The life cycle of the Antarctic Dry Valley soil nematode, Scottnema lindsayae (Timm 1971) was studied in laboratory culture at two temperatures, 10°C and 15°C. Soil yeast and bacteria isolated with the nematodes were used as the food source. The species reproduced sexually. The higher temperature had a negative effect on the life cycle. The number of eggs per female and the number of juveniles developing per female were greater at 10°C than at 15°C. Juveniles developed faster at 10°C and four juvenile stages were observed outside of the egg at both temperatures. The unusually long life cycle (218 d at 10°C) suggests that more than one austral summer may be required for successful completion. An increase in Dry Valley soil temperatures associated with potential global environmental change may have detrimental effects on soil nematodes.  相似文献

A fungal infection occurred in juvenile coonstripe shrimps,Pandalus hypsinotus, cultured at Hokkaido Institute of Mariculture, Hokkaido, Japan. The fungus was identified asLagenidium myophilum, the same fungus that had previously been isolated from the abdominal muscle of adult northern shrimps,Pandalus borealis, and larvae of the coonstripe shrimp. Histopathologically, numerous nonseptate hyphae were observed in the lesions, and melanized hemocytes were present within the blackened areas. The optimum temperature for growth of the present strain was 25–30°C, and the optimum NaCl concentration for growth was 0.5–1.0%. Its biological characteristics were compared with those ofLagenidium myophilum isolated from diseased larval coonstripe shrimp and adult northern shrimp. The fungus was pathogenic toward shrimps of the genusPandalus, which live in deep sea areas. The fungus could infect shrimps at various stages, from larva to adult.  相似文献

A nitrogen fixing organism containing a plasmid has been isolated from the rhizosphere fraction ofLeptochloa fusca (L) Kunth (kallar grass) growing on saline soils in the Punjab area. This bacterium can grow aerobically in a medium containing 1M NaCl and can fix nitrogen efficiently under microaerobic conditions on semi-solid medium with glucose or sucrose as a carbon source. Maximum N2-fixation in batch cultures occurred with 100 mM NaCl at pH 8.0 and 35°C. DNA hybridization and analysis of the protein pattern were carried out to establish its taxonomic position. On the basis of protein electrophoretic pattern, physiological characteristics, DNA relatedness, and better growth in the presence of high NaCl concentration, we regard this strain as a new species ofKlebsiella.  相似文献

Changes in polyol production and the intracellular amino acid pool were followed during the growth cycle of Debaryomyces hansenii in 4 mM and 2.7 M NaCl media. The intracellular levels of polyols were markedly enhanced by high salinity, the dominant solutes being glycerol in log phase cells and arabinitol in stationary phase cells. At low salinity arabinitol was the most prominent intracellular solute throughout the growth cycle. There were no major changes in the composition of the total amino acid pool with changes in cultural salinity. The amount of total free amino acids related to cell dry weight was 15–50% lower in cells cultured in 2.7 M NaCl as compared to 4 mM NaCl media.After subtraction of contributions from intracellular polyols the calculated cellular C/N ratio was found to be unaffected by cultural age and salinity during the late log and early stationary phase. On prolonged incubation of stationary phase cells, this ratio decreased, particularly at high salinity. The sensitivity of cells towards exposure to high salinity was measured in terms of the length of the lag phase after transference to 2.7 M NaCl media. This lag phase decreased with increasing intracellular polyol concentrations. At a given polyol content, stationary phase cells were considerably less sensitive than were log phase cells.When cultured at high salinity the mutant strain, 26-2b, grew more slowly and retained less of the total polyol produced during the early growth stages than did the wildtype. Exogenously supplied mannitol, arabinitol, and glycerol stimulated the growth of the mutant in saline media. The New Zealand strain of Tolypocladium cylindrosporum was cultured on Sabouraud dextrose agar medium under varying regimes of growth conditions. The isolate exhibited good tolerances to temperature (4–35 °C), pH (3–10) and salinity (0–7% NaCl). Optimal vegetative growth and sporulation were recorded between a temperature range of 20–30° C, pH of 5–6 and a salinity level of 0–2% NaCl. The North American isolate of the fungus showed similar tolerances, while the European isolate was less tolerant.  相似文献

