Relatively high levels of dihydrozeatin and trans-zeatin were detected in unstratified seeds of Acer saccharum Marsh. Both cytokinins increased substantially over the first 20 days of stratification at 5°C and then fell rapidly to values well below original levels by first germination on day 55. In seeds held at 20°C, a non-afterripening temperature, cytokinin levels remained constant for the first 10 days and then declined to their lowest levels by day 20. Levels of putrescine, spermidine and spermine in the radicles and cotyledons did not change during the full course of the afterripening process, but large increments were noted during radicle emergence. A large increase in ethylene production at germination suggests that competition for S-adenosyl-methionine by the ethylene and polyamine biosynthetic pathways did not inhibit synthesis of ethylene or polyamines during seedling emergence and establishment. In seeds stratified at 20°C, ethylene showed an exceptionally large peak early in the stratification period, but polyamine contents remained low throughout the test. The present results are consistent with the hypothesis that cytokinins play a significant role in overcoming the metabolic block present in dormant seeds. This conclusion is supported by data showing that high levels of cytokinins develop concurrently with the start of tissue differentiation and at the time when abscisic acid and phenolic inhibitors decline in stratifying seeds. Changes in ethylene and polyamine contents did not correlate with any events in the afterripening process; however, large increases in levels of these substances were closely associated with the germinative process and, in the case of polyamines, specifically with the start of cell division.  相似文献

 The numbers and types of spores of vesicular-arbuscular mycorrhizal fungi occurring in the top 15 cm of the soil in three maple forests in Eastern Canada were investigated using traditional wet-sieving/decanting methods. In the most acid site, at St. Hippolyte, Québec, where the soil had been amended with base cations, after 1 year there was no effect on the numbers of spores present. Vesicular-arbuscular mycorrhizal spores present at St. Hippolyte consisted of Glomus rubiforme, other Glomus spp. and Acaulospora spp. Although the sporocarpic species, G. aggregatum, G. macrocarpum and G. rubiforme occurred at St. Hippolyte, they were not found at the two less acid sites (Waterloo, Ontario and Lacolle, Québec) . Spores of Acaulospora spp. were found at all three sites, but were most abundant at St. Hippolyte. At St. Hippolyte the total number of spores was much higher than at the other two sites; at Waterloo numbers were an order of magnitude lower than at St. Hippolyte. It is suggested that G. rubiforme and Acaulospora species may be adapted to acid conditions. Seasonal patterns of spore abundance suggested that Acaulospora spp. may sporulate during the spring, whereas G. rubiforme may sporulate during the fall. Accepted: 6 September 1995  相似文献

Due to climate change, the ranges of many North American tree species are expected to shift northward. Sugar maple (Acer saccharum Marshall) reaches its northern continuous distributional limit in northeastern North America at the transition between boreal mixed‐wood and temperate deciduous forests. We hypothesized that marginal fragmented northern populations from the boreal mixed wood would have a distinct pattern of genetic structure and diversity. We analyzed variation at 18 microsatellite loci from 23 populations distributed along three latitudinal transects (west, central, and east) that encompass the continuous–discontinuous species range. Each transect was divided into two zones, continuous (temperate deciduous) and discontinuous (boreal mixed wood), based on sugar maple stand abundance. Respective positive and negative relationships were found between the distance of each population to the northern limit (D_north), and allelic richness (AR) and population differentiation (FST). These relations were tested for each transect separately; the pattern (discontinuous–continuous) remained significant only for the western transect. structure analysis revealed the presence of four clusters. The most northern populations of each transect were assigned to a distinct group. Asymmetrical gene flow occurred from the southern into the four northernmost populations. Southern populations in Québec may have originated from two different postglacial migration routes. No evidence was found to validate the hypothesis that northern populations were remnants of a larger population that had migrated further north of the species range after the retreat of the ice sheet. The northernmost sugar maple populations possibly originated from long‐distance dispersal.  相似文献

The phenolic acids and abscisic acid (ABA) of sugar pine ( Pinus lambertiana Dougl.) embryos and megagametophytes, separated by high-pressure liquid chromatography, were analyzed during 90 days stratification of the seeds. The phenolic acids occurred mainly as glycosides. Following hydrolysis, the majority of phenolics present could be identified as common benzoic and ciranamic acid derivatives. Levels of phenolic acids were relatively low in dormant seeds, but increased substantially in the embryos during stratification at 5°C, particularly cinnamic acid, p -coumaric acid, ferulic acid, and one unknown. This active synthesis during stratification did not support an inhibitory function for phenolic acids. During stratification at 5°C, changes in ABA levels in both tissues followed a triphasic pattern, with no loss during the first 30 days, a significant decrease the second 30 days, and a lesser decrease the last 30 days. Loss of ABA from moist seeds at 25°C occurred three times as rapidly, so that by 30 days the ABA level of these seeds was equivalent to that of seeds stratified 90 days at 5°C; however, dormancy was not alleviated at 25°C. Application of exogenous ABA (10−7 to 10−4M) to stratified seeds did not significantly reduce germination. Together, the above results did not support a primary role for ABA in the maintenance of dormancy in sugar pines.
A correlated increase in phenylpropanoid metabolism and respiratory capacity with increased germinability during stratification suggests that loss of dormancy may be more closely dependent on increased levels of growth promoters or shifts in metabolic pathways.  相似文献

