During the last decade, typical symptoms of Phytophthora diseases were observed in beech stands of several European countries. The main symptoms were the presence of bleeding cankers on the stem, a low crown density as well as the yellowing of foliage and the small size of leaves. Several species of Phytophthora, such as Phytophthora citricola, P. cambivora and P. cactorum, were reported as the causal agents. In order to evaluate the implication of the different Phytophthora species in beech decline in the southern part of Belgium (Wallonia), a monitoring was undertaken with the help of managers of public and private forests. Phytophthora strains isolated from beech of different stands as well as from soil were characterized through morphological and molecular analyses (PCR-RFLP of ITS). All the isolated strains were identified as P. cambivora, except for one strain whose identification is ongoing. Molecular analysis was also directly applied to necrosed tissues of bleeding beeches and enabled the detection of additional cases. All positive cases exhibited a profile characteristic of the species P. cambivora, except for one of the sampled trees showing a different Phytophthora profile also corresponding to the unidentified isolated strain. Identification of the Phytophthora species linked to this different RFLP profile is also ongoing. Both complementation types (A1 and A2) of P. cambivora were identified, sometimes in the same sampling site. Ornamented oogonia characteristic of this species were produced by pairing A1 and A2 strains isolated from the same site.  相似文献

The different subspecies of Phytophthora alni, P. alni subsp. alni (Paa), P. alni subsp. uniformis (Pau), and P. alni subsp. multiformis (Pam), are recent and widespread pathogens of alder in Europe. They are believed to be a group of emergent heteroploid hybrids between two phylogenetically close Phytophthora species. Nuclear and mitochondrial DNA analyses were performed, using a broad collection of P. alni and two closely related species, P. cambivora and P. fragariae. Paa possesses three different alleles for each of the nuclear genes we studied, two of which are present in Pam as well, whereas the third matches the single allele present in Pau. Moreover, Paa displays common mtDNA patterns with both Pam and Pau. A combination of the data suggests that Paa may have been generated on several occasions by hybridization between Pam and Pau, or their respective ancestors. Pau might have P. cambivora as a species ancestor, whereas Pam seems to have either been generated itself by an ancient reticulation or by autopolyploidization.  相似文献

Cytoplasmic and cell-wall antigens and antisera were prepared from four Phytophthora species, and cell-wall antigens were prepared from two Pythium species. Immunodiffusion of the Pythium and Phytophthora cell-wall antigens showed that the two Pythium species did not cross-react with the Phytophthora cell-wall antisera. Immunodiffusion analysis of both cell-wall and cytoplasmic antigens of Phytophthora revealed some degree of specificity between species but not between A1 and A2 mating types in Phytophthora cinnamomi. Species specificity was improved by using indirect fluorescent antibody techniques and by the use of cross-absorbed sera. Agglutination and quantitative precipitation techniques did not significantly improve specificity. It was possible to distinguish serologically between Phytophthora cinnamomi and Phytophthora cambivora and the Phytophthora cryptogea-Phytophthora drechsleri group. The absence of consistent serological variation between P. cryptogea and P dreschsleri is consistent with the suggestion (Bumbieris, 1974) that P. cryptogea and P. drechsleri should be considered as one species.  相似文献

Phytophthora alni subsp. alni, P. alni subsp. multiformis, and P. alni subsp. uniformis are responsible for alder disease in Europe. Class I and II elicitin gene patterns of P. alni subsp. alni, P. alni subsp. multiformis, P. alni subsp. uniformis, and the phylogenetically close species P. cambivora and P. fragariae were studied through mRNA sequencing and 3' untranslated region (3'UTR)-specific PCRs and sequencing. The occurrence of multiple 3'UTR sequences in association with identical elicitin-encoding sequences in P. alni subsp. alni indicated duplication/recombination events. The mRNA pattern displayed by P. alni subsp. alni demonstrated that elicitin genes from all the parental genomes are actually expressed in this allopolyploid taxon. The complementary elicitin patterns resolved confirmed the possible involvement of P. alni subsp. multiformis and P. alni subsp. uniformis in the genesis of the hybrid species P. alni subsp. alni. The occurrence of multiple and common elicitin gene sequences throughout P. cambivora, P. fragariae, and P. alni sensu lato, not observed in other Phytophthora species, suggests that duplication of these genes occurred before the radiation of these species.  相似文献

