Distribution and ontogeny of digestive enzymes in larval yellowtail and winter flounder 总被引:5,自引:0,他引:5
The distribution of digestive enzymes was studied using histoenzymological methods in yellowtail and winter flounder larvae reared on three different diets: live food, weaned at day 15, or starved. Alkaline phosphatase, dipeptidyl peptidase IV, aminopeptidase M and esterase were present at 3 days post-hatch and became differentially distributed coinciding with morphological development. For larvae fed a live diet, activity of these enzymes was present in the intestine of both species and rectum of yellowtail flounder. Alkaline phosphatase was also present in the post-oesophageal swelling (stomach anlage) of winter flounder. Activity of all enzymes was absent in starved winter flounder larvae and a decrease in aminopeptidase M and esterase activity occurred in starved yellowtail flounder larvae. Acid phosphatase was not identified in either species. The eVect of weaning on enzymatic activity was not evaluated fully as the larvae did not survive long enough after the introduction of the artificial diet to complete experimentation. 相似文献

The white sea bream, Diplodus sargus (Teleostei, Sparidae), is a species with a high commercial importance in Mediterranean aquaculture. There is currently little information available about the genetic characteristics of cultured populations. In this survey, we have developed eight polymorphic microsatellites for the white sea bream using an enriched genome library protocol. All of them were polymorphic in the 67 individuals tested, 32 of which were wild specimens, and 35 were individuals from a captive F(1) broodstock. These markers can potentially be useful tools for use in population genetic studies. 相似文献

The life history of the white sea bream Diplodus sargus in the Azores showed a pattern consistent with digynic hermaphroditism achieving sexual maturity during the second year of life, at 16·7 cm L T . Spawning occurred from March to June at temperatures between 15 and 17° C and the onset and duration of spawning season in the sea bream appeared to be influenced by sea water temperatures. As latitude decreased, both in the northern and southern hemispheres, the spawning season of D. sargus populations started earlier and extended longer, highlighting the potential importance of temperature to the onset and duration of reproduction in this species. Settlement took place from late May to July, and settlers remained in the nursery area for c . 2·5 months. Emigration from the nursery area to join shoals of juveniles occurred from late July to September. 相似文献

Ontogeny of the digestive tract during larval development of yellowtail flounder: a light microscopic and mucous histochemical study 总被引:6,自引:0,他引:6
The histological development and mucous histochemistry of the alimentary tract in larval yellowtail flounder were studied using light microscopy. Samples were taken when the larvae were first offered food at 3 days post-hatch, then at 7, 10, 29, 36, and 46 days post-hatch, at which time they were metamorphosing. Regional partitioning of the digestive tract into the buccal cavity, pharynx, oesophagus, post-oesophageal swelling (PES), intestine, and rectum was complete by day 10. Goblet cells were present only in the buccal cavity, pharynx and intestine by day 7, but increased in number and distribution as development continued. By day 29, the posterior zone of the oesophagus had a marked increase in goblet cell density and mucosal folding. At the transition from oesophagus to PES/stomach stratified epithelium with goblet cells changed abruptly to a columnar epithelium with no goblet cells. Multicellular glands in the PES of 36-day larvae allowed it to be defined as a stomach. The distinct brush border of columnar epithelium and the presence of goblet cells characterize the intestine and rectum. All goblet cells throughout the digestive tract were strongly positive for acid mucins as was the luminal layer of the stratified epithelia lining the buccal cavity, pharynx and oesophagus. The PES/stomach epithelium stained weakly for neutral mucins. No mucin staining was associated with the gastric glandular epithelium. The brush borders of the intestine and rectum were strongly positive for combinations of neutral and acid mucins. 相似文献

An allozymic study was conducted on 190 individuals of white sea bream, Diplodus sargus sampled on five locations throughout the Lion's Gulf and the Ligurian Sea (Mediterranean Sea), in order to identify genetic structure. Electrophoretic surveys carried out on muscle and liver tissues identified 25 loci of which 12 were polymorphic ( P 0.95). G -test analysis shows significant differences on allelic frequencies between the five stations at six loci. Examination of the spatial structure was performed using Nei's distances and F statistics, and indicated significant genetic differences between three groups. A group which clustered Blanes (Spain), Marseille (France) and Livorno (Italy) where the absence of genetic difference can be explained by migration of larvae and adults along a coastal 'continuum'. The Elba sample (Italy) shows genetic divergence from other samples and this difference may result from isolation due to limited migration of larvae and adults. Banyuls (France), differs from all other stations. Several hypotheses are proposed to explain genetic patterns including local current systems, larval dispersal, geographic isolation and historical effects, and variation in the size classes of sampled individuals between sites. 相似文献

