Sieved agricultural soil samples were treated with the anti-knock agent tetraethyl lead (Et4Pb), and the resulting effects were analyzed by microcalorimetry. Et4Pb additions resulted in an increase of the heat production rate, provided that oxygen was present and that the soil was not autoclaved. The increased heat production rate was accompanied by degradation of Et4Pb, as verified by speciation analysis (GC-MS) of the remaining Et4Pb and its ionic degradation products (triethyl lead and diethyl lead cations). Conclusive evidence was obtained that these transformations were mediated mainly by microbes. At an initial Et4Pb concentration of 2 g Pb/kg dry weight the biodegradation rate was about 780 μmol day−1 kg dry weight−1, whilst the chemical decomposition was only 50 μmol day−1 kg dry weight−1. A fivefold rise of the initial Et4Pb concentration resulted in a decrease of the biodegradation rate to 600 μmol day−1 kg dry weight−1 and an increase of the chemical decomposition to 200 μmol day−1 kg dry weight−1. The biodegradation rate was not influenced by the addition of glucose, which means that no indication for a cometabolic attack of Et4Pb was found.   Elimination of n-butanol from the gas phase was examined with a mixed culture in a compact bioscrubber. The extent of the cell concentration was limited by the supply of n-butanol, phosphate or potassium, and the growth rate was determined by the dilution rate. With n-butanol as the limiting substrate the cellular yield was 0.53 g dry cell weight/g n-butanol. Phosphate limitation decreased this yield to 0.34 g and potassium limitation to 0.31 g dry cell weight/g n-butanol at a dilution rate of 0.1/h. Under these conditions n-butanol was eliminated from the gas phase by 84%–100%. In the same order of limitations the specific degradation rate ranged from 0.19 g to 0.32 g n-butanol g dry cell weight−1 h−1. The fraction of n-butanol required to satisfy the needs for maintenance energy increased significantly depending on the limiting nutrient. Limitation by n-butanol, phosphate or potassium caused a maintenance requirement of 0.07, 0.16 and 0.34 g n-butanol g dry cell weight−1 h−1, thus showing a fivefold increase. This high demand for the carbon source demonstrated the feasibility of operating a bioscrubber under mineral limitation to reduce biomass formation significantly, and to maintain a high degree of substrate elimination from the gas phase. The physiological characteristics of growth and pentachlorophenol degradation of the bacteria Sphingomonas chlorophenolica RA2 and Mycobacterium chlorophenolicum PCP-1 were studied quantitatively in liquid culture under various conditions of pH, temperature, pO2, pCO2 and PCP concentration. Concerning their metabolic properties, RA2 and PCP-1 can be regarded as r-strategist and K-strategist, respectively. RA2 showed a higher activity concerning growth and PCP degradation than PCP-1 under optimum conditions. However, PCP-1 performed better under extreme conditions. Maximum growth rates or RA2 and PCP-1 on glucose were 0.21 h−1 and 0.024 h−1 and maximum PCP degradation rates 315 and 40 μmol (g of dry cells)−1 h−1, respectively. Optimized cultivation for RA2 on a technical scale led to the production of 40 g L−1 of cell dry mass within 55 h. The cultivation strategy including pH-controlled ammonium feeding can be used to effectively produce sufficient biomass of both strains for both research and application as inoculants in soil clean-up. Fructose and H2 were compared as electron donors for hydrogenation of carbon-carbon double bonds using Acetobacterium woodii. Caffeate was used as a model substrate. An electron donor was required and both fructose and H2 were suitable. With fructose as the donor, the K s for caffeate was 0.5 mM and the V max was 678 mmol kgdry weight −1 h−1.