Sugar maple (Acer saccharum) is a shade-tolerant, late successional dominant species in the North American eastern deciduous forest. The objective of this study was to quantify the relationship between climate and radial growth in sugar maple and to identify spatial and temporal patterns in dendroclimatic response. We used a combination of archived sugar maple tree-ring chronologies and newly sampled sites to calculate dendroclimatic response of sugar maple at 13 sites in the United States and Canada. At all sites, sugar maple growth was significantly correlated to monthly temperature, precipitation, or Palmer Drought Severity Index. However, there were no generalizable patterns in sugar maple’s growth response to climate. Individual sites had unique dendroclimatic responses with respect to: a) which climatic variables were correlated to radial growth; b) what months had significant correlations between climate and radial growth; and c) what years had significant correlations between climate and radial growth. The individualistic dendroclimatic response of sugar maple appears to reflect a plastic response of the species to changes in climate perhaps related to its status as a strong competitor in late-successional forests. This ability to survive a wide range of environmental conditions may bode well for the species persistence under variable future climatic conditions. It also points to the need for more research on late-successional species in examining forest response to potential climate change scenarios because these species may be more resilient than early-successional species.  相似文献

Embryos from the mature unstratified loblolly pine ( Pinus taeda L.) seeds used in this study were nondormant: however, they failed to germinate in situ because of constraints imposed by the surrounding tissues. During a stratification period of 35 days of moist chilling at 2°C, seed germinability increased from 19 to 76%. The total lipid content of the megagametophyte did not change during stratification, whereas the total protein content of both megagametophyte and embryo was more variable. The rate of synthesis of buffer soluble proteins in these two tissues increased and electrophoretic analysis showed that while similar proteins were synthesized during the stratification period, changes in the patterns of synthesis of some proteins did occur. In both the embryo and megagametophyte the synthesis of a set of proteins with molecular masses below 46 kDa decreased markedly after 14 days of chilling (DOC). In the megagametophyte, the synthesis of a more diverse set of proteins with molecular masses ranging from 16 to 78 kDa increased after 14 DOC. It is noteworthy that these changes in the patterns of protein synthesis coincided with the greatest relative increase in seed germinability of 35%.  相似文献

Changes in the patterns of gene expression were examined during loblolly pine ( Pinus taeda L.) seed stratification, germination, and post-germinative growth. In both the megagametophyte and the embryo, DNA contents remained relatively constant at all stages examined. RNA contents, however, increased in both tissues following seed germination, particularly in the embryo where a 7-fold increase in the RNA content was observed 5 days after germination. Poly(A)+ RNA, extracted from megagametophytes and embryos, was translated in vitro in a rabbit reticulocyte lysate cell-free system. Analysis of [35S]-methionine-labelled translation products by two-dimensional electrophoresis/fluorography indicated that there were changes in the populations of mRNAs during all developmental stages examined. In both the megagametophyte and the embryo several distinct mRNA populations, including one constitutively present at all stages examined, were identified. One mRNA population, present in the mature seed, decreased during seed stratification. Another population, not present in the mature seed, rose during the period of stratification that coincided with an increase in seed germinability. A third population, which appeared during seed germination, increased steadily during post-germinative growth. Besides these similarities, specific differences between megagametophyte and embryo were noted. For example, one mRNA population, which was present in the megagametophyte of the mature seed and remained constant during the stratification period, disappeared immediately following seed germination. In the embryo, one set of messages was germination specific. In total, these results show that mRNA populations change in a temporal fashion that is consistent with the patterns of de novo protein synthesis known to occur in loblolly pine during the same developmental periods.  相似文献

