The aim of this study was to investigate the effect of processing route (i.e., quench cooling and ball milling) on the surface energy heterogeneity and surface chemistry of indomethacin (IMC). Recently developed inverse gas chromatography (IGC) methodology at finite concentrations was employed to determine the surface energy distributions of crystalline, quench cooled and milled IMC samples. Surface properties of crystalline and processed IMC were measurably different as determined by the IGC and other conventional characterization techniques: differential scanning calorimetry and powder X-ray diffraction. Quench cooled IMC was in fully amorphous form. Milled IMC showed no amorphous character by calorimetric or X-ray diffraction studies. It was demonstrated that both processed IMC samples were energetically more active than the crystalline IMC. In particular, milled IMC exhibited a relatively higher dispersive surface energy and higher surface basicity (electron donor capability). This may be attributed to the creation of surface defect sites or exposure of higher energy crystal facets during the milling process. This study confirms that processing route has notable influence on the surface energy distribution and surface acid–base character. Amorphous drugs are used to improve the solubility, dissolution, and bioavailability of drugs. However, these metastable forms of drugs can transform into more stable, less soluble, crystalline counterparts. This study reports a method for evaluating the effect of commonly used excipients on the surface crystallization of amorphous drugs and its application to two model amorphous compounds, nifedipine and indomethacin. In this method, amorphous samples of the drugs were covered by excipients and stored in controlled environments. An inverted light microscope was used to measure in real time the rates of surface crystal nucleation and growth. For nifedipine, vacuum-dried microcrystalline cellulose and lactose monohydrate increased the nucleation rate of the β polymorph from two to five times when samples were stored in a desiccator, while d-mannitol and magnesium stearate increased the nucleation rate 50 times. At 50% relative humidity, the nucleation rates were further increased, suggesting that moisture played an important role in the crystallization caused by the excipients. The effect of excipients on the crystal growth rate was not significant, suggesting that contact with excipients influences the physical stability of amorphous nifedipine mainly through the effect on crystal nucleation. This effect seems to be drug specific because for two polymorphs of indomethacin, no significant change in the nucleation rate was observed under the excipients.KEY WORDS: amorphous, drugs, growth rate, nucleation rate, tablet excipients  相似文献

Atorvastatin calcium (ATC), an anti-lipid BCS class II drug, is marketed in crystalline and amorphous solid forms. The objective of this study was to perform solid state characterization of commercial crystalline and amorphous ATC drug samples available in the Indian market. Six samples each of crystalline and amorphous ATC were characterized using X-ray powder diffractometry (XRPD), differential scanning calorimetry (DSC), thermogravimetric analysis, Karl Fisher titrimetry, microscopy (hot stage microscopy, scanning electron microscopy), contact angle, and intrinsic dissolution rate (IDR). All crystalline ATC samples were found to be stable form I, however one sample possessed polymorphic impurity, evidenced in XRPD and DSC analysis. Amongst the amorphous ATC samples, XRPD demonstrated five samples to be amorphous ‘form 27’, while, one matched amorphous ‘form 23’. Thermal behavior of amorphous ATC samples was compared to amorphous ATC generated by melt quenching in DSC. ATC was found to be an excellent glass former with Tg/Tm of 0.95. Residual crystallinity was detected in two of the amorphous samples by complementary use of conventional and modulated DSC techniques. The wettability and IDR of all amorphous samples was found to be higher than the crystalline samples. In conclusion, commercial ATC samples exhibited diverse solid state behavior that can impact the performance and stability of the dosage forms.  相似文献

Magnetotactic multicellular aggregates consist of several bacteria that produce iron sulfide magnetosomes through a complex and poorly understood process. We observed new amorphous mineral particles within the cytoplasm of magnetotactic multicellular aggregates. Elemental mapping and electron energy loss spectroscopy detected iron and oxygen, but not sulfur, in these particles. These amorphous particles were about the same size as mature magnetosomes, around 50-70 nm in diameter. No membranes were observed surrounding the amorphous minerals. Partially crystalline inclusions composed of a crystalline core and an amorphous region around them similar in texture to the amorphous particles were also present. The shape of these amorphous regions followed the shape of the crystalline cores they enveloped. These regions also contained oxygen and iron. The crystalline phase, as previously reported, contained sulfur and iron. The presence of independent amorphous particles has not been reported before in magnetotactic multicellular aggregates.  相似文献

