The distribution of NO3? reduction between roots and shoots was studied in hydro-ponically-grown peach-tree seedlings (Prunus persica L.) during recovery from N starvation. Uptake, translocation and reduction of NO3?, together with transport through xylem and phloem of the newly reduced N were estimated, using 15N labellings, in intact plants supplied for 90 h with 0.5 mM NH4+ and 0.5, 1.5 or 10 mM NO3?. Xylem transport of NO3? was further investigated by xylem sap analysis in a similar experiment. The roots were the main site of NO3? reduction at all 3 levels of NO3? nutrition. However, the contribution of the shoots to the whole plant NO3? reduction increased with increasing external NO3? availability. This contribution was estimated to be 20, 23 and 42% of the total assimilation at 0.5, 1.5 and 10 mM NO3?, respectively. Both 15N results and xylem sap analysis confirmed that this trend was due to an enhancement of NO3? translocation from roots to shoots. It is proposed that the lack of NO3? export to the shoots at low NO3? uptake rate resulted from a competition between NO3? reduction in the root epidermis/cortex and NO3? diffusion to the stele. On the other hand, net xylem transport of newly reduced N was very efficient since ca 70% of the amino acids synthesized in the roots were translocated to the shoots, regardless of the level of NO3? nutrition. This net xylem transport by far exceeded the net downward phloem transport of the reduced N assimilated in shoots. As a consequence, the reduced N resulting from NO3? assimilation, principally occurring in the roots, was mainly incorporated in the shoots.  相似文献

Picea sitchensis (Bong.) Carr. seedlings were exposed to SO2, NO2 and SO2+ NO2 during dormancy in controlled environments, and were taken to night temperatures of 4, 0, −5, −10 and −15 °C in a freezer. Conditions in the freezer were carefully monitored during the low–temperature treatments. In two experiments, different photoenvironments and temperature regimes were imposed prior to the cold treatments, and different effects were observed. In the first, only limited frost hardiness was achieved and night temperatures of −15 °C were lethal. Temperatures of −5 and − 10 °C led to poor survival of lateral buds, particularly in plants exposed to 45 ppb SO2. The poor bud break in plants exposed to SO2 and to − 5 °C resulted in a loss of the effectiveness of this temperature as a chill requirement. Pressure-volume analysis showed that the shoots of plants exposed to NO2 had greater elasticity (lower elastic moduli, e), so that loss of turgor occurred at lower relative water contents. In contrast, a hardening period (2 weeks in night/day temperatures of 3/10 °C and 8 h days at 50 μmol m−2 s−1 PAR) gave decreased elasticity and lower solute potentials of spruce shoots. In the second experiment, exposure to 30 ppb SO2 and SO2+ NO2 led to slight, but consistent, increases in frost injury to the needles of plants frozen to − 5 and − 10 °C. The results suggest that the main interaction of low temperatures and winter pollutants may be on bud survival rather than on needle damage, but that effects are subtle, only occurring with certain combinations of pollutant dose and cold treatment.  相似文献

The physiological consequences for NO3 utilization by the plant of underexpression and overexpression of nitrate reductase (NR) were investigated in nine transformants of Nicotiana tabacum and Nicotiana plumbaginifolia. The in vitro NR activities (NRAs) in both roots and leaves of low- and high-NR tobacco transformants ranged from 5–10% to 150–200%, respectively, of those measured in wild-type plants. The level of NR expression markedly affected the NO3 reduction efficiency in detached leaves and intact plants. In both species, 15NO3 reduction ranged from 15–45% of 15NO3 uptake in the low-NR plants, to 40–80% in the wild-type, and up to 95% in high-NR plants. In the high-NR genotypes, however, total 15NO3 assimilation was not significantly increased when compared with that in wild-type plants, because the higher 15NO3 reduction efficiency was offset by lower 15NO3 uptake by the roots. The inhibition of NO3 uptake appeared to be the result of negative feedback regulation of NO3 influx, and is interpreted as an adjustment of NO3 uptake to prevent excessive amino acid synthesis. In genotypes underexpressing NR, the low 15NO3 reduction efficiency also was generally associated with a decrease in net 15NO3 uptake as compared with the wild type. Thus, underexpression of NR resulted in an inhibition of reduced 15N synthesis in the plant, although the effect was much less pronounced than that expected from the very low NRAs. The restricted NO3 uptake in low-NR plants emphasizes the point that the products of NO3 assimilation are not the only factors responsible for down-regulation of the NO3 uptake system.  相似文献