Aim To evaluate whether seedlings of sugar maple (Acer saccharum Marsh.) can establish beyond the species northern range limit in adjacent boreal forest. Location The hardwood–boreal forest transition zone on the north‐east shore of Lake Superior, Ontario, Canada. Methods Seed fall of sugar maple was monitored for 5 years in a stand of this species at its northern range limit, and seed from this stand was transplanted to five micro‐habitat types in an adjacent boreal forest. The establishment and survival of sugar maple seedlings there, and in the seed‐source stand, was monitored for the following 7–11 years. Soil‐surface light levels were measured in both forest types. Results Most seed fell in the final year of monitoring, when c. 250 seeds m?2 were recorded. First‐year seedling establishment rates in the maple stand, deriving from this mast seed year, was approximately double that deriving from seed transplanted to the boreal forest sites; this is tentatively attributed to seed predator satiation in the maple stand. However, subsequent seedling survivorship in the boreal forest was greater than that in the maple stand, resulting in comparable seedling densities by the end of 6 years. This difference is tentatively attributed to better illumination in the boreal forest sites, and canopy‐opening disturbances appear to be especially facilitative of seedling survival. Main conclusions There is no fundamental impediment to sugar maple seedlings establishing in boreal forest communities if climate warming occurs and seed is available. If management intervention is needed to accelerate seed availability in a rapidly warming boreal forest, then diffuse seed application to disturbed boreal forest sites during mast years of local boreal tree species is recommended as the most effective way of avoiding seed predation and increasing seedling survival.  相似文献

Germinability of isolated embryos from developing fruits of Acer platanoides was high at the earliest developmental stage assessed (90 dpa), but fell subsequently and at seed maturity was very low. These observations showed an inverse correlation with changes in endogenous free abscisic acid (ABA) levels in the embryo, which were low during early ontogeny, but reached maximum levels late in development (150–160 dpa). These observations suggest the possibility that dormancy may be induced during development as a result of ABA accumulation in the embryo, an argument strengthened by the obvious inhibitory effect of added ABA on the germinability of isolated embryos. The cotyledons appear to exert an inhibitory influence on embryo germinability that may result from their free ABA content although the embryonic axis itself possesses an innate dormancy that may reflect its own free ABA content. The increased germinability of isolated embryos resulting form added kinetin serves only to emphasise the complexity of the system and the dangers of simplistic interpretation.The correlation between germinability and ABA content is not complete, however, since much of the reduction in germinability had occurred before any appreciable increase in free ABA levels in the embryo was observed. Indeed the failure of the intact seed to respond to endogenous changes in embryonal ABA levels suggests that even though free ABA in the embryo may influence embryo germinability, it has little effect in the intact seed, where the presence of an intact testa may be a more important factor.The absence of a desiccation phase in the embryo during the late stages of development suggests that the large increases in endogenous free ABA did not cause dormancy by inhibiting water uptake, nor did they result from water stress in the embryonal tissues.  相似文献

During stratification at 5°C indole-3-acetic acid (IAA) levels in embryos of Acer platanoides decreased during the early stages but subsequently increased again throughout the remainder of a 144 day period. The reduction in IAA levels in embryos of fruits stored at 17°C was even more pronounced, and in addition, no increase was observed after longer storage periods at this temperature, the levels of IAA remaining very low. Germination in seeds maintained at 5°C was not observed until after 120 days or longer, but germination potential increased at an earlier stage, as shown by the fact that seeds transferred to 20°C gave appreciable increases in germination after much shorter chilling periods. Endogenous IAA levels in embryos from seeds transferred to 20°C after a chilling period, long enough to break dormancy, increased within 24 h, i.e. before visible germination, to levels similar to those observed in embryos from seeds chilled continuously for 144 days. Embryos from seeds chilled for 120 days, i.e. when the samples already showed visible germination and when the endogenous IAA content was already high, showed no further increase in endogenous IAA during a three day incubation at 20°C. None of the treatments employed was effective in inducing germination of seeds or embryos from fruits stored at 17°C.  相似文献

Seeds of Norway maple ( Acer platanoides L.) differing in water content (10, 20 and 30%) were stored for 6 weeks at 20 to 30°C. During this period changes in phospholipids and fatty acids as well as in seed viability and germination capacity were studied. A considerable decrease in the phospholipid content was observed, which depended on the water content in the seeds and was related to the decrease of the seed germination capacity. The level of linoleic (18:2) and linolenic (18:3) acids in the phospholipid fraction decreased considerably in the course of the accelerated seed ageing. The results obtained suggest that phospholipid degradation and peroxidation of unsaturated fatty acids, followed by membrane destruction, play a considerable role in maple seed ageing.  相似文献

The interactive effects of ozone and light on leaf structure, carbon dioxide uptake and short-term carbon allocation of sugar maple ( Acer saccharum Marsh.) seedlings were examined using gas exchange measurements and 14C-macroautoradiographic techniques. Two-year-old sugar maple seedlings were fumigated from budbreak for 5 months with ambient or 3 × ambient ozone in open-top chambers, receiving either 35% (high light) or 15% (low light) of full sunlight. Ozone accelerated leaf senescence, and reduced net photosynthesis, 14CO2 uptake and stomatal conductance, with the effects being most pronounced under low light. The proportion of intercellular space increased in leaves of seedlings grown under elevated ozone and low light, possibly enhancing the susceptibility of mesophyll cells to ozone by increasing the cumulative dose per mesophyll cell. Indeed, damage to spongy mesophyll cells in the elevated ozone × low light treatment was especially frequent. 14C macroautoradioraphy revealed heterogeneous uptake of 14CO2 in well defined areole regions, suggesting patchy stomatal behaviour in all treatments. However, in seedlings grown under elevated ozone and low light, the highest 14CO2 uptake occurred along larger veins, while interveinal regions exhibited little or no uptake. Although visible symptoms of ozone injury were not apparent in these seedlings, the cellular damage, reduced photosynthetic rates and reduced whole-leaf chlorophyll levels corroborate the visual scaling of whole-plant senescence, suggesting that the ozone × low light treatment accelerated senescence or senescence-like injury in sugar maple.  相似文献

