The development of advanced methods for accurately monitoring cell apoptosis has extensive significance in the diagnostic and pharmaceutical fields. In this study, we developed a rapid, sensitive and selective approach for the detection of cell apoptosis by combining the site‐specific recognition and cleavage of the DEVD–peptide with quantum dots (QDs)‐based fluorescence resonance energy transfer (FRET). Firstly, biotin‐peptide was conjugated on the surface of AuNPs to form AuNPs‐pep through the formation of an Au‐S bond. Then, AuNPs–pep–QDs nanoprobe was obtained through the connection between AuNPs–pep and QDs. FRET is on and the fluorescence of QDs is quenched at this point. The evidence of UV–vis spectra, transmission electron microscopy (TEM), and Fourier transform infrared (FT‐IR) spectroscopy revealed that the connection was successful. Upon the addition of apoptosis cell lysis solution, peptide was cleaved by caspase‐3, and AuNPs was dissociated from the QDs. At this time, FRET is off, and thus the QDs fluorescence was recovered. The experimental conditions were optimized in terms of ratio of peptide to AuNPs, buffer solution, and the temperature of conjugation and enzyme reaction. The biosensor was successfully applied to distinguishing apoptosis cells and normal cells within 2 h. This study demonstrated that the biosensor could be utilized to evaluate anticancer drugs.  相似文献

Novel CdTe/ZnS quantum dot (QD) probes based on the quenching effect were proposed for the simple, rapid, and specific determination of ammonium in aqueous solutions. The QDs were modified using 3‐mercaptopropionic acid, and the fluorescence responses of the CdTe/ZnS QD probes to ammonium were detected through regularity quenching. The quenching levels of the CdTe/ZnS QDs and ammonium concentration showed a good linear relationship between 4.0 × 10?6 and 5.0 × 10?4 mol/L; the detection limit was 3.0 × 10?7 mol/L. Ammonium contents in synthetic explosion soil samples were measured to determine the practical applications of the QD probes and a probable quenching mechanism was described. The interaction of glucose‐derived carbon quantum dots (CQDs) with silver (Ag) and gold (Au) nanoparticles (NPs) was explored by fluorescence spectroscopy. Both metal NPs cause an efficient quenching of CQD fluorescence, which is likely due to the energy transfer process between CQDs as donors and metal NPs as acceptors. The Stern–Volmer plots were evaluated and corresponding quenching constants were found to be 1.9 × 1010 and 2.2 × 108 M?1 for AgNPs and AuNPs, respectively. The analytical applicability of these systems was demonstrated for turn‐on fluorescence detection of the anti‐cancer drug, 6‐thioguanine. Because the CQD–AgNP system had much higher sensitivity than the CQD–AuNP system, we used it as a selective fluorescence probe in a turn‐on assay of 6‐thioguanine. Under optimum conditions, the calibration graph was linear from 0.03 to 1.0 μM with a detection limit of 0.01 μM. The developed method was applied to the analysis of human plasma samples with satisfactory results.  相似文献