␣Fructose oxidation was coupled very efficiently to caffeate reduction by an alteration in the fructose fermentation so that acetate was no longer produced. Oxygen consumption was measured in five Dermophis mexicanus and averaged (±SEM) 0.047 ± 0.004 ml O2 g−1 h−1. Carbon dioxide production averaged 0.053 ± 0.005 ml CO2 g−1 h−1 in the same five animals 1 week later. This metabolic rate is similar to metabolic rates of other Gymnophionans but lower than metabolic rates reported for Anurans and Urodeles. Total nitrogen excretion averaged 1.37 μmol N g−1 h−1 which is higher than that found for other amphibians. Of this, 82.5% (1.13 μmol N g−1 h−1) was in the form of urea while 17.5% (0.24 μmol N g−1 h−1) was in the form of NH3 + NH+ 4. Such ureotelism is typical of terrestrial amphibians like D. mexicanus. Osmotic water flux averaged 0.0193 ml g−1 h−1 in control (sham injected) animals and was not significantly altered by injection of either arginine vasotocin or mesotocin. This osmotic flux is similar to osmotic fluxes found for other terrestrial amphibians. The combined data suggest that metabolism in D. mexicanus is, like most other Gymnophionans, lower than other amphibians. The high rates of nitrogen (especially urea) excretion suggests that this fossorial animal accumulates urea like other burrowing amphibians. Accepted: 27 June 2000  相似文献

A thermostable lipase was produced in continuous cultivation of a newly isolated thermophilic Bacillus sp. strain IHI-91 growing optimally at 65 °C. Lipase activity decreased with increasing dilution rate while lipase productivity showed a maximum of 340 U l−1 h−1 at a dilution rate of 0.4 h−1. Lipase productivity was increased by 50% compared to data from batch fermentations. Up to 70% of the total lipase activity measured was associated to cells and by-products or residual substrate. Kinetic and stoichiometric parameters for the utilisation of olive oil were determined. The maximal biomass output method led to a saturation constant K S of 0.88 g/l. Both batch growth data and a washout experiment yielded a maximal specific growth rate, μmax, of 1.0 h−1. Oxygen uptake rates of up to 2.9 g l−1h−1 were calculated and the yield coefficient, Y X/O, was determined to be 0.29 g dry cell weight/g O2. From an overall material balance the yield coefficient, Y X/S, was estimated to be 0.60 g dry cell weight/g olive oil. The research into kinetics of styrene biodegradation by bacterial strain Pseudomonas sp. E-93486 coming from VTT Culture Collection (Finland) was presented in this work. Microbial growth tests in the presence of styrene as the sole carbon and energy source were performed both in batch and continuous cultures. Batch experiments were conducted for initial concentration of styrene in the liquid phase changed in the range of 5–90 g m−3. The Haldane model was found to be the best to fit the kinetic data, and the estimated constants of the equation were: μ m = 0.1188 h−1, K S = 5.984 mg l−1, and K i = 156.6 mg l−1. The yield coefficient mean value Y\textxs\textapp Y_{\text{xs}}^{\text{app}} for the batch culture was 0.72 gdry cells weight (gsubstrate)−1. The experiments conducted in a chemostat at various dilution rates (D = 0.035–0.1 h−1) made it possible to determine the value of the coefficient for maintenance metabolism m d = 0.0165 h−1 and the maximum yield coefficient value Y\textxs\textM = 0.913 Y_{\text{xs}}^{\text{M}} = 0.913 . Chemostat experiments confirmed the high value of yield coefficient Y\textxs\textapp Y_{\text{xs}}^{\text{app}} observed in the batch culture. The conducted experiments showed high activity of the examined strain in the styrene biodegradation process and a relatively low sensitivity to inhibition of its growth at higher concentrations of styrene in the solution. Such exceptional features of Pseudomonas sp. E-93486 make this bacterial strain the perfect candidate for technical applications.  相似文献

