In 1-, 2- to 3-, 7- to 8-, and 10-day old intact and spinal rat puppies, studies have been made of the effect of l-DOPA (100 mg/kg, intraperitoneally) on autogenic periodic motor activity in the gastrocnemius muscle. In 1- to 3-day old pups, strong stimulating effect was observed up to a prolonged continuous activity for 5-10 or even more minutes. This effect decreases with age. Traces of the inhibitory effect are observed at early stages in the form of total decrease of the activity. The inhibitory effect increases with age. In all spinal animals, at the background of a decreased activity, stimulating effect of l-DOPA was predominantly observed. Age peculiarities of the effect of l-DOPA are discussed in relation to ontogenetic development of catecholaminergic innervation in rats. It is suggested that stimulating effect of l-DOPA is associated with its action on the descending noradrenergic system, whereas the inhibitory one is mediated by the brain structures.  相似文献

At the 1st, 2nd and 3rd days after birth, subcutaneous injections of 6-oxydopamine in physiological solution containing 0.1% of ascorbic acid have been made to rat puppies in a dose 100 mg/kg per one injection. Within first three weeks, electromyographic studies were made on outbursts of autogenic periodic motor activity which is typical of animals at this age. It was shown that at the 3rd day of postnatal life, after 6-oxydopamine administration, total duration of motor excitation is significantly lower, whereas mean duration of the outbursts of continuous activity is lower even at the 7th day. Beginning from the 10th day, these parameters undergo opposite changes at the background of the increase in the number of outbursts and the decrease of intervals between the latter. The data obtained are discussed in relation to the role of catecholaminergic systems in regulation and realization of age dynamics of autogenic motor activity.  相似文献

In experiments on 3-day rat puppies, studies have been made of the effect of a stimulator of noradrenaline receptors--clonidine, and a stimulator of dopamine receptors--apomorphine on autogenic motor activity. It was shown that clonidine injections result in a significant increase of this activity, whereas apomorphine slightly decreases the latter. The data obtained in the present work together with those described earlier for l-DOPA effects, suggest that double regulation of autogenic activity is realized at early stages of ontogenesis. This regulation includes excitatory noradrenergic mechanisms and inhibitory influences which are mediated presumably by dopaminergic systems of the brain.  相似文献

Studies have been made on the role of the thyroid in the development of rats. In the first group of experiments, newborn rat received within a month mercazolyl which inhibits the activity of the thyroid; in animals of the second group, mercazolyl injections were combined with cold exposures which stimulated motor activity in animals. It was found that hypothyroid rats in both groups exhibit retardation of growth as compared to normal animals. However, retardation is less significant in animals of the second group, as it is indicated by smaller changes in the protein content and total mass of skeletal muscles.  相似文献

In experiments on 5-30-day rat puppies, under slight urethane narcosis, studies have been made on the late prolonged discharges in extensor muscles evoked by stimulation of contralateral tibial nerve before and after injection of DOPA. Within the first 16 days, the drug completely abolished or significantly reduced late discharges, increasing the spontaneous motor activity. Beginning from the 16th day, periods of potentiation of late discharges were observed. The data obtained are discussed in relation to the development of mechanisms of control of the activity of spinal generators which are responsible for late discharges and spontaneous motor activity.  相似文献

In experiments on 2-3-, 7-8-, 10- and 16-day old rat pups, basic age differences have been observed in the effect of a precursor of catecholamine mediator, i.e. L-DOPA on the activity on the spinal and brain mechanisms of autogenic periodic motor excitation. Strong stimulation of the spinal motor rhythm was observed during the first week. At this period, supraspinal rhythm becomes even less evident. On further development, the effect is an opposite one. In 10- and 16-day rat pups, at the background of total inhibition of the spontaneous activity, administration of L-DOPA significantly increases the activity of supraspinal generator of the motor rhythm. The data obtained reveal ontogenetic changes in regulatory mechanisms of autogenic motor activity.  相似文献

