Non-motile cells of Haematococcus pluvialis grow slowly, whereas motile cells grow fast and divide frequently. Cultivation from non-motile cells to motile cells of H. pluvialis was implemented to promote semi-continuous production. When old cultures which consist of non-motile cells were inoculated in fresh medium with an inoculation amount less than 15%, zoospores were produced in the non-motile cells and developed into motile cells, as the concentration of astaxanthin inducer in the medium was below the threshold value. This process was accomplished within 3 days after inoculation. Furthermore, enhancing KNO3 content to 1200 mg L?1 or reducing light intensity to 20 μmol photons m?2?s?1 could increase growth during the late culturing period of H. pluvialis and postpone the next round of transformation from motile cells to non-motile cells. A semi-continuous cultivation method for H. pluvialis from non-motile cells to motile cells is proposed in order to regulate the life cycle and promote industrial production. This cultivation mode shortens the inoculum cultivation stage and simplifies the production process of H. pluvialis, showing considerable commercial potential.  相似文献

The combined effect of macronutrients in the extraction medium on α-amylase produced by Bacillus subtilis were studied by using response surface methodology in shaken flask cultures. The production of amylase was significantly affected by the interaction between wheat bran and the cotton seed extract in the extraction medium and by the interaction between the cotton seed extract and starch. The optimal combination in the extraction medium for maximum α-amylase production was determined as 10.80 g·L?1 of wheat bran, 9.90 g·L?1 of cotton seed extract, 0.5 g·L?1 of starch, 2.0 g·L?1 of yeast extract, 5.00 g·L?1 of NaCl and 2.00 g·L?1 of CaCl2. A 12.55-fold increase of enzyme activity was recorded in the optimized medium compared to the result acquired in a minimum essential medium. The optimized medium was used to compare different cultivation strategies in fermenters. The pH-stat strategy for reducing cellular stress response and the substrate concentration-stat strategy for reducing substrate inhibition were independently investigated. The temperature-limited strategy has been proposed to solve the proteolytic digestion problem, although the high-pressure strategy resulted in high productivity. A hybrid strategy simultaneously controlling pH, temperature, substrate concentration and pO2 was finally investigated to enhance the efficiency of the process. This hybrid strategy resulted in high activity of α-amylase, increasing the productivity almost three-fold as compared to an ordinary fed-batch culture.  相似文献

The effects of incubation time and deficiencies of calcium and nitrate on zoospore production were investigated in Botrydiopsis arhiza Borzi, B. intercedens Vischer et Pascher, Bumilleriopsis peterseniana Vischer, Pseudobumilleriopsis pyrenoidosa Deason et Bold, and Ophiocytium maius Naegeli. Release of zoospores occurred after the transfer of stationary phase, vegetative cells into fresh Bold's basal medium. Maximum yields of zoospores were produced on the second day after transfer, 0·5–1·0 h after the start of the light period of a 12–12 h light-dark cycle. Lack of calcium or nitrate reduced the yield of zoospores in each taxon. Only O. maius produced large numbers of motile cells in nitrate-free medium.  相似文献

L-Asparaginase amidohydrolase (EC 3.5.1.1) has received significant attention owing to its clinical use in acute lymphoblastic leukemia treatment and non-clinical applications in the food industry to reduce acrylamide (toxic compound) formation during the frying of starchy foods. In this study, a sequential optimization strategy was used to determine the best culture conditions for L-asparaginase production from filamentous fungus Aspergillus terreus CCT 7693 by submerged fermentation. The cultural conditions were studied using a 3-level, central composite design of response surface methodology, and biomass and enzyme production were optimized separately. The highest amount of biomass (22.0?g·L?1) was obtained with modified Czapek–Dox medium containing glucose (14?g·L?1), L-proline (10?g·L?1), and ammonium nitrate (2?g·L?1) fermented at 37.2?°C and pH 8.56; for maximum enzyme production (13.50?U·g?1), the best condition was modified Czapek–Dox medium containing glucose (2?g·L?1), L-proline (10?g·L?1), and inoculum concentration of 4.8?×?108 espore·mL?1 adjusted to pH 9.49 at 34.6?°C. The L-asparaginase production profile was studied in a 7?L bench-scale bioreactor and a final specific activity of 13.81?U·g?1 was achieved, which represents an increase of 200% in relation to the initial non-optimized conditions.  相似文献

