Phenolic compounds have increasingly attracted global concerns in recent years due to their strong bioaccumulation and potential toxicity. In this study, the adsorption of 2, 4-Dichlorophenol (2, 4-DCP), 2, 4-Dinitrophenol (2, 4-DNP) and 2, 4-Dimethyphenol (2, 4-DMP) on sediment at different temperatures was studied. Adsorption isotherms fitted well to the Freundlich model and the adsorption capacity was increased when the temperature increased from 5°C to 25°C. It was found that the adsorption process could be modeled well using pseudo-second-order kinetic equation. The thermodynamic parameter ΔG° for 2, 4-DCP, 2, 4-DNP and 2, 4-DMP varied between ?8.82 and ?13.68, ?4.9 and ?8.05, and ?7.52 and ?10.55 kJ/mol, respectively, with ΔH° (kJ/mol) and ΔS°(J/(mol·K)) measuring 55.397 and 0.2263, 40.121 and 0.1585, and 38.012 and 0.16, respectively. The calculated thermodynamic parameters suggested that adsorption of the three selected phenolic compounds was a spontaneous (ΔG°< 0), endothermic (ΔH°> 0), and entropy-driven reaction (ΔS° > 0). The thermodynamic data also suggested that the three selected phenolic compounds to sediment were closed to chemisorption because ΔH° was around 40.  相似文献

The present study was conducted to investigate the effect of decomposition site and plant litter species on the colonizing microbial communities. For this, litter bag technique using beech and spruce litter was combined with RNA-based fingerprinting and cloning. Litter bags were incubated for 2 and 8 weeks in the Ah horizon of beech and beech–spruce mixed forest sites. Although sugars and starch were rapidly lost, lignin content increased by more than 40% for beech and more than doubled for spruce litter at both soil sites at the end of the experiment. Denaturing gradient gel electrophoresis analysis of 16S and 18S rRNA RT–PCR products was used for screening of differences between bacterial and fungal communities colonizing the two litter types. Development of the microbial community over time was observed to be specific for each litter type and decomposition site. RT–PCR products from both litter types incubated in beech–spruce mixed forest site were also cloned to identify the bacterial and fungal colonizers. The 16S rRNA clone libraries of beech litter were dominated by γ-proteobacterial members, whereas spruce libraries were mainly composed of α-, β-, and γ-proteobacterial members. Ascomycota members dominated the 18S rRNA clone libraries. Clones similar to Zygomycota were absent from spruce, whereas those similar to Basidiomycota and Glomeromycota were absent from beech libraries. Selective effects of litter quality were observed after 8 weeks. The study provides an insight into the bacterial and fungal communities colonizing beech and spruce litter, and the importance of litter quality and decomposition site as key factors in their development and succession.  相似文献

