Hybrid peptides were constructed from endothelin B receptor (ETB) selective antagonist RES-701-1 (1) and endothelin (ET-1). They have N-terminal 10 amino acids derived from 1 and C-terminal 10 amino acids derived from ET-1. RES-701-1(1-10)-[Ala15]ET-1(12-21) and its analogues substituted or truncated at the residues derived from RES-701-1 had proved to possess high receptor binding activity selective for ETB as well as 1. Substitutions at the residues derived from ET-1 had produced some analogues that possessed high affinity not only for ETB but for ETA. Although all analogues had antagonistic effects on ETA, some analogues had proved to function as agonist on ETB confirmed by the changes in intracellular calcium concentrations of ET receptor-transfected COS-7 cells. We have found four types of ET receptor-binding peptides: (1) ETB-selective agonist with weak ETA antagonism (3, KT7421); (2) ETB-selective antagonist with weak ETA antagonism (29, KT7539); (3) ETB agonist with potent ETA antagonism (27, KT7538); and (4) non-selective ETA/ETB antagonist (26, KT7540).  相似文献

The substrate range of the haem monooxygenase cytochrome P450cam (CYP101) has been broadened by site-directed mutagenesis. The hydroxylation selectivity of five mutants at the 96 position towards a range of substrates has been used to investigate P450cam -substrate molecular recognition. The substrates contained aromatic and activated and unactivated aliphatic C---H bonds, as well as reactive functional groups. Diphenylmethane, diphenylether, diphenylamine, and 1,1-di-phenylethylene were all hydroxylated regiospecifically at the para position, with no attack at the amine or the olefinic double bond. With benzylcyclohexane the activated benzylic and tertiary C---H bonds were not attacked, and the reactions catalysed by the Y96G and Y96A mutants were highly diastereoselective, with 4-trans-benzylcyclohexanol constituting 90% of the products. 1-Phenyl-1-cyclohexylethylene was oxidised predominantly at the 4-position of the cyclohexane ring without attack at the olefinic double bond, and approximately equal amounts of cis- and trans-4-phenylethenylcyclohexanol were formed. These results show that P450cam can be engineered to oxidise C---H bonds without attacking more reactive functional groups.  相似文献

A model series of 5-HT2C antagonists have been prepared by rapid parallel synthesis. These N-substituted phenyl-N′-pyridin-3-yl ureas were found to have a range of 5-HT2C receptor affinities and selectivities over the closely related 5-HT2A receptor. Extrapolation of simple SAR, derived from this set of compounds, to the more active but synthetically more complex 1-(3-pyridyl-carbamoyl)indoline series allowed us to target optimal substitution patterns and identify potent and selective 5-HT2C/2B antagonists.  相似文献

2-Chloro-5′-N-methylcarboxamidoadenosine analogues containing the (N)-methanocarba (bicyclo[3.1.0]hexane) ring system as a ribose substitute display increased selectivity as agonists of the human A3 adenosine receptor (AR). However, the selectivity in mouse was greatly reduced due to an increased tolerance of this ring system at the mouse A1AR. Therefore, we varied substituents at the N6 and C2 positions in search of compounds that have improved A3AR selectivity and are species independent. An N6-methyl analogue was balanced in affinity at mouse A1/A3ARs, with high selectivity in comparison to the A2AAR. Substitution of the 2-chloro atom with larger and more hydrophobic substituents, such as iodo and alkynyl groups, tended to increase the A3AR selectivity (up to 430-fold) in mouse and preserve it in human. Extended and chemically functionalized alkynyl chains attached at the C2 position of the purine moiety preserved A3AR selectivity more effectively than similar chains attached at the 3-position of the N6-benzyl group.  相似文献

Novel 5-HT1 autoreceptor ligands based on the N-4-aryl-piperazinyl-N′-ethyl-5,6,7,8-tetrahydropyrido[4′, 3′:4,5]thieno[2,3-d]pyrimidin-4(3H)-one core are described. Aiming at antidepressants with a novel mode of action our objective was to identify potent antagonists showing balanced affinities and high selectivity for the 5-HT1A and 5-HT1B receptors. Strategies for the development of dual 5-HT1A and 5-HT1B antagonists based on 1 and 2 as leads and the corresponding results are discussed. Isoquinoline analogue 33 displayed high affinity and an antagonistic mode of action for the 5-HT1A and the 5-HT1B receptors and was characterized further with respect to selectivity, electrically stimulated [3H]5-HT release and in vivo efficacy.  相似文献

