The effects of some selected arbuscular mycorrhizal (AM) fungi, Gigaspora margarita and Glomus mossae on the growth and the role of soluble amino acids of two contrasting cocoa cultivars (ICS84 tolerant and SNK10 sensitive) against black pod disease caused by Phytophthora megakarya were investigated. Root colonization by AM fungi is between 50 and 70% 18 weeks after planting. Tested AM fungi significantly increased all the plant growth parameters (height, number of leaves, shoot and root matter) and P uptake as compared to non‐inoculated plants in pot experiments. AM fungi inoculated cocoa reduced the disease severity. Compared to the control, the soluble amino acid levels increased with inoculation of the AM fungi strains in the necrotic stems of disease on inoculated cocoa plants. Significant relationships between amino acids and disease severity observed for two cocoa cultivars imply that the induction of specific amino acids synthesized by leaves, such as arginine, cysteine and glutamic acid, may represent potential candidate molecules for adaptation of such cultivars to P. megakarya disease. Inoculating seedlings with AMF in nurseries could enhance the development of cocoa plants protected against P. megakarya.  相似文献

The present study was undertaken to assess the benefit and compare the functioning of AM fungi on wheat grown conventionally and on beds. Ten treatment combinations were used, treatments 1 and 2: no fertilizers with and without arbuscular mycorrhizal (AM) fungi (In vitro produced Glomus intraradices); 3:100% of recommended NPK: (120 kg ha−1 N; 60 kg ha−1 P; 50 kg ha−1 K), and 4 and 5: 75% of recommended NPK dose with and without AM inoculation in a 5 × 2 split-plot design on wheat using conventional/flat system and elevated/raised bed system. The maximum grain yield (3.84 t ha−1) was obtained in AM fungi inoculated plots of raised bed system applied with 75% NPK and was found higher (although non- significant) than the conventional (3.73 t ha−1) system. The AM inoculation at 75% fertilizer application can save 8.47, 5.38 kg P and 16.95, 10.75 kg N ha−1, respectively, in bed and conventional system. While comparing the yield response with 100% fertilizer application alone, AM inoculation was found to save 20.30, 15.79 kg P and 40.60, 31.59 kg N ha−1, respectively, in beds and conventional system. Mycorrhizal inoculation at 75% NPK application particularly in raised bed system seems to be more efficient in saving fertilizer inputs and utilizing P for producing higher yield and growth unlike non-mycorrhizal plants of 100% P. Besides the yield, mycorrhizal plants grown on beds had higher AM root colonization, soil dehydrogenases activity, and P-uptake. The present study indicates that the inoculation of AM fungi to wheat under raised beds is better response (although non-significantly higher) to conventional system and could be adopted for achieving higher yield of wheat at reduced fertilizer inputs after field validation.  相似文献

在黄土高原的陕西省甘泉、绥德、米脂、榆林4县(市)选取4个样地,研究紫穗槐不同深度土层丛枝菌根真菌(Arbuscular mycorrhizal fungi,AMF)侵染率、孢子密度、球囊霉素和土壤因子之间的关系。结果表明:(1)AMF侵染率平均值绥德样地最大(100%),米脂样地最小(75.02%);孢子密度平均值米脂样地最大(5.91个/g),榆林样地最小(1.57个/g);4个样地孢子密度最大值均在0~10cm土层,且随土层加深而降低;AMF侵染率与孢子密度在各样地间差异显著,同一样地侵染率与孢子密度变化规律不一致。(2)榆林样地的有机碳、铵态氮、速效磷、脲酶和碱性磷酸酶活性平均值显著高于其他3个样地;除绥德、米脂样地的蔗糖酶和甘泉样地的碱性磷酸酶外,其他样地的土壤酶活性和球囊霉素含量均随土层加深而降低,且各土层之间差异显著,最大值均在0~10cm土层。(3)AMF侵染率与pH、总球囊霉素含量、易提取球囊霉素含量呈极显著正相关,与有机碳、硝态氮、速效磷含量呈显著正相关;孢子密度与速效磷、蔗糖酶、脲酶活性呈显著正相关,与铵态氮含量呈显著负相关。(4)第1、2主成分的累积方差贡献率达到56.4%,第1主成分主要综合了球囊霉素、AMF侵染率和孢子密度,第2主成分综合了pH和部分土壤因子信息。因此,球囊霉素、AMF侵染率、孢子密度、pH、部分土壤因子对决定土壤生态起主要作用。  相似文献

