(1) Octylglucoside stimulates an Mg2+-specific ATPase activity with CF1 preparations from different higher plants and the alga Chlamydomonas reinhardii. (2) Tentoxin at high concentrations (10?4–10?3 M) in the presence of octylglucoside further stimulates the Mg2+-ATPase activity of CF1 from tentoxin-sensitive species and inhibits the activity of CF1 from tentoxin-resistant species. The extent of tentoxin stimulation and inhibition varies among species. A maximal stimulation of over 2-fold was obtained with spinach CF1 and a maximal inhibition of 50% was obtained with C. reinhardii CF1. In Nicotiana spp., tentoxin had only a marginal effect on the Mg2+-ATPase activity induced by octylglucoside.  相似文献

Ferrocene reacts with hexafluoroacetone trihydrate in refluxing octane to afford >80% yields of [CpFe(η5-C5H4C(CF3)2OH)] (X-ray), carrying out the reactions at 180 °C gives an additional 5% yield of [Fe(η5-C5H4C(CF3)2OH)2] (X-ray).The mono alcohol is lithiated with ButOK/BunLi/TMEDA affording partial conversion to mixtures of [CpFe(1,2-η5-C5H3C(CF3)2OH)(X)] and [Fe(η5-C5H4X)(1,2-η5-C5H3C(CF3)2OH)(X)] (X = SMe, CPh2OH) upon reaction with Me2S2 or OCPh2.For X = CPh2OH both structures are crystallographically characterised.Enantiopure [CpFe(1,2-η5-C5H3C(CF3)2OH)(SMe)] can be prepared from (R)-[CpFe(η5-C5H4S(O)C6H4Me)] via [CpFe(1,2-η5-C5H3S(O)C6H4Me)(C(CF3)2OH)] (X-ray) or [CpFe(1,2-η5-C5H3S(O)C6H4Me)(SMe)].Related procedures allow the preparation of [CpFe(1,2-η5-C5H3CPh2OH)(Y)] (Y = SMe, CHO (X-ray), C(CF3)2OH) and[CpFe(1,2-η5-C5H3C(CF3)2OH)(CHO)].  相似文献

In order to improve metabolic stability, a ring structure with a cystine moiety was introduced into TY027 (Tyr-d-Ala-Gly-Phe-Met-Pro-Leu-Trp-NH-[3′,5′-(CF3)2Bzl]), which is a lead compound of our developing bifunctional peptide possessing opioid agonist and NK1 antagonist activities. TY038 (Tyr-cyclo[d-Cys-Gly-Phe-Met-Pro-d-Cys]-Trp-NH-[3′,5′-(CF3)2Bzl]) was found as a highly selective δ opioid agonist over μ receptor in conventional tissue-based assays, together with an effective NK1 antagonist activity and good metabolic stability with more than 24 h half life in rat plasma.  相似文献

Complexes of the type [UO2(β-diket)2·nB] where β-diket=CF3COCHCOR (R=CF3, CH3, C4H3S or C6H5 and denoted by HFAA, TFAA, TTA or BTA, respectively), B=tetrahydropyran (THP) and tetrahydrothiophene (THT) and n= 0.5–2.0, have been synthesized and characterised by infrared, 1H NMR and mass spectral techniques. Vapour pressure of UO2(HFAA)2·THP was measured by the transpiration method between the temperature range of 323–391 K. The enthalpy of vaporisation of UO2(HFAA)2·THP was estimated to be 18.4 kcal/mol.  相似文献

