The cell is presented as a biological ion exchanger resin. The similarities between ion accumulating cells and ion exchanger resins are correlated. The kinetic characteristics of biological ion exchange are shown to be amenable to analysis by a model commonly used for ion exchanger resins. The theories of ion exchange equilibria currently in use with ion exchanger resins are reviewed with their suitability for adaptation to biological ion exchange in mind.  相似文献

Cupric sulfate is reduced by ascorbate to the cuprous ion. The cuprous ion is then oxidized back to the cupric form by oxygen. A steady state concentration of the cupric ion is thus established to maintain a continuous oxidation of ascorbate in the presence of a trace amount of copper. In the presence of urate there is an instantaneous oxidation of ascorbate by the cupric ion. However, urate complexes with the cuprous ion and thus reduces the steady state concentration of the cupric ion. This decrease in cupric ion concentration interrupts ascorbate oxidation. The interaction of urate and cuprous ion was documented by analysis of uv absorption spectrum and the isolation of urate-Cu+ by high-pressure liquid chromatography.  相似文献

Quantum chemical model calculations were carried out for modeling the ion transport through an isolated ion channel of a cell membrane. An isolated part of a natural ion channel was modeled. The model channel was a calixarene derivative, hydrated sodium and potassium ions were the models of the transported ion. The electrostatic potential of the channel and the energy of the channel-ion system were calculated as a function of the alkali ion position. Both attractive and repulsive ion-channel interactions were found. The calculations - namely the dependence of the system energy and the atomic charges of the water molecules with respect to the position of the alkali ion in the channel - revealed the molecular-structural background of the potassium selectivity of this artificial ion channel. It was concluded that the studied ion channel mimics real biological ion channel quite well.  相似文献

The rat diaphragm has been used to investigate the neuromuscular blocking action of acetylcholine mustard which yields a potent nicotinic agonist, an aziridinium ion, in aqueous medium. Evidence was obtained that the acetylcholine mustard aziridinium ion impaired neuromuscular activity when the phrenic nerve was stimulated and that the ion did not directly inhibit muscle contraction. Impairment of neuromuscular activity was characterized by a latent period and depended both on the concentration of aziridinium ion and the frequency of stimulation of the phrenic nerve. Elevated concentrations of Ca-2+ and choline changed the response of the rat diaphragm to the aziridinium ion, the former increasing the rate of development of neuromuscular block and the latter protecting against neuromuscular block. These results indicated that the aziridinium ion may act either at the site of choline uptake or have an effect on acetylcholine synthesis in the nerve ending and that impairment of neuromuscular transmission in the rat diaphragm involved the availability of acetylcholine. Similar results were obtained with acetylcholine mustard aziridinium ion subjected to alkaline hydrolysis. This substance is thought to be choline mustard aziridinium ion. Although difficult to prove with the rat diaphragm it is possible that acetylcholinesterase of this preparation could hydrolyze acetylcholine mustard aziridinium ion at the neurotransmitter site and the resultant choline mustard aziridinium ion would interfere with the uptake of choline and eventually prevent neuromuscular transmission. This hemicholinium-like hypothesis for the mechanism of action of choline mustard aziridinium ion is compatible with reported date for toxicity of acetylcholine mustard aziridinium ion in the mouse.  相似文献

Plant nutrient acquisition from concentrated soil solutions is governed by low-affinity transport systems in the plasma membranes of root cells. In this Opinion article, we illustrate that for six major nutrient ions, in addition to influx mediation by low-affinity transporters, high rates of ion cycling at the plasma membrane are a hallmark of nutrient transport at elevated external concentrations. This phenomenon is characterized by high rates of ion influx and concurrent high efflux of the same ion, resulting in efflux:influx ratios close to 1. Prolonged maintenance of futile cellular ion cycling can be energetically unfavorable and detrimental to plant growth and survival. We discuss how ion cycling can contribute to the toxicities of ions such as Na(+) and NH(4)(+) in the low-affinity range of ion provision. We also argue that cellular ion cycling makes the reliable measurement of ion influxes in the low-affinity range a formidable challenge.  相似文献