The influence of (NH4)2SO4 concentration and dilution rate (D) on actual and potential H2 photoproduction has been studied in ammonium-limited chemostat cultures of Rhodobacter capsulatus B10. The actual H2 production in a photobioreactor was maximal (approx. 80 ml h−1 l−1) at D = 0.06 h−1 and 4 mM (NH4)2SO4. However, it was lower than the potential H2 evolution (calculated from hydrogen evolution rates in incubation vials), which amounted to 100–120 ml h−1 l−1 at D = 0.03–0.08 h−1. Taking into account the fact that H2 production in the photobioreactor under these conditions was not limited by light or lactate, another limiting (inhibiting) factor should be sought. One possibility is an inhibition of H2 production by the H2 accumulated in the gas phase. This is apparent from the non-linear kinetics of H2 evolution in the vials or from its inhibition by the addition of H2; initial rates were restored in both cases after the vials had been refilled with argon. The actual H2 production in the photobioreactor at D = 0.06 h−1 was shown to increase from approximately 80 ml h−1 l−1 to approximately 100 ml h−1 l−1 under an argon flow at 100 ml min−1. Under maximal H2 production rates in the photobioreactor, up to 30% of the lactate feedstock was utilised for H2 production and 50% for biomass synthesis. Continuous hydrogen gas evolution by self-flocculated cells of Enterobacter aerogenes, a natural isolate HU-101 and its mutant AY-2, was performed in a packed-bed reactor under glucose-limiting conditions in a minimal medium. The flocs that formed during the continuous culture were retained even when the dilution rate was increased to 0.9 h−1. The H2 production rate increased linearly with increases in the dilution rate up to 0.67 h−1, giving maximum H2 production rates of 31 and 58 mmol l−1 h−1 in HU-101 and AY-2 respectively, at a dilution rate of more than 0.67 h−1. The molar H2 yield from glucose in AY-2 was maintained at about 1.1 at dilution rates between 0.08 h−1 and 0.67 h−1, but it decreased rapidly at dilution rates more than 0.8 h−1. The role of gibberellins (GAs) in the regulation of shoot elongation is well established but the phytohormonal control of dry-matter production is poorly understood. In the present study, shoot elongation and dry-matter production were resolved by growing Brassica napus L. seedlings under five light intensities (photon flux densities) ranging from 25 to 500 μmol m−2 s−1. Under low light, plants were tall but produced little dry weight; as light intensity was increased, plants were progressively shorter but had increasing dry weights. Endogenous GAs in stems of 16- and 17-d-old plants were analyzed by gas chromatography-selected ion monitoring with [2H2] internal standards. The contents of GAs increased dramatically with decreasing light intensity: GA1, GA3, GA8 and GA20 were 62, 15, 16 and 32 times higher, respectively, under the lowest versus highest light intensities. Gibberellin A19 was not measured at 25 μmol m−2 s−1 but was 9␣times greater in the 75 compared to 500 μmol m−2 s−1 treatment. Shoot and hypocotyl lengths were closely positively correlated with (log) GA concentration (for example: r 2 = 0.93 for GA1 and hypocotyl length) but shoot dry matter was negatively correlated with GA concentration. The application of gibberellic acid (GA3) produced elongation of plants grown under high light, indication that their low level of endogenous GA was limiting shoot elongation. Although endogenous GA20 showed the greatest influence of light treatment, metabolism of [3H]GA20 and of [3H]GA1 was only slightly influenced by light intensity, suggesting that neither 2β- nor 3β-hydroxylation were points of metabolic regulation. The results of this study indicate that GAs control shoot elongation but are not directly involved in the regulation of shoot dry weight in Brassica. The study also suggests a role of GAs in photomorphogenesis, serving as an intermediate between light condition and shoot elongation response. Many fungi (particularly the white rot) are well suited for treatment of a broad range of textile dye effluents due to the versatility of the lignin-degrading enzymes produced by them. We have investigated decolourization of a number of recalcitrant reactive azo and acid dyes using the