The patterns of serum luteinizing hormone (LH), follicle stimulating hormone (FSH), progesterone and estradiol-17β during the estrous cycle of six crossbred (Alpine × Nubian × Native) and six native goats showing a 21 day estrous cycle in a semiarid zone of Venezuela are presented. In the crossbred goats, FSH had two significant peaks on Days 19 and 0 (33 ± 8.6 ng ml−1 and 25 ± 6 ng ml−1, respectively); in contrast, native goats only had one significant peak on the day of estrus (22 ± 2 ng ml−1), with the increase beginning on Day 17. During the follicular phase of crossbred goats, estradiol-17β and LH increased to 28 ± 6 pg ml−1 and 23 ± 6.9 ng ml−1, respectively, on Day 0. Prior to Day 0, LH increased to 10.0 ± 4.9 ng ml−1 on Day 18, decreasing to 1.5 ng ml−1 on Day 19, while estradiol-17β was increasing. This relationship between estradiol-17β and LH was not found to exist in native does, which presented a LH peak on Day 0 (30 ± 8 ng ml−1 and 35 ± 10 ng ml−1 in first and second estrus, respectively). LH basal levels were notably higher in native does. The highest concentrations of progesterone (10 and 12 ng ml−1) were detected on Days 12 and 15 in crossbred and native females, respectively. In conclusion, the relationship between estradiol-17β and gonadotropins during the follicular phase in crossbred goats suggests negative and positive feedback effects on both LH and FSH. Serum concentrations of LH were higher in native than in crossbred goats, whereas concentrations of FSH were higher in crossbred does. Thus, genetic factors need to be taken into account when comparing blood levels of gonadotropins in goats raised in tropical semiarid zones.  相似文献

The objective was to determine the effect of gonadotrophin-releasing hormone (GnRH), GnRH analogue (GnRH-A) or oestradiol administration on luteinising hormone (LH) and follicle-stimulating hormone (FSH) release in GnRH-immunised anoestrous and control cyclic heifers. Thirty-two heifers (477 ± 7.1 kg) were immunised against either human serum albumin (HSA; controls; n = 8), or a HSAGnRH conjugate. On day 70 after primary immunisation, control heifers (n = 4 per treatment; day 3 of cycle) received either (a) 2.5 μg GnRH or (b) 2.5 μg of GnRH-A (Buserelin®) and GnRH-immunised heifers (blocked by GnRH antibody titre; n = 6 per treatment) received either (c) saline, (d) 2.5 μg GnRH, (e) 25 μg GnRH or (f) 2.5 μg GnRH-A, intravenously. On day 105, 1 mg oestradiol was injected (intramuscularly) into control (n = 6) and GnRH-immunised anoestrous heifers with either low (13.4 ± 1.9% binding at 1:640; n = 6) or high GnRH antibody titres (33.4 ± 4.8% binding; n = 6). Data were analysed by ANOVA. Mean plasma LH and FSH concentrations on day 69 were higher (P < 0.05) in control than in GnRH-immunised heifers (3.1 ± 0.16 vs. 2.5 ± 0.12 ng LH ml−1 and 22.5 ± 0.73 vs. 17.1 ± 0.64 ng FSH ml−1, respectively). The number of LH pulses was higher (P < 0.05) in control than in GnRH-immunised heifers on day 69 (3.4 ± 0.45 and 1.0 ± 0.26 pulses per 6 h, respectively). On day 70, 2.5 μg GnRH increased (P < 0.05) LH concentrations in control but not in GnRH-immunised heifers, while both 25 μg GnRH and 2.5 μg GnRH-A increased (P < 0.05) LH concentrations in GnRH-immunised heifers, and 2.5 μg GnRH-A increased LH in controls. FSH was increased (P < 0.05) in GnRH-immunised heifers following 25 μg GnRH and 2.5 μg GnRH-A. Oestradiol challenge increased (P < 0.05) LH concentrations during the 13–24 h period after challenge with a greater (P < 0.05) increase in control than in GnRH-immunised heifers. FSH concentrations were decreased (P < 0.05) for at least 30 h after oestradiol challenge. In conclusion, GnRH immunisation decreased LH pulsatility and mean LH and FSH concentrations. GnRH antibodies neutralised low doses of GnRH (2.5 μg), but not high doses of GnRH (25 μg) and GnRH-A (2.5 μg). GnRH immunisation decreased the rise in LH concentrations following oestradiol challenge.  相似文献

