A protocol has been standardized for establishment and characterization of cell suspension cultures of Stevia rebaudiana in shake flasks, as a strategy to obtain an in vitro stevioside producing cell line. The effect of growth regulators, inoculum density and various concentrations of macro salts have been analyzed, to optimize the biomass growth. Dynamics of stevioside production has been investigated with culture growth in liquid suspensions. The callus used for this purpose was obtained from leaves of 15-day-old in vitro propagated plantlets, on MS medium fortified with benzyl aminopurine (8.9 μM) and naphthalene acetic acid (10.7 μM). The optimal conditions for biomass growth in suspension cultures were found to be 10 g l?1 of inoculum density on fresh weight basis in full strength MS liquid basal medium of initial pH 5.8, augmented with 2,4-dichlorophenoxy acetic acid (0.27 μM), benzyl aminopurine (0.27 μM) and ascorbic acid (0.06 μM), 1.0× NH4NO3 (24.7 mM), 3.0× KNO3 (56.4 mM), 3.0× MgSO4 (4.5 mM) and 3.0× KH2PO4 (3.75 mM), in 150 ml Erlenmeyer flask with 50 ml media and incubated in dark at 110 rpm. The growth kinetics of the cell suspension culture has shown a maximum specific cell growth rate of 3.26 day?1, doubling time of 26.35 h and cell viability of 75 %, respectively. Stevioside content in cell suspension was high during exponential growth phase and decreased subsequently at the stationary phase. The results of present study are useful to scale-up process and augment the S. rebaudiana biological research.  相似文献

Stevia rebaudiana Bertoni is a unique medicinal plant which is mostly utilized as a sugar substitute for diabetic patients. In this research, regenerated plantlets of stevia in tissue culture is transferred to pots in greenhouse and inoculated with plant-growth-promoting rhizobacteria (PGPRs) (Bacillus polymixa, Pseudomonas putida, and Azotobacter chroococcum) and arbuscular mycorrhizal fungus (AMF) (Glomus intraradices). The results showed that in comparison to control, inoculation with a single microorganism, significantly increased root and shoot biomass as well as stevioside, chlorophyll, and NPK content in plants. However, such increased effects have been found to be further enhanced significantly due to dual compatible mixtures of inoculants resulting from their strong synergistic relationships among themselves. All growth parameters recorded the highest in 60-days-old plants in the treatment of Glomus+Azotobacter and followed with Glomus+Bacillus and Azotobacter+Pseudomonas treatments, respectively. Furthermore, high-performance liquid chromatography (HPLC) chromatograms revealed that the highest stevioside content have been produced in same treatments. Triple treatments had less positive effects compared to dual inoculations. Probably competence between microorganisms in triple inoculations has reduced their efficiency. Thus, suitable combination of mycorrhizal fungi and PGPR as biotic elicitors can enhance growth and stevioside content in tissue culture-regenerated plantlets of stevia.  相似文献

A very rapid and efficient regeneration method has been established using mature expiants ofStevia rebaudiana Bert. Adventitious shoots were induced from nodal expiants of field-grown plants on four basal media supplemented with various combinations of auxins and cytokinins. The best performance (23.4 ± 2.1 shoots per expiant) was obtained on a Murashige and Skoog (MS) medium containing 2 mg L-1 IAA and 0.5 mg L-1 kinetin. Roots were then produced when thesein vitro- regenerated shoots were transferred to an MS medium supplemented with 3% sucrose and 2 mg L-1 IBA. When acclimatized to soil, the rooted plants had a 98.4% survival rate. Following transplantation in the field, stevioside contents were similar between the regenerated plants (10.68 mg g-1 dry weight) and the mother plants (12.01 mg g-1 dw).  相似文献

