In the biosphere, bacteria can function as geo-chemical agents, promoting the dispersion, fractionation and/or concentration of materials. Microbial mineral precipitation is resulted from metabolic activities of microorganisms. Based on this biomineralogy concept, an attempt has been made to develop bioconcrete material incorporating of an enrichment culture of thermophilic and anaerobic bacteria within cement-sand mortar/concrete. The results showed a significant increase in compressive strength of both cement-sand mortar and concrete due to the development of filler material within the pores of cement sand matrix. Maximum strength was observed at concentration 10(5)cell/ml of water used in mortar/concrete. Addition of Escherichia coil or media composition on mortar showed no such improvement in strength.  相似文献

Antifungal cement mortar or microbiological calcium carbonate precipitation on cement surface has been investigated as functional concrete research. However, these research concepts have never been fused with each other. In this study, we introduced the antifungal calciteforming bacteria (CFB) Bacillus aryabhattai KNUC205, isolated from an urban tunnel (Daegu, South Korea). The major fungal deteriogens in urban tunnel, Cladosporium sphaerospermum KNUC253, was used as a sensitive fungal strain. B. aryabhattai KNUC205 showed CaCO3 precipitation on B4 medium. Cracked cement mortar pastes were made and neutralized by modified methods. Subsequently, the mixture of B. aryabhattai KNUC205, conidiospore of C. sphaerospermum KNUC253, and B4 agar was applied to cement cracks and incubated at 18 degrees C for 16 days. B. aryabhattai KNUC205 showed fungal growth inhibition against C. sphaerospermum. Furthermore, B. aryabhattai KNUC205 showed crack remediation ability and water permeability reduction of cement mortar pastes. Taken together, these results suggest that the CaCO3 precipitation and antifungal properties of B. aryabhattai KNUC205 could be used as an effective sealing or coating material that can also prevent deteriorative fungal growth. This study is the first application and evaluation research that incorporates calcite formation with antifungal capabilities of microorganisms for an environment-friendly and more effective protection of cement materials. Sporosarcina pasteurii, a common soil bacterium has been tested for microbial treatment of cement mortar. The present study also seeks to investigate the effects of growth medium, bacterial concentration and different buffers concerning the preparation of bacterial suspensions on the compressive strength of cement mortar. Two growth media, six different suspensions and two bacterial concentrations were used in the study. The influence of growth medium on calcification efficiency of S. pasteurii was insignificant. Significant improvement in the compressive as well as the tensile strength of cement mortar was observed. Microbial mineral precipitation visualized by Scanning Electron Microscopy (SEM) shows fibrous material that increased the strength of cement mortar. Formation of thin strands of fillers observed through SEM micrographs improves the pore structure, impermeability and thus the compressive as well as the tensile strengths of the cement mortar. The type of substrate and its molarity have a significant influence on the strength of cement mortar.  相似文献

Fly ash acts as a partial replacement material for both Portland cement and fine aggregate. An innovative approach of microbial calcite precipitation in fly ash-amended concrete has been investigated. This is the first report to discuss the role of microbial calcite precipitation in enhancing the durability of fly ash-amended concrete. The present study investigated the effects of Bacillus megaterium ATCC 14581 on compressive strength, water absorption and water impermeability of fly ash-amended mortar and concrete. Mortar specimens were used for compressive strength and water absorption tests, while concrete specimens were used for water impermeability tests. At the fly ash concentrations of 10%, 20% and 40% in mortars, bacterial cell enhanced mortar compressive strength by 19%, 14% and 10%, respectively, compared to control specimens. Treated mortar cubes absorbed more than three times less water than control cubes as a result of microbial calcite deposition. Microbial deposition of a layer of calcite on the surface of the concrete specimens resulted in substantial decrease of water uptake and permeability compared to control specimens without bacteria. Microbial cells also prevented ingress of water effectively in different concentrations of fly ash-amended concrete. Scanning Electron Micrography (SEM) analyses evidenced the direct involvement of bacteria in calcite precipitation. The approach of the present study gives us dual environment friendly advantages. First, use of fly ash-a recovered resource reduces depletion of natural resources and also reduces the energy-intensive manufacturing of other concrete ingredients, leading to savings in both energy usage and emissions of greenhouse gases. And second, use of bacterial cells to improve strength and durability of fly ash-amended concrete further provides greener and economic options.  相似文献

