Previous work had indicated that the molecular weight calculated from the equilibrium distribution of DNA in a CsCl density gradient appears to be one half the estimated value of the true molecular weight. In the present study, the sedimentation equilibrium of a polyclectrolyte in a density gradient generated by a low molecular weight electrolyte is treated in terms of the representation according to which a constant fraction of the counterions is considered permanently immobilized and the remaining fraction completely free. Equations for the preferential hydration and the molecular weight are obtained. The validity of the treatment is tested by applying it to experimental data on the equilibrium sedimentation of ?X 174 DNA in CsCl, and it is found that the molecular weight calculated in this way is in agreement with the accepted value for this molecule. Also, recalculation of published data on density gradient centrifugation of T2 DNA in CsCl according to the present treatment brings up the molecular weight to within the range of values given by other methods.  相似文献

To model centrifugal sedimentation of biological suspensions, the time history of sedimentation of particles in a centrifugal field was considered for two geometries: a tube and a cylindrical container. The Kynch theory for batch gravitational settling in Cartesian coordinates based on mass conservation was extended to include a centrifugal sedimentation force, cylindrical coordinates, and the Hawksley-Vand hindered settling model. The resulting quasi-linear partial differential equation was solved by the method of characteristics. The combination of radial dependence of the sedimentation force and cylindrical geometry in the centrifugal case results in several differences in the time-position history diagram of the sedimentation process compared to the gravitational case. First, instead of a region of uniform concentration equal to the initial concentration, a region of concentration that is continuously decreasing with time results. Second, in the region of particle accumulation, curved constant concentration contours result instead of straight lines. Finally, a secondary shock that is dependent upon the initial concentration and the radius ratio of the rotating vessel appears in the centrifugal case. The time history of the concentration for a particle suspension with an initial concentration typical of blood is presented.  相似文献

The opportunity to trace the evolution of a triplet repeat is rare, especially for seed-plant lineages with a well-defined fossil record. Microsatellite PtTX2133 sequences from 18 species in 2 conifer genera were used to calibrate the birth of a CAGn repeat, from its protomicrosatellite origins to its repeat expansion. Birth occurred in the hard-pine genome ~ 136 million years ago, or 14 million generations ago, then expanded as a polymorphic triplet repeat 136-100 million years before a major North American vicariance event. Calibration of the triplet-repeat birth and expansion is supported by the shared allelic lineages among Old and New World hard pines and the shared alleles solely among North American diploxylon or hard pines. Five CAGn repeat units appeared to be the expansion threshold for Old and New World diploxylon pines. Haploxylon pine species worldwide did not undergo birth and repeat expansion, remaining monomorphic, with a single imperfect 198-bp allele. A sister genus, Picea, had only a region of cryptic simplicity, preceding a proto-microsatellite region. The polymorphic triplet repeat in hard pines is older than some long-lived microsatellites reported for reptiles, yet younger than those reported for insects. Some cautionary points are raised about phylogenetic applications for this long-lived microsatellite.  相似文献

Splicing of mammalian introns requires that the intron possess at least 80 nucleotides. This length requirement presumably reflects the constraints of accommodating multiple snRNPs simultaneously in the same intron. In the free-living nematode, C. elegans, introns typically are 45 to 55 nucleotides in length. In this report, we determine whether C. elegans introns can obviate the mammalian length requirement by virtue of their structure or sequence. We demonstrate that a 53 nucleotide intron from the unc-54 gene of C. elegans does not undergo splicing in a mammalian (HeLa) nuclear extract. However, insertion of 31 nucleotides of foreign, prokaryotic sequence into the same intron results in efficient splicing. The observed splicing proceeds by the same two-step mechanism observed with mammalian introns, and exploits the same 3' and 5' splice sites as are used in C. elegans. The branch point used lies in the inserted sequence. We conclude that C. elegans splicing components are either fewer in number or smaller than their mammalian counterparts.  相似文献

130 Ram testes have been used in the study, and the following methods were applied to fill and study the testical arteries: r?ntgenography, stereor?ntgenography, corosive method, Indian ink and gel injections. Variabilities occur as to the site where the a.testicularis arises in rams. Individual differences can also be observed in the arrangement of the epididymal arteries, in most cases, however, they arise from the first loops of the convolution. Both epididymal arteries are considerably thinner than the testicular artery, forming 2 independent vessel convolutions, located at both sides of the convolution of the a.testicularis, and not interfering with the loops of the latter. A.accessoria testis has been found in one ram only. 2 to 3 large waved loops can be observed in the pars marginalis and the a.testicularis of the ram. Bifurcation mostly occurs in the second third of the margo epididymidis. Individual variations have been found at further ramification of the rr.testiculares. The shape of the lobulus testis is indicated by the centripetal branch with its centrifugal twigs. In rams the a.ductus deferentis forms anastomoses both with the branches running from the a.epididymidis caudalis and the a.cremasterica.  相似文献

The role of cells capable of formation of megakaryocytic colonies (CFUm) in regulation of thrombocytopoiesis was studied using a simulation model of megakaryocytopoiesis. The CFUm were shown to be the least differentiated cells involved in regulation of megakaryocytopoiesis in mice upon immune thrombocytopenia and after intravenous injection of hydroxyurea. A correlation was found between the CFUm population productivity and the number of cells in a transit population of the megakaryocytic line which makes it possible to reproduce experimental kinetics in the model. The duration of cell development from CFUm to megakaryocytes is about 3 days.  相似文献

In the preceding article equations describing relevant ion flows in illuminated suspensions of bacteriorhodopsin liposomes have been derived. Here these equations are subjected to experimental tests. Changes in permeability characteristics of the liposomal membrane are brought about by addition of specific ionophores and change of medium composition. Using light-driven proton uptake and electrochemical potential differences for protons across the membrane as observation parameters, ridig attempts to falsify the derived equations are unsuccessful. Agreement between equations and experimental results is established on the point of: (i) the antagonistic effect of valinomycin and nigericin on the two components of the proton-motive force, (ii) the time dependence of the changes in transmembrane electrical and chemical potential differences after the onset of illumination. In three independent experimental systems evidence was obtained for the correctness of the postulated dependence of the turnover rate of the photochemical cycle on back pressure by the transmembrane electrochemical potential difference for protons.  相似文献

A stand of Stratiotes aloides L. in a riverside lagoon, Abborravan on the Vindelälven river in N Sweden, was studied for 3 years (1975–1977). The stand was vegetative, at all times submerged and therefore never flowering. On average each plant produced 3 adventitious roots and 40 leaves. One or two turions were also produced each year, together with stolons and associated offsets. At the beginning of June 1977 the population density was 42 plants/m2 and the biomass was 290 kg/ha (dry wt. basis). At the end of September that year the respective values were 90/m2 and 756 kg/ha. The dry wt./fresh wt. ratio of the biomass changed little during that period, from 6.4 to 6.5%. These and other results are compared with the respective data for Stratiotes stands in Central European lakes. In Abborravan Stratiotes seems well-adapted to survive the prevailing, hard, winter conditions. Freezing in situ is quite normal and is only lethal if the overwintering basal rosettes become frozen solid. The turions are quite frost-hardy.  相似文献
