Diatomaceous earth was modified by in situ precipitation of metallic hydroxides. Modification decreased the negative charge on the diatomaceous earth and increased its ability to adsorb viruses in water. Electrostatic interactions were more important than hydrophobic interactions in virus adsorption to modified diatomaceous earth. Filters containing diatomaceous earth modified by in situ precipitation of a combination of ferric chloride and aluminum chloride adsorbed greater than 80% of enteroviruses (poliovirus 1, echovirus 5, and coxsackievirus B5) and coliphage MS2 present in tap water at ambient pH (7.8 to 8.3), even after filtration of 100 liters of tap water. Viruses adsorbed to the filters could be recovered by mixing the modified diatomaceous earth with 3% beef extract plus 1 M NaCl (pH 9).  相似文献

The increased cell density and product titer in biomanufacturing have led to greater use of depth filtration as part of the initial clarification of cell culture fluid, either as a stand-alone unit operation or after centrifugation. Several recent studies have shown that depth filters can also reduce the concentration of smaller impurities like host cell proteins (HCP) and DNA, decreasing the burden on subsequent chromatographic operations. The objective of this study was to evaluate the HCP removal properties of the Pall PDH4 depth filter media, a model depth filter containing diatomaceous earth, cellulose fibers, and a binder. Experiments were performed with both cell culture fluid (CCF) and a series of model proteins with defined pI, molecular weight, and hydrophobicity chosen to match the range of typical HCP. The location of adsorbed (fluorescently labeled) proteins within the depth filters was determined using confocal scanning laser microscopy. Protein binding was greater for proteins that were positively charged and more hydrophobic, consistent with adsorption to the negatively charged diatomaceous earth. The lowest degree of binding was seen with proteins near their pI, which were poorly removed by this filter. These results provide new mechanistic insights into the factors governing the filter capacity and performance characteristics of depth filters containing diatomaceous earth that are widely used in the clarification of CCF.  相似文献

Silica gel, aluminium oxide, diatomaceous earth, polyamide, cyano, diol and amino plates have been tested for their capacity to separate the color pigments of six chili powders of different origin by both adsorption and reversed-phase thin-layer chromatography. The plates were evaluated at 340 and 440 nm wavelengths. Best separation of color pigments was obtained on impregnated diatomaceous earth layer using acetone-water 17:3 v/v eluent. It was found that the pigment composition of chili powders showed marked differences. Principal component analysis employed for the classification of the chili powders according to their pigment composition indicated that these differences can be used for the determination of the similarity or dissimilarity of the chili powders.  相似文献

The use of this technique allows a maximum recovery of 71.2% compared with 46.7% phage recovery obtained by the standard elution procedure. High percentages (over 83%) of phage adsorption were obtained with different filters from 1-liter aliquots of the samples, except for Virosorb 1-MDS filters (between 1.6 and 32% phage adsorption). Phage recovery by using the slow passing of the eluent depended on the filter type, with recovery ranging between 1.6% for Virosorb 1-MDS filters treated with polyethyleneimine and 103.2% for diatomaceous earth filters treated with 0.1% Nalco.  相似文献

This work investigates the adsorption of paraquat from aqueous medium using a methacrylic acid (MAA)-modified rice husk. The carboxyl groups were chemically bound to the surface of the rice husk by graft copolymerization using Fenton's reagent as a redox initiator. The graft copolymerization was examined to determine the H(2)O(2) concentration and the amount of MAA monomer used. FT-IR spectra confirmed the presence of carbonyl groups on the structural units of the rice husk derivative. The MAA-modified rice husks were hydrolyzed to sodium salt and used to adsorb paraquat. The adsorption was rapid in the first few minutes and quickly reached equilibrium. Equilibrium adsorption data are more consistent with the Langmuir isotherm equation than with the Freundlich equation. The maximum adsorption capacity of modified rice husks was 317.7mg/g-adsorbent. This value clearly exceeds the 60mg/g of Fuller's earth and the 90mg/g of activated carbon, which are the most commonly used binding agents for paraquat.  相似文献