The ontogeny of the digestive system of cobia Rachycentron canadum from hatching to 22 days post-hatch (dph) (20·1 mm standard length) was examined with light microscopy. The activities of selected pancreatic enzymes were also determined during this period in order to optimize current rearing methods for this species. At hatching (3·6 mm), the digestive tract consisted of a relatively undifferentiated, straight tube positioned dorsally to the yolk sac. The major morphological changes in the digestive tract primarily occurred over the first 1–4 dph (3·6–4·4 mm). During this time, larvae began exogenous feeding (3 dph) and the digestive tract differentiated into five histologically distinct regions: buccopharynx, oesophagus, stomach anlage, anterior intestine and posterior intestine. Yolk reserves were exhausted by 5 dph (4·5 mm) and the oil globule began rapidly decreasing in size disappearing entirely by 9–10 dph (6·3–6·8 mm). Gastric glands differentiated at this time, and by 12 dph (8·1 mm) surface mucous cells of the stomach anlage stained positive for neutral mucosubstances. By 16 dph (11·6 mm), the blind sac (fundic region) of the stomach formed as did the pyloric caecae which initially appeared as a single protrusion of the anterior intestine just ventral to the pyloric sphincter. Generally, enzyme activities (U larva−1 ) for amylase (0·0–1·8), chymotrypsin (0·0–7902·4), trypsin (0·2–16·6) and lipase (9·3–1319·0) were measurable at or soon after hatching and increased steadily from c. 8–22 dph (5·7–20·1 mm). The results of this study are discussed in terms of current and future weaning practices of this species. 相似文献

The amylase and trypsin activities of Artemia reared on synthetic axenic media were studied during development and compared with earlier results obtained with phytoplankton-fed Artemia. The enzymatic activity responses are also analysed with regard to the Provasoli growth index, protein growth and survival during starvation experiments or during ones involving specific deletions (starch or albumin). These experiments demonstrated a repressive regulatory mechanism for amylase and trypsin under substrate-saturating conditions. Differences in response of amylase and trypsin to starch and albumin deletions are discussed in terms of the existence of internal carbohydrate and protein pools in crustaceans. The ability of Artemia to shift from a high metabolism with exponential growth to a low metabolism without any growth but permitting survival is observed. The related enzyme responses are discussed. The importance of interactions between nutritional conditions, metabolic requirements, and the regulation of digestive enzymes is stressed. The response times and implications for the field studies on Zooplankton ecology are discussed. 相似文献

The cotton boll weevil, Anthonomus grandis, is a major pest of cotton crops in South America. In this work, partial biochemical characterizations of (hemi) cellulases and pectinases activities in the digestive system (head- and gut- extracts) of A. grandis were evaluated. Gut extract section from third instar larvae exhibited endoglucanase, xylanase, β-glucosidase, and pectinase activities. The endoglucanase and xylanase activities were localized in the foregut, whereas β-glucosidase activity was mainly detected in the hindgut. In addition, no difference in pectinase activity was observed across the gut sections. Thus, A. grandis digestive system is a potentially interesting reservoir for further lignocellulolytic enzymes research. 相似文献

The antioxidant enzymes superoxide dismutase (SOD; EC 1.15.1.1), catalase (EC 1.11.1.6), selenium-dependent glutathione peroxidase (SeGPX; EC 1.11.1.9), glutathione reductase (EC 1.6.4.2) and DT-diaphorase (EC 1.6.99.2), plus total GPX activity (sum of SeGPX and Se-independent GPX activities), were studied in 13 500 g supernatants of embryos and 3-day and 11-day post-hatch larvae of turbot Scophthalmus maximus L. SOD activity decreased progressively during development from embryos to 11-day-old larvae, indicative of a decreased need to detoxify superoxide anion radical (O2 − ). In contrast, catalase, SeGPX and glutathione reductase activities increased progressively from embryos to 11-day-old larvae, indicative of an increased need to metabolize hydrogen peroxide (H2 O2 ) and organic peroxides. Consistent with the latter changes, levels of lipid peroxides (i.e. thiobarbituric acid reactive substances) increased 13-fold from embryos to 3-day-old larvae, whilst total peroxidizable lipid was indicated to decrease. Increases were seen for NADPH-dependent DT-diaphorase (after hatching) and total GPX (between 3 and 11 days post-hatch) activities, whilst no change was found in NADH-dependent DT-diaphorase activity. Overall, the results demonstrate a capacity for early life-stages of S. maximus to detoxify reactive oxygen species (O2 − and H2 O2 ) and other pro-oxidant compounds (organic peroxides, redox cycling chemicals). Furthermore, qualitative and quantitative antioxidant changes occur during hatching and development, possibly linked to such events as altered respiration rates (SOD changes) and tissue reorganization and development (catalase, SeGPX, lipid peroxidation). 相似文献

