Environmental DNA (eDNA) promises to ease noninvasive quantification of fish biomass or abundance, but its integration within conservation and fisheries management is currently limited by a lack of understanding of the influence of eDNA collection method and environmental conditions on eDNA concentrations in water samples. Water temperature is known to influence the metabolism of fish and consequently could strongly affect eDNA release rate. As water temperature varies in temperate regions (both seasonally and geographically), the unknown effect of water temperature on eDNA concentrations poses practical limitations on quantifying fish populations using eDNA from water samples. This study aimed to clarify how water temperature and the eDNA capture method alter the relationships between eDNA concentration and fish abundance/biomass. Water samples (1 L) were collected from 30 aquaria including triplicate of 0, 5, 10, 15 and 20 Brook Charr specimens at two different temperatures (7 °C and 14 °C). Water samples were filtered with five different types of filters. The eDNA concentration obtained by quantitative PCR (qPCR) varied significantly with fish abundance and biomass and types of filters (mixed‐design ANOVA,< 0.001). Results also show that fish released more eDNA in warm water than in cold water and that eDNA concentration better reflects fish abundance/biomass at high temperature. From a technical standpoint, higher levels of eDNA were captured with glass fibre (GF) filters than with mixed cellulose ester (MCE) filters and support the importance of adequate filters to quantify fish abundance based on the eDNA method. This study supports the importance of including water temperature in fish abundance/biomass prediction models based on eDNA.  相似文献

The effects of rearing and feeding conditions, as well as of body size and origin on sexual differentiation have been examined in elvers collected on the Atlantik coast (River Ems) and in the Tyrrhenian Sea. Raised at 17°, 20°, 23°, 26° or 29° C and at constant photoperiod, the elvers received either commercial fish food or cod roe. Eels from the Tyrrhenian Sea mostly developed into males. To some extent, at temperatures optimal for growth (26° C) and feeding a cod-roe diet, a shift of sex ratio in favour of females was observed in Atlantic eels. The results obtained contradict those ofFidora (1951), who, rearing elvers in ponds, found the percentage of male eels to be positively correlated with increasing stock density. Although in the present study, stock density was 200 times higher than inFidora's experiments, the percentage of females was much higher than that of males. The results obtained supportBellini's (1907) findings that big elvers preferentially develop into females, but small elvers into males. Irrespective of elvers' origin, water temperature and diet, the onset of sexual differentiation dependends upon attaining a certain body length (15–25 cm), but not on age. Whereas under natural conditions females are supposed to grow faster than males, under the conditions tested the males grew faster than the females. It is concluded, that, in addition to genetic factors, various environmental factors may influence sex determination in eels.  相似文献

The movements and habitat use of resident yellow eels were studied in a stream stretch having both natural and minimum flow zones. N = 12 individuals (total length 505–802 mm) were surgically tagged with radio transmitters and released at their capture sites. They were located using manual radio receivers during the daytime from 2 to 5 days/week over periods ranging from 200 to 329 days, for a total of 1,098 positions. Eels showed home ranges ranging from 33 to 341 m (median value, 62 m), displayed strong fidelity to sites and demonstrated a great degree of plasticity in habitat use. Eels were slightly mobile throughout the year, but their movements were season and temperature dependent, with a maximum during the spring (mean water temperature, 12 °C) and a minimum in winter (3 °C). Stones and roots (utilization rate greater than 50 % of eels for more than 30 % of location days) were significantly the most frequently used habitats. Between the two flow zones, the natural flow was the most occupied, with a significantly higher proportion of resident eels (66.7 % of radio-tagged yellow eels) and longer occupation (81 % of location days) than the minimum flow zone with less suitable habitats.  相似文献