四纹豆象Callosobruchus maculatus (F.)是伊朗豇豆种子上的主要贮藏害虫。控制这一害虫时, 用生物杀虫剂比用常规杀虫剂更为合适。本研究评价了室内条件下在玻璃表面上硅藻土和多杀菌素对四纹豆象成虫的致死效应和亚致死效应。结果表明: 硅藻土处理24 h和48 h后, 对四纹豆象成虫的LC50 值分别为 1.47和0.2 g/m2; 多杀菌素处理24 h和48 h后, 对四纹豆象成虫的LC50 值分别为102.9 和68.8 mg ai/L, 说明两种化合物都对四纹豆象成虫具有较高的急性毒性。通过检测生物学参数, 研究了LC20浓度的硅藻土和多杀菌素对四纹豆象的亚致死效应。LC20浓度的硅藻土和多杀菌素使四纹豆象成虫的繁殖力分别比对照降低了71.5%和17.2%, 卵孵化率降低了57.5%和27.8%, 成虫寿命缩短了74.7%和17.1%。接触LC20浓度的硅藻土和多杀菌素使这一害虫的蛹期分别比对照延长了4.8%和2.3%。亚致死效应研究表明, 硅藻土和多杀菌素对四纹豆象的生命参数均产生了负面影响。总之, 致死效应和亚致死效应综合显示, 硅藻土在防治四纹豆象上具有较大的潜力。  相似文献

This research tested the suppressive ability of Beauveria bassiana (Balsamo) Vuillemin alone and in combination with diatomaceous earth against the red flour beetle, Tribolium castaneum (Herbst). Adults did not show a dose response to B. bassiana, and the addition of diatomaceous earth (DE) did not result in a significant increase in mortality. Against larvae, however, DE at 190 mg/kg grain enhanced the efficacy of B. bassiana at all concentrations ranging from 33 to 2,700 mg of conidia per kilogram of grain. The presence of DE resulted in 17- and 16-fold decreases in the median lethal concentration of B. bassiana at 56 and 75% RH, respectively. No significant differences in larval mortality in response to B. bassiana and diatomaceous earth alone or in combination were found between 56 and 75% RH. Conidial attachment to larvae was significantly greater with 190 mg/kg DE than without it. The partial analysis of lipids taken up by DE from the larvae revealed the removal of phospholipids and long-chain fatty acids. These results support the hypothesis that diatomaceous earth enhances the efficacy of B. bassiana against larval T. castaneum, at least in part by damaging the insect cuticle, thus increasing conidial attachment and making nutrients more available to conidia for their germination.  相似文献

A series of experiments was conducted to determine the effect of a flour food source on survival of red flour beetle, Tribolium castaneum (Herbst), and confused flour beetle, Tribolium confusum (DuVal), exposed to the labeled rate (0.5 mg/cm2) of Protect-It, a marine formulation of diatomaceous earth. Beetles were exposed at 27 degrees C, and 40, 57, and 75% RH in 62-cm2 petri dishes. When beetles were exposed for 1 or 2 d in dishes with the labeled rate (0.5 mg/cm2, or 31 mg per dish) of diatomaceous earth or in dishes containing flour at varying levels from 0 to 200 mg mixed with the labeled rate of diatomaceous earth, survival of both species increased as the amount of flour increased, and quickly plateaued at levels approaching 100%. In a second set of experiments, beetles were transferred to dishes containing flour at varying levels from 0 to 200 mg after they were exposed for 1 or 2 d in dishes with the labeled rate of diatomaceous earth alone. There were no significant differences in beetle survival among the levels of flour, however, survival in dishes with flour was usually greater than survival in dishes with diatomaceous earth alone. In a third test, beetles were exposed for 1, 2, and 3 d in dishes with either the labeled rate of diatomaceous earth alone (clean dishes), dishes with diatomaceous earth and empty straws, or dishes with diatomaceous earth and approximately 300 mg of flour packed in the straws. Survival was not significantly different between clean dishes or dishes with straws, but survival in dishes containing the straws with flour was usually 100%, regardless of exposure interval. In all experiments, confused flour beetles were less susceptible to diatomaceous earth than red flour beetles. In addition, survival was negatively related to exposure interval and positively related to relative humidity.  相似文献