In the present study, we investigated whether growth and main nutrient ion concentrations of cabbage (Brassica campestris L.) could be increased when plants were subjected to different NH4^+/NO3- ratios. Cabbage seedlings were grown in a greenhouse in nutrient solutions with five NH4^+/NO3- ratios (1:0; 0.75:0.25; 0.5:0.5; 0.25:0.75; and 0:1). The results showed that cabbage growth was reduced by 87% when the proportion of NH4^+-N in the nutrient solution was more than 75% compared with a ratio NH4^+/NO3- of 0.5:0.5 35 d after transplanting, suggesting a possible toxicity due to the accumulation of a large amount of free ammonia in the leaves. When the NH4+/NO3- ratio was 0.5:0.5, fresh seedling weight, root length, and H2PO4- (P), K^+, Ca^2+, and Mg^2+ concentrations were all higher than those in plants grown under other NH4^+/NO3- ratios. The nitrate concentration in the leaves was the lowest in plants grown at 0.5: 0.5 NH4^+/NO3-. The present results indicate that an appropriate NH4^+/NO3- ratio improves the absorption of other nutrients and maintains a suitable proportion of N assimilation and storage that should benefit plant growth and the quality of cabbage as a vegetable.  相似文献

The (C2H4+ H2(C2H2))/15N2 ratios of 15 clover- Rhizobium symbionts. soybean, and black medick symbionts were measured. Relative efficiency based on the C2H4 production and on 15N2 incorporation were compared, and in most symbionts there was little difference between the two measures of relative efficiency. Total measurable electron flux through nitrogenase during acetylene reduction and 15N2 incorporation were nearly equal for most symbionts studied. The relative efficiency and the (C2H4+ H2(C2H2))/15N2 ratio showed an inverse correlation. Use of this ratio appears preferable to use of the ratio of C2H2 reduction/N2 reduction. Some evolution of H2 was observed in the presence of C2H2.  相似文献

The quantitative relationship between C2H2 reduction, H2 evolution and 15N2 fixation was investigated in excised root nodules from pea plants ( Pisum sativum L. cv. Bodil) grown under controlled conditions. The C2H2/N2 conversion factor varied from 3.31 to 5.12 between the 32nd and the 67th day after planting. After correction for H2 evolution in air, the factor (C2H2-H2)/N2 decreased to values near the theoretical value 3, or in one case to a value significantly ( P < 0.05) below 3. The proportion of the total electron flow through nitrogenase, which is not wasted in H2 production but used for N2 reduction, is often stated as the relative efficiency (1-H2/C2H2). This factor varied significantly ( P < 0.05) during the growth period. The actual allocation of electrons to H2 and N2, expressed as the H2/N2 ratio, was independent of plant age, however. This discrepancy and the observation that the (C2H2-H2)/N2 conversion factor tended to be lower than 3, suggests that the C2H2reduction assay underestimates the total electron flow through nitrogenase.  相似文献