The involvement of endogenous opioids in modulation of prolactin (PRL) secretion during pregnancy in the pig was studied. Twenty-four crossbred pregnant gilts (150 ± 10 kg) were cannulated via the cephalic vein 24–48 h before treatment with 1 mg kg−1 body weight of naloxone (NAL) or 3 ml of saline (CONT) i.v. at Day 40 (NAL, n = 6; CONT, n = 6) or Day 70 (NAL, n = 6; CONT, n = 6) of pregnancy. Blood plasma was collected at 15 min intervals from 1 h before to 3 h after treatment with NAL or saline. At Day 40 of pregnancy, administration of NAL caused a decrease in mean plasma PRL concentrations at 60 min, 120 min and 180 min post-treatment (NAL, 19.1 ± 1.3 ng ml−1, P < 0.05; 15.8 ± 0.6 ng ml−1, P < 0.001; 14.6 ± 0.7 ng ml−1, P < 0.001, respectively) when compared with the CONT group (22.9 ± 0.7 ng ml−1, 21.6 ± 0.6 ng ml−1 and 22.4 ± 0.5 ng ml−1, respectively). Mean plasma estradiol concentration was higher (P < 0.01) in the NAL group during the second and third hour post-treatment than in the CONT group. At Day 70 of pregnancy, infusion of NAL also decreased (P < 0.001) plasma PRL concentrations at 60 min, 120 min and 180 min after treatment (20.1 ± 1.6 ng ml−1, 16.2 ± 1.5 ng ml−1 and 14.8 ± 0.4 ng ml−1, respectively) compared with the CONT group (33.4 ± 1.7 ng ml−1, 34.1 ± 1.3 ng ml−1 and 29.1 ± 0.9 ng ml−1, respectively). Estradiol concentrations were not different (P > 0.05) between groups in this stage of gestation. Mean concentrations of progesterone were similar during the pre- and post-treatment periods in both stages of pregnancy.These data would suggest a possible role of the opioids in modulation of PRL secretion at these stages of pregnancy in the pig.  相似文献

Gonadotropin-releasing hormone (GnRH) has been found to be expressed within the ovary and to modulate cell differentiation in ovarian cells. In the present study we have analyzed the influence of GnRH on DNA synthesis in rat granulosa cells. Cells were obtained from immature DES-treated rats and cultured in defined medium (DMEM:F12) containing combinations of FSH, estradiol, and transforming growth factor-β (TGF-β), both in the presence and absence of GnRH. A GnRH analog, Leuprolide (GnRHa), caused a dose-dependent inhibition of 3H-thymidine incorporation in cells cultured in the presence of FSH (20 ng/ml) and TGFβ (2.5 ng/ml), at concentrations as low as 5 × 10−11 M. Similarly, a complete inhibition of hormonally stimulated DNA synthesis were observed with another analog (Buserelin, ED50 = 1.58 ± 0.22 × 10−10 M) and native GnRH (ED50 = 1.4 ± 0.3 × 10−6 M). A competitive antagonist of GnRH (Antide) was used to neutralize the GnRH agonist effects. Antide 10−8 M could prevent the inhibition elicited by 10−7 M of Leuprolide. These results suggest that GnRH may play a role in the regulation of rat granulosa cell proliferation during follicular development. Mol. Reprod. Recently, cadmium has been described to disturb ovarian function in rats. In this paper the direct influence of cadmium on steroid production of ovarian cellsin vitro has been studied. Granulosa and luteal cells were obtained from proestrous and pregnant rats, and incubated with 0, 5, 10, 20 or 40 g ml–1 CdCl2 in the presence or absence of 0.1–1000 ng ml–1 follicle stimulating hormone (FSH) or luteinizing hormone (LH) for 24 or 48 h. Production of progesterone (P) and 17-estradiol (E2) by granulosa and that of P by luteal cells were measured by radioimmunoassay. In FSH-stimulated granulosa cell cultures, 5 and 40 g ml–1 CdCl2 suppressed P accumulation to 65 and 10%, respectively; accumulation of E2 (at 5 g ml–1 CdCl2) decreased to 44%. P production of LH-supported luteal cells dropped to 86 and 66%, respectively, when 5 and 40 g ml–1 CdCl2 was added to the medium. No alteration in basal P accumulation occurred in granulosa and luteal cell cultures following incubations with 20 and 40 g ml–1 CdCl2, whereas basal E2 production of granulosa cells was markedly diminished. It is concluded that CdCl2 suppressing steroid synthesisin vitro exerts a direct influence on granulosa and luteal cell function.  相似文献