Laboratory bioassays were carried out to evaluate the insecticidal effect of Beauveria bassiana (Balsamo) Vuillemin (Ascomycota: Hypocreales) germinated conidia, a commercial formulation of diatomaceous earth and a liquid formulation of spinosad, applied alone or in all possible combinations on wheat, against adults of the confused flour beetle, Tribolium confusum Jacquelin du Val (Coleoptera: Tenebrionidae). Diatomaceous earth was applied at one (200 ppm), the fungus at two (100 and 300 ppm) and spinosad at three (0.01, 0.1 and 0.5 ppm) dose rates, whereas all combinations of two or three of the insecticide concentrations were also tested. Insect mortality was determined after 2, 7 and 14 d of exposure. The simultaneous use of B. bassiana germinated conidia with diatomaceous earth notably increased mortality of T. confusum adults by sixfold 7 d after exposure, as compared with either B. bassiana or diatomaceous earth alone. When diatomaceous earth and spinosad were combined, diatomaceous earth did not increase the efficacy of spinosad. In general, the combination of all three agents did not differ from either the application of the agents tested alone, or their binary combinations, in most treatments examined.  相似文献

A method for the isolation of Echinococcus granulosus DNA from germinal layers of hydatid cysts is described. The method includes a hexadecyltrimethylammonium bromide/chloroform extraction and an adsorption to diatomaceous earth suspension. DNA suitable for polymerase chain reaction was obtained and used for parasite strain determination by mitochondrial cytochrome c oxidase I gene sequencing. Fertile and nonfertile cyst isolates from sheep, cattle, pigs, and humans were characterized. Hitherto, no direct parasite strain characterization has been made on nonfertile hydatid cysts, whereas here we report that nonfertile hydatid cysts were produced by sheep strain (G1 genotype) in sheep, cattle, and humans and by pig strain (G7 genotype) in pigs.  相似文献

This study, consisting of three experiments, was designed to assess whether diatomaceous earth, when applied to the surface of growing media, reduces adult fungus gnat Bradysia sp. nr. coprophila (Diptera: Sciaridae) emergence or inhibits the females from laying eggs; and whether fungus gnat adults are attracted to the fungus Trichoderma harzianum T-22 (Rifai strain KRL-AG2) under laboratory conditions. In the first two experiments, diatomaceous earth was applied at two different thicknesses (3.1 and 6.3 mm) and conditions (dry and moist) to the surface of a growing medium (Universal SB 300 Mix) after the growing medium had been artificially inoculated with second or third instars of fungus gnats, or before female fungus gnat adults were released into each deli squat container. In the third experiment, preparations of the fungus T. harzianum at the highest recommended label rate (0.889 kg/m3) were amended into the growing medium and processed 24, 48, or 72 h before use in a series of three two-choice trials with a two-armed experimental arena. In the first two experiments, the dry or moist layers of diatomaceous earth, in general, did not affect fungus gnats in terms of preventing adult emergence or egg laying by the females. During the course of these experiments, we observed that the diatomaceous earth dry treatments expanded as a result of absorbing moisture from the growing medium, creating fissures that allowed the fungus gnat larvae to pupate and females to lay eggs. In the third experiment, fungus gnat adults were not attracted to the T. harzianum treatments in any of the trials.  相似文献

Effects of diatomoceous earth on water content of Sitophilus granarius (L.) (Col., Curculionidae) and its possible use in stored product protection Increased problems caused by application of chemical pesticides within pest protection has led to the demand for non‐polluting, biologically specific active countermeasures. This study aimed to test the possible use of diatomaceous earth against Sitophilus granarius. It will also examine the physiological effects of amorphous diatomaceous earth against this insect. After 2 weeks, 100% mortality was observed of the adults treated with 2 g Fossil Shield® per kilogram wheat, at 11–12% corn moisture content and relative humidity of about 62 ± 2%. The rise in grain moisture and relative humidity decreased the effectiveness of the Fossil Shield®. It was observed, that the diatomaceous earth reduced the number of adult granary weevils and increased the mortality rate at development stages, but they did not completely prevent the hatching of the next generation. After contact with diatomaceous earth, the individuals of S. granarius had lost weight and reduced their water content, respectively. The adults significantly lost weight and body water after 1 day exposure to Fossil Shield®. This showed that silica dusts break the water barrier of the cuticle. However, it increases the transpiration rate of water across the cuticle and several insects died through dehydration. At higher relative humidity diatomaceous earth was less effective toward S. granarius, because there was a lower permeability for water across the cuticle. This delays or rather prevents the drying action of diatomaceous earths. Furthermore, the addition of food affected the effectiveness of dusts against S. granarius. A lot of stored product beetles are able to produce metabolic water in the feed substrates. This ability protects them and works contrary to the loss of water.  相似文献