Single-strand-conformation polymorphism (SSCP) of ribosomal DNA of 29 species (282 isolates) of Phytophthora was characterized in this study. Phytophthora boehmeriae, Phytophthora botryosa, Phytophthora cactorum, Phytophthora cambivora, Phytophthora capsici, Phytophthora cinnamomi, Phytophthora colocasiae, Phytophthora fragariae, Phytophthora heveae, Phytophthora hibernalis, Phytophthora ilicis, Phytophthora infestans, Phytophthora katsurae, Phytophthora lateralis, Phytophthora meadii, Phytophthora medicaginis, Phytophthora megakarya, Phytophthora nicotianae, Phytophthora palmivora, Phytophthora phaseoli, Phytophthora pseudotsugae, Phytophthora sojae, Phytophthora syringae, and Phytophthora tropicalis each showed a unique SSCP pattern. Phytophthora citricola, Phytophthora citrophthora, Phytophthora cryptogea, Phytophthora drechsleri, and Phytophthora megasperma each had more than one distinct pattern. A single-stranded DNA ladder also was developed, which facilitates comparison of SSCP patterns within and between gels. With a single DNA fingerprint, 277 isolates of Phytophthora recovered from irrigation water and plant tissues in Virginia were all correctly identified into eight species at substantially reduced time, labor, and cost. The SSCP analysis presented in this work will aid in studies on taxonomy, genetics, and ecology of the genus Phytophthora.  相似文献

The cyanobacterium Nostoc strain ATCC 53789, a known cryptophycin producer, was tested for its potential as a source of natural pesticides. The antibacterial, antifungal, insecticidal, nematocidal, and cytotoxic activities of methanolic extracts of the cyanobacterium were evaluated. Among the target organisms, nine fungi (Armillaria sp., Fusarium oxysporum f. sp. melonis, Penicillium expansum, Phytophthora cambivora, P. cinnamomi, Rhizoctonia solani, Rosellinia, sp., Sclerotinia sclerotiorum, and Verticillium albo-atrum) were growth inhibited and one insect (Helicoverpa armigera) was killed by the extract, as well as the two model organisms for nematocidal (Caenorhabditis elegans) and cytotoxic (Artemia salina) activity. No antibacterial activity was detected. The antifungal activity against S. sclerotiorum was further studied with both extracts and biomass of the cyanobacterium in a system involving tomato as a host plant. Finally, the herbicidal activity of Nostoc strain ATCC 53789 was evaluated against a grass mixture. To fully exploit the potential of this cyanobacterium in agriculture as a source of pesticides, suitable application methods to overcome its toxicity toward plants and nontarget organisms must be developed.  相似文献

Plant Reactions to Inoculation of Roots with Fungi and Bacteria   总被引:1,自引:0,他引:1  
The potential of 120 isolates of fungi and bacteria from plant rhizospheres to interfere with plant development and growth was studied in greenhouse experiments. The pure cultured isolates were obtained from plant roots in the field and applied as suspensions to the roots of eight test plant species. 10–20% of the isolates caused distinct symptoms on shoots, growth retardations without other symptoms or growth promotions. Responses of treated plants ranged from death of plants soon after treatment to up to about 40% higher shoot fresh weight than in control plants. Two bacterial isolates induced strong reactions in most of the plant species tested while other isolates showed a more or less pronounced specificity by giving reactions in only some of the plant species tested.  相似文献