In the first of three experiments, eight ovariectomised Greyface ewes primed with exogenous progesterone were used to provide quantitative data on the effects of two contrasting feeding levels (0.3 vs. 1.4 × maintenance) on plasma progesterone concentrations. Over the 9 day study period, mean (± SEM) daily progesterone concentrations were 4.3 ± 0.13 and 3.3 ± 0.17 μg l−1 for the low and high feeding regimens, respectively (P = 0.06), indicating that high feed intake suppressed circulating progesterone levels. The second experiment examined the effect in superovulated Finn-Dorset ewes of a diet supplying either 0.6 (Group L, n = 8) or 2.3 (Group H, n = 8) times their daily energy needs for maintenance, from 1 day before introduction of exogenous progesterone to the time of insemination, on plasma progesterone concentrations and the viability of ova recovered 4 days after insemination. Mean (± SEM) plasma progesterone concentrations were 4.5 ± 0.17 μg l−1 and 2.8 ± 0.16 μg l−1 for L and H ewes, respectively, during the 12 day priming period (P < 0.001). Eight hours after progesterone withdrawal, levels had fallen to 0.9 ± 0.06 μg l−1 and 0.8 ± 0.07 μg l−1, respectively, then rose to 17.8 ± 3.01 μg l−1 and 12.9 ± 2.50 μg l−1 (P > 0.10) at ovum collection. Intervals (mean ± SEM) to oestrous onset (14.5 ± 0.38 h) and the luteinising hormone (LH) surge (27.1 ± 0.98 h) were unaffected by feed intake. Mean (± SEM) ovulation rates (8.1 ± 1.57 vs. 7.8 ± 1.10) and numbers of ova recovered (5.0 ± 1.39 vs. 4.8 ± 1.11) were also similar for each group. However, the proportions of ova considered viable (over 32 cells) at recovery were 0.53 and 0.22 for L and H groups, respectively (P < 0.005). Following 72 h culture (Tissue Culture Medium-199 (M199) + 10% foetal calf serum (FCS)), 0.55 and 0.27, respectively, had developed to blastocysts (P < 0.025). Of ova assessed as viable at recovery, similar proportions (0.86 vs. 0.75) from L and H treatments developed to blastocysts, with corresponding nuclei counts (mean ± SEM) of 55 ± 5.2 and 55 ± 13.2. The third experiment used 12 superovulated Greyface ewes, each offered a different feed level within the range 0.6–2.5 × maintenance, to determine the nature of the relationship between feeding level, pre-ovulatory progesterone concentrations and ovum development at Day 2 following insemination and subsequently during 7 day co-culture (M199 + FCS). Increases in feeding level were accompanied by linear decreases in plasma progesterone (r2 = 0.79, P < 0.001), the interval to oestrous onset (r2 = 0.52, P < 0.01) and timing of the LH surge (r2 = 0.32, P < 0.06). Although undetectable at ovum collection, and somewhat equivocal after 4 day culture, high feeding levels prior to ovulation reduced the proportion of ova (0.16 vs. 0.58) developing to or beyond the expanding blastocyst stage after 7 day culture. Quantitative indices of cell division and protein synthesis confirmed this. In conclusion, excessive feeding during follicular recruitment and oocyte maturation in superovulated ewes imparts a legacy of embryonic loss and developmental retardation.  相似文献