We determined the kind of seed dormancy in Schisandra arisanensis, an ANA grade ([A]mborellales [N]ymphaeales [A]ustrobaileyales) angiosperm with medicinal value. Seeds have small underdeveloped embryos, and following seed maturity their length increased approximately 360% before radicle emergence. Germination was delayed 6–8 weeks, and the percentage and rate were much higher at 15/6, 20/10 and 25/15°C than at 30/20°C. For seeds incubated at 5/5°C (8 weeks) → 15/6°C (4 weeks) → 20/10°C (8 weeks) → 25/15°C (12 weeks) → 20/10°C (5 weeks), embryos grew at 15/6°C → 20/10°C, and almost all seeds that germinated (89%) did so at 20/10°C → 25/15°C. When seeds were incubated in a complementary temperature sequence, 25/15°C (12 weeks) → 20/10°C (8 weeks) → 15/6°C (4 weeks) → 5/5°C (9 weeks) → 15/6°C (4 weeks), embryos grew at 25/15°C → 20/10°C. Nearly all seeds that germinated (93%) did so at 25/15°C → 20/10°C and at 15/6°C following 9 weeks at 5/5°C. Based on the temperature requirements for embryo growth and seed germination, seeds of this species have non‐deep simple morphophysiological dormancy (C1bB).  相似文献

Aims The physiological responses during dormancy removal and multi-phasic germination were investigated in seeds of Paeonia corsica (Paeoniaceae).Methods Seeds of P. corsica were incubated in the light at a range of temperatures (10–25 and 25/10°C), without any pre-treatment, after W (3 months at 25°C), C (3 months at 5°C) and W + C (3 months at 25°C followed by 3 months at 5°C) stratification, and a GA 3 treatment (250 mg·l-1 in the germination substrate). Embryo growth, time from testa to endosperm rupture and radicle emergence were assessed as separate phases. Epicotyl–plumule emergence was evaluated incubating the germinated seeds at 15°C for 2 weeks, at 5 and 25°C for 2 months on agar water before transplanting to the soil substrate at 10, 15 and 20°C and at 15°C for 2 months on the surface agar water with GA 3 .Important findings Embryo growth, testa rupture, endosperm rupture (radicle emergence) and growth of the epicotyl were identified as four sequential steps in seeds of P. corsica. Gibberellic acid alone and warm stratification followed by 15°C promoted embryo growth and subsequent seed germination. Cold stratification induced secondary dormancy, even when applied after warm stratification. After radicle emergence, epicotyl–plumule emergence was delayed for ca. 3 months. Mean time of epicotyl–plumule emergence was positively affected by cold stratification (2 months at 5°C) and GA 3. P. corsica seeds exhibited differential temperature sensitivity for the four sequential steps in the removal of dormancy and germination processes that resulted in the precise and optimal timing of seedling emergence.  相似文献

A study was conducted in the Lower Laurentians of southern Quebec to test the hypothesis that base cation fertilization would improve the nutrient status of declining sugar maples (Acer saccharum Marsh.) and alter the partitioning of leaf C and N. Six 40×40 m plots were delineated in an 80 year old stand of sugar maple. Three plots received a mixture of fertilizer and liming materials (500 kg ha–1 of K2SO4, 250 kg ha–1 of CaCO3 and 250 kg ha–1 of CaMg(CO2)2) in the spring of 1989. Leaves from mid crown of dominant or co-dominant maples were sampled monthly during the 1990 growing season. Trees were cored in 1992 to measure their response in diameter growth. Fertilization increased diameter growth and foliar K concentration of trees but reduced foliar Ca concentration. Fertilization resulted in lower starch concentrations and higher ratios of soluble sugars to starch in June and September, and in higher free amino acid concentrations but lower ratio of total non-structural carbohydrates to free amino acids in September. Leaf proline concentration was correlated with leaf starch concentration (r=0.39). The results suggest that amelioration of K deficiencies in sugar maple through fertilization with a mixture of base cations can increase tree growth and affect the seasonal dynamics of foliar C and N pools.Abbreviations FAA free amino acids - TNC total non-structural carbohydrates  相似文献