We evaluated the response of the Antarctic soil nematodes Scottnema lindsayae and Plectus antarcticus to various salts (NaCl, MgSO4, KNO3 and NaCl + MgSO4) and salt concentrations in prepared salt solutions ranging from 0.1 to 3 M, and in saturation paste extracts of soils collected from multiple locations where nematode abundance varied from zero to numerous, and where electrical conductivity ranged from 108 to >12,000 μS/cm. Nematode salt tolerance was salt specific; both nematode species survived in low-experimental concentrations of NaCl and MgSO4, and neither species survived in KNO3 solutions of any concentration. There was no survival of nematodes in the saturation paste extracts of highly saline soils (4,100 μS/cm), while survival was over 80–97% in less saline soils (1,945 μS/cm). A 1:1 dilution of these highly saline saturation paste extracts increased S. lindsayae survival to 80%, while survival of P. antarcticus was not observed until dilutions of greater than 200%. The results complement previous studies demonstrating niche partitioning of S. lindsayae and P. antarcticus across salinity gradients and strengthen interpretations of the physiological mechanisms underlying previously reported spatial correlation between soil salinity and nematodes abundance in the Antarctic Dry Valleys.  相似文献

Novel alkaliphilic, mesophilic bacteria were isolated from subseafloor alkaline serpentine mud from the Ocean Drilling Program (ODP) Hole 1200D at a serpentine mud volcano, South Chamorro Seamount in the Mariana Forearc. The cells of type strain ODP1200D-1.5T were motile rods with a single polar flagellum. Growth was observed between 10 and 45–50°C (optimum temperature: 30–35°C, 45-min doubling time), between pH 6.5 and 10.8–11.4 (optimum: pH 8.5–9.0), and between NaCl concentrations of 0 and 21% (w/v) (optimum NaCl concentration: 2.5–3.5%). The isolate was a facultatively anaerobic heterotroph utilizing various complex substrates, hydrocarbons, carbohydrates, organic acids, and amino acids. Nitrate or fumarate could serve as an electron acceptor to support growth under anaerobic conditions. The G+C content of the genomic DNA was 57.5 mol%. Phylogenetic analysis based on 16S rRNA gene sequences indicated that the isolate belonged to the genus Marinobacter and was the most closely related to M. aquaeolei strain VT8T and M. hydrocarbonoclasticus strain SP.17T, while DNA–DNA hybridization demonstrated that the new isolate could be genetically differentiated from the previously described species of Marinobacter. Based on the physiological and molecular properties of the new isolate, we propose the name Marinobacter alkaliphilus sp. nov., type strain: ODP1200D-1.5T (JCM12291T and ATCC BAA-889T).  相似文献

Summary The effect of temperature on the growth rates of Halomonas subglaciescola and a Halobacterium sp., halotolerant and halophilic bacteria isolated from Antarctic saline lakes, was predicted accurately by models which propose a linear relationship between temperature and the square root of the reciprocal of the growth rate. The cardinal temperatures of the strains examined were in general 5–10°C lower than those of taxonomic counterparts isolated from temperate and tropical environments. Temperature dependence data for strains of the Halobacterium sp. and annual temperature profiles of Deep Lake allowed estimation of the in situ potential for growth.  相似文献

A Gram-positive, motile, endospore-forming and rod-shaped halophilic bacterial strain MSS-155 (KCTC 3788 and KCCM 41687) was isolated from a marine solar saltern of the Yellow Sea in Korea and was subjected to a polyphasic taxonomic study. This organism grew at temperature of 10.0–42.0°C with an optimum of 35°C. Strain MSS-155 grew optimally in the presence of 10% NaCl and did not grow in the absence of NaCl. The cell wall peptidoglycan type of strain MSS-155 was A4 based on l-Orn-d-Asp. Strain MSS-155 was also characterized chemotaxonomically by having menaquinone-7 (MK-7) as the predominant isoprenoid quinone and anteiso-C15:0 as the major fatty acid. The DNA G+C content was 44.0 mol%. Phylogenetic analysis based on 16S rDNA sequences showed that strain MSS-155 falls within the radiation of the cluster comprising Halobacillus species. Levels of 16S rDNA sequence similarity between strain MSS-155 and the type strains of four Halobacillus species were in the range 97.6–98.8%. Strain MSS-155 exhibited levels of DNA-DNA relatedness of 6.2–11.2% to the type strains of Halobacillus species described previously. On the basis of phenotypic properties, phylogeny, and genomic data, strain MSS-155 should be placed in the genus Halobacillus as a member of a novel species, for which we propose the name Halobacillus locisalis sp. nov.Communicated by W.D. Summary Isolated flexor muscles of the shore crab,Hemigrapsus edwardsi were maintained in saline solutions for periods of 2–12 h.In hypotonic saline solutions containing less than 400 mM sodium chloride, the fibres rapidly died. In 400 mM sodium chloride saline the fibres showed partial volume readjustment associated with reduction in the amount of intracellular ninhydrin-positive substances (NPS).In saline (460 mM sodium chloride) containing ouabain (2–5 mM) the fibres lost water and potassium, gained sodium and chloride, but the amount of NPS was not significantly changed.In hypertonic saline (550 mM sodium chloride) the fibres showed a partial recovery of volume during the 8 h experimental period. Associated with this was a rise in the amount of intracellular NPS.It was concluded that the ability of the muscle fibres ofHemigrapsus edwardsi to respond to a hyperosmotic challenge in an amino acid free medium, by an increase in intracellular amino acid levels, must represent a synthesis of these compounds from an intracellular source. This may be an adaptive feature of osmotic readjustment in this rather permeable crab.  相似文献

