The cephalic portion of the foregut of Neopachygaster is clearly subdivided into the pharynx and oesophagus. The precerebral pharynx is transformed into a well-developed anterior pharyngeal pump with two unpaired dilators and an intrinsic layer of the muscle fibers. There are two pairs of the posterior pharyngeal dilators, attached to the pharynx just behind the brain. According to these features, the pharynx of Neopachygaster has the most plesiomorphic structure among all the members of Stratiomyidae studied so far.  相似文献

Zebrafish (Danio rerio) have become a particularly effective tool for modeling human diseases affecting skeletal muscle, including muscular dystrophies1-3, congenital myopathies4,5, and disruptions in sarcomeric assembly6,7, due to high genomic and structural conservation with mammals8. Muscular disorganization and locomotive impairment can be quickly assessed in the zebrafish over the first few days post-fertilization. Two assays to help characterize skeletal muscle defects in zebrafish are birefringence (structural) and touch-evoked escape response (behavioral).Birefringence is a physical property in which light is rotated as it passes through ordered matter, such as the pseudo-crystalline array of muscle sarcomeres9. It is a simple, noninvasive approach to assess muscle integrity in translucent zebrafish larvae early in development. Wild-type zebrafish with highly organized skeletal muscle appear very bright amidst a dark background when visualized between two polarized light filters, whereas muscle mutants have birefringence patterns specific to the primary muscular disorder they model. Zebrafish modeling muscular dystrophies, diseases characterized by myofiber degeneration followed by repeated rounds of regeneration, exhibit degenerative dark patches in skeletal muscle under polarized light. Nondystrophic myopathies are not associated with necrosis or regenerative changes, but result in disorganized myofibers and skeletal muscle weakness. Myopathic zebrafish typically show an overall reduction in birefringence, reflecting the disorganization of sarcomeres.The touch-evoked escape assay involves observing an embryo''s swimming behavior in response to tactile stimulation10-12. In comparison to wild-type larvae, mutant larvae frequently display a weak escape contraction, followed by slow swimming or other type of impaired motion that fails to propel the larvae more than a short distance12. The advantage of these assays is that disease progression in the same fish type can be monitored in vivo for several days, and that large numbers of fish can be analyzed in a short time relative to higher vertebrates.  相似文献

The gross morphology and histology of the alimentary tracts of three species of glassy perchlet; Ambassis productus, A. natalensis, and A. gymnocephalus from estuaries on the southeast coast of Africa were investigated. The anatomy of the digestive tracts in all three species was found to be similar. Well-developed dentition and pharyngeal teeth together with a distensible stomach and a low relative gut length (RGL) suggest a predatory and carnivorous habit for all three species. The relative gut lengths of Ambassis species from different estuarine systems are compared‥ Differences in RGL for A. productus and A. natalensis from the Kosi and St Lucia systems with fish from Mdloti estuary are discussed. It is suggested that decreased RGL for fish at Mdloti is attributable to decreased food availability and not to a lack in the calorific content of their diet. Histological investigation revealed the presence of the following regions: a pharynx; an oesophagus; a stomach differentiated into cardiac and pyloric regions; a duodenum or upper intestine; an ileum or lower intestine; and a rectum. Pyloric and rectal sphincters are present. The tunics of the above regions are described. The epithelium of the oesophagus contains taste buds and numerous mucus cells, and varies from stratified anteriorly to simple columnar posteriorly. The muscularis comprises dorsally and ventrally located inner muscle bundles and an outer circular layer. Both layers consist of striated fibres. Gastric glands are present in the mucosa of the cardiac stomach but are absent in the pylorus. Columnar absorbing cells and goblet cells are present in the epithelium of the upper and lower intestine. The rectum is distinguished from the intestine by the proliferation of mucous-secreting cells which are thought to aid defecation.  相似文献

The cuticular oesophagus is a simple expansion of the dorsal pharyngeal wall of the mastax. The ciliary oesophagus is the cellular anterior wall of the stomach lumen, but seems to have the same embryological origin as the pharynx.In Brachionus calyciflorus, its cilia are surrounded by cuticular velums which have the same myelin-like structure and the same function as the buccal velum of Philodina roseola. In all cases, the oesophagus prevents the return of food particles from the stomach to the mastax lumen.  相似文献