The S-2,2,2-trifluoroethyl residue (-SCH2CF3) has been incorporated into human hemoglobin, Hb4(SH)2, as an extrinsic probe at Cys-β93 through formation of a disulfide bond. The thiol group was activated by reaction with 5,5′-dithiobis(2-nitrobenzoic acid), Hb4(SSCH2CF3)2 then being obtained by reaction with 2,2,2-trifluoroethanethiol. Both disulfide interchange reactions proceed to completion with a modest excess of reagents using dilute solutions of hemoglobin (0.005 m heme). The stoichiometry of each disulfide interchange reaction is readily determined by measurement of 5-thio-2-nitrobenzoic acid, a product of each reaction. The functional properties of Hb4 (SSCH2CF3)2 were found to be similar to those of Hb4(SH)2. At pH 7.0 and 20°C the partial pressure of oxygen required for half saturation was 0.45 mm Hg in 0.050 m 2,2-bis(hydroxymethyl)-2,2′,2″-nitrilo-triethanol, 4.1 mm Hg in 0.050 m potassium phosphate, and 16.5 mm Hg in 0.010 m inositol hexaphosphate. The n values of the Hill plots were 1.45, 1.80, and 2.3, respectively. The equilibrium constant for the tetramer-dimer dissociation reaction, K4,2, of the carbon monoxide derivative was 2.1 × 10?7m. The time course for combination with carbon monoxide was homogeneous at 432 nm. In the presence of inositol hexaphosphate the time course of combination with carbon monoxide was wavelength dependent.  相似文献

The effects of solvents on the ATPase activity of chloroplast coupling factor 1 (CF1) isolated from wild-type Chlamydomonas reinhardii have been studied. Of the solvents examined, the following order summarizes their maximal ability to stimulate the ATPase activity of CF1: ethanol > methanol>allyl alcohol >n-propanol > acetone≈dioxane > ethylene glycol. Glycerol inhibits the CF1 activity at all concentrations. In the absence of organic solvents, 50% of the activity of the enzyme is irreversibly lost after a 10 min incubation at 65–70°C. Ethanol (23%) causes a 30°C drop in the temperature required for 50% inactivation. ATP partially stabilizes the CF1 in the presence, but not in the absence, of ethanol. In the absence of organic solvents, both free Mg2+ and ADP inhibit the CF1-ATPase. Mg2+ is a noncompetitive inhibitor with respect to MgATP, and the kinetic constants are: V, 6.3 μmol ATP hydrolyzed/mg protein per min; Km(MgATP), 0.23 mM; Kii(Mg2+), 27 μM; and Kis(Mg2+), 50 μM. In the presence of ethanol, double-reciprocal plots are no longer linear and have a Hill coefficient of about 1.8±0.1. V increases about 10–12-fold. The pattern of inhibition by Mg2+ appears to change from noncompetitive to competitive with respect to MgATP. In addition, ADP no longer inhibits the MgATPase activity of CF1.  相似文献

Nickel (Ni) is a trace element essential for the growth and development of plants. Conversely, when in excess, Ni inhibits seed germination and reduces seedling growth. Therefore, we investigated the effect of Ni+2 (5–50 μM; supplied as nickel sulfate: NiSO4·6H2O) on the activity of enzymes involved in sugar metabolism of wheat (Triticum aestivum L.) seedlings after 96 h of exposure to the metal. Ni+2 treatment reduced root and coleoptile length of emerging wheat seedlings and the effect was more pronounced on the root length. Ni+2 (5–50 μM) treatment significantly enhanced carbohydrate content by 21–100 % over that of the control. In contrast, protein and reducing sugar contents declined by 17–43 and 22–69 %, respectively. The reduction in total protein content was confirmed by SDS-PAGE analysis. The activities of starch-metabolizing enzymes declined upon Ni+2 stress in a concentration-dependent manner. Activities of α- and β-amylases, acid and alkaline invertases, acid and alkaline phosphatases, and starch phosphorylase declined by 18–74 and 24–85 %, 42–76 and 21–73 %, 15–54 and 28–72 %, and 50–83 %, respectively, when compared to the control. The study concludes that Ni+2 impairs sugar metabolism as indicated by decline in the activity of sucrose and starch hydrolyzing enzymes. It resulted in decrease in the availability of biochemical energy and sugars required for the synthesis, leading to inhibition of radicle growth in germinating wheat seeds.  相似文献