Effect of ectomycorrhizal fungi on chestnut ink disease   总被引:4,自引:0,他引:4  
 Seedlings of Castanea sativa were inoculated at transplanting time with four ectomycorrhizal (ECM) fungi, Laccaria laccata, Hebeloma crustuliniforme, H. sinapizans and Paxillus involutus. At the end of the first vegetative season, 7 months after sowing, half of the mycorrhizal and nonmycorrhizal seedlings were challenged with a zoospore suspension of Phytophthora cambivora and the other half with P. cinnamomi. Five months later, mycorrhizal plants infected with P. cambivora or P. cinnamomi showed no sign of pathogen infection. The ECM fungi increased plant biomass also in the presence of the pathogen. Mycorrhizal seedlings inoculated with the pathogens showed greater shoot and root development than nonmycorrhizal chestnut plants. All the fungi tested reduced the negative effect of the ink disease pathogens on the plant host in vivo. The mechanisms by which the ECM fungi protect chestnut seedlings are discussed. Between 2002 and the end of 2009, more than 4000 samples from hardy ornamental plants, collected in surveys for Phytophthora ramorum, were examined to establish the occurrence and diversity of Phytophthora species in Scotland. The samples were gathered from more than 77 plant genera in nurseries, gardens and amenity landscapes. Fifteen different Phytophthora spp. were isolated and identified either by polymerase chain reaction (PCR) or by sequencing of the ITS1, 5.8S subunit and ITS2 region of the ribosomal RNA gene. The most widespread Phytophthora spp. were P. ramorum and P. syringae, followed by P. cactorum, P. kernoviae, P. plurivora, P. cambivora, P. citrophthora, P. taxon Pgchlamydo’, P. pseudosyringae and some single isolates of P. cinnamomi, P. cryptogea, P. gonapodyides, P. nicotianae and P. hibernalis. One isolate did not match any known species. In relation to the number of samples, Phytophthora was found more frequently in trade premises than in gardens or amenity landscapes and the species diversity was higher, highlighting the risks involved in plant trade.  相似文献

Soil borne Phytophthora cinnamomi and Phytophthora cambivora are considered the most pathogenic species associated with chestnut (Castanea sativa) decline in Europe. Mapping their incidence and distribution from nursery and plantation soils may offer valuable information for limiting spread. As conventional biological baiting and taxonomic confirmation is generally time consuming, labour, logistically and space intensive, we have focused on the development of a specific touchdown nested multiplex Polymerase Chain Reaction (PCR) approach for the simultaneous detection of both species direct from soil. Pre-existing and novel primers, based on Internal Transcribed Spacer (ITS) sequences, have been evaluated for their specificity and use in a multiplex capacity in various combinations. Coupled to this we have modified a mechanical lysis procedure for DNA extraction from up to 10 g of chestnut under storey soils (ranging from 0.5 to 25 μg DNA g(-1)fresh soil). Using serial dilutions and/or polyvinylpolypyrrolidone chromatography purification, both species have been successfully detected, in artificially and naturally infected soils. Levels of assay detection are comparable to other Phytophthora species where PCR based diagnostic systems have been reported. A qualitative evaluation of this approach against conventional baiting is presented.  相似文献

Actively foraging lizards are capable of identifying prey using only chemical cues sampled by tongue-flicking, and the relatively few omnivorous and herbivorous lizards tested similarly can identify both animal and plant foods from chemical cues. Whether lizards that eat plants respond to cues specific to preferred plant types and whether there is geographic variability in responses to cues from various plants correlated with the importance of those plants in local diets is unknown. In three populations of an omnivorous lacertid, the Balearic lizard Podarcis lilfordi, we studied chemosensory sampling and feeding responses to chemical cues from plant and animal foods presented on cotton swabs. Each lizard population is endemic to one islet off the coast of Menorca, Balearic Islands, Spain. Lizards in all three populations discriminated chemical cues from plant and animal foods from control substances. Our results extend findings of prey chemical discrimination and plant chemical discrimination in omnivores, increasing confidence that correlated evolution has occurred between plant diet and chemosensory response to palatable plants. There were no consistent differences among populations in tongue-flicking and biting responses to stimuli from flowers of syntopic and allopatric plant species. The lizards may respond to cues indicative of palatability in a wide range of plant species rather than exhibiting strong responses only to locally available plant species. Nevertheless, tongue-flicking and biting frequencies varied among plant species, perhaps indicating food preferences. In addition, there were differences among populations in tongue-flick rates, latency to bite, and licking behavior. Licking was observed in only one lizard population as a response to floral chemicals from only one of the plants species tested, raising the possibility of a population-specific linkage between identification of a particular plant species and performance of an appropriate feeding response.  相似文献

