Multiple lines of evidence support a central role of hormones in the etiology of breast cancer. In epidemiologic studies, considerable effort has focused on delineating the role of endogenous hormones in risk of breast cancer among postmenopausal women. Recently, substantial additional data has accrued from prospective studies where endogenous hormones are measured in study subjects prior to disease diagnosis. In this review, the epidemiologic evidence linking sex steroids—estrogens, testosterone, and progesterone, specifically—with subsequent risk of breast cancer in both premenopausal and postmenopausal women is summarized. Overall, a strong positive association between breast cancer risk and circulating levels of both estrogens and testosterone has now been well confirmed among postmenopausal women; women with hormone levels in the top 20% of the distribution (versus bottom 20%) have a two- to three-fold higher risk of breast cancer. Evidence among premenopausal women is more limited, though increased risk associated with higher levels of testosterone is consistent. However, both positive and null associations have been observed with estrogens and progesterone and clearly more evaluation is needed.  相似文献

Peroxidase and isoperoxidase patterns of tobacco have been comparatively analyzed in relation to parental origin and ploidy level (including hypohaploidy status) at different ages and physiological states. The interspecific hybrid N. sylvestris × N. tomentosiformis shows an additive pattern of parental peroxidases, which also resembles that of N. tabacum cv. Wisconsin and cv. Samsun. Enzyme activity (on dry weight and protein bases, decreases from the diploid to the hypohaploid forms of Wisconsin and Samsun with respective quantitative differences in the zymograms, when the plants are compared at the same chronological and vegetative stages. Abnormally vegetatively ageing hypohaploids recover all peroxidasic isoenzymes and reach an enzyme activity as high as those of haploids and diploids. Flowering hypohaploids, however, generally behave like haploids and diploids in changing their balance between acidic and basic peroxidases.  相似文献

Summary The abscisic acid contents of birch, maple and sycamore plants growing under long and short photoperiods were measured by gas-liquid chromatography. No increase was observed in the abscisic acid content of extracts when plants were transferred to dormancy-inducing conditions.A preliminary account of this work was presented at the XI International Botanical Congress, 1969.  相似文献

Summary The retinal rods, cones and epithelial pigment of most lower vertebrates display rhythmic photomechanical (retinomotor) migrations in response to changes in ambient lighting conditions. This study examines the extent of these migrations in the absence of the daily changes in illumination (constant darkness and constant light) in three species of teleosts. Salmo trutta, a crepuscularly active fish, showed two peaks of light adaptation occurring around dawn and dusk when kept in constant darkness. Tinca tinca, a nocturnal species, also showed an endogenous rhythm during extended periods of darkness, but, unlike Salmo trutta, it was light-adapted throughout what would normally have been day. At the maximal extent of migration under conditions of continual darkness, the pigment migrated 59% as much as it did during a normal light/dark cycle. Nannacara anomala, a tropical diurnally active species, showed a similar but more pronounced rhythm than Tinea tinea for all 3 days of experimental darkness, behaving essentially identically to fish exposed to a light/dark cycle. Nannacara anomala also displayed a weak rhythm when kept in constant light.It is concluded from these and previous results that the pattern of endogenous photomechanical movement depends both on the activity pattern of a species and on the constancy of the lighting conditions to which it has been exposed during its lifetime.  相似文献

An effect of age of quarry lakes on rotifer abundance and species composition has been evaluated. Rotifers occurred in all lakes under study. They were even found in the youngest (2 years of age) one, Rogonica 4, but both rotifer density and species richness were low there. Rotifer communities of much higher density and species diversity were noted in lakes only 4–6 years older. Lakes of over 30 years of age were strongly differentiated in rotifer numbers and species structure. In general, age of quarry lakes has an impact on rotifer communities only at the very beginning of the process of colonization. Several years later other factors become more important, e.g., depth or trophic state of the lakes.  相似文献