Trichosia pubescens larval midgut ceca cells display in their plasma membranes α-glucosidases (Mr 95,000; pHo 5.5; Km 5.7 mM; Ki for TRIS 8.9 mM), trehalases (Mr 69,000; pHo 5.3; Km 0.92 mM; Ki for TRIS 57 mM), and aminopeptidases (Mr 95,000; pHo 8.7; Km 0.19 mM) which are solubilized by Triton X-100. The enzymes were purified by electrophoresis and used to raise antibodies in a rabbit. T. pubescens imaginal midgut cells display in their plasma membranes an α-glucosidase (Mr 156,000; pHo 5.8; Km 2.3 mM; Ki for TRIS 0.2 mM), a trehalase (Mr 93,000; pHo 5.5; Km 0.72 mM; Ki for TRIS 45.5 mM), and an aminopeptidase (Mr 210,000; pHo 9.0; Km 0.47 mM). Antiserum produced against the larval enzymes shows no precipitation arc when tested by double immunodiffusion or by immunoelectrophoresis with Triton X-100-solubilized membrane proteins from imaginal midguts. Otherwise, a similar test showed that larval midgut cecal enzymes and larval ventriculus enzymes display complete immunological identity. The data suggest that, despite the fact the larval and imaginal aminopeptidase, α-glucosidase, and trehalase probably have similar functions, the genes coding for them in larvae and imagoes must differ. 相似文献

Histological changes of the digestive system and its associated glands, and structures of the jaw were studied in summer flounder Paralichthys dentatus from hatching (day 0) until day 44. Specimens for this study were hatched from artificially spawned broodstock and maintained in the laboratory (20 ± 1° C). During the first 3 days after hatching, the formation of the oral jaw apparatus, lengthening of the digestive tube, yolk resorption, and mucosae differentiation are the most conspicuous elements of development. The larval digestive system is morphologically ready to process external food at the time of mouth opening (3–4 days after hatching). Epithelial cells of the anteromedian and the posterior intestine show evidence of lipid and protein absorption, respectively, after first feeding. The most noticeable events occurring during the next month of independent life are an increase in mucosal folding, cellular differentiation in the luminal epithelia, gut segmentation and looping, and liver growth. Gastric glands and pyloric caeca appear by day 31 and complete the morphological digestive features characteristic of the juvenile stage. 相似文献

为了阐明草地螟Loxostege sticticalis大发生种群幼虫取食行为特征, 在室内条件下(温度22±1℃, 相对湿度70%)对不同幼虫密度[1, 10, 30头/瓶(650 mL)]饲养草地螟幼虫的食物利用率及消化酶活性进行了研究。结果表明: 幼虫中等(或高)密度对草地螟幼虫相对中肠重量、 相对取食量、 粪便干重、 食物利用率和近似消化率及总蛋白酶和亮氨酸氨肽酶活性影响显著。幼虫相对中肠重量以10头/瓶的幼虫密度最大, 1头/瓶的幼虫密度最小。随着幼虫密度的增加, 幼虫相对取食量和粪便干重增加, 而虫体干重减轻, 幼虫食物利用率降低。幼虫密度30头/瓶的幼虫相对取食量和粪便干重显著高于1和10头/瓶的, 而30头/瓶的幼虫食物利用率显著低于1头/瓶的。幼虫近似消化率随幼虫密度的逐渐增加而显著降低。幼虫密度10头/瓶的幼虫总蛋白酶和亮氨酸氨肽酶的活性显著高于1和30头/瓶的, 而淀粉酶的活性受幼虫密度影响不显著。随幼虫密度的增加, 幼虫相对中肠重量与总蛋白酶和亮氨酸氨肽酶活性变化趋势较为一致, 消化酶活性的变化可能与相对中肠重量大小有关。因此, 幼虫密度是影响草地螟幼虫取食行为的重要因子, 这些结果为阐明草地螟大发生种群与一般种群的为害特征提供了重要理论依据。 相似文献