The high biodiversity observed in coastal lagoons can often be related to a very high degree of environmental heterogeneity (β-biodiversity). In this article, I describe many physical factors that contribute to create environmental heterogeneity and different habitats within the Bassin d’Arcachon, a mesotidal lagoon in S.W. France. The microbial biodiversity has been studied in this coastal lagoon by culture-dependent and culture-independent techniques. The sediments of the meadows of the seagrass Zostera noltii have received a specific focus. During the last 15 years, several novel bacterial species have been isolated from these sediments and obtained in culture. Among these, five different species have been fully described as novel species according the Bacteriological Code and two of them represent also the type strain for a novel genus. The bacterial biodiversity has also been studied by molecular ecology techniques. An early study was focused on water column bacteria and a comparison has been made with the same community from the more eutrophic Prévost lagoon (France, Mediterranean coast). A clone library was also constructed for 16-S rRNA genes isolated and amplified from the 1–2 cm depth layer of the Z. noltii sediment. These studies showed a high degree of biodiversity and the importance of sulfate-reducing bacteria in the sediment, pointing to species that have not been obtained in culture. Collectively, these data show that there is still a large discrepancy between culture-dependent and culture-independent prokaryotic biodiversity. It is important to link the functional biogeochemical approach with the biodiversity approach. For the Bassin d’Arcachon studies, the biogeochemical approach has successfully nurtured the biodiversity studies, while the reverse has not yet been the case as the biogeochemical studies have lumped prokaryotes in functional groups. Specimens of Annalepis have been found from the Middle Triassic Series in the Yangtze River valley and described, including a new species. Annalepis is similar to the living plant, Isoëtes , in many aspects, such as the herb with a corm, more or less clustering sporophylls, presence of a ligule, monolete suture microspores of Aratrisporites -type and trilete megaspores, etc. The similarities of features between the two plants indicate their close affmities and that A. brevicystis might be the ancestry of Isoëtes . Since there is a ligule on the sporophyll of Pleuromeia, the Pleuromeiaceae represented by Pleuromeia could be allied to the Isoëtaceae, and both families should belong to Isoëtales. Isëtales represents a distinct evolutionary sequence and is inferred as being evolved probably from Palaeozoic SigiUaria . In Mesozoic age, two evolutionary lines are proposed: one as a line from Pleuromeia to Nathorstiana in relation to wood-monopolar rhizophore; the other as a line from Annalenis to lsoOtes related to herb-bipolar corm. Annalepis appeared almost simultaneously in the Yangtze River valley in early Middle Triassic, but the distribution of Annalepis was concentrated from east to west during middle-late Middle Triassic, in relation to the marine regression which occurred cyclically from east to west on the Yangtze Platform during that time.  相似文献

Objective

Teachers'' attitudes toward inclusion are often based on the practical implementation of inclusive education rather than a specific ideology and understanding of inclusiveness. This study aimed to identify the factors associated with primary school teachers'' attitudes towards inclusion of students with all disabilities in regular schools.

Method

Seventy four primary school teachers participated in a cross-sectional survey conducted in Western Australia. Teachers'' attitudes and efficacy toward integration of students with disabilities were measured using the Opinions Relative to Integration of Students with Disabilities scale and Bandura''s Teacher Efficacy scale respectively.

Results

Four teacher attributes—age, gender, teaching self-efficacy and training collectively explained 42% of the variability in teachers'' attitude toward including students with disabilities.

Conclusion

The current study further contributes to the accumulation of knowledge that can unpack the complex pattern of factors that should be considered to promote positive attitudes towards inclusive schools.  相似文献

Large soluble oligomeric species are observed as probable intermediates during fibril formation in aggregations of Parkinson’s disease (PD). Fibrillar deposits occur in PD. Amyloid forms α-Synuclein is one of the main compounds aggregations. β-Casein, a member of the Casein family, has been demonstrated to inhibit α-Synuclein aggregation by chaperone-like activity. In this study, we investigated the effect of chaperone activity of β-Casein in preventing the aggregation of α-Synuclein protein. We have examined the effect of β-Casein in preventing α-Synuclein aggregation by using from Thioflavin T-binding assay, transmission electron microscopy, ANS-binding assay, circular dichroism spectroscopy and FTIR spectroscopy. Results from the ThT binding assay demonstrated an increase in the ThT fluorescence intensity of α-Synuclein incubated in absence of β-Casein but in its presence fluorescence intensity is decreased. Electron microscopy data also indicated that β-Casein decreased the aggregation content of α-Synuclein. ANS results also showed that β-Casein significantly decreased the the hydrophobic area in α-Synuclein incubated. Circular dichroism spectroscopy (CD) results also showed that β-sheet structures of α-Synuclein incubated change to structural α-helical and β-turn in presence of β-Casein. FTIR spectroscopy indicates the presence of β-sheet structures in α-Synuclein incubated in absence of β-Casein and β-sheet structures decreased in its presence. Thus, our results suggest that in vitro, β-Casein interacts with α-Synuclein fibrils, changes the α-Synuclein structure and prevents amyloid fibril formation. This means that β-Casein could be essential for therapies inhibiting aggregation and to be an important therapeutic drug against PD.  相似文献