The chloro complexes trans-[Pt(Me)(Cl)(PPh3)2], after treatment with AgBF4, react with 1-alkynes HC---C---R in the presence of NEt3 to afford the corresponding acetylide derivatives trans-[Pt(Me) (C---C---R) (PPh3)2] (R = p-tolyl (1), Ph (2), C(CH3)3 (3)). These complexes, with the exception of the t-butylacetylide complex, react with the chloroalcohols HO(CH2)nCl (n = 2, 3) in the presence of 1 equiv. of HBF4 to afford the alkyl(chloroalkoxy)carbene complexes trans-[Pt(Me) {C[O(CH2)nCl](CH2R) } (PPh3)2][BF4] (R = p-tolyl, N = 2 (4), N = 3 (5); R=Ph, N = 2 (6)). A similar reaction of the bis(acetylide) complex trans-[Pt(C---C---Ph)2(PMe2Ph)2] with 2 equiv. HBF4 and 3-chloro-1-propanol affords trans-[Pt(C---CPh) {C(OCH2CH2CH2Cl)(CH2Ph) } (PMe2Ph)2][BF4] (7). T alkyl(chloroalkoxy)-carbene complex trans-[Pt(Me) {C(OCH2CH2Cl)(CH2Ph) } (PPh3)2][BF4] (8) is formed by reaction of trans-[Pt(Me)(Cl)(PPh3)2], after treatment with AgBF4 in HOCH2CH2Cl, with phenylacetylene in the presence of 1 equiv. of n-BuLi. The reaction of the dimer [Pt(Cl)(μ-Cl)(PMe2Ph)]2 with p-tolylacetylene and 3-chloro-1-propanol yields cis-[PtCl2{C(OCH2CH2CH2Cl)(CH2C6H4-p-Me}(PMe2Ph)] (9). The X-ray molecular structure of (8) has been determined. It crystallizes in the orthorhombic system, space group Pna21, with a = 11.785(2), B = 29.418(4), C = 15.409(3) Å, V = 4889(1) Å3 and Z = 4. The carbene ligand is perpendicular to the Pt(II) coordination plane; the PtC(carbene) bond distance is 2.01(1) Å and the short C(carbene)-O bond distance of 1.30(1) Å suggests extensive electronic delocalization within the Pt---C(carbene)---O moietry.  相似文献

Stimulation of rat cerebral cortex with endothelin-1 (ET-1) caused an increase in the tyrosine phosphorylation of several proteins. Two of these phosphoproteins were identified by the immunoprecipitation assays as being the focal adhesion kinase p125FAK and crk-associated substrate p130Cas. This effect was time- and dose-dependent, with an EC50 value of 3.9×10−8 M. In addition, the cerebral cortex ET receptor subtype involved in this action was determined by using BQ-123 and BQ-788, which are ETA and ETB receptor antagonists respectively. Our results indicate that the ET-1 effect on protein tyrosine phosphorylation occurred through ETB receptors. The requirement for extracellular Ca2+ on ET-1 action was also studied. ET-1-stimulated tyrosine phosphorylation of both p125FAK and p130Cas was abolished in the absence of external Ca2+ or in the presence of nimodipine, a Ca2+ channel-blocker. These results suggest that the ET-1-stimulated protein tyrosine phosphorylation was secondary to Ca2+ influx through the dihydropyridine Ca2+-channel. In slices where protein kinase C was inhibited, ET-1-stimulated tyrosine phosphorylation of both proteins was reduced. These results indicate that ET-1 modulates the tyrosine phosphorylation of specific proteins, which may be involved in adhesion processes in the brain.  相似文献

