We studied the role of spinal afferent pathways in the hyperpnea of electrically induced muscle contractions (ExE). The ventilatory (VE) and arterial CO2 partial pressure (PaCO2) responses were measured at rest and during two levels of ExE in awake human paraplegic subjects with clinically complete lesions of the spinal cord (range T4-T11). We hypothesized that if peripheral neural drive is critical to a normal ventilatory response, then ExE in the absence of intact pathways should cause a lower ventilatory response resulting in hypercapnia at the onset of ExE. ExE was induced by stimulation of the quadriceps and hamstring muscles that approximately doubled the resting level of CO2 production (VCO2). PaCO2 during work transitions and in the latter stages of ExE did not differ significantly from that at rest. Arterial pH progressively declined over time during ExE (P less than 0.01) as a result of increased lactate concentration (P less than 0.01). The linear relationship between VE and VCO2 was similar to that found for normal human subjects during ExE (P = 0.73). These data suggest that VE and presumably alveolar ventilation (VA) can be appropriately matched to VCO2 during low-intensity muscle contractions of the lower extremities in the absence of intact spinal afferent pathways. Moreover, since it is unlikely that postulated "central command" mechanisms were initiated during ExE in these paraplegic subjects, the data provide support for our previous conclusion that central command is not obligatory for matching VA to VCO2 (J. Spinal muscular atrophy (SMA) is caused by a drastic reduction in the ubiquitously expressed SMN protein, which is critical for the correct assembly of the snRNP complexes required for RNA splicing. However, it is unclear why loss of SMN and altered snRNP assembly only seem to affect motor neurons. Reporting in this issue, Zhang et al. (2008) challenge prior assumptions about the housekeeping function of SMN and demonstrate that loss of SMN leads to highly tissue-specific effects on splicing.  相似文献

MN (motor neuron) death in amyotrophic lateral sclerosis may be mediated by glutamatergic excitotoxicity. Previously, our group showed that the microdialysis perfusion of AMPA (α-amino-3-hydroxy-5-methyl-4-isoxazole propionate) in the rat lumbar spinal cord induced MN death and permanent paralysis within 12 h after the experiment. Here, we studied the involvement of energy metabolic deficiencies and of oxidative stress in this MN degeneration, by testing the neuroprotective effect of various energy metabolic substrates and antioxidants. Pyruvate, lactate, β-hydroxybutyrate, α-ketobutyrate and creatine reduced MN loss by 50–65%, preserved motor function and completely prevented the paralysis. Ascorbate, glutathione and glutathione ethyl ester weakly protected against motor deficits and reduced MN death by only 30–40%. Reactive oxygen species formation and 3-nitrotyrosine immunoreactivity were studied 1.5–2 h after AMPA perfusion, during the initial MN degenerating process, and no changes were observed. We conclude that mitochondrial energy deficiency plays a crucial role in this excitotoxic spinal MN degeneration, whereas oxidative stress seems a less relevant mechanism. Interestingly, we observed a clear correlation between the alterations of motor function and the number of damaged MNs, suggesting that there is a threshold of about 50% in the number of healthy MNs necessary to preserve motor function.  相似文献

Summary It has been known for many years that when a wing disc ofDrosophila is bisected, and the fragments cultured in adult females, regulation occurs and either a complete disc is regenerated or the fragment is duplicated. We have investigated how this regeneration process occurs. To establish which cells contribute to the regenerate, and thus determine if regeneration is the result of epimorphic regulation, fragments of discs, after culture in an adult for one to five days, were exposed to3H-thymidine to label replicating cells. Imaginal discs, both whole and as regenerating fragments, undergo some DNA replication which is distributed throughout the disc, but cut discs frequently show clusters of labelled cells around the wound, indicating that regeneration is probably epimorphic.  相似文献

The effects of a single and repetitive administration of m-cholinoblocker scopolamine (Sc) to male rats on retention of step-through passive avoidance (PA) or active avoidance (AA) in a shuttle-box were compared. In case of PA Sc (1 mg/kg) was injected i.p. only 30 min before training, only 30 min before testing, or both before training and before testing. In case of AA Sc (0.5 mg/kg/day) was injected i.p. only 15 min before each training session or both before training and before testing (44 days after achievement of learning criterion). The PA and AA retention were impaired only in the experiments, where the drug was administered before training, but did not differ from control, when Sc was injected twice. The Sc-induced amnesia (like many other cases of memory deficits) is suggested to be a manifestation of state-dependent learning. Similarity between the brain state during memory consolidation and during the retention test is necessary for recollection.  相似文献

Dyneins are the largest and most complex of the three classes of linear motor proteins in eukaryotic cells. The mass of the dynein motor domain is about ten times that of the other microtubule motor, kinesin. Dynein's homology with the AAA+ superfamily of mechanoenzymes distinguishes it from both kinesin and myosin, which share a common fold and ancestry as members of the G-protein superfamily. In contrast to the other motor proteins, little is known about the mechanism of dynein; its three-dimensional structure is unknown even at low resolution. Recent two-dimensional images from electron microscopy have revealed new details of its structure and how this changes to produce movement. These and the recently solved crystal structure of another AAA+ protein, ClpB, offer tantalising hints about dynein's mechanism, suggesting it may act like a molecular winch.  相似文献

