The circadian movement of the lamina of primary leaves of Phaseolus coccineus L. is mediated by antagonistic changes in the length of the extensor and flexor cells of the laminar pulvinus. The cortex of the pulvinus is a concentric structure composed of hexagonal disc-like cells, arranged in longitudinal rows around the central stele. Observations with polarization optics indicate that the cellulose microfibrils are oriented in a hoop-like fashion in the longitudinal walls of the motor cells. This micellation is the structural basis of the anisotropic properties of the cells: tangential sections of the extensor and flexor placed in hypotonic mannitol solutions showed changes only in length. As a consequence a linear correlation between length and volume was found in these sections. Based on the relationship between the water potential (which is changed by different concentrations of mannitol) and the relative volume of the sections and on the osmotic pressure at 50% incipient plasmolysis, osmotic diagrams were constructed for extensor and flexor tissues (cut during night position of the pulvinus). The bulk moduli of extensibility, , were estimated from these diagrams. Under physiological conditions the values were rather low (in extensor tissue below 10 bar, in flexor tissue between 10 to 15 bar), indicating a high extensibility of the longitudinal walls of the motor cells. They are strongly dependent on the turgor pressure at the limits of the physiological pressure range.In well-watered plants, the water potentials of the extensor and flexor tissues were surprisingly low,-12 bar and-8 bar, respectively. This means that the cells in situ are by no means fully turgid. On the contrary, the cell volume in situ is similar to the volume at the point of incipient plasmolysis: the cell volumes of extensor and flexor cells in situ were only 1.01 times and 1.1 times larger, respectively, than at the point of incipient plasmolysis, whereas at full turgidity (cells in water) the corresponding factors were 1.8 and 1.5. Protoplasts isolated from the laminar pulvinus of Phaseolus vulgaris and bathed in a medium containing KCl as the major salt were found to swell in response to IAA and to shrink in response to ABA. The protoplasts of flexor cells and those of extensor cells responded similarly. The results indicate that the cellular content of osmotic solutes is enhanced by IAA and reduced by ABA. The IAA-induced swelling was abolished when either the K(+) or the Cl(-) of the bathing medium was replaced by an impermeant ion or when the medium was adjusted to neutral pH (instead of pH 6). The response was inhibited by vanadate. It is concluded that the swelling is caused by enhanced influxes of K(+) and Cl(-), which probably occur through K(+) channels and Cl(-)/H(+) symporters, respectively. The ABA-induced shrinking was inhibited by 5-nitro-2-(3-phenylpropylamino)-benzoic acid, an anion-channel inhibitor, suggesting that it is caused by Cl(-) efflux through anion channels and charge-balancing K(+) efflux through outward-rectifying K(+) channels. It appears that the two plant hormones act on pulvinar motor cells to regulate their turgor pressure, as they do in stomatal guard cells. The findings are discussed in relation to the pulvinar movements induced by environmental stimuli.  相似文献

Measurements with microelectrodes implanted into Samanea saman (Jacq.) Merrill leaf pulvini showed that membrane potentials were rhythmically sensitive to the application of sucrose. The magnitude of the electrical depolarizations induced by sucrose were dependent on the concentration of H+ in the medium, yet changes in [H+] alone did not greatly affect the potential. During sucrose-induced electrical depolarization, there was a slight increase in the pH of the bathing medium; both effects were abolished by high levels of K+, Na+ or Ca2+ in the medium. These observations indicate that H+ enter the cells by some cooperative action with sucrose. A model of H+-substrate cotransport is proposed in which a sugar carrier in the membrane is made more permeable by the attachment of a proton. The rhythmic nature of this proposed cotransport may be related to circadian leaf-movements in this plant.  相似文献

The internal potential of flexor cells in slices of the laminar pulvini of Phaseolus coccineus has been recorded by standard microelectrode techniques in 100 eq m-3 external salt solutions of various ionic compositions. The measured values are between-15 and-60 mV depending on the external medium. Treating the results with the Goldman equation yields the following relative permeabilities: K+, 1.00; Na+, 0.24; Cl-, 0.19; NO 3 - , 1.6. The membrane potential was only slightly sensitive to external pH and Ca2+. Metabolic inhibitors (azide, cyanide and salicylhydroxamic acid, carbonyl cyanid m-chlorphenyl hydrazone) caused only slight depolarizations (ca. 4 mV), which differed from the ion-induced changes by their slow time courses. The results are consistent with the hypothesis that the relatively impermeable Cl- is actively transported and osmotically efficient, whereas the well-permeable K+ passively follows Cl- to maintain electroneutrality and is osmotically of only minor significance.Abbreviations SHAM salicylhydroxamic acid - CCCP carbonyl cyanid m-chlorphenyl hydrazone  相似文献

