Monte Carlo values of the density profiles and related properties of the double layer formed by an electrolyte near a charged electrode are reported for the cases where the electrode has a dielectric coefficient greater, equal, and smaller than that of the electrolyte that causes a surface polarization that can be represented by electrostatic images. As expected, compared to the case where there is no dielectric boundary the ions near the electrode are attracted or repelled by the electrode if the dielectric coefficient is greater or smaller, respectively, than that of the electrolyte. This effect is most pronounced near the electrode and is stronger for 2:2 electrolytes than for 1:1 electrolytes. For both monovalent and divalent ions the effect of the dielectric boundary is stronger at low concentrations. 相似文献

A study was made of the adhesion of liposomes, composed of dipalmitoyl- or di-stearoylphosphatidycholine, on the surface of epithelial cells in culture. Sodium fluorescein was entrapped in liposomes for their visualization by fluorescence microscopy. It is found that sonicated unilamellar liposomes adhere predominantly along the sheet margins. Multilamellar liposomes and lipid-coated carmine particles adhere over the whole cellular surface. However, their adhesion along sheet margins was stronger, as evidenced by a brief trypsin treatment. A prolonged trypsin treatment removed all types of liposomes from the cell surface. After the cells were partly detached from each other, small liposomes readily adhered to the newly accessible cell margins. The existence of special lipid membrane-binding proteins on the cell surface is suggested. 相似文献

Equations for the motion of an individual dust grain in the double layer of a negatively charged cylindrical probe in a glow discharge plasma are derived and solved numerically. The distribution of the electric potential near the probe is determined, and the grain charge is calculated as a function of the distance from the probe for different probe potentials. The trajectories of grains with different initial energies are traced. An analysis of the grain trajectories shows that, at a certain distance from the probe, high-energy grains may be recharged; i.e., the grain charge may change sign. The grains are found to have no direct effect on the probe current in a dusty plasma of a glow discharge. 相似文献

Photosystem II (PSII) catalyzes the oxidation of water to O2 at the manganese-containing, oxygen-evolving complex (OEC). Photoexcitation of PSII results in the oxidation of the OEC;
four sequential oxidation reactions are required for the generation and release of molecular oxygen. Therefore, with flash
illumination, the OEC cycles among five S
n
states. Chloride depletion inhibits O2 evolution. However, the binding site of chloride in the OEC is not known, and the role of chloride in oxygen evolution has
not as yet been elucidated. We have employed reaction-induced FT-IR spectroscopy and selective flash excitation, which cycles
PSII samples through the S state transitions. On the time scale employed, these FT-IR difference spectra reflect long-lived
structural changes in the OEC. Bromide substitution supports oxygen evolution and was used to identify vibrational bands arising
from structural changes at the chloride-binding site. Contributions to the vibrational spectrum from bromide-sensitive bands
were observed on each flash. Sulfate treatment led to an elimination of oxygen evolution activity and of the FT-IR spectra
assigned to the S3 to S0 (third flash) and S0 to S1 transitions (fourth flash). However, sulfate treatment changed, but did not eliminate, the FT-IR spectra obtained with the
first and second flashes. Solvent isotope exchange in chloride-exchanged samples suggests flash-dependent structural changes,
which alter protein dynamics during the S state cycle.

The discrete cilia approach is employed to describe the flow field above an active ciliated surface in a layer of finite depth. The infinite-size cilia surface model predicts a uniform flow above the cilia surface. Modifications as a result of a finitely extending cilia surface inside a finite-size dish predict a backward parabolic profile above an active cilia surface. Experiments are described which demonstrate a good fit between theoretical predictions and observations. The results provide a sound physical basis for the proper interpretation of fluid-flow observations close to an active ciliary field. 相似文献

Data are presented on the conductivity of bilayer lipid membranes of phosphatidilethanole amine with different surface charges in the presence of tetrapentylammonium chloride. It follows from the analysis of experimental data that to describe the effect of surface charge on membrane conductivity it is necessary to use a complicated model of double electric layer. This study was designed to analyze the diversity of aerobically growing bacteria in a landfill area, compared to those from a forest and a cultivated area at four different depths. The viable cell number of aerobes in the incineration ash layer (3.5(+/-0.4)x10(2)/g) was 10(3)- to 10(4)-fold lower than those in the other areas. On 16S ribosomal DNA similarity analysis of a total of 727 colonies, only the class Bacilli was detected in the incineration ash layer whose pH was extremely high (12.8), while five to seven classes were detected in the forest and the cultivated area. Of the genus Bacillus, B. licheniformis and the recently discovered Bacillus were predominant in the incineration ash layer. These analyses indicate that the incineration ash layer of a landfill might be a source that includes valuable or hitherto unreported Bacillus species. 相似文献