CdTe quantum dots (QDs) capped with different stabilizers, i.e. thioglycolic acid (TGA), 3‐mercaptopropionic acid (MPA) and glutathione (GSH) were investigated as fluorescent probes for the determination of Cu2+. The stabilizer was shown to play an important role in both the sensitivity and selectivity for the determination of Cu2+. TGA‐capped CdTe QDs showed the highest sensitivity, followed by the MPA and GSH‐capped CdTe QDs, respectively. The TGA‐ and MPA‐capped CdTe QDs were not selective for Cu2+ that was affected by Ag+. The GSH‐capped CdTe QDs were insensitive to Ag+ and were used to determine Cu2+ in water samples. Under optimal conditions, quenching of the fluorescence intensity (F0/F) increased linearly with the concentration of Cu2+ over a range of 0.10–4.0 µg/mL and the detection limit was 0.06 µg/mL. The developed method was successfully applied to the determination of Cu2+ in water samples. Good recoveries of 93–104%, with a relative standard deviation of < 6% demonstrated that the developed simple method was accurate and reliable. The quenching mechanisms were also described. A novel fluorescence assay system for glucose was developed with thioglycollic acid (TGA)‐capped CdTe quantum dots (QDs) as probes. The luminescence quantum yield of the TGA‐capped CdTe QDs was highly sensitive to H2O2 and pH. In the presence of glucose oxidase, glucose is oxidized to yield, gluconic acid and H2O2. H2O2 and H+ (dissociated from gluconic acid) intensively quenched the fluorescence of QDs. The experimental results showed that the quenched fluorescence was proportional to the glucose concentration within the range of 0.01–5.0 mm under optimized experimental conditions. Compared with most of the existing methods, this newly developed system possesses many advantages, including simplicity, low cost, high flexibility, and good sensitivity. A novel and sensitive method for the determination of ceftazidime and cefepime in an active pharmaceutical ingredient (API) has been developed based on the fluorescence quenching of poly(ethylene glycol) (PEG)2000‐capped carbon quantum dots (CQDs) prepared using a chemical oxidation method. The quenching of fluorescence intensity is proportional to the concentration of ceftazidime and cefepime over the range of 0.33–3.30 and 0.24–2.40 µg/mL, respectively. The mode of interaction between PEG2000‐capped CQDs and ceftazidime/cefepime in aqueous solutions was investigated using a fluorescence, UV/Vis and Fourier transform infrared spectrometry (FTIR) at physiological pH. UV/Vis and FTIR spectra demonstrated that ground state compounds were formed through hydrophobic interaction the fluorescence quenching of CQDs caused by ceftazidime and cefepime. Development of the fluorescent pH detection method is promising due to the sensitivity, easy operation, and low‐cost, etc. However, traditional organic fluorophores have still some disadvantages such as the tedious preparation and purification as well as low photostability and water solubility, which limits the rapid detection application. Semiconductor quantum dots (QDs) have recently risen to prominence as an alternative for organic fluorophores in fluorescence analysis by virtue of their convenient synthesis and superior optical properties. In this study, we report on sodium 4‐mercaptophenolate functionalized CdSe/ZnS QDs (denoted as ?OPhS‐QDs), which can serve as a selective “on–off” fluorescence probe for aqueous media pH. ?OPhS‐QDs exhibit strong fluorescence in near neutral medium. As a Lewis organic base, ?OPhS‐ moieties on QDs surface easily binds to proton under acidic conditions to yield 4‐mercaptophenol capped QDs (i.e. HOPhS‐QDs), which acts as an efficient hole trapper. As a result, the QDs photoluminescence (PL) is switched off. Under optimal conditions, the present probe exhibits a good linear relationship between fluorescence response and pH values in the pH range 3.0–5.2. Furthermore, the present probe exhibits a high selectivity for proton over other common cations and has been successfully used for pH detection in real water samples.  相似文献

We report a fluorescence resonance energy transfer (FRET) system in which the fluorescent donor is fluorescein isothiocyanate (FITC) dye and the fluorescent acceptor is CdTe quantum dot (QDs). Based on FRET quenching theory, we designed a method to detect the concentration of silver ions (Ag+). The results revealed a good linear trend over Ag+ concentrations in the range 0.01–8.96 nmol/L, a range that was larger than with other methods; the quenching coefficient is 0.442. A sensitive and simple analytical strategy for the detection of sodium cromoglycate (SCG) has been established based on a readily detectable fluorescence quenching effect of SCG for glutathione‐capped (GSH‐capped) CdTe quantum dots (QDs). The fluorescence of GSH‐capped CdTe QDs could be efficiently quenched by SCG through electron transfer from GSH‐capped CdTe QDs to SCG. Under optimum conditions, the response was linearly proportional to the concentration of SCG between 0.6419 and 100 µg/mL, with a correlation coefficient (R) of 0.9964; the detection limit (3δ/K) was 0.1926 µg/mL. The optimum conditions and the influence of coexisting foreign substances on the reaction were also investigated. The very effective and simple method reported here has been successfully applied to the determination of SCG in synthetic and real samples. We here for the first time demonstrate an analytical approach for the highly selective and sensitive detection of amoxicillin (Amox) in aqueous medium based on the fluorescence quenching of quantum dots (QDs). The change in fluorescence intensity of mercaptopropionic acid‐capped cadmium sulphide (MPA‐CdS) QDs is attributed to the increasing concentration of Amox. The results show that the fluorescence quenching of QDs by Amox takes place through both static and dynamic types of quenching mechanism. The fluorescence quenching of QDs with increase in concentration of Amox shows the linear range between 5 μg ml?1 and 30 μg ml?1 and the limit of detection (LOD) is 5.19 μg ml?1. There is no interference of excipients, which are commonly present in pharmaceutical formulation and urine samples. For the practical application approach, the developed method has been successfully applied for the determination of Amox in pharmaceutical formulations and urine samples with acceptable results.  相似文献