Streptococcus thermophilus strain CNRZ 455 produces a type II restriction endonuclease designated Sth455I. This enzyme was isolated from cell extracts by anionic and cationic exchange chromatography. This yielded an enzyme preparation free of non-specific nucleases. The optimal reaction conditions for Sth455I are: MgCL2, 30 mm; pH range, 8–9; incubation temperature, 37–42°C; and a high NaCl concentration, 100–200 mm. The results of single- and double-digestion experiments indicates that Sth455I is an isoschizomer of BstNI and EcoRII showing different sensitivity to methylation. The enzyme exhibits restriction activity on the DNA of three bacteriophages of S. thermophilus and no activity on the phage lytic for strain CNRZ 455. The restriction/modification system associated with this strain is discussed.  相似文献

AMethanosarcina species, designated strain ChGul, was isolated from goat feces; this is the first fully described pure culture ofMethanosarcina obtained from feces. Antigenic fingerprinting suggests that isolate ChGul is a new immunotype. The mol% G + C content of DNA was 42.2%. Strain ChGul grew on methanol, methylamines, and acetate in a minimal salts medium. It grew on H2-CO2 only after adaptation. Growth occurred as a milky-white suspension and contained cells mostly in doublets and quadruplets of irregular cocci; many cells contained phase bright spots typical of gas vacuoles. The isolate did not grow on formate, or CO2 plus isopropanol, ethanol, or acetone as substrates and did not produce methane from formate. The optimum growth temperature was 35–37°C, and optimum pH was 6.2–6.8. ChGul is unusually sensitive to sulfide and has low tolerance for NaCl. Optimal levels of total sulfide and NaCl for growth were 0.5 mM and 20–40 mM, respectively. Since ChGul requires adaptation for growth on H2-CO2 and cannot use formate, it may be restricted to methylotropic or acetoclastic methanogenesis in the rumen, a function not observed in previously isolated rumen methanogens that use H2-CO2 and formate. Our work suggests that improper NaCl and sulfide concentrations, and cell lysis, may have made isolation of rumenMethanosarcina difficult in the past. It also underscores the need to evaluate feed compositions and media components for most probable number studies, with respect to NaCl and sulfide levels, to understand the role ofMethanosarcina in the rumen.  相似文献

Extraction of nematodes from Dry Valley Antarctic soils   总被引:9,自引:3,他引:6  
Nematode density and taxonomic composition from Dry Valley soil processed by the sugar centrifugation (SC) method in Antarctica was compared to those extracted from soils shipped frozen to the USA and processed by either the SC or Baermann Funnel (BF) (at 5°C and 10°C) techniques. Soil selected for the extraction comparisons represented a wide range of soil properties found in the Dry Valleys. More nematodes were recovered from freshly collected Antarctic soil and from stored frozen soil using the SC technique than from BF at either temperature (P<0.05). Temperature had no effect on nematode densities extracted by the BF. Scottnema lindsayae was the most abundant species recovered by all extraction methods, but recovery was significantly lower from stored soils. Thus, nematodes can be extracted qualitatively following frozen storage using SC, but quantitative studies of nematode populations should be based on soils extracted following field sampling.  相似文献

A new alkaliphilic and moderately halophilic chemoorganotrophic anaerobic bacterium (strain Z-7986), which is spore-forming, rod-shaped, and has a gram-negative cell wall pattern, was isolated from the coastal lagoon mud of the highly mineralized Lake Magadi (Kenya). The organism is an obligatorily carbonate- and sodium chloride-dependent motile peritrichously flagellated rod that grows within a 3–17% NaCl concentration range (with an optimum at 7–12% NaCl) and within a pH range of 7.7–10.3 (with an optimum at pH values of 8–8.5). It is a moderate thermophile with a broad temperature optimum at 36–55°C; maximum growth temperature is 60°C. The bacterium catabolizes glucose, fructose, sucrose, maltose, starch, glycogen, N-acetyl-D-glucosamine, and, to a slight degree, peptone and yeast extract. Its anabolism requires yeast extract or casamino acids. Glucose fermentation yields formate, acetate, ethanol, H2, and CO2. The bacterium is sulfide-tolerant and capable of the nonspecific reduction of S0 to H2S. The G+C content of the DNA is 34.4 mol %. The analysis of the 16S rRNA sequence revealed that strain Z-7986 belongs to the order Haloanaerobiales and represents a new genus in the family Halobacteroidaceae. We suggest the name Halonatronum saccharophilum gen. nov. sp. nov. The type strain of this species is Z-7986T (= DSM13868, = Uniqem*211).  相似文献