The immediate effects of tillage on protected soil C and N pools and on trace gas emissions from soils at precultivation levels of native C remain largely unknown. We measured the response to cultivation of CO2 and N2O emissions and associated environmental factors in a previously uncultivated U.S. Midwest Alfisol with C concentrations that were indistinguishable from those in adjacent late successional forests on the same soil type (3.2%). Within 2 days of initial cultivation in 2002, tillage significantly (P=0.001, n=4) increased CO2 fluxes from 91 to 196 mg CO2‐C m?2 h?1 and within the first 30 days higher fluxes because of cultivation were responsible for losses of 85 g CO2‐C m?2. Additional daily C losses were sustained during a second and third year of cultivation of the same plots at rates of 1.9 and 1.0 g C m?2 day?1, respectively. Associated with the CO2 responses were increased soil temperature, substantially reduced soil aggregate size (mean weight diameter decreased 35% within 60 days), and a reduction in the proportion of intraaggregate, physically protected light fraction organic matter. Nitrous oxide fluxes in cultivated plots increased 7.7‐fold in 2002, 3.1‐fold in 2003, and 6.7‐fold in 2004 and were associated with increased soil NO3? concentrations, which approached 15 μg N g?1. Decreased plant N uptake immediately after tillage, plus increased mineralization rates and fivefold greater nitrifier enzyme activity, likely contributed to increased NO3? concentrations. Our results demonstrate that initial cultivation of a soil at precultivation levels of native soil C immediately destabilizes physical and microbial processes related to C and N retention in soils and accelerates trace gas fluxes. Policies designed to promote long‐term C sequestration may thus need to protect soils from even occasional cultivation in order to preserve sequestered C.  相似文献

The present study aims at clarifying the impact of oxidative stress on type B trichothecene production. The responses to hydrogen peroxide (H2O2) of an array of Fusarium graminearum and Fusarium culmorum strains were compared, both species carrying either the chemotype deoxynivalenol (DON) or nivalenol (NIV). In both cases, levels of in vitro toxin production are greatly influenced by the oxidative parameters of the medium. A 0.5 mM H2O2 stress induces a two- to 50-fold enhancement of DON and acetyldeoxynivalenol production, whereas the same treatment results in a 2.4- to sevenfold decrease in NIV and fusarenone X accumulation. Different effects of oxidative stress on toxin production are the result of a variation in Fusarium 's antioxidant defence responses according to the chemotype of the isolate. Compared with DON strains, NIV isolates have a higher H2O2-destroying capacity, which partially results from a significant enhancement of catalase activity induced by peroxide stress. A 0.5 mM H2O2 treatment leads to a 1.3- to 1.7-fold increase in the catalase activity of NIV isolates. Our data, which show the higher adaptation to oxidative stress developed by NIV isolates, are consistent with the higher virulence of these Fusarium strains on maize compared with DON isolates.  相似文献

Abstract: NH4+‐grown plants are more sensitive to light stress than NO3?‐grown plants, as indicated by reduced growth and intervenal chlorosis of French bean (Phaseolus vulgaris L.). Measuring the time course of Fv/Fm ratios under photoinhibitory light regimes did not reveal any difference in PS II damage between NO3?‐ and NH4+‐grown plants, in spite of some indications of higher energy quenching in NO3?‐grown plants. Also, a direct action of NH4+ as an uncoupler at the thylakoid membrane could be excluded. Instead, biochemical analysis revealed enhanced lipid peroxidation and higher activity of scavenging enzymes in NH4+‐grown plants indicating that these plants make use of metabolic pathways with stronger radical formation. Evidence for higher rates of photorespiration in NH4+‐grown plants came from experiments showing that electron flux and O2 evolution were decreased by SHAM in NH4+‐grown plants, and by antimycin A in NO3?‐grown plants. Further, the comparison of electron flux and of photoacoustic measurements of O2 evolution suggested that in NH4+‐grown plants the Mehler reaction was also increased, at least in the induction phase. However, the major cause of N form‐dependent stress sensitivity is assumed to be in the coupling between photosynthesis and respiration, i.e., NO3?‐grown plants can utilize the TCA cycle for the generation of C skeletons for amino acid synthesis, thus improving the ATP: reductant balance, whereas NH4+‐grown plants have enhanced rates of photorespiration.  相似文献