The production of extracellular polysaccharide, EPS WN9, fromPaenibacillus sp. and its suitability as a viscosity modifying admixture for cement mortar mixing were investigated. After 48 h culture in an optimized medium, cell growth and EPS production were 1,2 g/L and 4.0 g/L, respectively. By adding EPS WN9 to mortar, it was possible to prepare a homogeneous mortar without material segregation and excess air entrapment. The optimal amount of EPS addition to mortar was found to be 0.02 to 0.05%(w/w) of the cement used. Increasing the dosage of EPS WN9 from 0 to 0.05%(w/w) resulted in a setting retardation of 0.14 h to 0.8 h and an increase in the compressive strength of mortar of 10 to 20%.  相似文献

Biofilms of selected bacteria strains were previously used on metal coupons as a protective layer against microbiologically influenced corrosion of metals. Unlike metal surfaces, concrete surfaces present a hostile environment for growing a protective biofilm. The main objective of this research was to investigate whether a beneficial biofilm can be successfully grown on mortar surfaces. Escherichia coli DH5α biofilm was grown on mortar surfaces for 8 days, and the structure and characteristics of the biofilm were studied using advanced microscopy techniques such as scanning electron microscopy and confocal laser scanning microscopy in combination with fluorescence in situ hybridization, live/dead, extracellular polymer staining, ATP analysis, and membrane filtration. A biofilm layer with a varying thickness of 20–40 μm was observed on the mortar surface. The distribution of live and dead bacteria and extracellular polymers varied with depth. The density of the live population near the mortar surface was the lowest. The bacteria reached their highest density at three fourths of the biofilm depth and then decreased again near the biofilm–liquid interface. Overall, the results indicated a healthy biofilm growth in the chosen growth period of 8 days, and it is expected that longer growth periods would lead to formation of a more resistant biofilm with more coverage of mortar surfaces.  相似文献

Various types of microorganisms have been found to inhabit the inner surfaces of asbestos cement (AC) pipe and their activities can cause significant structural damage. They cause a patina to form on the inside surface of AC pipes as a distinctively continuous coating, commonly 2-5 mm in thickness and generally pigmented as yellow, orange, brown or black depending on the metallic cations that have been incorporated into the surface of biofilm (bioaccumulation). Four sublayers can be identified in the patina, from the outer sublayer that directly interacts with the conveyed drinking water to the inner sublayer that is in proximity of the intact cement matrix. The microbes in the outer sublayer are composed mainly of inactive biomass that separates the aerobic environment of the flowing water from the anaerobic conditions inside the patina. The bacteriological community structure shifts from mixed heterotrophic bacteria (HAB), iron-related bacteria (IRB) and slime-forming bacteria (SLYM) in the outer layer, to a more diverse community with IRB, acid-producing bacteria (APB) and SLYM and HAB in the middle sublayer, and further to the SLYM dominated in the inner sublayer. By directly interacting with cementitious materials, including generating organic acids, IRB and APB play important roles in the leaching of free lime and the dissolution of calcium (Ca)-bearing hydrated components of AC pipes, creating porous structure and reducing the pipe strength. Scanning electron microscopy with an energy dispersive X-ray has revealed that bacterial activity on the internal AC pipe wall had resulted in a significant loss of hydrated cement matrix, which can cause pipe failure when stresses imposed on the pipe exceed the remaining pipe strength.  相似文献

Biofilm formation is an increasing problem in medicine, due to the intrinsic resistance of microorganisms in the biofilm mode of growth against the host immune system and antimicrobial therapy. Adhesion is an important step in biofilm formation, influenced, among other factors, by the surface hydrophobicities and charges of both the substratum and the adhering microorganisms. Enterococcus faecalis strains generally display subpopulations with different surface charges, expressed as bimodal zeta potential distributions. Two-thirds of E. faecalis strains isolated from clogged biliary stents displayed such heterogeneity of surface charges in culture. In this study, the influence of this culture heterogeneity on initial adhesion and subsequent biofilm formation was investigated. Heterogeneous strains were retained in higher numbers on polystyrene than homogeneous strains. Also, biofilm formation was much more pronounced for heterogeneous strains than for homogeneous strains. In a population enriched to display only one subpopulation, fewer bacteria were retained than in its original heterogeneous culture. Also, the enriched subpopulation formed less biofilm than its original heterogeneous culture. The presence of ox bile during adhesion resulted in fewer retained bacteria, although heterogeneous strains were still retained in significantly higher numbers than were homogeneous strains, and, in general, the presence of ox bile reduced biofilm formation. The initial adhesion and biofilm formation were independent of the presence of the gene encoding the enterococcal surface protein (esp) or the expression of gelatinase (GelE). It is concluded that heterogeneity in cell surface charge represents an advantage for bacteria in the colonization of surfaces.  相似文献

