In a special habitat near Lunz (Mittersee) long time epiphytism ofCocconeis placentula var.tenuistriata and var.klinoraphis onFontinalis antipyretica leaves causes characteristic, strongly localized alterations in the walls of affected cells. They turn brown, may thicken, and may produce cystolithe-like protuberances into the cell lumina. These reactions obviously are brought about by a specific property ofCocconeis, for other epiphytes do not cause any change. The comparison of theCocconeis vegetation on younger and olderFontinalis leaves shows a considerable variability of the reactions mentioned, and in another habitat none could be found. Apparently several factors cause these reactions. Altogether,Cocconeis placentula is not a completely harmless epiphyte under certain circumstances. The alterations of the moss cells may be interpreted partly as a premature aging process, partly as a defensive action corresponding to alterations induced by some parasitic fungi. Furthermore, the reactions ofCocconeis onFontinalis are remarkably parallel to the influence of the dinococcalean algaRaciborskia inermis onSpirogyra grevilleana.

1. The effect of storm-flow on light- and dark-grown biofilms from a North Wales river was monitored using electron transport system (ETS) activity and epifluorescence cell counts. 2. Artificial substrata were colonized in the river, exposed to ambient-flow (60 cms?1) or storm-flow (235 cms?1 with suspended sediment addition) for 12h in a laboratory flow tank, and returned to the river to monitor the recovery period. 3. Total cell densities decreased in both the light- and dark-grown biofilms as a result of storm-flow. In addition, storm-flow ETS activity per cell increased significantly in the light-grown samples (2.7 times) but did not increase significantly in the dark-grown samples (1.6 times). 5. Within 48 h, storm-flow total cell densities and ETS activity levels had returned to ambient-flow levels in both light- and dark-grown biofilms. 6. Within each light regime, despite their different histories, both ambient and storm-flow samples responded the same to daily changes in the river environment, including a second disruption by natural causes, for the remainder of the 24-day experiment.  相似文献

The effects of herbivore size and hunger level were tested on lotic periphyton community structure and ash-free dry mass (AFDM). My hypotheses were 1) that small herbivores would remove more periphyton per unit biomass than large herbivores of the same species because of energetic demands and 2) that within the same size class, starved herbivores would remove more periphyton than nonstarved herbivores. The herbivore used was the prosobranch snail Elimia clavaeformis Lea. Seven treatments were employed: 1) no snails (control); 2) small “starved” snails; 3) large “starved” snails; 4) small and large “starved” snails; 5) small fed snails; 6) large fed snails; and 7) small and large fed snails. Although snails removed significant amounts of periphyton AFDM relative to controls, neither snail size nor degree of starvation had a significant effect on loss of total AFDM. Small snails removed significantly more erect forms of Stigeoclonium tenue (C. A. Ag.) Kütz. than large snails, but snail size had no other significant effect. Starved snails removed significantly more Cocconeis placentula Ehr. than fed snails, suggesting that after the more susceptible growth forms (e.g. erect Stigeoclonium) were removed, Cocconeis cells became more vulnerable to grazing by hungry snails. When small and large snails were combined in chambers, large snails gained weight over time, whereas small snails lost weight. These results suggest that intraspecific competition may occur within populations of Elimia.  相似文献

The valve structure of three marine diatom species, Cocconeis molesta var. crucifera, C. dirupta and C. pellucida was studied by transmission and scanning electron microscopy. The seasonal colonization and vertical zonation of adpressed and upright algae on Phragmites stems in Lake Belau, Germany, was investigated from November 1988 to October 1991. During the autumns and winters from 1988 to 1990 the dominant adpressed species included the diatoms Epithemia adnata, E. sorex, E. turgida and Rhopalodia gibba, while during the same seasons in 1990 and 1991, Cocconeis placentula var. euglypta was abundant. The upright diatoms were most abundant on the middle sections of the stems, whereas those epiphytes with most of their cell surfaces directly in contact with the substratum constituted the greatest part of the biomass on the lowest sections of the stems. Epithemia spp. and R. gibba attained their highest abundances near the sediment as well as buried beneath the most dense periphyton aggregations, while there were dense populations of C. placentula var. euglypta on the entire surface of the stems. Cocconeis is able to tolerate higher light intensities, with photosynthetically active radiation (PAR) values averaging 38?μmol?m?2?s?1, than Epithemia and Rhopalodia, which settle at a significantly lower PAR (26 and 20?μmol?m?2?s?1, respectively). The change in dominance among the adpressed species from Epithemia and Rhopalodia to Cocconeis can be explained by changes in the grazing habits of the snail Acroloxus lacustris.  相似文献