culture filtrate and purified laccase from the fungus Cyathus bulleri. For this, the enzyme was purified from the culture filtrate to a high specific activity of 4,022 IU mg−1 protein, produced under optimized carbon, nitrogen and C/N ratio with induction by 2,6-dimethylaniline. The protein was characterized as a monomer of 58±5.0 kDa with carbohydrate content of 16% and was found to contain all three Cu(II) centres. The three internal peptide sequences showed sequence identity (80–92%) with laccases of a number of white rot fungi. Substrate specificity indicated highest catalytic efficiency (k cat/K M) on guaiacol followed by 2,2′-azino-bis(3-ethylthiazoline-6-sulfonic acid) (ABTS). Decolourization of a number of reactive azo and acid dyes was seen with the culture filtrate of the fungus containing predominantly laccase. In spite of no observable effect of purified laccase on other dyes, the ability to decolourize these was achieved in the presence of the redox mediator ABTS, with 50% decolourization in 0.5–5.4 days.  相似文献

The effects of the substrate conditions on the volumetric productivity of Lactobacillus helveticus at different cell densities up to 60 g l−1 in a continuous stirred-tank reactor with microfiltration to retain the biomass were investigated. At low dilution rates, D, the steady-state volumetric productivity, r p, gradually increased to a maximum at D = 1.2–1.5 h−1, because of reduced product inhibition. At higher D values, r p unexpectedly decreased, although the substrate conditions further improved. The maxima of r p at different cell densities coincided with a critical specific substrate utilization rate beyond which the cell metabolism seems to be controlled through a catabolic modulator factor, and r p decreases. The feeding dynamics and oxygen uptake of the bottom-dwelling caridean shrimp Nauticaris marionis were studied during the April/May 1984, 1996 and 1997 cruises to Marion Island (Prince Edward Islands, Southern Ocean). N. marionis is thought to have an opportunistic feeding mode. Prey composition varied considerably between the years and sites investigated. Overall, benthic (mainly hydrozoans and bottom-dwelling polychaetes) and, at times, pelagic (largely euphausiids and copepods) prey items dominated in the stomachs of N. marionis both by occurrence and by volume. Generally, pelagic prey contributed more to the diets of smaller shrimps, while benthic prey was a more important component in the guts of larger specimens. Wet, dry and ash-free dry weight were determined for specimens used in respiration experiments. The respiration rates of N. marionis females with carapace length 6.6–11.1 mm ranged from 80 to 250 μl O2 individual−1 · h−1, or from 0.588 to 2.756 μl O2 · mg−1 dry weight h−1. Regression analyses showed highly significant correlations between oxygen consumption and carapace length for N. marionis. Daily ingestion rates estimated using an in situ gut content analysis technique (4.4% of body dry weight) and an energy budget approach (average 4.7% of body dry weight, range 2.0–7.5%) showed good agreement with each other. To assess the energetics of migration in an anadromous fish, adult American shad (Alosa sapidissima) were swum in a large respirometer at a range of speeds (1.0–2.3 body lengths (BL) s−1, 13–24 °C). Metabolic rate (MO2) was logarithmically related to swimming speed (Bl s−1; r 2 = 0.41, slope = 0.23 ± 0.037) and tailbeat frequency (beats × min−1; r 2 = 0.52, slope = 0.003 ± 0.0003). Temperature had a significant effect on metabolic rate (r 2 = 0.41) with a Q10 of 2.2. Standard metabolic rate (SMR), determined directly after immobilization with the neuroblocker gallamine triethiodide, ranged from 2.2–6.2 mmolO2 kg−1 h−1 and scaled with mass (W) such that SMR = 4.0 (±0.03)W0.695(±0.15). Comparison of directly determined and extrapolated SMR suggests that swimming respirometry provides a good estimate of SMR in this species, given the differences in basal activity monitored by the two methods. Overall, American shad metabolic rates (MO2 and SMR) were intermediate between salmonids and fast-swimming perciforms, including tunas, and may be a result of evolutionary adaptation to their active pelagic, schooling life history. This study demonstrates variability in metabolic strategy among anadromous fishes that may be important to understanding the relative success of different migratory species under varying environmental conditions. Biodegradation of propanol and isopropanol by a mixed microbial consortium   总被引:1,自引:0,他引:1  
