Three lines of evidence indicated that methionine sulfoxide is transported by the high-affinity methionine and glutamine transport systems in Salmonella typhimurium. First, methionine-requiring strains (metE) which have mutations affecting both of these transport systems (metP glnP) were unable to use methionine sulfoxide as a source of methionine. These strains could still grow on L-methionine because they possessed a low-affinity system (or systems) which transported L-methionine but not the sulfoxide. A methionine auxotroph with a defect only in the metP system, which was dependent upon the glnP+ system for the transport of methionine sulfoxide, was inhibited by L-glutamine because glutamine inhibited the transport of the sulfoxide by the glnP+ system. Second, a metE metP glnP strain could be transduced at either the metP or glnP genes to restore its ability to grow on methionine sulfoxide. Third, the transport of [14C]methionine sulfoxide was inhibited by methionine and by glutamine in the metP+ glnP+ strain. No transport was detected in the metP glnP double-mutant strain.  相似文献

The systems which transport methionine in Salmonella typhimurium LT2 have been studied. Fourteen mutants, isolated by three different selection procedures, had similar growth characteristics and defects in the specific transport process showing a Km of 0.3 microM for L-methionine, and therefore lack the high-affinity, metP transport system. The sites of mutation in four of the mutants were shown by P1-mediated transduction to be linked (0.3 to 1.1%) with a proline marker located at unit 7 on the S. typhimurium chromosome. The high-affinity system was subject to both repression and transinhibition by methionine, and it may also be regulated by the metJ and metK genes. There appeared to be at least two additional transport systems with relatively low affinities for methionine in the metP763 mutant strain, with apparent Km values for methionine of 24 microM and approximately 1.8 mM. The latter system, with a very low affinity for methionine, was inhibited by leucine. In addition, methionine inhibited leucine transport, suggesting that one of the low-affinity methionine transport systems may actually be a leucine transport system.  相似文献

The transport and utilization of D-methionine was investigated in several strains of Escherichia coli K-12. Wild-type cells exhibit a single transport system with a Km of 1.16 muM. This activity exhibits a specificity similar to that of the uptake of L-methionine. The activity toward the D-isomer and the high-affinity uptake of L-methionine are lost in strains mutant in metD, along with the ability to utilize D-methionine as methionine source. Both activities respond identically to gene dosage of metD and are both restored in revertants or transductants. However, although L-methionine is a potent inhibitor of D-methionine uptake, D-methionine has little or no effect on the uptake of the L-isomer. No mutants altered in the uptake of only one of the two isomers were found in a screening. Regulation of both activities was similar in their response to the internal methionine pool, and some evidence was suggestive of partial repressive control of these activities. The evidence is most consistent with the role of the metD product as a common step for two methionine-specific uptake systems, but other gene products may represent the initial substrate binding sites. This system also appears to be involved in the uptake of N-acetyl methionine and methionine sulfoxide and methionine sulfoximine. The uptake of the keto analogue of methionine, alpha-keto-gamma-methiol butyrate, appears to be mediated by a separate system specific for alpha-keto straight-chain acids 5- to 6-carbon units in length.  相似文献

The formation of the high-affinity (Km equal to 0.2 muM) L-glutamine transport system of Escherichia coli strain 7 (Lin) appears to be subject to the same major control as the glutamine synthetase (EC 6.3.1.2) of this gram-negative organism. Culture of cells under nitrogen-limited conditions provides maximum derepression of both the glutamine synthetase and the glutamine transport system. Nutritional conditions providing a rich supply of ammonium salts or available sources of nitrogen, i.e., conditions which repress the formation of glutamine synthetase, provide three- and 20-fold repression, respectively, of the glutamine transport system. Culture of cells with glutamine supplements of 2 mM does not increase the repression of high-affinity glutamine transport system beyond the level observed in the absence of glutamine. A second kinetically distinct low-affinity component of glutamine. A second kinetically distinct low-affinity component of glutamine uptake is observed in cells cultured with a glutamine-depleted nutrient broth. This second component is associated with the appearance of glutaminase A (EC 3.5.1.2) and asparaginase I (EC 3.5.1.1), a periplasmic enzyme. Parallel changes were observed in the levels of the high-affinity glutamine transport system and the glutamine synthetase when cells were cultured with the carbon sources: glucose, glycerol, or succinate.  相似文献