1. The objective was to determine the thermal limits for feeding and survival in the bullhead, Cottus gobio, using juveniles (total length 20–30 mm, live weight 0.5–1.5 g) from one population and adults (50–70 mm, 3.5–5.5 g) from three populations. 2. Fish were acclimated to constant temperatures (3, 7, 10, 15, 20, 25 or 27 °C) and the temperature was then changed at a rate of 1 °C /30 min to determine the critical limits for feeding, survival over 7 days (incipient lethal temperature), or survival for 10 min or less (ultimate lethal temperature). The rate of 1 °C/30 min was the optimum value from preliminary experiments, using nine rates from 0.5 °C/48 h to 18 °C h?1. As values for adults were not significantly different between populations, they were pooled to provide arithmetic means (with 95% CL) for the thermal limits at each acclimation temperature. 3. Feeding limits increased with acclimation temperature to upper and lower mean values (± 95% CL) of 26.5 ± 0.16 °C and 4.2 ± 0.20 °C for adults, 26.6 ± 0.59 °C and 5.0 ± 0.55 °C for juveniles. Incipient lethal levels defined a tolerance zone within which fish survive indefinitely; upper limits increased with acclimation temperature to a plateau of 27.6 ± 0.22 °C for adults and 27.5 ± 0.47 °C for juveniles, lower limits increased from near 0 °C to 2.5 ± 0.31 °C for adults and 2.7 ± 0.47 °C for juveniles. Ultimate lethal levels increased with acclimation temperature to a plateau of 32.5 ± 0.24 °C for adults and 32.6 ± 0.46 °C for juveniles, whilst the lower limits increased from near 0 to 0.9 ± 0.29 °C. Upper feeding, incipient and ultimate lethal values were significantly lower for juveniles than those for adults at acclimation temperatures < 20, < 20 and < 15 °C, respectively. 4. The thermal tolerance of bullheads was slightly lower than that of stone loach, similar to that of juvenile Atlantic salmon and higher than that of brown trout; the thermal limits for feeding were much wider than those for salmon or trout.  相似文献

The effect of incubation temperature (2, 4, 6, 8 and 10° C) on haddock Melanogrammus aeglefinus development and growth during the embryonic period and in subsequent ontogeny in a common post‐hatch thermal environment (6° C) was investigated. Hatching times were inversely proportional to incubation temperature and ranged from 20·3 days at 2° C to 9·1 days at 10° C. Growth rates were directly proportional to incubation temperature during both the embryonic and larval periods. There was a significant decline in growth rates following hatch in all temperature groups. Compared to the endogenously feeding embryos, growth rates in the exogenous period declined by 4·4‐fold at 4° C to 3·9‐fold at 8° C, indicative of the demarcation between the endogenous and exogenous feeding periods. Yolk utilization varied from 17 days at 2° C to 6 days at 10° C and followed a three‐stage sigmoidal pattern with the initial lag period inversely proportional to incubation temperature. Time to 50% yolk depletion varied inversely with temperature but occurred 1–1·5 days post‐hatch at all temperatures. Additionally, the period between 10 and 90% yolk depletion also decreased with increased temperature. Overall developmental rate was sequential with and directly proportional (2·3‐fold increase) to incubation temperature while the time spent in each developmental stage was inversely proportional to temperature. Larger embryos tended to be produced at lower temperatures but this pattern reversed following hatch, as larvae from higher temperature groups grew more rapidly than those from other temperature groups. Larvae from all temperatures achieved a similar length (c.total length 4·5 mm) upon complete yolk absorption. The study demonstrated the significant impact that temperature has upon developmental and growth rates in both endogenous and exogenous feeding periods. It also illustrated that temperature changes during embryogenesis had significant and persistent effects on growth in subsequent ontogeny.  相似文献

Upper limits for the ultimate lethal level increased with acclimation temperature to a maximum plateau of 33.5°C for adults (95% CL ± 0.19) and juveniles (95% CL ± 0.40), whilst the lower limits increased from near 0°C to 2.5 ± 0.30°C. At acclimation temperatures below 20°C, upper incipient and ultimate lethal values were significantly lower for juveniles than those for adults. 4. The thermal tolerance of stone loach was higher than that of juvenile Atlantic salmon or brown trout, one or both of these species often being dominant in streams with stone loach.  相似文献

