Addenda to the Revisions of the Genera <Emphasis Type="Italic">Gergithus</Emphasis> Stål and <Emphasis Type="Italic">Hemisphaerius</Emphasis> Schaum (Hemiptera,Auchenorrhyncha, Fulgoroidea: Issidae)

The genus Gnezdilovius Meng, Webb et Wang, 2017 is revised. Maculergithus Constant et Pham, 2016, which was described as a subgenus of Gergithus Stål, 1870, is upgraded to a genus. Ishiharanus Hori, 1969 is reinstalled from synonymy with Gergithus and considered a valid name. Two new genera are erected, Ceratogergithus Gnezdilov, gen. n. (type species: Gergithus spinosus Che, Zhang et Wang, 2007) and Ophthalmosphaerius Gnezdilov, gen. n. (type species: Hernisphaerius trilobulus Che, Zhang et Wang, 2006). Hernisphaerius bistriatus Schumacher, 1915, Gergithus carbonarius Melichar, 1906, G. rosticus Chan et Yang, 1994, G. nummarius Chan et Yang, 1994, and G. rotundus Chan et Yang, 1994 are transferred to the genus Epyhemisphaerius Chan et Yang, 1994, Gergithus quinquemaculatus Che, Zhang, Wang, 2007—to the genus Maculergithus, Gergithus chelatus Che, Zhang, Wang, 2007 and G. pseudotessellatus Che, Zhang, Wang, 2007—to the genus Ceratogergithus, Hernisphaerius binocularis Chen, Zhang, Chang, 2014—to the genus Ophthalmosphaerius, and Gergithus robustus hoozanensis Schumacher, 1915—to the genus Gnezdilovius. The male genitalia of Gergithus herbaceus (Kirby, 1891) and Hernisphaerius interclusus Noualhier, 1896 are illustrated for the first time.

Genus <Emphasis Type="Italic">Oxymetopon</Emphasis> (Microdesmidae: Ptereleotrinae) in the coastal areas of Province Khanh Hoa (Vietnam,South China Sea)

Species of the genus Oxymetopon of the fauna of Vietnam are revisited. New species O. curticauda sp. n. is described. New synonymy of O. compressum Chan, 1966 = O. formosum Fourmanoir, 1967 is established. New traits for the species identification are revealed. A key to the species was compiled.     

Isolation and characterization of restriction endonuclease EclHKI from an Enterobacter cloacae strain

An Enterobacter cloacae strain, producing restriction enzyme EclHKI, was isolated from a decaying potato. The enzyme is an isoschizomer of Eam1105I, which recognizes and cleaves GACNNN!NNGTC. EclHKI was produced at high activity (4×10⁴ U/g wet cells) and was purified from contaminants which interfere with restriction digestion by passing the cell lysate through DEAE-Sephacel and heparin columns. Effects of Chan Su,a traditional Chinese medicine,on the calcium transients of isolated cardiomyocytes: cardiotoxicity due to more than Na,K-ATPase blocking

Extracts of Chan Su, a traditional Chinese medication used as a topical anesthetic and cardiac medication, were incubated with cardiomyocytes that had been loaded with a calcium specific fluorescent probe. Calcium transients were measured by real-time fluorescence spectrophotometry following treatment. The transients were rapidly abolished following addition of a moderate concentration of the extract (400 ng/ml), resulting in high levels of intracellular calcium, while the lower amount (40 ng/ml) blocked the sodium-potassium adenosine triphosphatase. Treatments with ouabain and nifedipine were also made, either prior to, or after the addition of the Chan Su, in an attempt to better delineate the site(s) of action. The moderate concentration of Chan Su (400 ng/ml) extract caused the myocytes to cease beating within seconds of addition, even in experiments when saturating concentrations of nifedipine or ouabain had been previously added to the cells. As expected bufalin, the active component of Chan Su has similar effects. Our findings indicate that this compound is extremely cardiotoxic, even in small dose and acts rapidly to alter intracellular calcium stores in cardiomyocytes and possibly acts at sites other than the Na(+)+K(+) ATPase, either directly, or indirectly via changes in calcium concentrations.     