Feeding rates of Brachionus plicatilis were studied for two types of food — algae Monochrysis lutheri and baker's yeast Saccharomyces cerevisae. The main regularities of changes in filtration rate and ration were studied in small culture volumes (1 ml) for adult amictic females depending on food concentration (1, 2, 4, 8 and 16 · 106 cells · ml−1), ambient temperature (16 and 26 °C), and salinity (5, 10, 15, 20, 25 and 30 ppt). B. plicatilis ration did not depend on the salinity, but was largely determined by temperature and food concentration. It was found that at 16 and 26 °C the dependence of the ingestion rate (ration) on food concentration differed greatly. A hypothesis was suggested to explain this phenomenon. A critical concentration of both types of food at which the increase in the rotifer ration ceased is 4 · 106 cells · ml−1. This is the minimum “background” food concentration for B. plicatilis mass cultivation. The average rations measured at the concentration of M. lutheri and S. cerevisae of 4 · 106 cells · ml−1 where 1.3 ± 0.1 and 4.8 ± 1.3 μg dry weight. · ind−1 · day−1 at 26 °C and 0.54 ± 0.1 and 1.9 μg d. w. · ind−1 · day−1 at 16 °C, respectively. The rations obtained in the laboratory were corrected for the conditions of rotifer commercial production in the open field in summer time. The correct values were 0.86 and 0.72 μg d. w. · ind−1 · day−1 for algae and yeast, respectively.  相似文献

An electro-transformation procedure was established for Bacillus cereus ATCC 14579. Using early growth-stage culture and high electric field, the ectroporation efficiency was up to 2 × 109 cfu μg 1 ml 1 with pC194 plasmid DNA. The procedure was tested with three other plasmids, of various sizes, replication mechanisms and selection markers, and the transformation efficiencies ranged between 2 × 106 and 1 × 108 cfu μg 1 ml 1. The effects of two wall-weakening agents on electroporation rates were also evaluated. The transformation rate that was reached with our procedure is 103 times higher than that previously obtained with members of the Bacillus genus with similar plasmids, and 106 times superior than that achieved with available protocols for B. cereus. The proposed method is quick, simple, efficient with small rolling circle plasmids and large theta replicating plasmids with low copy number per cell, and suitable for many genetic manipulations that are not possible without high-efficiency transformation protocols.  相似文献

This study was undertaken to characterize the relationship between changes in steroid production, cell cycle activity (ie, cell proliferation) and apoptosis in antral and mural bovine granulosa cells cultured in vitro. This was done to select conditions promoting optimal estradiol production by bovine granulosa cells cultured in completely defined conditions. In the first experiment, antral granulosa cells were cultured over the entire 4 days of the culture period in the presence of either 0, 2, or 10 ng/ml of FSH (chronic conditions) or were maintained under minimal FSH support (0.5 ng/ml FSH) for the first 3 days of culture and then were challenged over the fourth day of culture with either 0, 2, or 10 ng/ml FSH (challenged conditions). Compared with cells exposed to constant FSH levels (chronic conditions), the FSH-induced production of estradiol was higher (P < 0.006) and that of progesterone was lower (P < 0.02) over the last 24 h of culture, when antral granulosa cells were maintained under minimal FSH support during the first 3 days of culture (challenged conditions). In the second experiment, dynamics of estradiol and progesterone productions, conversion of [14C]androstenedione into subsequent steroid metabolites, DNA content, cell cycle activity, and apoptosis (as assessed by flow cytometry) of antral and mural granulosa cells over the first 3 days of culture under minimal FSH support and in response to a challenge with FSH during the last 24 h of culture were evaluated. Estradiol production as well as the conversion of androstenedione into testosterone and estradiol were greater (P< 0.01) in antral than in mural granulosa cells cultured under challenged conditions. A higher proportion of mural than antral granulosa cells were in the proliferative state at the end of culture (P < 0.03). This may be related to the decreased ability of mural cells to produce estradiol. FSH suppressed (P < 0.05) the spontaneous onset of apoptosis in both cell types. These results suggest that functional differences between these two cell compartments need to be considered in studying bovine granulosa cells in vitro. Because of their large (400 to 600%) FSH-induced estradiol production, antral granulosa cells cultured under challenged conditions provide a model that can be used to examine substances for their ability to alter estradiol production and apoptosis in bovine granulosa cells. Chroococcoid cyanobacteria, (mean size = 0.79 μm, likely Synchetocystis limnetica Popovsk) and total eubacteria (mean size = 0.33 μm), from Lake Baikal, USSR, were enumerated using epifluorescence microscopy and sized with image analysis. Bacterial densities ranged from 0.44 · 106 cells ml−1 at 250 m to 2.3 · 106 cells ml−1 at the surface. Mean eubacterial abundance was 1.3 · 106 cells ml−1. Cyanobacterial densities were more variable, ranging from 0.42 · 104 cells ml−1 at 250 m to 9.8 · 104 cells ml−1 at the surface, with a mean abundance of 2.7 · 104 cells ml−1. The cyanobacteria, in particular, occurred in clusters resembling “marine snow”. Our results indicate that Lake Baikal picoplankton size and density are similar to other large lakes but may have a more diverse community structure than in other large oligotrophic lakes.  相似文献