1. Benthic chironomid larvae and the amphipod Gammarus lacustris have been observed in the pelagic habitats of many mountain lakes. The main goal of this study was to determine if chironomid larvae and gammarids potentially affect predator–prey and nutrient dynamics in pelagic food webs of mountain lakes. 2. Eighty‐six mountain lakes were surveyed in Alberta and eastern British Columbia during the years 1965–1984, 1991–2004 and 2005–2007. Pelagic chironomid larvae were found in 86% of these lakes, and pelagic gammarids were found in 29% of lakes. Densities of pelagic chironomid larvae were 92% lower in lakes with pelagic gammarids and 76% lower in lakes with trout (P < 0.05). Intraguild predation of trout on gammarids appeared to reduce predation pressure on chironomid larvae. Gammarids consumed in vitro about 1 chironomid per gammarid per day or about 20% of their body mass in chironomid biomass per day. 3. Concentrations of total dissolved P and N, particulate C, and chlorophyll‐a increased with increasing densities of pelagic gammarids and chironomid larvae in situ (R2 = 0.14 ± 0.19 SD, P < 0.1) and in vitro (P < 0.001). 4. Our findings suggest that gammarids and chironomid larvae are linked as predators and prey in pelagic food webs, possibly stimulating phytoplankton abundance via nutrient release.  相似文献

The alimentary canals of Trochonerilla mobilis and Nerillidium troglochaetoides consist of a ventral pharyngeal organ, oesophagus, stomach, intestine, and rectum. Prominent salivary glands lying lateral to the oesophagus discharge their secretions into the buccal cavity. Ciliated canals, the enteronephridia, embedded in the intestinal epithelium, open into the stomach near its border to the intestine. The ventral pharynx comprises a muscle bulb connected to a tonguelike organ by an investing muscle. The whole alimentary canal is ciliated except for the intestine of T. mobilis. The stomach is built up of absorptive cells and posteriorly also of secretory cells, whereas the intestine consists of only one cell type which is considered to be mainly absorptive. A typical microvillar brush border is present only in the intestine of T. mobilis; elsewhere the density of microvilli is low or the cells have irregular apical processes. In N. troglochaetoides the intestine has a ventral ciliary gutter laterally bordered by cells with highly specialized microvilli. The enteronephridia — 3 in N. troglochaetoides and 13 in T. mobilis — are unicellular tubes up to 130 μm long with a microvillar brush border and other cytological features typical for nephridial ducts. To characterize the process of vertebral segmentation and disc formation in living animals, we analyzed tiggy-winkle hedgehog (twhh):green fluorescent protein (gfp) and sonic hedgehog (shh):gfp transgenic zebrafish models that display notochord-specific GFP expression. We found that they showed distinct patterns of expression in the intervertebral discs of late stage fish larvae and adult zebrafish. A segmented pattern of GFP expression was detected in the intervertebral disc of twhh:gfp transgenic fish. In contrast, little GFP expression was found in the intervertebral disc of shh:gfp transgenic fish. Treating twhh:gfp transgenic zebrafish larvae with exogenous retinoic acid (RA), a teratogenic factor on normal development, resulted in disruption of notochord segmentation and formation of oversized vertebrae. Histological analysis revealed that the oversized vertebrae are likely due to vertebral fusion. These studies demonstrate that the twhh:gfp transgenic zebrafish is a useful model for studying vertebral segmentation and disc formation, and moreover, that RA signaling may play a role in this process.  相似文献