Thermogravimetric analyses (TGA) was applied to study the effects of enzymatic bleaching of eucalyptus pulp with xylanase and a laccase-mediator system. The thermal degradation profile of the pulps was sensitive to the enzymatic treatments. Xylanase treatment produced an ordered and clean microfibril, whereas laccase oxidized surface cellulose chains and increased the amorphous (paracrystalline) cellulose content. In this case, pulp viscosity decreased from 972 to 859 mL/g and apparent pulp crystallinity calculated from TGA data decreased almost 50%. Alkaline extraction was necessary to recover pulp crystallinity and to remove oxidized lignin in the laccase-treated samples. TGA data allowed differentiating and quantifying crystalline and amorphous cellulose. This thermogravimetric approach is a simple method in order to monitor superficial changes in cellulosic microfibrils.  相似文献

Oxygen vacancies are demonstrated to be beneficial to various electrocatalytic reactions. However, integrating oxygen vacancies into an amorphous catalyst with a large specific surface area, and investigating its effect on the oxygen evolution reaction remains a great challenge. Herein, oxygen vacancies are introduced into an amorphous N, P, and F tri‐doped CoFe2O4 using ionic liquid as a dopant. Simultaneously, ultrafine MoS2 nanoclusters are anchored onto its surface to increase the specific surface area. The vacancy‐rich MoS2/NPF‐CoFe2O4 exhibits an overpotential of 250 mV and a small Tafel slope of 41 mV dec?1, which is the best spinel‐based oxygen evolution reaction (OER) electrocatalysts so far. The excellent performance is attributed to massive oxygen vacancies, amorphous structure, large surface area, and synergistic coupling effects among active species. Density‐functional theory calculations reveal that the electronic structure of the catalyst can be modulated in the presence of heteroatoms and MoS2 nanoclusters, and then the energy barriers of intermediates are decreased as well, which enhances the OER performance. This design not only provides a simple strategy to construct amorphous structures with abundant oxygen vacancies using ionic liquid‐dopants, but also presents an in‐depth insight into the OER mechanism in alkaline solution.  相似文献

Supercapacitors or electrochemical capacitors, as energy storage devices, require very stable positive electrode materials for useful applications. Although most positive electrodes are based on crystalline mixed‐metal hydroxides, their pseudocapacitors usually perform poorly or have a short cycle life. High activities can be achieved with amorphous phases. Methods to produce amorphous materials are also not typically amenable towards mixed‐metal compositions. It is demonstrated that electrochemistry in an ambient environment can be used to produce a series of amorphous mixed‐metal hydroxides with a homogeneous distribution of metals for use as positive electrode materials in a supercapacitor. The integrated performance of the amorphous ternary mixed‐metal hydroxide pseudocapacitor is superior to that of crystalline materials. The amorphous Ni‐Co‐Fe hydroxide supercapacitor is characterized by a long‐term cycling stability that retained 94% of its capacity after 20 000 cycles. This is much higher than the cycle life of crystalline devices. These results show the broad applicability of this methodology towards new electrode materials for high‐performance supercapacitors, especially amorphous mixed‐metal hydroxides, as advanced electrode materials.  相似文献

Zopiclone is a poorly soluble psychotherapeutic agent. The aim of this study was to prepare and characterize an amorphous form of zopiclone as well as the characterization and performance of a stable amorphous solid dispersion. The amorphous form was prepared by the well-known method of quench-cooling of the melt. The solid dispersion was prepared by a solvent evaporation method of zopiclone, polyvinylpyrrolidone-25 (PVP-25), and methanol, followed by freeze-drying. The physico-chemical properties and stability of amorphous zopiclone and the solid dispersion was studied using differential scanning calorimetry (DSC), infrared spectroscopy (FT-IR), thermogravimetric analysis (TGA), scanning electron microscopy (SEM), hot-stage microscopy (HSM), X-ray diffractometry (XRD), solubility, and dissolution studies. The zopiclone amorphous solid-state form was determined to be a fragile glass; it was concluded that the stability of the amorphous form is influenced by both temperature and water. Exposure of amorphous zopiclone to moisture results in rapid transformation of the amorphous form to the crystalline dihydrated form. In comparison, the amorphous solid dispersion proved to be more stable with increased aqueous solubility.KEY WORDS: amorphous, fragile, solid dispersion, stability, zopiclone  相似文献

The hierarchical structure of spider dragline silk is composed of two major constituents, the amorphous phase and crystalline units, and its mechanical response has been attributed to these prime constituents. Silk mechanics, however, might also be influenced by the resistance against sliding of these two phases relative to each other under load. We here used atomistic molecular dynamics (MD) simulations to obtain friction forces for the relative sliding of the amorphous phase and crystalline units of Araneus diadematus spider silk. We computed the coefficient of viscosity of this interface to be in the order of 102 Ns/m2 by extrapolating our simulation data to the viscous limit. Interestingly, this value is two orders of magnitude smaller than the coefficient of viscosity within the amorphous phase. This suggests that sliding along a planar and homogeneous surface of straight polyalanine chains is much less hindered than within entangled disordered chains. Finally, in a simple finite element model, which is based on parameters determined from MD simulations including the newly deduced coefficient of viscosity, we assessed the frictional behavior between these two components for the experimental range of relative pulling velocities. We found that a perfectly relative horizontal motion has no significant resistance against sliding, however, slightly inclined loading causes measurable resistance. Our analysis paves the way towards a finite element model of silk fibers in which crystalline units can slide, move and rearrange themselves in the fiber during loading.  相似文献