When grown on iron-salt medium supplemented with the bisulfite ion, Leptospirillum ferrooxidans was much more sensitive to the ion than was Thiobacillus ferrooxidans. The causes of the sensitivity of L. ferrooxidans to the bisulfite ion were studied. The bisulfite ion completely inhibited the iron-oxidizing activities of L. ferrooxidans and T. ferrooxidans at 0.02 and 0.2 mM, respectively. A trapping reagent for the bisulfite ion, formaldehyde, completely reversed the inhibition. The treatment of intact cells with 1.0 mM bisulfite ion for 1 h and washing the bisulfite ion from the cells had no harmful effects on the iron-oxidizing activity of T. ferrooxidans. However, the treatment of L. ferrooxidans with 0.1 mM bisulfite ion for 1 h completely destroyed the iron-oxidizing activity. T. ferrooxidans had sulfite:ferric ion oxidoreductase activity. In contrast, a quite low level of sulfite:ferric ion oxidoreductase activity was found in L. ferrooxidans, suggesting that it is much more difficult for L. ferrooxidans to oxidize the bisulfite ion to the less harmful sulfate than it is for T. ferrooxidans. These results suggest that the sensitivity of L. ferrooxidans to the bisulfite ion is due to a lack of an active sulfite:ferric ion oxidoreductase and the sensitivity of its iron oxidase to bisulfite ion.  相似文献

离子通道亚型与其基因共表达的关联对研究离子通道功能有重要意义。文章采用主成分分析和模糊C-均值聚类算法对数据进行分析, 将方法应用到人类和小鼠两套表达谱数据, 结果发现离子通道亚型中钾离子通道、钙离子通道、氯离子通道和受体激活型离子通道的表达谱聚类结果与生物学分类有较好的一致性, 体现了离子通道亚型在mRNA水平上的共表达, 并证实了通过离子通道表达谱能很好的对离子通道的功能亚型进行分类。  相似文献

溶酶体离子通道蛋白异常引起溶酶体功能障碍是导致阿尔茨海默病(Alzheimer’s disease,AD)和帕金森病(Parkinson’s disease,PD)等神经退行性疾病的重要因素.溶酶体离子通道蛋白调节溶酶体内离子稳态、溶酶体膜电压以及溶酶体的酸度.溶酶体离子通道蛋白的结构或功能缺陷会引起溶酶体降解功能障碍,导致神经退行性疾病的发生发展.在这篇综述中,我们总结了各种离子通道蛋白调节溶酶体功能的过程及机制,以及离子通道蛋白异常参与神经退行性疾病的过程和机制.调节离子通道蛋白改善溶酶体的功能、促进异常聚集蛋白的清除,是神经退行性疾病治疗的潜在途径.  相似文献

Ion channels form a complex class of membrane transport proteins. They are often classified according to their selective permeability for particular ion species as well as to their gating properties, which are controlled by either membrane voltage, ligand binding or physical stimuli. Ion transport through membrane pores embedded in protein channel complexes possesses both a chemical and an electrical dimension with ion flux causing both charge separations as well as changes in ionic concentrations. This electrochemical double-nature of ion transport is reflected in the two main physiological domains of ion channel function: in excitable cells many ion channels predominately control membrane voltage to generate fast electrical signaling, while epithelial or intracellular ion channels are mainly involved in directional ion transport. Given this framework, individual channelopathies display their major deficiencies either in fast electrical signaling or ion transport itself.  相似文献

This highlight article summarizes the current published literature of ion channels and ion transport in type I cells. Twenty years ago, the general theory of ion and fluid transport in the lung was that the alveolar type II cells, known to contain ion channels, governed ion transport and that the type I cells, believed to be incapable of ion transport, only allowed passive movement of water. Unable to reconcile the extraordinarily large surface area covered by type I cells (95% of the internal surface area of the lung) with such minimal biological activity, investigators set out to demonstrate that type I cells were capable of ion transport and played a role in regulating lung fluid balance. Various methods were employed to show that type I cells contained ENaC (HSC and NSC channels), CNG and K(+) channels, and CFTR, further necessitating a revision of the current theories of ion and fluid transport in the lung.  相似文献

Molecular dynamics simulations were performed to study thermal stabilization of proteins via electrostatic interactions of ion pairs. Dynamic motions of four ion pairs previously proposed to be important in thermal stability of adenylate kinase from the thermophile Bacillus stearothermophilus were monitored during the simulation. One of the four ion pairs identified in the crystal structure, Lys180-Asp114, was not maintained in close contact suggesting that the ion pair does not contribute to thermal stability. Among the other three ion pairs, the ion pair Arg116-Glu198 was proposed to be the most important for stability. To predict behaviors of the ion pairs when engineered into a mesophilic homologue to increase stability, in silico mutants of adenylate kinase from the mesophile Bacillus subtilis were generated, and their molecular dynamics simulations were carried out. The ion pairs in the mutant simulations displayed similar behaviors to those in the simulation of the thermophilic protein. To validate the results of the simulations experimentally, the same mutants were produced in vitro and their thermal stabilities were measured using differential scanning calorimetry. In agreement with the simulations, the Lys180-Asp114 did not result in any increase in stability by itself or additive effect with other ion pairs, whereas a mutant with the Arg116-Glu198 exhibited the highest stability among the mutants having one of the four ion pairs. These results provide specific knowledge about stability in adenylate kinases and more generally suggest that molecular dynamics simulations can provide valuable information for identifying and engineering ion pairs.  相似文献