AimsExplore the effect of compound lobelia oral liquid on acute pharyngitis rabbits model caused by ammonium hydroxide, evaluate its curative effect.MethodsAcute pharyngitis rabbit model was prepared by ammonia spray at pharyngeal. After 1 h of the last administration, score the pharyngeal tissue symptoms of rabbits under the condition of naked eyes (score according to the score criteria). Then, take blood from the marginal auricular vein for routine blood tests and serum TNF-α level measure. Take pharyngeal tissue for HE staining, and observe its pathological changes under the microscope.ResultsCompared with the blank group, the pharyngeal tissue symptom score and serum TNF-α level in the model group were significantly increased (P < 0.01). The results of blood routine showed that the number of white blood cells and lymphocytes in the model group increased significantly (P < 0.01), and neutrophilic granulocyte percentage decreased significantly (P < 0.01). The infiltration of inflammatory cells in the pharynx tissue was obvious and pathological changes were observed. Compared with the model group, large, middle and small doses of compound lobelia oral liquid group could reduce the scores of pharyngeal tissue symptoms in rabbits with acute pharyngitis induced by ammonia. The large and middle dose compound lobelia oral liquid group could significantly reduce serum TNF-α level and neutrophilic granulocyte percentage, significantly increase the number of white blood cells and lymphocytes (P < 0.01); The small dose compound lobelia oral liquid group could significantly reduce the number of white blood cells and lymphocytes (P < 0.01), there was an increasing tendency to neutrophils and a decreasing tendency to serum TNF-α, but no statistical significance. The large, middle and small doses of compound lobelia oral liquid group all could improve the pathological changes of pharyngeal tissue inflammatory infiltration.ConclusionCompound lobelia oral liquid has a good effect on rabbit acute pharyngitis model caused by ammonia.  相似文献

It has been widely documented that fish oil attenuates inflammatory responses partially via down-regulation of T-lymphocyte function. To determine the anti-inflammatory role of fish oil in weanling pigs, we investigated the effects of fish oil and its functional constituents on peripheral blood lymphocyte proliferation, cytokine production and subsequent intracellular signalling in inflammatory-challenged weanling pigs and in in vitro cultured lymphocytes. Fish oil (7%) or corn oil (7%) was supplemented to 72 crossbred pigs (7.6 ± 0.3 kg BW and 28 ± 3 days of age) in a 2 × 2 factorial experiment that included an Escherichia coli lipopolysaccharide (LPS) challenge (challenged or not challenged). On day 14 and 28 of the experiment, 200 μg/kg BW of LPS or an equivalent amount of sterile saline was administered to the pigs by intraperitoneal injection. Blood samples were collected on days 15 and 29 to determine peripheral blood lymphocyte proliferation, interleukin-1β (IL-1β) and interleukin-2 (IL-2) production. The results showed that inflammatory challenge decreased average daily gain (P < 0.05) and average daily feed intake (P < 0.05) during days 15 - 28. Fish oil supplementation had no effect on growth performance. Inflammatory challenge increased lymphocyte proliferative response to concanavalin A (Con A) (P < 0.05) following each challenge. Fish oil tended to suppress (P < 0.1) the proliferation following the first challenge. Similarly, fish oil tended to reduce IL-1β production (P < 0.1) following the second challenge and IL-2 (P < 0.1) production following the first challenge in both challenged and unchallenged pigs compared with corn oil. In parallel in vitro experiments, peripheral blood lymphocytes of weanling pigs were incubated with various concentrations of eicosapentaenoic acid (EPA), docosahexaenoic acid (DHA) or linoleic acid (LA) (0, 20, 40, 60, 80, 100 μg/ml). EPA, DHA and high levels of LA predominantly suppressed IL-1β (P < 0.05), IL-2 (P < 0.05) production and subsequent lymphocyte proliferation (P < 0.05). Low levels of LA increased (P < 0.05) IL-2 production. Compared with LA, EPA resulted in a stronger inhibition of lymphocyte proliferation (P < 0.05) and IL-2 (P < 0.01), and DHA resulted in a stronger inhibition of IL-1β (P < 0.05) and IL-2 (P < 0.01). To elucidate the mechanism(s) by which fish oil and its functional constituents suppressed lymphocyte function, the kinetics of intracellular [Ca2 +]i and protein kinase C activity were determined in in vitro experiments. EPA, DHA and LA exerted very similar dose-dependent stimulatory effects on intracellular Ca2 +. EPA and DHA inhibited protein kinase C activity (P < 0.05), while LA had no significant effect (P > 0.05). These results suggest that fish oil and its functional constituents (EPA and DHA) exerted an anti-inflammatory effect by down-regulation of lymphocyte activation in weanling pigs, possibly by manipulation of intracellular signalling.  相似文献