The ability of the unicellular rhodophyte Porphyridium cruentum to grow mixotrophically on the soluble fraction of Solarium tuberosum meal was tested. At the beginning of stationary phase Porphyridium cruentum produced 7 μg ml−1 of phycoerythrin and 129 μg ml−1 of total soluble exopolysaccharide when cultured autotrophically. When cultured mixotrophically with the soluble fraction of Solanum tuberosum meal, the productivity increased to 10 μg ml−1 of phycoerythrin and 330 μg ml−1 of total soluble exopolysaccharide. When the soluble fraction of S. tuberosum meal was supplied together with nitrate and phosphate, the productivity of phycoerythrin increased to 21 μg ml−1 while the production of total soluble exopolysaccharide decreased to 195 μg ml−1. Results demonstrate that the soluble fraction of S. tuberosum meal can be used as substrate for the production of phycoerythrin and exopolysaccharide by P. cruentum improving the results obtained with the autotrophic culture medium.  相似文献

We have evaluated the possibility that mouse oocyte growth in vitro could be achieved under the influence of soluble compound(s) released by different somatic cell types. For this purpose, zona-free denuded oocytes from 12-day-old mice were cultured on monolayers of NIH-3T3 fibroblasts, which are able to establish gap junctional communications with them, in the presence or absence of media conditioned by preantral granulosa cells or by Sertoli cells, plated at increasing concentrations from 0.3–1 × 106 ml−1 cells. After 3 days, no increase in vitellus diameter was recorded from fibroblast-coupled oocytes maintained in culture medium or in the presence of media conditioned by 0.3 × 106 ml−1 Sertoli cells. By contrast, increasing proportions of coupled oocytes grew, provided the continuous presence of media conditioned by 0.5 or 1 × 106 ml−1 Sertoli cells, or by 0.3, 0.5, and 1 × 106 ml−1 preantral granulosa cells. Since the ligand of c-kit, the growth factor KL, promotes the growth in vitro of oocytes cultured in follicles from 8-day-old mice, an antibody against mouse KL was used to evaluate whether in our culture conditions KL might also be responsible for the growth of oocytes from 12-day-old mice. No inhibition of growth was evident in oocytes cultured directly on preantral granulosa or Sertoli-cell monolayers. Furthermore, the growth of fibroblast-coupled oocytes cultured in media conditioned by preantral granulosa cells was not significantly affected by the presence of this antibody during culture. By contrast, a high percentage of oocytes cultured on fibroblasts in the presence of media conditioned by Sertoli cells showed a significant inhibition of growth and no metabolic cooperativity. It was concluded that, besides KL, other bioactive factor(s) released by either preantral granulosa or Sertoli cells can induce a significant stimulation of mouse oocyte growth in vitro. © 1996 Wiley-Liss, Inc.  相似文献

When BmN-4 and M-BmN cells were grown in shake flasks, the pH initially dropped and later increased. The increase in pH signaled a ‘metabolic switch’ that was used here as an indicator for initiating a supplemental glucose and glutamine feed. Using the pH-based fed-batch culture method described, the maximum cell densities of BmN-4 cells and M-BmN cells were increased from 30×105 cells ml−1 to 43×105 and 52×105 cells ml−1, respectively. Correspondingly, the production of polyhedra (4·5×105 OBs ml−1) and HBsAg (574 ng ml−1), from the infection of BmN-4 and M-BmN by wild-type and recombinant BmNPV viruses, respectively, were both significantly enhanced 50% and 100%, respectively. This feeding strategy was implemented with no advanced instrumentation yet facilitated significantly increased yield in shake flasks. The technique should benefit those in research laboratories employing the baculovirus expression system as a rapid and efficient production system.  相似文献