Water‐soluble ZnS:Mn quantum dots (QDs) were synthesized using a hydrothermal method with 3‐mercaptopropionic acid as stabilizer. The optical properties of ZnS:Mn QDs were thoroughly investigated by tuning the doping concentration of Mn2+ and the Zn/S precursor ratio, to obtain an optimal parameter for QDs with excellent fluorescence characteristics. ZnS:Mn QDs excited at only one wavelength, however, which seriously limited their further application. Here, a trace Cd ion was doped into a ZnS host, resulting in QD excitation covering a wide adjustable waveband. Furthermore, when a ZnS shell was coated onto the surface of the ZnCdS:Mn QDs, photoluminescence intensity and stability were further enhanced. After coupling with an anti‐CK 19 antibody, the ZnCdS:Mn/ZnS core/shell QDs were able to function by labeling cancer cells, indicating that they could be considered as a suitable bio‐probe for cells and tissue imaging.  相似文献

This mini‐review describes the recent developments in quantum dots‐based nanoprobes in liquid‐phase chemiluminescence (CL) analysis. In the referenced reports, multiple quantum dots (QDs) were adopted as final emission species either after direct oxidation reactions (direct CL) or after chemiluminescence resonance energy transfer (indirect CL). This review does not include papers in which QDs have been used as enhancers, catalysts, carriers or quenchers in chemiluminescence systems. A brief overview on the CL mechanisms of various QDs‐based nanoprobes and their analytical applications over the last decade is given, followed by comments on the future challenges and prospects in this field.  相似文献

A method for quantitative analysis of nitrite was achieved based on fluorescence quenching of graphene quantum dots. To obtain reliable results, the effects of pH, temperature and reaction time on this fluorescence quenching system were studied. Under optimized conditions, decrease in fluorescence intensity of graphene quantum dots (F0/F) showed a good linear relationship with nitrite concentration between 0.007692–0.38406 mmol/L and 0.03623–0.13043 μmol/L; the limits of detection were 9.8 μmol/L and 5.4 nmol/L, respectively. Variable temperature experiments, UV absorption spectra and thermodynamic calculations were used to determine the quenching mechanism, and indicated that it was an exothermic, spontaneous dynamic quenching process. This method was used to analyse urine samples, and showed that it could be applied to analyse biological samples.  相似文献

The continuous spread of highly pathogenic avian influenza virus (AIV) subtype H5N1 is threatening the poultry industry and human health worldwide. Rapid and sensitive diagnostic methods are required for the H5N1 surveillance. In this study, the fluorescent (FL) probe of CdTe quantum dots (QDs) was designed using covalently linked rabbit anti‐AIV H5N1 antibody. Based on these QD–antibody conjugates, a novel sandwich FL‐linked immunosorbent assay (sFLISA) was developed for H5N1 viral antigen detection. The sFLISA allowed for H5N1 viral antigen determination in a linear range of 8.0 × 10?3 to 5.1 × 10?1 μg mL?1 with the limit of detection (LOD) of 1.5 × 10?4 μg mL?1. In comparison with virus isolation for 103 clinic samples, the sensitivity and specificity of sFLISA were found to be 93.6 and 91.1% respectively. The sFLISA supplied a novel approach to rapid and sensitive detection of AIV subtype H5N1 and showed great potential for biological applications in immunoassays. Glibenclamide (GB), as a sulfonylurea‐based medication is commonly prescribed for the treatment of type 2 diabetes. Due to its increasing consumption, there is a need to develop a simple, fast, and reliable detection method to follow its concentration in pharmaceutical and biological samples. Herein, a novel fluorometric method is developed for the sensitive measurement of GB. The method is based on the enhancing effect of GB on the fluorescence emission of mercaptopropionic acid (MPA) capped cadmium telluride quantum dots (CdTe QDs). QDs were synthesized in aqueous solution and were characterized by fluorescence spectroscopy, transmission electron microscopy (TEM), X‐ray diffraction (XRD) and Fourier transform infrared spectroscopy (FT‐IR). Fluorescence intensity of QDs was enhanced by adding GB in a very low concentration. The effect of operative factors such as pH, buffer, contact time and concentration of CdTe QDs were investigated and in the optimized condition, a linear increase was achieved for the emission intensity of QDs by increasing GB concentration in the range 49–345 ng mL?1, with a detection limit of 17.84 ng mL?1. The offered method has an acceptable precision (relative standard deviations were < 2.8%) and was satisfactorily applied for the determination of GB in pharmaceutical products and human urine samples.  相似文献