Biochemical analyses were made on anthers and pistils at various developmental stages of both male-sterne and fertile plants of Taigu wheat. Analyses ineluded total free amino aeids and free proline. The following results were obtained: 1. There was no significant difference between the content of free proline in anthers of male-sterile and fertile plants at reduction division of mierospore mother cells. 2. In anthers with early uninucleate miorospores, the content of free proline of fertile plants was remarkably higher than that of male-sterile plants. It is interesting to note that at this stage the content of free proline in fertile plants rose to 1.65% of the dry weight of the anther, constituting 50% of the total free amino acids, and amounted to 7-fold of that in male-sterile plants. This result is in line with the results obtained with most cytoplasmic malesterile plants reported by other workers, although malesterility in Taigu wheat is controlled by the nueleus. 3. In pistils, at the stages eorresponding to the early uninneleate and the binueleate stages of the pollen, the free proline content of fertile plants was twice as much as that of the male-sterile plants. This differenee disappeared gradually after fertilization. 4. Tile content of total free amino aoids did not fluetuate as much as the free proline content. There was no differenee in anthers of both types of plants during reduction division of mierospore mother cells. In anthers with early uninueleate pollen grains, total free amino acid content of fertile plants exceeded that of male-sterile plant, the difference levelled off at latter stages. In pistils, before fertilization, the content of total free amino acids of the fertile phmts was slightly higher than that of the male-sterile plants. We evaluated the roles of arbuscular mycorrhizal (AM) fungi in growth and phosphorus (P) nutrition of wheat (Triticum aestivum L.) in a highly calcareous soil and compared the responses of wheat with those of clover (Trifolium subterraneum L). In the first experiment wheat (cv. Brookton) was harvested at 6 wk. Colonisation by four AM fungi was low (<20%). Clover was harvested at 8 wk. Colonisation varied with different fungi, with the highest value (52%) obtained with Glomus intraradices. Although suffering from P deficiency, non-mycorrhizal (NM) wheat grew relatively well with no added P (P0) and application of P at 100 mg kg−1 (P100) increased the dry weight (DW). Shoot P concentrations increased with P application and there were positive effects of all AM fungi at P100. In contrast, NM clover grew very poorly at P0 and did not respond to P application. Clover responded positively to all AM fungi at both P levels, associated with increases in P uptake. In the second experiment colonisation by a single AM fungus (G. intraradices) of two wheat cultivars (Brookton and Krichauff) was well established at 6 wk (~50% in P0 plants) and continued to increase up to maturity (~70%), but decreased greatly at both harvests as P supply was increased (up to 150 mg P kg−1: P150). Addition of P significantly increased plant growth, grain yield and P uptake irrespective of cultivar and harvest time, and the optimum soil P for grain yield was P100. In both cultivars, a growth depression in AM plants occurred at 6 wk at all P levels, but disappeared at 19 wk with added P. At P0, AM plants also produced lower grain yield (weight) per plant, but with higher P, AM plants produced higher grain yields than NM plants. There was a significant positive effect of AM on grain P concentration at P0, but not at other P levels. Brookton was somewhat more P efficient than Krichauff, and the latter responded more to AM fungi. This study showed that responses of wheat to AM inoculation and P supply were quite different from those of clover, and changed during development. Results are discussed in relation to the underlying soil properties.  相似文献

This work investigates the effects of abscisic acid (ABA) on physiologic parameters related to yield in wheat (Triticum aestivum) grown under field conditions with water restriction ranging between 45.7% and 49.5% of field capacity during anthesis and postanthesis. ABA (300 mg L−1) was sprayed onto the plants at the beginning of shoot lengthening which significantly promoted leaf area and higher concentrations of chlorophylls and carotenoids in flag leaf at anthesis. ABA also increased soluble carbohydrates in shoots at anthesis, which were then re-exported to the grains at maturity. This correlated with a yield increase that was achieved by a higher number and weight of grains per spike, but protein content was not significantly affected.  相似文献