Optimum production of l-tryptophan by Escherichia coli depends on pH. Here, we established conditions for optimizing the production of l-tryptophan. The optimum pH range was 6.5–7.2, and pH was controlled using a three-stage strategy [pH 6.5 (0–12 h), pH 6.8 (12–24 h), and pH 7.2 (24–38 h)]. Specifically, ammonium hydroxide was used to adjust pH during the initial 24 h, and potassium hydroxide and ammonium hydroxide (1:2, v/v) were used to adjust pH during 24–38 h. Under these conditions, NH4 + and K+ concentrations were kept below the threshold for inhibiting l-tryptophan production. Optimization was also accomplished using ratios (v/v) of glucose to alkali solutions equal to 4:1 (5–24 h) and 6:1 (24–38 h). The concentration of glucose and the pH were controlled by adjusting the pH automatically. Applying a pH-feedback feeding method, the steady-state concentration of glucose was maintained at approximately 0.2 ± 0.02 g/l, and acetic acid accumulated to a concentration of 1.15 ± 0.03 g/l, and the plasmid stability was 98 ± 0.5 %. The final, optimized concentration of l-tryptophan was 43.65 ± 0.29 g/l from 52.43 ± 0.38 g/l dry cell weight.  相似文献

A method was developed for quantitative determination of 5α,7α-dihydroxy-11-ketotetranor-prostane-1,16-dioic acid, the major urinary metabolite of prostaglandins F and F in man. The method was based on the use of the O-methyloxime derivative of [5β-3H; 10,10,12,12-2H4]5α,7α-dihydroxy-11-ketotetranor-prostane-1,16-dioic acid as internal standard and determination of ratios between unlabeled and deuterium-labeled molecules by multiple-ion analysis. Excretion values found for healthy human subjects were: males, 10.8–59.0 μg24 hr (n = 10, mean value, 24.0 ± 17.2 (SD) μg) and females, 7.6–13.6 μg24 hr (n = 10, mean value, 10.5 ± 2.1 (SD) μg).  相似文献

Electron poor cationic complexes [(CF3PCP)Pt(L)]+ (where L = CO, NC5F5, or acetone) react with H2O in polar solvents via selective hydrolysis of a single P-CF3 substituent to afford the spectroscopically-characterized phosphinoyl-bridged complex {k3-P,C,P,k1-O-(CF3)2PCH2C6H3CH2P(CF3)O}2Pt2 (1) in good yield. X-ray diffraction confirms the presence of a six-member Pt-P-O-Pt-P-O ring in a chair conformation. The presumed intermediate aqua complex, (CF3PCP)Pt(H2O)+, is stable in dichloromethane, but when dissolved in more polar solvents readily converts to 1.  相似文献

The isolation of the chloroplast ATP synthase complex (CF0-CF1) and of CF1 from Dunaliella bardawil is described. The subunit structure of the D. bardawil ATPase differs from that of the spinach in that the D. bardawil α subunit migrates ahead of the β subunit and ε-migrates ahead of subunit II of CF0 when separated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The CF1 isolated from D. bardawil resembles the CF1 isolated from Chladmydomonas reinhardi in that a reversible, Mg2+-dependent ATPase is induced by selected organic solvents. Glycerol stimulates cyclic photophosphorylation catalyzed by D. bardawil thylakoid membranes but inhibits photophosphorylation catalyzed by spinach thylakoid membranes. Glycerol (20%) also stimulates the rate of ATP-Pi exchange catalyzed by D. bardawil CF0-CF1 proteoliposomes but inhibits the activity with the spinach enzyme. The ethanol-activated, Mg2+-ATPase of the D. bardawil CF1 is more resistant to glycerol inhibition than the octylglucoside-activated, Mg2+-ATPase of spinach CF1 or the ethanol-activated, Mg2+-dependent ATPase of the C. reinhardi CF1. Both cyclic photophosphorylation and ATP-Pi exchange catalyzed by D. bardawil CF0-CF1 are more sensitive to high concentrations of NaCl than is the spinach complex.  相似文献