N‐acetyl‐l ‐cysteine‐capped Mn‐doped ZnS quantum dots (QDs) were prepared by hydrothermal methods. It could emit phosphorescence at 583 nm with the excitation wavelength at 315 nm. The phosphorescence intensity of QDs could be quenched dramatically by increasing the concentration of Co2+ ion. The novel phosphorescence sensor based on N‐acetyl‐l ‐cysteine‐capped QDs was developed for detecting Co2+ ion with a linear dynamic range of 1.25 × 10–6–3.25 × 10–5 m . The limit of detection and RSD were 6.0 × 10–8 m and 2.3%, respectively. Interference experiments showed excellent selectivity over numerous cations such as alkali, alkaline earth and transitional metal ions. The possible quenching mechanism was also examined by phosphorescence decays. The proposed phosphorescence method was further applied to the trace determination of Co2+ ion in tap and pond water samples with recoveries of 97.75–103.32%. Mercaptopropionic acid (MPA)–capped cadmium telluride (CdTe) quantum dot (QDs) fluorescent probes were synthesized in aqueous solution and used for the determination of salicylic acid. The interaction between the MPA–capped CdTe QDs and salicylic acid was studied using fluorescence spectroscopy and some parameters that could modify the fluorescence were investigated to optimize the measurements. Under optimum conditions, the quenched fluorescence intensity of MPA–capped CdTe QDs was linearly proportional to the concentration of salicylic acid in the range of 0.5–40 µg mL–1 with a coefficient of determination of 0.998, and the limit of detection was 0.15 µg mL–1. The method was successfully applied to the determination of salicylic acid in pharmaceutical products, and satisfactory results were obtained that were in agreement with both the high pressure liquid chromatography (HPLC) method and the claimed values. The recovery of the method was in the range 99 ± 3% to 105 ± 9%. The proposed method is simple, rapid, cost effective, highly sensitivity and eminently suitable for the quality control of pharmaceutical preparation. In this work, a turn‐off photoluminescent sensing proof‐of‐concept based on blue luminescent graphene quantum dots (GQDs) as the fluorescent probe was developed. For that purpose, GQDs optical response was related with the catalytic enzymatic activity of alkaline phosphatase (ALP), in the presence of hydroquinone diphosphate (HQDP). The hydrolysis of HQDP by ALP generated hydroquinone (HQ). The oxidation of HQ, enzymatically produced, to p‐benzoquinone (BQ) resulted in the quenching of GQDs fluorescence (FL). Therefore, the developed luminescent sensing mechanism allowed the FL quenching with ALP activity to be related and thus quantified the concentration of ALP down to 0.5 nM of enzyme. This innovative design principle appears as a promising tool for the development of enzymatic sensors based on ALP labeling with fluorescent detection or even for direct ALP luminescent quantification in an easy, fast and sensitive manner.  相似文献

In this paper, based on the fluorescence of carbon quantum dots (CQDs) quenched by mercury ions (Hg2+) and the nonresponse of Hg2+ to rhodamine B fluorescence, a dual emission ratio fluorescence sensor was constructed to realize the quantitative detection of Hg2+. Under excitation at 365 nm, the fluorescence spectrum showed double emission peaks at 437 nm and 590 nm, corresponding to the fluorescence emissions of CQDs and rhodamine B, respectively. This method quantitatively detected Hg2+ based on the linear relationship between the ratio of the intensities of the two emission peaks F437/F590 and the concentration of Hg2+. The detection range was 10–70 nM, and the limit of detection (S/N = 3) was 3.3 nM. In addition, this method could also realize the qualitative and semiquantitative detection of Hg2+ according to the fluorescence colour change of the probe under ultraviolet light. After various evaluations, the method could be successfully applied to the quantitative and visual detection of Hg2+ in tap water, and demonstrated excellent selectivity, anti-interference performance, and repeatability of the method.  相似文献

A sensitive and simple method for the determination of enoxacin (ENX) was developed based on the fluorescence quenching effect of ENX for glutathione (GSH)‐capped CdTe quantum dots (QDs). Under optimum conditions, a good linear relationship was obtained from 4.333 × 10?9 mol?L?1 to 1.4 × 10?5 mol?L?1 with a correlation coefficient (R) of 0.9987, and the detection limit (3σ/K) was 1.313 × 10?9 mol?L?1. The corresponding mechanism has been proposed on the basis of electron transfer supported by ultraviolet–visible (UV) light absorption, fluorescence spectroscopy, and the measurement of fluorescence lifetime. The method has been applied to the determination of ENX in pharmaceutical formulations (enoxacin gluconate injections and commercial tablets) with satisfactory results.
