)-like and nitrate reductase (NR) were monitored in the needles of Pinus thunbergii infected by Bursaphelenchus xylophilus. The results showed that the content of NO increased significantly only 8?h after the invasion of B. xylophilus, while H(2)O(2) increased 12?h after invasion. NO donor SNP could promote and NO scavenger cPTIO could prevent the production of NO and H(2)O(2). The content of NO changed earlier than that of H(2)O(2). In addition, the symptoms appeared 9, 5 and 12?days, respectively, after the inoculation with B. xylophilus, SNP pre-treatment and cPTIO pre-treatment followed by B. xylophilus infection. After B. xylophilus infection, the content of NO in P. thunbergii changed fiercely more earlier than the appearance of external symptoms, which indicated that the content of NO was related with the appearance and the development of the symptoms. The treatment with L-NNA (NOS inhibitor) inhibited the content of NO significantly, whereas, Na(2)WO(4) (NR inhibitor) had no effect. The further analysis of NOS revealed that NO changed in consistent with cNOS activity. To sum up, NO, as the upstream signal molecule of H(2)O(2), was involved in the pine early response to the invasion of B. xylophilus and influenced the accumulation of the content of H(2)O(2). Moreover, NOS-like rather than NR was responsible for the endogenous NO generation, which was modulated by cNOS during the interaction between P. thunbergii and B. xylophilus. Key message NO is involved in early response of P. thunbergii to the invasion of B. xylophilus and NOS is the key enzyme responsible for NO generation in P. thunbergii.  相似文献

本文依据捕食线虫真菌的捕食特性以及松材线虫的生长习性,对松材线虫进行了定时,定量的测定试验,试图通过培养后线虫的种群数量变化,来摸索定量测定捕食率的方法.在漏斗法基础上进行了相应的改进,成功地摸索出3种方法捕食率测定的新方法.各方法各具优缺点,滤膜镜检法和双层镜检法的数据更精确,条切镜检法和漏斗法所测数值较粗略.因此,可依据实验的侧重点以及实验所需的精密程度,而采用最适宜的方法进行捕食率测定实验.  相似文献

This study uncovered the potential clinical value and molecular driving mechanisms of circular RNAs (circRNAs) in gallbladder cancer (GBC). Differentially expressed circRNAs in GBC cells were screened by high-throughput sequencing. CircRNA_CDKN1A (circBase ID: hsa_circ_0076194) was knocked out in BGC-SD cells through transfection with sh-circRNA_CDKN1A. Then, proliferation was investigated via CCK8 and EdU assays, apoptosis via flow cytometry, migration via wound healing assays, and invasion via Transwell assays. Bioinformatics analysis of circRNA_CDKN1A-related signaling pathways was performed using MetScape and g:Profiler. Results showed that the knockdown of circRNA_CDKN1A enhanced the proliferation, migration, and invasion of GBC cells and inhibited apoptosis. In addition, knocking out circRNA_CDKN1A promoted GBC cell proliferation and enhanced the dry indices of the OCT4 protein and CD34 expression levels. The knockdown of circRNA_CDKN1A activated the epithelial–mesenchymal transition pathway. Bioinformatics analysis revealed that the biological role of circRNA_CDKN1A in GBC cells involved the NF-κB pathway. LY2409881, which is an NF-κB inhibitor, reversed the effects induced by the knockdown of circRNA_CDKN1A in GBC-SD cells. In summary, the knockdown of circRNA_CDKN1A promoted the progression of GBC by activating the NF-κB signaling pathway. For the first time, this study revealed the mechanism of circRNA_CDKN1A-mediated regulatory action in GBC and identified the newly discovered circRNA_CDKN1A–NF-κB signaling axis as a potentially important candidate for clinical therapy and prognostic diagnosis of GBC.  相似文献

Two pectate lyase genes (Bx-pel-1 and Bx-pel-2) were cloned from the pine wood nematode, Bursaphelenchus xylophilus. The deduced amino acid sequences of these pectate lyases are most similar to polysaccharide lyase family 3 proteins. Recombinant BxPEL1 showed highest activity on polygalacturonic acid and lower activity on more highly methylated pectin. Recombinant BxPEL1 demonstrated full dependency on Ca2+ for activity and optimal activity at 55 degrees C and pH 8 to 10 like other pectate lyases of polysaccharide lyase family 3. The protein sequences have predicted signal peptides at their N-termini and the genes are expressed solely in the esophageal gland cells of the nematode, indicating that the pectate lyases could be secreted into plant tissues to help feeding and migration in the tree. This study suggests that pectate lyases are widely distributed in plant-parasitic nematodes and play an important role in plant-nematode interactions.  相似文献