Background

Mothers that mate with multiple males often produce higher quality offspring than mothers that mate with a single male. By engaging in polyandry, mothers may increase their chances of mating with a compatible male or promote sperm competition - both of which act to increase maternal fitness via the biasing of the paternity of offspring. Surprisingly, mating with multiple males, can carry benefits without biasing paternity and may be due simply to differences in genetic diversity between monandrous and polyandrous clutches but role of genetic diversity effects in driving the benefits of polyandry remains poorly tested. Disentangling indirect, genetic benefits from genetic diversity effects is challenging but crucial if we are to understand the selection pressures acting to promote polyandry.

Methodology/Principal Findings

Here, we examine the post-fertilisation benefits of accessing the sperm of multiple males in an externally fertilising polychaete worm. Accessing the sperm of multiple males increases offspring performance but this benefit was driven entirely by genetic diversity effects and not by the biasing of paternity at fertilisation.

Conclusions/Significance

Previous studies on polyandry should be interpreted cautiously as genetic diversity effects alone can explain the benefits of polyandry yet these diversity effects may be difficult to disentangle from other mechanisms. We suggest that future studies use a modified experimental design in order to discriminate between genetic diversity effects and indirect, genetic benefits.  相似文献

Signal transduction within the DNA damage response is driven by the flux of protein-protein interaction cascades that ultimately recruit repair complexes to sites of damage. The protein RAP80 plays a central role in the damage response by targeting BRCA1/BRCA2 tumor suppressors to DNA damage foci through multivalent binding of Lys-63-linked polyubiquitin chains. Mutations within the high penetrance BRCA1/BRCA2 genes account for ∼20% of familial breast cancers. The genetic basis for the remaining cancers remains unknown, but may involve defects in binding partners for BRCA1 and BRCA2 that lead to impaired targeting to foci and a concomitant role in the pathogenesis of cancer. Recently, an in-frame deletion mutation (ΔE81) in a conserved region from the first ubiquitin interaction motif of RAP80 has been linked to an increase in chromosomal abnormalities. Using NMR spectroscopy, we demonstrate that the N-cap motif within the α-helix of the first ubiquitin interaction motif from ΔE81 undergoes a structural frameshift that leads to abolishment of multivalent binding of polyubiquitin chains. Loss of this single glutamate residue disrupts favorable electrostatic interactions between RAP80 and ubiquitin, establishing a plausible molecular basis for a potential predisposition to cancer unrelated to mutations within BRCA1/BRCA2 genes.  相似文献

Some molecular properties of αs1-κ-casein complex, αs1- and κ-casein polymers were examined by gel filtration, ultracentrifugation, and viscometry at pH 7.1. The Stoke’s radii of αs1-κ-casein complex, αs1- and κ-casein polymers were 99, 44 and 108 Å, respectively. The molecular weight of the above proteins were approximately 45 × 104, 10 × 104 and 80 × 104, respectively. The stokes radius of αs1-κ casein complex reduced compared with that of κ-casein polymer and the molecular weight of the complex was about half that of κ-casein polymer. These results suggest that κ-casein polymer dissociates into 4 smaller particles when αs1-κ-casein complex is formed. The frictional coefficient and Scheraga-Mandelkern constants for each protein suggest that the molecular shape of αs1-casein polymer is globular and that of αs1-κ-casein complex and κ-casein polymer is rod-like.  相似文献