At hatching, the yolk-sac matrix of Siberian sturgeon Acipenser baeri contained neutral glycoconjugates, glycogen, proteins rich in arginine, lysine, tyrosine, cysteine and cystine, glycoproteins containing mannose (Man) and/or glucose (Glc), N -acetyl-D-galactosamine (GalNAc), L-fucose (Fuc), sialic acid and/or N -acetyl-D-glucosamine (GlcNAc) residues, as well as neutral and acidic lipids. Buccopharyngeal and anterior oesophageal goblet cellls produced a combination of neutral and acid sialoglycoproteins, while those from the posterior oesophagus secreted only neutral glycoproteins; both types of secretions contained tryptophan and -S-S- groups and were unreactive to lectin techniques. Most intestinal goblet cells secreted mainly carboxylated and sulphated sialoglycoproteins with some rests of neutral glycoconjugates, while few of them produced only acid or neutral glycoproteins. Intestinal glycoproteins were rich in GalNAc, GlcNAc and sialic acid residues. Close relationships between digestive enzymes and morphological development of digestive organs were observed. Histochemistry of enzymes revealed that just after hatching, alkaline and acid phosphatase, ATP -ase and non-specific esterase activities were detected in the yolk sac. From the onset of exogenous feeding to the juvenile stage (30 days post-hatch), an enhancement of enzymatic activities was observed, as alkaline and acid phosphatase, ATP -ase, aminopeptidase M and nonspecific esterase sharply increased. However, lipase activity decreased in the liver and brush border of enterocytes by 13–14 days post-hatch. Two types of lipase were detected in the alimentary canal, a non-pancreatic lipase that was secreted in the cardiac stomach by gastric glands, and a pancreatic lipase, which activity was mainly detected in the brush border of the intestinal epithelium. 相似文献

Abstract. .Reproducing females of the firebug, Pyrrhocoris apterus (Linnaeus) (Heteroptera: Pyrrhocoridae), reared under long-day (LD 18:6h) photocycle (long-day females), and diapausing females reared under short-day (LD 12:12h) conditions (short-day females) were analysed for temporal patterns of feeding, drinking and gut digestive enzyme activities. In long-day females the highest activities of feeding and drinking were found at the beginning of each reproductive cycle. Significant correlation between feeding and drinking activities in both reproducing long-day females and short-day females during pre-diapause state was found. In pre-diapausing short-day females the cyclical pattern of feeding and drinking was maintained despite the inhibition of reproduction. After entering the diapause their feeding activity became very low. Analysis of five gut enzymes revealed that short-day females differ significantly from long-day females in higher protease, amylase and esterase activities, in case of lipase only at certain times of their adult life; no significant difference was found in the aminopeptidase activity. Positive correlations between intensity of feeding and gut enzyme activities were observed in short-day females. Using several protease inhibitors most protease activity in the gut of P.apterus was identified as cysteine and aspartyl proteases, and only a small part of the activity could be caused by some serine proteases. 相似文献

Mouthpart and alimentary canal development was examined in Lysmata amboinensis larvae using scanning electron microscopy and histology. The gross morphological features of external mouthparts and internal digestive tract structures of larvae at different developmental stages indicate that ingestive and digestive capabilities are well developed from early on. With increasing age of the larvae the mouthpart appendages increased in size, the hepatopancreas in tubular density and the midgut in length. The density of setae and robustness of teeth and spines of individual structures increased. The most pronounced changes from early to late stage larvae involved formation of pores on the paragnaths and labrum, transformation of the mandibular spine‐like teeth to molar cusps, development of the filter press in the proventriculus and of infoldings in the previously straight hindgut. The results suggest that early stage L. amboinensis larvae may benefit from soft, perhaps gelatinous prey, whereas later stages are better equipped to handle larger, muscular or more fibrous foods. Two investigations were carried out with 150 g gilthead sea bream Sparus aurata to determine the relative activity of six digestive enzymes (pepsin, trypsin, chymotrypsin, carboxypeptidase A, carboxypeptidase B and amylase) and the pH variation in the lumen of different parts of the gut of fish fed one or two meals per day. Pepsin activity was found exclusively in the stomach, whereas activities of the other enzymes studied were found in all regions of the gut, including the stomach. The lack of localization of enzyme production in the digestive tract of S. aurata is similar to many other species as reported in the literature. The pH variations found in the different regions of the gut could be explained by general digestive physiology following the flow of digesta along the digestive tract. The range of pHs recorded in the various regions of the gut were generally outside the cited optima for many digestive proteases in this species. 相似文献