The synthesis and structure-activity relationships of a series of 5,7-diarylcyclopenteno[1,2-b]pyridine-6-carboxylic acids are described. Our efforts have been focused on modification of the aryl ring at the 5-position and the alkyl substituent at the 2-position of the bottom 4-methoxyphenyl ring in an effort to develop orally available ET(A) selective antagonists with safer profiles in terms of the P-450 enzyme inhibitory activity. Incorporation of a hydroxymethyl group as an alkyl substituent in methylenedioxyphenyl and 6-dihydrobenzofuran derivatives led to the identification of orally bioavailable ET(A) selective antagonists 1f and 7f. These compounds also showed not only excellent binding affinity (IC(50) < 0.10nM, more than 800-fold selectivity for the ET(A) receptor over the ET(B) receptor) but also sufficient oral bioavailability, 48% and 56%, respectively, in rats. Furthermore, these compounds did not exhibit either competitive or mechanism-based inhibition of human cytochrome P450 enzymes.  相似文献

Cyclizations of alkylhydrazines with N-acyl-S-methylisothioureas, readily synthesized from acyl chlorides, sodium thioisocyanate, dialkylamines then methyl iodide in a one-pot reaction, gave 1-alkyl-3-dialkylamino-5-phenyltriazoles 7 as major products. The regioisomers were assigned through the use of NOE NMR experiments. While bearing a N-bis(cyclopropyl)methyl-N-propylamino group, this series of compounds shows very good binding affinity on the human CRF1 receptor. Among them, 1-methyl-3-[N-bis(cyclopropyl)methyl-N-propylamino]-5-(2,4-dichlorophenyl)-1H-[1,2,4]triazole 7a had the best binding affinity for the CRF1 receptor (Ki=9 nM).  相似文献

Structure-activity relationships have been investigated through substitutions at the 9-position of the 2-amino-6-(2-furanyl) purine (5) to identify novel and selective A(2A) adenosine receptor antagonists. N-Arylated chitosans were synthesized via Schiff bases formed by the reaction between the primary amino group of chitosan with aromatic aldehydes followed by reduction of the Schiff base intermediates with sodium cyanoborohydride. Treatment of chitosan containing N,N-dimethylaminobenzyl and N-pyridylmethyl substituents with iodomethane under basic conditions led to quaternized N-(4-N,N-dimethylaminobenzyl) chitosan and quaternized N-(4-pyridylmethyl) chitosan. Methylation occurred at either N,N-dimethylaminobenzyl and N-pyridylmethyl groups before the residual primary amino groups of chitosan GlcN units were substituted. The total degree of quaternization of each chitosan varied depending on the extent of N-substitution (ES) and the sodium hydroxide concentration used in methylation. Increasing ES increased the total degree of quaternization but reduced attack at the GlcN units. N,N-dimethylation and N-methylation at the primary amino group of chitosan decreased at higher ES’s. Higher total degrees of quaternization and degrees of O-methylation resulted when higher concentrations of sodium hydroxide were used. The molecular weight of chitosan before and after methylation was determined by gel permeation chromatography under mild acidic condition. The methylation of the N,N-dimethylaminobenzyl derivative with iodomethane was accompanied by numerous backbone cleavages and a concomitant reduction in the molecular weight of the methylated product was observed. The antibacterial activity of water-soluble methylated chitosan derivatives was determined using Escherichia coli (Gram-negative) and Staphylococcus aureus (Gram-positive) bacteria; minimum inhibitory concentrations (MIC) of these derivatives ranged from 32 to 128 μg/mL. The presence of the N,N-dimethylaminobenzyl and N-pyridylmethyl substituents on chitosan backbone after methylation did not enhance the antibacterial activity against S. aureus. However, N-(4-N,N-dimethylaminobenzyl) chitosan with degree of quaternization at the aromatic substituent and the primary amino group of chitosan of 17% and 16–30%, respectively, exhibited a slightly increased antibacterial activity against E. coli.  相似文献