The early proliferative stages of the microsporidian parasite, Pseudoloma neurophilia were visualized in larval zebrafish, Danio rerio, using histological sections with a combination of an in situ hybridization probe specific to the P. neurophilia small‐subunit ribosomal RNA gene, standard hematoxylin‐eosin stain, and the Luna stain to visualize spores. Beginning at 5 d post fertilization, fish were exposed to P. neurophilia and examined at 12, 24, 36, 48, 72, 96, and 120 h post exposure (hpe). At 12 hpe, intact spores in the intestinal lumen and proliferative stages developing in the epithelial cells of the anterior intestine and the pharynx and within hepatocytes were observed. Proliferative stages were visualized in the pancreas and kidney at 36–48 hpe and in the spinal cord, eye, and skeletal muscle beginning at 72 hpe. The first spore stages of P. neurophilia were observed at 96 hpe in the pharyngeal epithelium, liver, spinal cord, and skeletal muscle. The parasite was only observed in the brain of larval fish at 120 hpe. The distribution of the early stages of P. neurophilia and the lack of mature spores until 96 hpe indicates that the parasite gains access to organs distant from the initial site of entry, likely by penetrating the intestinal wall with the polar tube.  相似文献

We aimed to investigate the possible role of creatine (CR) supplementation in counteracting dexamethasone-induced muscle wasting and insulin resistance in rats. Also, we examined whether CR intake would modulate molecular pathways involved in muscle remodeling and insulin signaling. Animals were randomly divided into four groups: (1) dexamethasone (DEX); (2) control pair-fed (CON-PF); (3) dexamethasone plus CR (DEX-CR); and (4) CR pair-fed (CR-PF). Dexamethasone (5 mg/kg/day) and CR (5 g/kg/day) were given via drinking water for 7 days. Plantaris and extensor digitorum longus (EDL) muscles were removed for analysis. Plantaris and EDL muscle mass were significantly reduced in the DEX-CR and DEX groups when compared with the CON-PF and CR-PF groups (P < 0.05). Dexamethasone significantly decreased phospho-Ser473-Akt protein levels compared to the CON-PF group (P < 0.05) and CR supplementation aggravated this response (P < 0.001). Serum glucose was significantly increased in the DEX group when compared with the CON-PF group (DEX 7.8 ± 0.6 vs. CON-PF 5.2 ± 0.5 mmol/l; P < 0.05). CR supplementation significantly exacerbated hyperglycemia in the dexamethasone-treated animals (DEX-CR 15.1 ± 2.4 mmol/l; P < 0.05 vs. others). Dexamethasone reduced GLUT-4 translocation when compared with the CON-PF and CR-PF (P < 0.05) groups and this response was aggravated by CR supplementation (P < 0.05 vs. others). In conclusion, supplementation with CR resulted in increased insulin resistance and did not attenuate muscle wasting in rats treated with dexamethasone. Given the contrast with the results of human studies that have shown benefits of CR supplementation on muscle atrophy and insulin sensitivity, we suggest caution when extrapolating this animal data to human subjects.  相似文献

The ontogeny of larval body density and the morphological and histological events during swimbladder development were investigated in two cohorts of yellowtail kingfish Seriola lalandi larvae to understand the relationship between larval morphology and body density. Larvae <3 days post hatch (dph) were positively buoyant with a mean ± s.d . body density of 1·023 ± 0·001 g cm?3. Histological evidence demonstrated that S. lalandi larvae are initially transient physostomes with the primordial swimbladder derived from the evagination of the gut ventral to the notochord and seen at 2 dph. A pneumatic duct connected the swimbladder to the oesophagus, but degenerated after 5 dph. Initial swimbladder (SB) inflation occurred on 3 dph, and the inflation window was 3–5 dph when the pneumatic duct was still connected to the gut. The swimbladder volume increased with larval age and the epithelial lining on the swimbladder became flattened squamous cells after initial inflation. Seriola lalandi developed into a physoclist with the formation of the rete mirabile and the gas‐secreting gland comprised low‐columnar epithelial cells. Larvae with successfully inflated swimbladders remained positively buoyant, whereas larvae without SB inflation became negatively buoyant and their body density gradually reached 1·030 ± 0·001 g cm?3 by 10 dph. Diel density changes were observed after 5 dph, owing to day time deflation and night‐time inflation of the swimbladder. These results show that SB inflation has a direct effect on body density in larval S. lalandi and environmental factors should be further investigated to enhance the rate of SB inflation to prevent the sinking death syndrome in the early life stage of the fish larvae.  相似文献