The aerobic biodegradation of high concentrations of 1-propanol and 2-propanol (IPA) by a mixed microbial consortium was investigated. Solvent concentrations were one order of magnitude greater than any previously reported in the literature. The consortium utilized these solvents as their sole carbon source to a maximum cell density of 2.4 × 109 cells ml−1. Enrichment experiments with propanol or IPA as carbon sources were carried out in batch culture and maximum specific growth rates (μmax) calculated. At 20 °C, μ max values were calculated to be 0.0305 h−1 and 0.1093 h−1 on 1% (v/v) IPA and 1-propanol, respectively. Growth on propanol and IPA was carried out between temperatures of 10 °C and 45 °C. Temperature shock responses by the microbial consortium at temperatures above 45 °C were demonstrated by considerable cell flocculation. An increase in propanol substrate concentration from 1% (v/v) to 2% (v/v) decreased the μ max from 0.1093 h−1 to 0.0715 h−1. Maximum achievable biodegradation rates of propanol and IPA were 6.11 × 10−3% (v/v) h−1 and 2.72 × 10−3% (v/v) h−1, respectively. Generation of acetone during IPA biodegradation commenced at 264 h and reached a maximum concentration of 0.4% (v/v). The results demonstrate the potential of mixed microbial consortia in the bioremediation of solvent-containing waste streams. Songbirds are widely studied to investigate the hormonal control of behavior. However, little is known about the effects of steroids on neurotransmission in these birds. We used electrophysiological and pharmacological techniques to characterize γ-aminobutyric acid (GABA) type A receptors (GABAA) of primary cultured telencephalic and hippocampal neurons from developing zebra finches. Additionally, their modulation by 17β-estradiol(E2), 5α- and 5β-dihydrotestosterone (DHT), 5α- and 5β-pregnan-3α-ol-20-one, and corticosterone was examined. Whole-cell GABA-evoked currents were inhibited by picrotoxin (10 μmol l−1) and bicuculline methiodide (10 μmol l−1) and potentiated by pentobarbital (100 μmol l−1) and propofol (3 μmol l−1). Loreclezole (10 μmol l−1) potentiated GABA-evoked currents, suggesting the presence of β2, β3 and/or β4 subunits. Diazepam (1 μmol l−1) potentiated currents, while Zn2+ (1 μmol l−1) caused no inhibition, indicating the presence of γ subunits. 5α- and 5β-Pregnan-3α-ol-20-one (100 nmol l−1) potentiated currents, whereas E2 (1 μmol l−1), 5α- and 5β-DHT (1 μmol l−1), and corticosterone (10 μmol l−1) had no detectable effect. We conclude that zebra finch telencephalic and hippocampal GABAA receptors include α, β, and γ subunits and are similar to their mammalian counterparts in both their biophysical and pharmacological properties. Additionally, GABA-evoked currents are greatly potentiated by 5α- and 5β-pregnan-3α-ol-20-one but show little or no acute modulation by sex steroids or corticosterone. Polyhydroxybutyrate (PHB) was produced by Ralstonia eutropha DSM 11348 (formerly Alicaligenes eutrophus) in media containing 20–30 g l−1 casein peptone or casamino acids as sole sources of nitrogen. In fermentations using media based on casein peptone, permanent growth up to a cell dry mass of 65 g l−1 was observed. PHB accumulated in cells up to 60%–80% of dry weight. The lowest yields were found in media without any trace elements or with casamino acids added only. The residual cell dry masses were limited to 10–15 g l−1 and did not contain PHB. The highest productivity amounted to 1.2 g PHB l−1 h−1. The