Laminaria longissima and Zaohoucheng No.1 (a commercial variety selected from Laminaria japonica) differ to a certain extent in their morphological characteristics and biological habits. It was assumed that varieties bred through their hybridization should exhibit high yield potential and tolerate relatively high seawater temperatures. Female gametophyte clones isolated from L. longissima were crossed with male clones isolated from Zaohoucheng No.1. Laminaria variety 90-1 was obtained after gametophyte crossing, continuous self-crossing and selection. This variety was genetically homozygous; the indices of variation of blade length, width and thickness of the final two selection cycles were 7–8%; i.e., not different significantly. Variety 90-1 grew faster, lost less tissue and had higher yield potential than two widely used commercial varieties of L. japonica (all commercial varieties currently used in China originate from this latter species). The blade of variety 90-1 increased 3.71 cm day−1 on average during the whole period of cultivation, almost two-fold that of two controls, and growth was maintained even when seawater temperature was higher than 18°C–3°C higher than the temperature tolerated by other Laminaria varieties. Variety 90-1 increased yield by more than 70% over two controls and also synthesized desirable amounts of iodine, mannitol and algin. In blade length, variety 90-1 was more similar to L. longissima than to L. japonica, but more similar to L. japonica in blade width and thickness. Since the adoption of variety 90-1 in 1999, its culturing area has increased each year to reach its current area of 7,000 ha, i.e., almost one-third of the total cultivation acreage of Laminaria in China. Breeding of variety 90-1 has demonstrated that it is feasible to develop elite Laminaria varieties by crossing gametophytes from different Laminaria species in combination with successive self-crossing and selection.  相似文献

Laminaria is the main seaweed species under commercial cultivation in China, which utilizes 80 x 109 artificially cultured seedlings each year. This paper reports the large-scale application of seedling production using vegetative gametophytes in Laminaria cultivation. Key techniques, such as the attachment of gametophytes to the collectors and tank culture of seedlings in the hatchery, are examined. The problems of detachment and irregular dispersion of seedlings were also investigated. An efficient and practical seedling production technology was developed, which consists of seeding gametophytes on collectors, control of seawater flow in tanks, washing of collectors and regulation of ambient light, seawater temperature and nutrition supplement. After 2 months of culture, seedlings grown on woven palm rope collectors 0.5 cm in diameter measured up to the national technical standard of China (i.e., > 20 individuals cm−1 at a length of > 1.5 cm, and > 100 individuals cm−1 at a length of 0.2 cm–1.5 cm, desolation rate < 1% and detachment rate < 5%). Compared with spore-derived seedlings, no significant differences in sporophyte recruitment and dispersion pattern were observed. Large-scale production of seedlings for commercial cultivation of 4,000 ha from 2001 to 2006 was conducted successfully.  相似文献

This study assessed the possibility of using drift and subtidal seaweeds from St Lawrence Island, Alaska (lat. 63°N) for sale by the native population after simple processing. Over 125 km of coastline were surveyed for distribution of both drift and subtidal seaweeds. Drift seaweed wet weight densities ranged from 0.2 to over 9 kg m−2, with an average of over 4 t km−1 in the areas sampled. Attached, benthic seaweed densities ranged from 0.15 to 0.32 kg m−2. Thirty and 35% of the biomass was composed of Agarum cribrosum and species of Laminaria, respectively, both as drift and as benthic seaweed. Data from tagged Laminaria indicated that growth rates were relatively slow for most of the year. The drift seaweed resource on the coasts south and west of the city of Gambell appeared to have good potential for a small-scale commercial harvest. (*author for correspondence)  相似文献

Background

We evaluated the Ziehl-Neelsen staining (ZNS) technique for the diagnosis of paragonimiasis in Laos and compared different modifications of the ZNS techniques.