In this study, the effects of carbon source, nitrogen source, and metal ions on cell growth and Bacillus aryabhattai β-amylase production in recombinant Brevibacillus choshinensis were investigated. The optimal medium for β-amylase production, containing glucose (7.5?g·L?1), pig bone peptone (40.0?g·L?1), Mg2+ (0.05?mol·L?1), and trace metal elements, was determined through single-factor experiments in shake flasks. When cultured in the optimized medium, the β-amylase yield reached 925.4?U mL?1, which was 7.2-fold higher than that obtained in the initial medium. Besides, a modified feeding strategy was proposed and applied in a 3-L fermentor fed with glucose, which achieved a dry cell weight of 15.4?g L?1. Through this cultivation approached 30?°C with 0?g·L?1 initial glucose concentration, the maximum β-amylase activity reached 5371.8?U mL?1, which was 41.7-fold higher than that obtained with the initial medium in shake flask.  相似文献

This paper deals with the production of 2,3- butanediol by K. oxytoca in batch cultures. The effect of urea on various kinetic parameters was studied by replacing the ammonium salts in the medium with the corresponding nitrogen equivalent in the form of urea. The specific growth rate and the product yield in an unacclimatised batch culture were found to be 0.29 h?1 and 0.26 g·g?1 respectively. The acclimatised batch cultures on the other hand behaved similar to that grown using the original medium with a specific growth rate of 0.66 h?1 and the product yield of 0.345 g·g?1. However the cultures were unable to grow when urea was used both as the carbon and nitrogen source.  相似文献

以红花草莓叶片为外植体,通过筛选诱导愈伤组织、不定芽及壮苗、生根的培养基,建立一套实用且易推广的红花草莓组培快繁技术体系。结果表明:在愈伤组织的诱导过程中TDZ的诱导效果优于6-BA,TDZ与NAA配合使用效果优于与IBA的组合。6-BA浓度为0.5 mg·L~(-1)时不定芽诱导率高达86.6%。低浓度的6-BA和8 g·L~(-1)的琼脂更有利于壮苗培养,NAA比IBA更有利诱导生根。综上述,最适红花草莓愈伤组织的诱导培养基为MS+1.0 mg·L~(-1)TDZ+0.5 mg·L~(-1)NAA+30 g·L~(-1)蔗糖+7 g·L~(-1)琼脂;最适不定芽分化的培养基为MS+0.5 mg·L~(-1)6-BA+0.1 mg·L~(-1)NAA+30 g·L~(-1)蔗糖+7 g·L~(-1)琼脂;最适壮苗培养基为MS+0.1 mg·L~(-1)6-BA+0.1 mg·L~(-1)NAA+30 g·L~(-1)蔗糖+8g·L~(-1)琼脂;最适生根培养基为MS+0.5mg·L~(-1)NAA+30 g·L~(-1)蔗糖+8 g·L~(-1)琼脂。试管苗移栽生长20 d后,成活率高达93%,且后期草莓苗生长壮健。此体系的建立为优质红花草莓种苗大规模生产提供了科学依据和技术支持。  相似文献