In conscious 2-6-day rat puppies, studies have been made on the bioelectrical activity in the visual and sensorimotor cortex. ECG in newborn rat puppies exhibits characteristic intermittence of complexes of the electrical activity with intervals of partial or almost complete absence of the activity in a minute scale. This phenomenon reflects the ancient property of immature nervous system, i.e. a capacity to autogenic periodic excitation. The structure of these complexes may be different, since it reflects the condition of animals at the given moment, the degree of maturation of elements involved in realization of the bioelectrical activity and interrelationship with other parts of the brain. With respect to amplitude-frequency parameters, age dynamics and the relationship to the spontaneous motor activity, four distinct types of complexes were revealed in the ECG of rat puppies during the first week of their postnatal life.  相似文献

The metabolic clearance rate (MCR) of immunoreactive rat prolactin (rPRL) was determined in urethane-anaesthetized 14 to 16-day-old rat pups using the constant infusion to equilibrium method. Two different doses of rPRL were used, 329 and 472 ng/min each at a volume of 0.0025 ml/min for 30 min. The MCR was 0.216 ml/min following infusion of 329 ng PRL/min dose and 0.337 ml/min following infusion of the 472-ng/min dose. When calculated on the basis of metabolic body weight the MCR of rPRL in rat pups was comparable to that of adult rats.  相似文献

Study of parameters of the cardiac, respiratory, and motor activity (MA) was carried out on newborn rat pups for the first day after birth (P0) and at the 14th day of postnatal development (P14) after change of the activity levels of dopaminergic and noradrenergic systems. To provide an excessive level of catecholamines, the animals were administered individually with L-DOPA (25–100 mg/kg) and with the indirect adrenomimetic isoamine (3 and 10 mg/kg). Additionally there were studied effects of L-DOPA and isoamine after blockade of D1 and D2 dopamine receptors (antagonists SCH-23390 and sulpirid). The L-DOPA administration produced a dose-dependent MA enhancement with its subsequent possible conversion into the continuous generalized activity. At P0 the release of monoamines was accompanied by development of weak bradycardia. There was noted a tendency for acceleration of respiration at administration of the low dose both of L-DOPA and of isoamine and for its deceleration at high doses. At P14 the L-DOPA administration was accompanied by deceleration of the heart rate (HR) by 8% and by acceleration of respiration rate (RR) by 26%. The isoamine administration produced an insignificant decrease of HR and an increase of RR by 8% at the low dose and by 21% at the high dose of the agent. At the blockade of D1 receptors, RR remained close to the background values, while at the blockade of D2 receptors it decreased insignificantly. Blockade of D1 and D2 receptors did not cause significant HR changes. Analysis of the HR variability has shown that both after L-DOPA administration and at blockade of dopamine receptors no unidirectional reaction was observed: in 80% of rat pups the portion of nerve mechanisms of HR regulation increased, while in the rest-of sympathetic and humoral factors at a decrease of parasympathetic effects. In all rat pups the isoamine administration was accompanied by a shift of the specter power into the higher frequency area; in 60% of the animals there were enhanced sympathetic effects. At P14 in rat pups after administration both of L-DOPA and of isoamine, the sympathetic nervous effects were predominant. Thus, at P0 both at release of endogenous catecholamines and at their excessive concentration in rat pups there occurs a qualitative change of character of the catecholaminergic effects on functional activity of excitable structures, particularly of those connected with regulation of respiration.  相似文献

Previous studies in our laboratory have shown that even short-term separation of preweanling rat pups from the mother produces adverse effects in the pup. These include alterations in ornithine decarboxylase activity and in the secretion of growth hormone and corticosterone. The present study demonstrates that while intermittent heavy stroking effectively reverses or prevents all the changes associated with maternal deprivation neither kinesthetic nor vestibular stimulation affects these responses. The results verify earlier findings from this laboratory indicating that tactile interactions between rat pups and their mother modulate pup physiology and provide experimental support for the hypothesized role of tactile stimuli on early infant development.  相似文献