Porcine granulosa cells have been shown previously to both secrete and respond to insulin-like growth factor-I (IGF-I), suggesting an autocrine function of this peptide in the follicle. The present work was undertaken to determine possible effects of IGF-I on in vitro maturation, in vitro fertilization, and early embryonic development in culture. Granulosa and cumulus cell proliferation and differentiation based on 3H-thymidine uptake and progesterone production, respectively, were also assessed. The results showed that the cleavage rate of oocytes was markedly stimulated in a dose-dependent manner by the addition of IGF-I to the oocyte maturation medium (P < 0.05). Embryo development beyond the 8-cell stage was improved by IGF-I, reaching a maximum of 22% at 200 ng/ml IGF-I. Treatment with IGF-I after fertilization increased the percentage of total oocyte cleavage (P < 0.05) to approximately 52%, 43%, and 57% at, respectively, 25, 50, and 100 ng/ml IGF-I. 3H-thymidine incorporation by granulosa cells was significantly increased in cultures treated with FSH (3-fold) or IGF-I (6-fold) compared to the control. For the cumulus cells, FSH caused a similar increase (3-fold) in 3H-thymidine incorporation while IGF-I stimulated a 15-fold increase. Progesterone production by the granulosa cells was increased to the same extent by treatment with FSH or IGF-I (4.7 and 5.1-fold, respectively). However, for the cumulus cells, while FSH caused a marked 16-fold increase in progesterone production, IGF-I caused only a marginal increase of 2.5-fold. These results indicate a beneficial effect of IGF-I on in vitro porcine oocyte maturation and pre-implantation embryo development, suggesting a physiological role for IGF-I in vivo. The in vivo effect of IGF-I may be indirect via autocrine stimulation of cumulus and/or granulosa cells resulting in enhanced oocyte maturation and fertilization. © 1994 Wiley-Liss, Inc.  相似文献

Experiments were carried out to investigate the effects of varying physiological concentrations (0, 10, 100, and 1000 ng ml−1) of oestradiol or testosterone on [3H]-thymidine incorporation by porcine granulosa and theca cells in vitro. Granulosa cells only were recovered from small (1–3-mm) follicles and both granulosa and theca cells recovered from large (4–8-mm) porcine follicles. Cells were cultured for 72 h in medium containing 10% foetal calf serum, 24 h in serum-free medium, and finally 40 h in serum-free medium containing [3H]-thymidine and appropriate steroid treatment. Although DNA per well was significantly higher (P < 0.05) at the end of culture in the theca cells than in the granulosa cells, neither steroid treatment had a significant (P > 0.1) effect on DNA concentration in either cell type. Overall, cells from small follicles incorporated significantly (P < 0.01) more [3H]-thymidine than those from medium follicles. Both oestradiol and testosterone significantly (P < 0.01) inhibited thymidine incorporation by cells from both follicle size categories, with a significant (P < 0.05) hormone × dose interaction. Finally, there was a highly significant (P < 0.001) interaction between the response of cells to different hormone concentrations and the follicle size from which they were recovered. These results indicate that both oestradiol and testosterone may act in an autocrine/paracrine manner to inhibit proliferation and encourage differentiation in follicular cells and thus are likely regulators of the later stage of antral follicle development in the pig.  相似文献