随机共振是一种非零噪声优化系统响应的现象。运用信噪比的评价方式,研究单个Hodgkin-Huxley神经元及其所构建的双层神经元网络中的随机共振,来模拟生物感觉系统中检测微弱信号的随机共振现象。结果表明,单个神经元在阈值下存在噪声优化系统检测性能的随机共振现象,但是最优的噪声强度却随外部信号性质的改变而变化;双层神经元网络不但可以在固定的噪声强度上对一定幅度范围内的阈下信号进行优化检测,而且噪声的存在并没有降低网络对阈上信号的检测能力。 相似文献

Molecular techniques, based on clone library of 18S rRNA gene, were employed to ascertain the diversity of microeukaryotic organisms in sediments from the East Sea. A total of 261 clones were recovered from surface sediments. Most of the clone sequences (90%) were affiliated with protists, dominated by Ciliates (18%) and Dinoflagellates (19%) of Alveolates, phototrophic Stramenopiles (11%), and Cercozoa (20%). Many of the clones were related to uncultivated eukaryotes clones retrieved from anoxic environments with several highly divergent 18S rRNA gene sequences. However, no clones were related to cultivated obligate anaerobic protists. Protistan communities between subsurface layers of 1 and 9 cm shared 23% of total phylotypes which comprised 64% of total clones retrieved. Analysis of diversity indices and rarefaction curve showed that the protistan community within the 1 cm layer exhibited higher diversity than the 9 cm layer. Our results imply that diverse protists remain to be uncovered within marine benthic environments. 相似文献

Surface layers (S-layers) form regular crystalline structures on the outermost surface of many bacteria. Clostridium difficile possesses such an S-layer consisting of two protein subunits. Treatment of whole cells of C. difficile with 5 M guanidine hydrochloride revealed two major proteins of different molecular masses characteristic of the S-layer on SDS-PAGE. In this study 25 isolates were investigated. A high degree of variability in the molecular mass of the two S-layer proteins was evident. Molecular masses ranged from 48 to 56 kDa for the heavier protein and from 37 to 45 kDa for the lighter protein. A further protein component of 70 kDa was detectable in all isolates. No cross-reaction was seen between the two major proteins from isolates that produced different S-layer patterns, and most S-layer proteins from isolates with the same or similar banding patterns did not cross-react. The S-layer proteins, when detected by a combination of Coomassie blue staining and immunoblotting, are a useful marker for phenotyping. 相似文献

The effects of lowering pH from 7 to 5 on the absorption, circular dichroism (MCD) and EPR spectra were studied for Paracoccus halodenitrificans nitric oxide reductase (NOR). Intensities of the characteristic bands for the high spin heme b, that at 592 nm in the absorption spectrum and those at 591 (+) and 606 (-) in the MCD spectrum decreased considerably. Concomitant cryogenic EPR spectrum indicated a drastic increase in the signal intensity due to the high spin heme b at g approximately 6, of which less than 5% had been EPR detectable at pH 7. Cyanide (x40) bound to the high spin heme b center in the reduced NOR irrespective of pH, while a much larger amount of azide (x1000) was necessary to bind to the reduced NOR at an acidic pH, ca. 5. Based on these results the structure and function of the high spin heme b center as the active site of NOR was discussed. 相似文献

The surface energy of the alveolar surfactant layer is determined in the scope of a modification of the structural model of Larsson et al. [(1999) J Disp Sci Technol 20:1-12], according to which this layer is built up of a lipid monolayer adsorbed at the hypophase/air interface and supported by a network of lipid bilayers immersed into the hypophase, i.e., the alveolar liquid. Formulae are derived for the dependence of the specific surface energy of the surfactant layer on the distance between the bilayers constituting the layer. It is shown that at equilibrium this energy can have values comparable with or less than 1 mJ/m2 needed for normal functioning of the alveolus during the respiration cycle. The specific surface energy of the surfactant layer with monolayer-bilayer structure can have such low values only if the layer is of optimal thickness and if the specific line energy of the monolayer-bilayer contact lines is negative and that of the bilayer-bilayer contact lines is positive. It is found that in dynamic regime the change in the specific surface energy of the alveolar surfactant layer with bilayer-monolayer structure is in qualitative agreement with that determined experimentally during lung inflation and deflation. 相似文献