The first estimation of the heterozygous carrier rates for the SMN1 gene deletions and SMN2 gene duplications in populations of Russia has been performed. The numbers of SMN gene copies have been deter-mined in samples from Chuvash and Udmurt populations, as well the population of the Moscow region, by means of multiplex ligation-dependent probe amplification. The heterozygous carrier rates for the CMA gene were 2.7% (1: 37 people), 2.8% (1: 36 people), and 2.8% (1: 36 people) in Chuvashes, Udmurts, and residents of the Moscow region, respectively. The SMN2 duplication frequencies have been determined in the studied groups. It is 1.5, 4, and 2.5% in Chuvashes, Udmurts, and residents of the Moscow region, respectively. The high SMN2 duplication frequency in Udmurts may explain why the SMN1 heterozygous carriage frequency in this population was overestimated in earlier PCR-RFLP analyses.  相似文献

Many authors have referred to the important role of vegetation in the consolidation of salt marsh sediments, but experiments previously carried out by us have shown results that do not always agree with these statements. In other words, the type of salt marsh surface coverage is not the main factor that contributes to the consolidation of sediments. To test this hypothesis different Portuguese salt marsh stations (species/unvegetated areas) from two sites, Tagus estuary (Corroios and Pancas) and Ria de Aveiro (Barra and Verdemilho), were compared to evaluate their influence on suspended matter deposition on the salt marsh surface. A short-term sedimentation study was performed within stands of Spartina maritima, Halimione portulacoides, Sarcocornia perennis subsp. perennis and unvegetated areas, by analysing the deposition of sediment material on nylon filters anchored to the marsh surface. Numerical results obtained from hydrodynamic models coupled to a Lagrangean module implemented for the Ria de Aveiro and the Tagus Estuary, namely the root-mean square velocity (V rms) and residual velocity of tides, were also used. Average sedimentation rates (mean value between the different surface cover in a salt marsh) showed a seasonal trend more or less defined but with significantly different values between sites and salt marshes. Sedimentation rates varied between marshes: there are significant differences between Pancas and the other three marshes, but only significant differences in sedimentation rates between Spartina and Sarcocornia. Despite the important role of vegetation in the consolidation of salt marsh sediments, our results suggest that, the position of stations and related abiotic conditions in the salt marshes are determining factors of variation to take into account in the studies related with the stabilization and survival of salt marshes facing sea level rise. The epigean amphipod Gammarus lacustris possessed a rate of oxygen uptake (MO2) three times greater that of the hypogean amphipod Stygobromus sp. While much of the difference between these two species could be attributed to different feeding regimes, the hypogean amphipod still exhibited a lower rate of metabolism than the epigean species. This suggests that there can be both a genetic and an environmental (reduced food availability and/or hypoxia) component to the reduced metabolism characteristic of many hypogean animals. The results of this study have re-emphasised the pitfall of making species comparisons without knowledge of the extent of physiological variation within a species. The olfactory system is a fascinating and beguiling sensory system: olfactory sensory neurons detect odors underlying behaviors essential for mate choice, food selection, and escape from predators, among others. These sensory neurons are unique in that they have dendrites contacting the outside world, yet their first synapse lies in the central nervous system. The information entering the central nervous system is used to create odor memories that play a profound role in recognition of individuals, places, and appropriate foods. Here, the structure of the olfactory epithelium is given as an overview to discuss the origin of the olfactory placode, the plasticity of the olfactory sensory neurons, and finally the origins of the gonadotropin‐releasing hormone neuroendocrine cells. For the purposes of this review, the development of the peripheral sensory system will be analyzed, incorporating recently published studies highlighting the potential novelties in development mechanisms. Specifically, an emerging model where the olfactory epithelium and olfactory bulb develop simultaneously from a continuous neurectoderm patterned at the end of gastrulation, and the multiple origins of the gonadotropin‐releasing hormone neuroendocrine cells associated with the olfactory sensory system development will be presented. Advances in the understanding of the basic mechanisms underlying olfactory sensory system development allows for a more thorough understanding of the potential causes of human disease. Theories of beauty were evaluated by requiring subjects to “evolve” a beautiful female face using a Genetic Algorithm. In this procedure, a computer program generated a small population of faces (first generation of phenotypes) from a set of random binary strings (genotypes). Genotypes specified the shapes and soft tissue anthropometrics of facial features. Each of the first generation of faces was rated by a subject (relative fitness measure) for beauty. The fittest genotypes then bred in proportion to their fitness, with crossover and mutation of the binary strings, to produce offspring which were again rated by the subject. This process continued until the most beautiful face, for that subject, was evolved. Forty Caucasian subjects (20 M, 20 F) were required to evolve their idealized beautiful female face using this procedure. The features and soft tissue anthropometrics of their final composites were compared to population norms. Also, the final composites, and different faces generated from the same data base, were rated for beauty by independent judges. The results support the conclusion that the concept of facial beauty is the result of sexual selection, and a beautiful female face has features and proportions indicative on high fertility.  相似文献