以马来沉香茎段为外植体,分别对外植体的消毒、启动培养、增殖培养、壮苗培养、生根培养、炼苗移栽环节进行研究,着重探索马来沉香组织培养技术各个环节的最佳培养基配方,为马来沉香的工厂化育苗提供技术指导。结果表明:马来沉香最佳消毒方法是用0.1%升汞消毒4~5min;启动率最高的培养基配方是1/2MS+6-BA 0.2mg·L-1+NAA 0.1mg·L-1+蔗糖30g·L-1+琼脂5.8g·L-1,启动率达70.5%;增殖系数最高的培养基是1/2MS+0.1mg·L-16-BA+25g·L-1蔗糖+5.8g·L-1琼脂,增殖系数达2.9;最佳壮苗培养基是1/2MS+30g·L-1蔗糖+5.8g·L-1琼脂;最佳生根培养基为1/2MS+NAA 5.0mg·L-1+20g·L-1糖+6g·L-1琼脂,培养2d后移入1/2MS培养基继续培养,生根率为83%;马来沉香移栽较难成活,在泥炭土∶黄泥土(2∶1)的基质上成活率最高,移栽成活率65%。  相似文献

Several factors affecting erythritol production from glycerol by Yarrowia lipolytica Wratislavia K1 strain were examined in batch fermentations. Ammonium sulfate, monopotassium phosphate, and sodium chloride were identified as critical medium components that determine the ratio of polyols produced. The central composite rotatable experimental design was used to optimize medium composition for erythritol production. The concentrations of ammonium sulfate, monopotassium phosphate, and sodium chloride in the optimized medium were 2.25, 0.22, and 26.4 g L?1, respectively. The C:N ratio was found as 81:1. In the optimized medium with 100 g L?1 of glycerol the Wratislavia K1 strain produced 46.9 g L?1 of erythritol, which corresponded to a 0.47 g g?1 yield and a productivity of 0.85 g L?1 hr?1. In the fed-batch mode and medium with the total concentration of glycerol at 300 g L?1 and C:N ratio at 81:1, 132 g L?1 of erythritol was produced with 0.44 g g?1 yield and a productivity of 1.01 g L?1 hr?1.  相似文献

The effect of dissolved oxygen partial pressure on the accumulation of astaxanthin in the green alga Haematococcus lacustris ( Gir.) Rostaf (UTEX16) was studied in N-limited continuous chemostat cultures. The steady-state astaxanthin content measured against culture volume, cell number, and biomass dry weigh of Haematococcus cultures was proportional to the dissolved O2 partial pressure in the culture medium, over the range of 0–50% O2 The steady-state biomass dry weight concentrations remained at between 0.52 and 0.57 g. L-1 over the range of dissolved O2 partial pressure studied. Steady-state cell densities at dissolved O2 partial pressures above the air saturation level (1.13–1.58 × 105 cells.mL-1) were about half of that measured at lower dissolved O2 partial pressures (2.42–2.63 × 105 cells.mL-1). Both biflagellated zoospores and nonmotile aplanospores were found at steady state. The fraction of nonmotile cells was higher at dissolved O2 partial pressures above the air saturation level (94.44–98.01%) than at dissolved O2 partial pressure below the air level (79.64–86.12 and 91.75% ).  相似文献

采用正交试验等设计,系统开展了光皮桦组织培养高效再生体系研究。结果表明:光皮桦茎段外植体最佳诱导培养基及激素组合为MS+0.50mg.L-1 6-BA+0.10mg.L-1 TDZ+30mg.L-1蔗糖+5.50g.L-1琼脂,丛生芽最佳生根培养基为1/2MS+1mg.L-1 IBA+20g.L-1蔗糖+5.50g.L-1琼脂;丛生芽在最佳生根培养基上培养15d后获健壮生根苗,移栽成活率达90%。该实验结果为光皮桦的优良品种快繁以及遗传转化体系建立奠定了良好的基础。  相似文献

In this study, a hybrid system of response surface methodology followed by genetic algorithm has been adopted to optimize the production medium for L-glutamic acid fermentation with mixed cultures of Corynebacterium glutamicum and Pseudomonas reptilovora. The optimal combination of media components for maximal production of L-glutamic acid was found to be 49.99 g L?1 of glucose, 10 g L?1 of urea, 18.06% (v/v) of salt solution, and 4.99% (v/v) of inoculum size. The experimental glutamic acid yield at optimum condition was 19.69 g L?1, which coincided well to the value predicted by the model (19.61 g L?1). Using this methodology, a nonlinear regression model was developed for the glutamic acid production. The model was validated statistically and the determination coefficient (R 2) was found to be 0.99.  相似文献