The objective of this study was to evaluate ether extract (EE) concentrations, pigments, and wax in forages (n = 14), concentrates (n = 23), and cattle feces (n = 100) using extraction methods recommended by Association of Official Analytical Chemists (AOAC; method 920.39) and American Oil Chemist's Society (AOCS; method Am 5-04). The EE contents were compared by adjusting a linear regression model for each sample type. For the feces and forage samples, the EE contents produced by the AOCS method were greater (P<0.05) compared to those obtained using the AOAC method. No differences between methods were observed in EE content of concentrates (P>0.05). Concentration of vegetable pigments and wax were evaluated by using analysis for variance. Vegetable pigments were lower (P<0.05) in the post-extraction residues using the AOCS method, than the AOAC method, indicating greater participation of vegetable pigments in the EE. No differences were observed between the methods in wax concentration of the post-extraction residues (P>0.05). The quantification method of the EE content that is recommended by AOCS is not suggested for analyses of forage and feces of ruminants because it possibly increases the removal of non-fatty material, mostly pigments, in comparison to the method recommended by AOAC.  相似文献

The activities of enzymes which catalyze one step in each of the five major carbon pathways in green plants were measured in secondary pulvini and other tissues of Phaseolus coccineus L. leaves. We were able to detect activities of fumarase (EC 4.2.1.2; tricarboxylic-acid pathway), NAD-glyceraldehyde-phosphate dehydrogenase (NAD-GAPDH, EC 1.2.1.12; glycolysis), 6-phosphogluconate dehydrogenase (6-PGDH, EC 1.1.1.44; oxidative pentose-phosphate pathway), ribulose-1, 5-bisphosphate carboxylase (Rubisco, EC 4.1.1.39; photosynthetic carbon-reduction pathway), and of hydroxypyruvate reductase (HP-R, EC 1.1.1.81; photosynthetic carbon-oxidation pathway). On a protein basis the activities of Rubisco and HP-R in pulvinar regions were very low (below 1 and 2 mol · (kg protein) –-1 · h–-1, respectively), but the activities of fumarase and NAD-GAPDH were between 10- and 5-fold higher compared with the laminar tissue (up to 7 and 50 mol · (kg protein)–-1 · h–-1, respectively). Similarly, the protein specific activities of 6-PGDH were increased in the pulvinus (3–4 compared with approx. 1 mol · (kg protein)–-1 · h–-1 in the leaf blade). No differences in specific activities were detected between day and night positions of the leaves. By applying quantitative histochemical techniques we determined the longitudinal and transversal compartmentation of the activities of fumarase, NAD-GAPDH, and 6-PGDH in pulvinar tissues. Levels of activity of all three enzymes increased towards the middle part of the pulvinus. Here, expressed on a dry-weight (DW) basis, the analysis of cross sections showed highest activities in the outer parts of the extensor in the order given, approx. 0.6, 5, and 0.25 mol · (kg DW)–-1 · h–-1 for fumarase, NAD-GAPDH and 6-PGDH. When related to protein, levels of activity were comparably high within the inner parts of extensor and flexor, and partly also in the abaxial part of the bundle (fumarase, 6-PGDH). Hemoglobin variants are the protein mutations most often encountered in the clinical scene. They have been useful for developing methods to analyze mutant proteins because of their size and ease of collection in large amounts. Improvements in analytical methods have been directed toward higher resolution in electrophoresis and shorter elution times in chromatography. More importantly, in the last 20 years, hemoglobin variants have been used in the development of mass spectrometric strategies for analyzing protein mutations. This approach consists of a series of steps: measurement of the molecular mass of globins to detect or confirm the presence of mutations, peptide mass mapping or peptide mass fingerprinting of an enzymatic digest to identify mutated peptides, and tandem mass spectrometry to determine or confirm the site and type of mutation. The mass spectrometric strategy has enabled rapid analysis and demonstrated a superb ability to detect a number of hemoglobin variants, particularly those without a change in electrophoretic or chromatographic properties. Even with the recent advances in DNA analysis, protein analysis is still essential, because post-translational modifications following amino acid substitutions can occur including N-terminal acetylation, deamidation and oxidation-mediated processes.  相似文献

