Spin-echo NMR comparative study of water diffusion in the cortex and stele of maize (Zea mays L.) roots was made with the aim to determine predominant pathways of radial water movement in the root. The root parts examined differed in terms of water diffusion coefficients and sensitivity to HgCl2, the aquaporin blocker. These differences are discussed from the viewpoint of unequal contributions of separate transport pathways (apoplastic, symplastic, and transmembrane) to the overall water flow. Characteristics of water diffusion in roots with the endodermis damaged suggest an inconsiderable contribution of the endodermis into resistance to water movement.  相似文献

Insulin resistance results, in part, from impaired insulin signaling in insulin target tissues. Consequently, increased levels of insulin are necessary to control plasma glucose levels. The effects of elevated insulin levels on pancreatic beta (β) cell function, however, are unclear. In this study, we investigated the possibility that insulin may influence survival of pancreatic β cells. Studies were conducted on RINm, RINm5F and Min-6 pancreatic β-cells. Cell death was induced by treatment with H2O2, and was estimated by measurements of LDH levels, viability assay (Cell-Titer Blue), propidium iodide staining and FACS analysis, and mitochondrial membrane potential (JC-1). In addition, levels of cleaved caspase-3 and caspase activity were determined. Treatment with H2O2 increased cell death; this effect was increased by simultaneous treatment of cells with insulin. Insulin treatment alone caused a slight increase in cell death. Inhibition of caspase-3 reduced the effect of insulin to increase H2O2-induced cell death. Insulin increased ROS production by pancreatic β cells and increased the effect of H2O2. These effects were increased by inhibition of IR signaling, indicative of an effect independent of the IR cascade. We conclude that elevated levels of insulin may act to exacerbate cell death induced by H2O2 and, perhaps, other inducers of apoptosis.  相似文献

This study employed confocal laser scanning microscopy to monitor the effect of H2O2 on cytosolic as well as mitochondrial calcium (Ca2+) concentrations, mitochondrial inner membrane potential (m) and flavine adenine dinucleotide (FAD) oxidation state in isolated mouse pancreatic acinar cells. The results show that incubation of pancreatic acinar cells with H2O2, in the absence of extracellular Ca2+ ([Ca2+]o) led to an increase either in cytosolic and in mitochondrial Ca2+ concentration. Additionally, H2O2 induced a depolarization of mitochondria and increased oxidized FAD level. Pretreatment of cells with the mitochondrial inhibitors rotenone or cyanide inhibited the response induced by H2O2 on mitochondrial inner membrane potential but failed to block oxidation of FAD in the presence of H2O2. However, the H2O2-evoked effect on FAD state was blocked by pretreatment of cells with the mitochondrial uncoupler, carbonyl cyanide p-trifluoromethoxy-phenylhydrazone (FCCP). On the other hand, perfusion of cells with thapsigargin (Tps), an inhibitor of the SERCA pump, led to an increase in mitochondrial Ca2+ concentration and in oxidized FAD level, and depolarized mitochondria. Pretreatment of cells with thapsigargin inhibited H2O2-evoked changes in mitochondrial Ca2+ concentration but not those in membrane potential and FAD state. The present results have indicated that H2O2 can evoke marked changes in mitochondrial activity that might be due to the oxidant nature of H2O2. This in turn could represent the mechanism of action of ROS to induce cellular damage leading to cell dysfunction and generation of pathologies in the pancreas. Mercuric chloride (HgCl2) is a highly toxic compound, which can cause nephrotoxic damage. In the present study effects of HgCl2 on mitochondria integrity and energy metabolism, as well as antidotal effects of 2,3-dimercaptopropane-1-sulfonate (DMPS) were investigated in the opossum kidney derived cell line (OK). OK cell monolayers were incubated during 0, 1, 3, 6, and 9 h in serum-free culture medium containing 15 μM HgCl2, either in the absence or in the presence of 60 μM DMPS in a 1:4 ratio. Intracellular ATP content, MTT reduction, and HSP70/HSP90 induction were studied; confocal, transmission electron microscopy, and light microscopy studies were also performed. For confocal analysis, a mitochondrial selective probe (MitoTracker Red CMXH2Ros) was used. Antioxidant activity of DMPS was also studied by the scavenging of the free radical 2, 2-diphenyl-1-picrylhydrazyl (DPPH) technique. A decrease of ATP content, an impaired ability to reduce tetrazolium, and dramatic changes on cellular and mitochondrial morphology, and energetic levels were found after either 6 or 9 h of HgCl2 exposure. Increased expression of HSP90 and HSP70 were also seen. When OK cells were co-incubated with HgCl2 and DMPS, cellular morphology, viability, intracellular ATP, and mitochondrial membrane potential were partially restored; a protective effect on mitochondrial morphology was also seen. DMPS also showed potent antioxidant activity in vitro. Mitochondrial protection could be the cellular mechanism mediated by DMPS in OK cells exposed to a toxic concentration of HgCl2.  相似文献