A laboratory investigation was carried out into the effects of light and heat on the feeding and resting behaviour of Glossina morsitans morsitans Westwood. Flies placed in small cages on the ears of rabbits were tested for feeding responsiveness at light intensities of 0, 10, 100, 1000 and 25 000 lux. Feeding occurred at the same rate in darkness as it did in light at up to 1000 lux, but at 25 000 lux was largely inhibited. At 26° C, flies flew to and settled in the light end of a 1.2 m-long choice chamber. As the temperature was raised (by 10° C/hour) they eventually took off and flew directly to the dark end. The temperature at which this skototaxis occurred was strongly negatively correlated with the light intensity: each log unit increase in light intensity (in lux) caused a 2.2° C fall in take-off temperature. Skototaxis occurred at higher temperatures in recently fed female flies than in starved ones.  相似文献

The presence of two phenotypes in a single species is a widespread phenomenon, also observed in European eel (Anguilla anguilla). This dimorphism has been related to dietary differences in the subadult elver and yellow eel stages, with broad‐heads generally feeding on harder and/or larger‐bodied prey items than narrow‐heads. Nevertheless, both broad‐ and narrow‐headed phenotypes can already be found among glass eels, the stage preceding the elver eel stage. As these glass eels are considered nonfeeding, we investigate here to what degree the observed variation in head width is reflected in variation in the musculoskeletal feeding system, as well as whether this reflects the same variation observed in the older, dimorphic yellow eels. Additionally, we investigate whether musculoskeletal differences between broad‐ and narrow‐headed glass eels have implications on their feeding performance and could thus impact prey preference when eels start feeding. Therefore, we compared the cranial musculoskeletal system of five broad‐ and narrow‐headed glass eels using 3D‐reconstructions and simulated the glass eel's bite force using the data of the muscle reconstructions. We found that the variation in the musculoskeletal system of glass eels indeed reflects that of the yellow eels. Broader heads were related to larger jaw muscles, responsible for mouth closure. Accordingly, broad‐heads could generate higher bite forces than narrow‐headed glass eels. In addition, broader heads were associated with higher coronoid processes and shorter hyomandibulae, beneficial for dealing with higher mechanical loadings and consequently, harder prey. We, thus, show that head width variation in glass eels is related to musculoskeletal differences which, in turn, can affect feeding performance. As such, differences in prey preference can already take place the moment the eels start feeding, potentially leading to the dimorphism observed in the elver and yellow eel stage.  相似文献

The population dynamics of Metopolophium dirhodum were studied on winter wheat seedlings at constant (10°C, 15°C, 20°C, 25°C and 30°C) and fluctuating (12(night)-22(day)°C) temperatures, and during booting to early inflorescence, and anthesis to early milky ripe stage, at 19°C. The pre-reproductive development time was decreased by increasing the temperature from 10°C to 25°C. It was significantly shorter when the aphids were feeding during booting to early inflorescence than during anthesis to early milky ripe stage but was similar when the aphids were feeding on the flag, second or third leaves. The total number of nymphs produced/apterous adult was not significantly affected by temperature from 10°C to 25°C but adult reproductive lifespan was reduced by increasing temperature from 10°C and 12–22°C to 15°C, 20°C and 25°C. The daily intrinsic rate of increase changed from 0.11 to 0.25, and the cohort generation time decreased from 31 to 12 days, with increase of temperature from 10°C to 25°C. Reproductive rate was similar when the aphids were feeding on the flag, second or third leaves during booting to early inflorescence at 19°C. The reproductive rate was higher when the aphids fed from mid-inflorescence to mid-milky ripe stage than from mid-milky to early ripe stage. These results were compared with those from other studies. Predictions from a simulation model using development and reproductive rates from this study and literature were compared and the former rates resulted in a more accurate prediction of field observations in 1979, an outbreak year.  相似文献

