KAAT1 is a lepidopteran neutral amino acid transporter belonging to the NSS super family (SLC6), which has an unusual cation selectivity, being activated by K+ and Li+ in addition to Na+. We have previously demonstrated that Asp338 is essential for KAAT1 activation by K+ and for the coupling of amino acid and driver ion fluxes. By comparing sequences of NSS family members, site-directed mutagenesis, and expression in Xenopus laevis oocytes, we identified Lys102 as a residue likely to interact with Asp338. Compared with wild type, the single mutants K102V and D338E each showed altered leucine uptake and transport-associated currents in the presence of both Na+ and K+. However, in K102V/D338E double mutant, the K102V mutation reversed both the inhibition of Na+-dependent transport and the block in K+-dependent transport that characterize the D338E mutant. K+-dependent leucine currents were not observed in any mutants with D338E. In the presence of the oxidant Cu(II) (1,10-phenanthroline)3, we observed specific and reversible inhibition of K102C/D338C mutant, but not of the corresponding single cysteine mutants, suggesting that these residues are sufficiently close to form a disulfide bond. Thus both structural and functional evidence suggests that these two residues interact. Similar results have been obtained mutating the bacterial transporter homolog TnaT. Asp338 corresponds to Asn286, a residue located in the Na+ binding site in the recently solved crystal structure of the NSS transporter LeuTAa (41). The highly homologous neutral amino acid transporters KAAT1 and CAATCH1, cloned from the midgut epithelium of the Manduca sexta larva, are members of the Na+/Cl-dependent transporter family. Recent evidence indicates that transporters of this family form constitutive oligomers. CAATCH1 and KAAT1 give rise to specific kinds of current depending on the transported amino acid, cotransported ion, pH, and membrane voltage. Different substrates induce notably distinct transport-associated currents in the two proteins that represent useful tools in structural-functional studies. To determine whether KAAT1 and CAATCH1 form functional oligomers, we have constructed four concatameric proteins for electrophysiological analysis, consisting of one KAAT1 protein covalently linked to another KAAT1 (K-K concatamer) or to CAATCH1 (K-C concatamer) and vice versa (C-C concatamer and C-K concatamer), and eight constructs where the two transporters were linked to yellow or cyan fluorescent protein in the NH2 or COOH terminus, to determine the oligomer formation and the relative distance between the different subunits by fluorescence resonance energy transfer (FRET) analysis. Heterologous expression of the concatenated constructs and coinjection of the original proteins in different proportions allowed us to compare the characteristics of the currents to those of the oocytes expressing only the wild-type proteins. All the constructs were fully active, and their electrophysiological behavior was consistent with the activity as monomeric proteins. We have clonedand functionally characterized the human Na+-dependenthigh-affinity dicarboxylate transporter (hNaDC3) from placenta. ThehNaDC3 cDNA codes for a protein of 602 amino acids with 12 transmembrane domains. When expressed in mammalian cells, the clonedtransporter mediates the transport of succinate in the presence ofNa+ [concentration of substrate necessary for half-maximaltransport (Kt) for succinate = 20 ± 1 µM]. Dimethylsuccinate also interacts with hNaDC3. TheNa+-to-succinate stoichiometry is 3:1 and concentration ofNa+ necessary for half-maximal transport(KNa+0.5) is 49 ± 1 mM as determined by uptake studies withradiolabeled succinate. When expressed in Xenopuslaevis oocytes, hNaDC3 induces Na+-dependent inwardcurrents in the presence of succinate and dimethylsuccinate. At amembrane potential of 50 mV,KSuc0.5 is 102 ± 20 µM andKNa+0.5 is 22 ± 4 mM as determined by the electrophysiological approach. Simultaneous measurements of succinate-evoked charge transfer andradiolabeled succinate uptake in hNaDC3-expressing oocytes indicate acharge-to-succinate ratio of 1:1 for the transport process, suggestinga Na+-to-succinate stoichiometry of 3:1. pH titration ofcitrate-induced currents shows that hNaDC3 accepts preferentially thedivalent anionic form of citrate as a substrate. Li+inhibits succinate-induced currents in the presence of Na+.Functional analysis of rat-human and human-rat NaDC3 chimeric transporters indicates that the catalytic domain of the transporter lies in the carboxy-terminal half of the protein. The humanNaDC3 gene is located on chromosome20q12-13.1, as evidenced by fluorescent in situ hybridization. Thegene is >80 kbp long and consists of 13 exons and 12 introns.