A strain of Bacillus sp. coded JMa5 was isolated from molasses contaminated soil. The strain was able to grow at a temperature as high as 45°C and in 250 g/l molasses although the optimal growth temperature was 35–37°C. Cell density reached 30 g/l 8 h after inoculation in a batch culture with an initial concentration of 210 g/l molasses. Under fed-batch conditions, the cells grew to a dry weight of 70 g/l after 30 h of fermentation. The strain accumulated 25–35%, (w/w) polyhydroxybutyrate (PHB) during fermentation. PHB accumulation was a growth-associated process. Factors that normally promote PHB production include high ratios of carbon to nitrogen, and carbon to phosphorus in growth media. Low dissolved oxygen supply resulted in sporulation, which reduced PHB contents and dry weights of the cells. It seems that sporulation induced by reduced supply of nutrients is the reason that PHB content is generally low in the Bacillus strain.  相似文献

Three psychrotrophic Arthrobacter strains, isolated from Antarctic lake sediment (An24, An25T) and deep-sea sediment (ZX6) were studied. Their 16S rRNA gene sequences showed highest similarities (97.0–97.9%) with those of A. nicotianae and A. protophormiae. All three strains underwent rod–coccus morphological change, had high mol% G+C content, were aerobic to slightly anaerobic, and grew between 0°C and 30°C, with optimal growth temperature around 25°C. The cell wall peptidoglycan was A4 variant. DNA–DNA hybridization, physiological and chemotaxonomic studies indicated that these three strains constituted a new homogeneous genomic species within the genus Arthrobacter, for which the name Arthrobacter ardleyensis, with the type strain An25T (CGMCC 1.3685, JCM 12921) was proposed.M. A Bacillus sp. producing a high level of intracellular penicillin G acylase (PAC) was isolated. The PAC production in this strain was induced by phenylacetic acid. Various carbon and nitrogen sources were evaluated for their effect on growth and PAC production at 28 °C and pH 7.0. Cells grown in medium supplemented with sucrose as carbon source and tryptone as nitrogen source produced maximum activity of 6.45 and 8.92 U mg–1, respectively. Maximum concentration of PAC (10.1 Umg–1) was produced by the cells grown in the medium containing sucrose and tryptone, which was twofold higher than the production in basal medium.  相似文献

Candida shehatae NCL-3501 utilized glucose and xylose efficiently in batch cultures. The specific rate of ethanol production was higher with mixtures of glucose and xylose (0.64–0.83 g g–1 cells d–1) compared to that with individual sugars (0.38–0.58 g g–1 cells d–1). Although the optimum temperature for growth was 30°C, this strain grew and produced appreciable levels of ethanol at 45°C. A stable ethanol yield (0.40–0.43 g g–1 substrate utilized) was obtained between 10 g L–1 and 80 g L–1 of initial xylose concentration. Conversion efficiency was further improved by immobilization of the cells in calcium alginate beads. Free or immobilized cells ofC. shehatae NCL-3501 efficiently utilized sugars present in rice straw hemicellulose hydrolysate, prepared by two different methods, within 48 h. Ethanol yields of 0.45 g g–1 and 0.5 g g–1 from autohydrolysate, and 0.37 g g–1 from acid hydrolysate were produced by free and immobilized cells, respectively.  相似文献

A representative of the purple sulfur bacteria was isolated from organic-rich intertidal sediments of Hamelin Pool, Shark Bay, Australia. The isolate, strain HPC, is nutritionally versatile, being capable of photoheterotrophic growth in the absence of reduced sulfur sources and of dark microaerophilic growth, either heterotrophically or lithotrophically. Vitamins are not required. Nine organic carbon substrates, including the C2–C5 fatty acids, support photoheterotrophic growth. The isolate is an obligate halophile capable of growth over a wide salinity range (0.5%–8.5% NaCl). On the basis of its morphology, physiology, pigmentation, and DNA base ratio, strain HPC is considered to be an obligately halophilic representative ofChromatium vinosum.  相似文献