Interface response functions (IRFs) for amorphous and crystalline forms of Si have been determined for several empirical atomic-scale models using Molecular Dynamics and compared to available experimental results fitted to a Wilson-Frenkel equation form. Stillinger–Weber (SW), the environment-dependent intermolecular potential (EDIP), and a version of the modified embedded atom method (MEAM) models were found to produce unacceptable representations of the IRFs of both solid phases; they were either unable to predict the amorphous melting point and/or the maximum solidification velocity. The best of these models was judged to be the SW potential, known to produce a very accurate IRF for crystalline silicon. Increasing the strength of the three-body term by up to 25% above that of the original SW potential improves the prediction of the melting characteristics of the amorphous phase. Above this limit, liquid phase properties are impaired. The resultant IRFs provide an important backdrop to understand the kinetics of explosive crystallization (EC) processes, as we shall show in comparison to recent experimental data on the EC of amorphous Ge. A cost effective hydrogen evolution reaction (HER) catalyst that does not use precious metallic elements is a crucial demand for environment‐benign energy production. The family of earth‐abundant transition metal compounds of nitrides, carbides, chalcogenides, and phosphides is one of the promising candidates for such a purpose, particularly in acidic conditions. However, its catalytic performance is still needed to be enhanced through novel material designs and crystalline engineering. Herein, a chemically and electronically coupled transition metal phosphosulfide/N‐doped carbon nanotubes (NCNT) hybrid electrocatalyst is fabricated via a two‐step synthesis. The uniquely designed synthesis leads to the material morphology featuring a core–shell structure, in which the crystalline metal phosphide core is surrounded by an amorphous phosphosulfide nanoshell. Notably, due to the favorable modification of chemical composition and surface properties, core–shell CoP@PS/NCNT exhibits the noticeable HER activity of approximately ?80 mV @ ?10 mA cm?2 with excellent durability, which is one of the highest active nonnoble metal electrocatalysts ever reported thus far.  相似文献

In view of the sluggish kinetics suppressing the oxygen evolution reaction (OER), developing efficient and robust OER catalysts is urgent and essential for developing efficient energy conversion technologies. Herein, hybrid amorphous/crystalline FeCoNi layered double hydroxide (LDH)-supported single Ru atoms (Ru SAs/AC-FeCoNi) are developed for enabling a highly efficient electrocatalytic OER. The amorphous outer layer in Ru SAs/AC-FeCoNi is composed of abundant defect sites and unsaturated coordination sites, which can serve as anchoring sites to stabilize single Ru atoms. The crystalline inner has a highly symmetric rigid structure, thereby strengthening the stability of support for a long-lasting OER. The synergistic effects endow this hybrid catalyst with extremely low overpotential (205 mV at 10 mA cm−2). Density functional theory calculation indicates that single Ru atoms stabilized by hybrid amorphous/crystalline FeCoNi LDH facilitate the formation of Ru–O* (rate-determining step), thus accelerating the OER process.  相似文献

Swelling behavior and rearrangements of an amorphous ultrathin cellulose film (20 nm thickness) exposed to water and subsequently dried were investigated with grazing incidence X-ray diffraction, neutron reflectivity, atomic force microscopy, and surface energy calculations obtained from contact angle measurements. The film swelled excessively in water, doubling its thickness, but shrunk back to the original thickness upon water removal. Crystallinity (or amorphousness) and morphology remained relatively unchanged after the wetting/drying cycle, but surface free energy increased considerably (ca. 15%) due to an increase in its polar component, that is, the hydrophilicity of the film, indicating that rearrangements occurred during the film's exposure to water. Furthermore, stability of the films in aqueous NaOH solution was investigated with quartz crystal microbalance with dissipation monitoring. The films were stable at 0.0001 M NaOH but already 0.001 M NaOH partially dissolved the film. The surprising susceptibility to dissolve in dilute NaOH was hypothetically attributed to the lack of hierarchical morphology in the amorphous film.  相似文献