Vitellogenin and vitellin of Manduca sexta and some other insect species were purified by immobilized metal ion affinity chromatography. Ferric ion was chosen as the immobilized metal ion. Agarose-bound carboxymethylpicolylamine was used as the chelating adsorbent for the ferric ion. Vitellogenin and vitellin, both phosphorylated lipoproteins, were shown to bind specifically to the iron. The general applicability of immobilized ferric ion affinity chromatography for the purification of insect vitellogenin and vitellin is suggested.  相似文献

Constant electric fields and constant ion currents are often considered in theories of ion transport. Therefore, it is important to understand the validity of these helpful concepts. The constant field assumption requires that the charge density of permeant ions and flexible polar groups is virtually voltage independent. We present analytic relations that indicate the conditions under which the constant field approximation applies. Barrier models are frequently fitted to experimental current-voltage curves to describe ion transport. These models are based on three fundamental characteristics: a constant electric field, negligible concerted motions of ions inside the channel (an ion can enter only an empty site), and concentration-independent energy profiles. An analysis of those fundamental assumptions of barrier models shows that those approximations require large barriers because the electrostatic interaction is strong and has a long range. In the constant currents assumption, the current of each permeating ion species is considered to be constant throughout the channel; thus ion pairing is explicitly ignored. In inhomogeneous steady-state systems, the association rate constant determines the strength of ion pairing. Among permeable ions, however, the ion association rate constants are not small, according to modern diffusion-limited reaction rate theories. A mathematical formulation of a constant currents condition indicates that ion pairing very likely has an effect but does not dominate ion transport.  相似文献

An effect of ion exchange capacity of petri dish on cell attachment and growth of an anchorage-dependent animal cell was studied. We proposed a new idea to analyze cell attachment kinetics. As the experimental result of cell attachment to a petri dish, the attachment rate constant and the maximum number of attached cells both increase with increasing the ion exchange capacity. However, no direct correlation between the ion exchange capacity and the cell growth was observed. An influence of ion exchange capacity on the cell growth of initially attached cells onto the petri dish with different ion exchange capacities was examined. The specific growth rate of initially attached cells decreased with increasing the ion exchange capacity. The detrimental effect of the ion exchange capacity on the specific growth rate of the initially attached cells exists during the cell cultivation period. In the ion exchange capacity of 27 meq/m2, the specific growth rate drastically decreased. It is considered that there is optimum value of the ion exchange capacity for the cell growth and attachment. It is concluded that the optimum ion exchange capacity is 18 meq/m2.  相似文献

It is shown that the NH4 ion acts in cases of antagonism on the egg of Fundulus more like the K ion than the Na ion; this corresponds to the fact that in its general chemical behavior the NH4 ion resembles the K ion more closely than the Na ion. 2. It is shown that the tolerance of sea urchin eggs towards the Li ion can be increased 500 per cent or more if at the same time a certain amount of Na ion is replaced by K, Rb, or Cs ions. Since in the periodic table Na occupies a position between K and Li it is inferred that the Li and K ions deviate in their physiological action in the opposite direction from the Na ion. 3. These data indicate that the behavior of the K ion in antagonistic salt action (which forms the basis of the physiologically balanced action of ions) is due to its purely chemical character, i.e. its position in the periodic table or rather to its atomic number, and not to those explosions in its nucleus which give rise to a trace of radioactivity.  相似文献

The quasi-steady ion distribution in a plasma with a single ion species and with low-intensity ion acoustic turbulence is found. Conditions are determined under which the stimulated scattering of ion acoustic waves by ions leads to the formation of a superthermal ion distribution function that decreases with increasing velocity more gradually than does a Maxwellian distribution function. It is found that the plasma conductivity increases as a result of a decrease in the turbulence level due to an enhancement of the Cherenkov damping of ion acoustic waves by resonant ions, whose number increases because of the formation of a gradually decreasing distribution of superthermal ions.  相似文献

Mass spectra of a series of chloro- and nitrophenylglucuronides by liquid secondary ion (LSI) mass spectrometry were obtained. In the positive ion mode class characteristic fragmentations and adduct ions are observed only in the presence of alkali salt additives. No additives were necessary in the negative ion mode to see abundant class characteristic [M-H]- and aglycone fragment ions. Cluster ion formation was found to be prominent but only in the negative ion mode.  相似文献
