Surface microlayer and subsurface algal communities were comparedon 14 sample dates over a 5 month period in a small Wisconsinpond. Similarities in species composition indicate that themicrolayer community had many species in common with the subsurfacecommunity. Relative species abundances, however, were differentfrom surface to subsurface. Analysis of the depth distributionof individual species indicates that the response to the uniquephysical/chemical conditions in the microlayer is species specific,with flagellates often in low numbers in the surface microlayeras a result of negative phototaxis; and diatoms often in highnumbers in the surface microlayer, as a result of a specificphysical/chemical affinity. All Enterococcus spp., isolated from environmental water samples (n=81), emitted a high chemiluminescence signal in the presence of luminol (10(-2) M). Kinetic studies of chemiluminescence show a close correlation between chemiluminescence and growth curves during the exponential phase, with a maximum chemiluminescence reached just before bacterial growth entered in the stationary phase. On the other hand, genera closely related to Enterococcus such as Streptococcus or Lactococcus produced a very weak chemiluminescent signal. Chemiluminescence of enterococci could therefore offer a rapid test, in aiding the identification of the genus Enterococcus and in the survey of the microbiological quality of water supplies.  相似文献

Intensively cultured fish stock when fed protein-rich feeds typically excrete high concentrations of total ammoniacal-nitrogen (TAN) into the water column which can have adverse effects on productivity, and upon the environment when aquaculture water is discharged. An immobilized culture of nitrifying bacteria isolated from prawn pond water and known to effectively remove TAN from saline water was tested for its ability to remove TAN from freshwater. The culture was readily adaptable to non-saline conditions and maintained TAN at less than 0.25 mg l–1, even with a daily addition of 3.2 to 4.2 mg TAN l–1 per d. The use of the immobilized culture of nitrifying bacteria represents an innovative and economical in situ treatment technology for removal of TAN in both saline and freshwater.  相似文献

Expanding primary leaves of 11 cultivars of Phaseolus vulgaris, and one each of Phaseolus coccineus, Phaseolus lunatus and Vigna unguiculata were inoculated with 23 isolates of Elsinoe phaseoli from Phaseolus spp. and one from Vigna unguiculata. On the basis of the macroscopic and microscopic appearance of the lesions, symptoms were placed into five classes. Spore yields from the lesions in each class were used to classify the types of lesions as representing resistance or susceptibility to each isolate. There was a differential response of cultivars to isolates. According to the reaction of five cultivars of P. vulgaris to infection, isolates were placed in four pathogenicity groups. In addition to the differential reaction of cultivars, there was some evidence of host specificity among the isolates. Thus, P. lunatus was susceptible to most of the P. vulgaris isolates but resistant to the ones from P. coccineus. Vigna unguiculata was susceptible only to the V. unguiculata isolate and was resistant to all the isolates from Phaseolus spp. The isolate from V. unguiculata failed to infect any of the bean cultivars or the other Phaseolus spp. The need to select suitable isolates for challenging cultivars in a breeding programme is stressed.  相似文献

A survey was carried out on the species of fungi associated with mycotic infections of fish in a Nigerian freshwater fish pond. A total of 24 fungal species belonging to 6 genera of aquatic phycomycete were isolated from the infected fishes. Achlya racemosa, Aphanomyces laevis, Dictyuchus sterile, Saprolegnia ferax, S. litoralis and S. parasitica had 100% frequency of occurrence amongst the infected fishes. There were similarities in the species of fungi isolated from the infected fishes in the fish pond and those isolated from the hatchery. Of the infected fishes, Clarias lazera had the highest number of fungal isolates. It was followed by Tilapia zilli, then by Chanos chanos, Ethmalosa fimbriata and finally by Cyprinus carpio. The implications of the results obtained on the development of fish farming in Nigeria are discussed.  相似文献

Summary Temperature of air and surface water showed a double oscillation curve. Physicochemical conditions and their diurnal amplitudes varied depending on the depth of water, amount of phyton and total activity of organisms. Permanent hardness, temporary hardness, acidity and total carbondioxide varied interdependently and also closely related to the amounts of plant matter. The abundance of gastropods was correlated with the abundance of food associated with permanent hardness. The abundance of bottom soil fauna was related to the temperature. Floral succession and abundance were correlated with the variations in silicates, phosphates and other chemical conditions. Microcystis bloom was recorded during winter months.Data formed part of Ph. 1. Cryptic species diversity poses evolutionary questions about its origin and maintenance, and ecological questions about the coexistence of seemingly identical species. 2. We examine patterns of morphological and life history differentiation in three sympatric cryptic species of freshwater amphipods within the Hyalella azteca species complex. These species are separated by extensive molecular evolution, but appear similar in phenotype. Species were collected from the littoral zone of a small kettle lake in Michigan, U.S.A., and identified to species by molecular genetic methods. 3. Two of the species were similar, differing only in female size, whereas the third was larger in body size and had larger clutches of smaller eggs than the other two. There were differences between the species also in pleon spine length and antennal segment number. 4. An analysis of allozyme variation among the cryptic species in three lakes suggests that the species are reproductively isolated within lakes. 5. We suggest that phenotypic similarity of these species is maintained by size‐selective predation by fish. The small, but statistically significant, differences in body size may form the axis for a tradeoff between resource exploitation and predator avoidance, a condition that can foster coexistence of phenotypically similar species.  相似文献