Stevia rebaudiana (S. rebaudiana) is the most important therapeutic plant species and has been accepted as such worldwide. It has a tendency to accumulate steviol glycosides, which are 300 times sweeter than marketable sugar. Recently, diabetic patients commonly use this plant as a sugar substitute for sweet taste. In the present study, the effects of different spectral lights were investigated on biomass accumulation and production of secondary metabolites in adventitious root cultures of S. rebaudiana. For callus development, leaf explants were excised from seed-derived plantlets and inoculated on a Murashige and Skoog (MS) medium containing the combination of 2,4-dichlorophenoxy acetic acid (2, 4-D, 2.0 mg/l) and 6-benzyladenine (BA, 2.0 mg/l), while 0.5 mg/l naphthalene acetic acid (NAA) was used for adventitious root culture. Adventitious root cultures were exposed to different spectral lights (blue, green, violet, red and yellow) for a 30-day period. White light was used as control. The growth kinetics was studied for 30 days with 3-day intervals. In this study, the violet light showed the maximum accumulation of fresh biomass (2.495 g/flask) as compared to control (1.63 g/flask), while red light showed growth inhibition (1.025 g/flask) as compared to control. The blue light enhanced the highest accumulation of phenolic content (TPC; 6.56 mg GAE/g DW), total phenolic production (TPP; 101 mg/flask) as compared to control (5.44 mg GAE/g DW; 82.2 mg GAE/g DW), and exhibited a strong correlation with dry biomass. Blue light also improved the accumulation of total flavonoid content (TFC; 4.33 mg RE/g DW) and total flavonoid production (TFP; 65 mg/flask) as compared to control. The violet light showed the highest DPPH inhibition (79.72%), while the lowest antioxidant activity was observed for control roots (73.81%). Hence, we concluded that the application of spectral lights is an auspicious strategy for the enhancement of the required antioxidant secondary metabolites in adventitious root cultures of S. rebaudiana and of other medicinal plants.  相似文献

采用盆栽法,以甜菊( Stevia rebaudiana Bertoni)品种‘中山4号'(‘Zhongshan No.4')当年生扦插苗为研究对象,研究不同形态氮肥(硫酸铵、硝酸钠和尿素)及不同施氮量(纯氮)、施磷量( P2 O5)和施钾量( K2 O)对幼苗生长及糖苷含量和单株积累量的影响。结果显示:随氮肥、磷肥和钾肥施用量的提高,甜菊幼苗的株高、茎粗、叶长、叶宽、单株叶干质量和单株茎干质量均呈先升后降的变化趋势,且总体上与对照无显著差异,仅施磷量300 mg·kg-1处理组的叶长显著高于对照;根据施肥量与单株叶干质量的回归方程,确定硫酸铵、硝酸钠、尿素、磷肥和钾肥的施用量分别为64.87、660.21、735.84、211.54和775.92 mg·kg-1时,幼苗单株叶干质量最高。在硫酸铵处理组中,300 mg·kg-1处理组甜菊叶片中的莱鲍迪苷A( R-A)含量及甜菊苷( St)、R-A和总苷的单株积累量以及600 mg·kg-1处理组的St单株积累量高于对照,多数处理组的St、R-A和总苷含量及单株积累量均低于对照;在硝酸钠处理组中,1200 mg·kg-1处理组的R-A和总苷含量、600和900 mg·kg-1处理组的St单株积累量以及300~900 mg·kg-1处理组的R-A和总苷单株积累量高于对照,其他处理组的St、R-A和总苷含量及单株积累量均低于对照;在尿素处理组中,1500 mg·kg-1处理组的R-A和总苷的含量和单株积累量以及600和900 mg·kg-1处理组的R-A和总苷单株积累量高于对照,其他处理组的St、R-A和总苷含量及单株积累量均低于对照;各施氮处理组中,仅1500 mg·kg-1处理组的R-A含量与对照差异显著,其他指标均与对照无显著差异。在施磷处理组中,100 mg·kg-1处理组的R-A含量以及100和200 mg·kg-1处理组的St、R-A和总苷的单株积累量高于对照,多数处理组的St、R-A和总苷含量及单株积累量低于对照且与对照均无显著差异。在施钾处理组中,各处理组的St、R-A和总苷含量及单株积累量均高于对照,其中仅900 mg·kg-1处理组的St、R-A和总苷含量与对照显著差异。各施肥处理组的St含量占总苷含量的百分率均低于对照、R-A含量占总苷含量的百分率均高于对照,且总体上与对照无显著差异。经过综合分析,建议在甜菊生育期内的施肥量为纯氮600~900 mg·kg-1、P2 O5200~300 mg·kg-1和K2 O 600~900 mg·kg-1,其中氮肥以尿素为宜。  相似文献

