ABSTRACT. Cells of Paramecium bursaria, which harbor a symbiotic alga of the genus Chlorella, reverse the effective beat of their cilia and swim backward when stimulated in either of two ways. Ionic stimulation is introduction of cells into a solution high in K+ while step-down photostimulation is a sharp reduction in the intensity of light falling on the culture. Much is known about the mechanisms of ionic stimulation of ciliary reversal, but little is known about step-down photobehavior. Inhibitors of ionically stimulated ciliary reversal were applied to cells undergoing step-down photobehavior; Ca-channel inhibitors, neomycin and W-7, inhibit both behaviors. Activation of Ca-channels in the ciliary membrane is involved in step-down photobehavior, suggesting that the algae may alter the Paramecium membrane to make it more excitable. 相似文献

Summary— Regulated exocytosis of defensive secretory organelles, the trichocysts, as well as a transient Ca2+-influx can be induced in Paramecium by aminoethyldextran (Kerb?uf and Cohen, J Cell Biol (1990) 111, 2527). Knoll et al (Febs Lett (1992) 304, 265) reported that veratridine was also a secretagogue for Paramecium. Here we show that, like aminoethyldextran, veratridine induces a transient Ca2+-influx. Both aminoethyldextran-and veratridine-induced exocytosis and associated Ca2+-influx were: i) blocked in the nd12 thermosensitive mutant at the non-permissive temperature; and ii) inhibited by amiloride and four divalent cations, Ba2+, Mg2+, Sr2+ and Co2+. This suggests that, although of different chemical nature, aminoethyldextran and veratridine act through the same physiological pathway. In addition, the inhibitory doses are comparable to the ones found to inhibit a hyperpolarization-sensitive Ca2+-current described in Paramecium (Preston et al (1992) J Gen Physiol 100, 233). The possibility that the activation of this Ca2+-current by the secretagogue represents an early step in the regulation of trichocyst exocytosis is discussed. 相似文献

SYNOPSIS. Methods are described for the recovery of highly purified DNA from Paramecium aurelia and Tetrahymena pyriformis in high yields. Our DNA is only slightly contaminated with RNA and carbohydrate, and little or no protein can be detected. We could not reduce the RNA (orcinol-positive material) by further treatment (sephadex or hydroxyapatite chromatography and preparative CsCl gradients). At the extreme our DNA is contaminated with 15–20% RNA but the real value is most likely considerably lower than this. The DNA we have prepared from Paramecium and Tetrahymena shows all the properties of double-stranded, high molecular weight DNA when characterized by temperature melting, CsCl density gradient centrifugation and hydroxyapatite and sephadex chromatography. When denatured, it absorbs to nitrocellulose filters. The 2 major results of importance from our work reported here are: (1) There is similarity in base composition of DNA from different syngens of Paramecium (28% G+C for syngens 1, 2, 4, 5, and 9 and 29–30% G+C for syngen 8) while there is variation between the syngens of Tetrahymena (24–31% G+C for syngens 1, 4, 7, 10, 11, and 12); (2) the density of any Paramecium DNA varies depending upon whether the cells are grown in the presence of bacteria or in axenic medium. Our results are compatible with observations previously reported for Tetrahymena but contradict those made for Paramecium. The earlier reports of differences in base composition between syngens of Paramecium are probably due in large part to the use of stocks grown on bacteria. 相似文献

Discs large 1 (Dlg1) is a modular scaffolding protein implicated in the control of cell polarity through assembly of specific multiprotein complexes, including receptors, ion channels and signaling proteins, at specialized zones of the plasma membrane. Recent data have shown that in addition to these well‐known interaction partners, Dlg1 may also recruit components of the vesicle trafficking machinery either to the plasma membrane or to transport vesicles. Here, we discuss Dlg1 function in vesicle formation, targeting, tethering and fusion, in both the exocytotic and endocytotic pathways. These pathways contribute to cell functions as major and diverse as glutamatergic activity in the neurons, membrane homeostasis in Schwann cell myelination, insulin stimulation of glucose transport in adipocytes, or endothelial secretion of the hemostatic protein, von Willebrand factor (VWF). 相似文献

The spatial and seasonal distribution of Paramecium bursaria in two small Indiana ponds was studied using a sampling grid. Very small (5.0 ml) samples were taken so that the individual microhabitats could be studied. The results were evaluated in comparison to the data collected for the P. aurelia complex collected in the same manner and at the same sites. It was found that P. bursaria exist in a clumped distribution, but that the distribution was not very different from random. Paramecium bursaria also exist at the surface and at the mud-water interface. Temperature does not seem to play a statistically significant role in determining population size. The breeding system of P. bursaria is optimized for an outbreeding population of low density. In comparison, the species of the P. aurelia complex exist in a very clumped distribution, are found only at the mud-water interface, and are inbreeders. The evolutionary strategies of the two types of paramecia are discussed. 相似文献