Monomeric G-protein, Rhes, is not an imidazoline-regulated protein in pancreatic beta-cells

The monomeric G-protein, Rhes, is a candidate imidazoline-regulated molecule involved in mediating the insulin secretory response to efaroxan [S.L. Chan, L.K. Monks, H. Gao, P. Deaville, N.G. Morgan, Identification of the monomeric G-protein, Rhes, as an efaroxan-regulated protein in the pancreatic beta-cell, Br. J. Pharmacol. 136 (1) (2002) 31-36]. This suggestion was based on observations regarding changes in Rhes mRNA expression in rat islets and pancreatic beta-cells after prolonged culture with efaroxan, leading to desensitization of the insulin response to the compound. To verify this report, we have evaluated the effects of the imidazoline compounds efaroxan and BL11282 on Rhes mRNA expression in isolated rat pancreatic islets maintained in conditions identical to those used by Chan et al. The results demonstrate that desensitization of the insulin response to efaroxan, or to another imidazoline, BL11282, does not change Rhes mRNA expression levels. Transfection of MIN6 cells with plasmids containing Rhes or Rhes-antisense also does not alter efaroxan- or BL11282-induced insulin secretion. Together, these data do not support the hypothesis that Rhes is an imidazoline-regulated protein.

Simultaneous determination of five main active bufadienolides of Chan Su in rat plasma by liquid chromatography tandem mass spectrometry

To study the pharmacokinetics of Chan Su, a sensitive and selective method was developed and validated for the determination of five main bufadienolides (cinobufagin, resibufogenin, bufalin, bufotalin and arenobufagin) in rat plasma. The analytes were extracted by liquid-liquid extraction with ethyl acetate after internal standard (IS, caudatin) spiked. The separation was performed by a ZORBAX SB-C18 column (3.5 microm, 2.1 mmx100 mm) and a C18 guard column (5 microm, 4.0 mmx2.0 mm) with an isocratic mobile phase consisted of acetonitrile-water-formic acid (50:50:0.05, v/v/v) at a flow rate of 0.3 mL/min. The Agilent G6410A triple quadrupole LC/MS system was operated under the multiple reaction monitoring mode (MRM) using the electrospray ionization technique in positive mode. The nominal retention times for cinobufagin, resibufogenin, bufalin, bufotalin, arenobufagin and caudatin were 3.07, 3.55, 2.30, 1.62, 1.22 and 3.43 min, respectively. All analytes showed good linearity in a wide concentration range (r>0.995) and their lower limits of quantification (LLOQ) were all 1.0 ng/mL. The method was linear for all analytes with correlation coefficients>0.995 for all analytes. The average extract recoveries of the five analytes from rat plasma were all over 85%, the precisions and accuracies determined were all within 15%. This method has been successfully applied to pharmacokinetic study of Chan Su in rats following oral administration.     

Does daily climate variation have an effect on species’ elevational range size?

In their recent paper published in Science (2016, 351 , 1437–1439), Chan et al. analysed 137 montane gradients, concluding that they found a novel pattern—a negative relationship between mean elevational range size of species and daily temperature variation, which was claimed as empirical evidence for a novel macrophysiological principle (Gilchrist's hypothesis). This intriguing possibility was their key conceptual contribution. Unfortunately, as we show, the empirical evidence was flawed because of errors in the analyses and substantial sampling bias in the data. First, we re‐ran their analyses using their data, finding that their model should have been rejected. Second, we performed two additional re‐analyses of their data, addressing biases and pseudoreplication in different ways, both times again rejecting the evidence claimed to support Gilchrist's hypothesis. These results overturn the key empirical findings of Chan et al.'s study. Therefore, the “macrophysiological principle” should be regarded as currently remaining unsupported by empirical evidence.