Single-species microbial biofilm screening for industrial applications   总被引:2,自引:0,他引:2  
While natural microbial biofilms often consist of multiple species, single-species biofilms are of great interest to biotechnology. The current study evaluates biofilm formation for common industrial and laboratory microorganisms. A total of 68 species of biosafety level one bacteria and yeasts from over 40 different genera and five phyla were screened by growing them in microtiter plates and estimating attached biomass by crystal violet staining. Most organisms showed biofilm formation on surfaces of polystyrene within 24 h. By changing a few simple conditions such as substratum characteristics, inoculum and nutrient availability, 66 strains (97%) demonstrated biofilm formation under at least one of the experimental conditions and over half of these strains were classified as strong biofilm formers, potentially suitable as catalysts in biofilm applications. Many non-motile bacteria were also strong biofilm formers. Biofilm morphologies were visualized for selected strains. A model organism, Zymomonas mobilis, easily established itself as a biofilm on various reactor packing materials, including stainless steel.  相似文献

Biofilms are sessile microbial aggregates on the interfaces, and they were usually considered as microbial contamination sources in medical care and various industries. We studied the control and application of biofilms formed by food-related microorganisms, and mechanism of the biofilm formation was also investigated. We studied the biofilm formation in mixed cultures using various combinations of two strains of food-related microorganisms. There were various microorganisms that showed decreased or increased biofilm formation in the mixed culture in comparison with that in a single culture. Biofilm formed by lactic acid bacteria and yeast isolated from traditional fermented food, Fukuyama pot vinegar, exhibited unique feature in that structure and formation mechanism, and expected to be used as an immobilized microorganism in fermentation production. Here our studies on the control and application of biofilms and the mechanisms of its formation were described.  相似文献

目的:微生物被膜是一种具有协调性、功能性和高度结构性的膜状复合物,它可以为微生物提供良好的生存环境,免受外界因素的干扰。研究发现,具有产生微生物被膜能力的细菌治病性明显增强,而现今缺乏一种分析微生物被膜的有效手段。本文探讨利用COMSTAT软件对微生物被膜进行定量分析的方法和意义,为对微生物被膜定性定量分析提供支持手段,从而为研究微生物被膜致病性提供方法理论基础。方法:以葡萄球菌为研究模型,利用激光扫描共聚焦显微镜成像技术,结合COMSTAT微生物被膜分析软件对微生物被膜的单位面积生物量、基质覆盖率、平均厚度、粗糙系数等方面进行定量分析,研究了该葡萄球菌的生物被膜生长变化过程,并考察了抗生素对其生物被膜的抑制作用。结果:在葡萄球菌生物被膜生长过程中,生物量、平均厚度以及平均扩散距离等结构指标数值都有明显增加,而粗糙度和表面积与生物量比值呈现降低趋势,表明了微生物被膜由发生向成熟的转化过程。与此同时,经10μg/mL和100μg/mL的卡那霉素处理得到的葡萄球菌微生物被膜生长受到明显抑制,且随着卡那霉素的浓度增加,抑制效果随之增加。结论:本文运用COMSTAT软件的分析方法首次从生物量、平均厚度等结构指标数值的角度描述了葡萄球菌生物被膜,从而有效评价微生物被膜发生、发展、成熟以及崩解的生长过程。该技术在研究微生物被膜形成的理论机制方面存在潜在价值,可以为研究微生物被膜治病性提供理论基础,具有理论指导意义。  相似文献

Microbiologically induced calcite precipitation by the bacterium Sporosarcina pasteurii (NCIM 2477) using the industrial effluent of the dairy industry, lactose mother liquor (LML) as growth medium was demonstrated for the first time in this study. The urease activity and the calcite precipitation by the bacterium was tested in LML and compared with the standard media like nutrient media and yeast extract media. Calcite constituted 24.0% of the total weight of the sand samples plugged by S. pasteurii and urease production was found to be 353 U/ml in LML medium. The compressive strength of cement mortar was increased by S. pasteurii in all the media used compared to control. No significant difference in the growth, urease production and compressive strength of mortar among the media suggesting LML as an alternative source for standard media. This study demonstrates that microbial calcite acts as a sealing agent for filling the gaps or cracks and fissures in constructed facilities and natural formations alike.  相似文献