A bacterial strain, designated cfT was isolated from surface water of a freshwater pond for shrimp (Macrobrachium rosenbergii) culture at Ping-Tung (Southern Taiwan). Cells of this organism were Gram-negative, slightly curved rods which were motile by means of a single polar flagellum. Strain cfT utilized chitin as the exclusive carbon, nitrogen, and energy source for growth, both under aerobic and anaerobic conditions. Optimum conditions for growth were between 25 and 37 degrees C, 0 and 1% NaCl and pH 6 to 8. Strain cfT secreted two chitinolytic enzymes with approximate molecular weight 52 and 64 kDa, which hydrolyzed chitin to produce chitotriose as major product. Sequence comparison of an almost complete 16S rDNA gene showed less than 92% sequence similarity with known bacterial species. Phylogenetic analysis based on the neighbour-joining and other methods indicated that the organism formed a distinct lineage within the beta-subclass of Proteobacteria. The predominant cellular fatty acids of strain cfT were hexadecanoic acid (about 29%), octadecenoic acid (about 12%) and summed feature 3 (16:1 omega7c or 15 iso 2-OH or both [about 49%]). Its DNA base ratio was 62.8 mol% G+C. We propose to classify strain cfT (= CCRC 17210T = LMG 22011T) as Chitinimonas taiwanensis gen. nov., sp. nov.  相似文献

Two freshwater crayfish species, Astacus astacus L. and Pacifastacus leniusculus Dana, co-occur in some Swedish lakes. Observational studies indicate that the introduced, North American species P. leniusculus may gradually replace the native A. astacus, but the mechanism behind the replacement is not known. This study examined the direct effects of interspecific competition between the crayfish, and indirect effects of competitive interactions and fish (European perch, Perca fluviatilis L.) predation. Three different experiments with young-of-the-year (YOY) crayfish were performed. P. leniusculus was strongly dominant over similar-sized A. astacus in interference competition for shelter in a laboratory experiment. However, in a 35-day experiment in outdoor pools, A. astacus growth and survival were about equally affected by interactions with conspecifics and P. leniusculus. In contrast, P. leniusculus was significantly more affected by intraspecific competition than by competition with A. astacus, suggesting asymmetric competition between the two species. The presence of perch in outdoor ponds with mixed-species groups of the two crayfish species resulted in considerably higher predation rates on A. astacus than on P. leniusculus. Both species showed strong antipredator responses to perch by increasing refuge use. I suggest that higher perch predation rates on A. astacus originate from P. leniusculus being the superior species in interspecific competition for shelter. Because of displacement from refuges, A. astacus individuals become more exposed to the predator. This indirect effect of interactions among the two cray-fish species and the predator may be important in the observed in situ replacement of A. astacus by P. leniusculus.  相似文献

A new Klebsiella species, K. variicola, is proposed on the basis of total DNA-DNA hybridization, on the monophyly observed in the phylogenetic analysis derived from the sequences of rpoB, gyrA, mdh, infB, phoE and nifH genes and on distinct phenotypic traits. The bacteria from this new species seem to be genetically isolated from K. pneumoniae strains, do not ferment adonitol and were obtained from plants (such as banana, rice, sugar cane and maize) and hospitals. 为了探讨栖息于不同生境中鱼类的游泳能力和偏好游泳速度及其生理机制,本研究以中华倒刺鲃(Spinibarbus sinensis)、异育银鲫(Carassius auratus gibelio)、岩原鲤(Procypris rabaudi)、青鱼(Mylopharyngodon piceus)和胭脂鱼(Myxocryprinus asiaticus) 5种鱼的幼鱼为对象,在(25±1)℃条件下测定了5种鱼类的标准代谢率(SMR)、最大代谢率(MMR)、有氧代谢范围(MS)、临界游泳速度(Ucrit)、最大匀加速游泳速度(Ucat)和偏好游泳速度(Upref)。结果发现:5种实验鱼中,中华倒刺鲃的游泳能力最强,游泳能力较差的为青鱼和胭脂鱼; 5种鱼之间的代谢和游泳能力差异显著,其偏好游泳速度主要集中在(10~24.5cm·s-1)区域。研究表明,鱼类游泳能力的种间差异可能主要由心鳃系统相关的呼吸能力和体型相关的游泳效率所决定。本研究提供的有关鱼类游泳能力、偏好游泳速度等资料对于鱼道设计等有一定的参考价值...  相似文献