The aim of this study was to analyse the development of the digestive tract of Siamese fighting fish larvae (Betta splendens Regan, 1910), from hatching to 92 hours post‐hatching (hph) at 28.0°C, in order to determine the most appropriate time to begin exogenous feeding (live feed) and to investigate the digestive tract and its function during larval development. At hatching (29 hours post‐fertilization), the digestive tract was a simple and straight undifferentiated tube lined by a single layer of columnar epithelial cells; the mouth and anus were closed. At 18 hph, the mouth was open. At 32 hph, the tongue was located in the depression of the buccopharyngeal cavity floor and supported by basal cartilage. Goblet cells were present in the epithelium lining the pharynx and oesophagus. At 56 hph, the midgut had four distinct layers: the mucosa with typical villi, the submucosa, the smooth muscle layer, and the serosa. Histological analysis revealed that the larvae retained endogenous yolk reserves until 74 hph. Lipid accumulation was observed in the liver, which coincided with complete yolk absorption. At this time, the digestive tract was fully open and functional. Thus, it can be concluded that live feed should be given to B. splendens larvae from 74 hph onward, when the larvae are able to consume the food provided. This study also provides useful data for the improvement of husbandry techniques and for the formulation of diets specific to ornamental fish larvae.  相似文献

A study was conducted to compare growth and survival of Hucho taimen larvae from 21 to 76 days after hatch (DAH) fed one of three diets: formulated feed alone (group F); a co‐feeding diet of water fleas, tubifex and formulated feed (group C); or live food of water fleas and tubifex (group L), and to investigate the potential use of dietary L‐alanyl‐L‐glutamine (L‐AG) in larval taimen for a more nutritious starter diet. Triplicate groups of 5000 fish were randomly assigned to each aquarium provided with water from a flow‐through system, and fed to apparent satiation. The results show that larvae can feed efficiently on floating crumbled particles of formulated feed. Weight gain of larvae fed only formulated feed was significantly lower than other groups at 34 DAH (P < 0.05). At the end of the experiment, weight gain reached the highest value in group F and was lowest in group L (P < 0.05). Condition factor reached the highest values in group F and lowest in group C (P < 0.05). Specific growth rate was in accordance with weight gain at 76 DAH. Survival showed no differences among the groups (P > 0.05). In conclusion, H. taimen larvae can be fed formulated feed alone and L‐AG may be used as a feeding attractant during the weaning process, which should lead to a better understanding in the rearing improvement in the feeding of larvae.  相似文献

Myoretronectes paranaensis, gen. et sp. n., from lotic sandy bottoms of the Paraná river, Argentina, is described. The establishment of the new genus is based on the presence of a non-ciliated pharynx, a discrete oesophagus, and a strong muscle system surrounding the distal pharynx. A new emended diagnosis of the family Retronectidae is presented. Myoretronectes paranaensis apparently cohabits with the oligochaete Narapa bonettoi Righi & Varela, 1983 and the copepod Parastenocaris sp.  相似文献

Summary Structural and ultrastructural data are presented for the myoepithelial pharynges of 20 species of Gastrotricha representing the marine Macrodasyida and marine and freshwater Chaetonotida. A comparative analysis reveals that pharynges with several plesiomorphic characters occur in Chordodasys among the Macrodasyida and Neodasys among the Chaetonotida. The Gastrotricha are systematized based on pharyngeal characters and the system is shown to be concordant with all recent classifications of the group. The plesiomorphic design of the Gastrotrich pharynx is given as: a cylindrical pharyngeal pump composed of monociliated myoglanduloepithelial cells surrounding a circular or oval lumen; radial myofibrils organized into several cross-striated sarcomeres with Z-discs composed of planar aggregations of dense bodies; excitation-contraction coupling is achieved by peripheral couplings of SR with the sarcolemma; apical cell surface with microvilli protruding through the two-layered cuticle; pharynx with at least 3 longitudinal tracts of monociliated sensory cells; nerves as at least 4 basal intraepithelial, longitudinal tracts of neurites. As a model for muscle cell evolution, the investigation postulates a monociliated, cross-striated myoglanduloepithelial cell as the original muscle cell design within the Gastrotricha. Triradiate myoepithelial foreguts occur only in Bryozoa (Ectoprocta), Gastrotricha, Nematoda and Tardigrada. The potential homology of pharyngeal organization of the latter three phyla is discussed. Based on pharyngeal structure, it is concluded that Gastrotricha (Chaetonotida-Paucitubulatina) and Nematoda share several apomorphic characters and share, therefore, a most recent common ancestor. Affinities of Tardigrada with Aschelminthes are considered feasible but currently inconclusive for lack of sufficient comparative ultrastructural data for the Tardigrada.  相似文献