Methodology

We applied the following approach: We (1) examined a paragonimiasis index case''s sputum with wet film direct examination (WF) and ZNS; (2) re-examined stored ZNS slides from two provinces; (3) compared prospectively WF, ZNS, and formalin-ether concentration technique (FECT) for sputum examination of patients with chronic cough; and (4) compared different ZNS procedures. Finally, we assessed excess direct costs associated with the use of different diagnostic techniques.

Principal Findings

Paragonimus eggs were clearly visible in WF and ZNS sputum samples of the index case. They appeared brownish-reddish in ZNS and were detected in 6 of 263 archived ZNS slides corresponding to 5 patients. One hundred sputum samples from 43 patients were examined with three techniques, which revealed that 6 patients had paragonimiasis (13 positive samples). Sensitivity per slide of the FECT, ZNS and the WF technique was 84.6 (p = 0.48), 76.9 (p = 0.25) and 61.5% (p = 0.07), respectively. Percentage of fragmented eggs was below 19% and did not differ between techniques (p = 0.13). Additional operational costs per slide were 0 (ZNS), 0.10 US$ (WF), and 0.79 US$ (FECT). ZNS heated for five minutes contained less eggs than briefly heated slides (29 eggs per slide [eps] vs. 42 eps, p = 0.01). Bloodstained sputum portions contained more eggs than unstained parts (3.3 eps vs. 0.7 eps, p = 0.016).

Conclusions/Significance

Paragonimus eggs can easily be detected in today''s widely used ZNS of sputum slides. The ZNS technique appears superior to the standard WF sputum examination for paragonimiasis and eliminates the risk of tuberculosis transmission. Our findings suggest that ZNS sputum slides should also be examined routinely for Paragonimus eggs. ZNS technique has potential in epidemiological research on paragonimiasis.  相似文献

The growth of 22 strains of Azolla pinnata R. Br., 3 strains of A. filiculoides Lam. and one strain each of A. mexicana Presl and A. caroliniana Willd. was tested separately in liquid culture media kept in controlled, artificial light (30 klux) growth cabinets. Three temperature levels were used: 33°C (37/29°C day/night), 29°C (33/25°C) and 22°C (26/18°C)/ Photoperiod was 12 h a day.For most A. pinnata strains (except three) and an A. mexicana strain the maximum weekly relative growth rate was higher at 33°C than at 22°C, but not for A. filiculoides and A. caroliniana. The highest value of maximum relative growth rate corresponded to 1.9 doubling days and in most strains this occurred in the first week. As the plants grew, the growth rate slowed down more severely at higher temperatures. The maximum biomass was higher at 22°C than at 33°C in all strains. At 22°C, it took 30–50 days to attain maximum biomass and the highest value was 14 g N m?2 or 320 g dry m?2 by A. caroliniana, followed by 12 g N m?2 or 290 g dry wt. m?2 by one strain of A. filiculoides. At 29°C, the maximum biomass was attained in 20–35 days. The highest value was 6.3 g N m?2 or 154 g dry wt. m?2 by A. caroliniana. At 33°C, most A. pinnata strains gave a maximum biomass of less than 4 g N m?2 after 13–23 days, while some strains grew up to 30 days, resulting in a higher maximum biomass. The highest maximum biomass at 33°C was 5.5 g N m?2 or 140 g m?2 dry wt. by A. pinnata from Cheng Mai while the maximum biomass of A. filiculoides and A. caroliniana was much less. Azolla filiculoides requires lower temperature than other species for its growth. Azolla pinnata has the best tolerance to high temperatures among the four species. Azolla mexicana could not be discriminated from A. pinnata in its response to temperature. Azolla caroliniana may keep an intermediate position between A. filiculoides and A. pinnata in temperature response.The formation of ammonia in the medium was examined and it occurred mostly under stationary growth conditions, but, at 33°C, some strains of A. pinnata and A. mexicana released or formed ammonia at 0.3–0.8 μg N ml?1 per week during their initial exponential growth stage.  相似文献