The ghost crab Ocypode stimpsoni displays waving and sound production. Sounds are produced by thumping the sand substratum with the major cheliped, and two types of sounds can be discriminated; one with a low frequency of about 12 Hz, called rapping, and another with a higher frequency (about double), called quivering. In our observations, a sequence of waving and sound emission would sometimes terminate abruptly, or appear as independent components but the component order never changed. The most frequently observed patterns were “waving with rapping and quivering”;, “waving with quivering”; and “quivering only”; quivering sounds being involved in more than 80% displays. Quivering sometimes occurred immediately after crabs emerged from the burrow, or when they returned to the entrance after discarding an excavated sand mass. The occurrence frequency of waving and sounds, the wave amplitude, and the frequency of the sound increased when other crabs approached.  相似文献

Coral sands off Réunion Island and Rodrigues (Mascarene archipelago, Western Indian Ocean) support diverse diatom communities, particularly rich in Monoraphidineae. Recent surveys of reef environments (June 2005 and June 2007) permitted the recognition of several taxa belonging to the diatom genus Cocconeis, among which are two new, relatively small taxa, epipsammic on coral sand. Cocconeis coronata Riaux-Gobin et Romero sp. nov. is similar to C. scutellum but has its own distinctive features. In external view, the concave raphe valve possesses uniseriate striae and a subsident hyaline marginal rim, while the strongly convex sternum valve has a submarginal, generally continuous crest, a hexagonal pattern of areolation, short, rounded spines that are often regularly arranged around valve face areolae, a complex areola hymen and small marginal chambers. Some morphometric differences were noted between the populations of C. coronata at Réunion and Rodrigues. Cocconeis margaritata Riaux-Gobin et Al-Handal, sp. nov. is smaller, possesses a sternum valve like that of C. placentula and C. neothumensis, but has denser striation and small pearl-like concretions around the areola aperture on the sternum valve. Several other taxa that resemble, or may be varieties of, C. margaritata require further study before being formally described. Cocconeis margaritata and C. coronata are small and relatively rare, so their study requires scanning electron microscopy.  相似文献

The role of epiphytes in an intertidal Zostera noltii seagrass bed in Marennes-Oléron Bay was assessed in comparison with the other main benthic primary producers (Z. noltii, microphytobenthos) at two bathymetric levels and on a seasonal basis. Assemblage and biomass of epiphytes were studied using scanning electron microscopy (SEM). Z. noltii and its detrital matter followed a typical seasonal pattern: microphytobenthos was present in large quantities throughout the year representing 21% of the total biomass while detrital matter, above-ground parts and below-ground parts accounted for 65, 9 and 5%, respectively. Only two species of epiphytic diatoms, Cocconeis scutellum and Cocconeis placentula, were observed on seagrass leaves. Epiphyte biomass was very low, representing on average less than 0.001% of that of microphytobenthos or leaves. This low epiphyte biomass is linked with the absence of macroalgae and also with the low biovolume of Cocconeis, which formed a monolayer of cells on leaves. This can be explained by the severe conditions of the intertidal and the high leaf turn-over of Z. noltii leaves.  相似文献