With increasing industrialization, numerous air pollutants are generated. This research aimed to investigate the effects of inhalation of oxidative pollutants. H2O2 was used to simulate oxidative air pollutants, and glutathione, a reducing agent that is widely distributed in organisms, was used as an antagonist, to protect cells from oxidative stress. H2O2 was diluted using two gradients (0.05 mM, 0.20 mM, 0.80 mM, 3.20 mM and 0.05 mM, 0.10 mM, 0.15 mM, 0.20 mM) and GSH was dissolved at 20 μM. MTT, MDA, ROS, GSH, and TSLP were used as biomarkers to evaluate oxidative stress and possible resulting molecular events. A dose–response relationship was observed between H2O2 concentrations and the above-mentioned biomarkers. Glutathione significantly reduced levels of oxidative stress.  相似文献

Natural bond orbital (NBO) analyses and dissected nucleus-independent chemical shifts (NICS π z z ) were computed to evaluate the bonding (bond type, electron occupation, hybridization) and aromatic character of the three lowest-lying Si2CH2 (1-Si, 2-Si, 3-Si) and Ge2CH2 (1-Ge, 2-Ge, 3-Ge) isomers. While their carbon C3H2 analogs favor classical alkene, allene, and alkyne type bonding, these Si and Ge derivatives are more polarizable and can favor “highly electron delocalized”? and “non-classical”? structures. The lowest energy Si 2CH2 and Ge 2CH2 isomers, 1-Si and 1-Ge, exhibit two sets of 3–center 2–electron (3c-2e) bonding; a π-3c-2e bond involving the heavy atoms (C–Si–Si and C–Ge–Ge), and a σ-3c-2e bond (Si–H–Si, Ge–H–Ge). Both 3-Si and 3-Ge exhibit π and σ-3c-2e bonding involving a planar tetracoordinated carbon (ptC) center. Despite their highly electron delocalized nature, all of the Si2CH2 and Ge2CH2 isomers considered display only modest two π electron aromatic character (NICS(0) π z z =--6.2 to –8.9 ppm, computed at the heavy atom ring center) compared to the cyclic-C 3H2 (–13.3 ppm).

The genotoxicity of reactive oxygen species (ROS) is well established. The underlying mechanism involves oxidation of DNA by ROS. However, we have recently shown that hydrogen peroxide (H2O2), the major mediator of oxidative stress, can also cause genomic damage indirectly. Thus, H2O2 at pathologically relevant concentrations rapidly induces higher order chromatin degradation (HOCD), i.e. enzymatic excision of chromatin loops and their oligomers at matrix-attachment regions. The activation of endonuclease that catalyzes HOCD is a signalling event triggered specifically by H2O2. The activation is not mediated by an influx of calcium ions, but resting concentrations of intracellular calcium ions are required for the maintenance of the endonuclease in an active form. Although H2O2-induced HOCD can efficiently dismantle the genome leading to cell death, under sublethal oxidative stress conditions H2O2-induced HOCD may be the major source of somatic mutations.  相似文献

