1. The routine oxygen consumption of Tilapia sparrmanii without the addition of any toxicants over a 72 hr period showed a decrease for the first 48 hr, but stabilised thereafter.2. Addition of zinc (98 mg l−1) resulted in a drastic decrease of oxygen consumption for 3 hr. The routine oxygen consumption showed a significant decrease for the first 24 hr, while the second and third 24 hr revealed significant differences with great individual variance.3. The decrease in oxygen consumption observed after exposure to zinc, could be caused by gill damage as well as the internal action of zinc.4. An increase in oxygen consumption was noted for almost 3 hr after addition of iron (88mg l−1). During the first-, second- and third 24 hr the oxygen consumption increased significantly, compared to the control values.5. The increase in routine oxygen consumption of T. sparrmanii when compared to control values after exposure to iron, could be attributed to stress and possible gill changes.6. The study revealed that after acute (72 hr) exposure to sublethal concentrations of zinc and iron, the routine oxygen consumption of T. sparrmanii was altered.  相似文献

Atrazine is the most commonly used agricultural herbicide in the midwestern cornbelt of the U.S.A. This project investigated atrazine bioconcentration in Cladophora glomerata (L.) Kütz, the dominant algae by biomass in nutrient enriched rivers. C. glomerata concentrates many organic and inorganic materials from solution, and by considering the partitioning coefficient of atrazine (Log K ow=2.56), the uptake of atrazine by this green algae was expected. The bioconcentration factor of C. glomerata ranged from 29 to 5223 over a one year sampling period, and bioconcentration was found to be a function of maximum biomass increase (due to high nutrients) and discharge. Uptake increased at low discharges as a result of increased contact time between cells and water. We found that C. glomerata was able to bioconcentrate a yearly average of 54% of the available atrazine from the water. Efficiency of uptake increased during the summer growing season of the algae (81%) and was 34% during the winter months. This field study demonstrates the ability of C. glomerata to bioconcentrate atrazine to a high degree and suggests an alternative and quicker biological removal of atrazine from agriculturally polluted rivers.  相似文献

Zinc plays a critical role in a diverse array of biochemical processes. However, excess of zinc is deleterious to cells. Therefore, cells require finely tuned homeostatic mechanisms to balance uptake and storage of zinc. Here we show that iron starvation affects zinc metabolism by downregulating expression of the plasma membrane zinc importer encoding zrfB and upregulating the putative vacuolar zinc transporter-encoding zrcA in Aspergillus fumigatus. Nevertheless, the zinc content of iron-starved mycelia exceeded that of iron replete mycelia, possibly due to unspecific metal uptake induced by iron starvation. In agreement with increased zinc excess and zinc toxicity during iron starvation, deficiency in siderophore-mediated high-affinity iron uptake caused hypersensitivity to zinc. Moreover, an increase of zinc uptake by conditional overexpression of zrfB was more toxic under iron depleted compared to iron replete conditions. This deregulated zinc uptake under iron starvation caused a decrease in heme production and an increase in protoporphyrin IX accumulation. Furthermore, zinc excess impaired production of the extracellular siderophore triacetylfusarinine C but not the intracellular siderophore ferricrocin. Taken together, these data demonstrate a fine tuned coordination of zinc and iron metabolism in A. fumigatus.  相似文献

AimsThe aim of this study was to investigate the effect of iron or/and zinc supplementation and termination of this treatment on the antioxidant defence of the male reproductive system and sperm viability in rats.MethodsThe study consisted of 3 stages: I) 4-week adaptation to the diets (C-control or D-iron deficient); II) 4-week iron and/or zinc supplementation (10-times more than in the C diet of iron: CSFe, DSFe; zinc: CSZn, DSZn; or iron and zinc: CSFeZn, DSFeZn; and III) 2-week post-supplementation period (the same diets as during stage I). Parameters of antioxidant status (total antioxidant capacity and SOD, GPx, and CAT activiy), oxidative damage (lipid and protein peroxidation), and sperm viability were measured.ResultsSimultaneous iron and zinc supplementation compared to iron supplementation (CSFeZn vs CSFe) increased SOD activity in the testes and decreased the level of malondialdehyde in the epididymis after stage II, and increased the percentage of live sperm after stage III. After discontinuation of the iron and zinc supplementation and a return to the control diet, the following was observed a decrease of SOD activity in the testes and GPx activity in the epididymis, and a increase malondialdehyde concentration in prostates. After stage III, in DSFeZn vs DSFe rats, an increase of SOD and CAT activity in the epididymis was found.ConclusionZinc supplementation simultaneous with iron may protect the male reproductive system against oxidative damage induced by high doses of iron and may have a beneficial effect on sperm viability. The effect of this supplementation was observed even two weeks after the termination of the intervention.  相似文献