The Na+-dependent nucleoside transporter 2 (CNT2) mediates active transport of purine nucleosides and uridine as well as therapeutic nucleoside analogs. We used the two-electrode voltage-clamp technique to investigate rat CNT2 (rCNT2) transport mechanism and study the interaction of nucleoside-derived drugs with the transporter expressed in Xenopus laevis oocytes. The kinetic parameters for sodium, natural nucleosides, and nucleoside derivatives were obtained as a function of membrane potential. For natural substrates, apparent affinity (K0.5) was in the low micromolar range (12–34) and was voltage independent for hyperpolarizing membrane potentials, whereas maximal current (Imax) was voltage dependent. Uridine and 2'-deoxyuridine analogs modified at the 5-position were substrates of rCNT2. Lack of the 2'-hydroxyl group decreased affinity but increased Imax. Increase in the size and decrease in the electronegativity of the residue at the 5-position affected the interaction with the transporter by decreasing both affinity and Imax. Fludarabine and formycin B were also transported with higher Imax than uridine and moderate affinity (102 ± 10 and 66 ± 6 µM, respectively). Analysis of the pre-steady-state currents revealed a half-maximal activation voltage of about –39 mV and a valence of about –0.8. K0.5 for Na+ was 2.3 mM at –50 mV and decreased at hyperpolarizing membrane potentials. The Hill coefficient was 1 at all voltages. Direct measurements of radiolabeled nucleoside fluxes with the charge associated showed a ratio of two positive inward charges per nucleoside, suggesting a stoichiometry of two Na+ per nucleoside. We havefunctionally characterized Na+-driven bicarbonatetransporter (NBC)4, originally cloned from human heart by Pushkin etal. (Pushkin A, Abuladze N, Newman D, Lee I, Xu G, and Kurtz I. Biochem Biophys Acta 1493: 215-218, 2000). Of the fourNBC4 variants currently present in GenBank, our own cloning efforts yielded only variant c. We expressed NBC4c (GenBank accession no.AF293337) in Xenopus laevis oocytes and assayed membrane potential (Vm) and pH regulatory function withmicroelectrodes. Exposing an NBC4c-expressing oocyte to a solutioncontaining 5% CO2 and 33 mM HCOelicited a large hyperpolarization, indicating that the transporter iselectrogenic. The initial CO2-induced decrease inintracellular pH (pHi) was followed by a slow recovery thatwas reversed by removing external Na+. Two-electrodevoltage clamp of NBC4c-expressing oocytes revealed largeHCO- and Na+-dependent currents. When wevoltage clamped Vm far from NBC4c's estimatedreversal potential (Erev), the pHirecovery rate increased substantially. Both the currents andpHi recovery were blocked by 200 µM4,4'-diisothiocyanostilbene-2,2'-disulfonic acid (DIDS). We estimatedthe transporter's HCO:Na+ stoichiometryby measuring Erev at different extracellularNa+ concentration ([Na+]o)values. A plot of Erev againstlog[Na+]o was linear, with a slope of 54.8 mV/log[Na+]o. This observation, as well asthe absolute Erev values, are consistent with a2:1 stoichiometry. In conclusion, the behavior of NBC4c, which wepropose to call NBCe2-c, is similar to that of NBCe1, the firstelectrogenic NBC.