养虾池好氧反硝化细菌新菌株的分离鉴定及特征   总被引:14,自引:0,他引:14  
利用间歇曝气选择性富集并对所获菌株的好氧反硝化活性进行检测。筛选到一株亚硝酸盐去除活性较高的好氧反硝化细菌。在溶解氧(D0)为3.80-5.21mg/L的培养条件下。该菌株10h内将亚硝态氮由26.18mg/L降至0;在盐度为0—25之间20h内均可达到同样的去除效果。通过形态学特征、生理生化反应及部分长度16SrDNA序列分析对筛选菌株进行鉴定,初步判定它为嗜麦芽寡养单胞菌Stertotrophomonas maltophilia。亚硝酸盐还原酶基因分析结果表明。该菌株只含亚硝酸盐还原酶nitS基因,其序列与Alcaligenes faecalis A15(后来被重新鉴定为Pseudomonas stutzeri)的nirS基因序列相似。  相似文献

Profiles of dissolved O(2) and methane with increasing depth were generated for Lake Washington sediment, which suggested the zone of methane oxidation is limited to the top 0.8 cm of the sediment. Methane oxidation potentials were measured for 0.5-cm layers down to 1.5 cm and found to be relatively constant at 270 to 350 micromol/liter of sediment/h. Approximately 65% of the methane was oxidized to cell material or metabolites, a signature suggestive of type I methanotrophs. Eleven methanotroph strains were isolated from the lake sediment and analyzed. Five of these strains classed as type I, while six were classed as type II strains by 16S rRNA gene sequence analysis. Southern hybridization analysis with oligonucleotide probes detected, on average, one to two copies of pmoA and one to three copies of 16S rRNA genes. Only one restriction length polymorphism pattern was shown for pmoA genes in each isolate, and in cases where, sequencing was done, the pmoA copies were found to be almost identical. PCR primers were developed for mmoX which amplified 1.2-kb regions from all six strains that tested positive for cytoplasmic soluble methane mono-oxygenase (sMMO) activity. Phylogenetic analysis of the translated PCR products with published mmoX sequences showed that MmoX falls into two distinct clusters, one containing the orthologs from type I strains and another containing the orthologs from type II strains. The presence of sMMO-containing Methylomonas strains in a pristine freshwater lake environment suggests that these methanotrophs are more widespread than has been previously thought.  相似文献

Geographical gradients in species richness and the degree to which different taxa show congruent patterns remain unknown for many taxonomic groups. Here, I examined broad-scale species richness patterns in five groups of freshwater organisms; macrophytes, dragonflies, stoneflies, aquatic beetles and fishes. The analyses were based on provincial distribution records in Denmark, Norway, Sweden and Finland. In general, variation in species richness across provinces was concordant among the groups, but stoneflies showed weaker negative relationships with the other taxonomic groups. Species richness in most groups decreased with increasing latitude and altitude, and a considerable part of the variation was explained by mean July temperature. However, stoneflies showed a reversed pattern, with species richness correlating positively, albeit more weakly, with mean provincial altitude. Nevertheless, combined species richness of all five taxa showed a strong relationship with mean July temperature, accounting for 74% of variation in provincial species richness alone. Such temperature-controlled patterns suggest that regional freshwater biodiversity will strongly respond to climate change, with repercussions for local community organization in freshwater ecosystems in Fennoscandia.  相似文献

Little is known about the epidemiology of Cryptosporidium in Jordan and no genotyping studies have been conducted on Cryptosporidium isolates from humans or animals from Jordan. Genotyping of 44 Cryptosporidium isolates from Jordanian children at the 18S rRNA locus and a unique diagnostic locus identified four Cryptosporidium species; C. parvum (22), C. hominis (20), C. meleagridis (1) and C. canis (1). Sub-genotype analysis of 29 isolates at the 60-kDa glycoprotein (GP60) locus identified three C. parvum, two C. hominis subtype families and one C. meleagridis subtype. Several rare and novel subtypes were identified indicating unique endemicity and transmission of Cryptosporidium in Jordan.  相似文献

The microbiome of freshwater sponges is rarely studied, and not a single novel bacterial species has been isolated and subsequently characterized from a freshwater sponge to date. A previous study showed that 14.4% of the microbiome from Ephydatia fluviatilis belong to the phylum Planctomycetes. Therefore, we sampled an Ephydatia sponge from a freshwater lake and employed enrichment techniques targeting bacteria from the phylum Planctomycetes. The obtained strain spb1T was subject to genomic and phenomic characterization and found to represent a novel planctomycetal species proposed as Planctopirus ephydatiae sp. nov. (DSM 106606 = CECT 9866). In the process of differentiating spb1T from its next relative Planctopirus limnophila DSM 3776T, we identified and characterized the first phage – Planctopirus phage vB_PlimS_J1 – infecting planctomycetes that was only mentioned anecdotally before. Interestingly, classical chemotaxonomic methods would have failed to distinguish Planctopirus ephydatiae strain spb1T from Planctopirus limnophila DSM 3776T. Our findings demonstrate and underpin the need for whole genome-based taxonomy to detect and differentiate planctomycetal species.  相似文献