该文选用牛角瓜茎段为外植体,通过组织培养方法探索牛角瓜组织培养和种苗快繁技术。结果表明:最佳外植体表面消毒方法是以0.1%HgCl_2处理7 min,外植体存活率为32.3%;初代培养基为MS+蔗糖30 g·L~(-1)+琼脂3.5 g·L~(-1),培养20 d后形成3~4 cm高的再生芽。增殖培养前期筛选的较为适宜的增殖培养基为MS+6-BA 1.5 mg·L~(-1)+NAA 0.05 mg·L~(-1)+蔗糖30 g·L~(-1)+琼脂3.5 g·L~(-1),增殖系数4.6。但在后续的培养过程中发现,牛角瓜组培苗易玻璃化,且随着世代更迭,玻璃化程度加重,到了第四代几乎全部玻璃化。因此在上述增殖培养的基础上,以AgNO_3作为玻璃化抑制剂,筛选出最终的增殖培养基为MS+6-BA1.5 mg·L~(-1)+NAA 0.05 mg·L~(-1)+AgNO_31.0 g·L~(-1)+蔗糖30 g·L~(-1)+琼脂3.5 g·L~(-1)。用此增殖培养基,培养25 d,苗高5~8 cm,增殖系数5.8,玻璃化率低于10%,且连续培养多代,玻璃化率维持在10%以下。生根壮苗培养基为1/2MS+NAA 1.0 mg·L~(-1)+蔗糖20 g·L~(-1)+琼脂3.6 g·L~(-1),培养14 d,生根率98%;将生根苗移栽于70%遮阴度的大棚中,30 d后,苗高20 cm左右,成活率85%。利用该方法可对牛角瓜优良种苗进行规模化生产。  相似文献

Studies were performed on the effect of CaCO3 and CaCl2 supplementation to fermentation medium for ethanol production from xylose, glucose, or their mixtures using Scheffersomyces (Pichia) stipitis. Both of these chemicals were found to improve maximum ethanol concentration and ethanol productivity. Use of xylose alone resulted in the production of 20.68 ± 0.44 g L?1 ethanol with a productivity of 0.17 ± 0.00 g L?1 h?1, while xylose plus 3 g L?1 CaCO3 resulted in the production of 24.68 ± 0.75 g L?1 ethanol with a productivity of 0.21 ± 0.01 g L?1 h?1. Use of xylose plus glucose in combination with 3 g L?1 CaCO3 resulted in the production of 47.37 ± 0.55 g L?1 ethanol (aerobic culture), thus resulting in an ethanol productivity of 0.39 ± 0.00 g L?1 h?1. These values are 229 % of that achieved in xylose medium. Supplementation of xylose and glucose medium with 0.40 g L?1 CaCl2 resulted in the production of 44.84 ± 0.28 g L?1 ethanol with a productivity of 0.37 ± 0.02 g L?1 h?1. Use of glucose plus 3 g L?1 CaCO3 resulted in the production of 57.39 ± 1.41 g L?1 ethanol under micro-aerophilic conditions. These results indicate that supplementation of cellulosic sugars in the fermentation medium with CaCO3 and CaCl2 would improve economics of ethanol production from agricultural residues.  相似文献