The target tracer [11C]methyl 4-(2-methyl-5-(methoxycarbamoyl)phenylamino)-5-methylpyrrolo[2,1-f][1,2,4]triazine-6-carboxylate ([11C]10a) and [11C]methyl 4-(2-methyl-5-(ethoxycarbamoyl)phenylamino)-5-methylpyrrolo[2,1-f][1,2,4]triazine-6-carboxylate ([11C]10b) were prepared from their corresponding precursors with [11C]CH3OTf under basic condition through O-[11C]methylation and isolated by a simplified solid-phase extraction (SPE) method in 50–60% radiochemical yields at end of bombardment (EOB) with 185–555 GBq/μmol specific activity at end of synthesis (EOS).  相似文献

9,10-Difluoropalmitic acid (DFPA) labeled with the cyclotron produced, positron emitting radionuclide 18F has been synthesized as a potential analogue of 9,10-[3H]palmitic acid, a fatty acid which has been used to study lipid metabolism in rat brain and pituitary. [18F]DFPA was prepared by the direct and stereoselective addition of [18F]F2 to the double bond of cis-9,10-palmitoleic acid. The fluorination was carried out in FCCl3 at −70 °C using a low concentration of F2 (0.5%) in neon. [18F]DFPA has been obtained in radiochemical yields of 12–16% from end-of-bombardment (EOB) in approx. 2.5 h. Chemical and radiochemical purity exceeded 95%, and specific activities calculated to EOB ranged from 500 to 1000 mCi/mmol. [18F]DFPA crosses the blood-brain barrier and is incorporated into rat brain at about twice the level of that of 9,10-[3H]palmitic acid. The synthesis of [18F]DFPA permits us to study the biological disposition and metabolism of a vicinal-difluoro fatty acid.  相似文献

The reference standard methyl (2-amino-5-(benzylthio)thiazolo[4,5-d]pyrimidin-7-yl)-d-leucinate (5) and its precursor 2-amino-5-(benzylthio)thiazolo[4,5-d]pyrimidin-7-yl)-d-leucine (6) were synthesized from 6-amino-2-mercaptopyrimidin-4-ol and BnBr with overall chemical yield 7% in five steps and 4% in six steps, respectively. The target tracer [11C]methyl (2-amino-5-(benzylthio)thiazolo[4,5-d]pyrimidin-7-yl)-d-leucinate ([11C]5) was prepared from the acid precursor with [11C]CH3OTf through O-[11C]methylation and isolated by HPLC combined with SPE in 40–50% radiochemical yield, based on [11C]CO2 and decay corrected to end of bombardment (EOB). The radiochemical purity was >99%, and the specific activity (SA) at EOB was 370–1110 GBq/μmol with a total synthesis time of ~40-min from EOB. The radioligand depletion experiment of [11C]5 did not display specific binding to CX3CR1, and the competitive binding assay of ligand 5 found much lower CX3CR1 binding affinity.  相似文献