Summary Surgical experiments on Helianthus annuus and Phaseolus multiflorus seedlings involving the application of auxin and gibberellin to decapitated plants, suggested that internode extension growth occurs under the controlling influence of apically synthesised gibberellin rather than auxin. Studies were made of diffusible gibberellins from sunflower apical buds in relation to geotropic stimulation. Approximately ten times as much gibberellin was obtained from lower than from upper tissues of horizontal shoot-tips, whereas approximately equal quantities were obtained from the two halves of upright tips. Evidence was obtained suggesting both lateral transport of gibberellin in the young internode, and also enhanced gibberellin synthesis in buds maintained in a horizontal position during the collection of diffusible gibberellins into agar. The results are discussed in relation to current concepts of the role of auxin in geotropism.  相似文献

Comparative analyses of reproductive and vegetative tissues of the olive (Olea europaea L. cv. Manzanillo) for endogenous hormones, particularly inhibitors and gibberellin like substances, were made to study the relation between such hormones and thermoinduction of flowering. Qualitative and quantitative changes in gibberellin-like subtance(s) were observed in lateral buds (potential flower buds) but not in leaves or terminal buds (potential vegetative buds) sampled from orchard trees at intervals during the winter and spring. At least two types of gibberellin-like substances were found in extracts of lateral buds; their levels increased progressively during the low temperature induction period, reaching a maximum shortly before floral initiation. Two types of inhibitors were extracted from buds and leaves. A nonacidic type did not change during the induction stage but decreased considerably during the initiation period. An acidic inhibitor, which was identified as an abscisic acid-like substance, was present at a relatively lower level in lateral (flower) buds than in terminal (vegetative) buds during the induction period.  相似文献

Realizing the potential of Embryo transfer (ET) for rapid, cheap and widespread dissemination of genetic material, the risk of transmission disease through the embryos must be considered. The aim of this paper is to evaluate theses risks at each step of production, storage and transfer. The pathogen agent may potentially originate from the donor male (semen) or the donor female (oocytes, embryos) and finally from the environmental conditions. As the differences between in vivo and in vitro derived embryos have been well described, evaluation of the potential risks should be assessed separately for in vivo and in vitro produced embryos. Even if this paper insist on the diseases or diseases agents that are more questionable, it clearly appears that ET remains the more safety way to transfer gene, provided prevention measures are properly handled (use of donor that are specific pathogen free, washing of embryos, additional treatment...) and furthermore it can be easily seen as the best way to prevent some disease transmissions (TSEs, leukosis, foot-and-mouth disease...).  相似文献

In various vegetation types in Zimbabwe, the catches of Glossina pallidipes Austen and G. morsitans morsitans Westw. (Diptera: Glossinidae) at a target baited with odour (acetone, 1-octen-3-ol and two phenols) were positively correlated with catches of the same species at an unbaited net. No correlation existed between target catches and hand net catches of tsetse flies sitting on the vegetation. G. pallidipes females caught at a target and at an unbaited net were older than those caught from vegetation. Of the female G. pallidipes caught at the target, 46% were in the first 3 days of pregnancy. Of those caught at the unbaited net, significantly fewer, 21%, were in this stage. G. pallidipes males caught from vegetation contained more fat (3.07±0.333 mg) than those caught at the unbaited net (2.06±0.339 mg) or at the target (2.19±0.218 mg). It is inferred that target catches consisted predominantly of tsetse which were already in flight when they sensed the stimuli from the target, and that target catches were biased towards female G. pallidipes in the first 3 days of pregnancy.  相似文献