Growth-curvature responses of hypocotyls of Arabidopsis thaliana (L.) Heynh. were measured in double mutants between msg1 and axr1, both of which are auxin-resistant and defective in hypocotyl growth curvature induced upon unilateral application of auxin.
The msg1 axr1 double mutants showed no auxin-induced growth curvature, that is, they exhibited the msg1 phenotype, though the axr1 defects were partial. Hypocotyls of both the msg1 and axr1 mutants were partially defective in second-positive phototropism, whereas the double mutants lost the response completely.
When grown on vertically held agar plates, the axr1 mutant showed normal hypocotyl gravitropism and the mutation did not affect the reduced hypocotyl gravitropism of msg1. Hypocotyls of msg1 and axr1 mutants grew upward like wild-type ones when grown along an agar surface, while they grew more randomly when grown without
an agar support, suggesting that axr1 hypocotyls are not completely normal in gravitropism. The extent of defects in growth orientation increased in the order:
msg1 axr1 double mutants > msg1 > axr1 > wild type. The hypocotyls of these mutants showed auxin resistance in the order: msg1 axr1 > axr1 > msg1 > wild type. The msg1 mutant had epinastic leaves and axr1 had wrinkled leaves; leaves of the msg1 axr1 double mutants were epinastic and wrinkled. These results suggest that MSG1 and AXR1 act independently in separate pathways
of the reactions tested in the present study. In contrast, the phenotype of the msg1 aux1 double mutants shows that AUX1 is not significantly involved in these phenomena.

We evaluated the role of VAMP-2/synaptobrevin, VAMP-7/TI-VAMP, and VAMP-8/endobrevin in exocytic pathways of HSY cells, a
human parotid epithelial cell line, by coexpressing these VAMP proteins tagged with green fluorescent protein (GFP) and human
growth hormone (hGH) as a secretory cargo. Exocytosis of hGH was constitutive and the fluorescent signal of hGH–GFP was observed
in the Golgi area and small vesicles quickly moving throughout the cytoplasm. The cytoplasmic vesicles containing hGH overlapped
well with VAMP-7-GFP, but did so scarcely with VAMP-2-GFP or VAMP-8-GFP. However, when the vesicle transport from the trans-Golgi network to the plasma membrane was arrested by incubation at 20°C for 2 h and then released by warming up to 37°C;
VAMP-2-GFP and hGH were clearly colocalized together in small cytoplasmic vesicles. Neither VAMP-7-GFP nor hGH–GFP was colocalized
with LAMP-1, a marker for lysosomes and late endosomes. These results suggest that (1) VAMP-2 can be one of the v-SNAREs for
constitutive exocytosis; (2) VAMP-7 is involved in the constitutive exocytosis as a slow, minor v-SNARE, but not in the lysosomal

Compassion, the emotional response of caring for another who is suffering and that results in motivation to relieve suffering, is thought to be an emotional antecedent to altruistic behavior. However, it remains unclear whether compassion enhances altruistic behavior in a uniform way or is specific to sub-types of behavior such as altruistic helping of a victim or altruistic punishment of a transgressor. We investigated the relationship between compassion and subtypes of altruistic behavior using third-party paradigms where participants 1) witnessed an unfair economic exchange between a transgressor and a victim, and 2) had the opportunity to either spend personal funds to either economically a) help the victim or b) punish the transgressor. In Study 1, we examined whether individual differences in self-reported empathic concern (the emotional component of compassion) was associated with greater altruistic helping or punishment behavior in two independent samples. For participants who witnessed an unfair transaction, trait empathic concern was associated with greater helping of a victim and had no relationship to punishment. However, in those who decided to punish the transgressor, participants who reported greater empathic concern decided to punish less. In Study 2, we directly enhanced compassion using short-term online compassion meditation training to examine whether altruistic helping and punishment were increased after two weeks of training. Compared to an active reappraisal training control group, the compassion training group gave more to help the victim and did not differ in punishment of the transgressor. Together, these two studies suggest that compassion is related to greater altruistic helping of victims and is not associated with or may mitigate altruistic punishment of transgressors. 相似文献

A review of field and laboratory investigations suggests thatmany ectothermic vertebrates can exploit the spatiotemporaldistribution of environmental temperatures to maximize energyutilization and to enhance survivability. Diel and seasonalcycles in thermal preference, acclimation rate, thermal toleranceand heat-hardening may well be adapted to temporal variationsin environmental temperature. In addition, many ectoiherms behaviorallyexploit thermal heterogeneity in the environment. Such behavioraladaptations are synergistic with various degrees of physiologicalregulation. Voluntary brief exposures to temperatures that wouldbe lethal upon prolonged exposure can result in heat-hardening.Heat-hardening, distinct from acclimation to high temperature,is a short-term increase in thermal tolerance while toleranceacclimation is a longer lasting response within normal rangesof environmental temperatures; both are reversible nongeneticresponses. The physiological and ecological significance ofbehaviorally mediated heat-hardening may be greater than previouslyrealized and suggest new approaches for future study. 相似文献