The aim of this study was to evaluate the concentrations of copper, iron and zinc in blood serum of cats experimentally infected with Trypanosoma evansi. Animals were divided into two groups: control and infected with T. evansi. The animals were infected with 108 trypomastigotes each and parasitemia was estimated daily for 56 days by microscopic examination of smears. Hematological and biochemical parameters were evaluated for monitoring of the disease. Serum metal levels were determined in blood samples collected at days 7, 28 and 56 of the experiment. Inductively coupled plasma optical emission spectrometry was used to measure the levels of copper, iron and zinc. Significant differences were observed among groups (P < 0.05). Increased levels of copper and decreased iron and zinc levels were observed. A decrease in the number of red blood cells was also observed 7 days after inoculation. Biochemical parameters were not altered. Therefore, the infection by T. evansi might alter the serum metal levels, causing metabolic disturbances in cats.  相似文献

Biofortification through genetic manipulation is the best approach for improving micronutrient content of the staple food crops to alleviate hidden hunger, namely, the deficiency of Fe and Zn affecting more than two billion people worldwide. An interspecific hybridization was made between T. aestivum line Chinese Spring (CS) and Aegilops kotschyi accession 3790 selected for high grain iron and zinc concentration. The CS × Ae. kotschyi F1 hybrid with low chromosome pairing was highly male and female sterile. This was backcrossed with wheat cultivars to get seed set. The selfed BC1F1 and BC2F1 plants with high grain iron and zinc concentration were selected in subsequent generations. The selected derivatives showed 60–136% enhanced grain iron and zinc concentration and 50–120% increased iron and zinc content per seed as compared to the recipient wheat cultivars. Thirteen cytologically stable, fertile and agronomically superior plants with high grain iron and zinc concentrations were selected for molecular characterization. The application of anchored wheat SSR markers, transferable to Ae. kotschyi, to the high grain iron and zinc containing derivatives indicated introgression of group 2 and group 7 chromosomes of Ae. kotschyi. GISH and FISH analysis of some derivatives confirmed the substitution of chromosomes 2S and 7U for their homoeologues of the A genome, suggesting that some of the genes controlling high grain micronutrient content in the Ae. kotschyi accession are on these chromosomes.  相似文献

Despite recurrent exposure to zinc through inhalation of ambient air pollution particles, relatively little information is known about the homeostasis of this metal in respiratory epithelial cells. We describe zinc uptake and release by respiratory epithelial cells and test the postulate that Zn2+ transport interacts with iron homeostasis in these same cells. Zn2+ uptake after 4 and 8 h of exposure to zinc sulfate was concentration- and time-dependent. A majority of Zn2+ release occurred in the 4 h immediately following cell exposure to ZnSO4. Regarding metal importers, mRNA for Zip1 and Zip2 showed no change after respiratory epithelial cell exposure to zinc while mRNA for divalent metal transporter (DMT)1 increased. Western blot assay for DMT1 protein supported an elevated expression of this transport protein following zinc exposure. RT-PCR confirmed mRNA for the metal exporters ZnT1 and ZnT4 with the former increasing after ZnSO4. Cell concentrations of ferritin increased with zinc exposure while oxidative stress, measured as lipid peroxides, was decreased supporting an anti-oxidant function for Zn2+. Increased DMT1 expression, following pre-incubations of respiratory epithelial cells with TNF-α, IFN-γ, and endotoxin, was associated with significantly decreased intracellular zinc transport. Finally, incubations of respiratory epithelial cells with both zinc sulfate and ferric ammonium citrate resulted in elevated intracellular concentrations of both metals. We conclude that exposure to zinc increases iron uptake by respiratory epithelial cells. Elevations in cell iron can possibly affect an increased expression of DMT1 and ferritin which function to diminish oxidative stress. Comparable to other metal exposures, changes in iron homeostasis may contribute to the biological effects of zinc in specific cells and tissues.  相似文献