The context‐dependent defence (CDD) hypothesis predicts that defence levels of plant species against herbivory are not fixed but vary with environmental conditions, in a way that is specific for plant species that share evolutionary adaptations to resource conditions exemplified by similar maximum relative growth rates. More specifically, we expected plants from resource‐poor environments to display high defence levels but not when grown under resource‐rich conditions, whereas the reverse – plants from resource‐rich conditions displaying low defence levels but not when grown under resource‐poor conditions – is not necessarily the case. In this study, we used multiple‐choice bioassays in which leaf discs were fed to larvae of Spodoptera exigua (Hübner) (Lepidoptera: Noctuidae) as an efficient and effective way of indicating plant defence levels. This generalist herbivore was capable of detecting both inter‐ and intraspecific differences in defence among plant species. The CDD was tested by exploring the effects of various experimental resource conditions (light, nutrients) upon the herbivore preferences and by comparing these preferences with the maximum relative growth rate of plant species. The experimental results provide general support for the CDD hypothesis with respect to nutrient‐level variation but the effects were not related to the origin of the plant species tested. Variation in light conditions did not result in consistent effects upon herbivore preferences. The CDD therefore can be formulated more precisely as: defence levels of plant species vary under different environmental conditions but in a way that is specific for plant species that share evolutionary adaptations to similar nutrient conditions. This more precise CDD hypothesis is a useful addition to existing optimal‐defence theory because of its focus on the possible plastic effects of resource conditions upon plant defence levels. This is relevant when designing experimental plant–herbivore studies.  相似文献

This meta-analysis summarizes published information on the use of various plant species by parasitoid wasps. Trials measured either a physiological response such as wasp longevity when supplied with flowers from a single plant species, or a behavioral response like attraction of wasps to field plantings. The log response ratio was the effect size used to standardize estimates and meta-analyses were conducted to make overall estimates for plant species included in multiple trials. Physiological response trials have been conducted on 126 different plant species and behavioral response trials tested 104 plant species. The log response ratio of different plant species ranged from 0 to 2.7 for longevity, and up to 4.1 for attraction. The longevity response ratio estimate is equivalent to a nearly 15-fold increase in the ratio of days longevity with the flower to days longevity in the starvation control. Wasp longevity increased the most with plants in the Polygonaceae, Apiaceae, Brassicaceae, Boraginaceae, Solanaceae, and Rosaceae. Plant species in the Onagraceae, Caryophyllaceae, Lamiaceae, Scrophulariaceae, Asteraceae, and Fabaceae tended to slightly increase wasp life spans, but their effect was lower and less consistent among species. There were a number of families which did not increase wasp longevity over the controls at all such as Chenopodiaceae. This review can help identify plant species which have been proven to supply nectar for parasitoids for potential use in a conservation biological control program, but plant selection should not be limited to the small list of species that were included in these studies.  相似文献

Selected rhizosphere bacterial isolates, previously determined as plant growth deleterious, were tested for their ability to suppress plant growth after foliar spray applications, for selectivity with regard to plant species, and in pilot field experiments for their potential as weed biocontrol agents. Inundative foliar applications of aqueous bacterial suspension were performed on a range of weed and crop species. Plant symptoms after spraying ranged from rapid necrosis and wilting to an overall growth suppression or stunting. Significant and selective reductions in biomass of up to 90% fresh weight, as well as large reductions in plant survival and plant height were recorded in greenhouse pot experiments. However, monocotyledonous plants were affected weakly or not at all by two isolates extensively tested. Effects of these were dose- and plant age-dependent, and were for some plants enhanced by high relative humidity. For one isolate, A153, effects were also expressed in cell-free culture filtrates pointing to involvement of specific metabolites. In pilot field experiments, strong growth suppression was observed on broad-leaved plants, while barley crop plants were unaffected.  相似文献