The process of hyperhydricity in tissue cultured plants of Aloe polyphylla is affected by both applied cytokinins (CKs) and the type of gelling agent used to solidify the medium. Shoots were grown on media with agar or gelrite and supplemented with different concentrations of N6-benzyladenine (BA) or zeatin (0, 5 and 15 μM). Endogenous CKs were measured in in vitro regenerants after an 8-weeks cycle to examine whether the hyperhydricity-inducing effect of exogenous CKs and gelling agents is associated with changes in the endogenous CK content. On media with agar a reduction in hyperhydricity occurred, while the gelrite treatment produced both normal and hyperhydric shoots (HS). The content of endogenous CKs, determined by HPLC-mass spectrometry, in the shoots grown on CK-free media comprised isopentenyladenine-, trans-zeatin- and cis-zeatin-type CKs. The application of exogenous CKs resulted in an increase in the CK content of the shoots. Following application of zeatin, dihydrozeatin-type CKs were also detected in the newly-formed shoots. Application of BA to the media led to a transition from isoprenoid CKs to aromatic CKs in the shoots. Shoots grown on gelrite media contained higher levels of endogenous CKs compared to those on agar media. Total CK content of HS was higher than that of normal shoots grown on the same medium. We suggest that the ability of exogenous CKs and gelrite to induce hyperhydricity in shoots of Aloe polyphylla is at least partially due to up-regulation of endogenous CK levels. However, hyperhydricity is a multifactor process in which different factors intervene.  相似文献

Changes in ornithine decarboxylase, putrescine N-methyltransferaseand N-methylputrescine oxidase activities in response to sometreatments were investigated using hydroponically grown tobaccoplants. Decapitation of shoots brought about marked elevationof the three enzyme activities, which reached their peaks 24hr after decapitation, then declined. An excellent correlationwas observed between the accumulation of nicotine and changesin the three root enzyme activities. Administration of IAA at2.5 to 5 µconcentration significandy increased these enzymeactivities in roots of decapitated plants but higher concentrationsof IAA prevented the rise in enzyme activities promoted by decapitation.Nicotine strongly inhibited the rise in enzyme activities inroots of decapitated plants in all cases. The results suggestthat these enzymes are under the control of a common regulatorysystem, in which auxin and nicotine are important components.Ornithine decarboxylase was present in all the plants examined,but putrescine N-methyltransferase and N-mediylputrescine oxidasewere detected only in the roots of tobacco, Datura and Atropaplants. 1Part XVI of the series "Phytochemical Studies on Tobacco Alkaloids". We have shown previously that transgene expression can be suppressed in hematopoietic cells using vectors that are responsive to microRNA (miRNA) regulation. Here we investigate the potential of this approach for more sophisticated control of transgene expression. Analysis of the relationship between miRNA expression levels and target mRNA suppression suggested that suppression depends on a threshold miRNA concentration. Using this information, we generated vectors that rapidly adjust transgene expression in response to changes in miRNA expression. These vectors sharply segregated transgene expression between closely related states of therapeutically relevant cells, including dendritic cells, hematopoietic and embryonic stem cells, and their progeny, allowing positive/negative selection according to the cells' differentiation state. Moreover, two miRNA target sites were combined to restrict transgene expression to a specific cell type in the liver. Notably, the vectors did not detectably perturb endogenous miRNA expression or regulation of natural targets. The properties of miRNA-regulated vectors should allow for safer and more effective therapeutic applications.  相似文献

Neurodegenerative diseases (NDs) are characterized by primary symptoms, such as cognitive or motor deficits. In addition, the presence of neuropsychiatric symptoms (NPS) in patients with ND is being increasingly acknowledged as an important disease feature. Yet, their neurobiological basis remains unclear and mostly centered on neurons while overlooking astrocytes, which are crucial regulators of neuronal function underlying complex behaviors. In this opinion article, we briefly review evidence for NPS in ND and discuss their experimental assessment in preclinical models. We then present recent studies showing that astrocyte-specific dysfunctions can lead to NPS. Because many astrocyte alterations are also observed in ND, we suggest that they might underlie ND-associated NPS. We argue that there is a need for dedicated preclinical studies assessing astrocyte-based therapeutic strategies targeting NPS in the context of ND.  相似文献