Soil and crop management practices may influence biomass growth and yields of cotton (Gossypium hirsutum L.) and sorghum (Sorghum bicolorL.) and sequester significant amount of atmospheric CO2in plant biomass and underlying soil, thereby helping to mitigate the undesirable effects of global warming. This study examined the effects of three tillage practices [no-till (NT), strip till (ST), and chisel till (CT)], four cover crops [legume (hairy vetch) (Vicia villosa roth), nonlegume (rye) (Secale cerealeL), hairy vetch/rye mixture, and winter weeds orno covercrop], and three N fertilization rates (0, 60–65, and 120–130 kg N ha –1) on the amount of C sequestered in cotton lint (lint + seed), sorghum grain, their stalks (stems + leaves) and roots, and underlying soil from 2000 to 2002 in central Georgia, USA. A field experiment was conducted on a Dothan sandy loam (fine-loamy, kaolinitic, thermic, Plinthic Kandiudults). In 2000, C accumulation in cotton lint was greater in NT with rye or vetch/rye mixture but in stalks, it was greater in ST with vetch or vetch/rye mixture than in CT with or without cover crops. Similarly, C accumulation in lint was greater in NT with 60 kg N ha –1 but in stalks, it was greater in ST with 60 and 120 kg N ha –1 than in CT with 0 kg N ha –1. In 2001, C accumulation in sorghum grains and stalks was greater in vetch and vetch/rye mixture with or without N rate than in rye without N rate. In 2002, C accumulation in cotton lint was greater in CT with or without N rate but in stalks, it was greater in ST with 60 and 120 kg N ha –1 than in NT with or without N rate. Total C accumulation in the above- and belowground biomass in cotton ranged from 1.7 to 5.6 Mg ha –1 and in sorghum ranged from 3.4 to 7.2 Mg ha –1. Carbon accumulation in cotton and sorghum roots ranged from 1 to 14% of the total C accumulation in above- and belowground biomass. In NT, soil organic C at 0–10 cm depth was greater in vetch with 0 kg N ha –1 or in vetch/rye with 120–130 kg N ha –1 than in weeds with 0 and 60 kg N ha –1 but at 10–30 cm, it was greater in rye with 120–130 kg N ha –1 than in weeds with or without rate. In ST, soil organic C at 0–10 cm was greater in rye with 120–130 kg N ha –1 than in rye, vetch, vetch/rye and weeds with 0 and 60 kg N ha –1. Soil organic C at 0–10 and 10–30 cm was also greater in NT and ST than in CT. Since 5 to 24% of C accumulation in lint and grain were harvested, C sequestered in cotton and sorghum stalks and roots can be significant in the terrestrial ecosystem and can significantly increase C storage in the soil if these residues are left after lint or grain harvest, thereby helping to mitigate the effects of global warming. Conservation tillage, such as ST, with hairy vetch/rye mixture cover crops and 60–65 kg N ha –1 can sustain C accumulation in cotton lint and sorghum grain and increase C storage in the surface soil due to increased C input from crop residues and their reduced incorporation into the soil compared with conventional tillage, such as CT, with no cover crop and N fertilization, thereby maintaining crop yields, improving soil quality, and reducing erosion.  相似文献

The aim of this study was to analyze the performance of Acaena elongata colonized by arbuscular mycorrhizal fungi (AMF) to different phosphorus (P) concentrations, as a measure of AMF dependency. A. elongata, is a species from soils where P availability is limited, such as temperate forests. Our research questions were: 1) How do different P concentrations affect the AMF association in Acaena elongata, and 2) How does the AMF association influence A. elongata’s growth under different P concentrations? A. elongata’s growth, P content in plant tissue, AMF colonization and dependency were measured under four P concentrations: control (0 g P kg−1 ), low (0.05 g P kg−1 ), intermediate (0.2 g P kg−1 ) and high (2 g P kg−1 ) in different harvests. A complete randomized block design was applied. A. elongata’s growth was higher under -AMF in intermediate and high P concentrations, and the lowest growth corresponded to +AMF in the low and intermediate P concentration. We observed a negative effect on the root biomass under +AMF in intermediate P concentration, while the P concentration had a positive effect on the leaf area ratio. The AMF colonization in A. elongata decreased in the highest P concentration and it was favored under intermediate P concentration; while the low and the high concentrations generated a cost-benefit imbalance. Our results suggest that the performance of some plant species in soils with low P availability may not be favored by their association with AMF, but a synergy between AMF and intermediate P concentrations might drive A. elongata’s growth.  相似文献