The effects of 14 days exposure to sublethal concentrations of endosulfan (10 and 40 μg L?1) were investigated in mRNA‐ P450 1A expression, antioxidant enzymes (SOD and CAT) activity and histopathological alterations of Persian sturgeon (Acipenser persicus) fingerlings with weights of 3–5 g. The results illustrated that the relative mRNA‐ P450 1A expression level significantly increased (P < 0.05) compared to the control group. Highest significant increase (P < 0.05) was observed on the first day, then decreased towards day 14 of exposure. The SOD and CAT activity showed a significant increase in fish exposed to different concentrations up to day 7, then activity decreased on day 14 in fish of all treated groups. Although signs of tissue lesions were observed on day 4, they increased from day 7 and reached the highest level on day 14. The magnitude of all changed studied parameters (gene expression, enzymes and histopathological) follows a concentration‐dependent manner.  相似文献

Fish larvae in aquaculture have high mortality rates due to pathogenic bacteria, especially the Vibrio species, and ineffective prophylactic strategies. Vaccination is not feasible in larvae and antibiotics have reduced efficacy against multidrug resistant bacteria. A novel approach to controlling Vibrio infections in aquaculture is needed. The potential of phage therapy to combat vibriosis in fish larvae production has not yet been examined. We describe the isolation and characterization of two bacteriophages capable of infecting pathogenic Vibrio and their application to prevent bacterial infection in fish larvae. Two groups of zebrafish larvae were infected with V. anguillarum (∼106 CFU mL−1) and one was later treated with a phage lysate (∼108 PFU mL−1). A third group was only added with phages. A fourth group received neither bacteria nor phages (fish control). Larvae mortality, after 72 h, in the infected and treated group was similar to normal levels and significantly lower than that of the infected but not treated group, indicating that phage treatment was effective. Thus, directly supplying phages to the culture water could be an effective and inexpensive approach toward reducing the negative impact of vibriosis in larviculture.  相似文献

The effect of benzo(a)pyrene on regeneration of the pharynx in Bdellocephala brunnea Ijima et Kaburaki was studied by inserting a 5-µg crystal of this carcinogen into the pharyngeal cavity of worms from which the pharynx had been extirpated 2 d previously. Multiple pharynges formed in the regenerates, and neoplasia was induced in a few cases. The malformed pharynges appeared as protrusions overlapping the anterior part of the main pharynx. Electron microscopy of the pharynx in normal, intact worms shows eight layers comprising inner and outer ciliated epithelia, muscle, and glands.  相似文献

Zebrafish (Danio rerio) have become an important model organism for developmental biology and human health studies. We recently demonstrated differential growth patterns between the zebrafish and a close relative the giant danio (Danio aequipinnatus), where the giant danio appears to exhibit indeterminate growth similar to most fish species important for commercial production, while zebrafish exhibit determinate growth more similar to mammalian growth. This study focused on evaluating muscle growth regulation differences in adult zebrafish and giant danio utilizing growth hormone treatment as a mode of growth manipulation. Growth hormone treatment resulted in increased overall growth in giant danio, but failed to increase growth in the zebrafish. Growth hormone treatment increased muscle IGF-I and GHrI gene expression in both species, but to a larger degree in the giant danio. In contrast, zebrafish exhibited a larger increase in IrA and IGF-IrB gene expression in muscle in response to GH treatment. In addition muscle myostatin levels were differentially regulated between the two species, with a down-regulation observed in rapidly growing, GH-treated giant danio and an up-regulation in zebrafish not actively growing in response to GH. This is the first report of differential expression of growth-regulating genes in closely related fish species exhibiting opposing growth paradigms. These results further support the role that the zebrafish and giant danio can play important model organisms for determinate and indeterminate growth.  相似文献
