目的研究PLAGL2(pleiomorphic adenoma genelike-2,PLAGL2)对SP-C基因(surfactant pro-tein—C,SP—C)启动子的结合与影响作用。方法定点诱变技术(site—directed mutagenesis)及凝胶电泳迁移实验(electrophoresis mobility shiftassay,EMSA)研究PLAGL2与SP—C基因启动子之间的相互结合作用;体外培养MLE12细胞共转染和荧光素酶报告基因活性值检测技术,研究PLAGL2对SP—C基因启动子表达活性的影响;结果EMSA及竞争性EMSA实验表明:PLAGL2能通过其核心和簇结合位点序列与同位素标记的SP—C基因启动子片段相结合形成蛋白-DNA复合物;细胞共转染及荧光素酶报告基因活性值检测结果显示:PLAGL2能明显促进SP—C基因启动子的表达。结论在肺Ⅱ型细胞中,PLAGL2能与SP—C基因启动子相互作用并促进其表达。 相似文献

锌指蛋白(zinc finger protein,ZNF/ZFP)是一类具有手指状结构域的转录因子,广泛存在于动、植物及微生物体内。ZNF在多种肿瘤组织中表达,通过调控下游靶基因的表达参与肿瘤的发生发展过程。ZNF表达异常与结直肠癌临床病理特征和预后密切相关。本文从结直肠癌的增殖、迁移和侵袭、凋亡、癌细胞干性等4种恶性行为角度,阐述了近年来锌指蛋白家族成员(ZEB1/2、Gli1/3、PLAGL1/2、KLF4、Snail1、ZBTB7A、ZBTB7C、ZBTB16)在结直肠癌中的作用机制,旨在为临床靶向治疗结直肠癌提供参考依据。 相似文献

作为大脑最基本和重要的功能之一,学习记忆始终是脑科学研究的热点领域。尽管相关研究已经取得了很大进展,但其具体机制尚不完全清楚。能产生促红细胞生成素的肝癌细胞(eryth-ropoietin-producing hepatocellular carcinoma cell,Eph)受体与其配体肝配蛋白被统称为Eph家族蛋白,其分布十分广泛且功能复杂。越来越多的研究表明,Eph家族蛋白能够调控包括突触发生及突触可塑性等多种细胞行为,在学习记忆中发挥不可或缺的作用。本文重点介绍Eph家族蛋白对学习记忆的作用并对其可能的作用机制进行概述。 相似文献

胞膜窖 (Caveolae) 是细胞膜内陷形成的一种特殊的脂筏结构,含有丰富的胆固醇和鞘磷脂,并在调节细胞信号转导中发挥重要作用。大量的研究显示caveolae相关蛋白窖蛋白 (Caveolins) 在维持caveolae的结构及其功能的调节中发挥重要作用。然而,最近研究发现caveolae的形成同样需要另一类蛋白家族cavins蛋白家族的参与。此外,cavins蛋白家族成员之一cavin-1能够与caveolins主要成员窖蛋白-1 (Caveolin-1) 相互作用参与调节caveolae的结构和功能。文中将就近年来cavins与caveolins的关系及其在调节caveolae中的作用进行综述。 相似文献

Wnt信号通路高度保守,对细胞命运决定、迁移、极性建立、原始体轴形成和器官的发生以及干细胞的自我更新等起着关键的作用。Dishevelled (DVL)作为第一个被发现的胞浆蛋白,是Wnt信号通路的核心组分,在Wnt信号传导通路中都起着重要作用。通过对不同物种(从单细胞动物到哺乳动物)基因组中DVL家族基因进行检索发现:DVL蛋白家族起源于早期的多细胞动物,与大堡礁海绵有共同的祖先,其成员可分为四个亚家族,分别是DVL1亚家族、DVL2亚家族、DVL3亚家族和无脊椎动物DVL亚家族,并发现DVL蛋白家族演化过程中存在基因复制现象;在演化历程中,DVL蛋白家族Motif的组成存在着规律性的变化,随着物种的演化Motif的数量有所增加;DVL家族基因存在着内含子的插入和缺失现象,其相位在高等动物中较为保守。本研究为进一步理解DVL蛋白家族的起源与演化动态提供一定的参考。 相似文献

The activation of the pleomorphic adenoma gene 1 (PLAG1) is the most frequent gain-of-function mutation found in pleomorphic adenomas of the salivary glands. To gain more insight into the regulation of PLAG1 function, we searched for PLAG1-interacting proteins. Using the yeast two-hybrid system, we identified karyopherin alpha2 as a PLAG1-interacting protein. Physical interaction between PLAG1 and karyopherin alpha2 was confirmed by an in vitro glutathione S-transferase pull-down assay. Karyopherin alpha2 escorts proteins into the nucleus via interaction with a nuclear localization sequence (NLS) composed of short stretches of basic amino acids. Two putative NLSs were identified in PLAG1. The predicted NLS1 (KRKR) was essential for physical interaction with karyopherin alpha2 in glutathione S-transferase pull-down assay, and its mutation resulted in decreased nuclear import of PLAG1. Moreover, NLS1 was able to drive the nuclear import of the cytoplasmic protein beta-galactosidase. In contrast, predicted NLS2 of PLAG1 (KPRK) was not involved in karyopherin alpha2 binding nor in its nuclear import. The residual nuclear import of PLAG1 after mutation of the NLS1 was assigned to the zinc finger domain of PLAG1. These observations indicate that the nuclear import of PLAG1 is governed by its zinc finger domain and by NLS1, a karyopherin alpha2 recognition site. 相似文献

The PLAGL1 gene encodes a homonymous zinc finger protein that promotes cell cycle arrest and apoptosis through multiple pathways. The protein has been implicated in metabolic, genetic, and neoplastic illnesses, but the molecular mechanisms by which the protein PLAGL1 participates in such diverse processes remains to be elucidated.