Summary An attempt has been made to find out the productivity and periodicity of the phytoplankton in relation to the physical and chemical characteristics of the river Ganges, between University Ghat and Raj Ghat at Varanasi. The observations were taken during a single year (i.e., March 1957 to March 1958). The chemical analyses of the water and estimates of the number of phytoplankton were made on samples collected fortnightly.The phytoplankton has proved to be eutrophic and polymixic in nature. Diatoms formed the largest bulk, blue-greens being the second major constituent while the green algae formed the third in order of abundance with, however, large number of species. A definite phytoplankton periodicity has been noticed and found to be seasonal in character.Correlations between the phytoplankton periodicity and population maxima, with the habitat factors revealed depletion of nutrients like, nitrogen, phosphate, silica, carbon, magnesium and sulphates.Changes in the phytoplankton populations were clearly evident more in relation to physical than to chemical conditions of the water. Changes in water-level, transparency and temperature affected the growth of the phytoplankton.High alkalinity and buffering capacity resulted in preferential growth of diatoms and blue-green algae. Rich phosphates and silicates coupled with moderate nitrogen contents were responsible for high phytoplankton yields in summer and winter seasons.The abundance of blue-greens may be due to the higher values of pH, temperature, dissolved organic matter, phosphate, nitrogen, and relatively high values of dissolved oxygen. Green algae showed a wide range of adaptability but could not develop in any abundance.  相似文献

Behçet’s disease (BD), a multi-organ inflammatory disorder, is associated with the presence of the human leukocyte antigen (HLA) HLA-B*51 allele in many ethnic groups. The possible antigen involvement of the major histocompatibility complex class I chain related gene A transmembrane (MICA-TM) nonapeptide (AAAAAIFVI) has been reported in BD symptomatic patients. This peptide has also been detected in HLA-A*26:01 positive patients. To investigate the link of BD with these two specific HLA alleles, molecular dynamics (MD) simulations were applied on the MICA-TM nonapeptide binding to the two BD-associated HLA alleles in comparison with the two non-BD-associated HLA alleles (B*35:01 and A*11:01). The MD simulations were applied on the four HLA/MICA-TM peptide complexes in aqueous solution. As a result, stabilization for the incoming MICA-TM was found to be predominantly contributed from van der Waals interactions. The P2/P3 residue close to the N-terminal and the P9 residue at the C-terminal of the MICA-TM nonapeptide served as the anchor for the peptide accommodated at the binding groove of the BD associated HLAs. The MM/PBSA free energy calculation predicted a stronger binding of the HLA/peptide complexes for the BD-associated HLA alleles than for the non-BD-associated ones, with a ranked binding strength of B*51:01 > B*35:01 and A*26:01 > A*11:01. Thus, the HLAs associated with BD pathogenesis expose the binding efficiency with the MICA-TM nonapeptide tighter than the non-associated HLA alleles. In addition, the residues 70, 73, 99, 146, 147 and 159 of the two BD-associated HLAs provided the conserved interaction for the MICA-TM peptide binding.  相似文献

Plant species diversity is surveyed in the southern part of the Taï National Park (TNP), Côte d’Ivoire. This park is the largest remaining tract of pristine forest in the West African rainforest biodiversity hotspot. Plant assemblages and environment factors were surveyed in 39 temporary 625-m2 plots within three areas. The species list was completed thanks to itinerant prospections. TWINSPAN classification and detrended correspondence analysis were applied to the 39 relevés. We recorded a total of 916 plant species, including 169 West African endemics, that rises the floristic richness of TNP in 1233 species. Most plant species recorded were Guineo-Congolian species (90.9%) among which 8% were ‘Sassandrian species’, i.e. endemics of the extreme southwest Côte d’Ivoire. Eight of these endemics were exclusive of the southern part of TNP. The forest is very species-rich, particularly in Rubiaceae, Leguminosae and Euphorbiaceae. Most of the sampled species (59.5%) were erect phanerophytes with a dbh≤10 cm. We distinguished 7 plant community types, mainly determined by geomorphology and disturbance history. Lowland forests were typical dense evergreen forests with different degrees of maturity related to historical human impact. Inselbergs and swampy depressions provide an original azonal vegetation. We conclude that the southern part of TNP contributes significantly to the regional biodiversity hotspot since it includes the last remnants of primary evergreen rainforest and contains species assemblages from both higher rainfall biomes on moist soils derived from schist parent material and lower rainfall biomes on inselberg slopes and top. As anthropogenic disturbance clearly reduces biodiversity, these last remnants should be integrally protected.  相似文献