The effect of Na+ on phosphate uptake was studied in four strainsof cyanobacteria: Synechococcus PCC 7942, Gloeothece PCC 6501,Phormidium sp. and Chlorogloeopsis PCC 6912. Phosphate uptakewas stimulated by Na+ in all cases. Li+ and K+ acted as partialanalogues for Na+. Half-saturation [K1/2(Na+)] of phosphateuptake was reached with Na+ concentrations ranging from 317µM in Chlorogloeopsis to 659 µM in Phormidium. High concentrations of cytosolic Na+ ions induce the time-dependent formation of an inactive state of the Na+/Ca2+ exchanger (NCX), a process known as Na+-dependent inactivation. NCX activity was measured as Ca2+ uptake in fura 2-loaded Chinese hamster ovary (CHO) cells expressing the wild-type (WT) NCX or mutants that are hypersensitive (F223E) or resistant (K229Q) to Na+-dependent inactivation. As expected, 1) Na+-dependent inactivation was promoted by high cytosolic Na+ concentration, 2) the F223E mutant was more susceptible than the WT exchanger to inactivation, whereas the K229Q mutant was resistant, and 3) inactivation was enhanced by cytosolic acidification. However, in contrast to expectations from excised patch studies, 1) the WT exchanger was resistant to Na+-dependent inactivation unless cytosolic pH was reduced, 2) reducing cellular phosphatidylinositol-4,5-bisphosphate levels did not induce Na+-dependent inactivation in the WT exchanger, 3) Na+-dependent inactivation did not increase the half-maximal cytosolic Ca2+ concentration for allosteric Ca2+ activation, 4) Na+-dependent inactivation was not reversed by high cytosolic Ca2+ concentrations, and 5) Na+-dependent inactivation was partially, but transiently, reversed by an increase in extracellular Ca2+ concentration. Thus Na+-dependent inactivation of NCX expressed in CHO cells differs in several respects from the inactivation process measured in excised patches. We examined 1) whether the effects of lowered trans-sarcolemmal Na+ gradient on force differed between nonfatigued fast- and slow-twitch muscles of mice and 2) whether effects on action potentials could explain the decrease of force. The Na+ gradient was reduced by lowering the extracellular [Na+] ([Na+]o). The peak force-[Na+]o relationships for the twitch and tetanus were the same in nonfatigued extensor digitorum longus and soleus muscles: force was maintained over a large range of [Na+]o and then decreased abruptly over a much smaller range. However, fatigue was significantly exacerbated at a lowered [Na+]o that had little effect in nonfatigued soleus muscle. This finding suggests that substantial differences exist in the Na+ effect on force between nonfatigued and fatigued muscle. The general phosphate need in mammalian cells is accommodated by members of the Pi transport (PiT) family (SLC20), which use either Na+ or H+ to mediate inorganic phosphate (Pi) symport. The mammalian PiT paralogs PiT1 and PiT2 are Na+-dependent Pi (NaPi) transporters and are exploited by a group of retroviruses for cell entry. Human PiT1 and PiT2 were characterized by expression in Xenopus laevis oocytes with 32Pi as a traceable Pi source. For PiT1, the Michaelis-Menten constant for Pi was determined as 322.5 ± 124.5 µM. PiT2 was analyzed for the first time and showed positive cooperativity in Pi uptake with a half-maximal activity constant for Pi of 163.5 ± 39.8 µM. PiT1- and PiT2-mediated Na+-dependent Pi uptake functions were not significantly affected by acidic and alkaline pH and displayed similar Na+ dependency patterns. However, only PiT2 was capable of Na+-independent Pi transport at acidic pH. Study of the impact of divalent cations Ca2+ and Mg2+ revealed that Ca2+ was important, but not critical, for NaPi transport function of PiT proteins. To gain insight into the NaPi cotransport function, we analyzed PiT2 and a PiT2 Pi transport knockout mutant using 22Na+ as a traceable Na+ source. Na+ was transported by PiT2 even without Pi in the uptake medium and also when Pi transport function was knocked out. This is the first time decoupling of Pi from Na+ transport has been demonstrated for a PiT family member. Evidence suggests that 1) ischemia-reperfusion injury is due largely to cytosolic Ca2+ accumulation resulting from functional coupling of Na+/Ca2+ exchange (NCE) with stimulated Na+/H+ exchange (NHE1) and 2) 17-estradiol (E2) stimulates release of NO, which inhibits NHE1. Thus we tested the hypothesis that acute E2 limits myocardial Na+ and therefore Ca2+ accumulation, thereby limiting ischemia-reperfusion injury. NMR was used to measure cytosolic pH (pHi), Na+ (Na), and calcium concentration ([Ca2+]i) in Krebs-Henseleit (KH)-perfused hearts from ovariectomized rats (OVX). Left ventricular developed pressure (LVDP) and lactate dehydrogenase (LDH) release were also measured. Control ischemia-reperfusion was 20 min of baseline perfusion, 40 min of global ischemia, and 40 min of reperfusion. The E2 