A method for determination of α-ketoisocaproic acid (KIC) and [4,5,5,5,6,6,6-2H7]α-ketoisocaproic acid ([2H7]KIC) in rat plasma was developed using gas chromatography–mass spectrometry-selected ion monitoring (GC–MS-SIM). [5,5,5-2H3]α-Ketoisocaproic acid ([2H3]KIC) was used as an analytical internal standard to account for losses associated with the extraction, derivatization and chromatography. The keto acids were extracted by cation-exchange chromatography using BondElut SCX cartridge and derivatized with N-phenyl-1,2-phenylenediamine to form N-phenylquinoxalinone derivatives. Quantitation was performed by SIM of the respective molecular ions at m/z 278, 281 and 285 for the derivatives of KIC, [2H3]KIC and [2H7]KIC on the electron impact method. The limit of detection was found to be 70 fmol per injection (S/N=3) and the limit of quantitation for [2H7]KIC was around 50 nM in rat plasma. Endogenous KIC concentrations in 50 μl of rat plasma were measured with relative intra- and inter-day precision of 4.0% and 3.3%, respectively. The intra- and inter-day precision for [2H7]KIC spiked to rat plasma in the range of 0.1 to 10 μM gave good reproducibility with relative standard deviation (RSD) of 6.5% and 5.4%, respectively. The intra- and inter-day relative errors (RE) for [2H7]KIC were less than 6.4% and 3.8%, respectively. The method was applied to determine the plasma concentration of [2H7]KIC after an intravenous administration of [2H7]KIC in rat.  相似文献

Abiotic stress caused by insecticide treatment is an interesting and challenging topic in plant research. Here a simultaneous extraction method with methyl tert-butyl ether for metabolomics and lipidomics analysis by using rapid resolution liquid chromatography quadrupole time-of-flight mass spectrometry was developed and validated. The extraction efficiency of lipidome and metabolome based on the current developed method showed an obvious improvement compared with literatures. About 300 metabolites were identified in rice leaf. Method validation was performed and analytical properties including the linearity (R2, 0.991–1.000), repeatability (over 87 % peaks with CV < 20 % accounting for over 92 % of total response) and recovery (74.4–119.6 %) were satisfactory. The method was then applied to investigate time-course changes caused by insecticide treatment in transgenic rice with cry1Ac and sck genes and its wild counterpart. Antioxidants including ferulic acid and sinapic acid were down-regulated at 24 h after insecticide treatment. Signaling metabolites including salicylic acid and nicotinamide adenine dinucleotide were significantly up-regulated at 12–24 h after treatment in transgenic rice. More flavonoids were significantly changed in transgenic plants. Results indicated that gene transformation affected the metabolic response of rice to insecticide stress.  相似文献

The title compound, 2-{4-[3-(2,5-dimethylphenyl)-3-methylcyclobutyl]thiazol-2-yl}isoindoline-1,3-dione (C24H22N2O2S), was synthesized and characterized by IR-NMR spectroscopy and single-crystal X-ray diffraction. The compound crystallizes in the monoclinic space group P21/c with a?=?19.7799(13) Å, b?=?6.7473(4) Å, c?=?15.7259(9) Å and β?=?103.416(5)°. In addition, the molecular geometry, vibrational frequencies and gauge including atomic orbital (GIAO) 1H and 13C chemical shift values of the title compound in the ground state have been calculated by using the Hartree-Fock (HF) and density functional method (DFT/B3LYP) with 6–31G(d), 6–31 + G(d,p) and LANL2DZ basis sets, and compared with the experimental data. To determine conformational flexibility, molecular energy profile of the title compound was obtained by semi-empirical (AM1) calculations with respect to two selected degrees of torsional freedom, which were varied from ?180° to +180° in steps of 5°. Besides, molecular electrostatic potential, frontier molecular orbitals (FMO) analysis and thermodynamic properties of the title compound were investigated by theoretical calculations.  相似文献

Repeated washes of isolated chloroplasts with dilute sodium pyrophosphate solution results in the removal of carboxydismutase and some minor proteins from the thylakoid membranes. A subsequent treatment of the membranes with hypertonic sucrose solution yields pure coupling factor CF1 in the supernatant. Purity of the protein was demonstrated by disc electrophoresis.The amount of CF1 protein liberated was quantitatively determined. The percentage of CF1 removed by this treatment was calculated from the Ca2+-dependent ATPase activity retained at the thylakoid membranes. From these data the total CF1 content of chloroplasts was calculated. An average value of 0.42 mg CF1 protein/mg chlorophyll was obtained. Based on a molecular weight for CF1 of 326000 (see Farron, F. (1970) Biochemistry 9, 3823–3828), a ratio of 1 mole CF1 per 860 moles chlorophyll was computed.  相似文献