In this study, the effect of aucubin on H2O2-induced apoptosis was studied by using a rat pheochromocytoma (PC12) cell line. We have analyzed the apoptosis of H2O2-induced PC12 cells, H2O2-induced apoptosis appeared to correlate with lower Bcl-2 expression, higher Bax expression and sequential activation of caspase-3 leading to cleavage of poly-ADP-ribose polymerase (PARP). Aucubin not only inhibited lower Bcl-2 expression, high Bax expression, but also modulated caspase-3 activation, PARP cleavage, and eventually protected against H2O2-induced apoptosis. These results indicated that aucubin can obstruct H2O2-induced apoptosis by regulating of the expression of Bcl-2 and Bax, as well as suppression of caspases cascade activation.  相似文献

A density functional theory (DFT) study of cct-As, ccc, and cct-CO isomers of the ruthenium dihydride complex RuH2(CO)2(AsMe2Ph)2 is reported (see Scheme for the labeling isomer 34 structures of RuH2(CO)2(AsMe2Ph)2). Complex geometries and relative energies of different isomers have been calculated with both B3LYP and M06-2X functionals. The results show that the B3LYP calculated Boltzmann populations of cct-As, ccc, and cct-CO isomers are 65.5, 34.2, and 0.3%, respectively. These are in better agreement with the experimental data than those calculated at the M06-2X level. However, the calculations of 1H NMR chemical shifts were found to be better described with M06-2X than with B3LYP or with HF level of theories. In addition, a transition state between the two most stable isomers was determined through DFT/(B3LYP or M06-2X) calculations.

Chlorella vulgaris is a unicellular green alga resistant to heavy metals. Chlorella is a rich nutritional ingredient because it contains high levels of antioxidants. The objective of this research was to study if C. vulgaris protect renal cells against mercury-chloride-caused oxidative stress and cellular damage in the kidney. Our results demonstrated that HgCl2 causes oxidative stress and cellular damage, and that C. vulgaris administration prevents oxidative stress and cellular damage in kidney  相似文献

An experimental study has been carried out on the stability of adenine (one of the five nucleic acid bases) under hydrothermal conditions. The experiments were performed in sealed autoclaves at 300 degrees C under fugacities of CO(2), N(2) and H(2) supposedly representative of those in marine hydrothermal systems on the early Earth. The composition of the gas phase was obtained from the degradation of oxalic acid, sodium nitrite and ammonium chloride, and the oxidation of metallic iron. The results of the experiments indicate that after 200 h, adenine is still present in detectable concentration in the aqueous phase. In fact, the concentration of adenine does not seem to be decreasing after approximately 24 h, which suggests that an equilibrium state may have been established with the inorganic constituents of the hydrothermal fluid. Such a conclusion is corroborated by independent thermodynamic calculations.  相似文献

Using the microfluorescent method of identification of damaged plasmatic membranes, the influence of hydrogen peroxide on mice peritoneal macrophages was determined. H2O2 in vitro produced a destructive effect on the cells depending on the temperature, time of incubation, and concentration of the agent. Arsenic trioxide (As2O3) has potential anti-cancer activity against a wide range of carcinomas via apoptosis induction or oncoprotein degradation. The mechanisms involved are not fully elucidated. Here, we demonstrated that As2O3 induced-apoptosis in HeLa and MCF-7 cancer cells was in part triggered by tubulin polymerization. High expression of JWA promoted tubulin polymerization and increased the sensitivity of the cancer cells to As2O3. The activation of the p38 MAPK (mitogen-activated protein kinases) signaling pathway was found to contribute to JWA-promoted tubulin polymerization. Our results suggest that JWA may serve as an effective enhancer of microtubule-targeted As2O3 anti-cancer therapy.  相似文献