Quantitative histochemistry was used to investigate the tissue-specific compartmentation of soluble carbohydrates (sucrose, glucose, fructose), starch and malate in the laminar pulvinus, leaf blade and petiole of Phaselous coccineus L. at day and night positions of diurnal leaf movement. Total carbohydrate levels measured in a series of cross sections along individual pulvini of 24-d-old plants showed only small differences between the day and night positions of the respective leaf. In contrast, the level of malate changed during diurnal leaf movement, especially in the central part of a pulvinus. The levels of glucose and fructose in the pulvinus increased towards the transition zones between the pulvinus and lamina, and pulvinus and petiole, and this trend was even more pronounced for starch. By contrast, sucrose levels were highest in the pulvinus proper. The transverse compartmentation of metabolites was studied in distinct, approx. 0.5-mm-thick tissue slices from the central part of a pulvinus. These were dissected further into up to 14 distinct subsamples (bundle, bundle sheath, motor tissues, flanks). Irrespective of the position of the leaf (day or night), the central vascular core and the surrounding bundle sheath had high levels of sucrose (up to 500 mmol-(kg DW)–1) and low levels of glucose and fructose (below 100 mmol-(kg DW)–1), while in the cortex the situation was reversed. In the night position the level of sucrose decreased by approx. 30% in the bundle sheath and the central vascular core but not in the other sections. We thus suggest that because of the relatively small diurnal changes in their cortical pools, soluble sugars are not involved in the osmotic processes resulting in leaf movement. In contrast, pulvini from 14-d-old plants showed an interesting diurnal change in starch and malate pools in the outermost layer of the extensor. Here starch increased at night while the malate pool was lowered nearly stoichiometrically. Inverse pool sizes were found in the day position of the respective leaves. Although less significant, the opposite diurnal variation occurred in samples taken from the flexor region. We thus were able to locate areas of different carbohydrate activities in the laminar pulvinus of P. coccineus. The central vascular core, including the bundle sheath, is involved in temporary storage of photoassimilates, and the cortical regions are responsible for osmotically driven leaf movement. Legume plants, due to their distinctive botanical characteristics, such as leaf movements, physiological characteristics, such as nitrogen fixation, and their abilities to endure environmental stresses, have important roles in sustainable pastures development. Leaf movement of legume plants is turgor regulated and osmotically active fluxes of ions between extensor and flexor of pulvinus cause this movement. To determine the role of calcium ions in circadian leaf movements of Phaseolus vulgaris L., a radiotracer technique experiment using 45Ca ions were employed. Measurements were taken during circadian leaf movements, and samples were taken from different parts of the leaflet. The 45Ca β-particle activity reduced from leaflet base pulvinus to leaf tip. The pulvinus had the highest activity, while the leaf tip had the lowest. By increase of the ratio of 45Ca β-particle activity within flexor to extensor (Fl/Ex) the midrib-petiole angle, as an indicator of leaf movement, increased linearly during circadian leaf movement (r = 0.86). The 45Ca β-particle activity of Flex/Ext ratio reduced linearly (r = −0.88) toward midnight. In conclusion, it was found that calcium ions accumulation is opposite to the fluxes of osmatically active ions and water movement. Whereas genome sequencing has given us high-resolution pictures of many different species of bacteria, microarrays provide a means of obtaining information on genome composition for many strains of a given species. Genome-composition analysis using microarrays, or 'genomotyping', can be used to categorize genes into 'present' and 'divergent' categories based on the level of hybridization signal. This typically involves selecting a signal value that is used as a cutoff to discriminate present (high signal) and divergent (low signal) genes. Current methodology uses empirical determination of cutoffs for classification into these categories, but this methodology is subject to several problems that can result in the misclassification of many genes.  相似文献

In this paper, we present a new analytical method to evaluate the temperature dependence of the thermal line shift and thermal line width of spectral lines in the Raman process using a simple approximation for the Debye functions. The proposed formulae guarantee the accurate and fast calculation of the thermal line shift and thermal line width. As an example of application, the analytical expression obtained is used to calculate the line shift and line width of the 2E4A2 transitions in V2+:MgO at temperatures from 0 K up to 500 K. This analytical evaluation shows that our results are satisfactory for the wide range temperature variations.  相似文献

Thigmonastic or seismonastic movements in Mimosa pudica, such as the response to touch, appear to be regulated by electrical, hydrodynamical and chemical signal transduction. The pulvinus of Mimosa pudica shows elastic properties, and we found that electrically or mechanically induced movements of the petiole were accompanied by a change of the pulvinus shape. As the petiole falls, the volume of the lower part of the pulvinus decreases and the volume of the upper part increases due to the redistribution of water between the upper and lower parts of the pulvinus. This hydroelastic process is reversible. During the relaxation of the petiole, the volume of the lower part of the pulvinus increases and the volume of the upper part decreases. Redistribution of ions between the upper and lower parts of a pulvinus causes fast transport of water through aquaporins and causes a fast change in the volume of the motor cells. Here, the biologically closed electrochemical circuits in electrically and mechanically anisotropic pulvini of Mimosa pudica are analyzed using the charged capacitor method for electrostimulation at different voltages. Changing the polarity of electrodes leads to a strong rectification effect in a pulvinus and to different kinetics of a capacitor discharge if the applied initial voltage is 0.5 V or higher. The electrical properties of Mimosa pudica''s pulvini were investigated and the equivalent electrical circuit within the pulvinus was proposed to explain the experimental data. Various nuclear analytical methods have been developed and applied to determine the elemental composition of calcified tissues (teeth and bones). Fluorine was determined by prompt gamma activation analysis through the19F(p, α ψ)16O reaction. Carbon was measured by activation analysis with He-3 ions, and the technique of Proton-Induced X-ray Emission (PIXE) was applied to simultaneously determine Ca, P, and trace elements in well-documented teeth. Dental hard tissues: enamel, dentine, cementum, and their junctions, as well as different parts of the same tissue, were examined separately. Furthermore, using a Proton Microprobe, we measured the surface distribution of F and other elements on and around carious lesions on the enamel. The depth profiles of F, and other elements, were also measured right up to the amelodentin junction.