Here, we examine the distribution, habitat use, and migratory destinations of North Pacific humpback whales wintering off Central America. Coastal boat surveys were conducted off Costa Rica and Panama between 1996 and 2003. In 1999, a broader survey was conducted along most of Central America. Over 23,000 km were surveyed, with the greatest effort off southern Costa Rica. We made 191 sightings of 320 individual humpback whales. Whales were seen between 14°N and 8°N, making this the most southerly of the North Pacific wintering areas. Encounters included singles, adult pairs, singers, and mother/calf pairs. Mother/calf pairs accounted for 27% of all groups sighted, which is one of the highest sighting rates reported among North Pacific wintering areas. Sixty percent of sightings occurred in depths <50 m. Average sea surface temperature was 28.6°C (±1.0 SD). Ninety percent of the 77 unique whales photo‐identified were also seen in the California–Oregon–Washington feeding aggregation. The 1999 survey showed that humpback whales were widely distributed along the Central American coast at relatively low densities. The extensive distribution of animals, the higher proportion of calves, and the almost exclusive migration to a single feeding area contrast with observations in other regions.  相似文献

Fish marking is an essential tool for fisheries management, especially for evaluating the stocking of endangered fish species to support conservation and sustainable use of fish stocks. Batch marking of young European eels Anguilla anguilla (L.) prior to stocking is recommended as the benefits of stocking for the spawning stock can be evaluated by recapturing marked fish over time, therefore mass marking of young eels with substances such as alizarin red S (ARS) is becoming increasingly important. To improve the marking method and reduce marking costs when immersing glass eels in an ARS solution, eight laboratory experiments under varying conditions (e.g., temperature, ARS concentration, immersion time, osmotic induction, fish density) and with ARS from different suppliers were carried out. The results show that optimal marking of glass eels can be carried out in the field or during transport by putting approximately 50 g of glass eels per liter in 150 mg L−1 ARS solution for 3 h at 10–15°C. Lower concentrations did not result in reliable marking. Water temperatures of 5°C and below can have a stunning effect on the eels and increase mortality significantly, regardless of the concentration of ARS. Glass eel densities below 50 g L−1 in the marking bath increase marking costs unnecessarily, while a higher density of 100 g L−1 resulted in significantly higher mortality and lower marking success. A somewhat more difficult but less expensive alternative is to bathe the fish in a saline solution of 1% (10 PSU) of 80 mg L−1 ARS for 3 h at 10°C. Costs can also be significantly reduced by choice of supplier for ARS, but care should be taken as the quality of the powder appears to vary (mean percentage of sufficiently marked eels ranged from 59% to 91% among suppliers in the present study) and can lead to marking failure. The optimal marking conditions can help ensure that stocked glass eels can be reliably identified in future studies to assess stocking benefits while reducing costs.  相似文献

Invasive species pose a major threat to aquatic ecosystems. Their impact can be particularly severe in tropical regions, like those in northern Australia, where >20 invasive fish species are recorded. In temperate regions, environmental DNA (eDNA) technology is gaining momentum as a tool to detect aquatic pests, but the technology's effectiveness has not been fully explored in tropical systems with their unique climatic challenges (i.e. high turbidity, temperatures and ultraviolet light). In this study, we modified conventional eDNA protocols for use in tropical environments using the invasive fish, Mozambique tilapia (Oreochromis mossambicus) as a detection model. We evaluated the effects of high water temperatures and fish density on the detection of tilapia eDNA, using filters with larger pores to facilitate filtration. Large‐pore filters (20 μm) were effective in filtering turbid waters and retaining sufficient eDNA, whilst achieving filtration times of 2–3 min per 2‐L sample. High water temperatures, often experienced in the tropics (23, 29, 35 °C), did not affect eDNA degradation rates, although high temperatures (35 °C) did significantly increase fish eDNA shedding rates. We established a minimum detection limit for tilapia (1 fish/0.4 megalitres/after 4 days) and found that low water flow (3.17 L/s) into ponds with high fish density (>16 fish/0.4 megalitres) did not affect eDNA detection. These results demonstrate that eDNA technology can be effectively used in tropical ecosystems to detect invasive fish species.  相似文献

