Five new cembranoids, sarcocrassocolides A–E (1–5), along with three known cembranoids 6–8, have been isolated from a Formosan soft coral Sarcophyton crassocaule. The structures of the new metabolites were elucidated by extensive spectroscopic analysis and the absolute configuration of 1 was determined by a modified Mosher’s method. Compounds 1–4 exhibited significantly cytotoxic activity against a limited panel of cancer cell lines. Compounds 1–4, 6 and 8 were shown to exert significant in vitro anti-inflammatory activity in LPS-stimulated RAW264.7 macrophage cells. Compound 6 also significantly inhibited the accumulation of pro-inflammatory COX-2 protein. 相似文献

Two new diterpenes, lobocompactols A (1) and B (2), and five known compounds (3-7) were isolated from the methanol extract of the soft coral Lobophytum compactum using combined chromatographic methods and identified based on NMR and MS data. Each compound was evaluated for cytotoxic activity against A549 (lung) and HL-60 (acute promyelocytic leukemia) human cancer cell lines. Among them, compound 5 exhibited strong cytotoxic activity against the A549 cell line with an IC50 of 4.97 ± 0.06 μM. Compounds 3, 4, and 7 showed moderate activity with IC50 values of 23.03 ± 0.76, 31.13 ± 0.08, and 36.45 ± 0.01 μM, respectively. The cytotoxicity of 5 on the A549 cells was comparable to that of the positive control, mitoxantrone (MX). All compounds exhibited moderate cytotoxicity against the HL-60 cell line, with IC50 values ranging from 17.80 ± 1.43 to 59.06 ± 2.31 μM. Their antioxidant activity was also measured using oxygen radical absorbance capacity method, compounds 1 and 2 exhibiting moderate peroxyl radical scavenging activity of 1.4 and 1.3 μM Trolox equivalents, respectively, at a concentration of 5 μM. 相似文献

Chemical investigation of the soft coral Lobophytum durum resulted in the isolation of seven new cembranolides, durumolides F–L (1–7), as well as one previously characterized cembranolides, sinularolide D (8). The molecular structures of these isolated metabolites were determined mainly through NMR techniques and HRESIMS analysis. Moreover, the absolute configurations of 1 and 5 were established by application of modified Mosher’s method. The antibacterial activities, anti-inflammatory effects, and anti-HCMV (Human cytomegalovirus) endonuclease activity of metabolites 1–8 were also evaluated in vitro. Anti-inflammatory activity of metabolites 1 and 6 (10 μM) significantly reduced the levels of the iNOS protein to 0.8 ± 0.6% and 5.7 ± 2.2%, respectively, and COX-2 protein to 47.8 ± 9.0% and 71.6 ± 5.8%, respectively. Metabolites 1–8 (100 μg/disk) exhibited weak antibacterial activity against Salmonella enteritidis. 相似文献

A new 10-membered-ring diterpene, cyclolobatriene (1), along with three other known diterpenes, lobatriene (2), eunicol (3), and fuscol (4), were isolated from the Okinawan soft coral Lobophytum pauciflorum. Their structures were established by extensive NMR spectroscopic analyses. Cyclolobatriene (1) is an additional example of rare prenylated germacrenes. Although 1, due to a 10-membered-ring structure, exists as an equilibrium mixture of three conformers, the NMR measurement in CDCl(3) at 7°C enabled us to assign the NMR signals of the three, which is the first example of the complete NMR assignment of all the existing conformers of germacrene-type compounds. Cyclolobatriene (1) was thermally unstable and converted into 2 through Cope rearrangement upon heating at 70°C. Eunicol (3) also possesses the same prenylated germacrene structure as 1, showing similar physico-chemical properties to 1. All four compounds 1-4 showed cytotoxic effect with IC(50)'s of 0.64, 0.41, 0.35 and 0.52 μM, respectively, against human epidermoid carcinoma A431 cells. 相似文献