A circadian rhythm in growth was detected by computer-aided image analysis in 3–4-cm-long, juvenile sporophytes of the kelp species Pterygophora California Rupr. and in seven Laminaria spp. In P. californica, the free-running rhythm occurred in continuous white fluorescent light, had a period of 26 h at 10°or 15°C, and persisted for at least 2 weeks in white or blue light. The rhythm became insignificant in continuous green or red light after 3 cycles. Synchronization by white light-dark regimes, e.g. by 16 h light per day, resulted in an entrained period of 24 h and in a shift of the circadian growth minimum into the middle of the light phase. A morning growth peak represented the decreasing portion of the circadian growth curve, and an evening peak the increasing portion. The circadian growth peak was not visible during the dark phase, because growth rate decreased immediately after the onset of darkness. At night, some growth still occurred at 16 or 12 h light per day, whereas growth stopped completely at 8 h light per day, as in continuous darkness. During 11 days of darkness, the thallus area became reduced by 3.5%, but growth rate recovered in subsequent light–dark cycles, and the circadian growth rhythm reappeared in subsequent continuous light.  相似文献

Encarsia tricolor Foërster is a facultative autoparasitoid that develops on the important pestTrialeurodes vaporariorum (Westwood) in outdoor crop conditions, which makes this aphelinid species promising for biological control programs in regions where field and protected crops coexist. In this paper we report the results obtained in the study of daily and totalE. tricolor egg laying and of adult female preference for different host stages in which to lay eggs at constant temperatures in the range 10 to 32 °C. Only whitefly nymphs were present in the searching arena (tomato leaflets). The mean number of eggs laid per female in one day ranged from 4.0 (10 °C and 32 °C) to 15.2 (24 °C). The mean total number of eggs increased with temperature from 10 to 28 °C, reaching a maximum of 123 eggs per female at 28 °C, and decreased sharply from 28 to 32 °C. The relation between the intrinsic rate of increase (rm) and temperature in the range 10 to 28 °C followed a straight line whose equation was rm=?0.076+0.011*T (R2=0.99). The rm ofE. tricolor was greater than the rm ofT. vaporariorum when temperature was higher than 9.2 °C. The preference for any particular host instar in which to lay eggs was not always significant. However, N4 was the host instar preferred whenever preference was statistically significant.  相似文献

Studies were conducted to determine the effect of insecticide residues on germination of primary conidia ofentomophthora muscae (Cohn) Fresenius and to determine the relative susceptibility of healthy and infectedMusca domestica L. to insecticides. Primary conidia were discharged fromM. domestica cadavers onto glass microscope slides treated with insecticides. All insecticides significantly inhibited germination relative to control slides. Permethrin and naled had the least inhibition (36–40 %), while malathion and dimethoate almost completely inhibited germination (87–94 %). Tetrachlorvinphos/dichlorvos and carbaryl were intermediate (57–59 %). Healthy flies were not significantly different from infected flies in terms of susceptibility to naled, dimethoate, or permethrin. Infected flies exposed on the last day of pathogen incubation died on the bottom of the Petri dishes, rather than climbing and attaching as did untreated, infected flies. Both control and treated groups produced large numbers of primary conidia.  相似文献

Algae biofuel has the potential to replace fossil fuels. However, cultivation and productivity of target algae need improvement, while controlling undesired organisms that can lower the efficiency of production systems. A central composite design and response surface model were utilized to predict cultivation optima of marine microalga, Nannochloropsis salina, under a suite of environmental parameters. The effects of salinity, pH, and temperature and their interactions were studied on maximum sustainable yield (MSY, a measure for biomass productivity), lipid content of N. salina, and invading organisms. Five different levels of each environmental predictor variable were tested. The environmental factors were kept within ranges that had previously been determined to allow positive N. salina growth (14.5–45.5 PSU; pH 6.3–9.7; 11–29 °C). The models created for this experiment showed that N. salina’s MSY and lipid content are not strongly affected over the broad range of salinity and temperature values. Calculated optima levels were 28 PSU/20 °C for MSY and 14.5 PSU/20 °C for lipid accumulation, but neither value significantly influenced the model. However, pH was the most important factor to influence algae productivity, and pH optimum was estimated around 8. Both MSY and lipid content were strongly reduced when pH deviated from the optimum. Occurrence of invading organisms seemed stochastic, and none of the environmental factors studied significantly influenced abundance. In conclusion, pH should be kept around 8 for maximum productivity of N. salina. Temperature and salinity should be kept around 20 °C and 28 PSU; however, moderate variations are not too much of a concern and might enhance lipid content of N. salina.  相似文献