The presence of a sodium-dependent, saturable uptake process is described in basolateral membranes of rat renal cortex for L-glutamine. Concentration-dependence studies indicate the presence of multiple transport systems withK m 1 of 0.032 mM and V1 of 0.028 nmol/mg of protein per min, andK m 2 of 17.6 mM and V2 of 17.6 nmol/mg of protein per min. Lysine completely inhibits the high-affinity, low-capacityK m system and partially inhibits the low-affinity, high-capacity system. Cystine and other dibasic amino acids also affect glutamine uptake.  相似文献

Alanine and glutamine transport have been studied during red blood cell maturation in the rat. Kinetic parameters of Na+-dependent L-alanine transport were:K m 0.43 and 1.88 mM andV max 158 and 45 nmoles/ml ICW/min for reticulocytes and erythrocytes, respectively. During red cell maturation in the rat there is a loss of capacity and affinity of the system ASC for L-alanine transport. The values for Na+-dependent L-glutamine transport in reticulocytes wereK m 0.51 mM andV max 157 nmoles/ml ICW/min. On the other hand, a total loss of L-glutamine transport mediated by both N and ASC systems is demonstrated in mature red cells. This seems to indicate that during rat red cell maturation the system N disappears. Furthermore, the system ASC specificity in mature cells changes, and glutamine enters the red cell by non-mediated diffusion processes.  相似文献

Summary Certain metH mutants (which lack the B12-dependent homocysteine transmethylase) give rise to revertants resistant to the methionine analogue, ethionine. The revertants retain the original metH mutation and its suppression is due to two mutations, supI and supII. The supI mutation, which confers ethionine resistance, appears to be a mutation in the methionine regulatory gene, metJ, but the location and nature of supII have not been determined. It is possible that suppression results from a direct association between the metH and metJ gene products or by the introduction of an alternative pathway of homocysteine methylation.  相似文献

Abstract: Uptake of L-glutamine (2 mM) by rat brain cortex slices against a concentration gradient is markedly inhibited (40%) by branched-chain Lamino acids (1 mM), L-phenylalanine (1 mM), or L-methionine (1 mM); that of L-asparagine (2 mM) is much less affected by these amino acids. Other amino acids investigated have little or no effect on cerebral L-glutamine uptake. The suppressions of L-glutamine uptake by the inhibitory amino acids are apparently blocked by high [K+], which itself has little or no effect on glutamine uptake. This abolition of suppression is partly explained by high [K+] retention of endogenous glutamine; in the absence of Ca2+ such retention disappears. The inhibitory amino acids (1 mM) also enhance the release of endogenous glutamine, exogenous glutamine with which slices have been loaded, or glutamine synthesized in the slices from exogenous glutamate. The enhanced release of endogenous glutamine is diminished by high [K+]. The suppression of glutamine uptake by the branched-chain amino acids is independent of the concentration of glutamine at low concentrations (0.25–0.5 mM), indicating non-competition, but is reduced with high concentration of glutamine. The inhibition by L-phenylalanine is noncompetitive. L-Glutamine (2 mM) exerts no inhibition of the cerebral uptakes of the branched-chain L-amino acids or Lphenylalanine (0.25–2 mM). The inhibitory amino acids are as active in suppressing L-glutamine uptake with immature rat brain slices as with adult, although the uptake, against a gradient, of L-glutamine in the infant rat brain is about one-half that in the adult. They are also just as inhibitory on the concentrative uptake of L-glutamine by a crude synaptosomal preparation derived from rat brain cortex. Such a nerve ending preparation takes up L-glutamine (0.25 mM), against a gradient, at about ninefold the rate at which it is taken up by cortex slices (for equal amounts of protein), and the uptake process is markedly suppressed by high [K+] in contrast to the effects of high [K+] with slices. The possible physiological and pathological consequences of the suppression of glutamine uptake are discussed.  相似文献

Summary A low-affinity and a high-affinity sylose proton symport operated simultaneously in both starved and non-starved cells of Pichia stipitis. Glucose competed with xylose for transport by the low-affinity system and inhibited xylose transport by the high-affinity system non-competitively. The low affinity system was subject to substrate inhibition when glucose but not when xylose was the substrate. The differences between the characteristics of monosaccharide transport by Pichia stipitis and its imperfect state, Candida shehatae, are discussed.  相似文献