The amount of free glucose and sucrose within the leaves of Stevia rebaudiana has been evaluated under different photoperiods and related to steviol glycoside (SVgly) accumulation. The diurnal dynamics of glucose and sucrose in leaves, internodes, apices, and roots revealed considerable fluctuations. Within leaves, glucose and sucrose content decreased up to three-fold during the night whereas SVgly amounts did not show any significant diurnal fluctuations. Ontogenetic variations in glucose content was markedly different under long-day (LD) and short-day (SD) regimes. Under LD, glucose content increased two-fold after the onset of flower bud formation, whereas under SD, a stagnation or small decrease was observed. This increase under LD was already noticed in the upper sink leaves at the end of the vegetative stage, possibly acting as a metabolic trigger for the phase transition to reproductive development. Under SD, glucose content appeared more controlled resulting in a better linear relationship with dry matter and SVgly content. The increased sink demands of the transitional apex led to a rapid decline in leaf sucrose which showed little or no correlation with SVgly amounts. From our results, it becomes clear that the correlation between glucose or sucrose as substrates and SVglys as products is very dynamic and is significantly influenced by day-length and ontogeny.  相似文献

The aim of this study was to determine whether steviol glycoside accumulation is under phytochrome control. The results indicate that Stevia rebaudiana Bertoni plants grown under short-day conditions showed precocious flowering and stagnation of steviol glycoside accumulation. Long night interruption by red LED light stimulated and sustained the vegetative growth as well as the accumulation of steviol glycosides in the leaves. After 7 weeks of treatment, steviol glycoside content was about two-fold higher in LED-treated plants than in the short-day control group. The effects of red LED light were measured both in a greenhouse and in a phytotron, irrespective of cultivar-specific differences. Therefore, it can be concluded that a mid-night interruption by red LED light during short photoperiods provides an easy and inexpensive method to increase vegetative leaf biomass production with an increased steviol glycoside yield.  相似文献

Although the effectiveness of topolins in plant tissue culture systems has recently been highlighted, there is a dearth of information on their interactions with auxins in relation to shoot organogenesis and secondary metabolite production. The current study evaluated the role of topolins singly or in combination with an auxin in comparison to 6-benzyladenine (BA) on shoot proliferation and secondary metabolite production of Huernia hystrix, a medicinal and ornamental stem-succulent of the endemic flora of southern Africa. Meta-topolin (mT) was more effective in improving shoot proliferation and phenolic production compared to BA. In general, the exogenous addition of α-naphthalene acetic acid (NAA) significantly increased shoot proliferation. The highest number of regenerated shoots (12.2 ± 0.98 shoots per explant) was recorded with medium containing 20 μM mT supplemented with 10 μM NAA and was three-times higher when compared to the treatments with cytokinin only. This suggests a synergistic interaction of auxin with cytokinin. On the other hand, supplementation with low NAA concentrations resulted in reduced in vitro flavonoid production in most cases, when compared to treatments with cytokinin only. Moreover, differences in cytokinin concentrations (even when used in combination with NAA equimolar concentrations) significantly affected secondary metabolite production in some cases. The current findings highlighted the differential effects of auxin-cytokinin interactions on shoot proliferation and the production of secondary metabolites in H. hystrix.  相似文献