1. The role of synaptophysin in the exocytotic release of dopamine (DA) was examined in Xenopus laevis oocytes injected with rat brain mRNA.2. The mRNA-injected oocytes showed DA uptake which depended on the incubation time and external DA concentrations.3. Stimulation with KCl (10–50 mM) of mRNA-injected oocytes preloaded with DA evoked external Ca2+-dependent release of DA. Paramecium tetraurelia, like some other ciliate species, uses an alternative nuclear genetic code where UAA and UAG are translated as glutamine and UGA is the only stop codon. It has been postulated that the use of stop codons as sense codons is dependent on the presence of specific tRNAs and on modification of eukaryotic release factor one (eRF1), a factor involved in stop codon recognition during translation termination. We describe here the isolation and characterisation of two genes, eRF1-a and eRF1 b, coding for eRF1 in P. tetraurelia. The two genes are very similar, both in genomic organization and in sequence, and might result from a recent duplication event. The two coding sequences are 1,314 nucleotides long, and encode two putative proteins of 437 amino acids with 98.5% identity. Interestingly, when compared with the eRF1 sequences either of ciliates having the same variant genetic code, or of other eukaryotes, the eRF1 of P. tetraurelia exhibits significant differences in the N-terminal region, which is thought to interact with stop codons. We discuss here the consequences of these changes in the light of recent models proposed to explain the mechanism of stop codon recognition in eukaryotes. Besides, analysis of the expression of the two genes by Northern blotting and primer extension reveals that these genes exhibit a differential expression during vegetative growth and autogamy. 相似文献

The Sec1/Munc18 (SM) proteins constitute a conserved family with essential functions in SNARE‐mediated membrane fusion. Recently, a new protein–protein interaction site in Sec1p, designated the groove, was proposed. Here, we show that a sec1 groove mutant yeast strain, sec1(w24), displays temperature‐sensitive growth and secretion defects. The yeast Sec1p and mammalian Munc18‐1 grooves were shown to play an important role in the interaction with the SNAREs Sec9p and SNAP‐25b, respectively. Incubation of SNAP‐25b with the Munc18‐1 groove mutant resulted in a lag in the kinetics of SNARE complex assembly in vitro when compared with wild‐type Munc18‐1. The SNARE regulator SRO7 was identified as a multicopy suppressor of sec1(w24) groove mutant and an intact Sec1p groove was required for the plasma membrane targeting of Sro7p–SNARE complexes. Simultaneous inactivation of Sec1p groove and SRO7 resulted in reduced levels of exocytic SNARE complexes. Our results identify the groove as a conserved interaction surface in SM proteins. The results indicate that this structural element is important for interactions with Sec9p/SNAP‐25 and participates, in concert with Sro7p, in the initial steps of SNARE complex assembly. 相似文献

被动游泳运动可诱发小鼠抑郁样行为,游泳环境的改变已成为抑郁样行为严重程度影响因素之一。观察不同水质、水温及持续时间对被动游泳小鼠抑郁样行为的影响,并初步探讨肠道菌群组成与抑郁样行为的关系。通过不同条件下的被动游泳运动建立抑郁样行为小鼠模型。采用糖水偏好实验及强迫游泳实验评价其行为学变化;采用16S rDNA高通量测序技术及实时荧光定量PCR技术对小鼠肠道菌群进行分子生态学分析。被动游泳16周后,各模型组小鼠体质量及糖水偏爱度均较正常对照组降低,而不动时间则有所延长。其中,室温海水游泳15 min小鼠体质量及糖水偏爱度降低程度最大,不动时间最长,与正常对照组比较,均具有显著性差异(P<0.05)。各模型组小鼠肠道菌群Chao指数、Shannon指数及PCoA分析均较正常对照组具有显著性差异(P<0.05),其从门水平到属水平的丰度也发生不同程度改变。其中,室温海水游泳15 min小鼠肠道菌群组成变化程度最大,并发生拟杆菌属、普氏菌属等多个菌属的富集以及乳杆菌属丰度的减少,实时荧光定量PCR实验也得到了较为一致的结果(P<0.05)。以上结果表明,被动游泳运动可导致小鼠抑郁样行为的发生,其肠道菌群组成也发生明显改变;同时,菌群组成的改变会随着抑郁样行为的严重程度而有所变化。 相似文献

In the laboratory, courtship behavior and carotenoid pigmentation of male guppies are condition-dependent traits, since their expression is affected by physical vigor and environmental factors such as water velocity and diet. Whether these relationships exist in guppies under field conditions is yet to be determined. We compared the swimming performance, courtship behavior, and carotenoid pigmentation of guppies from headwater and downstream localities in four rivers of Trinidad. Swimming performance and courtship behavior of males differed among rivers and between headwater and downstream sites. Guppies from headwater sites swam significantly faster and had higher display rates than those from downstream sites. Mean swimming performance across sites was positively correlated with mean water velocity, but was correlated with the number of orange color spots (carotenoid pigment) in only one river. These results indicate that the courtship behavior of Trinidadian guppies is condition-dependent because the amount of display behavior is positively correlated to swimming performance, a measure of physical endurance. The proximal cause for this condition-dependence may be predator induced variation in microhabitat use by guppies in headwater and downstream locations. 相似文献