Algal responses to nutrients, grazing by Helicopsyche borealis, and concurrent grazing by Helicopsyche and Baetis tricaudatus were examined in recirculating stream chambers. Alagl communities, dominated by Achnanthes minutissima, Cocconeis placentula, and Synedra ulna, were primarily phosphorus-limited. Algal populations responded after only 6 days of nutrient enrichment. Initially, both the adnate diatom Cocconeis and erect diatom Synedra showed positive response to nutrient enrichment. Accumulation of algal biomass between day 3 and 6 in the P enriched treatment was resulted primarily from the growth of Synedra, an overstory rosette-like diatom colony. Such a shift in dominant growth from adnate to erect diatoms is a general phenomenon in periphyton succession in the absence of disturbance. Algal species showed differential responses to an increase of Helicopsyche densities. The accrual rate of Achnanthes continuously decreased with increasing grazer densities. The accrual rates of both Cocconeis and Synedra declined but reached plateaus between medium and high grazing densities. Baetis effectively and exclusively depressed Synedra and had no significant impact on Cocconeis. After concurrent grazing, algal communities were mainly dominated by Cocconeis (approximately 80% of total algal biovolume). The grazer' s mouth structures, grazing efficiencies, and mobility may account for the differential effects of concurrent grazing on algal communities. Significant interactive effects of P and grazing by Helicopsyche indicated that both nutrient addition and grazing may exert significant impact on algal communities. However, grazing may have a much stronger effect on algae than nutrients. Our results indicate that enhancement of algal biomass by P was dampened by grazing activities and that P had no effect on algal biomass in the presence of grazers.  相似文献

Grooming is a proactive method to keep a ship’s hull free of fouling. This approach uses a frequent and gentle wiping of the hull surface to prevent the recruitment of fouling organisms. A study was designed to compare the community composition and the drag associated with biofilms formed on a groomed and ungroomed fouling release coating. The groomed biofilms were dominated by members of the Gammaproteobacteria and Alphaproteobacteria as well the diatoms Navicula, Gomphonemopsis, Cocconeis, and Amphora. Ungroomed biofilms were characterized by Phyllobacteriaceae, Xenococcaceae, Rhodobacteraceae, and the pennate diatoms Cyclophora, Cocconeis, and Amphora. The drag forces associated with a groomed biofilm (0.75 ± 0.09 N) were significantly less than the ungroomed biofilm (1.09 ± 0.06 N). Knowledge gained from this study has helped the design of additional testing which will improve grooming tool design, minimizing the growth of biofilms and thus lowering the frictional drag forces associated with groomed surfaces.  相似文献

This study was performed in order to characterize the relationship between adhesion and biofilm formation abilities of drinking water-isolated bacteria (Acinetobacter calcoaceticus, Burkholderia cepacia, Methylobacterium sp., Mycobacterium mucogenicum, Sphingomonas capsulata and Staphylococcus sp.). Adhesion was assessed by two distinct methods: thermodynamic prediction of adhesion potential by quantifying hydrophobicity and the free energy of adhesion; and by microtiter plate assays. Biofilms were developed in microtiter plates for 24, 48 and 72 h. Polystyrene (PS) was used as adhesion substratum. The tested bacteria had negative surface charge and were hydrophilic. PS had negative surface charge and was hydrophobic. The free energy of adhesion between the bacteria and PS was > 0 mJ/m2 (thermodynamic unfavorable adhesion). The thermodynamic approach was inappropriate for modelling adhesion of the tested drinking water bacteria, underestimating adhesion to PS. Only three (B. cepacia, Sph. capsulata and Staphylococcus sp.) of the six bacteria were non-adherent to PS. A. calcoaceticus, Methylobacterium sp. and M. mucogenicum were weakly adherent. This adhesion ability was correlated with the biofilm formation ability when comparing with the results of 24 h aged biofilms. Methylobacterium sp. and M. mucogenicum formed large biofilm amounts, regardless the biofilm age. Given time, all the bacteria formed biofilms; even those non-adherents produced large amounts of matured (72 h aged) biofilms. The overall results indicate that initial adhesion did not predict the ability of the tested drinking water-isolated bacteria to form a mature biofilm, suggesting that other events such as phenotypic and genetic switching during biofilm development and the production of extracellular polymeric substances (EPS), may play a significant role on biofilm formation and differentiation. This understanding of the relationship between adhesion and biofilm formation is important for the development of control strategies efficient in the early stages of biofilm development.  相似文献