D’Orbigny’s stratigraphic contributions were enormous, both practical and theoretical. In his practice, d’Orbigny realised the central importance of using fossil faunas to correlate strata across different countries. His ‘succession chronologique des âges du monde’ (of 1852) recognised 5 periods and 27 stages (ignoring his ‘époque actuelle’). The great majority of the latter were from two Periods, Jurassic (stages 7 to 16) or Cretaceous (17 to 23). D’Orbigny described the palaeontological characteristics, and geographical extensions, of each. Some stages he based on countries he never visited and although he never specified ‘stratotype’ localities, he referred to some as ‘étalon’, ‘le mieux’ or ‘le plus beau’. These leads proved crucial when, in the mid 20th century, stratigraphers realised that definitions of chrono-stratal units must be attempted. The first attempts at typification simply followed zoology, where a type specimen tries to define a central position within the morphological variation of a species. In 1962, an early attempt was made to define Jurassic stages by three type horizons at different type localities (or stratotypes as they were now called) for each. A central horizon was chosen as ‘lectotype section’ and upper and lower boundaries for each were defined elsewhere. In France, it was preferred to investigate original intentions at named localities, through unit-stratotypes. These allowed some nationalistic pride, since France was the home of so many d’Orbignyan stages. 1980 saw the publication Les Étages français et leurs Stratotypes. However, British Silurian stratigraphers had realised that such zoologically based concepts would produce conflict, when any defined upper boundary stratotype conflicted with the defined lower boundary of a superjacent unit. They suggested, from 1962, to define only lower boundaries, via ‘golden spikes’, at basal-boundary stratotypes. This was the method chosen for the basal Devonian stratotype at Klonk, Czech Republic, defined in 1972, and a method adopted globally from 1986. However, this was when correlations were still largely based on fossils. The explosion of so many other stratigraphies since, based on magnetic or chemical records, events etc, has produced a first reaction against the idea that such stratotypes should be so defined, while a second, potentially more major claim, is that any ‘golden spike’ concept may no longer ‘glitter’, as competing stratotype concepts may be holding back stratigraphic progress.  相似文献

Summary A detailed and systematic study of the algae present in the guts of several fishes from the river and their percentage composition was carried out. Only a few fishes like Hilsa ilisha, Gadusia chapra and Barbus stigma showed herbivorous and omnivorous habits. Definite grazing of the phytoplankton by fishes was clearly envisaged. A protist tentatively designated “Pseudoperkinsus tapetis” belonging to the eukaryotic group Ichthyosporea (Mesomycetozoa) was previously isolated from carpet shell clams in Galicia (northwest Spain). In the present study, based on molecular data, a potential P. tapetis specimen was identified from the gill tissues of the mussel Adipicola pacifica associated with whale carcasses (generating chemosynthetic-based ecosystems) collected at shelf depths in the northwest Pacific (southwest Japan). Small subunit ribosomal DNA sequences (1751 sites) of the genotypes of P. tapetis from Spain and Japan were almost identical (only one substitution and one insertion/deletion difference). On the other hand, differences of 10 and 8 substitutions were found in two internal transcribed spacer regions of ribosomal DNA, ITS1 (288 sites) and ITS2 (251 sites) between these two genotypes, respectively, indicating that they are genetically different at the population level. These findings suggest that P. tapetis occurs worldwide and can associate with (and possibly infect) various types of bivalves. Further, a PCR method to specifically detect the P. tapetis cells in the host was also established.  相似文献

