Summary The bioleaching of cobalt from domestic, industrial smelter wastes was studied.Thiobacillus ferrooxidans solubilized Co from sulfidic dross furnace mattes. At pulp densities of 4% (w/v) up to 600 mg of Co per liter of leaching solution was released from nickel matte, corresponding to removal of about two-thirds of the original amount of Co in the matte. Bioleaching methods may be useful as a component of a process for solubilization and recovery of Co from sulfidic smelter mattes.  相似文献

Thiobacillus ferrooxidans was cultivated on 100-nm-thick synthetic pyrite (FeS2) films. The steps of biooxidation were studied with high-resolution transmission electron microscopy. The crystallized sulfide was transformed into colloidal sulfur (4–70 nm, depending on the age of the cell and the degree of substrate oxidation; 70nm initially and 4nm after oxidation of the pyrite substrate), which was taken up and distributed over an organic capsule around the bacteria. This colloidal sulfur acted as intermediate energy storage and was transferred by contact to daughter cells not directly attached to the sulfide substrate.  相似文献

Oxidation of Fe(II) iron and bioleaching of pyrite and chalcopyrite by Thiobacillus ferrooxidans was adversely affected by isopropylxanthate, a flotation agent, and by LIX 984, a solvent-extraction agent, each at 1 g/l. The reagents/l were adsorbed on the bacterial surface, decreasing the bacteria's development and preventing biooxidation. Both reagents inhibited the bioleaching of pyrite and LIX 984 also inhibited the bioleaching of chalcopyrite.  相似文献

A strain ofThiobacillus ferrooxidans MAL-4-1 was adapted to grow at higher concentrations of copper by repeated subculturing in the presence of increasing levels of added cupric ions in 9K medium. The strains adapted to copper were found to be more efficient in bioleaching of copper from concentrates. When copper tolerant strains were back cultured repeatedly in 9K medium without cupric ions, the initially developed metal tolerance was observed to be lost. This indicates that the copper tolerance developed is stress-dependent and not a permanent trait of the adapted strain.  相似文献

Summary Uranyl sulphate (0.2–0.9 mM) inhibited ferrous iron oxidation by growing cultures ofThiobacillus ferrooxidans. The addition of 5–100 mM uranium to the cultures caused immediate cessation of carbon dioxide fixation, rapid loss of viability and gradual depression of ferrous iron oxidation. Virtually no uranium was found in washed cells grown in the presence of subtoxic to toxic amounts of uranyl sulphate. Uranium-poisoned organisms appeared plasmolyzed in electron micrographs. Cultures tolerant to 5 mM UO2 2+ were develoepd by successive subculturing in increased uranium concentrations. The tolerance was maintained during subculturing in uranium-free medium. Frequency of mutants resistant to 1.0 and 1.5 mM UO2 2+ was 1 per 1.3×106 and 1 per 9.0×108, respectively. The frequency was increased in the presence of 15–150 mM nickel, zinc and manganese. In liquid cultures, bivalent cations and EDTA alleviated the toxicity of 2 mM uranyl sulphate.  相似文献

The effects of naturally-occurring organic compounds on ferrous iron oxidation by the bacterium Thiobacillus ferrooxidans were examined with a view to using these compounds to treat or prevent acid mine/rock drainage. The compounds glucose, cellobiose, galacturonic acid, and citric acid were added to the growth medium of five different strains of the bacterium and growth studies were done to determine whether or not strain differences existed with respect to organic compound sensitivity. The effects of these compounds were compared to the effects of sodium dodecyl sulfate (SDS) an anionic detergent. Each of the compounds tested had an inhibitory effect on the strains of the bacterium and sensitivity to these compounds was strain dependent. All strains appeared to be equally susceptible to SDS. Inhibitory concentrations ranged from 70 mM to >280 mM for glucose, 7.5 mM to 150 mM for cellobiose, 20 mM to 230 mM for galacturonic acid, and 50 mM to 130 mM for citric acid. SDS effectively inhibited iron oxidation for all strains at a concentration of 0.3 mM, the lowest concentration tested. Some naturally-occurring organic compounds, therefore, might be candidates for the growth control of T. ferrooxidans.  相似文献