protocol was identical, except that 1 nM E2 was included in the perfusate before ischemia and during reperfusion. E2 significantly limited the changes in pHi, Na and [Ca2+]i during ischemia (P < 0.05). In control OVX vs. OVX+E2, pHi fell from 6.93 ± 0.03 to 5.98 ± 0.04 vs. 6.96 ± 0.04 to 6.68 ± 0.07; Na rose from 25 ± 6 to 109 ± 14 meq/kg dry wt vs. 25 ± 1 to 76 ± 3; [Ca2+]i changed from 365 ± 69 to 1,248 ± 180 nM vs. 293 ± 66 to 202 ± 64 nM. E2 also improved recovery of LVDP and diminished release of LDH during reperfusion. Effects of E2 were diminished by 1 µM N-nitro-L-arginine methyl ester. Thus the data are consistent with the hypothesis. Serum- and glucocorticoid-induced kinase 1 (SGK1) is thought to be an important regulator of Na+ reabsorption in the kidney. It has been proposed that SGK1 mediates the effects of aldosterone on transepithelial Na+ transport. Previous studies have shown that SGK1 increases Na+ transport and epithelial Na+ channel (ENaC) activity in the apical membrane of renal epithelial cells. SGK1 has also been implicated in the modulation of Na+-K+-ATPase activity, the transporter responsible for basolateral Na+ efflux, although this observation has not been confirmed in renal epithelial cells. We examined Na+-K+-ATPase function in an A6 renal epithelial cell line that expresses SGK1 under the control of a tetracycline-inducible promoter. The results showed that expression of a constitutively active mutant of SGK1 (SGK1TS425D) increased the transport activity of Na+-K+-ATPase 2.5-fold. The increase in activity was a direct consequence of activation of the pump itself. The onset of Na+-K+-ATPase activation was observed between 6 and 24 h after induction of SGK1 expression, a delay that is significantly longer than that required for activation of ENaC in the same cell line (1 h). SGK1 and aldosterone stimulated the Na+ pump synergistically, indicating that the pathways mediated by these molecules operate independently. This observation was confirmed by demonstrating that aldosterone, but not SGK1TS425D, induced an 2.5-fold increase in total protein and plasma membrane Na+-K+-ATPase 1-subunit abundance. The ability of the two highly homologous Na+/Cl-dependent neutral amino acid transporters KAAT1 and CAATCH1, cloned from the midgut epithelium of the larva Manduca sexta, to transport different amino acids depends on the cotransported ion, on pH, and on the membrane voltage. Different organic substrates give rise to transport-associated currents with their own characteristics, which are notably distinct between the two proteins. Differences in amplitude, kinetics, and voltage dependence of the transport-associated currents have been observed, as well as different substrate selectivity patterns measured by radioactive amino acid uptake assays. These diversities represent useful tools to investigate the structural determinants involved in the substrate selectivity. To identify these regions, we built four chimeric proteins between the two transporters. These proteins, heterologously expressed in Xenopus laevis oocytes, were analyzed by two-electrode voltage clamp and uptake measurements. Initially, we exchanged the first three domains, obtaining the chimeras C3K9 and K3C9 (where numbers indicate the transmembrane domains and letters represent the original proteins), which showed electrophysiological and [3H]amino acid uptake characteristics resembling those of KAAT1 and CAATCH1, respectively. Subsequent substitution of the last four domains in C3K9 and K3C9 gave the proteins C3K5C4 and K3C5K4, which showed the same behavior as KAAT1 and CAATCH1 in electrophysiological and transport determinations. This study characterized theNa+-dependent transport of L-glutamine by ahuman neuroblastoma cell line, SK-N-SH. The Na+-dependentcomponent represented >95% of the total glutamine uptake. Kineticstudies showed a single saturable high-affinity carrier with aMichaelis constant (Km) of 163 ± 23 µMand a maximum transport velocity (Vmax) of13,713 ± 803 pmol · mgprotein1 · min1. Glutamine uptakewas markedly inhibited in the presence of L-alanine, L-asparagine, and L-serine. Li+ didnot substitute for Na+. These data show thatL-glutamine is predominantly taken up through systemASC. Glutamine deprivation resulted in the decrease of glutamine transport by a mechanism that decreasedVmax without affectingKm. The expression of the system ASC subtypeASCT2 decreased in the glutamine-deprived group, whereas glutaminedeprivation did not induce changes in system ASC subtype ASCT1 mRNAexpression. Adaptive increases in Na+-dependent glutamate,Na+-dependent 2-(methylamino)isobutyric acid, andNa+-independent leucine transport were observed underglutamine-deprived conditions, which were completely blocked byactinomycin D and cycloheximide. These mechanisms may allow cells tosurvive and even grow under nutrient-deprived conditions.