mean molecular mass of the biopolymer was determined as 750 kDa. The proportion of polyhydroxyvalerate was less than 0.2% in PHB. The bioprocess was scaled up to a 300-l plant. During a fermentation time of 39 h the cells accumulated PHB to 78% w/w. The productivity was 0.98 g PHB l−1 h1. Instead of the conventional carbon sources used for propionic acid biosynthesis, the utilization of glycerol is considered here, since the metabolic pathway involved in the conversion of glycerol to propionic acid is redox-neutral and energetic. Three strains, Propionibacterium acidipropionici, Propionibacterium acnes and Clostridium propionicum were tested for their ability to convert glycerol to propionic acid during batch fermentation with initially 20 g/l glycerol. P. acidipropionici showed higher efficiency in terms of fermentation time and conversion yield than did the other strains. The fermentation profile of this bacterium consisted in propionic acid as the major product (0.844 mol/mol), and in minimal by-products: succinic (0.055 mol/mol), acetic (0.023 mol/mol) and formic (0.020 mol/mol) acids and n-propanol (0.036 mol/mol). The overall propionic acid productivity was 0.18 g l−1h−1. A comparative study with glucose and lactic acid as carbon sources showed both less diversity in end-product composition and a 17% and 13% lower propionic acid conversion yield respectively than with glycerol. Increasing the initial glycerol concentration resulted in an enhanced productivity up to 0.36 g l−1h−1 and in a maximal propionic acid concentration of 42 g/l, while a slight decrease of the conversion yield was noticed. Such a propionic acid production rate was similar or higher than the values obtained with lactic acid (0.35 g l−1h−1) or glucose (0.28 g l−1h−1). These results demonstrated that glycerol is a carbon source of interest for propionic acid production. Gamma linolenic acid (GLA) degradation in Spirulina followed first-order reaction kinetics. At an accelerated temperature range of 45 to 55°C, the degradation rate constants (k r) of GLA obtained were 4.0 × 10−2 to 8.8 × 10−2 day−1. The energy of activation (E a) was 16.53 kcal mol−1, and the Q10 was 2.22. Based on 20% GLA degradation, the shelf life of sun-dried Spirulina at 30°C is 263 days or 8.6 months using the Arrhenius plot, and 258 days or 8.5 months using the Q 10 approach. This study compared the cardiorespiratory responses of eight healthy women (mean age 30.25 years) to submaximal exercise on land (LTm) and water treadmills (WTm) in chest-deep water (Aquaciser). In addition, the effects of two different water temperatures were examined (28 and 36°C). Each exercise test consisted of three consecutive 5-min bouts at 3.5, 4.5 and 5.5 km · h−1. Oxygen consumption (O2) and heart rate (HR), measured using open-circuit spirometry and telemetry, respectively, increased linearly with increasing speed both in water and on land. At 3.5 km · h−1 O2 was similar across procedures [χ = 0.6 (0.05) l · min−1]. At 4.5 and 5.5 km · h−1 O2 was significantly higher in water than on land, but there was no temperature effect (WTm: 0.9 and 1.4, respectively; LTm: 0.8 and 0.9 l · min−1, respectively). HR was significantly higher in WTm at 36°C compared to WTm at 28°C at all speeds, and compared to LTm at 4.5 and 5.5 km · h−1 (P ≤ 0.003). The HR-O2 relationship showed that at a O2 of 0.9 l · min−1, HR was higher in water at 36°C (115 beats · min−1) than either on land (100 beats · min−1) or in water at 28°C (99 beats · min−1). The Borg scale of perceived exertion showed that walking in water at 4.5 and 5.5 km · h−1 was significantly harder than on land (WTm: 11.4 and 14, respectively; LTm: 9.9 and 11, respectively; P ≤ 0.001). These cardiorespiratory changes occurred despite a slower cadence in water (the mean difference at all speeds was 27 steps/min). Thus, walking in chest-deep water yields higher energy costs than walking at similar speeds on land. This data has implications for therapists working in hydrotherapy pools.