Different chemotypes of Sinularia flexibilis exist in the Hainan island. Thus, a collection of this soft coral from a location different from the one of our previous study afforded three novel cembranoid esters featuring a n-butyl alcohol moiety, a structural element rare in natural products of both terrestrial and marine origin. The structures of the new compounds were elucidated by detailed spectroscopic analysis and by the comparison of their spectroscopic data with those reported in the literature. In addition, the absolute stereochemistry of the previously reported diepoxycembrene (9) was first time determined by the X-ray diffraction analysis. In bioassays, compounds 6–8 exhibited strong anti-inflammatory effect with IC50 values of 2.7, 4.7, and 4.2?μM, respectively, whereas compound 5 displayed cytotoxicity against several cancer cells with IC50 values ranging from 8.9 to 27.4?μM. Abstract. The alcyonacean soft coral genera Sarcophyton and Lobophytum are conspicuous, ecologically important members of shallow reef communities throughout the Indo-West Pacific. Study of their ecology is, however, hindered by incomplete knowledge of their taxonomy: most species cannot be identified in the field and the two genera cannot always be distinguished reliably. We used a 735-bp fragment of the octocoral-specific mitochondrial protein-coding gene msh1 to construct a phylogeny for 92 specimens identified to 19 species of Lobophytum and 16 species of Sarcophyton . All phylogenetic methods used recovered a tree with three strongly supported clades. One clade included only morphologically typical Sarcophyton species with a stalk distinct from the polypary, poorly formed club-shaped sclerites in the colony surface, and large spindles in the interior of the stalk. A second clade included only morphologically typical Lobophytum colonies with lobes and ridges on the colony surface, poorly formed clubs in the colony surface, and interior sclerites consisting of oval forms with regular girdles of ornamental warts. The third distinct clade included a mix of Sarcophyton and Lobophytum nominal species with intermediate morphologies. Most of the species in this mixed clade had a polypary that was not distinct from the stalk, and the sclerites in the colony surface were clubs with well-defined heads. Within the Sarcophyton clade, specimens identified as Sarcophyton glaucum belonged to six very distinct genetic sub-clades, suggesting that this morphologically heterogeneous species is actually a cryptic species complex. Our results highlight the need for a complete taxonomic revision of these genera, using molecular data to help confirm species boundaries as well as to guide higher taxonomic decisions. 相似文献

The first and in-depth chemical investigation of the South China Sea soft coral Sinularia scabra has resulted to the isolation of a library of diverse cembrane type diterpenoids, including six new compounds, namely xiguscabrates A and B (1 and 2), xiguscabral A (3), xiguscabrols A and B (4 and 5), and 8-epi-xiguscabrol B (6), and twenty-seven known analogs (7–33). Their structures were elucidated by extensive spectroscopic analysis and by the comparison with literature data. In bioassay, several isolates exhibited inhibitory effects on the ConA-induced T lymphocytes and/or LPS-induced B lymphocytes proliferation. Among them, compound 24 showed considerable specific inhibition on B cell proliferation, with IC50 value of 4.4 μM and selectivity index (SI) of 10.9. The structure-activity relationship (SAR) of the tested metabolites was analyzed, and the further mechanism study of the specific B-cell targeted immunosuppressive compound 24 on purified CD19+ B cells was also performed to uncover the effects on the function and maturity of B cells, including cytokines production, abnormal activation, antigen presenting capacity and plasma cells formation. 相似文献

We document long-term effects of a simulated bleaching event on the reproductive output and offspring viability of the soft
coral Lobophytum compactum. Corals were subjected to temperature and solar radiation treatments to produce both moderately (48–60%) and heavily (90–95%)
bleached colonies. Although bleached colonies recovered their zooxanthellae within 10 to 18 weeks, impacts on reproductive
output were significant for at least two annual spawning seasons. In the first year, both polyp fecundity and mean oocyte
diameter were reduced and inversely correlated with the degree of bleaching, with complete failure of fertilization in the
group of heavily bleached colonies. For moderately bleached soft corals, survival and growth of sexual offspring did not differ
significantly from those of unbleached colonies. Although no further reductions in zooxanthellae densities in experimental
soft corals were recorded throughout the subsequent second year, egg size and fecundity of the heavily bleached soft corals
were still significantly reduced 20 months later. Severe bleaching clearly has long-term sub-lethal impacts, reducing overall
reproductive output for at least two spawning seasons.

