1植物名称樱桃种间杂交后代。杂交组合有甜樱桃(Prunus avium)×酸樱桃(P.cerasus),甜樱桃(P.avium)×欧洲李(P.domestica)、甜樱桃(P.avium)×中国樱桃(P.pseudocerasus)、酸樱桃(P.cerasus)×欧洲李(P.domestica)、酸樱桃(P.cerasus)×中国樱桃(P.pseudocerasus)。2材料类别幼胚。3培养条件基本培养基为F14。(1)幼胚培养培养基:F14+6-BA0.5mg·L-1(单位下同)+IBA1.0+GA1.0;(2)胚芽继代增殖培养基:F14+6-BA0.5+IBA0.2+GA0.2;(3)生根培养基:F14+IBA0.5+NAA0.05。培养基中蔗糖为2%,琼脂为0.7%,pH5.4。培养温度(25±2)℃,光照1…  相似文献

实验所用的基本培基中以B_5培养基对向日葵幼胚培养为最适。激素萘乙酸主要促进幼胚的胚性生长。吲哚乙酸(IAA)与激动素(KT)的配合使用,对幼胚的分化起明显的调节作用。天然提取物则明显地促进幼胚生长、其中荸荠汁的培养基可使向日葵球形胚进行正常发育。 用两种改良的B_5培养基做胚胎培养系统,使向日葵种间杂交胚生长发育。先将杂交后的心形胚培养在胚胎生长培养基上,进行充分的胚性生长。然后把胚转移到萌发培养基,使其萌发形成幼苗,最后移入土壤直至开花。  相似文献

本文报道美味猕猴桃(Actintdia delictosa cv.Hayward)(6x)和毛花猕猴桃(A.eria-ntha)(2x)杂交当代种子的胚胎学分析和胚援救的结果:1.根据杂交种子外部形态和胚胎发育程度,区分为正常的和败育的两类,其比率接近1:1。正常种子的胚发育分化正常,且含有适量胚乳。败育种子中,除少数不含胚和胚乳外,绝大多数种子的胚发育终止于球形、心形和早子叶阶段,胚体畸形。胚乳处于消失或完全解体。2.在被试8组培养基中,最适合胚萌发和生长的是:MS+IAA0.5ppm+GA_30.5ppm,MS+2ip2ppm+IAA0.5ppm+GA_30.5ppm和 MS+2ip2ppm+GA_30.5ppm。适于实生苗生长的培养基为 MS+GA_30.5ppm 和 MS 培养基。在 MS+BAP2ppm 和 MS+IAA0.5ppm+GA_30.5ppm 培养基上,一些正常种子和少数不正常种子胚的下胚轴直接形成了许多不定芽,从而诱导产生了更多的杂种植株。但是在MS十BAP2ppm+GA_30.5ppm,MS+BAP2ppm+IAA0.5ppm 和 MS+2ip2ppm+GA_30.5ppm培养基上,虽然胚产生了愈伤组织,但都未分化出器官。杂种实生苗根尖细胞近半数的含有近4倍性染色体数(4x=116),约半数为其他倍性。  相似文献

采用人工授粉的方法,以‘小桃红’建兰为母本、纹瓣兰为父本进行杂交,获得了种间远缘杂交后代植株。杂交果苹与母本自交果荚的大小差异大小,但鲜重明显大于母本,果荚内的种子量却少于母本。杂种胚离体培养90d出现绿色原球胚,210d可萌发出大量植株;而‘小桃红’自交种子培养210d只形成少量绿色根状茎。杂种胚的萌发具有杂种优势,不仅萌发快,而且出苗率高,萌发时先形成原球胚,原球胚上长出大量小根毛,继而形成大量假鳞茎,假鳞茎经转瓶培养后,分化形成大量健壮植株,分化率高。  相似文献

以不同类型的大白菜与紫甘蓝为材料,运用蕾期授粉结合胚挽救技术获得大白菜与紫甘蓝的种间杂种,并对其进行细胞学鉴定.结果表明,大白菜与紫甘蓝杂交,获得了57株F1代幼苗;对根尖染色体数量进行观察和杂种花粉特性调查发现,其中47株具有预期的染色体数目,2n =19,鉴定为真杂种,其花粉败育;另有6株具有38条染色体,鉴定为种间异源双二倍体,应该是发生了染色体自然加倍,其花粉可育.可育的杂种F1代与大白菜回交,获得了大白菜-紫甘蓝BC1代材料.田间观测结果显示,杂种F1代植株综合性状均介于双亲之间,BC1代植株包球明显,综合性状偏向母本大白菜.  相似文献