The purpose of this research was to investigate the influence of changes in the amorphous state on the crystallization of trehalose. Amorphous trehalose is known to stabilize biomaterials; hence, an understanding of crystallization is vital. Amorphous trehalose, prepared by spray-drying, was exposed to either a single step (0–75%) in relative humidity (RH) or to modulated 0–75–0% RH to cause crystallization. For the single-step experiment, two samples crystallized in a predictable manner to form the dihydrate. One sample, while notionally identical, did not crystallize in the same way and showed a mass loss throughout the time at 75% RH, with a final mass less than that expected for the dihydrate. The idiosyncratic sample was seen to have a starting near infrared (NIR) spectra similar to that exhibited by anhydrous crystalline trehalose, implying that short-range order in the amorphous material (or a small amount of crystalline seed, not detectable using powder X-ray diffraction) caused the sample to fail to form the dihydrate fully when exposed to high RH. The modulated RH study showed that the amorphous material interacted strongly with water; the intensity of the NIR traces was not proportional to mass of water but rather the extent of hydrogen bonding. Subsequent crystallization of this sample clearly was a partial formation of the dihydrate, but with the bulk of the sample then shielded such that it was unable to show significant sorption when exposed to elevated RH. It has been shown that the nature of the amorphous form will alter the way in which samples crystallize. With oscillation in RH, it was possible to further understand the interactions between water and amorphous trehalose.  相似文献

Developing amorphous solid dispersions of water-insoluble molecules using polymeric materials is a well-defined approach to improve the dissolution rate and bioavailability. While the selected polymer plays a vital role in stabilizing the amorphous solid dispersion physically, it is equally important to improve the dissolution profile by inhibiting crystallization from the supersaturated solution generated by dissolution of the amorphous material. Furthermore, understanding the mechanism of dissolution rate enhancement is of vital importance. In this work, wetting kinetics was taken up as an alternative approach for understanding the enhanced dissolution rate for amorphous solid dispersion of a poorly soluble drug. While cilostazol (CIL) was selected as the model drug, povidone (PVP), copovidone, and hypromellose (HPMC) were the polymers of choice. The concentrations against time profiles were evaluated for the supersaturated solutions of CIL in the presence and absence of the selected polymers. The degree of supersaturation increased significantly with increase in polymer content within the solid dispersion. While povidone was found to maintain the highest level of supersaturation for the greatest length of time both in dissolution and solution crystallization experiments, copovidone and hypromellose were found to be the less effective as crystallization inhibitor. The ability of polymers to generate and maintain supersaturated drug solutions was assessed by dissolution studies. The time-dependent stress-strain behavior of spider dragline silk was already observed decades ago, and has been attributed to the disordered sequences in silk proteins, which compose the soft amorphous matrix. However, the actual molecular origin and magnitude of internal friction within the amorphous matrix has remained inaccessible, because experimentally decomposing the mechanical response of the amorphous matrix from the embedded crystalline units is challenging. Here, we used atomistic molecular dynamics simulations to obtain friction forces for the relative sliding of peptide chains of Araneus diadematus spider silk within bundles of these chains as a representative unit of the amorphous matrix in silk fibers. We computed the friction coefficient and coefficient of viscosity of the amorphous phase to be in the order of 10−6 Ns/m and 104 Ns/m2, respectively, by extrapolating our simulation data to the viscous limit. Finally, we used a finite element method for the amorphous phase, solely based on parameters derived from molecular dynamics simulations including the newly determined coefficient of viscosity. With this model the time scales of stress relaxation, creep, and hysteresis were assessed, and found to be in line with the macroscopic time-dependent response of silk fibers. Our results suggest the amorphous phase to be the primary source of viscosity in silk and open up the avenue for finite element method studies of silk fiber mechanics including viscous effects.  相似文献

The achievement of effective alkaline hydrogen production from water electrolysis is an active field of research. Herein, an integrated electrode composed of crystalline Ni(OH)2 and amorphous NiMoOx is fabricated onto nickel foam (denoted as Ni(OH)2–NiMoOx/NF). The hydrogen evolution reaction (HER) kinetics are optimized along with phase transformation process during soaking operation. An overpotential of 36 mV to drive 10 mA cm?2 along with the low Tafel slope of 38 mV dec?1 reveals the catalyst's excellent HER performance and a Heyrovsky‐step‐controlled HER mechanism. When assembled into a urea‐assisted water electrolyzer, a voltage of 1.42 V can reach 10 mA cm?2. Further experiments and Fourier transform infrared spectroscopy (FTIR) results illustrate the synergy effect between crystalline and amorphous areas and the optimized water dissociation step. Crystalline Ni(OH)2 serves as the scissor for water dissociation in an alkali environment to produce H*, while the amorphous NiMoOx layer serves as the location for H* adsorption and H2 desorption.  相似文献