Summary A total of 30 metP mutations defective in the high-affinity methionine transport system were linked in P1 transduction to the zaf-1351::Tn10 insertion mutation at min 5–6 on the Salmonella typhimurium chromosome map. The relationship of metP to several other markers in this region was studied. Methionine transport was strongly inhibited by arsenate, suggesting that the metP system belongs to the shock-sensitive category and possesses a periplasmic binding protein. However, other experiments provided less clear cut evidence. Transport activity was only slightly reduced by osmotic shock; a methionine binding activity was detected in shock fluids from the wildtype strain, and although this activity was reduced by 50% in 3 frameshift mutants, mutants without any activity were not found. No differences were detected in the shock fluids of the 30 mutants when examined by SDS-polyacrylamide gel electrophoresis.  相似文献

Ethylene production by axial and cotyledonary tissues excised from Xanthium pennsylvanicum Wallr. seeds was markedly (up to 5-fold) stimulated by the D-isomers of phenylalanine, valine, leucine, threonine, methionine and eithionine while the L-isomers caused no such effect. Responsiveness of these seed tissues to D-methionine appeared soon after the beginning of imbibition, reached a maximum after 6–12 and 12–24 h for the axial and cotyledonary tissues, respectively, and then decreased sharply. D-Phenylalanine and D-methionine also stimulated ethylene production in seed tissues of X. canadense Mill. and in cotyledonary segments from seeds of Helianthus annuus L., Cucurbita moschata Duch. and Vigna radiata (L.) Wilczek. The endogeneous ethylene production and the D-amino-acid-stimulated ethylene production by the seed segments was strongly inhibited by aminoethoxyvinyl glycine, a potent inhibitor of ethylene synthesis from L-methionine.  相似文献

The escape of several enzymes from “ammonia catabolite repression” in gdhA? (NADP-linked glutamate-dehydrogenase-less) mutants, as well as in gdhCR mutants of Saccharomyces cerevisiae, does not involve glutamine synthetase, either as a positive or as a negative control element. A glutamine-synthetase-less mutant (gln?) was used in this demonstration.In addition to its derepressing effect on the NAD-linked glutamate dehydrogenase, the gdhCR mutation releases “nitrogen catabolite repression” on arginase and allatoinase, as well as glutamine repression on glutamine synthetase. A gdhCS mutation was used to demonstrate that these effects are not mediated through the NAD-linked glutamate dehydrogenase.  相似文献

The fresh water polyp Hydra is noted for its ability to regenerate missing body parts. Transplantation experiments indicate that the control of regeneration includes signalling over long distances. These signals appear to include diffusible morphogens, activators and inhibitors. In order to elucidate the nature of such signals, tissue of polyps was homogenized and fractionated. The fractions were tested for their ability to hinder head regeneration. The active factor within these fractions was determined to be methionine. Both the active fractions and L-methionine were found to antagonize not only head regeneration but also foot regeneration. Budding, the asexual means of reproduction, is antagonized. L-methionine acts in micromolar concentrations while the stereoisomer D-methionine does not. L-methionine may act by providing a methyl group in transmethylation processes.  相似文献

Summary Recombinant DNA plasmids containing inserts from the glnA region of Escherichia coli were used to study the expression of gln, hut, and nif operons in a regulation defective mutant (GlnHutNif) of Klebsiella pneumoniae, KP5060. Genes adjacent to the C-terminal end of glnA on the E. coli chromosome were able to derepress hut and nif operons in K. pneumoniae in the absence of glnA product. However, complete derepression of nif operons required inclusion of the segment adjacent to the N-terminal end of the glnA region of the E. coli chromosome along with the C-terminal end segment. In the absence of functional glnA, such a fully derepressed strain expressed nif and hut constitutively indicating a role for the catalytic activity of glutamine synthetase in repression of the genes under nitrogen control.  相似文献