The present study was designed to investigate the effect of mercuric chloride administration on copper, zinc, and iron concentrations in the liver, kidney, lung, heart, spleen, and muscle of rats. The results showed that after dose and time exposure to mercuric chloride, the concentration of mercury in the six tissues was significantly elevated. Data showed that there were no interaction between mercury and tissue iron. There was a considerable elevation of the content of copper in the kidney and liver. The most significant changes in the copper concentration took place in the kidneys. About a twofold increase in the copper content of the kidney was noted after exposure to mercuric chloride (3 mg and 5 mg/kg). Only slight elevations in the copper content occurred in the liver, especially in high dose and longer exposure time. In the remaining organs, the copper content was not changed significantly (p>0.05). The most significant changes in the zinc concentration took place in liver, kidney, lung, and heart (5 mg/kg). Marked changes in kidney zinc concentrations were observed at any of the specified doses. Zinc concentrations were significantly increased in kidney of rats sacrificed 9–48 h after sc injection of HgCl2 (5 mg/kg); in liver obtained from rats at 18, 24, or 48 h after injection; and in lung after 24 or 48 h of treatment. The heart and spleen zinc concentrations were elevated at 24 and 48 h after injection of HgCl2 (5 mg/kg), respectively. The results of this study implicate that effects on copper and zinc concentrations of the target tissues of mercury may play an important role in the pathogenesis of acute mercuric chloride intoxication.  相似文献

Hyperthermia in boars reduces growth performance and sperm production. Zinc is an essential trace element in animal nutrition. Here we investigate the effects of dietary zinc on epididymal structure and function in Bama miniature pigs treated with heat exposure and investigate approaches to improve the reproductive performance in summer. Male Bama miniature pigs (n=18; aged 6 months; bodyweight=10.79±0.06 kg) were randomly allocated to 3 groups: control group (Control), heat treatment group (HT), and the diet-supplemented and heat treatment group (H+Zn). The Control and HT groups were fed with basal diet and the H+Zn group were fed with basal diet plus 1500 mg/kg zinc daily. After being fed with these 2 different diets for 30 days, pigs in the HT and H+Zn groups were exposed to 5 h of 40 °C heat treatment for 8 days. Rectal temperature and jugular venous blood were collected 3 h after onset of heat exposure on days 1, 4 and 8. Pigs were sacrificed after the termination of heat exposure. Heat treatment increased serum testosterone concentration on day 1 and 4 (P<0.01). In addition, the HT group displayed an increase in the clear cell count and a decrease in epithelium thickness in the caput epithelium (P<0.01, P<0.05), and dietary zinc protected the boars from these impairments (P<0.01, P=0.29). Evaluation of oxidative states showed that heat exposure increased the levels of malondialdehyde (MDA) and glutathione (GSH) in the epididymis (P<0.01, P<0.05), while dietary zinc reduced this elevation (P<0.01, P<0.01). Heat exposure enhanced the glucocorticoid receptor (GR) expression in the nuclei of principal and basal cells (P<0.01, P<0.01) while dietary zinc attenuated the GR immunoreactivity intensity (P<0.01, P<0.01). These results demonstrate that dietary zinc protects the epididymis from high temperature-induced impairment, alleviates oxidative stress, restores the integrity of the caput epithelium and decreases the stress response.  相似文献

Studies were performed regarding the effect of cadmium accumulation on the levels of essential elements (copper, zinc and iron) in the tissues of a marine bivalve mollusc, Mizuhopecten yessoensis, exposed to cadmium at 250 ppb during 2 weeks. It was found that the concentration of cadmium in the tissues increased in the order gonads < gills < hepatopancreas < kidney during exposure time. However, the highest value of concentration factor was recorded in the gills. Our data demonstrate that cadmium accumulation in all mollusc tissues is followed by the alterations in copper, zinc and iron concentration, but that the pattern of these changes varies with each tissue. Cadmium had the most pronounced effects on essential trace element homoeostasis in the kidney. The present study suggests that levels of the essential metals in a particular tissue can be modified depending on the level of cadmium accumulation. The possible mechanisms of the effects of cadmium on the essential trace elements are discussed.  相似文献