Manipulating objects (small rocks, mussel shells, etc.) to which limpets are clinging will cause many spontaneously to release their hold ("bail out"). Experiments demonstrated that in the owl limpet (Lottia gigantea) bail out was only triggered by rotation of the limpet around a horizontal axis at rates of 0.13 to 1.00rps. Limpets reattached more vigorously to a surface following a “bail out” than when they were forcibly removed from the substratum. In the field a variety of intertidal gastropods were observed to “bail out”. “Bail out” appears to be an adaptive response allowing an individual to abandon a dislodged substratum for a more stable one.  相似文献

Abstract

The objective of this work was to develop a subgingival biofilm model using a stirred bioreactor. Discs of bovine teeth were adapted to a stirred bioreactor filled with a culture medium containing bacterial species associated with periodontal health or disease. After anaerobic incubation, the biofilms growing on the substratum surfaces were collected and analyzed. The mean number of Colony-forming Units (CFUs) varied, but with no difference between 3 and 7?days of biofilm formation (p?>?0.05). Scanning Electron Microscopy (SEM) analysis showed a uniform biofilm layer covering the cement layer of the root surface containing bacteria with diverse morphology. In checkerboard DNA-DNA hybridization, bacterial species were identified in both biofilms. In conclusion, a subgingival biofilm model was developed using a stirred bioreactor, allowing the in vitro reproduction of complex microbial communities. This is an advanced model that may be useful to mimic complex clinical periodontal biofilms.  相似文献

Studies on the ecology of microbial parasites and their hosts are predicated on understanding the assemblage of and relationship among the species present. Changes in organismal morphology and physiology can have profound effects on host–parasite interactions and associated microbial community structure. The marine rickettsial organism, “Candidatus Xenohaliotis californiensis” (WS-RLO), that causes withering syndrome of abalones has had a consistent morphology based on light and electron microscopy. However, a morphological variant of the WS-RLO has recently been observed infecting red abalone from California. We used light and electron microscopy, in situ hybridization and16S rDNA sequence analysis to compare the WS-RLO and the morphologically distinct RLO variant (RLOv). The WS-RLO forms oblong inclusions within the abalone posterior esophagus (PE) and digestive gland (DG) tissues that contain small rod-shaped bacteria; individual bacteria within the light purple inclusions upon hematoxylin and eosin staining cannot be discerned by light microscopy. Like the WS-RLO, the RLOv forms oblong inclusions in the PE and DG but contain large, pleomorphic bacteria that stain dark navy blue with hematoxylin and eosin. Transmission electron microscopy (TEM) examination revealed that the large pleomorphic bacteria within RLOv inclusions were infected with a spherical to icosahedral-shaped putative phage hyperparasite. TEM also revealed the presence of rod-shaped bacteria along the periphery of the RLOv inclusions that were morphologically indistinguishable from the WS-RLO. Binding of the WS-RLO-specific in situ hybridization probe to the RLOv inclusions demonstrated sequence similarity between these RLOs. In addition, sequence analysis revealed 98.9–99.4 % similarity between 16S rDNA sequences of the WS-RLO and RLOv. Collectively, these data suggest that both of these RLOs infecting California abalone are “Candidatus Xenohaliotis californiensis,” and that the novel variant is infected by a putative phage hyperparasite that induced morphological variation of its RLO host.  相似文献

A nuclear mutation of Glycine max (soybean) segregates 1:2:1 in regard to chlorophyll content. The heterozygous (LG) leaf blade contains about one-half the pigment content of the wild type (DG) per gram fresh weight. A lethal yellow (LY) type contains about 1 to 2% of the DG leaf pigment values. The chlorophyll a/b ratio in the LG is about 5 compared to about 2 in the DG. Protein/leaf values are lower in the LG and LY types when compared to DG. The LG plastid lamellae contain more protein/chlorophyll, cytochromes/chlorophyll, and quinones/chlorophyll than the DG. P700/chlorophyll values are similar in the DG and LG types.  相似文献