This work investigated the agronomic, physiological and biochemical response of Stevia rebaudiana Bertoni grown under different nitrogen (N) rates. A pot trial in open air conditions was set up in 2012 with the aim to evaluate the effect of four N rates on the biometric and productive characteristics, steviol glycoside (SG) content as well as on leaf gas exchanges, chlorophyll fluorescence, photosynthetic pigments, Rubisco activity and N use efficiency. N deficiency caused a decrease in leaf N content, chlorophylls and photosynthetic CO2 assimilation, resulting in a lower dry matter accumulation as well as in reduced SG production. The application of 150 kg N ha? 1 seems to be the most effective treatment to improve rebaudioside A (Reb A) content, Reb A/stevioside ratio, photosynthetic CO2 assimilation, stomatal conductance, N use efficiency, ribulose-1,5-bis-phosphate carboxylase/oxygenase (Rubisco) and PSII efficiency. The results demonstrate that by using an appropriate N rate it is possible to modulate the SG biosynthesis, with a significant increase in the Reb A content and in the ratio between Reb A and stevioside. This finding is of great relevance in order to obtain a raw material designed to meet consumer needs and bio-industry requirements for high-quality, Reb A content, and safe and environmentally friendly products.  相似文献

Medicinal plants contain a plethora of secondary metabolites, most of which are bioactive in nature. The role of a popular plant growth retardant CCC has been investigated to explore its impact on secondary metabolite production, particularly phenols and flavonoids from in vitro grown Stevia rebaudiana. CCC stimulated the production of total phenols and flavonoids in calli and leaves. Moreover, this elevated level of phenols and flavonoids was correlated with the antioxidant potential of the tissue extracts. Methanolic extracts from CCC-treated calli and leaves showed significant increment in antioxidant activity as determined by standard DPPH, ABTS, and hydroxyl radical scavenging assays. No significant antiproliferative effect of methanolic extracts from different tissue was noticed against THP-1 monocyte (ATCC-TIB202), Hela cell (ATCC-CCL2) lines endorses the issue of clinical safety of the extracts.  相似文献

This investigation was carried on to find out the changes occurred in Stevia rebaudiana in response to paclobutrazol (PBZ; 0–4 mg L?1) treatment and drought stress. Polyethylene glycol (PEG; 0–6 % w/v) was used to stimulate drought stress. Drought stress reduced fresh and dry weight, water content, chlorophylls, carotenoids, anthocyanins, water soluble carbohydrates, reducing sugar and proline amounts. Electrolyte leakage, MDA, α-tocopherol and glycine betaine contents increased in drought-stressed plants. The activity of P5CS and PDH enzymes and protein content showed no significant changes under drought stress. PBZ (with or without PEG) treatments decreased fresh and dry weight and water content. In PBZ-treated plants, less pigments was damaged by drought stress. PBZ treatment reduced the negative effect of drought stress on lipid peroxidation which resulted in lower electrolyte leakage and MDA content, compared to the same PEG level without PBZ. PBZ (with or without PEG) treatments increased glycine betaine, α-tocopherol, proline and protein contents. The amount of water soluble carbohydrates, reducing sugar and activity of P5CS and PDH were not affected by PBZ treatments. SDS-PAGE analysis revealed that drought stress increased a 25 kD protein with a critical function in plant development under stresses. According to the results, PEG provoked a severe drought stress in S. rebaudiana that could partly be restored by PBZ treatment.  相似文献