Members of the SLC20 family or type III Na+-coupled Pi cotransporters (PiT-1, PiT-2) are ubiquitously expressed in mammalian tissue and are thought to perform a housekeeping function for intracellular Pi homeostasis. Previous studies have shown that PiT-1 and PiT-2 mediate electrogenic Pi cotransport when expressed in Xenopus oocytes, but only limited kinetic characterizations were made. To address this shortcoming, we performed a detailed analysis of SLC20 transport function. Three SLC20 clones (Xenopus PiT-1, human PiT-1, and human PiT-2) were expressed in Xenopus oocytes. Each clone gave robust Na+-dependent 32Pi uptake, but only Xenopus PiT-1 showed sufficient activity for complete kinetic characterization by using two-electrode voltage clamp and radionuclide uptake. Transport activity was also documented with Li+ substituted for Na+. The dependence of the Pi-induced current on Pi concentration was Michaelian, and the dependence on Na+ concentration indicated weak cooperativity. The dependence on external pH was unique: the apparent Pi affinity constant showed a minimum in the pH range 6.2–6.8 of 0.05 mM and increased to 0.2 mM at pH 5.0 and pH 8.0. Xenopus PiT-1 stoichiometry was determined by dual 22Na-32Pi uptake and suggested a 2:1 Na+:Pi stoichiometry. A correlation of 32Pi uptake and net charge movement indicated one charge translocation per Pi. Changes in oocyte surface pH were consistent with transport of monovalent Pi. On the basis of the kinetics of substrate interdependence, we propose an ordered binding scheme of Na+:H2PO4:Na+. Significantly, in contrast to type II Na+-Pi cotransporters, the transport inhibitor phosphonoformic acid did not inhibit PiT-1 or PiT-2 activity. The effects of hypo- and hypersaline treatments ranging from7–68% on the intracellular inorganic ion and organic soluteconcentrations were determined in the eulittoral green macroalgaeUlothrix implexa, Ulothrix subflaccida, Enteromorpha bulbosa,Acrosiphonia arcta, and Ulva rigida from Antarctica and SouthernChile. The main inorganic cations were K+, Na+, and Mg2+ inall species. The major osmolyte in E. bulbosa, A. arcta, andU. rigida was K+ at increasing salinities. In both Ulothrixspecies, however, K+ levels declined during hypersaline stressand Na+ concentrations rose significantly. The main inorganicanions were Cl-, SO24-, and PO34- in all algae, while E. bulbosaand U. rigida also contained NO+3. A. arcta showed an extremelyhigh SO2-4 content. The organic solutes proline, sucrose, andß-dimethylsulphoniopropionate (DMSP) played an importantrole in osmotic acclimation. The occurrence of three organicosmolytes suggests an additional function of these solutes ascryoprotectants in the cold-water macroalgae investigated.  相似文献

Leaves of three C4 plants, Setaria italica, Pennisetum typhoides,and Amaranthus paniculatus possessed five- to ten-fold higheractivities of a (Na+-K+)-dependent ATPase than those of twoC3 plants, Oryza sativa and Rumex vesicarius. Na+-K+ ATPasefrom leaves of Amarathus exhibited an optimal pH of 7?5 andan optimal temperature of 35 ?C. It required 40 mM K+ and 80mM Na+ for maximal activity. Ouabain partially inhibited (Na+-K+)-dependentATPase activity in leaves of C4 plants. Ouabain also blockedthe movement of label from initially formed C4 acids into endproducts in leaves of only C4 plants, Setaria and Amaranthusbut not in a C3 plant, Rumex. We propose that Na+-K+ ATPasemay mediate transfer of energy during active transport of C4acids from mesophyll into the bundle sheath.  相似文献