Amyloid beta (Aβ) oligomers associated with Alzheimer’s disease (AD) form Ca2+-permeable plasma membrane pores, leading to a disruption of the otherwise well-controlled intracellular calcium (Ca2+) homeostasis. The resultant up-regulation of intracellular Ca2+ concentration has detrimental implications for memory formation and cell survival. The gating kinetics and Ca2+ permeability of Aβ pores are not well understood. We have used computational modeling in conjunction with the ability of optical patch-clamping for massively parallel imaging of Ca2+ flux through thousands of pores in the cell membrane of Xenopus oocytes to elucidate the kinetic properties of Aβ pores. The fluorescence time-series data from individual pores were idealized and used to develop data-driven Markov chain models for the kinetics of the Aβ pore at different stages of its evolution. Our study provides the first demonstration of developing Markov chain models for ion channel gating that are driven by optical-patch clamp data with the advantage of experiments being performed under close to physiological conditions. Towards the end, we demonstrate the up-regulation of gating of various Ca2+ release channels due to Aβ pores and show that the extent and spatial range of such up-regulation increases as Aβ pores with low open probability and Ca2+ permeability transition into those with high open probability and Ca2+ permeability.  相似文献

Background

Buruli ulcer (BU) is a cutaneous infectious disease caused by Mycobacterium ulcerans. The exact mode of transmission remains elusive; yet, some studies identified environmental, socio-sanitary, and behavioral risk factors. The purpose of this study was to assess the association of such factors to contracting BU in Tiassalé, south Côte d’Ivoire.

Methodology

A case-control study was conducted in 2012. Cases were BU patients diagnosed according to clinical definition put forth by the World Health Organization, readily confirmed by IS2404 polymerase chain reaction (PCR) analysis prior to our study and recruited at one of the health centers of the district. Two controls were matched for each control, by age group (to the nearest 5 years), sex, and living community. Participants were interviewed after providing oral witnessed consent, assessing behavioral, environmental, and socio-sanitary factors.

Principal Findings

A total of 51 incident and prevalent cases and 102 controls were enrolled. Sex ratio (male:female) was 0.9. Median age was 25 years (range: 5–70 years). Regular contact with unprotected surface water (adjusted odds ratio (aOR) = 6.5; 95% confidence interval (CI) = 2.1–19.7) and absence of protective equipment during agricultural activities (aOR = 18.5, 95% CI = 5.2–66.7) were identified as the main factors associated with the risk of contracting BU. Etiologic fractions among exposed to both factors were 84.9% and 94.6%, respectively. Good knowledge about the risks that may result in BU (aOR = 0.3, 95% CI = 0.1–0.8) and perception about the disease causes (aOR = 0.1, 95% CI = 0.02–0.3) showed protection against BU with a respective preventive fraction of 70% and 90%.

Conclusions/Significance

Main risk factors identified in this study were the contact with unprotected water bodies through daily activities and the absence of protective equipment during agricultural activities. An effective strategy to reduce the incidence of BU should involve compliance with protective equipment during agricultural activities and avoidance of contact with surface water and community capacity building through training and sensitization.  相似文献

The name of Natal’ya Nikolaevna Traugott is closely associated with the history of human higher nervous activity studies in Russia. Destiny gifted to N.N. Traugott the opportunity to communicate with many outstanding scientists; she regularly attended the “Pavlov Wednesdays”, took part in V.M. Bechterev’s clinical discussions, visited L.S. Vygotskii’s lectures on the problems of children higher psychic functions development. For many years N.N. Traugott had a professional relationship with A.R. Luriya and his colleagues, at the suggestion of L.A. Orbeli she had headed the laboratory of pathology of higher nervous function from the beginning of 1956. The paramount importance for the development of new directions in evolutionary physiology had N.N. Traugott’s works on the principles of cortical dysfunction in different psychopathological conditions and her recommendations aimed at a strategy for individual corrections of speech in children.  相似文献