以自选育的白花泡桐优树茎段为外植体,进行种苗组培快繁技术研究。结果表明:其最佳的外植体灭菌方法是以0.1%升汞处理7 min;合适的初代诱导培养基为MS+6-BA 2.0 mg·L~(-1)+IBA 0.2 mg·L~(-1)+糖30 g·L~(-1)+琼脂3.5 g·L~(-1)(pH 5.8),培养30 d,芽诱导率70%;合适的继代增殖方法为在高浓度植物生长物质培养基MS+6-BA 4.0 mg·L~(-1)+IBA 0.4 mg·L~(-1)+蔗糖30 g·L~(-1)+琼脂3.5 g·L~(-1)(pH 5.8)和低浓度植物生长物质培养基MS+6-BA 0.4 mg·L~(-1)+IBA 0.04 mg·L~(-1)+蔗糖30 g·L~(-1)+琼脂3.5 g·L~(-1)(pH 5.8)中交替培养,获得的丛生芽长势良好,玻璃化率低于5%,增殖系数大于6.0/25 d;最适的生根培养基为1/2MS+NAA0.2 mg·L~(-1)+蔗糖20 g·L~(-1)+卡拉胶3.4 g·L~(-1)(pH 5.8),培养14 d,得到白花泡桐生根苗,每株长根5~10条,根长3~5 cm,生根率98%,根系洁白、根毛少而短,易于清洗。将生根苗按照常规方法炼苗后移栽于温室大棚中,50 d后即可出圃,此时平均苗高1.0 m、地径1.0~2.0 cm,成活率在90%以上。  相似文献

为探究裸果木再生体系建立的影响因素,确定其不定芽发生的起源,该研究以裸果木健壮植株的茎段为外植体,采用6 BA和IBA不同浓度组合,筛选愈伤增殖及不定芽再生的最佳浓度组合,确定生根诱导的关键影响因素,建立再生体系,并对其不定芽分化进程进行解剖结构分析,以确认其起源。结果表明:(1)裸果木茎段的最佳愈伤增殖培养基为MS+1 mg·L-1 IBA+1 mg·L-1 6 BA+30 g·L-1蔗糖+7 g·L-1琼脂,主体间效应分析表明IBA为关键影响因素;愈伤大小随IBA浓度增加呈现先升高后下降的趋势。(2)最佳不定芽诱导培养基为MS+0.5 mg·L-1 6 BA+30 g·L-1蔗糖+7 g·L-1琼脂,诱导不定芽数量为4.9个/块,生芽率达92.3%。(3)生根诱导中,SH基本培养基和蔗糖浓度为关键因素,最佳生根培养基为SH+0~10 g·L-1蔗糖+7 g·L-1琼脂,生根率达91.3%。(4)解剖结构观察发现,不定芽起源于愈伤表层的分生细胞,为外起源。该研究通过器官发生途径建立了裸果木的再生体系,确定了不定芽为外起源,为裸果木这一珍稀濒危的林木种质资源保护及可持续利用奠定了研究基础,并为其未来的发展利用提供了有效途径。  相似文献

The fermentation of both glucose and xylose is important to maximize ethanol yield from renewable biomass feedstocks. In this article, we analyze growth, sugar consumption, and ethanol formation by the yeast Kluyveromyces marxianus UFV-3 using various glucose and xylose concentrations and also under conditions of reduced respiratory activity. In almost all the conditions analyzed, glucose repressed xylose assimilation and xylose consumption began after glucose had been exhausted. A remarkable difference was observed when mixtures of 5 g L?1 glucose/20 g L?1 xylose and 20 g L?1 glucose/20 g L?1 xylose were used. In the former, the xylose consumption began immediately after the glucose depletion. Indeed, there was no striking diauxic phase, as observed in the latter condition, in which there was an interval of 30 h between glucose depletion and the beginning of xylose consumption. Ethanol production was always higher in a mixture of glucose and xylose than in glucose alone. The highest ethanol concentration (8.65 g L?1) and cell mass concentration (4.42 g L?1) were achieved after 8 and 74 h, respectively, in a mixture of 20 g L?1 glucose/20 g L?1 xylose. When inhibitors of respiration were added to the medium, glucose repression of xylose consumption was alleviated completely and K. marxianus was able to consume xylose and glucose simultaneously.  相似文献