The shell of the bivalve Montacuta ferruginosa is covered with a rust-colored biofilm. This biofilm includes filamentous bacteria and protozoa encrusted with a mineral, rich in ferric ion and phosphate. The aim of this research was to study two possible microbial iron precipitation pathways in the biofilm, namely, microbial iron oxidation and microbial degradation of organic Fe(III) complexes. The iron-oxidizing activity was assayed spectrophotometrically by monitoring the formation of the dye Wurster blue in biofilm extracts. Iron-oxidizing activity was effectively detected in extracts obtained by oxalic acid treatment of biofilm fragments. Extracts obtained without oxalic acid treatment, heated extracts, or extracts supplemented with HgCl 2 did not show any activity. This suggests that an iron-oxidizing factor (IOF), possibly an enzyme, coprecipitated with the mineral. Additional information gathered by using sodium dodecyl sulfate-polyacrylamide gel electrophoresis, gel-filtration chromatography, and UV spectrophotometry indicate that the IOF would be a small peptide or glycopeptide (1,350 Da). Microbial degradation of organic Fe(III) complexes was assayed with biofilm fragments incubated in a medium containing ferric citrate. Analysis of the supernatants after various intervals revealed that the complex was degraded by living microorganisms much faster than in the heat-killed negative controls. We conclude that ferric iron precipitation in the biofilm may proceed by way of microbial Fe(II) oxidation as well as microbial degradation of organic Fe(III) complexes.  相似文献

As a high priority of waste management and recycling by the Hong Kong government, Recycled Aggregate (RA) has been used in various construction applications, mainly as sub-grade, roadwork, and unbound materials. However, higher-grade applications are rare. The major barrier encountered is the variation of quality within RA, which causes lower strength, resulted from crystallization of Recycled Aggregate Concrete (RAC). Therefore, the objective of this study is to examine the crystallization of RAC in a Two-Stage Mixing Approach. Following are the five areas of interest: (i) investigate the waste problems in construction activities; (ii) examine the crystal development on the hydration of cement paste; (iii) develop a two-stage mixing approach (TSMA) for improving the performance of RAC; (iv) explore the crystallization of TSMA in comparison with the Normal Mixing Approach (NMA) through use of Differential Scanning Calorimetry (DSC); and (v) verify the results obtained from DSC analysis with those obtained from compressive strength testing. This study adopted 0, 20, and 100% RA substitution in virgin aggregate and measured by DSC and compressive strength on both TSMA and NMA. TSMA uses only half the water for mixing, forming a thin layer of cement slurry on the surface of RA that will permeate into the porous old cement mortar and fill old cracks and voids in the pre-mix process. The results from DSC analysis clearly demonstrated that TSMA can give a better crystallization of CaO·SiO2·H2O [CSH] and Ca(OH)2[CH]. The optimal situation occurs on 20% RA substitution in virgin aggregate, balancing the advantages of each, a finding supported by the results from compressive strength testing. Therefore, TSMA is a superior methodology and opens a wider application for the use of RAC.  相似文献

Background

Vital pulp preservation in the treatment of deep caries is challenging due to bacterial infection. The objectives of this study were to synthesize a novel, light-cured composite material containing bioactive calcium-silicate (Portland cement, PC) and the antimicrobial quaternary ammonium salt monomer 2-methacryloxylethyl dodecyl methyl ammonium bromide (MAE-DB) and to evaluate its effects on Streptococcus mutans growth in vitro.

Methods

The experimental material was prepared from a 2∶1 ratio of PC mixed with a resin of 2-hydroxyethylmethacrylate, bisphenol glycerolate dimethacrylate, and triethylene glycol dimethacrylate (4∶3∶1) containing 5 wt% MAE-DB. Cured resin containing 5% MAE-DB without PC served as the positive control material, and resin without MAE-DB or PC served as the negative control material. Mineral trioxide aggregate (MTA) and calcium hydroxide (Dycal) served as commercial controls. S. mutans biofilm formation on material surfaces and growth in the culture medium were tested according to colony-forming units (CFUs) and metabolic activity after 24 h incubation over freshly prepared samples or samples aged in water for 6 months. Biofilm formation was also assessed by Live/Dead staining and scanning electron microscopy.

Results

S. mutans biofilm formation on the experimental material was significantly inhibited, with CFU counts, metabolic activity, viability staining, and morphology similar to those of biofilms on the positive control material. None of the materials affected bacterial growth in solution. Contact-inhibition of biofilm formation was retained by the aged experimental material. Significant biofilm formation was observed on MTA and Dycal.