Theeffects of different bathing solutions and fusicoccin (FC),a known activator of plasma membrane ATPase, on plasma membranepotential (Em) and net fluxes of Na+, K+and H+were studied inwheat suspension cells (Triticum aestivum) in response to differentNaCl treatments. Emof cells in Murashige and Skoog (MS) mediumwas less negative than in cells exposed to a medium containing10 mM KCl + 0.1 m M CaCl2(KSM) and to a basic salt medium (BSM),containing 1 m M KCl and 0.1 m M CaCl2. Multiphasic Na+accumulationin cells was observed, peaking at 13 min after addition of 120m M NaCl to MS medium. This time scale was in good agreementwith net Na+flux changes measured non-invasively by moving ion-selectivemicroelectrodes (the MIFE system). When 120 m M NaCl was addedto all media studied, a quick rise of Na+influx was reversedwithin the first 20 min. In both 120 and 20 m M NaCl treatmentsin MS medium, net Na+efflux was observed, indicating that activeNa+transporters function in the plant cell response to saltstress. Lower external K+concentrations (KSM and BSM) and FCpre-treatment caused shifts in Na+fluxes towards net influxat 120 m M NaCl stress. The growth of garden orache, A triplex hortensis was studiedunder conditions of mild NaCl or Na2SO4 salinity. Growth, drymatter production and leaf size were substantially stimulatedat 10 mM and 50 mM Na+ salts. Increased growth, however, appearedto be due to a K+-sparing effect of Na+ rather than to salinityper se. The distribution of K+ and Na+ in the plant revealeda remarkable preference for K+ in the roots and the hypocotyl.In the shoot the K/Na ratio decreased strongly with leaf age.However, the inverse changes in K+ and Na+ content with leafage were dependent on the presence of bladder hairs, which removedalmost all of the Na+ from the young leaf lamina. Measurementsof net fluxes of K+ and Na+ into roots and shoots of growingAtriplex plants showed a higher K/Na selectivity of the netion flux to the root compared to the shoot. With increasingsalinity the selectivity ratio SK, Na* of net ion fluxes tothe roots and to the shoots was increased. The data suggestthat recirculation of K+ from leaves to roots is an importantlink in establishing the K/Na selectivity in A. hortensis plants.The importance of K+ recirculation and phloem transport forsalt tolerance is discussed. Patch-clampstudies of mammalian skeletal muscleNa+ channels are commonly done atsubphysiological temperatures, usually room temperature. However, atsubphysiological temperatures, mostNa+ channels are inactivated atthe cell resting potential. This study examined the effects oftemperature on fast and slow inactivation ofNa+ channels to determine iftemperature changed the fraction of Na+ channels that were excitableat resting potential. The loose patch voltage clamp recordedNa+ currents(INa) in vitroat 19, 25, 31, and 37°C from the sarcolemma of rat type IIbfast-twitch omohyoid skeletal muscle fibers. Temperature affected thefraction of Na+ channels that wereexcitable at the resting potential. At 19°C, only 30% of channelswere excitable at the resting potential. In contrast, at 37°C, 93%of Na+ channels were excitable atthe resting potential. Temperature did not alter the resting potentialor the voltage dependencies of activation or fast inactivation.INa available atthe resting potential increased with temperature because thesteady-state voltage dependence of slow inactivation shifted in adepolarizing direction with increasing temperature. The membranepotential at which half of the Na+channels were in the slow inactivated state was shifted by +16 mV at37°C compared with 19°C. Consequently, the low availability ofexcitable Na+ channels atsubphysiological temperatures resulted from channels being in the slow,inactivated state at the resting potential.