The green alga Haematococcus pluvialis is the current best source of natural astaxanthin, a high-value carotenoid. Traditionally, the production process of astaxanthin by this algae is achieved by a two-stage system: during the first stage, vegetative “green” cells are produced and then converted, in the second stage, into cysts that accumulate astaxanthin. In this work, a medium screening strategy based on the mixing of a three-component hydroponic fertilizer was applied to identify a new formulation optimized for the vegetative stage. A maximal and high cell density of 2?×?106 cells mL?1 was obtained in a medium containing a high level of phosphate relative to nitrate, resulting in a N/P ratio much lower than commonly used media for H. pluvialis. In this medium, cells remained at the vegetative and motile stage during a prolonged period of time. Both high cell density culture and motile stage persistence was proved to be related to the N/P feature of this medium. We conclude that the macrozoid stage of H. pluvialis is favored under high-P and low-N supply and that low-cost hydroponic fertilizers can be successfully used for achieving high density cultures of vegetative cells of H. pluvialis.  相似文献

褐纹报春苣苔(Primulina glandaceistriata)是一种极具观赏价值的喀斯特地区野生花卉,目前尚未有褐纹报春苣苔组培快繁的研究报道。该研究以褐纹报春苣苔的叶片为外植体,通过两种途径建立其组培快繁体系。结果表明:适宜的不定芽诱导培养基为MS+6-BA 2.0 mg·L~(-1)+NAA 0.10 mg·L~(-1),适宜的不定芽增殖培养基为MS+ZT 2.0 mg·L~(-1)+NAA 0.10 mg·L~(-1)+活性炭0.05 g·L~(-1),增殖系数为11.09;适宜的愈伤组织诱导培养基为MS+TDZ 2.0 mg·L~(-1)+NAA 0.10 mg·L~(-1),适宜的愈伤组织分化培养基为MS+ZT 1.0 mg·L~(-1)+NAA 0.10mg·L~(-1),分化系数为12.46;适宜的生根培养基为1/2MS+IBA 0.1 mg·L~(-1)+活性炭0.05 g·L~(-1)或1/2MS+IBA0.5 mg·L~(-1)+活性炭0.05 g·L~(-1),生根率为100%。该研究结果成功建立了褐纹报春苣苔的组培快繁体系,为今后褐纹报春苣苔的种苗繁殖和遗传转化提供了技术支持。  相似文献

Citric acid was produced by five species of the yeast Candida after growth on a medium containing soy biodiesel-based crude glycerol. After growth on a medium containing 10 g L?1 or 60 g L?1 crude glycerol for 168 hr at 30°C, Candida parapsilosis ATCC 7330 and C. guilliermondii ATCC 9058 produced the highest citric acid levels. On 10 g L?1 or 60 g L?1 crude glycerol for 168 hr at 30°C, the citric acid level produced by C. parapsilosis ATCC 7330 was 1.8 g L?1 or 11.3 g L?1, respectively, while C. guilliermondii ATCC 9058 produced citric acid concentrations of 3.0 g L?1 or 10.4 g L?1, respectively. Biomass production by C. guilliermondii ATCC 9058 on 10 g L?1 or 60 g L?1 crude glycerol for 168 hr at 30°C was highest at 1.2 g L?1 or 6.9 g L?1, respectively. The citric acid yields observed for C. guilliermondii ATCC 9058 after growth on 10 g L?1 or 60 g L?1 crude glycerol (0.35 g g?1 or 0.21 g g?1, respectively) were generally higher than for the other Candida species tested. When similar crude glycerol concentrations were present in the culture medium, citric acid yields observed for some of the Candida species utilized in this study were about the same or higher compared to citric acid yields by Yarrowia lipolytica strains. Based on the findings, it appeared that C. guilliermondii ATCC 9058 was the most effective species utilized, with its citric acid production being similar to what has been observed when citric acid-producing strains of Y. lipolytica were grown on crude glycerol under batch conditions that could be of significance to biobased citric acid production.  相似文献
