The present study was undertaken to find out whether or not sexual dimorphism in biological activities and amino acid compositions of mouse prolactin might be due to heterogeneity in mRNA for mouse prolactin Cloned cDNAs for mouse prolactin were first isolated from a mouse pituitary cDNA library by hybridization with a rat prolactin cDNA. Then, one clone of about 140 positive clones obtained from 2000 transformants was subjected to nucleotide sequence analysis and verified to contain a nearly full length of cDNA sequence coding for mouse prolactin precursor. The deduced complete amino acid sequence indicates that the precursor molecule consists of 31 amino acids as the signal peptide and 197 amino acids of prolactin, in which two amino acids were found to be different from the amino acid sequence previously published elsewhere. S1 nuclease mapping analysis using male and female pituitary RNAs indicates that mouse preprolactin is encoded by two mRNAs in both sexes. The two mRNAs differ from each other based upon the deletion of three nucleotides in the coding region for the signal peptide determined by the nucleotide sequence analysis in other cDNA clones. In the present study, no sexual difference was revealed in murine prolactin mRNA. 相似文献

利用RACE技术获得了冠突散囊菌stf1基因的全长DNA序列。该序列全长3 029bp,开放阅读框长度为2 664bp,从114–2 908bp,在253bp处含有一个131bp的内含子,预测编码887个氨基酸。同源分析结果表明该基因与Snd1/p100转录因子同源。应用相对定量SYBR Green I荧光定量PCR技术对stf1基因在不同发育时期的表达量的差异进行了检测。结果表明,这个基因在子囊孢子时期的表达量最高,在分生孢子时期的表达量最低,相比子囊孢子时期下降了1倍。为深入研究冠突散囊菌的产孢调控机制奠定了一定的基础。 相似文献

DNA complementary to the rat hypothalamic mRNA coding for the corticotropin-releasing factor precursor (prepro-CRF) has been cloned by screening a cDNA library with a human genomic DNA probe. Nucleotide sequence analysis of the cloned cDNA has revealed that rat prepro-CRF consists of 187 amino acid residues including a putative signal peptide. The CRF and putative signal peptide regions are more highly conserved among rat, human and ovine prepro-CRF than is the cryptic portion. 相似文献

We screened a gene library constructed with human leukocyte chromosomal DNA fragments with an oligonucleotide probe complementary to the middle part of the cDNA for the human epidermal growth factor receptor. One of the genomic DNA clones obtained contains an insert of 36 kilobase pairs encoding 54% of the mature receptor protein. From the sequencing experiments, we were able to locate the exact site of the gene rearrangement, which had been previously suggested in some epidermoid carcinoma cell lines, in the intron preceding the exon coding for the hydrophobic transmembrane domain. Such a rearrangement must result in the removal of an authentic splicing acceptor site and the separation of the 3'-segment of the receptor gene which could encode a polypeptide homologous to the v-erb-B oncogene product responsible for chicken carcinogenesis. 相似文献

Metallothionein mRNA was purified from liver of mice injected with cadmium. The corresponding double-stranded cDNA was prepared and inserted into the PstI site of plasmid pBR322. The resulting recombinant DNA was used to transform the RR1 strain of . Clones resistant to tetracycline and sensitive to ampicillin were screened for the presence of metallothionein-specific restriction fragments in their plasmids. One plasmid, called M135, contains a cDNA insert covering the entire length of the mRNA for mouse liver metallothionein-I, except for the first 18 bases at the 5′ end. 相似文献