Differentially labelled 35S-thiosulphate was taken up by washed cells of Thiobacillus ferrooxidans which were previously grown on thiosulphate. The uptake was proportional to the biomass over the range 0.5–4.0 mg dry wt. of bacteria and showed typical saturation kinetics with an estimated K m value of 0.5 mM for 35S-thiosulphate. Dithionate and Group VI anions inhibited the uptake, which was under pH control and had a temperature optimum of 50°C. In the absence of thiosulphate, the cells bound 35S-sulphate but the binding did not increase on prolonged incubation and the label could be removed completely by washing with dilute sulphuric acid. Increasing amounts of the label were incorporated from [outer-35S]thiosulphate into cellular materials over a 60-min period, whereas little or no assimilation was observed from either the [inner-35S]thiosulphate or 35S-sulphate. The interaction of uranium with cells of three recently described eco-types of Acidithiobacillus ferrooxidans recovered from uranium mining wastes was studied. The uranium sorption studies demonstrated that the strains from these types possess different capabilities to accumulate and tolerate uranium. The amount of uranium biosorbed by all A. ferrooxidans strains increased with considerable concentrations. We have found that the representatives of type II accumulate significantly higher amounts of uranium in comparison to the other A. ferrooxidans strains. The investigations of the tolerance to uranium showed that the types I and III are resistant to 8 and 9 mM of uranium respectively, whereas the type II does not tolerate more than 2 mM of uranium. The recovery of the accumulated uranium by desorption was investigated using various desorbing agents as sodium carbonate, sodium citrate and EDTA at different concentrations. Sodium carbonate was the most efficient desorbing agent, removing 97% of the uranium sorbed from the cells of A. ferrooxidans type III, and 88.33 and 88.50% from the cells of the types I and II, respectively.  相似文献

Summary Microorganisms are currently used for the recovery of copper from mining dumps of low-grade ore. One of the most important microorganisms involved in copper-solubilization isThiobacillus ferrooxidans, although many other microbial genera are also thought to be implicated. A mining dump poses some special problems for the industrial microbiologist because it represents a non-sterile and heterogeneous substrate. Consequently, to enhance our knowledge of the role of microorganisms in metal recovery we must identify the indigenous microorganisms and understand their respective contributions to the process. In addition, when a superior strain of microorganism is developed in the laboratory, by genetic engineering or by other means, we must have a method to evaluate the maintenance of such a strain in the mining dump. In this paper, we describe DNA homology studies, using dot blot and Southern blot analysis of hybridizations of both whole genomic DNA and cloned DNA sequences, to identify and enumerate several bioleaching microorganisms. We demonstrate that it is possible to identify different species of microorganisms and, in one case, to discriminate between different strains of a single species. It is also possible to identify and quantitate certain species in a mixed culture. DNA hybridization analysis has several advantages over the more conventional bacteriological methods of identification, especially in a complex bioleaching situation.  相似文献

Two strains of Thiobacillus, T. ferrooxidans and T. thiooxidans, have been isolated from a bacterial inoculum cultivated during a one-year period in a 1001 continuous laboratory pilot for treatment of an arsenopyrite/pyrite concentrate. The optimum pH for the growth of both strains has been found to be between 1.7 and 2.5. Because of the high metal toxicity in bioleach pulps, the tolerance of T. ferrooxidans and T. thiooxidans with respect to iron and arsenic has been studied. The growth of both strains is inhibited with 10 g/l of ferric ion, 5 g/l of arsenite and 40 g/l of arsenate. 20 g/l of ferrous iron is toxic to T. ferrooxidans but 30 g/l is necessary to impede the growth of T. thiooxidans.  相似文献