We compared the ability of different plant-based expression platforms to produce geraniol, a key metabolite in the monoterpenoid branch of the terpenoid indole alkaloid biosynthesis pathway. A geraniol synthase gene isolated from Valeriana officinalis (VoGES) was stably expressed in different tobacco systems. Intact plants were grown in vitro and in the greenhouse and were used to generate cell suspension and hairy root cultures. VoGES was also transiently expressed in N. benthamiana. The highest geraniol content was produced by intact transgenic plants grown in vitro (48 μg/g fresh weight, fw), followed by the transient expression system (27 μg/g fw), transgenic plants under hydroponic conditions in the greenhouse and cell suspension cultures (16 μg/g fw), and finally hairy root cultures (9 μg/g fw). Differences in biomass production and the duration of cultivation resulted in a spectrum of geraniol productivities. Cell suspension cultures achieved a geraniol production rate of 1.8 μg/g fresh biomass per day, whereas transient expression produced 5.9 μg/g fresh biomass per day (if cultivation prior to agroinfiltration is ignored) or 0.5 μg/g fresh biomass per day (if cultivation prior to agroinfiltration is included). The superior productivity, strict process control and simple handling procedures available for transgenic cell suspension cultures suggest that cells are the most promising system for further optimization and ultimately for the scaled-up production of geraniol.  相似文献

The first capture of the Greenland (or Black) halibut Reinhardtius hippoglossoides in the Arctic Ocean on the border with the East Siberian Sea (79°03′–79°08′ N 139°59′–141°16′ E, 259–277 m) and the repeated findings of the species in the Laptev Sea (78°03′–78°04′ N 132°56′–133°04′ E, 307 m; 78°33′–78°35′ N 138°44′–138°48′ E, 125 m) are reported. Fish (79 specimens with the length of 15.0–44.5 cm) were caught during four bottom trawlings on the edge of the continental shelf of the two seas in the transformed waters of Atlantic origin, as evidenced by the discovery of the indicator species of these waters—the Glacier lanternfish Benthosema glaciale (Myctophidae)—in one of the stomachs of halibut. The other captures of Greenland halibut in the Arctic were also reviewed.  相似文献

The aims of the present study were to develop a parameterization of a one-year-long observed PAR time-series, apply the PAR parameterization in a primary production relation, and compare calculated and observed time-series of primary production. The PAR parameterization was applied in the generally used relation for the primary production (P d): P d = a(BI 0 Z 0) + b with observed photic depth (Z 0) and Chl-a concentrations (B). It was tested whether the PAR parameterization in combination with this simple relation for primary production was able to describe the actual measured primary production. The study is based on a one year long time-series of PAR, CTD-casts (n = 45), and primary production measurements (n = 24) from Århus Bay (56°09′ N; 10°20′ E), south west Kattegat. Results showed a high and positive correlation between observed and calculated primary production in the bay, as based on the present PAR parameterization combined with the simple primary production relation. The developed PAR parameterization, which calculates total daily surface irradiance per day (M photons m?2 d?1), can be applied in any ecological application taking into account that it was developed for the latitude of 56° N.  相似文献