Gastrotricha is a cosmopolitan group of aquatic invertebrates. To date, approximately 765 species have been described. This study is the first to deal with species delimitation and cryptic species of freshwater Gastrotricha. Three commonly encountered species, Heterolepidoderma ocellatum, Lepidochaetus zelinkai, and Lepidodermella squamata, are investigated for cryptic speciation. Most of the material is based on Swedish specimens but closely related species from other parts of the world are also included. Taxonomic revisions are supported by phylogenies based on 18S rRNA, 28S rRNA, and COI mtDNA of freshwater Chaetonotidae from several genera and inferred from Bayesian and maximum likelihood approaches. Heterolepidoderma ocellatum f. sphagnophilum is raised to species level, under the name H. acidophilum n. sp. Moreover, genetic data based on COI indicate large variation between two morphologically very similar groups of Lepidodermella squamata. The extent of cryptic speciation in L. zelinkai appears low. Based on the phylogenetic hypothesis presented in this article, the new species, Lepidodermella intermedia n. sp., from northern Sweden is also described. The phylogenetic hypothesis generated shows that Chaetonotidae is a non-monophyletic group.  相似文献

In an attempt to recycle the lignocellulosic wastes like Eichhornia crassipes, Salvinia cucullata and rice (Oryza sativa) straw as manurial inputs in freshwater fish pond ecosystem, a decomposition experiment was carried out in litter bags in an oligotrophic freshwater fish pond environment, with the above mentioned three substrates in unprocessed and microbially processed forms. The loss rates, associated microbial groups, oxygen consumption patterns and other related parameters like carbon, nitrogen, phosphorus, cellulose, hemicellulose and lignin were analysed. The mean daily dry matter loss rates (unprocessed: 10.44>6.97>1.97 and processed: 11.03>8.21>3.67) and oxygen uptake rate (unprocessed: 0.675>0.571>0.568 mg O2 g–1 h–1 and processed: 0.592>0.424>0.407 mg O2 g–1 h–1) in raw and processed substrates were in the sequence Eichhornia > rice straw > Salvinia. The oxygen consumption pattern almost covariated with variations in temperature of pond water, daily dry matter loss rates and fungal counts on substrates. During the decay, the percentage of N and P increased whereas that of C decreased, resulting in lowering of C/N and C/P ratios of the substrates. The structural polymeric fractions like cellulose and hemicellulose decreased along with dry matter whereas the lignin content increased after an initial decrease due to loss of other structural carbohydrates resulting in apparent per cent gain of lignin. A higher number of different heterotrophic bacterial groups was observed in the processed substrates as compared to their raw counterparts. However, cellulolytic bacterial numbers were found to fluctuate through the study period. The fungal load was found to be decreasing gradually as the decay progressed. In this study, bacteria were found to be the prominent microbial group responsible for the decay. The nitrogen-fixing, phosphatase-producing and phosphorus-solubilising bacterial groups were observed to play an important role in lowering the C/N and C/P ratios of the decomposing substrates during decay.  相似文献

Betanodaviruses, the causative agents of viral nervous necrosis in marine fish, have bipartite positive-sense RNA genomes. Because the genomes are the smallest and simplest among viruses, betanodaviruses have been well studied using a reversed genetics system as model viruses. However, studies of virus-host interactions have progressed slowly because permissive hosts for betanodaviruses (basically larvae and juveniles of marine fish) are only available for limited periods of the year and are not suitable for the construction of a genetic engineering system. To obtain a model fish species that are not subject to these problems, 21 freshwater fish species were injected intramuscularly with a betanodavirus (redspotted grouper nervous necrosis virus) and tested for their susceptibility to the virus. Based on their responses, the tested fish were classified into 3 groups: 4 susceptible fish, 10 less susceptible fish, and 7 resistant fish. The susceptible fish, celebes rainbowfish Telmatherina ladigesi, threadfin rainbowfish Iriatherina werneri, dwarf rainbowfish Melanotaenia praecox, and medaka Oryzias latipes, exhibited erratic swimming and eventually died within 10 d post-inoculation. The virus was specifically localized in the brains, spinal cords, and retinas of the infected fish, similar to the pattern of infection in naturally infected marine fish. We believe that these susceptible freshwater fish species could act as good host models for betanodavirus-fish interaction studies.  相似文献