利用GISH和RAPD检测黄毛草莓×凤梨草莓种间杂种   总被引:5,自引:2,他引:3  
以地高辛(DIG)标记的黄毛草莓基因组DNA为探针、凤梨草莓基因组DNA为封阻对黄毛草莓×凤梨草莓杂种F1的五倍体植株根尖染色体进行基因组原位杂交(GISH),结果表明利用GISH可将黄毛草莓染色质从凤梨草莓中检测出来,从而证实了黄毛草莓与凤梨草莓种间杂交的真实性.利用RAPD技术对来自黄毛草莓与凤梨草莓不同品种的2个杂交组合获得的14个株系及其亲本的DNA指纹图谱进行分析.结果表明,不同株系之间在基因组水平上具有高度异质性,以引物S1371和S1405能将黄毛草莓×春霄组合的10个杂交后代株系加以区别,以单一引物S1371、S1380、S1397、S1404、S1405可使黄毛草莓×硕香后代株系之间相互区别.在杂交后代中观察到了亲本黄毛草莓与凤梨草莓不同栽培品种的特征带,从而进一步证实黄毛草莓与凤梨草莓种间杂交后代的真实性.  相似文献

为提高姜花属种间杂交胚挽救中幼胚萌发率,以白姜花×金姜花的胚珠为试材,研究不同胚珠发育时期、不同培养基及低温处理果实对幼胚萌发率的影响.结果表明,白姜花×金姜花胚挽救的适宜培养基是MS+0.1 mg/L BA十0.1 mg/L NAA;接种时期以60 d的幼胚培养效果最佳;低温处理果实3~6 d能有效提高幼胚的萌发率.  相似文献

选用5个风信子品种进行花粉生命力测定,对完全双列杂交获得的杂种种子剥胚进行离体培养.试验表明:(1)风信子不同品种花粉萌发率差别较大,其中Fondante的花粉萌发率最高为74.45%,Amsterdam的最低为12.68%,其他3个品种的花粉萌发率介于二者之间.(2)品种间自交杂交亲和力存在一定差异,结实率大于30%的组合从高到低依次为Fondante×Atlantic(61.2%)、Fondante×Fondante(54.0%)、Carnegie×Carnegie(42.6%)、JanBos×JanBos(36.0%)、Carnegie×Atlantic(32.7%)、Atlantic×Fondante(30.0%),组合Atlantic×Amster-dam、Carnegie×JanBos、Amsterdam×Fondante、Amsterdam×JanBos、JanBos×Amsterdam未得到果实,其余组合结实率介于0~30%之间.(3)适宜风信子杂交幼胚离体培养的初代培养基为MS+100 mg/L谷氨酰胺,其次为MS+0.2 mg/LIBA+0.02 mg/L6-BA+100 mg/L谷氨酰胺...  相似文献

取兰属大花蕙兰为母本、墨兰为父本进行种间杂交并进行杂种胚拯救研究。结果显示,子房离体培养明显优于胚珠离体培养,是胚抢救培养的有效手段。不同取材时间及在不同培养基中子房培养情况差异明显。在授粉后60 d子房离体培养即可获得成功,但以授粉后90 d子房离体培养最佳,萌发率高,达80%。子房培养的最适培养基为VW+BA 0.2 mg/L+NAA 0.2 mg/L+蛋白胨3 g/L+水解酪蛋白500 mg/L+椰子汁10%。  相似文献

采用体细胞染色体计数和RAPD (Random Amplified Polymorphism DNA) 分子标记的方法鉴定甘蔗与蔗茅属间杂交种F1的真实性.结果表明,杂种F1材料01/47、01/85、01/120的2n=80~82,染色体遗传方式为n 2n.用110个随机引物进行RAPD扩增,63个引物可获得双亲的RAPD多态性,其中3个引物OPC-19、OPE-2、OPF-4可在杂种F1材料01/64和01/120的RAPD扩增标记中显示出双亲的特征谱带,分别为父本3500bp和母本1300bp、父本1350bp和母本900bp、父本550bp和母本900bp.  相似文献