Apanteles sp.group ultor [Hym.: Braconidae] is the dominant and most widely distributed parasite ofEctomyelois ceratoniae (Zeller). The parasitization percent was increased from 10 % during April to 35 % at the end of the pomegranate fruiting season at October. Female parasite preferred to infest the host larvae at the age of, 2–3 days than 7 day old. The average number of host larvae parasitized by single female parasite was 58 under the rearing conditions of 27±2 °C., 55±10% RH and photoperiod of 16 h light per day. Lydella thompsoni Hertin,Tachinidae, is an endoparasitoid frequently associated in southern France with larvae ofOstrinia nubilalis, Pyralidae, Sesamia nonagrioides, Noctuidae, Archanara geminipuncta andA. dissoluta, Noctuidae. The tachinid was reared successfully at 21°C in the laboratory by providing the adult flies with a high humidity, a light intensity of 8,000–10,000 lux for mating and a mixture of casein proteolysate and honey as food. Under such conditions, the flies lived for about 30 days and about half of them mated successfully. Hosts were infested by dissecting mature tachinid females and placing 1–2 of the extracted planidia onto each moth larva. Half of the planidia successfully entered their larval hosts. The biology of the tachinid larvae was studied on the 2 main hosts,O. nubilalis andS. nonagrioides. At 21°C, 25°C and 28°C, larval development took less time onO. nubilalis than onS. nonagrioides. At 25°C, female larvae onO. nubilalis required 9.0±0.5 days and onS. nonagrioides 10.5±0.3 days, male larvae onO. nubilalis required 8.3±0.5 days and onS. nonagrioides 10.6±0.3 days. Pupal duration was also influenced by the larval host. In winter, 2nd instar larvae ofL. thompsoni enter a resting or quiescent condition. This condition is terminated sooner (December–January) in larvae developing onS. nonagrioides than in those developing onO. nubilalis (February–March). The life cycle of the tachinid in the field was studied by trapping flies in water dishes and by collecting parasitized host larvae from various plants. Flies were caught from April to October, mainly in September. Larvae of the spring generation of the parasitoid developed on larvae of species ofArchanara that fed on the reed,Phragmites communis. From 1976 to 1982, parasitism averaged about 16%. Parasitism by summer generation onS. nonagrioides was highest (4–5%) in June. Parasitism ofO. nubilalis did not change by more than 2 fold in either of the 2 summers studied (10–17 % in 1981, 6–10 % in 1982). By September the numbers of host larvae had increased to 10–20 times the number available earlier in the season. Larval populations ofL. thompsoni similarly increased from 3–400 larvae per ha in June to 4–5,000 per ha in September. The stability of the relationship between this parasitoid and its hosts in southeastern France is discussed and compared to relationship described elsewhere.  相似文献

A gelatin film procedure was used to localize proteolytic activity in the lumen of the intestinal ceca of Leucochloridiomorpha constantiae (Trematoda) adults. Slides were coated with a 7·5% gelatin solution and then fixed in 10% neutral buffered formalin (NBF). Cryostat sections of isolated worms or those attached to the bursa of Fabricius of the domestic chick were affixed to the gelatin film. Experimental and control slides were incubated in a humid chamber for 30 min at 37·5 and 4°C, respectively. Slides were again fixed in NBF, and then stained for protein with mercuric bromphenol blue (MBB). In experimental slides, the lumen of the intestinal ceca was lysed and did not stain, whereas worm and host tissue and the gelatin were protein-positive. Control sections stained uniformly positive for protein. In this procedure tissue is retained on the slide and proteolytic activity can be correlated with a tissue site on the same slide.  相似文献