小线角木蠹蛾Streltzoviella insularis是一种重要的林木蛀干害虫,在我国北方地区广泛分布,其寄主范围广,尤其对城市园林树种威胁严重。本研究测定了小线角木蠹蛾幼虫分别取食3种喜食寄主(洋白蜡、银杏和国槐)和4种普通寄主(旱柳、栾树、香椿、毛桃)后的解毒酶(细胞色素P450酶、谷胱甘肽S转移酶和羧酸酯酶)和消化酶(内切-β-1,4-葡聚糖酶、外切-β-1,4-纤维二糖水解酶、β-葡萄糖苷酶和木聚糖酶)的活性。结果表明,不同寄主对小线角木蠹蛾幼虫的消化酶和解毒酶都存在显著影响,其中取食普通寄主的幼虫3种解毒酶的平均活性(P450、GST和CarE酶活性分别为21.12、4876.86、0.035μmoL/min/mg protein)均高于取食喜食寄主的幼虫(P450、GST和CarE酶活性分别为15.87、3760.55、0.027μmoL/min/mg protein),且普通寄主的平均次生代谢物质含量更高(普通寄主总酚、类黄酮和单宁含量分别为173.10、110.07、88.77 mg/g,喜食寄主为105.22、14.05、55.74 mg/g);而取食普通寄主与喜食寄主的幼虫EG和CBH酶活性之间没有显著差异,仅取食普通寄主的幼虫β-葡糖苷酶活性和木聚糖酶活性略高于取食喜食寄主的幼虫,幼虫的消化酶活性(纤维素酶和半纤维素酶)和寄主营养物质(纤维素和半纤维素)并未在普通寄主和喜食寄主中表现出明显趋势。因此本研究认为小线角木蠹蛾的寄主适应性与寄主次生代谢物质(总酚、类黄酮和单宁)含量的关联性更强,而与营养物质(纤维素和半纤维素)含量的相关性较弱。 相似文献

At hatching, the oesophagus of haddock Melanogrammus aeglefinus lacks goblet cells, the intestine is a simple undifferentiated tube, the liver is present as a rounded mass caudal to the heart, and numerous zymogen granules are present in the pancreas. The first intestinal convolution appears at day 2, at the posterior end of the digestive tract. The oesophagus displays alcian blue and PAS positive mucus secreting cells on day 12, which become numerous by day 15. By day 18, epithelial cells of the posterior intestine show evidence of protein absorption in the form of supranuclear vacuoles. The swimbladder inflates in 50% of the larvae by day 22, although inflation rate is highly variable. By day 35, or 10 mm, a pyloric caecal ridge appears which separates the presumptive stomach, which is now showing evidence of gastric gland formation, from the intestine. This marks the beginning of digestive features characteristic of the juvenile stage. 相似文献

为了明确继代饲喂不同寄主植物对草地贪夜蛾幼虫体内生理酶活性的影响,在室内分别用3种寄主植物叶片饲养草地贪夜蛾多代,测定其F3代5龄幼虫体内保护酶(SOD,POD,CAT)、消化酶(脂肪酶、胃蛋白酶和α-淀粉酶)和解毒酶(GSTs,CarEs,CYP450s)活性差异,并分析不同寄主植物次生代谢物含量与幼虫体内酶活及其生长发育的相关性。结果表明,取食马铃薯叶片的F3代幼虫总发育历期最长,取食小麦的最短,两者间差异显著;小麦叶片中的单宁、总酚、类黄酮和生物碱含量均显著低于玉米和马铃薯;草地贪夜蛾幼虫体内保护酶活性均为取食小麦叶片的最低,取食马铃薯叶片的最高,其中取食马铃薯叶片的草地贪夜蛾体内POD和SOD酶活性达取食小麦的1.34和1.26倍;以3种寄主为食的F3代幼虫脂肪酶和α-淀粉酶活性变化不大,但持续饲喂马铃薯叶片3个世代可导致草地贪夜蛾幼虫体内胃蛋白酶活性显著升高,达10.502 U/mg. prot,为持续饲喂玉米叶片幼虫(1.508 U/mg. prot)的6.96倍以上。连续饲喂3种寄主叶片后的幼虫GSTs和CarEs活性无显著性差异,但取食玉米叶片的幼虫体内细胞色素P450s(CYP450s)活性显著高于取食小麦和马铃薯的幼虫。可见,连续多代取食次生物质含量不同的寄主植物会使草地贪夜蛾体内部分酶活性发生变化,并对幼虫的生长发育产生影响。 相似文献