The development of nasal chemosensitivity was measured in rat pups that received exogenous l-thyroxine or a vehicle injection on postnatal Days 1–4. Unconditioned respiratory responses were used to measure detection of the olfactant, amyl acetate, which was presented via a dilution olfactometer in an ascending series of log-unit concentrations. Thyroxine-treated pups exhibited greater sensitivity than control subjects on Day 6. Sensitivity appeared to converge in hormone-treated and control pups by Day 8 and continued to improve in both groups until the last day of testing (Day 16).  相似文献

In the United State, 20% of pregnant women smoke. One of the most consistent adverse outcomes is reduced birth weight in the off-spring. Animal studies using chronic nicotine, the major psychoactive tobacco ingredient, have shown conflicting results, questioning the role of nicotine in growth retardation. To evaluate the direct effects of nicotine during a period equivalent to the human third trimester, we developed an oral gastric intubation model using neonatal rat pups. Nicotine (6 mg/kg/day) was dissolve in milk-formula and delivered during three feedings daily from postnatal day (P)1 to P7. Nicotine immediately and significantly [P<0.05] decreased weight gain per day (WGD) by 13.5% (+/-) 1 day after onset of treatment in both genders and throughout the treatment period. This resulted in significantly lower body weight at P4 and P5 in male and female pups, respectively. After nicotine withdrawal, WGD returned to control level within 1 day, whereas total body weight recovered by P18. There were no long-term consequences on body weight or growth pattern in either gender. The nicotinic acetylcholine receptor (nAChR) antagonist dihydro-beta-erythroidine (DHbetaE) reversed nicotine's effects on WGD suggesting an involvement of heteromeric alpha4beta2, whereas methyllycaconitine (MLA) an antagonist for the homomeric alpha7-type receptor was ineffective. The immediate decrease of growth in neonatal pups suggests that nicotine's effect on birth weight results from direct anorexic rather then indirect effects due to placental dysfunction or increased fetal hypoxia. The postnatal oral gastric intubation model seems to accurately reflect the direct effects of nicotine in neonates.  相似文献

Catecholamine level in the blood, heart and adrenal tissues, and heart sensitivity to catecholamines were determined in 3-week-old rats after chemical sympathectomy. This paper reviews the ontogeny of thermoregulation and diurnal rhythmicity in rats. Additionally, original data are presented that indicate the emergence of an endogenous circadian core temperature rhythm during the first postnatal week. Despite neurological immaturity, newborn rats display autonomic and behavioral thermoregulatory responses within 24 h of birth. Their "biological clock" is already running before birth. The thermal environment of pups changes cyclically owing to diurnal variations in maternal behavior, but the core temperatures of 1-week-old pups huddling in the absence of the dam also show marked diurnal fluctuations. Five- to 8-day-old lean Zucker rat pups artificially reared in the absence of 24-h cycles of ambient temperature and food intake show diurnal changes in core temperature similar to those in huddling mother-reared pups. Diurnal core temperature changes, evident only when regulatory effectors are not overwhelmed, are one of the first self-maintained diurnal rhythms to appear. Because thermoregulation and circadian rhythmicity both appear before maturation of the neural networks believed to be critical for their control in adult animals, studying the immature rat might increase our understanding of the control of these processes in the more complex mature central nervous system.  相似文献

With the aid of a pair-feeding procedure, two groups of pregnant Long-Evans rats were fed a liquid diet containing 35% or 0% ethanol-derived calories during days 6-20 of gestation. A third group was allowed free access to standard lab chow and water throughout pregnancy. At 6-7 or 9-10 days of age, suckling performance by male and female offspring representing the three prenatal treatment groups was examined. The test stimulus was a 6-10-day postparturient, anesthetized dam in which milk letdown was prevented. Compared to both pair-fed and lab chow controls, alcohol-exposed animals exerted a lower maximum suckling pressure, spent less time suckling during the test session, and displayed an altered suckling pattern. These data are consistent with existing clinical and experimental evidence documenting sucking deficits following prenatal alcohol exposure and are discussed in terms of prenatal alcohol-induced CNS impairment.  相似文献