Conclusion

The synthesized material containing HEMA-BisGMA-TEGDMA resin with MAE-DB as the antimicrobial agent and PC to support mineralized tissue formation inhibited S. mutans biofilm formation even after aging in water for 6 months, but had no inhibitory effect on bacteria in solution. Therefore, this material shows promise as a pulp capping material for vital pulp preservation in the treatment of deep caries.  相似文献

We have studied the differences in the organic matter processing and biofilm composition and structure between autoheterotrophic and heterotrophic biofilm communities. Microbial communities grown on artificial biofilms were monitored, following incubation under light and dark conditions and with or without the addition of glucose as a labile organic compound. Glucose addition greatly affected the microbial biofilm composition as shown by differences in 16S rRNA gene fingerprints. A significant increase in β-glucosidase and peptidase enzyme activities were also observed in glucose-amended biofilms incubated in the dark, suggesting an active bacterial community. Light enhanced the algal and bacterial growth, as well as higher extracellular enzyme activity, thereby indicating a tight algal–bacterial coupling in biofilms incubated under illumination. In these biofilms, organic compounds excreted by photosynthetic microorganisms were readily available for bacterial heterotrophs. This algal–bacterial relationship weakened in glucose-amended biofilms grown in the light, probably because heterotrophic bacteria preferentially use external labile compounds. These results suggest that the availability of labile organic matter in the flowing water and the presence of light may alter the biofilm composition and function, therefore affecting the processing capacity of organic matter in the stream ecosystem.  相似文献

Detecting changes in the formation dynamics of biofilms stemming from bacteria and unicellular microorganisms in their natural environment is of prime interest for biological, ecological as well as anti-fouling technology research. We developed a robust optical fiber-based biofilm sensor ready to be applied in natural aquatic environments for on-line, in situ and non-destructive monitoring of large-area biofilms. The device is based on the detection of the natural fluorescence of microorganisms constituting the biofilm. Basically, the intrinsic fluorescence of the amino acid tryptophan is excited at a wavelength of λ=280 nm and detected at λ=350 nm utilising a numerically optimized sensor head equipped with a UV-LED light source and optical fiber bundles for efficient fluorescence light collection. Calibration was carried out with tryptophan solutions and two characteristic marine bacteria strains revealing linear signal response, satisfactory background suppression, wide dynamic range, and an experimental detection limit of 4 × 10(3)cells/cm(2). Successful field experiments in the Baltic Sea accomplished over a period of twenty-one days provided for the first time continuous observation of biofilm formation dynamics in a natural habitat. Starting from the first adhering bacteria, the measurement yielded the characteristic three phases of biofilm formation up to a fully developed biofilm. The sensor system holds potential for applications in aquatic sciences including deep sea research and, after further miniaturisation, in the industrial and biomedical field.  相似文献

This work deals with the bacterial contamination of yeast, both as biofilm and in the planktonic phase. A model continuous system using self-fluorescent microorganisms was proposed to perform in vivo and in situ studies of a mixed biofilm. The yeast strain was inoculated first while the bacteria were added few days later to mimic a contamination. Supports sampled during the experiment were observed by scanning confocal laser microscopy. The behaviour of the microorganisms in real process conditions was then analysed without any treatment that could modify their physiology and/or damage the community structure. Using image analysis, the characteristics of biofilm development (microorganism ratio, 3D-organisation, growth rates) were studied and compared to the behaviour of the suspended cells in the bioreactor. Based on the biovolumes (volume occupied by each microorganism), the growth rates in biofilm for the bacteria and the yeasts were determined at 0.10 and 0.03 h(-1) respectively, while the imposed dilution rate was 0.10 h(-1). Even though the ability of yeast to develop biofilm was demonstrated, its capacity remained very low compared to that of the bacteria which quickly invaded and covered the whole yeast biofilm. This approach makes an original and powerful tool to study the competition phenomena occurring in model biofilms.  相似文献

The study characterized the sessile microbial communities on mortar and stone in Milan University's Richini's Courtyard and investigated the relationship between airborne and surface-associated microbial communities. Active colonization was found in three locations: green and black patinas were present on mortar and black spots on stone. Confocal laser scanning microscopy, scanning electron microscopy and culture-independent molecular methods revealed that the biofilm causing deterioration was dominated by green algae and black fungi. The mortar used for restoration contained acrylic and siloxane resins that could be used by microorganisms as carbon and energy sources thereby causing proliferation of the biofilm. Epifluorescence microscopy and culture-based methods highlighted a variety of airborne microflora. Bacterial and fungal counts were quantitatively similar to those reported in other investigations of urban areas, the exception being fungi during summer (1–2 orders of magnitude higher). For the first time in the cultural heritage field, culture-independent molecular methods were used to resolve the structure of airborne communities near discoloured surfaces, and to investigate the relationship between such communities and surface-associated biofilms.  相似文献