以白皮松成熟胚为外植体诱导再生小植株.试验结果表明,成熟胚不定芽诱导以MS培养基最佳,附加0.294 mg/L的NAA和3.56 mg/L的6-BA时,诱导率接近100%;MS培养基附加NAA(0.05 mg/L)时,平均增殖系数可达6.3以上.不定芽增殖率最大值(10)出现在SH培养基上,此时NAA浓度为0.05 mg/L、6-BA浓度为4 mg/L.MS培养基中加入适量活性炭和GA3能促进不定芽生长,随着活性炭和GA3浓度的增加,有效嫩梢(≥2cm和≥4 cm)的比率显著增高;当活性炭和GA3浓度过高时(分别超过2.75 g/L和4.1 mg/L),不定芽的伸长与生长受到抑制.在离体培养条件下,以种胚为外植体获得了无根苗.  相似文献

从100个随机引物中选取重复性好、条带清晰的随机引物分别对针叶树和阔叶树上的34个散斑壳(Lophodermiumspp.)菌株进行RAPD-PCR扩增反应,RAPD分析结果与表型性状表现出良好的一致性,从而表明RAPD技术可以较好的应用于散斑壳属种内及近似种间亲缘关系的分析。通过RAPD分析结果与表型性状的比较和分析,明确了部分表型性状在散斑壳属近似种区分方面的重要性。  相似文献

The shoot apices of ten groundnut interspecific hybrids, showing peanut stripe virus symptoms were used as explants. Murashige and Skoog (MS) medium containing 1 mg dm-3 each of naphthaleneacetic acid (NAA) and benzylaminopurine (BAP) supported maximum callusing of the hybrids J 11 × Arachis sp. PI 30008 as well as J 11 × A. sp. Manfredi # 5 (86.3 and 82.6 %, respectively). On MS medium with 3.5 mg dm-3 NAA and 0.2 mg dm-3 BAP, the meristems of J 11 × A. stenosperma as well as J 11 × A. otavioi gave good response (79.7 and 77.2 %). The regeneration frequencies ranged from 16.7 to 76.9 % and was more than 30 % in all hybrids except for J 11 × PI 30085. Shoots (5 cm) regenerated from the virus-free (ELISA tested) calli rooted well (60 – 80 %) on MS basal medium supplemented with 0.4 % activated charcoal and 200 mg dm-3 casein hydrolysate. In addition, long shoots which failed to produce roots, when transferred to soil during rainy season and protected from direct sun got established. Species of the genus Daphnia, in particular the subgenus Hyalodaphnia, represent a taxonomically problematic group due to their phenotypic plasticity, local races and the formation of interspecific hybrids and backcrosses. In this study, we present a genetic approach utilising nuclear DNA to unequivocally identify species and interspecific hybrids. Several nuclear loci (ITS1-ITS2, CA14 and GA13) were amplified by PCR and products were subjected to diagnostic restriction enzymes (restriction fragment length polymorphism; RFLP). The application of this approach to several populations across Europe revealed that the markers are highly consistent and reproducible. In addition, we illustrate with a number of examples how this approach contributed to unravel previously unrecognised taxa, increased the sensitivity of biodiversity studies or contributed to the analysis of resting egg banks. Advantages and disadvantages of this approach compared to existing techniques are discussed and several empirical studies and their results are summarised.  相似文献

Seed coats and fibers of aborted structures (termed “motes”) and mature seeds from an interspecific hybrid (ISH) of cotton (Gossypium hirsutum x G. barbadense) were studied by microscopy to determine (a) the developmental stages at which motes of various sizes stop growing and (b) the origin of lint-contaminating fragments. Small motes (1–2 mm wide, up to 3 mm long) were identified as ovules whose growth had been arrested before anthesis or soon afterwards, medium-size motes (1–3 mm wide, 3–5 mm long) as those whose growth had been arrested up to 10 days post-anthesis, and large motes (3–5 mm wide, 5–8 mm long) as those which had stopped growing about 20 days post-anthesis. Microscopic examination of lint-contaminating fragments showed that they derive mainly from medium-size motes and partly from mature seed coats. Reducing the number of medium-size motes by breeding can, therefore, improve the lint quality of ISHs of cotton.  相似文献