The effect of oxidoreductase inducers guaiacol and syringaldazine on the ability of Coriolus hirsutus, Coriolopsis fulvocinerea, Cerrena maxima, and cocultivated Coriolus hirsutus/Cerrena maxima to degrade atrazine in submerged cultures was studied. All the basidiomycetes reduced atrazine concentration with and without syringaldazine or guaiacol. The degree of atrazine degradation was higher in induced cultures (77–98% vs. 68–94% without induction). Of the four cultures, the highest detoxifying potential was observed in Coriolopsis fulvocinerea with and without an inducer (98% with guaiacol), and the lowest was in Cerrena maxima. Inducers decreased the residual atrazine concentration differently in the different cultures. A long-term increase of laccase production was observed in both induced and uninduced cultures, whereas the activity of Mn-peroxidase decreased. The results indicate that laccase plays a larger role in atrazine biodegradation than Mn-peroxidase.  相似文献

The effects of latent iron deficiency combined with parenteral subchronic or acute cadmium exposure during pregnancy on maternal and fetal tissue distribution of cadmium, iron and zinc, and on fetal viability were evaluated. Timed-pregnant Sprague-Dawley rats were fed on semisynthetic test diets with either high iron (240 mg kg) or low iron (10 mg kg), and concomitantly exposed to 0, 3 or 5 mg cadmium (as anhydrous CdCl2) per kilogram body weight. Animals were exposed to cadmium from gestation day 1 through 19 by subcutaneously implanted mini pumps (Subchronic exposure) or on gestation day 15 by a single subcutaneous injection (Acute exposure). All rats were killed on gestation day 19. Blood samples, selected organs and fetuses were removed and prepared for element analyses by atomic absorption spectrometry. Low iron diet caused decreases in maternal body weight, maternal and fetal liver weights, placental weights and tissue iron concentrations. By cadmium exposure, both subchronic and acute, tissue cadmium concentrations were increased and the increase was dose-related, maternal liver and kidney zinc concentrations were increased, and fetal zinc concentration was decreased. Cadmium concentration in maternal liver was additionally increased by low iron diet. Acute cadmium exposure caused lower maternal body and organ weights, high fetal mortality, and decreased fetal weights of survivors. In conclusion, parenteral cadmium exposure during pregnancy causes perturbations in essential elements in maternal and fetal compartments. Acute cadmium exposure in the last trimester of gestation poses a risk for fetal viability especially when combined with low iron in maternal diet.  相似文献

The macroalga Gracilaria lemaneiformis is an important and commercially valuable renewable resource. It is distributed widely in shallow marine waters but grows mostly on tropical or subtropical coasts. We investigated the accumulation of Cd, Cu, and Pb by live G. lemaneiformis under low concentrations. There was a positive correlation between the organisms’ metal concentrations and exposure concentrations. When exposed to both Cu and Cd, the concentrations of Cu and Cd in G. lemaneiformis were higher than those exposed to solutions of Cu and Cd alone. However, the concentrations of heavy metals in G. lemaneiformis were not markedly different (p?<?0.05) between the treatment groups and the control groups. We analyzed the results with nonlinear curve fitting and employed a two-compartment model to study the accumulation kinetics of heavy metals by G. lemaneiformis. The uptake rate constants and bioconcentration factors (BCFs) of the metals decreased with increased exposure concentration. The theoretical equilibrium concentrations increased significantly with the exposure concentrations. Our results suggested that G. lemaneiformis obviously accumulated heavy metals from seawater. As an important resource for food and pharmaceuticals, G. lemaneiformis should be cultivated in clear seawater.  相似文献

The interaction of dietary iron and zinc was studied in chicks. Zinc was found to be more toxic in iron-deficient animals than iron-supplemented animals as measured by hemoglobin concentrations and growth. Analyses of the kidney and liver for iron and zinc were carried out. As the level of iron was increased from 0-1000 ppm supplementation, the concentration of liver zinc increased. The organ levels of iron were decreased as the dietary zinc levels were increased from 0-5000 ppm. Radioisotope studies using65Zn revealed that the iron content of the diet did not affect absorption of zinc. Administration of the isotope, either in an intestinal segment or intravenously, resulted in more zinc being taken up by the liver in the iron supplemented animals. This was especially noted when the ratio of the isotope in liver to that in the blood was compared. Gel chromatography of kidney and liver homogenates revealed that iron deficiency resulted in less zinc being eluted in a volume characteristic of metallothionein compared to homogenates of organs from iron supplemented animals. The results indicate that iron-supplemented animals have a greater capacity for sequestering zinc on metallothionein than do iron-deficient animals. Conversely, iron-deficient chicks were more susceptible to the effects of zinc toxicity than are iron-adequate chicks.  相似文献