Single domain antibody fragments (sdAbs) exhibit a rapid tumor uptake and fast blood clearance amenable for labeling with 18F (t½ = 110 min) but suffer from high kidney accumulation. Previously, we developed a method for 18F-labeling of sdAbs via trans-cyclooctene (TCO)-tetrazine (Tz) inverse electron demand Diel’s Alder cycloaddition reaction (IEDDAR) that incorporated a renal brush border enzyme (RBBE)-cleavable linker. Although >15 fold reduction in kidney activity levels was achieved, tumor uptake was compromised. Here we investigate whether replacing the [18F]AlF-NOTA moiety with [18F]fluoronicotinyl would rectify this problem. Anti-HER2 sdAb 5F7 was first derivatized with a TCO-containing agent that included the RBBE-cleavable linker GlyLys (GK) and a PEG chain, and then subjected to IEDDAR with 6-[18F]fluoronicotinyl-PEG4-methyltetrazine to provide [18F]FN-PEG4-Tz-TCO-GK-PEG4-5F7 ([18F]FN-GK-5F7). For comparisons, a control lacking GK linker and 5F7 labeled using residualizing N-succinimidyl 3-guanidinomethyl-5-[125I]iodobenzoate (iso-[125I]SGMIB) also were synthesized. Radiochemical purity, affinity (KD) and immunoreactive fraction of [18F]FN-GK-5F7 were 99%, 5.4 ± 0.7 nM and 72.5 ± 4.3%, respectively. Tumor uptake of [18F]FN-GK-5F7 in athymic mice bearing subcutaneous SKOV3 xenografts (3.7 ± 1.2% ID/g and 3.4 ± 1.0% ID/g at 1 h and 3 h, respectively) was 2- to 3-fold lower than for co-injected iso-[125I]SGMIB-5F7 (6.9 ± 1.9 %ID/g and 8.7 ± 3.0 %ID/g). However, due to its 6-fold lower kidney activity levels, tumor-to-kidney ratios for [18F]FN-GK-5F7 were 3–4 times higher than those for co-injected iso-[125I]SGMIB-5F7 as well as those observed for the 18F conjugate lacking the RBBE-cleavable linker. Micro-PET/CT imaging of [18F]FN-GK-5F7 in mice with SKOV-3 subcutaneous xenografts clearly delineated tumor as early as 1 h with minimal activity in the kidneys; however, there was considerable activity in gallbladder and intestines. Although the tumor uptake of [18F]FN-GK-5F7 was unexpectedly disappointing, incorporating an alternative RBBE-cleavable linker into this labeling strategy may ameliorate this problem.  相似文献

A keratin-degrading bacterium of Bacillus licheniformis BBE11-1 was isolated and its ker gene encoding keratinase with native signal peptide was cloned and expressed in Bacillus subtilis WB600 under the strong P HpaII promoter of the pMA0911 vector. In the 3-L fermenter, the recombinant keratinase was secreted with 323 units/mL when non-induced after 24 h at 37 °C. And then, keratinase was concentrated and purified by hydrophobic interaction chromatography using HiTrap Phenyl-Sepharose Fast Flow. The recombinant keratinase had an optimal temperature and the pH at 40 °C and 10.5, respectively, and was stable at 10–50 °C and pH 7–11.5. We found this enzyme can retained 80 % activity after treated 5 h with 1 M H2O2, it was activated by Mg2+, Co2+ and could degraded broad substrates such as degraded feather, bovine serum albumin, casein, gelatin, the keratinase was considered to be a serine protease. Coordinate with Savinase, the keratinase could efficient prevent shrinkage and eliminate fibres of wool, which showed its potential in textile industries and detergent industries.  相似文献