A local strain of the pennate diatom Pinnularia cf. “nobilis” was investigated using cytochemistry and fluorescence and EM techniques. The regular perforation of the chloroplasts of P. “nobilis” and the lack of a typical diatom pyrenoid were confirmed at the ultrastructural level. Cavities and channels in the complex secondary plastid were found to harbor symbiotic bacteria, and their DNA elicited DAPI fluorescence. Wheat germ agglutinin, labeling bacteria walls, elicited a similar fluorescence pattern. Previous speculation that the apochlorotic DNA‐positive dots in the plastids of several Pinnularia species are “scattered ct‐nucleoids” is thus refuted. Bacteria were rod shaped and gram negative. They resided in the lumen of the endoplasmic reticulum (ER) during host interphase confined to the specialized ER compartment housing the secondary plastid, that is, to the space between the third and the fourth membrane profile, encircling the chloroplast. TEM images of chemically and cryofixed cells revealed that cavities resulted from the interaction of bacteria with the plastid according to the following sequence, alignment, attachment, deformation, and disintegration. This occurred without visible injury to the primary chloroplast envelope or the relict cell membrane of the reduced ancestral red alga that surrounds the chloroplast. The patterned arrangement of bacteria suggests recognition sites on the vestigial cell membrane, thought to interact with surface groups on the bacteria. The intimate association between bacteria and secondary plastid inside the common specialized ER cisterna suggests they form a functional unit. Comparison of thylakoid profiles, disrupted by bacteria in Pinnularia, with those disrupted by the pyrenoid in other pennate diatoms (e.g. Trachyneis) revealed a significant ultrastructural resemblance. No aposymbiotic Pinnularia cells were found at the sampling site.  相似文献

【目的】考察茎瘤固氮根瘤菌ORS571中c-di-GMP合成酶AZC-2412的编码基因缺失的突变表型,初步探究其功能机理。【方法】本实验构建基于cre-loxp重组酶系统的根瘤菌基因敲除系统,以及采用三亲接合技术构建突变株。测定野生型和突变株的生长速率、趋化能力、胞外多糖产量、生物膜形成等表型。【结果】突变株与野生型生长速率几乎相同。与野生型相比突变株由于细胞内c-di-GMP水平降低,胞外多糖、生物膜产量等均有所下降。【结论】实验表明,环二鸟苷酸合成酶AZC-2412缺失,使得c-di-GMP水平降低,对胞外多糖生成、细菌的运动能力、生物膜的形成、细胞絮凝、与植物的互作等均有调控作用。  相似文献

Abstract

Diatoms, which are early autotrophic colonisers, are an important constituent of the biofouling community in the marine environment. The effects of substratum and temporal variations on the fouling diatom community structure in a monsoon-influenced tropical estuary were studied. Fibreglass and glass coupons were exposed every month for a period of 4 days and the diatom population sampled at 24 h intervals, over a period of 14 months. The planktonic diatom community structure differed from the biofilm community. Pennate diatoms dominated the biofilms whilst centric diatoms were dominant in the water column. Among the biofilm diatoms, species belonging to the genera Navicula, Amphora, Nitzschia, Pleurosigma and Thalassionema were dominant. On certain occasions, the influence of planktonic blooms was also seen on the biofilm community. A comparative study of biofilms formed on the two substrata revealed significant differences in density and diversity. However species composition was almost constant. In addition to substratum variations, the biofilm diatom community structure also showed significant seasonal variations, which were attributed to physico-chemical and biological changes in both the water and substratum. Temporal variations in the tychopelagic diatoms of the water were also observed to exert an influence on the biofilm diatom community. Variations in diatom communities may determine the functional ecosystem of the benthic environment.  相似文献

Glass plates are frequently used as the substratum in flow cell experiments to allow continuous non-destructive observations of biofilm development via microscopy. The aim of this study was to evaluate hydroxyapatite-coated glass as a substratum for flow cell experiments, in comparison to plain glass, for modelling primary colonization of the tooth surface by Streptococcus sanguis. Glass plates were magnetron sputter coated with hydroxyapatite, producing a thin transparent layer. Biofilm development in the flow cell was recorded using image capture from a microscope, and images were analyzed to determine percentage coverage of the substratum over 24 h. Removal of biofilm by increasing the flow rate was also assessed. No statistically significant differences were detected between S. sanguis biofilms grown on the two different substratum materials. Hence, this work supports the proposal that the conditioning film reduces the influence of substratum surface properties.  相似文献