PB90 is a novel protein elicitor isolated from Phytophthora boehmeriae. Here, we report that treatment of PB90 stimulates hypericin production and hydrogen peroxide (H2O2) generation in Hypericum perforatum L. cells and demonstrate that H2O2 is essential for PB90-induced hypericin production. To further study the source of PB90-triggered H2O2, we have investigated activities of plasma membrane NADPH oxidase in Hypericum perforatum L. cells subjected to PB90 treatment. It is revealed that treatment of the cells with PB90 significantly increases NADPH oxidase activity. NADPH oxidase inhibitors suppress not only the PB90-stimulated NADPH oxidase activity but also the PB90-triggered H2O2 generation and PB90-induced hypericin production, showing that NADPH oxidase is involved in PB90-triggered H2O2 generation and hypericin production. Moreover, the suppression of NADPH oxidase inhibitors on PB90-induced hypericin production can be reversed by H2O2, although H2O2 per se has no effects on hypericin production of the cells. Together, the data demonstrate that PB90 may induce hypericin production of H. perforatum cells through the NADPH oxidase-mediated H2O2 signaling pathway.  相似文献

Acclimation to excess light is required for optimizing plant performance under natural environment. The present work showed that the treatment of Arabidopsis leaves with exogenous H2O2 can increase the acclimation of PSII to excess light. Treatments with H2O2 also enhanced the capacity of the mitochondrial alternative respiratory pathway and salicylic acid (SA) content. Our work also showed that the lack in alternative oxidase (AOX1a) in AtAOX1a antisense line and the SA deficiency in NahG (salicylate hydroxylase gene) transgenic mutant attenuated the H2O2-induced acclimation of PSII to excess light. It indicates that the H2O2-induced acclimation of PSII to excess light could be mediated by the alternative respiratory pathway and SA.  相似文献

In this paper, we report a study on the structure and first hyperpolarizability of C60Cl2 and C60F2. The calculation results show that the first hyperpolarizabilities of C60Cl2 and C60F2 were 172 au and 249 au, respectively. Compared with the fullerenes, the first hyperpolarizability of C60Cl2 increased from 0 au to 172 au, while the first hyperpolarizability of C60F2 increased from 0 au to 249 au. In order to further increase the first hyperpolarizability of C60Cl2 and C60F2, Li@C60Cl2 and Li@C60F2 were obtained by introducing a lithium atom to C60Cl2 and C60F2. The first hyperpolarizabilities of Li@C60Cl2 and Li@C60F2 were 2589 au and 985 au, representing a 15-fold and 3.9-fold increase, respectively, over those of C60Cl2 and C60F2. The transition energies of four molecules (C60Cl2, Li@C60Cl2, C60F2, Li@C60F2) were calculated, and were found to be 0.17866 au, 0.05229 au, 0.18385 au, and 0.05212 au, respectively. A two-level model explains why the first hyperpolarizability increases for Li@C60Cl2 and Li@C60F2.  相似文献

The interaction of cis-[PtCl2(Me2SO)2] with human serum albumin (HSA) and the sensitivity of the complex to heat denaturation as dependent on the duration of incubation have been studied by UV absorption and fluorescence spectroscopy. Optimal conditions for cis-[PtCl2(Me2SO)2] binding to HSA have been determined. The results are compared with the data for the HSA-cisplatin complex. It has been found that binding of HSA with cis-[PtCl2(Me2SO)2] does not result in significant structural changes of the protein.  相似文献

Functional and reversible plasmonic resonances across the visible and near-infrared spectrum have opened new avenues for developing advanced next-generation nanophotonic devices. In this study, by using optothermally controlled phase-change material (PCM) for plasmonic nanostructures, we successfully induced highly tunable charge transfer plasmon (CTP) resonance modes. To this end, we have chosen a two-member dimer assembly consisting of gold cores and Ge2Sb2Te5 (GST) shells in distant, touching, and overlapping regimes. We show that switching between amorphous (dielectric) and crystalline (conductive) phases of GST allows for achieving tunable dipolar and CTP resonances and enables an effective interplay between these modes along the near-infrared spectrum. By analyzing electromagnetically calculated spectral responses for the dimer antenna in tunneling and direct charge transfer regimes, we confirmed that the induced CTPs in touching and overlapping regimes are highly controllable and pronounced in comparison to the quantum tunneling regime. We also use the precise, fast, and controllable switching between dipolar and CTP resonant modes to develop a telecommunication switch based on a simple metallodielectric dimer. The proposed structures can help designing optothermally controlled devices without morphological variations in the geometry of the design, and having strong potential for advanced plasmon modulation and fast data routing.  相似文献