The European eel Anguilla anguilla (eel hereafter) is critically endangered and has a catadromous life cycle, which means adult eels that live in pumped catchments must pass through pumps during their downstream spawning migration. Policy makers are currently lacking detailed site-by-site eel distribution information to estimate the overall impact of individual pumping stations on eel escapement, and as such lack the data to enable informed prioritisation of pumping station management and targeted mitigation. This study investigated whether environmental DNA (eDNA) metabarcoding can provide increased detection sensitivity for eel and fish community structure in highly regulated pumped catchments, when compared directly to current standard practice fish survey protocols (seine netting/electric fishing). Eels were detected in 14 of 17 sites (82.4%) using eDNA metabarcoding in contrast to 3 of 17 sites (17.6%) using traditional catch methods. In addition, when using eDNA monitoring, species richness was higher in 16 of 17 sites (94.1%), and site occupancy was greater than or equal to traditional methods for 23 of 26 of the fish species detected (88.5%). Although eDNA methods presented significantly higher average species richness and species site occupancy overall, eDNA and catch methods were positively correlated in terms of species richness and site occupancy. It was therefore found that eDNA metabarcoding was a high-sensitivity method for detecting eels in pumped catchments while also increasing the detection of overall fish community structure compared to traditional catch methods. In addition, this study highlights how eDNA monitoring is especially suited to increase the detection of particular species, with traditional methods sufficient for others. This high sensitivity, coupled with the ability to sample multiple sites in a short time frame, suggests that eDNA metabarcoding workflows could be invaluable tools when prioritising pumping station management.  相似文献

The aim of the experiment was to determine the impact of heat stress on nutrient digestibility and nitrogen balance in sheep fed silages differing in fibre quality. The digestibility trial was conducted at three different ambient temperatures (15°C, 25°C and 35°C for 24 h/d). The tested brown-midrib maize (Bm) silage had a higher nutrient digestibility, except for ether extract (EE) and a higher metabolisable energy (ME) content than the control maize (Con) silage. Nitrogen (N) excretion with faeces was higher but N excretion with urine was lower for sheep fed Bm silage, subsequently N balance did not differ between the two silages. Temperature had no effect on nutrient digestibility, except for crude protein (CP), but N excretion with urine was lower at elevated temperatures. A diet by temperature interaction was found for dry matter (DM) and organic matter (OM) digestibility. When the ambient temperature increased from 15°C to 25°C, the DM and OM digestibility increased in animals fed Con silage, but decreased in animals fed Bm silage. Concomitantly, ME estimated from digestible nutrients was higher for Bm than for Con at 15°C, but no differences were found at 25°C and 35°C. Effects of diet by temperature interaction, furthermore, were observed for EE and CP digestibility. Therefore, forage quality has to be considered when feeding heat-stressed animals.  相似文献

We investigated the environmental factors that affected temporal variability of eel recruitment and upstream migration in a freshwater coastal river along the southeastern US. Glass eels Anguilla rostrata were collected through ichthyoplankton sampling in the lower Roanoke River, North Carolina. Monthly samples were taken from fixed stations from May 2001 through June 2003. There was no evidence of consistent seasonal migration patterns for glass eels in Roanoke River. From May through December in 2001, glass eels were captured only during August. In 2002, glass eels arrived in February and remained in ichthyoplankton samples through October, with the exception of samples from September. Peak catch occurred in March at 4.02 ± 1.2 and declined through June to 0.18 ± 0.07 (#/1,000 m3). By August, the mean density increased to 0.96 ± 0.82 and to 3.59 ± 2.77 by October. In 2003 from January through June, glass eels were captured only during February and March. Glass eels were routinely collected when river discharge rates were <150 m3 s−1. River discharge rates >650 m−3 s−1 resulted in no glass eels in our samples. Upstream migration during 2002 was not correlated with water temperature or related to lunar phase. Glass eel freshwater upstream migration was initiated when water temperatures exceeded a threshold range of 10°C to 15°C; however, glass eels continued to migrate when water temperatures approached 30°C. The overall negative effect of river discharge suggests that changes in the water release schedules of upstream hydroelectric facilities during glass eel migration could strongly influence their recruitment success.  相似文献