The free amino acid, protein, water and dry matter contents were determined during the seed development of Araucaria angustifolia. Soluble and insoluble proteins in the mature seed represent 4.2 % of the fresh matter. The embryonic axis stored the greatest amount of soluble proteins, while cotyledons both with the embryonic axis showed the largest quantities of insoluble proteins in the mature seed. The greatest concentration of free amino acids was detected during the stage when cotyledons start to develop. Glutamic acid, aspartic acid, alanine and serine were predominant in the whole seed while arginine, lysine and γ-aminobutyric acid were present in great amounts only in cotyledons and embryonic axis. Although megagametophyte was important as a source of free amino acids, it was not the major protein storage organ in the mature seed. In the embryogenetic process, the rise of cotyledons is closely related to physiological and biochemical changes. The distribution of an arbuscular mycorrhizal (AM) fungus between soil and roots, and between mycelial and storage structures, was studied by use of the fatty acid signature 16:1(omega)5. Increasing the soil phosphorus level resulted in a decrease in the level of the fatty acid 16:1(omega)5 in the soil and roots. A similar decrease was detected by microscopic measurements of root colonization and of the length of AM fungal hyphae in the soil. The fatty acid 16:1(omega)5 was estimated from two types of lipids, phospholipids and neutral lipids, which mainly represent membrane lipids and storage lipids, respectively. The numbers of spores of the AM fungus formed in the soil correlated most closely with neutral lipid fatty acid 16:1(omega)5, whereas the hyphal length in the soil correlated most closely with phospholipid fatty acid 16:1(omega)5. The fungal neutral lipid/phospholipid ratio in the extraradical mycelium was positively correlated with the level of root infection and thus decreased with increasing applications of P. The neutral lipid/phospholipid ratio indicated that at high P levels, less carbon was allocated to storage structures. At all levels of P applied, the major part of the AM fungus was found to be present outside the roots, as estimated from phospholipid fatty acid 16:1(omega)5. The ratio of extraradical biomass/intraradical biomass was not affected by the application of P, except for a decrease at the highest level of P applied.  相似文献

The growth response of Acacia mangium Willd. to inoculation with selected Bradyrhizobium strains was investigated in two field trials in the Ivory Coast (West Africa). In the first trial (Anguededou), four provenances (i.e., trees originating from seeds harvested in different geographical areas) of A. mangium were inoculated with four Bradyrhizobium strains from different origins. Six months after being transplanted in the field, the heights of all inoculated trees showed a statistically significant increase of 9 to 26% compared with those of uninoculated trees, with the most effective strain being Aust 13c. After 19 months, the positive effect of inoculation on tree growth was confirmed. The effect of A. mangium provenance on tree growth was also highly significant. Trees from the Oriomo provenance of Papua New Guinea had a mean height that was 25% greater than those of other provenances. Analysis of variance showed a highly significant effect of interaction between strain and host provenance factors. Thus, most effective strain × provenance combinations could be proposed. Immunological identification of strains clearly showed that 90 to 100% of nodules from trees inoculated with three of the four Bradyrhizobium strains or from uninoculated trees contained exclusively Aust 13c 23 months after tree transplantation. This predominance of Aust 13c in nodules was still observed 42 months after tree transplantation. The second experiment (Port-Bouët), performed with a different soil, confirmed the long-term positive effect of Aust 13c on plant growth, its high competitive ability against indigenous strains, and its persistence in soil. Strain Aust 13c should thus be of great interest for inoculating A. mangium under a wide range of field conditions.  相似文献

Free amino acids and S-methylmethionine (MMS, an anti-ulcer factor, Vitamin U) in green tea extracts were simultaneously determined with an HPLC-amino acid analyzer using lithium citrate buffers.

Rapid analysis of MMS and free amino acids was achieved with a high resolution column, MCI Gel CK-10U 0.46 </> × 15 cm, within 180 min. MMS in the column eluates was identified as dimethyl sulfide by a gas-chromatographic method with a flame photometric detector. The contents of MMS and free amino acids in various green teas were determined and compared with respect to the quality of commercial teas, blended teas and non-blended teas, and the place of cultivation of the tea. The described method is rapid and useful for checking the quality of green tea.  相似文献

Two methods were developed and used to study the root system dynamics of two species grown together or separately under field conditions. The first method, based on herbicide injection at different soil depths, was used to determine the rooting depth penetration rate of each species in pea–barley and pea–mustard mixtures. The roots absorbed the herbicide when they reached the treated zone leading to visible symptoms on the leaves which could be readily monitored. The second method used differences in 15N natural abundance and N concentration between legume and non-legume species to quantify the contribution of each species to root biomass of a pea–barley mixture. Each contribution was calculated using 15N abundance and N concentration of root mixtures and of subsamples of roots of individual species within mixtures. Both methods can indeed be used to distinguish roots of species in mixtures and thus to study belowground competition between associated species. The use of these methods demonstrated species differences in root system dynamics between species but also significant effects of interactions between species in mixtures. The rooting depth penetration rate was mainly species specific whereas root biomass was dependant on plant growth, allocation of dry matter between shoot and root components and growth factors such as N fertilization. Root biomass of each species may vary therefore with the level of competition between species.  相似文献