Fulvenes (1a–e) derived from condensation of cyclopentadiene with acetone or a variety of aldehydes were treated with LiPAr2 (Ar = phenyl, p-tolyl) to yield the respective substituted (diarylphosphinomethyl)cyclopentadienides (2, 3). Subsequent reaction with ZrCl4(THF)2 gave the respective bis[(diarylphosphinomethy])cyclopentadienyl]zirconium dichlorides ( Ar = phenyl (4), p-tolyl (5)). The complex rac-[C5H4-CH(CH3)-PPh2]2ZrCl2 (rac-4b) was characterized by X-ray diffraction. The reaction of complexes 4a and 5a [(Cp-CMe2-PAr2)2ZrCl2] with PdCl2(NCPh)2 or PtCl2(NCPh)2 leads to the formation of the trans-(metallocene-chelate-phosphane)metal complexes 6–9 (e.g. trans-Cl2Pd(Ph2P-CMe2-Cp)2ZrCl2]. Chloride abstraction from the reaction product of [Cp-CH(CMe3)PPh2]2ZrCl2 with PdCl2(NCPh)2 eventually gave the cationic complex [meso,trans-(Cp-CH(CMe3)PPh2)2(Cl)Zr(μ-Cl)Pd(Cl)]+ (10) that was also characterized by X-ray diffraction. It features a dimetallabicyclic framework with two Cp-CHR-PPh2 ligands and a chloride bridging between the early and the late transition metal center.  相似文献

This research was undertaken to study the enzymatic deacylation of l,2-diacyl-sn-glycero-3-phosphocholines (sn-1,2-PC) to sn-glycerol-3-phosphocholine (GPC); this compound could be an useful intermediate in the synthesis of “structured” sn-1,2-PC, after re-acylations of the two sn-positions of the glycerol backbone. The enzymatic reactions represent a valid alternative to the chemical deacylation that can be simply obtained in alkaline conditions. High conversion were achieved using a lipase selective for the sn-1-position of sn-1,2-PC (Lipozyme IM, from Mucor miehei) together with a Phospholipase A2 from hog pancreas, enzyme selective for the sn-2-position; the best results were obtained carrying out the enzymatic reaction in a microemulsion system.  相似文献

Herein we describe the SAR of 1,5-biaryl pyrrole derivatives, with substituents in the 6-position of the benzoic acid moiety, as EP(1) receptor antagonists. Substitution at this position was well tolerated and led to the identification of several analogues with high affinity for the EP(1) receptor that displayed good efficacy in the established FCA model of inflammatory pain. Furthermore, several analogues were prepared which combined substitution at the 5- and 6-positions as well as derivatives with an aromatic ring fused to the 5- and 6-positions.  相似文献

The electrocatalytic oxidation of NADH, ascorbate, urate, xanthine and H2O2 at different polypyrrole electrodes has been investigated. The conducting polymer was grown on platinum, glassy carbon, or graphite electrodes and modified by means of enclosed redox-active anions or other redox-active compounds covalently bound to either the N- or the β-position of the pyrrole. Copolymers of pyrrole and N-substituted pyrrole derivatives of chloranil or 2,3-dicholoro-1,4-naphthoquinone showed outstanding electrocatalytic properties for the oxidation of NADH. The application of these electrodes in amperometric steady-state measurements or flow-injection systems in combination with dehydrogenase reactions has been possible.  相似文献

The structure-activity relationship (SAR) of the vinyl pyridine region of himbacine derived thrombin receptor (PAR-1) antagonists is described. A 2-vinylpyridyl ring substituted with an aryl or a heteroaryl group at the 5-position showed the best overall PAR-1 affinity and pharmacokinetic properties. One of the newly discovered analogs bearing a 5-(3-pyridyl) substituent showed excellent PAR-1 affinity (Ki = 22 nM) and oral activity with reduced ClogP and improved off-target selectivity compared to an earlier development candidate.  相似文献

The synthesis of tetradentate cobalt(II) salicylaldiminato complexes and their catalytic evaluation in the oxidation of phenol is reported. Tetradentate cobalt complexes [Co(Ln)2] were obtained by reacting the N-(aryl)salicylaldimine ligands (HL1–HL6) with cobalt acetate in a 2:1 mole ratio. All complexes were characterized using FT-IR and elemental analysis. X-ray crystal structures of complexes 1, 2 and 5 have also been obtained. All complexes were found to be active for the oxidation process.  相似文献