PVA-cryogels entrapping about 109 cells of Acidithiobacillus ferrooxidans per ml of gel were prepared by freezing-thawing procedure, and the biooxidation of Fe2+ by immobilized cells was investigated in a 0.365 l packed-bed bioreactor. Fe2+ oxidation fits a plug-flow reaction model well. A maximum oxidation rate of 3.1 g Fe2+ l–1 h–1 was achieved at the dilution rate of 0.4 h–1 or higher, while no obvious precipitate was determined at this time. In addition, cell-immobilized PVA-cryogels packed in bioreactor maintained their oxidative ability for more than two months under non-sterile conditions. The loss of part of the lipopolysaccharides (LPS) of the outer membrane of T. ferrooxidans negatively influenced the attachment of the bacteria to minerals and the bioleaching process. LPS previously extracted from T. ferrooxidans and which had come into contact with pyrite inhibited the attachment of cells to minerals and also negatively affected the bioleaching. These results suggest that LPS play an important role in the attachment of the microorganisms and therefore, its presence or absence could affect the bioleaching process.  相似文献

Thiobacillus ferrooxidans has been cultivated on synthetic pyrite (FeS2) single crystals as the only energy source and the pyrite interface investigated with respect to characteristic morphological changes using scanning electron microscopy. Corrosion patterns of bacterial size were identified in different stages of development and correlated with bacterial activity. It appears that bacterial attack of the sulfide interface starts by secretion of organic substances around the contact area between the bacterial cell and the sulfide energy source. They might either be part of a pseudo capsule which shields the contact area or may form a sulfur absorbing and transporting organic film. Degradation of the sulfide occurs in the contact area below the bacterial cell leading to a corrosion pit which the bacterium may abandon after it has reached a depth of bacterial dimension. Electron spectroscopic (XPS) and X-ray fluorescence studies indicate a layer of organic substances covering the sulfide surface under bacterial leaching conditions, which is sufficiently thick for consideration in interfacial chemical mechanisms.  相似文献

The intermediary production of elemental sulfur during the microbial oxidation of reduced sulfur compounds has frequently been reported. Thiobacillus ferrooxidans, an acidophilic chemolithoautotroph, was found to produce an insoluble sulfur compound, primarily elemental sulfur, during the oxidation of thiosulfate, trithionate, tetrathionate and sulfide. This was confirmed by light and electron microscopy. Sulfur was produced from sulfide by an oxidative step, while the production from tetrathionate was initiated by a hydrolytic step, probably followed by a series of chemical reactions. The oxidation of intermediary sulfur was severely inhibited by sulfhydryl-binding reagents such as N-ethylmaleimide, by the addition of uncouplers or after freezing and thawing of the cells, which probably damaged the cell membrane. The mechanisms behind these inhibitions have not yet been clarified. Finally, it was observed that elemental sulfur oxidation by whole cells depended on the medium composition. Summary Copper contained in a solid industrial waste produced in a silicone manufacturing process was leached with spent iron medium from aThiobacillus ferrooxidans culture. Most effective leaching was observed in a continuously fed, dual reactor system. Spent iron medium was generated by growingT. ferrooxidans in 0.9 K iron medium at pH 1.5 in the first reactor, and was transferred to a second reactor in which it leached the copper from the waste. Leaching was effective at a pulp density of the waste material as high as 20%. In experiments run at a pulp density of 2.5%, the spent iron medium was most efficient in leaching copper when it was first diluted 100-fold with a mineral salts solution at pH 1.5. Removal of the copper from the waste appeared to involve its displacement by acid, dissolved mineral salts, and ferric iron. Potentials for practical application of this process are discussed.  相似文献

Time-resolved laser-induced fluorescence spectroscopy (TRLFS) was used to study the properties of uranium complexes (emission spectra and fluorescence lifetimes) formed by the cells of the three recently described eco-types of Acidithiobacillus ferrooxidans. The results demonstrated that these complexes have different lifetimes which increase in the same order as the capability of the strains to accumulate uranium. The complexes built by the cells of the eco-type II were the strongest, whereas, those of the eco-types I and III were significantly weaker. The emission spectra of all A. ferrooxidans complexes were almost identical to those of the uranyl organic phosphate compounds. The latter finding was confirmed by infrared spectroscopic analysis.  相似文献