The influences of water temperature and feeding regime on otolith growth in Anguilla japonica glass eels and elvers were investigated using individuals reared at 5, 10, 15, 20, 25 and 30° C and in fed or unfed conditions at salinity 32 after their otoliths were marked with alizarin complexone (ALC). To eliminate the difficulty of observing the edges of otoliths with optical (OM) or scanning electron (SEM) microscopes, three to 10 individuals were sampled from each tank at 10, 20 and 30 days during the experiment and reared for an additional 10 days at 25° C after their otoliths were marked a second time. Otolith growth and the number of increments were measured using both OM and SEM. Most A. japonica commenced feeding after 10 days at 20–30° C or after 20 days at 15° C, but no feeding occurred at 5 and 10° C. No otolith growth occurred at 5 and 10° C except in two individuals with minimal increment deposition at 10° C. Otolith growth was proportional to water temperature within 15–25° C and not different between 25 and 30° C. At 15, 25 and 30° C, the mean otolith growth rate in fed conditions was higher than in unfed conditions. The number of increments per day was significantly different among water temperatures (0·00–0·01 day−1 at 5 and 10° C, 0·43–0·48 day−1 at 15° C and 0·94–1·07 day−1 at 20–30° C). These results indicated that otolith growth in A. japonica glass eels and elvers was affected by temperature and ceased at ≤10° C under experimental conditions. Hence, future studies analysing the otoliths of wild-caught A. japonica glass eels and elvers need to carefully consider the water temperatures potentially experienced by the juveniles in the wild.  相似文献

Recruitment of Strongylocentrotus droebachiensis (Muller) was monitored for four successive years following a mass mortality of this species on a subtidal boulder bottom in St. Margaret's Bay, Nova Scotia. Recently recruited juveniles (1–2 mm diameter) were observed in the fall (September–November) between (1982) and (1984), but not in (1985). Juveniles grew to 6–8 mm diameter at 1-yr post-settlement and 19 mm diameter ((1983) cohort) at 2-yr post-settlement. Individuals > 18 mm diameter had macroscopic gonads in the spring of their third year and spawning probably occurred at 2.75-yr post-settlement. The growth rate of juveniles (3–6 mm initial diameter) maintained in cages with coralline-covered rocks was similar to that of the natural population. Growth and gonad index increased significantly when kelp (Laminaria longicruris (Pylaie)) was added to cages, suggesting that juveniles were food limited in St. Margaret's Bay. Despite an abundance of kelp and other fleshy algae, they remained cryptic under rocks and were not observed grazing on attached plants. The effects of temperature and food supply on juvenile growth and survival were examined in laboratory experiments using factorial designs. Recently-metamorphosed juveniles (from laboratory cultures), maintained at three temperature regimes (5°C, ambient, 16 °C) and two feeding regimes (no added food, coralline algae added), had similar growth rates (over 9 mth) among treatment combinations. However, growth was asymptotic at 16 °C and ambient temperatures, suggesting food limitation. Survival of these juveniles was higher in treatments with coralline algae, but survival was not affected by temperature. Field-collected juveniles (3–6 mm diameter), maintained at three temperature regimes (5°C, ambient, 16 °C) and three feeding regimes (no added food, coralline algae added, and kelp and coralline algae added), grew only in treatments with kelp added as food. In these treatments, growth rates increased with temperature. However, the growth curve at variable ambient temperature regimes was linear, suggesting seasonal acclimation. Survival of the field-collected juveniles was highest in treatments at 5 °C and/or with added kelp.  相似文献