Four size classes of both sexes of laboratory-cultured Streptocephalus proboscideus (post-metanauplii 4.7±0.4; juvenile virgins 8.7±0.7; adults I 13.8±0.9, and adults II 22.07±1.1 mm) were fed five concentrations (20 to 320 ml−1) of Anuraeopsis fissa, or six concentrations (20 to 640 ml−1) in adults I and adults II, for 30 minutes. Post-metanauplii consumed at maximum 66±9 rotifers ind.−1 min.−1 (mean±S.D.) while the largest adult females maximally ingested 347±37 rotifers min.−1. Regardless of predator size and sex, prey consumption was dependent on prey density. Functional response curves either plateaued or declined at 320 prey ml−1 in post-metanauplii, juveniles and adults I, and at 640 ml−1 in adults II. Females consumed c. 40% more prey than males. On a daily basis, adult II females consumed up to 1.05 mg rotifer dry weight (10% of their own body weight) while post-metanauplii consumed up to 0.2 mg DW (100% of their body weight). Intermediate stages had intermediate consumption rates. Filtration rates indicated that a fully grown S. proboscideus may filter as much as 2 1 of water per day, suggesting that fairy shrimps, in their natural environment, may often be food-limited.  相似文献

We have recently demonstrated that gonadotropin releasing hormone (GnRH) acts directly on ovarian granulosa cells to inhibit the follicle stimulating hormone (FSH)-induced increase in granulosa cell steroidogenesis invitro. A GnRH antagonist, [D-pGlu1, D-Phe2, D-Trp3,6] GnRH (A), which is known to antagonize GnRH-stimulated gonadotropin release by cultured pituitary cells, was tested in the granulosa cell system. GnRH (10?8M) inhibited estrogen and progesterone production by FSH-treated granulosa cells invitro, whereas the antagonist A (10?6M) did not affect FSH stimulation of steroidogenesis. Antagonist A, when added together with GnRH and FSH, blocked the GnRH inhibition of FSH-induced steroidogenesis. Estrogen and progesterone production by granulosa cells was increased by 50% at a molar ratio (IDR50) of 201and121 ([antagonist]/[GnRH]), respectively. At 10?6M, antagonist A completely prevented the GnRH (10?8M) inhibition. A similar effect of antagonist A was seen in FSH-induced increase of luteinizing hormone (LH) receptor content. FSH treatment for 2 days invitro induced an 8-fold increase in LH receptor content in cultured granulosa cells; concomitant treatment with 10?8M GnRH completely inhibited the FSH effect. Antagonist A (10?6M), by itself, had no effect on the FSH action. However, when added together with FSH and GnRH, antagonist A completely abolished the inhibitory effect of GnRH. These results demonstrate that the direct inhibitory effect of GnRH on granulosa cell function can be prevented by a GnRH antagonist and that the GnRH action at the ovarian level may require stringent stereospecific interactions of these peptides with putative GnRH recognition sites.  相似文献

This study examined the competence of oocytes from the tammar wallaby, Macropus eugeniio mature in vitro. Oocytes were collected from follicles >1 mm diameter 24 h after pregnant mare serum gonadotrophin (PMSG) treatment and incubated in Eagle's minimum essential medium supplemented with 10% fetal calf serum, at 35°C in 5% CO2 in air for 24, 36, or 48 h. Oocytes were incubated either granulosa cell-intact or granulosa cell-free or in the presence of 10 IU ml?1 PMSG or 10 μg ml?1 porcine luteinizing hormone (LH) + 10 μg ml?1 porcine follicle stimulating hormone (FSH). The ability of oocytes recovered from small (<1.5-mm-diameter) and large (≥1.5-mm-diameter) follicles to mature in vitro was also examined. The nuclear status of oocytes was assessed using the DNA-specific dye Hoechst 33342. Initially, all oocytes examined contained a germinal vesicle. After 24 h of culture, 60% of oocytes had progressed to metaphase I or anaphase I. After 36 h, approximately 20% of oocytes possessed metaphase II chromosomes, and 20% of oocytes were at metaphase I or anaphase I. At the completion of the 48 h culture period, 40% of oocytes had completed maturation to the metaphase II stage. In vitro oocyte maturation after 48 h was not affected by the presence of granulosa cells, PMSG, or LH and FSH. More oocytes from large follicles (55%) completed maturation by 48 h than from small follicles (20%). Approximately 50% of oocytes remained at the GV stage at all times under all conditions. Marsupial oocytes thus undergo spontaneous nuclear maturation once removed from the follicular environment, suggesting a basically similar control system to that in placental mammals. © 1993 Wiley-Liss, Inc.  相似文献