Fourteen strains of Tribolium castaneum (Herbst) (Coleoptera: Tenebrionidae) had mortalities ranging from 5 to 100% when exposed to diatomaceous earth at 600 ppm for seven days. The most tolerant strain had a lethal dose for 50% of the population (LD50) of 413 ppm and the most susceptible strain had a LD50 of 238 ppm. Adults of the tolerant strain were lighter (2.0 mg) than the susceptible strain (2.6 mg). Tolerant adults lost water at lower rate (6 g h–1 than susceptible adults (12 g h–1), when held in wheat treated with 600 ppm diatomaceous earth for 24 h, than held at 5% r.h. with no food. Tolerant adults that were not exposed to diatomaceous earth lost water at a lower rate (3 g h–1) than susceptible adults (5 g h–1). Both strains, exposed and not exposed to diatomaceous earth died when their water content was between 33 and 37% of their total weight. Insects taken directly from the cultures had 52% (tolerant) and 53% (susceptible) of their total weight as water. Tolerant adults moved slower through grain and across filter paper than susceptible adults. Tolerant adults avoided wheat treated with diatomaceous earth at concentrations as low as 75 ppm, whereas the adults from the susceptible strain did not avoid diatomaceous earth, even at 600 ppm. The consequences of a strain tolerant to diatomaceous earth is discussed with respect to the use of diatomaceous earth to control stored-product insect infestations.  相似文献

Wolinella succinogenes HAP-1 is a Gram-negative microaerophile which reduces perchlorate to chloride by the proposed pathway ClO4 to ClO3 to ClO2 to Cl + O2. A cost-effective perchlorate treatment process has been established using a consortium of facultative anaerobic organisms and W. succinogenes HAP-1. The mixed anaerobic bacterial culture containing W. succinogenes HAP-1 was examined for the ability to form a biofilm capable of perchlorate reduction. An up-flow anaerobic fixed bed reactor (UAFBR) was packed with diatomaceous earth pellets and operated at residence times of 1.17 and 0.46 h to insure a viable biofilm had attached to the diatomaceous earth pellets. Reduction rates of perchlorate to chloride in the UAFBR could be maintained at 1 g of perchlorate reduced h−1 L−1. Studies with the same bacterial consortium in continuously stirred tank reactors (CSTR) generally reduced 0.5–0.7 g of perchlorate h−1. Viable cell counts were performed periodically on the diatomaceous earth pellets and demonstrated that the W. succinogenes HAP-1 population was maintained at 28–47% of the total microbial population. Scanning electron micrographs demonstrated that the external and internal surfaces of the diatomaceous pellets were densely colonized with microbial cells of multiple cell types. Recombinant hepatitis B surface antigen (r-HBsAg) produced in yeast is adsorbed on a diatomaceous earth matrix for purification purposes. A pH dependence in the adsorption-elution behavior was found. The capacity of celite (Hyflo Super Cei) for adsorbing r-HBsAg increased with decreasing pH. Nonspecific proteins were also adsorbed, but a low pH dependence was found. Elution from the matrix was performed using a basic pH buffer, in which r-HBsAg is more specifically adsorbed/desorbed than contaminant proteins, permitting the purification of the r-HBsAg. A pH of 4.0 was used for adsorption and pH 8.2 was used for desorption. The described protocol allows a purification factor between three- and fivefold with respect to contaminant proteins and sixfold with respect to contaminant DNA. Analysis of the diatomaceous earth collected from the Miocene type section in the Shanwang Basin of Shandong shows that the diatom assemblage there contains 16 genera, 95 species and varieties (of which 3 being new species), This fairly typical Miocene diatom assemblage may be correlated with the Miocene assemblage in Yakima district and Otis basin of Oregon, U.S.A. and those in W. Siberia, USSR. It is proved from the characteristics of the present diatom assemblage that the palaeoeeologieal environment during the diatom deposition was typically fresh lacus- trine facies, and the original lake surface was fairly extensive and the lake water was comparatively stable, but not very deep. The diatom assemblage contains species living now in more northern water bodies. This may be further proved by the associated grown cysts of Chrysophyta. The thin-bedded papery diatomaceous earth may be contributed partly to the fall of volcanic ashes, but most probably it is closely related to the seasonal palaeoclimatic changes as reflected in the growth repreduction stages of the diatoms.  相似文献