The aim of this study was to determine if different methods of handling porcine, ovine, bovine and equine blood between collection and centrifugation influence measurable progesterone levels. A 2 × 2 × 5 factorial experiment was conducted for each species with heparin (with or without), temperature of incubation (4 and 22°C) and time of incubation (0, 6, 12, 24 and 48 h) as the main effects. Following centrifugation, plasma and serum samples were stored at ?20°C until progesterone concentrations were determined by radioimmunoassay. Method of handling porcine and equine blood between collection and centrifugation did not affect the levels of progesterone. However, heparinized blood held at 4°C resulted in the most consistent levels of progesterone over time. Progesterone levels were fairly consistent across time in the ovine blood by all methods of handling except heparinized blood incubated at 22°C. By 24 h after collection, plasma progesterone concentrations decreased by 50% for the ovine blood incubated at 22°C with heparin. Decreases were detected by all the methods of handling the bovine blood between collection and centrifugation. The rate of decline, however, was considerably faster for blood held at 22°C than blood held at 4°C. At 12–48 h after collection, the concentrations of progesterone averaged only 5% of the time 0 sample for blood incubated at 22°C. In contrast, at least 30% of the progesterone values in the time 0 sample were detected between 12 and 48 h of incubation for the blood held at 4°C.  相似文献

Current research targeting filtered macrobial environmental DNA (eDNA) often relies upon cold ambient temperatures at various stages, including the transport of water samples from the field to the laboratory and the storage of water and/or filtered samples in the laboratory. This poses practical limitations for field collections in locations where refrigeration and frozen storage is difficult or where samples must be transported long distances for further processing and screening. This study demonstrates the successful preservation of eDNA at room temperature (20 °C) in two lysis buffers, CTAB and Longmire's, over a 2‐week period of time. Moreover, the preserved eDNA samples were seamlessly integrated into a phenol–chloroform–isoamyl alcohol (PCI) DNA extraction protocol. The successful application of the eDNA extraction to multiple filter membrane types suggests the methods evaluated here may be broadly applied in future eDNA research. Our results also suggest that for many kinds of studies recently reported on macrobial eDNA, detection probabilities could have been increased, and at a lower cost, by utilizing the Longmire's preservation buffer with a PCI DNA extraction.  相似文献

In the laboratory, blood feeding rates, oögenesis and fat body development were utilized to assess the effects of photoperiod and temperature in the conditioning of adult female Culiseta inornata for aestivation. Long-day (16L:8D) and short-day (8L:16D) photoperiods were examined in combination with temperatures of 15°C, 20°C, and 25°C. All tested individuals were reared from egg to adult under one of six possible regimes.Blood feeding occurred in approximately 60% of the females subjected to short-day conditions. Females subjected to long-day regimes exhibited gonotrophic concordance (the inhibition of blood feeding), evidenced by a decrease in overall blood-feeding rate to 20%. Inhibition of blood feeding by long-day females decreased in a linear fashion with increasing temperature. No interaction of the effects of photoperiod and temperature upon blood feeding was apparent. Examination of the ovarioles of post-blood feeding females, reared under each of the above conditions, revealed no evidence of gonotrophic dissociation (the inhibition of oögenesis despite continued blood feeding).Fat body hypertrophy occurred in females reared under long-day conditions, whereas hypotrophy of this tissue was apparent in short-day females. No significant difference in fat body development occurred between parous and nulliparous females reared under long-day photoperiod conditions. Short-day blood-fed females reared at 15°C deposited a significantly greater amount of fat than short-day blood fed females reared at 20° and 25°C, and short-day nonbloodfed females reared at 15°, 20°, and 25°C. The primary stimulus for fat body hypertrophy appears to be long-day photoperiod conditions. Temperature exerted little discernible effect upon this process, and there was no interaction between the effects of photoperiod and temperature upon the degree of fat body development.  相似文献