Experimental bleaching reduces the levels of important biochemical parameters in adult tissues and eggs of the soft coral
Lobophytum compactum. Protein, lipid, mycosporine-like amino acids (MAAs) and carotenoid concentrations remained lower in bleached adults than
in controls for at least 8 months. Reductions in concentrations of all four parameters were greater in eggs than in maternal
tissues, potentially jeopardizing egg and larval viability. In particular, reductions in lipids, proteins and carotenoids
in tissues of heavily bleached soft corals were amplified approximately twofold in eggs. In comparison, amplification of maternal
tissue reductions were not as great for MAAs, suggesting that MAAs are given higher priority in egg provisioning. Our finding
that MAA levels are normally three times higher in eggs than in unbleached maternal tissues supports the importance of MAAs
for larval survival. Twenty months after experimental bleaching the biochemical composition of both adult tissues and their
eggs were indistinguishable from those of control (unbleached) soft corals.

The structures of the isolated compounds were elucidated based on extensive spectroscopic evidence including Fourier transform ion cyclotron resonance mass spectrometry (FTICR-MS) and both one- and two-dimensional nuclear magnetic resonance (1D and 2D NMR, respectively), in comparison with reported data. Compound 6 potently inhibited IL-12 and IL-6 production in LPS-stimulated bone marrow derived dendritic (BMDCs) with IC50 values of 5.30 ± 0.21 and 13.12 ± 0.64 μM, respectively. Chemical investigations of the soft coral Sinularia gyrosa resulted in the isolation of six new norcembranolides, gyrosanolides A–F (1–6), a new norcembrane, gyrosanin A (7), and 11 known norditerpenoids 8–18. The structures of the isolated compounds were elucidated through extensive spectroscopic data and by comparison with reported data in the literature. Compounds 1–3, 7–9, 12, and 13 at concentration of 10 μM did not inhibit the COX-2 protein expression, but significantly reduced the levels of the iNOS protein (55.2 ± 14.6%, 18.6 ± 6.7%, 10.6 ± 4.6%, 66.9 ± 5.2%, 10.2 ± 5.1%, 17.4 ± 7.2%, 47.2 ± 11.9%, and 56.3 ± 5.1%, respectively) by LPS stimulation. A repeated silica gel column chromatography followed by HPLC purification on the methanol extract of marine soft coral Sinularia dissecta, resulted in the isolation of fifteen polyhydroxylated steroids (1-15), involving six new C-18 functionalized steroids, 3beta-acetoxy-1alpha,11alpha-dihydroxygorgost-5-en-18-oic acid (1), gorgost-5-en-1alpha,3beta,11alpha,18-tetrol (2), 18-acetoxy-1alpha,3beta,11alpha-trihydroxygorgost-5-ene (3), 24(S)-3beta-acetoxy-1alpha, 11alpha-dihydroxyergost-5-en-18-oic acid (4), 24(S)-ergost-5-en-1alpha,3beta,11alpha,18-tetrol (5), and dissectolide (6). The structures of the new compounds were determined on the basis of extensive spectroscopic data (IR, MS, (1)H and (13)C NMR, HMQC, HMBC, and NOESY) analysis. Two new phenolic compounds, caesalpiniaphenols G–H (1 and 2), were isolated from Vietnamese Caesalpinia sappan heartwood. The chemical structures were established mainly by extensive spectroscopic studies and chemical evidence. Compounds 1 and 2 showed potent inhibitory activity against HL-60 cancer cell lines with respective IC50 values of 16.7 and 22.5 µg/mL. Treating HL-60 cells with various concentrations of 1 resulted in growth inhibition and the induction of apoptosis. 相似文献