Abstract

The accuracy of comparative genomic hybridization (CGH) analysis is affected by hybridization efficiency. We describe here a simple method for enhancing hybridization efficiency. The hybridization procedure is essentially the same as that of conventional methods. Hybridization solution containing denatured DNA probe mixture was applied to a metaphase chromosome slide or DNA chip slide and covered with a coverslip. In the new method, however, the slide was inverted by turning the coverslip downward prior to hybridization. We termed this method the inverted slide method. To estimate the efficiency of the new method, metaphase chromosome slides and DNA chip slides were treated by both the conventional and inverted slide methods and incubated in a moist chamber at 37°C for 12, 24, 48, and 72 h. Hybridization signals were approximately 1.5 to 2 times brighter on the slides using the inverted slide method than those using the conventional method after 48 and 72 h of incubation. Furthermore, topographical differences in fluorescence intensity were smaller in slides using the inverted-slide method than in those prepared by the conventional method. The inverted slide method is methodologically very simple and improves the resolution of CGH.  相似文献

The effects of day length on development and reproductive maturation of zoophytophagous bug Macrolophus pygmaeus (= M. nubilis) were investigated under laboratory conditions using two strains originated from the environs of Rome, Italy (41.75°N, 12.30°E and 41.95°N., 12.80°E) and Sochi, Krasnodar Territory, Russia (43.9°N, 39.3°E). The insects were kept under day length of either 10 or 16 h at a constant temperature of 20°C. Nymphs and adults were fed on the grain moth eggs. Embryonic development lasted 18–20 days, nymphal development, 25–30 days, and reproductive maturation of females, 4–6 days. The short day length (10 h) resulted in 1–2-day prolongation of nymphal development and a longer maturation time in individuals from both strains, whereas the duration of embryo development was independent of photoperiod. Under both photoperiods, males developed faster than females. Under the short day length, females of the Rome strain matured markedly faster than those of the Sochi strain. Faster development under low temperature and stronger tendency to delay the reproductive maturation observed in the Sochi strain could be explained by a relatively fast (when compared with Rome) autumnal decrease in temperature. The mechanism and adaptive value of the long-day acceleration of development are not yet clear.  相似文献

The reproductive tract of the parasitoidMicroctonus hyperodae was found to comprise 6.1±0.2 ovarioles containing a total of 40–60 oöcytes. After oviposition into its hostListronotus bonariensis, the parasitoid's egg volume increased by 205 times prior to hatching. At 19.1°C ovipositingM. hyperodae survived for a mean 21±4 days and laid a mean of 48±8 eggs.M. hyperodae collected from Colonia, Uruguay laid a mean of 62±15 eggs which was significantly more than the other ecotypes. Under caging conditions with an ample supply of hosts,M. hyperodae laid 51% of its eggs in the first 72 hours and on average 41% of the species' life-span occurred after the exhaustion of its egg supply. Minimum temperature forM. hyperodae oviposition was found to be c. 5°C; beyond this the rate of increase in egg-laying was approximately linear until 30°C whereafter the rate fell abruptly. There was effectively no egg-laying at 39°C. The parasitoid showed no preference for one host sex or the other either in the laboratory or under field conditions. Compared to otherMicroctonus spp.,M. hyperodae appeared to show low fecundity and high longevity. The adaptive implications of this are discussed.  相似文献

Egg maturation in Calliphora vicina is known to occur within a wide range of temperatures, from 12°C to nearly 30°C (Vinogradova, 1991). Photoperiodism has no effect on this process. Some females enter diapause already at 20°C; their fraction increases at lower temperatures and reaches 100% at 6°C. Reproducing females with eggs can survive for a long time and even lay eggs at low temperatures (4–5°C). Experiments with C. vicina from Leningrad Province revealed some effects of the diet (liver or fish) and temperature on the fly reproduction. At 20 and 25°C, 7–10-day old females begin to oviposit, but at 20°C egg maturation is observed in 98% of females feeding on liver and in only 5% of females feeding on fish. On the liver diet, the mean daily fecundity is significantly correlated with the day of oviposition but not with the temperature. At 20°C a significant correlation is observed between the mean daily fecundity and both the day of oviposition and food. The total number of eggs laid by flies after feeding on fish is half that produced after feeding on liver. The optimal conditions for Calliphora vicina cultivation are a 16-h light day, temperatures within the range from 20 to 25°C, and liver as food.  相似文献