In excess, iron can induce the production and accumulation of reactive oxygen species (ROS), causing oxidative stress. The objective of this work was to evaluate the impact of toxic concentrations of iron (Fe) on the antioxidative metabolism of young Eugenia uniflora plants. Forty-five-day-old plants grown in Hoagland nutrient solution, pH 5.0, were treated with three Fe concentrations, in the form of FeEDTA, during three periods of time. At the end of the treatment, the plants were harvested and relative growth rate, iron content, lipid peroxidation and enzymes and metabolites of the antioxidative metabolism were determined. Iron-treated plants showed higher iron contents, reduced relative growth rates and iron toxicity symptoms in both leaves and roots. There was an increase in lipid peroxidation with increasing Fe, only in the leaves. The enzymatic activities of superoxide dismutase (SOD) and glutathione reductase (GR) increased with increasing Fe concentration and treatment exposure time. The activities of catalase (CAT), peroxidase (POX) and ascorbate peroxidase (APX) also increased with increasing Fe concentration but decreased with increasing treatment exposure time. Glutathione peroxidase activity (GPX) decreased with increasing Fe concentration and exposure time. The ascorbate (AA) and reduced glutathione (GSH) contents and the AA/DHA and GSH/GSSG ratios, in general, increased with increasing Fe concentration and treatment exposure time. The results indicate that under toxic levels of Fe, young E. uniflora plants suffer increased oxidative stress, which is ameliorated through changes in the activities of antioxidative enzymes and in the contents of the antioxidants AA and GSH.  相似文献

The rice (Oryza sativa) GTs belong to a super family possibly with hundreds of members. However, which GTs are involved in plant response to toxic chemicals is unknown. Here, we demonstrated 59 novel GT genes screened from our recent genome-wide sequencing datasets of rice crops exposed to atrazine (a herbicide persistent in ecosystems). Analysis of GT genes showed that most of the GTs contain functional domains typically found in proteins transferring glycosyl moieties to their target compounds. A phylogenetic analysis revealed that many GT genes from different families have diverse cis-elements necessary for response to biotic and environmental stresses. Experimental validation for the GTs was undertaken through a microarray, and 36 GT genes were significantly detected with an expression pattern similar to that from deep-sequencing datasets. Furthermore, 12 GT genes were randomly selected and confirmed by quantitative real-time RT-PCR. Finally, the special activity of total GTs was determined in rice roots and shoots, with an increased activity under the atrazine exposure. This response was closely associated with atrazine absorption in the rice tissues. These results indicate that exposure to atrazine can trigger specific GT genes and enzyme activities in rice.  相似文献

There are numerous reports of the effect of inulin on the bioavailability of mineral compounds. However, there are no conclusive reports concerning its beneficial impact (or lack thereof) in the case of such essential trace elements as iron, copper, or zinc. The aim of the study was to compare the effects of inulin addition with different degrees of polymerization (DPs) on growth performance in fatteners as well as on blood plasma concentrations of iron, copper, and zinc and selected hematological indices. The experiment was conducted throughout the fattening period (up to a body weight of approximately 115 kg) on 112 weaners with an initial weight of 25.0 ± 0.5 kg divided into 7 groups. The first group served as a control, while the other groups received increasing doses (1, 2, and 3 %) of standard inulin (SI; DPav ≥ 10) or long-chain inulin (LCI, DPav ≥ 23) in complete mixtures. Compared with the control, the supplementation of the mixtures with inulin increased the average daily gains, the final body weight, and the plasma content of trace elements (P < 0.05). An increased plasma zinc concentration was noted after application of inulin with a lower polymerization degree (P < 0.05). In turn, at a higher inulin polymerization degree, a higher final body weight and increased copper (P < 0.05), iron (P < 0.1), hemoglobin, mean corpuscular hemoglobin concentration (MCHC), and packed cell volume (PCV) levels were detected in animal blood (P < 0.05). The inulin addition was found to have modified the analyzed indices, and the optimal supplementation level was estimated at 20  g·kg?1 diet. Inulin with the higher DP exerted a more pronounced effect on the analyzed properties.  相似文献