The use of exuding stylets holds considerable promise for the investigation of sieve-tube physiology. However, largely because of difficulties in cutting insect stylets, the technique has been applied to only a few plant species. Based on our experience, a comparison is made of the available means of obtaining sieve-tube exudate from the exuding stylets of phloem-feeding insects, including aphids, scale and mealybugs. Forty-one plant species and approx. 35 insect species were tested for their ability to provide stylet exudate. Stylets on all but a few of the plant species tested yielded at least some exudate, but the success rate and duration of exudation on many species were unsatisfactory for detailed investigations of phloem transport. Plant species appears to be the most important factor for obtaining reliably exuding stylets, although the size of the insect species used and the physiological condition of the plant are also important variables. Armored scale provide a simple and reliable source of exuding stylets, but are impractical for most experimental purposes. Radio-frequency microcautery of aphid stylets was substantially the most effective means of cutting stylets. Since N-acyl homoserine lactones (AHLs) are key mediators of cell density-dependent regulation of traits involved in virulence and epiphytic fitness in gram-negative bacteria such as Pseudomonas syringae, a variety of plant species were examined to determine their production of leaf surface compounds that could interact with these signaling systems. Leaf washings of 17 of 52 plant species tested stimulated or inhibited AHL-dependent traits in at least one of the bacterial reporter strains used. The active compounds from most plants could be distinguished from known AHLs due to different patterns of mobility during C8 and C18 reverse-phase thin-layer chromatography (TLC) and normal-phase TLC compared to the patterns for authentic bacterial AHLs. All plant extracts were also tested to determine their abilities to sequester iron and trigger bacterial siderophore synthesis on a medium containing abundant iron. Leaf washings from 16 of the 52 plant species, as well as tannic acid solutions, stimulated pyoverdine synthesis in P. syringae in a high-iron medium. These preparations also inhibited the growth of a P. syringae mutant unable to produce pyoverdine siderophores but not the growth of the wild-type bacterium. The stimulation of siderophore production and the growth inhibition by plant extracts and purified tannins were both reversed by addition of ferric chloride to culture media, indicating that iron was made unavailable by the compounds released onto the leaf surface.  相似文献

When colonizing new areas, alien plant species success can depend strongly on local environmental conditions. Microclimatic barriers might be the reason why some alien plant species thrive in urban areas, while others prefer rural environments. We tested the hypothesis that the climate in the native range is a good predictor of the urbanity of alien species in the invaded range. The relationship between climate in the native range and the percentage of artificially sealed surfaces (urbanity) at the occurrences of 24 emerging alien plant species, in European areas with a temperate climate (termed oceanic Europe) was evaluated. We found that alien species growing in more urban environments originated from warmer or drier native ranges than found in oceanic Europe. These results have strong conservation implications as climate-warming will likely lift climatic barriers that currently constrain numerous alien plant species to cities, boosting the role of cities as points of entry for invasive plants.

Plant individuals rely on pollinator services for their reproduction and often have to share these services with co‐occurring neighbours, creating complex indirect plant–plant interactions. Many current theoretical models focus on the effect of floral resources’ density on the net outcome of these indirect plant–plant interactions, often neglecting the identity of plant species in the communities and especially the species’ spatial pattern. To fill this gap, we created a spatially explicit model whose goal was to study the interplay between relative densities and spatial distribution patterns of two plant species differing in their attractiveness for pollinators. Since theory predicts that pollinator behaviour strongly governs the outcome of pollination, we allowed the pollinators to systematically change their plant preferences based on their foraging experience. Thus the interplay between density and spatial patterns of plants was tested over a continuum of behaviours from specialists to generalists. Our most striking finding was that reproductive success of the less attractive species was affected in an opposite way by spatial patterns depending on whether the species had relatively low or high densities. Namely, when the less attractive species was highly abundant, its survival was higher when aggregated in large monospecific patches than when uniformly distributed. On the other hand, when the attractive species was more abundant, the less attractive species survived better when uniformly distributed. These results were consistent as long as the scale of the plant spatial aggregation was similar to or larger than the pollinators’ detection range. Our results suggest that aggregated plant spatial patterns manipulate pollinator behaviour by trapping them within monospecific patches. This effect was sufficiently strong to enhance the survival of a competitively inferior species and hence to act in a way similar to the more familiar niche or temporal separation among plant species.  相似文献

Homogenates from several insect species were assayed for inhibition of acetylcholinesterase by the potato glycoalkaloid alpha-chaconine. Colorado potato beetle acetylcholinesterase was up to 150-fold less sensitive than other species tested. Acetylcholinesterase from an insecticide-resistant strain of Colorado potato beetles was more sensitive to inhibition than the susceptible strain. Most insect species tested had inhibitory concentrations causing a 50% reduction in activity in the 5 to 40 microM range. Sensitive insect acetylcholinesterases were similar to mammalian cholinesterases in their response to alpha-chaconine. The results indicate that pesticides and host plant resistance factors may interact at the same target. Changes in the target due to selection pressure from either pesticides or host plant resistance factors could affect the efficacy of both control strategies.  相似文献