Extracellular acylglycerols are hydrolysed by lipases active at the surface of intact fat cells isolated from rat or human adipose tissue. During short-term incubation, rat fat cells hydrolyse di-[3H]oleyl-[14C]glycerol at a rate of 70 +/- 7.7 mU/10(6) cells (mean +/- S.E.) versus 440 +/- 62 mU/10(6) cells for the hydrolysis of mono-[3H]oleylglycerol; these relatively high lipolytic potencies may serve, among other functions, to counteract the cytolytic effect of both esters. Reaction rates with both substrates are unchanged by addition of various apolipoproteins C and by the nutritional state of the animals. Fat cells incorporate 15-20 per cent of the total [3H]-oleic chains liberated by hydrolysis, with no correlation between uptake and hydrolysis rates. [3H]-oleic chains in cell lipids are found mainly as diacylglycerol (15 per cent) and triacylglycerol (80 per cent). Both lipolytic processes differ from the hydrolysis of trioleylglycerol by cell-bound lipoprotein lipase, which occurs at lower rates (6.5 +/- 0.6 mU/10(6) cells) and depends on apolipoprotein C-II and nutritional state of the animals. The results support the accepted view that lipoprotein lipase and monoacylglycerol lipase are distinct enzymes. Differences between lipoprotein lipase and diacylglycerol lipase activities raise the possibility of different catalytic entities. In conclusion, isolated fat cells in suspension hydrolyse and incorporate lipids. This model should approximate physiological conditions more closely than the use of lipases in the free state. 相似文献

Electron microscopy of the Azotobacter vinelandii tetragonal surface array, negatively stained with ammonium molybdate in the presence of 1 mM calcium chloride, showed an apparent repeat frequency of 12 to 13 nm. Image processing showed dominant tetrad units alternating with low-contrast cruciform structures formed at the junction of slender linkers extending from corner macromolecules of four adjoining dominant units. The actual unit cell showed p4 symmetry, and a = b = 18.4 nm. Distilled water extraction of the surface array released a multimeric form of the single 60,000 molecular-weight protein (S protein) which constitutes the surface layer. The molecular weight of the multimer was estimated at 255,000 by gel filtration, indicating a tetrameric structure of four identical subunits and suggesting that this multimer was the morphological subunit of the S layer. Tetrameric S protein exhibited low intrinsic stability once released from the outer membrane, dissociating into monomers when incubated in a variety of buffers including those which served as the base for defined media used to cultivate A. vinelandii. The tetramer could not be stabilized in these buffers at any temperature between 4 and 30 degrees C, but the addition of 2 to 5 mM Ca2+ or Mg2+ completely prevented its dissociation into monomers. Circular dichroism measurements indicated that the secondary structure of the tetramer was dominated by aperiodic and beta-sheet conformations, and the addition of Ca2+ did not produce any gross changes in this structure. Only the tetrameric form of S protein was able to reassemble in vitro in the presence of divalent cations onto the surface of cells stripped of their native S layer. 相似文献

The apparent viscosity of blood in glass tubes declines with decreasing diameter (F?hraeus-Lindqvist effect) and exhibits a distinctive minimum at 6-7 microm. However, flow resistance in vivo in small vessels is substantially higher than predicted by in vitro viscosity data. The presence of a thick endothelial surface layer (ESL) has been proposed as the primary cause for this discrepancy. Here, a physical model is proposed for microvascular flow resistance as a function of vessel diameter and hematocrit in vivo; it combines in vitro blood viscosity with effects of a diameter-dependent ESL. The model was developed on the basis of flow distributions observed in three microvascular networks in the rat mesentery with 392, 546, and 383 vessel segments, for which vessel diameters, network architecture, flow velocity, and hematocrit were determined by intravital microscopy. A previously described hemodynamic simulation was used to predict the distributions of flow and hematocrit from the assumed model for effective blood viscosity. The dependence of ESL thickness on vessel diameter was estimated by minimizing deviations of predicted values for velocities, flow directions, and hematocrits from measured data. Optimal results were obtained with a layer thickness of approximately 0.8-1 microm for 10- to 40-microm-diameter vessels and declined strongly for smaller diameters, with an additional hematocrit-dependent impact on flow resistance exhibiting a maximum for approximately 10-microm-diameter vessels. These results show that flow resistance in vivo can be explained by in vitro blood viscosity and the presence of an ESL and indicate the rheologically effective thickness of the ESL in microvessels. 相似文献