Plant roots accumulate potassium from a wide range of soil concentrations, utilizing at least two distinct plasma membrane uptake systems with different affinities for the cation. Although details on the structure and function of these transporters are beginning to emerge many prominent questions remain concerning how these proteins function in planta. Such questions can be addressed through the use of well-defined transport mutants. Csi52, a caesium-insensitive mutant of Arabidopsis thaliana which is defective in potassium transport, is further characterized here using conventional electrophysiology, patch-clamp and radiometric approaches to identify the nature of the potassium transport lesion. Rb+ uptake experiments reveal a reduced uptake in csi52 in both the high- and low-affinity uptake range. Patch-clamp analysis indicates that the activity of the predominant inward rectifying channel observed in wild-type cells is extremely low in root protoplasts isolated from csi52, whereas outward rectifying channel activity is comparable between wild-type and mutant. Rb+ uptake studies show that in both wild-type and csi52 the high-affinity uptake pathway is considerably less sensitive to Cs+ than the low-affinity pathway with K1/2 values for Cs+ of around 1.3 and 0.2 mM, respectively. Furthermore, K+ starvation leads to a larger relative increase in high-affinity K+ uptake in the mutant than the wild-type. The results demonstrate the Cs+ sensitivity of each individual uptake pathway is comparable in wild-type and csi52 but the high-affinity pathway is less Cs+ sensitive (in both wild-type and csi52). Therefore, the larger shift toward high-affinity uptake in the mutant compared with the wild-type under K+-starvation conditions will endow the mutant with a higher degree of overall Cs+ resistance. The data supply evidence for the hypothesis that the csi52 mutation lies within a gene that regulates the activity of several potassium transport systems and coordinates their relative contribution to overall root K+ uptake.  相似文献

Clonal lines of glial, neuronal, and nonneural origin accumulate choline via a high-affinity carrier-mediated transport system withK m in the range of 10–14 M. These cell lines also accumulate choline by a second system that is not saturable at 10 mM choline, and that may represent diffusion. The transport of choline in glial cells differs from that seen in neuronal cells with respect to its Na+ requirement. The omission of Na+ from the incubation medium reduces high-affinity choline transport in neuronal cells and enhances it in glial cells. Kinetic analysis of the data indicates that reversible cholinesterase inhibitors and hemicholinium-3 (HC-3) inhibit the high-affinity transport system for choline. On the other hand, the diffusional or low-affinity component of choline transport in either cell type appears to have no Na+ requirement and is unaffected by either cholinesterase inhibitors or 10–4 M HC-3. The neuronal-glial differences in the Na+ requirement of choline transport may be related to the coupling of transport to choline metabolism, which differs in the two cell types. The presence of a high-affinity transport system for choline in clonal glial lines used as models of normal glia suggest that glia may modulate the availability of choline for acetylcholine synthesis at cholinergic synapses.  相似文献

Abstract: Choline mustard aziridinium is a potent, irreversible and selective blocker of sodium-dependent, high-affinity transport of choline into rat forebrain synaptosomes; it was found to be 30 times less potent against low-affinity transport of choline. The IC50 value for high-affinity transport was 0.94 μM, compared to 29 μM for low-affinity uptake. The inhibitory action of choline mustard aziridinium ion on high-affinity transport of choline was graded with respect to time; a 12-fold increase in potency was obtained by increasing the inhibitor preincubation times from 1 to 30 min. Low concentrations of choline mustard aziridinium ion could produce significant blockade of choline carriers providing the exposure time was prolonged. The characteristics of the blockade of synaptosomal high-affinity choline transport by choline mustard aziridinium ion also changed depending upon preincubation time. The kinetics of inhibition of high-affinity choline transport by choline mustard aziridinium ion showed apparent competitive inhibition initially, followed by noncompetitive characteristics at longer preincubations with inhibitor. The rate of irreversible inhibition of carriers by this nitrogen mustard analogue would appear to be rapid; the rate constant was determined to be 5 × 10?2 s?1for micromolar concentrations of inhibitor. This action may preclude the transport of the mustard analogue into the nerve terminal, although initially some reversible binding with the carrier may result in the translocation of some choline mustard aziridinium ion into the presynaptic ending. The progressive alkylation of high-affinity carriers by the analogue could indicate the presence of excess carrier sites in the presynaptic membrane, or subpopulations of carriers in an inactive state in equilibrium with active carriers. A model is described for the inhibitory action of choline mustard aziridinium ion on synaptosomal high-affinity choline carriers.  相似文献