松材线虫病是重要的森林病害,该病与松材线虫携带的病原菌和松树的内生病原菌密切相关。在室内条件下,初步研究了从人工培养的松材线虫上分离到的菌株C对松材线虫病的抑制作用。在健康的水培马尾松枝上分别接种松材线虫接种液、菌株C接种液、松材线虫与菌株C的混合接种液。处理后松枝的相对重量与相对蒸腾强度均为:接种菌株C的松枝>混合接种的松枝>接种线虫的松枝。处理后15d时,接种线虫的松枝与混合接种的松枝的相对重量有显著性差异(p<0.05)。接种线虫松枝的存活期显著短于其它处理松枝的存活期(p<0.05)。接种菌株C的针叶褐变株数少于接种线虫的松枝,两者有显著性差异(p<0.05)。从接种线虫和混合接种的所有松枝中都分离到松材线虫,且分离出的线虫量没有显著性差异。将8个月生的断根马尾松苗插入菌株C的查彼培养液的滤液中培养,6d后松苗的平均感病指数和感病株率均显著少于对照(p<0.05)。这表明,菌株C对松材线虫病有抑制作用,菌株C培养液中产生的某些代谢物质有利于松苗的抗病和存活。菌株C可能抑制了松树上的内生病原菌和松材线虫携带的病原微生物,或提高了松树的生长力和抗逆能力。经电子显微镜观察并参照AP I 20 C AUX鉴定系统鉴定,菌株C为季也蒙假丝酵母C and id a gu ilierm ond ii。  相似文献

本试验测定了牛心朴子5种有机溶剂提取物以及经光照和高温处理后的甲醇提取物对松材线虫的毒力。结果表明,5种有机溶剂提取物对松材线虫的校正击倒率随极性的增加而升高:二氯甲烷〈乙酸乙酯〈无水乙醇〈丙酮〈甲醇。甲醇提取物在浓度为6,3、2g/L时,48h的校正击倒率达近100%。经光照和高温处理后的两组甲醇提取物比未处理组毒力总体下降,但当两处理组提取物的浓度为6,3、2g/L时,48h的校正击倒率均达80%以上。  相似文献

松材线虫是重要的外来有害生物,造成松树大量死亡,带来严重的经济损失和生态破坏.松材线虫与伴生微生物存在密切的生态关系.本文综述了松材线虫伴生微生物的种类及其在松材线虫繁殖和致病过程中的重要生态作用,从微生态系统角度对松材线虫病进行探讨.伴生真菌能为松材线虫提供食物,维持松材线虫次生侵染循环,提高分散型第三龄幼虫在种群中的比例,利于松材线虫侵染扩散;伴生细菌能够提高松材线虫致病性,促进其繁殖,并菌有助于松材线虫降解松萜类物质,从而提高松材线虫的适应性.  相似文献

Pathogenicity of the pine wood nematode, Bursaphelenchus xylophilus, to Japanese larch, Larix kaempferi, seedlings was tested with inoculation experiments under nursery conditions. Water suspensions of nematodes (mixed stages cultured on Botrytis cinerea or dispersal fourth-stage juveniles (DJ4) extracted from the adult Japanese pine sawyer, Monochamus alternatus) were injected into the stems of 2- and 3-year-old Japanese larch and Japanese black pine, Pinus thunbergii, seedlings growing in a nursery. In another treatment, Japanese pine sawyer adults holding DJ4 were released under a net that covered the upper half of the seedlings. Regardless of nematode inoculation method, Japanese larch seedlings were as susceptible as Japanese black pine seedlings to B. xylophilus under nursery conditions. The rate of disease development was similar on larch and pine seedlings. Nematode population densities were lower in the stems of dead larch seedlings than in the stems of dead pine seedlings. Histopathological observations revealed that the distribution of nematodes in the stems of dead larch seedlings was mostly limited to the cortex, phloem and cambial zone. Traumatic resin canal formation was one of the most characteristic symptoms in larch seedlings which was dissimilar to that in pine seedlings.  相似文献

The draft genome sequence of Pseudomonas sp. strain M47T1, carried by the Bursaphelenchus xylophilus pinewood nematode, the causative agent of pine wilt disease, is presented. In Pseudomonas sp. strain M47T1, genes that make this a plant growth-promoting bacterium, as well as genes potentially involved in nematotoxicity, were identified.  相似文献

松材线虫Bursaphelenchus xylophilus是我国重要的林业检疫性有害生物之一,由其引发的松材线虫病已造成巨大的经济损失,严重阻碍了林业的健康发展。研究并明确松材线虫在四川省的潜在适生区,对四川省有关部门制定该病害的早期监测、预警及防控具有一定的参考意义。本文基于2009—2018年四川省林业有害生物普查数据中松材线虫病和松墨天牛Monochamus alternatus的实际地理分布数据(松材线虫病:n=208,松墨天牛:n=803)及19个环境变量数据,利用MaxEnt模型和Arc GIS对松材线虫在四川省的潜在分布区进行预测,并用ROC曲线分析法检测模型模拟精度、用刀切法检测变量的重要性及其适宜值。结果表明:松材线虫在四川省的潜在最佳适生区主要分布在宜宾市、广安市、达州市、自贡市、西昌市,以及乐山市和眉山市的交界区,面积为36 541 km^2;影响松材线虫分布的主要环境变量为最干季均温(适值范围1. 5~8. 0℃,最适值6. 4℃)、季节性降水变异系数(适值范围22. 5%~34. 0%,最适值34. 0%)、最冷月最低温(适值范围0. 4~2. 5℃,最适值1. 9℃)、海拔(适值范围250~5 500 m,最适值450 m)、年温差(适值范围5. 9~9. 1℃,最适值5. 9℃)和年降水量(适值范围64~135 mm,最适值68 mm)。  相似文献