The ability to synthesize Indole-3-acetic acid (IAA) is widely associated with the plant growth promoting rhizobacteria (PGPR). The present work deals with isolation and characterization of such bacteria from the rhizosphere of medicinal plant Stevia rebaudiana and optimization of IAA production from its isolates. The optimization of IAA production was carried out at different pH and temperature with varied carbon and nitrogen sources of culture media. Out of different isolates obtained, three of them were screened as efficient PGPRs on the basis of different plant growth promoting attributes. Isolates CA1001 and CA2004 showed better production of IAA at pH 9 (91.7?µg?ml?1) and at temperature 37?°C (81.7?µg?ml?1). Dextrose (1%) was found to be the best carbon source for isolate CA1001 with 104?µg?ml?1 IAA production. Isolate CA 2004 showed best production of IAA 36?µg?ml?1 and 34?µg?ml?1 at 1.5% and 1% Beef extract as nitrogen source respectively. Isolate CA 1001 showed 32?µg?ml?1 IAA production at 0.5% nicotinic acid concentration. From the current study, CA1001 and CA2004 emerged as noble alternatives for IAA production further which also resulted in root and shoot biomass generation in crop plants, hence can be further used as bio-inoculants for plant growth promotion.  相似文献

Two greenhouse experiments were conducted to study the effect of two plant growth retardants, Chlorocholine chloride (CCC) and Paclobutrazol (PBZ), on growth, Steviol glycosides (SVglys) content and antioxidant capacity in Stevia (Stevia rebaudiana Bertoni). Five concentrations of CCC (0, 250, 500, 750 and 1,000 ppm) and PBZ (0, 6, 12, 18 and 24 ppm) with three replications were applied to Stevia plants as treatments based on completely randomized design. CCC was sprayed on Stevia shoots, but PBZ was applied as a drench. The obtained results showed that CCC reduced plant height but improved leaf and stem dry weight, especially with 750 ppm concentration. Total SVgly content and consequently SVglys yield were significantly reduced by CCC application, and 1,000 ppm of CCC concentration was more effective than other treatments. PBZ had no effect on Stevia height while it significantly enhanced stem and dry weight at 12 ppm. Moreover, PBZ remarkably increased total SVglys contents, SVglys yield, and Rebaudioside A/Stevioside ratio. Total antioxidant capacity significantly varied with CCC and PBZ and the highest activity was obtained with 1,000 and 12 ppm of CCC and PBZ, respectively. The results of these experiments indicated that, although CCC and PBZ are plant growth retardants and act as anti-gibberellins, only CCC reduced plant height and SVglys production in Stevia. On the contrary, PBZ at 12 ppm concentration, improved Stevia growth, SVglys production, and antioxidant capacity.  相似文献

Stevia rebaudiana (Bert.) is an emerging sugar alternative and anti-diabetic plant in Pakistan. That is why people did not know the exact time of propagation. The main objective of the present study was to establish feasible propagation methods for healthy biomass production. In the present study, seed germination, stem cuttings and micropropagation were investigated for higher productivity. Fresh seeds showed better germination (25.51–40%) but lost viability after a few days of storage. In order to improve the germination percentage, seeds were irradiated with 2.5, 5.0, 7.5 and 10 Gy gamma doses. But gamma irradiation did not show any significant change in seed germination. A great variation in survival of stem cutting was observed in each month of 2012. October and November were found the most suitable months for stem cutting survival (60%). In order to enhance survival, stem cuttings were also dipped in different plant growth regulators (PGRs) solution. Only indole butyric acid (IBA; 1000 ppm) treated cutting showed a higher survival (33%) than control (11.1%). Furthermore, simple and feasible indirect regeneration system was established from leaf explants. Best callus induction (84.6%) was observed on MS-medium augmented with 6-benzyladenine (BA) and 2,4-dichlorophenoxyacetic acid (2,4-D; 2.0 mg l−1). For the first time, we obtained the highest number of shoots (106) on a medium containing BA (1.5 mg l−1) and gibberellic acid (GA3; 0.5 mg l−1). Plantlets were successfully acclimatized in plastic pots. The current results preferred micropropagation (85%) over seed germination (25.51–40%) and stem cutting (60%).  相似文献
